The levels of 5-methylcytosine (5-mC) and 5-hydroxymethylcytosine (5-hmC) in DNA (5-mC-DNA and 5-hmC-DNA) are strongly correlated with cancer occurrence and development. The ability to distinguish and quantitatively detect them is important for cancer research. We have developed a hybridization chain reaction (HCR)-based electrochemical assay for the signal-amplified detection of the relative contents of 5-mC-DNA and 5-hmC-DNA. The DNA duplexes (containing 5-mC-DNA and 5-hmC-DNA with different percentages) were modified on a gold electrode. Electroactive [Ru(NH3)6]3+ (RuHex) was used as the signal reporter, because it binds to DNA double strands. The duplexes can be cleaved by MspJI endonuclease without HCR, and result in a small peak current. However, the cleavage can be blocked after the 5-hmC-DNA duplex is converted to β-glucosyl-5-hydroxymethylcytosine (β-glu-5-hmC) by T4 β-glucosyltransferase (T4 β-GT), and with the addition of helper DNA, a long double-helix DNA was formed through HCR. A significantly amplified peak current can be achieved due to the adsorption of numerous RuHex. The electrochemical signal of RuHex is correlated to the content of 5-hmC-DNA. Upon fixing the total quantity of 5-mC-DNA and 5-hmC-DNA on the electrode, the signals increase with the increase in the percentage of 5-hmC-DNA for the HCR. With this assay, a detection limit of 0.05% for 5-hmC-DNA was achieved.

Biomass-derived resources exploited in depth toward the synthesis of high value-added chemicals is very promising, but still limited by the applicable catalyst. In this paper, we report regulated robust multilayer-like polyoxometalates(Niobium molybdate) specific to the catalytic synthesis of promising 5-Ethoxymethylfurfural (EMF). The present solid acid catalysts resulted from precisely-tuned compositions of Nb and Mo toward acquiring controlled Brønsted acid amounts and interlayer space coupling with specific Lewis acid sites are demonstrated to be the determining factors for tuning conversion and selectivity in the synthesis of EMF, respectively. The characterization results indicate that the increment in Nb moiety is beneficial to produce more accessible acid sites and the enlarged interlayer space in the catalyst, thereby promoting the acid-catalyzed conversion. As a result, the full conversion(Conv. 100%) of 5-hydroxymethylfurfural into the 5-Ethoxymethylfurfural (Select. >99%) by etherification with ethanol within only 60 min was realized as the record-high yield under the solvent-free condition, far outperforming several traditional catalysts. In addition, the rational 67% yield of EMF was received when using fructose as an initial reactant. Ulteriorly, the resultant catalyst inherits the superior catalytic activity in the substrate expansion experiments in the presence of different alcohols. More importantly, the resulted catalyst shows durable catalytic activity without any activity-loss during the eight recycling, which reveals the huge potential industrial application prospect.

5-formylcytosine (5 fC) and 5-carboxylcytosine (5caC) serve as key intermediates in DNA demethylation process with significant implications for gene regulation and disease progression . In this study, we introduce a novel electrochemical sensing platform specifically designed for the sensitive and selective detection of 5 fC and 5caC in DNA . Protein A-modified magnetic beads (ProtA-MBs) coupled with specific antibodies facilitate the immunorecognition and enrichment of these modified bases. Signal amplification is achieved through several chemical reactions involving the interaction between N 3 -kethonaxl and guanine , copper-free click chemistry for the attachment of dibenzocyclooctyne (DBCO)-Biotin, and the subsequent recognition by streptavidin-conjugated horseradish peroxidase (SA-HRP). The assay's readout is performed on a disposable laser-induced graphene (LIG) electrode, modified with the bead-antibody-DNA complex in a magnetic field , and analyzed using differential pulse voltammetry in a system employing hydroquinone (HQ) as the redox mediator and H 2 O 2 as the substrate. This immunosensor displayed excellent sensitivity, with detection limits of 14.8 fM for 5 fC across a 0.1–1000 pM linear range and 87.4 fM for 5caC across a 0.5–5000 pM linear range, and maintained high selectivity even in the presence of interferences from other DNA modifications . Successful application in quantifying 5 fC and 5caC in genomic DNA from cell extracts, with recovery rates between 97.7% to 102.9%, underscores its potential for clinical diagnostics. N 3 -kethoxal was used for the first time in an electrochemical sensor. This work not only broadens the toolkit for detecting DNA modifications but also provides a fresh impetus for the development of point-of-care testing (POCT) technologies.

