Galangal extract (GE)-based hypouricemic functional food is under-developed due to ambiguous quality control standard that is closely associated with action mechanisms and interaction of key xanthine oxidase (XO) inhibitors (kaempferide and galangin) in GE. In terms of kinetics analysis, fluorescence quenching and molecular docking , kaempferide and galangin showed similar docking posture to xanthine in molybdopterin center, and formed flavonol-XO complexes driven by hydrogen bonding, hydrophobic interaction and van der Waals force, competitively inhibiting XO . Kaempferide, had stronger binding affinity for XO and three more hydrogen bonds with XO than galangin, interacting with critical amino acid residues (Arg880 and Glu802) in catalysis reaction of XO and showing stronger XO inhibitory activity than galangin. The combination of kaempferide and galangin enhanced their binding affinities for XO, showing synergistic inhibition on XO at optimal molar ratio 1:4 that could be quality control standard for GE. This study provided new insights into structure-XO inhibitory activity relationship of methoxylated flavonoids and quality control standard for GE-based hypouricemic functional food.

The trigonal planar unit possesses significant hyperpolarizability and polarizability anisotropy, which makes it useful for optimizing nonlinear optical (NLO) materials, however, chalcogenide with this unit has seldom been reported. In this work, a novel approach is introduced by integrating the unprecedented trigonal planar MS 3 (M = Cd/In, Hg/In) motifs into the nearly optically isotropic tetrahedral units, resulting in two novel chalcogenides CsM 2 In 2 S 6 (M = Cd/In, 1 ; Hg/In, 2 ). Notably, structures 1 and 2 feature nearly planar triangular units at the center, encircled by three trimers, further interconnecting each other to create 3D frameworks. Importantly, phases 1 and 2 display phase-matching (PM) capabilities, primarily attributed to incorporating trigonal planar MS 3 units that additionally enhance polarizability anisotropy. Furthermore, compounds 1 and 2 demonstrate moderate second-harmonic generation (SHG) signals (0.70 and 0.84 × AgGaS2@1.7 µm). This study pioneers an efficient strategy for the design of infrared NLO crystals with PM capabilities.

Zhuye Shigao Decoction (ZSD), a classic traditional Chinese medicine (TCM) formula, has demonstrated therapeutic efficacy in clinical settings for acute pneumonia; however, its mechanism of action remains elusive. This study aims to investigate the therapeutic effects and potential mechanisms of ZSD in lipopolysaccharide (LPS)‐induced acute pneumonia in mice. A liquid chromatography‐mass spectrometry (LC‐MS)‐based metabolomics approach, integrated with a gene expression omnibus (GEO) data mining technique, was employed in this investigation. The results indicated that ZSD administration significantly alleviated LPS‐induced pathological changes in lung tissue. In the model group, the concentrations of interleukin‐1 β (IL‐1 β ), interleukin‐6 (IL‐6), and tumor necrosis factor‐ α (TNF‐ α ) were increased, whereas the levels of immunoglobulin A (IgA) and immunoglobulin M (IgM) were significantly decreased. These indicators were reversed considerably in the ZSD‐high dose (ZSD‐H) group. A total of 118 metabolites exhibiting significant alterations in lung tissue were identified through metabolomics analysis. Following ZSD treatment, 84 of these metabolites were negatively regulated, and the associated metabolic processes implicated multiple pathways, including sphingolipid and arachidonic acid (AA) metabolism. Differentially expressed genes (DEGs) were identified through a comprehensive analysis of GEO datasets. Integrative pathway analysis identified signaling pathways associated with ZSD treatment effects in acute pneumonia. Notably, the Janus kinase/signal transducer and activator of transcription (JAK‐STAT) and Toll‐like receptor (TLR) signaling pathways emerged as critical contributors to these effects. This study indicates that ZSD can exert therapeutic effects on LPS‐induced acute pneumonia through a regulatory mechanism involving multiple components, targets, and pathways.

