Integration of Raman spectroscopy and machine learning for species-specific assessment of fish freshness with insights into biogenic amines

Yuxin Zhang,Xinping Lin,Sufang Zhang,Ningbo Qin,Chaofan Ji

Ensuring fish quality and safety is critical for protecting consumer health, particularly with respect to freshness loss and biogenic amines (BAs) accumulation. In this study, Raman spectroscopy was integrated with machine learning to enable rapid, non-destructive assessment of fish freshness and prediction of BAs. Raman spectra were collected from five fish species at different freshness stages and analyzed using a combination of unsupervised and supervised learning approaches to discriminate freshness levels and species. Spectral analysis revealed prominent freshness markers, characterized by intensity changes in specific Raman bands, particularly the Amide I band (1654 cm −1 ) and aliphatic C-H stretching vibrations (1451 cm −1 ), which were closely associated with protein degradation and lipid oxidation during storage and contributed significantly to freshness discrimination, and were further supported by two-dimensional correlation spectroscopy, confirming their sequential evolution during spoilage. Compared with conventional chemometric methods, machine learning models demonstrated superior classification performance, with a convolutional neural network model achieving the highest accuracy of 94.80%. For safety assessment, partial least squares regression and support vector regression models were developed to predict BAs, both exhibiting strong predictive performance (Rp > 0.9). Based on histamine concentrations, samples could be reliably classified into three categories: high histamine (>50 mg/kg), medium histamine (10-50 mg/kg) and low histamine (<10 mg/kg), with classification accuracies of 80.00%, 88.23% and 100.00%, respectively. Overall, the integration of Raman spectroscopy with machine learning provides an effective framework for rapid evaluation of fish freshness and early identification of safety risks, supporting quality monitoring and control in aquatic products.

Toxicity evaluation of MC-LR in different fish organs based on aptamer-recognized SERS tag coupled with magnetic separation

Wei Yue,Bingyong Lin,Yueyue Huang,Yueliang Wang,Yuanyuan Yao,Lifen Chen,Yanbo Zeng,Lei Li,Zhaosheng Qian,Longhua Guo

Microcystin-LR (MC-LR) is a strong liver toxin, which will enter the human body by eating MC-LR-contaminated aquatic products. Therefore, it is urgent to explore whether people are exposed to MC-LR-contaminated aquatic products. Herein, a sensing strategy based on aptamer-recognized SERS tag coupled with magnetic separation was developed for ultrasensitive and specific detection of MC-LR in fish organs. This sensor had a dynamic range of 0.1 ng/L to100 µg/L and a limit of detection down to 0.1 ng/L for MC-LR. This ultrasensitive and rapid separation strategy allow us to explore the bioaccumulated amount of MC-LR in different fish organs. The results show that the order of MC-LR content accumulated in different organs of local crucian carps were heart > liver > kidney > meat > spleen. The MC-LR content in fish meat was 1.1–8.5 times of the WHO proposed tolerable daily intake level (TDI), implying the aquatic products in local aquatic market had potential threats to human health.

Characterization of flavor perception and characteristic aroma of traditional dry-cured fish by flavor omics combined with multivariate statistics

Fangxue Chen,Lingwei Shen,Xujia Shi,Yi Deng,Yu Qiao,Wenjin Wu,Guangquan Xiong,Lan Wang,Xin Li,Anzi Ding,Liu Shi

Traditional dry-cured fish has a rich aroma and has been favored by consumers. In this study, headspace solid-phase microextraction (HS-SPME) combined with gas chromatography-olfactory-mass spectrometry (GC-O-MS) was used to identify the aroma components of traditional dry-cured fish. A total of 39 volatile compounds were identified by GC-O-MS, and 17 compounds had odor activity values greater than 1. Aroma recombination model was prepared by mixing 17 volatile compounds from sun-dried fish with an odorant activity value greater than 1 in an odorless matrix. The odorless base had a good similarity to the aroma of dry-cured fish. Seven sensory attributes were selected to describe the aroma of dry-cured fish, namely fishy, fatty, meaty, grassy, ground, rancid, and metallic. The electronic nose and multivariate statistical results showed that there were differences in the aroma of dried cured fish with different traditional drying methods. Sun-dried fish had a strong oily, meaty and metallic taste, while shade-dried fish had strong fishy, grassy, earthy, and rancid odors. Omission tests indicated that hexanal, nonanal, octanal, heptanal, 2-octanal,(E)-, 2-nonenal,(E), 3-methylbutanal, pentanol, hexanol, nonanol, heptanol, 1-octen-3-ol, 2-octen-1-ol,(E)-, 2-pentyl furan and d -limonene are characteristic aroma compounds of dry-cured fish.