A high-performance liquid chromatography method with a diode array and an electrochemical detection (HPLC-ECD/DAD) was developed to determine the levels of tryptophan (TRP), kynurenine (KYN), kynurenic acid (KYA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in rat plasma. The prepared samples were separated on a BDS column (4.6 mm × 250 mm, 5 mm) with column oven temperature of 25 °C. The mobile phase consisted of 5% acetonitrile and a buffer solution, which contained 25 mmol/L sodium acetate and 0.01 mmol/L EDTA, adjusting pH to 4.5 with acetic acid, and it was pumped at a flow-rate of 1.0 mL/min. KYN and KYA were measured by a variable wavelength detector at wavelengths 360 nm and 333 nm respectively, TRP and vanillic acid (as IS) both were measured at 280 nm. Determination of 5-HT and 5-HIAA was accomplished at the electrochemical working potential of 700 mV. Total run time was 14 min. Several parameters of the developed method were validated including linearity, accuracy precision, and stability. The results showed the established method had good LOD and separation for all of the five compounds and IS in the biological matrix. The method is simple, fast, economical and accurate. The analytical method and the results could provide a reference for the clinical and scientific research of depression.

The reaction of tributylphosphane with a 7-(2′-formyl-2-biphenylyl)phosphanorbornadiene P-W(CO) 5 complex leads to the 5-phosphaphenanthrene P-W(CO) 5 complex via an intramolecular phospha-Wittig condensation. This complex is stable enough for detection by 31 P NMR spectroscopy but cannot be isolated due to its high reactivity. It is trapped by addition of MeOH or cycloaddition with 2,3-dimethylbutadiene or a nitrileimine. The adducts were characterized by X-ray crystal structure analysis. Graphical The reaction of 4 with tributylphosphane at room temperature gives phosphaphenanthrene complex 5 , which can be detected by 31 P NMR and trapped by addition, and [2+4] or [2+3] cycloaddition.

In order to overcome the technical bottlenecks associated with the traditional synthesis of 5-nitrosalicylic acid, this study constructs a continuous nitration system using a microchannel reactor. Systematic process optimization of the nitration reaction of salicylic acid is conducted, focusing on the influence of reactor channel type, material ratio, residence time, and reaction temperature on product yield. The optimal process parameters identified are as follows: umbrella-shaped microchannel reactor is employed, using acetone as the solvent with a mass ratio of salicylic acid to acetone of 1:5.0. The molar ratio of salicylic acid to HNO 3 to H 2 SO 4 is controlled at 1:4:0.5. Under the conditions of 60 °C and a residence time of 4 min for continuous reaction, the product yield reaches 85.9% after recrystallization and purification with a 75% ethanol aqueous solution, achieving a purity of 97.2% as determined by liquid chromatography. Compared to traditional batch nitration processes, this method significantly reduces the consumption of concentrated sulfuric acid and substantially shortens reaction times, providing a technical reference for the safe and environmentally friendly industrial production of 5-nitrosalicylic acid.

Applications of terahertz time-domain spectroscopy (THz-TDS) in the fields of chemistry and biomedicine have recently received increased attention. Specifically, THz-TDS is particularly effective for the identification of alkaloid molecules, because it can distinguish the vibration types of base molecules in the THz band and provide a direct characteristic spectrum for the configuration and conformation of biomolecules. However, when THz-TDS technology is used to identify alkaloid molecules, most of them are concentrated in the 0.1–3.0 THz band, limiting the amount of information that can be obtained. In this work, a wide-spectrum THz-TDS system was independently built to explore the absorption spectra of uracil and its 5-substituents in the range of 1.3–6.0 THz. We found that, in the THz band, uracil and its 5-substituents have similar absorption peaks near 4.9 and 3.3 THz, while the 5-substituents have an additional absorption peak in the range of 1.5–2.5 THz. This absorption peak is red-shifted as the relative atomic mass of the 5-substituted atoms increases. Gaussian software was adopted to calculate the absorption spectra of the samples. The simulation conclusions were in good agreement with the experimental results, revealing that the vibration of the base molecule at low frequencies can be attributed to the inter-molecular vibration. This work demonstrates that THz-TDS technology can be used to accurately identify biomolecules with similar molecular structures, reflecting the importance of molecular structure in biological activity. Keywords:terahertz time domain spectroscopy;absorption peaks;5-substituents of uracil;vibration;broad spectrum;molecular structures

5-Hydroxymethylfurfural (5-HMF) is a heterocyclic compound with six carbons commonly found in heat-treated carbohydrate-rich foods. 5-HMF exceeding the specified limit is cytotoxic to the human body, and will be converted into carcinogenic substances (5-sulfoxide methyl furfural) after long-term accumulation in the body. Therefore, it is highly necessary to develop a sensitive and accurate detection method for 5-HMF in the field of food safety. In this study, a photoelectric sensing method was developed for the highly sensitive detection of 5-HMF using hollow TiO2 nanospheres successfully synthesized by template, sol–gel and lye etching methods. The structure and composition of the materials were studied by XRD, XPS, SEM and TEM. The electrochemical and photoelectrochemical properties of an h-TiO2 electrode probe based on indium tin oxide (ITO) slides were investigated. The results indicated that the linear relationship of 5-HMF is good in the concentration range of 10−11–10−7 M, and the detection limit of 5-HMF is 0.001 nM. Moreover, the PEC sensor shows high accuracy in the detection of actual samples.