All-inorganic perovskite quantum dots (PQD) have become the most promising next-generation luminescent material due to their excellent luminescent properties. However, as an ionic crystal, their stability is the biggest problem for their large-scale application. Especially, it is a huge challenge to use them in water or at high temperatures. This study proposes a strategy for the simultaneous synthesis of CsPbBr 3 quantum dots and silica aerogels (SiO 2 AG). During the formation of the aerogel network, the quantum dot precursor solution is confined within nanoscale silica aerogel “microcapsules”; upon heating, the quantum dots undergo nucleation and growth within these “capsules”, simultaneously completing the synthesis and protection of the quantum dots. The experimental results show that the photoluminescence properties of CsPbBr 3 quantum dots and SiO 2 aerogel (CsPbBr 3 @SiO 2 AG) Composites material prepared by this method remain unchanged after heat treatment at 400°C for 8 h, and the photoluminescence quantum yield (PLQY) can reach 99.1%. Even after direct immersion in water and natural evaporation, the luminescent properties remain above 39%, and the precipitation of Pb 2+ is effectively inhibited. This material provides strong support for the large-scale application of perovskite quantum dots, especially in various emerging fields under extreme conditions.

Tofacitinib is a widely used medication for the treatment of arthritis. It has been reported that some patients experience abnormal cholesterol levels following treatment, leading to recommendations for the coadministration of lipid-lowering drugs such as statins. In this study, we investigated the potential drug–drug interactions between tofacitinib and the statins simvastatin and lovastatin. In the in vitro experiments, rat liver microsomes were employed to evaluate the inhibitory effects of lipid-lowering agents on the metabolism of tofacitinib, with the primary metabolite M8 analyzed using ultra-performance liquid chromatography–tandem mass spectrometry. The results showed that simvastatin and lovastatin significantly inhibited the metabolism of tofacitinib, with IC 50 values of 5.837 and 10.68 μM, respectively. For the in vivo pharmacokinetic studies, Sprague–Dawley rats were pretreated with simvastatin or lovastatin via oral gavage for 7 days before the oral gavage of tofacitinib. This pretreatment led to an increased area under the concentration–time curve of tofacitinib, suggesting that a potential reduction in the first metabolism and/or systemic clearance takes place. These findings demonstrate significant interactions between tofacitinib and certain lipid-lowering agents in the rat model, particularly simvastatin and lovastatin, both in vitro and in vivo.

Oxidative stress, arising from a dysregulation between the generation of reactive oxygen species (ROS) and antioxidant defense mechanisms, is closely linked to the pathogenesis of numerous chronic diseases. To identify natural peptide-based antioxidants, rabbit meat proteins were enzymatically hydrolyzed using seven proteases. Among them, Hydrolysates produced by papain demonstrated the greatest radical scavenging activity, with DPPH and ABTS inhibition rates of 51.50 % and 61.50 %, respectively. Ultrafiltration and Sephadex G-15 fractionation revealed that the <3 kDa fraction (PH3) and its subfraction F5 possessed the strongest antioxidant activity. LC-MS/MS analysis of F5 identified 163 peptides, of which 56 were predicted to exhibit antioxidant potential using PeptideRanker and AnOxPePred algorithms. Notably, seven novel peptides showed strong binding affinities to Keap1 (binding energy < −8.0 kcal/mol) in molecular docking, suggesting possible modulation of the Keap1–Nrf2 antioxidant pathway. Quantum chemical analysis (HOMO–LUMO orbitals and electrostatic potential mapping) combined with radical scavenging assays demonstrated that electron-donating aromatic residues (Trp, Tyr) played key roles in antioxidant activity via hydrogen atom and electron transfer mechanisms, establishing a molecular-level structure–activity relationship. In vivo validation using an AAPH-induced zebrafish model confirmed the peptides' safety and efficacy, including reduced ROS levels and restored endogenous antioxidant enzyme activities (SOD and CAT). Collectively, these findings provide mechanistic insight into the antioxidant action of rabbit-derived peptides and support their potential application as natural antioxidants for functional food development and oxidative stress-related disease prevention.