A Simple and Selective Colorimetric Aptasensor for Detection of Toxins Microcystin-LR in Fish Tissue Using a Truncated Aptamer

Feng Junli,Wu Yuanyuan,Shen Qing

In the present study, a sensitive and simple approach for colorimetric detecting microcystin-LR (MC-LR) in fish tissue was developed based on gold nanoparticles (AuNPs) and a truncated aptamer Apt-910–2, which was obtained by mutation and rational truncation from the previously selected 60-mer one. The 23-mer Apt-910–2 presented improved binding affinity to MC-LR and higher specificity. Under the optimum experimental conditions, the Apt-910–2 colorimetric aptasensor worked well in 10 to 150 ng/mL MC-LR concentration ranges, and the limit of detection (LOD) was 0.38 ng/mL. This Apt-910–2 colorimetric aptasensor was also applied to spiked fish tissue samples and gave recovery rates ranged from 75.18 to 87.72%, with the visual LOD 10 μg/kg in fish tissue judged by the naked eye. Thus, this study provides a rapid, reliable, and convenient approach for visual detection of MC-LR, which is of great significance for food safety control by assessing potential health risks related to seafood consumption.

Branched poly(ethylenimine) carbon dots-MnO2 nanosheets based fluorescent sensory system for sensing of malachite green in fish samples

Xiaowei Mu,Xin Liu,Xiwen Ye,Wei Zhang,Lu Li,Pinyi Ma,Daqian Song

Malachite green (MG) is an organic dye compound that is frequently used as a fungicide and antiseptic in aquaculture. However, human or animal exposure to MG causes carcinogenic, teratogenic and mutagenic effects. Herein, a novel fluorescent assay was designed for the detection of MG using manganese dioxide nanosheets (MnO 2 NS) as an energy acceptor to quench the fluorescence of branched poly(ethylenimine) carbon dots (BPEI-CDs) via Förster resonance energy transfer. When butyrylcholinesterase is introduced to form thiocholine in the presence of S-butyrylthiocholine iodide, MnO 2 NS can be recovered by thiocholine to Mn 2+ , resulting in restoration of the fluorescence of BPEI-CDs. Exploiting these changes in fluorescence intensity in the above system, a fluorescence probe was successfully developed for the quantitative detection of MG. Besides, this assay was applied to fish samples, verifying the high potential for practical application of the proposed sensor for the monitoring of MG in aquatic products.

Active polylactic acid/tilapia fish gelatin-sodium alginate bilayer films: Application in preservation of Japanese sea bass (Lateolabrax japonicus)

Jingwen Chen,Yinxuan Li,Yifen Wang,Salimata Yakubu,Haibing Tang,Li Li

Most edible films/coatings including those made of gelatin cannot be used independently as packaging films. In this study, active fish gelatin-based bilayer packaging films were developed and applied to preserve Japanese sea bass. The coating method was applied to produce polylactic acid/tilapia fish gelatin-sodium alginate bilayer film (PLA/FGSA). β -cyclodextrin inclusion complexes of cinnamaldehyde ( β -CD-CI) and thymol ( β -CD-Ty) were added into the gelatin-based layer of the PLA/FGSA to obtain active bilayer films (PLA/FGSA-CI and PLA/FGSA-Ty). This combination improved the oxygen and water vapor barrier abilities of the films, compared with PLA and edible gelatin-based monolayer films. Both active bilayer films possessed strong antibacterial and antioxidant activities . PLA/FGSA-Ty film showed the most effective inhibition against the growth of Pseudomonas .spp and H 2 S-producing Shewanella , generation of total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) of fish meat. These results indicate the active bilayer films are promising in the preservation of seafood.

