Cellulose nanocrystals (CNCs) are inherently right-handed nanostructures that originate from nature, showing chirality in their fibrils, bundles, and self-assembled films. However, the enantio-specific interaction between CNCs and other chiral molecules has not been explored so far. In this study, we first demonstrated a chirality-related difference in the composite films of cellulose nanocrystals and histidine with a D- or L-configuration. The distinction is not only presented in the self-assembled nanostructures of CNCs, optical properties, and the thermal decomposition of composites but also in the crystallization of the amino acid. We suppose that it might have originated from the packing of amino acids on the twisted surface of CNCs. The knowledge about the enantio-specific interaction between the chiral amino acid and polysaccharide nanostructure is of significant importance for developing a new strategy for enantiomeric separation.

The synthesis of d -isoascorbyl stearate from d -isoascorbic acid and stearic acid with immobilized lipase (Novozym ® 435) as catalyst was studied. Response surface methodology and Box-Behnken design with six variables and three levels were employed to evaluate the effects of processing conditions on the conversion of d -isoascorbic acid. The results confirmed that the response surface method and statistical analysis were proved to be useful in developing optimal conditions for d -isoascorbyl stearate synthesis. The optimum conditions were predicted as follows: reaction temperature 48 °C, reaction time 17.7 h, immobilized lipase amount 50.0 % (w/w, of d -isoascorbic acid), substrate molar ratio 9:1 (stearic acid to d -isoascorbic acid), d -isoascorbic acid concentration 0.14 mol/L (based on solvent), 4A molecular sieve addition 200 g/L (based on solvent), and the optimal conversion was 90.6 %. Through the kinetics model fitting of the esterification, it was considered that the esterification conformed to a Ping–Pong bi–bi kinetic model with d -isoascorbic acid inhibition, and the obtained kinetic constants showed that the inhibition of d -isoascorbic acid and the enzyme affinity to substrate were abate with the increase of the reaction temperature.

The reaction kinetics of lithium-sulfur (Li-S) battery mainly depends on the capture and catalytic conversion efficiency of lithium polysulfide (LiPSs). The unique electron configuration of the 5d metal and the high electrical conductivity of MXene can enhance the interaction between the catalyst and LiPSs. However, the challenge of enhancing the adsorption of LiPSs by regulating the electronic structure of the catalyst with 5d metal and MXene remains unresolved and obscure. In this work, we have developed molybdenum disulfide (MoS 2 )-tungsten disulfide (WS 2 ) @MXene (MoS 2 -WS 2 @MX) heterostructure catalysts for separator coatings in Li-S batteries by introducing W and MXene for the first time. Theoretical calculations demonstrate that the 5 d metallic and conductive MXene coupling effectively modulate the orbital hybridization degree between the catalyst and LiPSs, thereby reducing the energy barrier for LiPSs conversion. Electrochemical test and in situ tests verify the effectiveness of the strategy on the adsorption and conversion of LiPSs. Consequently, the MoS 2 -WS 2 @MX based batteries exhibit high initial reversible capacity of 950.3 mAh g −1 and low capacity decay of 0.066 % per cycle over 500 cycles at 2 C, which is superior to most reported catalysts. This study provides a dual regulatory strategy for enhancing catalytic activity, offering valuable insights for the advancement of high-performance Li-S batteries.

The development of STING inhibitors for the treatment of STING-related inflammatory diseases continues to encounter significant challenges. The activation of STING is a multi-step process that includes binding with cGAMP, self-oligomerization, and translocation from the endoplasmic reticulum to the Golgi apparatus, ultimately inducing the expression of IRF3 and NF-κB-mediated interferons and inflammatory cytokines. It has been demonstrated that disruption of any of these steps can effectively inhibit STING activation. Traditional structure-based drug screening methodologies generally focus on specific binding sites. In this study, a TransformerCPI model based on protein primary sequences and independent of binding sites is employed to identify compounds capable of binding to the STING protein. The natural product Licochalcone D (LicoD) is identified as a potent and selective STING inhibitor. LicoD does not bind to the classical ligand-binding pocket; instead, it covalently modifies the Cys148 residue of STING. This modification inhibits STING oligomerization, consequently suppressing the recruitment of TBK1 and the nuclear translocation of IRF3 and NF-κB. LicoD treatment ameliorates the inflammatory phenotype in Trex1 −1− mice and inhibits the progression of DSS-induced colitis and AOM/DSS-induced colitis-associated colon cancer (CAC). In summary, this study reveals the potential of LicoD in treating STING-driven inflammatory diseases. It also demonstrates the utility of the TransformerCPI model in discovering allosteric compounds beyond the conventional binding pockets.