Bisphenol analogues (BPs), the endocrine-disrupting compounds, have been detected in water, sediment, and even agricultural products. Irrigation is the main way for material migration, and it has been suggested as one of the sources of BPs. But the attention to irrigation water and sediment, and further to the ecological risks, is rare. In a hilly irrigation area in southern China, the partition and accumulation, contribution sources, and potential ecological risk of four bisphenols (BPA, bisphenol S, F, and AF) in the irrigation canal were investigated in the spring of 2023 and 2024. All four BPs were detected in the irrigation water and sediment. BPF was the dominant of 19.6–60.9 ng/L in water, while BPA was still the prominent compound (46.2–54.4% of ∑BPs) in sediment. BPs in the irrigation canal were related to the direct discharge of aquaculture, domestic, and planting drainage. Although low risks for each compound, the comprehensive risk (∑RQ) of sediments went into medium risk. BPF and BPA were the main risk contributors. BPs in sediments increased steadily during the investigation. The special accumulative point was where slow flow maintained, especially caused by the hilly change. This comprehensive study of BPs in hilly irrigation canals should be valuable for reference in similar areas. And it will be beneficial for environmental protection and production safety.

Testosterone is a vital male hormone responsible for male sexual characteristics. The taste receptor family 1 subunit 3 (T1R3) regulates testosterone synthesis and autophagy in non-taste cells, and the links with the taste receptor family 1 subunit 1 (T1R1) for umami perception. However, little is known about these mechanisms. Thus, we aimed to determine the relationship between the umami taste receptor (T1R1/T1R3) and testosterone synthesis or autophagy in testicular Leydig cells of the Xiang pig. There was a certain proportion of spermatogenic tubular dysplasia in the Xiang pig at puberty, in which autophagy was enhanced, and the testosterone level was increased with a weak expression of T1R3. Silenced T1R3 decreased testosterone level and intracellular cyclic adenosine monophosphate (cAMP) content and inhibited the messenger RNA (mRNA) expression levels of testosterone synthesis enzyme genes [steroidogenic acute regulatory protein ( StAR ), hydroxy-delta-5-steroid dehydrogenase, 3 beta- and steroid delta-isomerase 1 ( 3β-HSD1 ), cytochrome P450 family 17 subfamily A member 1 ( CYP17A1 ) and hydroxysteroid 17-beta dehydrogenase 3 ( 17β-HSD3 )]. In addition, T1R3 increased the number of acidic autophagy bubbles and upregulated the expression levels of autophagy markers [Microtubule-associated protein 1 A/1B-light chain 3 (LC3) and Beclin-1] in testicular Leydig cells of the Xiang pig. Using an umami tasting agonist (10 mM L-glutamate for 6 h), the activation of T1R1/T1R3 enhanced the testosterone synthesis ability by increasing the intracellular cAMP level and upregulated the expression levels of StAR , 3β-HSD1 , CYP17A1 and 17β-HSD3 in Leydig cells. Furthermore, the number of acidic autophagy bubbles decreased in the T1R1/T1R3-activated group with the downregulation of the expression levels of the autophagy markers, including LC3 and Beclin-1 . These data suggest that the function of T1R1/T1R3 expressed in testicular Leydig cells of the Xiang pig is related to testosterone synthesis and autophagy.

A commercial formulation called prothioconazole·kresoxim-methyl 50% water dispersible granule (WG), used to control the powdery mildew, is under registration for use on cucumbers. Therefore it is urgent to validate the reliability of the recommended good agricultural practices (GAP) conditions (187.5 g a.i. ha−1, three sprays with a 7-d interval, and a pre-harvest interval of 3 d) by conducting field trials in 12 regions in China, as required by national regulations, with a subsequent risk assessment. The residues of prothioconazole-desthio and kresoxim-methyl in field samples were determined using QuEChERS coupled with high-performance liquid chromatography – tandem mass spectrometry (HPLC-MS/MS). On the suggested pre-harvest interval (PHI, 3 d), the residual concentrations of prothioconazole-desthio (no maximum residue limit set in China) and kresoxim-methyl (maximum residue limit = 0.5 mg/kg) in cucumbers were 0.01–0.020 and 0.01–0.050 mg/kg, respectively. The acute risk quotients of prothioconazole-desthio in cucumbers were no higher than 0.079% for Chinese consumers. The chronic dietary risk quotient of kresoxim-methyl and of prothioconazole-desthio for different groups of consumers in China ranged from 2.3% to 5.3% and from 1.6% to 4.6%, respectively. Thus, prothioconazole·kresoxim-methyl 50% WG sprayed on cucumbers under the recommended GAP information can pose a negligible risk for Chinese consumers.