Cu/zeolite catalysts have attracted considerable attention for cold-start applications; however, their efficiency in propylene adsorption remains limited. To address this challenge, Cu/ZSM-5 catalysts with enhanced metal-support interactions were successfully constructed via a seed-assisted, template-mediated synthesis method, leading to improved propylene capture and oxidation efficiency. The catalysts were systematically characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR), N 2 adsorption-desorption, scanning electron microscope (SEM), transmission electron microscope (TEM), NH 3 -TPD, X-ray photoelectron spectroscope (XPS), and in situ infrared spectroscopy to investigate the influence of acid site density and metal-support interactions on propylene adsorption behavior. The results demonstrated that Cu@ZSM-5 catalysts synthesized with different structure-directing agents exhibited distinct acidity profiles and varying strengths of metal-support interaction. Among them, the Cu@ZSM-5-130 sample prepared using the TPAOH template showed a high specific surface area (474 m 2 /g) and a more optimal acidity balance. It was found that an excessive density of strong acid sites can be detrimental to propylene adsorption and oxidation. The intimately associated Cu species within the zeolite matrix facilitated selective adsorption of hydrocarbons, particularly propylene. In situ infrared spectroscopy further revealed a temperature-dependent oxidation pathway of propylene over the Cu@ZSM-5-TPAOH-130 catalyst, starting with epoxide formation at low temperatures, progressing to partial oxidation products ( e.g. , acrolein) at intermediate temperatures, and ultimately yielding complete oxidation products, such as CO 2 and H 2 O at higher temperatures. The important bio-based platform chemical 5-Hydroxymethylfurfural (HMF) can be oxidized for use in several products that serve as versatile building blocks of polymers. Previously, we had demonstrated that Pseudomonas aeruginosa PC-1 can selectively produce 5-hydroxymethyl-2-furancarboxylic acid (HMFCA) from 800 mM HMF, with a yield greater than 90%. In this study, an aldehyde dehydrogenase (PaALDH64) was identified from P. aeruginosa PC-1 for selective HMF oxidation to HMFCA. The HMF tolerance level of E. coli /pET- PaALDH64 was over 300 mM, which was greater than the tolerance levels of all other reported enzymes. The proposed biocatalyst could be used to catalyze the biotransformation of other aldehydic furans, with carboxylic acid yields over 92%. Molecular docking revealed that the catalytic active site cleft in PaALDH64 formed more hydrogen bonds with HMF compared to other enzymes, which stabilized the enzyme–substrate interaction and enhanced the catalytic efficiency. In a fed-batch process, 547.8 mM HMFCA was produced from 600 mM HMF by E. coli /pET- PaALDH64 within 19 h only, which showed an excellent HMF conversion rate. Overall, PaALDH64 constitutes a promising enzyme for the industrial production of HMFCA.

BACKGROUND The conversion of glucose into 5-hydroxymethylfurfural (5-HMF) involves the isomerization of glucose to fructose catalyzed by Lewis acid and subsequent dehydration of fructose catalyzed by Brönsted acid. In this work, in order to obtain a high yield of 5-HMF, a series of Sn-Al-Beta catalysts containing both Lewis and Brönsted acid sites were prepared by a convenient solid state ion exchange (SSIE) method using partially dealuminated Beta zeolite and tin(II) acetate. RESULTS The obtained bifunctional catalysts were investigated for one-pot production of 5-HMF from glucose in the ionic liquid (IL) [C 4 mim]Cl. Among various catalysts, Sn-Al-Beta- 4 - 8 exhibited the best catalytic performance, giving a 5-HMF yield of 54% under optimal reaction conditions (393 K, 2 h), along with a glucose conversion of 81%. The Sn-Al-Beta- 4-8 catalyst and IL were regenerated by the extraction of 5-HMF, and the reactivity was reserved after five runs of recycling. In addition to glucose, the Sn-Al-Beta- 4-8 /[C 4 mim]Cl system was also proven to have the potential to effectively convert other carbohydrates into 5-HMF. CONCLUSION The results suggested that the catalyst is convenient to prepare and has preferable efficiency and stability in the conversion of carbohydrates into 5-HMF. © 2022 Society of Chemical Industry (SCI).