D-Allose, a rare sugar, has gained significant attention not only as a low-calorie sweetener but also for its anticancer, antitumor, anti-inflammatory, antioxidant, and other pharmaceutical properties. Despite its potential, achieving high-level biosynthesis of D-allose remains challenging due to inefficient biocatalysts, low conversion rates, and the high cost of substrates. Here, we explored the food-grade coexpression of Blautia produca D-allulose 3-epimerase (Bp-DAE) and Bacillus subtilis L-rhamnose isomerase (BsL-RI) within a single cell using B. subtilis WB800N as the host. Using this system, D-allose was synthesized via a simple, cost-effective, one-pot enzymatic process, employing whole cells as catalysts and D-fructose as the substrate. The system exhibited optimal activity at 65 °C, pH 8.5, with 1 mM Mn2+ and 20 g/L of whole-cell dry weight. Initial production reached 12.5 g/L D-allose with a 12.5% yield from 100 g/L D-fructose. Optimization of dual promoter combinations enhanced production, achieving 15.0, 29.1, and 43.2 g/L D-allose from 100, 200, and 300 g/L D-fructose, with yields of 15.00, 14.55, and 14.40%, respectively. This D-allose production biocatalyst offers a scalable and economically viable platform for the industrial production of rare sugar.

Oleanolic acid (OA) is a triterpenoid found in various fruits and vegetables and used in traditional Chinese medicine. OA plays a crucial role in the treatment of several cancers, but poor water solubility, low permeability, and significant efflux have limited its widespread clinical use. Vitamin E-d-α-tocopheryl polyethylene glycol succinate (vitamin E-TPGS) and Pluronic P105 were used to improve the solubility and permeability and to decrease the efflux of OA. OA-loaded mixed micelles were prepared by ethanol thin-film hydration. The physicochemical properties of the micelles, including zeta potential, morphology, particle size, solubility, drug loading, and drug entrapment efficiency were characterized. OA release from micelles was slower than that from the free drug system. OA uptake by A549 non-small-cell lung cancer (NSCLC) cells was enhanced by the micelles. A tumor model was established by injecting A549 cells into nude mice. In vivo imaging showed that OA-micelles could accumulate in the tumors of nude mice. Additionally, smaller tumor size and increased expression of pro-apoptotic proteins were observed in OA-micelle-treated mice, indicating that OA-micelles are more effective than free OA in treating cancer. In vitro experiments were performed using two NSCLC cell lines (A549 and PC-9). Cytotoxicity evaluations showed that the half-maximal inhibitory concentrations of free OA and OA-micelles were 36.8±4.8 and 20.9±3.7 μM, respectively, in A549 cells and 82.7±7.8 and 56.7±4.7 μM, respectively, in PC-9 cells. Apoptosis assays revealed that the apoptotic rate of OA-micelle-treated A549 and PC-9 cells was higher than that of cells treated with the same concentration of free OA. Wound healing and transwell assays showed that migration and invasion were significantly suppressed in OA-micelle-treated cells. Immunofluorescence and Western blot analyses confirmed that the epithelial–mesenchymal transition was reversed in OA-micelle-treated cells. Mixed micelles are a promising nano-drug delivery system for lung cancer treatment.