The treatment of dry eye mainly includes instillation of cyclosporine A (CsA) nanoemulsion or the use of punctal plugs. Therefore, in this study, a novel injectable in situ organogel plug was developed using CsA as a model drug, stearic acid , injectable soybean oil, and N -methyl-2-pyrrolidinone (NMP) (1.25:10:0.6, w/v/v) as gel materials, to provide a dual mechanism for dry eye treatment. The formulated CsA injectable in situ organogel (CsA-OG) was evaluated in terms of stability, in vitro release, rheology, ocular irritation, punctal occlusion tests, and ocular distribution assessment. In vivo ocular distribution investigations showed that CsA-OG achieved considerably higher C max (1.94, 1.92 and 1.97-fold respectively) and AUC 0-72h in the cornea, conjunctiva, and sclera (2.49, 2.27 and 2.15-fold respectively) than ciclosporin eye drops ( p < 0.05). In vitro model evaluation demonstrated significant decrease in flow flux to 52.78 % at 2 min after CsA-OG injection. According to evaluation of the in vivo model, the organogel plug can completely block the lacrimal passages and greatly decrease the lacrimal drainage rate ( p < 0.05). The above results suggest that these intracanalicular CsA-OG plugs can offer more extensive clinical applications than existing lacrimal drainage plugs and may act as a drug delivery system .

Headgroups in cationic lipids play very important roles in determining transfection efficiency and toxicity in gene delivery. To better understand the influence of headgroups on gene delivery, a tri-peptide-based lipid was synthesized, wherein the usual quaternary ammonium was replaced by a tri-peptide. Though both the tri-peptide-based lipid (DAO3) and the quaternary ammonium-based lipid (DDCTMA) successfully mediated gene transfection, DAO3 was superior to DDCTMA in both in vitro and in vivo studies. Following their preparation into liposomes, the particle size, zeta potential, and DNA-binding capacity of the liposomes and lipoplexes were characterized to evaluate the efficiency of DAO3 compared to DDCTMA with regard to gene interactions. The expression of luciferase from pDNA mediated by DAO3 was 2-fold greater than than that with DDCTMA in Hep-2 cells, and DAO3/siRNA lipoplexes could silence about 60% luciferase in A549 cancer cells expressing firefly luciferase. DAO3/Luc-siRNA treatment exhibited 3-fold the efficiency of DDCTMA/Luc-siRNA in terms of in vivo luciferase RNAi with the bare density ratio of 0.54 at 48 h. Furthermore, DAO3 could mediate IGF-1R siRNA to inhibit tumor growth through silencing the expression of the IGF-1R protein, whereas DDCTMA showed nearly no effects. Most importantly, DAO3 had no obvious toxicity in vitro and in vivo, due to the biocompatibility of the peptide headgroups. In conclusion, these results demonstrated that the replacement of the quaternary ammonium headgroup by tri-ornithine may increase transfection efficiency and decrease toxicity.

Zingiber officinale (ZO) is a traditional food condiment. The essential oils of Z. officinale (ZOEOs) are known to have multiple bioactivities. In this study, gas chromatography mass spectrometer (GC-MS) analytical method was used to identify active ingredient present in ZOEOs. A total of 41 compounds were identified in ZOEOs. Major components in ZOEOs were zingiberene (19.71%), (+)-β-cedrene (12.85%), farnesene (12.17%), α-curcumene (10.18%) and β-elemene (3.54%). Experimental results of 12-O-tetradecanoylphorbol-13 acetate (TPA) induced ear swelling validation mice model showed that ZOEOs treatment has better anti-inflammatory effect compared with ibuprofen (positive control) at high concentrations. Histological and immunohistochemical analysis showed that ZOEOs significantly decreased COX-2, IL-6 and NF-κB expression in a dose dependent manner. The mRNA levels of COX-2 and NF-κB were also down regulated by the application of ZOEOs. This indicated that ZOEOs exhibited positive effects in ear skin protection. Antibacterial experimental results showed that EOZOs had anti-bacterial effects on Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. DPPH radical scavenging, A549 cell line and LNCaP cell line inhibition results indicated that ZOEOs exhibited potential antioxidant and anti-tumor properties. The findings of these study provide scientific basis on therapeutic use of ZO in food, cosmetic and pharmaceutical industries.