Simultaneous determination of the residues of anesthetics and sedatives in fish using LC-QLIT-MS/MS combined with DSPE

Qin Huang,Hua Zhou,Xianglun Wu,Chenli Song,Jiangang Zheng,Meikang Lei,Pengqian Mu,Pinggu Wu

Liquid chromatography coupled with quadrupole linear ion trap tandem mass spectrometry (LC-QLIT-MS/MS) technology operated in electrospray ionization (ESI) was developed for tracing anesthetic (AETs) and sedatives (SDTs) in fish. Sampling procedure was achieved by using acetonitrile extraction followed by dispersive solid phase extraction (DSPE) clean-up. Under the optimized laboratory conditions, reliable qualitative confirmation was obtained through the multiple reaction monitoring-information dependent acquisition-enhanced product ion (MRM-IDA- EPI ) mode. Results indicated a favorable linear in the concentration range of 1–100 μg∙kg −1 (0.1–10 μg∙kg −1 for MS-222), with regression coefficient not less than 0.9997. The detection limit ranges from 0.03 to 0.4 μg∙kg −1 (S/N = 3). The validated method was applied to determine AETs and SDTs in fish with satidfied recoveries of 86.3 %–111.7 % and the relative standard deviations (RSD) of 1.9 %–8.9 % (n = 6). Practical samples analysis indicated that the proposed method is simple, rapid, sensitive and accurate for identification of AETs and SDTs.

Circulating exosome level of indigenous fish may be a novel biomarker for the integrated ecotoxicity effect of water environment

Meilin Tang,Ying Chen,Hongyi Xian,Suqin Tan,Zhenwei Lian,Xiaowu Peng,Dalin Hu

The deficiency of effective biomarker for the toxic effects of water pollutants greatly limits the application of biological monitoring. This study aimed to investigate the possibility of circulating exosomes of indigenous fish acting as biomarker for the ecotoxicity effect of water environment. The Helong Reservoir in Guangzhou, China, was chosen as the investigating field, of which the water quality belongs to Class V (2013) (GB 3838-2002, China). The clean drinking water source of the upper reaches of the Liuxihe Reservoir was selected as the control. Indigenous fishes including Oreochromis niloticus (Nile tilapia), Labeo rohita (Rohu), Carassius auratus (Crucian carp) were sampled during the period from July 2020 to April 2021. Circulating exosomes of fish samples were isolated by using ultracentrifugation, characterized with transmission electron microscopy (TEM) and quantified by using bicinchoninic acid (BCA) assay. Oxidative stress, DNA and chromosome damage in liver, kidney, brain, gill and blood of fish samples were measured. The results showed that there were significant differences in superoxide dismutase (SOD) activity, glutathione (GSH) and malondialdehyde (MDA) contents, DNA and chromosome damage in fish samples between the Helong Reservoir and the control. Interestingly, there were also significant differences in circulating exosome levels of fish samples between them. Our data suggested that circulating exosome level of indigenous fish may be a novel biomarker for the ecotoxicity effects of water environment.

Sulfur-doped reduced graphene oxide@chitosan composite for the selective and sensitive electrochemical detection of Hg2+ in fish muscle

Qingxia Ran,Fangfang Sheng,Guorui Chang,Min Zhong,Shuxia Xu

In this study, a glassy carbon electrode was modified with sulfur-doped reduced graphene oxide (S-rGO) and then combined with chitosan (CTS) to produce an electrochemical sensor. There is a favorable interaction between Hg 2+ and S-rGO, due to the strong affinity between sulfur and mercury. This effect was exploited for the selective and sensitive electrochemical determination of trace amounts of Hg 2+ in fish muscle samples by differential pulse anodic stripping voltammetry (DPASV). The factors that can affect the sensitivity and precision of the modified electrode were investigated through the optimization of various parameters, such as S-rGO concentration, solution pH, deposition potential, accumulation potential, accumulation time, and supporting electrolytes. By introducing S-rGO to enrich and amplify the electrochemical signal, the sensing strategy allowed to detect a range from 0.125 to 6 μM Hg 2+ with a detection limit of 1.6 nM. The sensor also showed excellent selectivity towards Hg 2+ in the presence of interfering metal ions, such as Fe 3+ , Zn 2+ , Mg 2+ , Mn 2+ , As 3+ , Na + , Cd 2+ and CO 2+ . The practical feasibility of the proposed sensor was assessed in fish muscle samples with satisfactory results compared with that of atomic fluorescence spectrometry. Moreover, this work demonstrated a simple and reliable method for improving the electrochemical performance of Hg 2+ sensor, which paves the way for many promising potential applications in the field of environmental testing.