Pteris cretica L. var. nervosa is one of the most well-known Chinese medicines. Although it is widely used to treat jaundice hepatitis, the main ingredient for its treatment was not thoroughly explored until recently. Essentially, the purpose of this study is to find the monomer compound in Pteris cretica L. var. nervosa , which is most likely to be effective in treating liver injury. Through the model of LPS/D-gal-induced liver injury in mice, the best therapeutic site of the total extract was explored, the chemical components of the parts with the best therapeutic effect were separated, a total of 10 flavonoids were isolated, and the RAW264.7 cells induced by LPS were used as the experimental model to explore the preliminary anti-inflammatory activity of NO production in vitro. Finally, the anti-inflammatory activity and the highest content in this plant Luteolin-7-O-rutinoside (LUT) were selected, as the object of study in vivo. It was found that LUT could not only reduce alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, but also significantly reduce the release of tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-1β (IL-1β), and inhibit PI3K/AKT/AMPK/NF-κB pathway. In addition, LUT can increase levels of SOD and GSH to reduce oxidative stress. It has an obvious therapeutic effect on acute liver injury induced by LPS/D-gal in mice. Therefore, infer LUT is a functional substance in Pteris cretica L. var. nervosa. Graphical abstract

Chlorogenic acids are important phenolics in the fruits of wolfberry, but little attention has been paid on their glucosylated forms. In the present study, a glucosylated form of chlorogenic acid was isolated from the fruits of Lycium barbarum L. var. auranticarpum K. F. Ching (also called yellow wolfberry) and identified to be (-)-5- O -(3- O -β- d -glucopyranosylcaffeoyl)-quinic acid (5-CQA-3′βG) by high resolution mass spectrometry and nuclear magnetic resonance spectrometry. The content of 5-CQA-3′βG in the dried fruit was determined as 0.0293 ± 0.0015% by HPLC. In addition, 5-CQA-3′βG showed a good scavenging capacity for 2,2′-azino-bis-(3-ethylben-zothiazoline-6-sulphonate) free radicals but had a relatively low reducing power and scavenging capacity for 2,2-diphenyl-1-picrylhydrazyl free radical. Moreover, the secretion of nitric oxide, tumor necrosis factor-α and interleukin-6 as well as related mRNA expression were reduced in lipopolysaccharide-stimulated RAW264.7 macrophage cells treated with 5-CQA-3′βG. This is the first report describing purification, identification and bioactivity of glucosylated CQA from yellow wolfberry.

Although lithium–oxygen batteries (LOBs) hold great promise as future energy storage systems, they are impeded by insulated discharge product Li 2 O 2 and sluggish oxygen reduction reaction/oxygen evolution revolution (ORR/OER) kinetics. The application of a highly efficient cathode catalyst determines the LOBs performance. The d-band modulation and catalytic kinetics promotion are important concept guidelines for the performance enhancement of cathode catalysts. In this work, the homogeneous in-plane distortion-derived synergistic catalytic capability of an Ag 2 Mo 2 O 7 catalyst with modulated d-band centers and promoted ORR/OER kinetics is demontrated. The uniform elongation of Ag─O bonds and compression of Mo─O bonds in (020) plane leads to d-band splitting and d-band center optimization and delivers improved adsorption behavior for high ORR/OER capability. Furthermore, the spatial and energy overlap of Ag d xz and O 2 anti-bonding π * orbitals facilitate electron injection during ORR process and reduce the energy barrier for charge transfer and O 2 desorption during OER process, accelerating the ORR/OER kinetics. As a result, the (020) plane-exposed Ag 2 Mo 2 O 7 cathode exhibits ultralong cycle stability of 817 cycles at 500 mA g −1 and large specific discharge/charge capacities of 15898/15180 mAh g −1 . This work provides facile concept guidance for optimizing catalytic capability through controlled lattice distortion in cathode catalysts for LOBs.

The development of 1-deoxy-d-xylulose-5-phosphate synthase (DXPS) inhibitors still needs an innovative extension with novel chemotypes to avoid herbicide resistance. Herein, a series of 2,4-diphenylquinoline derivatives were designed, synthesized, and structurally optimized from lead compound 24 via stepwise A/B/C-ring modification. Greenhouse assays identified a-3 (3-CF3 A-ring) as the most potent analogue, exhibiting the higher herbicidal activity (71.4% inhibition on DESSO at 375 g a.i./ha) and dosage tolerance than 24. The mechanism of compound a-3 targeting DXPS proteins has been also identified using surface plasmon resonance binding assay, physiological biomarker measurement, substrate accumulation, and molecular docking. The key active motifs between a-3 and DXPS have been further defined based on the structure–activity relationships. This work will further expand the structural diversity of DXPS inhibitors with higher bioactivity.

Enzyme–inorganic hybrid nanoflowers (HNFs) are promising immobilization platforms because of their simple fabrication and often enhanced catalytic performance. Here, a robust and highly efficient hybrid nanoflower (β-Gal-SC@ST-LFAI) for d -tagatose biosynthesis was developed using co-immobilizing β-galactosidase (β-Gal) and l -arabinose isomerase (LFAI) by ST/SC–mediated self-assembly and biomineralization. First, the effects of key immobilization conditions on the fabrication of β-Gal-SC@ST-LFAI were investigated. Under optimal conditions, β-Gal-SC@ST-LFAI exhibited higher catalytic activity and activity recovery than a conventional β-Gal@LFAI. Compared with the free enzyme system, β-Gal-SC@ST-LFAI showed enhanced thermal stability, pH tolerance and organic solvent resistance. The structure of β-Gal-SC@ST-LFAI was further characterized by SEM, CLSM, DLS, FTIR, CD and BET technology. The resulting HNFs displayed a typical flower-like architecture with an average diameter of 1.83 μm and specific surface area of 18.57 m 2 ·g −1 . CD spectroscopy showed a decrease in α-helix content and an increase in β-sheet content in β-Gal-SC@ST-LFAI compared to the free cascade enzymes. In addition, β-Gal-SC@ST-LFAI exhibited a lower K m than the free cascade enzymes, indicating enhanced substrate affinity. The HNFs also demonstrated better storage stability than the free enzymes and excellent operational stability, retaining more than 77.99% of their cascade catalytic activity after seven reuse cycles. Finally, β-Gal-SC@ST-LFAI can catalyze the conversion of d -lactose to d -tagatose with a yield of 37.22%. This study provides a practical strategy for constructing multienzyme HNFs with high catalytic efficiency and process robustness.

Pseudomonas aeruginosa , a significant opportunistic pathogen, poses a serious threat to food safety due to its antibiotic resistance and strong biofilm-forming ability. In this study, quercetagetin (Que) was identified as a novel quorum sensing (QS) inhibitor. Reporter strain assays, molecular docking, and dynamic simulation indicated that Que acts as an antagonist of the QS receptors. Que attenuated QS-regulated virulence phenotypes in a dose-dependent manner but showed insufficient antibiofilm activity ( p >0.05) at sub-minimum inhibitory concentration (sub-MIC) levels. In contrast, the combination of 0.5 mmol/L Que with 20 mmol/L D-leucine or D-tryptophan exhibited additive effects on planktonic cells, reducing its survival by 2.19–3.97 log compared to the control. For biofilm cells, the combined treatment potentiated the reduction in cell density by 0.48–1.42 log. Notably, these effects were strain-dependent but were equal to or outperformed those of the ε-polylysine (ε-PL) control. Mechanistically, this enhancement was driven by the disruption of biofilm matrix and cell membrane integrity. This was evidenced by the greater suppression of Acyl-homoserine lactones (AHLs) synthesis and virulence phenotypes, alongside substantial reductions in biofilm biomass and roughness and increased in membrane permeability compared to Que. Furthermore, the combination resulted in repression of key enzymes activities (Na + -K + -ATPase and succinate dehydrogenase) by 29%–38%. These changes indicated a promising strategy to control P. aeruginosa and enhance food safety.