Natural interface guiding cell: Directly using waste fish scales with rich micro/nano structures for control of cell behaviors

Jiao Yang Lu,Fu Rui Zhang,Xue Zhi Ding,Li Qiu Xia,Wei Tao Huang

Extracellular matrix (ECM) with various micro/nano features is an important chemical and physical clue to control cell behaviors. Fish scale surface has rich structures and similar components to ECM, but there are no reports on directly using the natural surface of fish scales to guide cell behavior. Herein, natural interface of waste fish scales can be directly, simply, green, and low-costly used to regulate cell behavior. A series of characterization showed that the natural surface of fish scales is rich in micro/nano structures (groove, short ridge, fibers), contain collagen and hydroxyapatite, has suitable wettability and no cytotoxicity. These natural structures on waste fish scales have adjustable cell guidance abilities, which are consistent with the anisotropy levels of these structures (ordered fibers > grooves > damaged fibers > short ridges). Moreover, cell contact interaction with micro/nano topographies on the fish scale surface is dynamically adjustable with growth time, and also increased the chemoresistance of cancer cells to drug (irinotecan and cisplatin). Due to its advantages of wide material sources, waste recycling, patterning-free, and pretreatment-free, directly using biowastes (fish scales) not only provides a new opportunity for the development of in vitro anisotropic culture platform, but also helps to deepen the understanding and application of natural materials.

Sodium caseinate particles with co-encapsulated resveratrol and epigallocatechin-3-gallate for inhibiting the oxidation of fish oil emulsions

Xin Yin,Huanhuan Dong,Hao Cheng,Chuye Ji,Li Liang

In oil-in-water emulsions, a single phenolic compound has been used as interfacial antioxidant. The co-encapsulated polyphenols may produce more supper stability or bioactivity than a single encapsulated one. This study used epigallocatechin-3-gallate (EGCG) and resveratrol as model polyphenols with different solubility. Their co-encapsulation in SC particles was analyzed on the basis of particle characterization, loading efficiency, antioxidant activity , polyphenolic microenvironment and retention. When the polyphenols’ concentration was 0.4 mM, 98% EGCG and 68% resveratrol were co-encapsulated in sodium caseinate (SC) particles. The SC particles containing dual polyphenols were then used to stabilize fish oil emulsions, which were analyzed in term of particle characterization, oil oxidation , polyphenolic partition and retention. When the content of fish oil was 10%, there were 77% resveratrol in the emulsified oil droplets and 87% EGCG at the oil-water interface. The co-encapsulated EGCG improved the stability of resveratrol in SC particles and protected fish oil emulsions against oxidation prior to resveratrol. These results are instructive for the combination of multiple bioactive compounds for the applications in foods and pharmaceuticals.

Superhydrophobic Poly(l-lactic acid) Membranes with Fish-Scale Hierarchical Microstructures and Their Potential Application in Oil–Water Separation

Lingqi Zhong,Haizheng Tao,Xiao Gong

In this work, superhydrophobic poly(l-lactic acid) (PLLA) hierarchical membranes exhibiting excellent oil-removal performance, which is of great importance in curbing the oil-pollution environment, were fabricated by a simple solvent-evaporation-induced precipitation method. PLLA membranes with hierarchical micro/nanostructures (fish scales, fibrous sheets, and petal-like morphology) can be conveniently prepared by adjusting the preparation parameters including PLLA concentration, precipitation temperature, type of solvent and nonsolvent, and the addition of nano-SiO2. The results show that the water contact angle of the fish-scale-structured PLLA membrane was 138.6°, revealing that water repellency was significantly improved compared to that of the solvent-casting PLLA membrane (∼72.8°). Moreover, the PLLA/SiO2 nanocomposite membrane with a dense hierarchical micro/nanostructure had a water contact angle greater than 167.1°, which has great potential in oil–water separation.
