Managing pneumonia caused by multidrug-resistant (MDR) bacteria presents significant clinical challenges. The near-infrared II (NIR-II) laser irradiance exhibits strong photothermal conversion capabilities, making it a promising candidate for photothermal and chemodynamic therapies as non-antibiotic strategies. However, traditional methods are often hindered by issues such as the uncontrollable production of reactive oxygen species and the low efficiency associated with NIR-II photothermal therapy. This underscores the necessity for precisely regulated and highly effective synergistic therapies. To address these limitations, we have innovatively developed a defect/interface dual-engineered nanozyme (PtPb@SbO 3-x ) and comprehensively characterized its atomic-scale structure and catalytic mechanisms using density functional theory calculations and synchrotron radiation techniques. In vitro experiments demonstrated that PtPb@SbO 3-x could efficiently eliminate drug-resistant bacteria and disrupt biofilm structures under low-concentration hydrogen peroxide and NIR-II irradiation while exhibiting excellent biocompatibility. In pneumonia models, the nanozyme enabled rapid infection clearance and significantly reduced inflammatory responses via synergistic photothermal and chemodynamic therapy effects. Furthermore, integrated multi-omics analyses-including metabolomics, transcriptomics, and proteomics-systematically uncovered the molecular mechanisms driving its therapeutic efficacy. This study successfully establishes a non-antibiotic nanozyme-based therapeutic strategy that is efficient, low in toxicity, and non-invasive for treating MDR bacterial infections. It also provides a solid theoretical basis and technical framework for the rational design of defect/interface dual-engineered nanoplatforms.

Infected wound healing remains a significant clinical challenge due to bacterial resistance and excessive inflammation. Herein, a multifunctional hydrogel dressing (Nb2C-Arg/Gel) is developed that integrates two-dimensional Nb2C nanosheets (photothermal agent) and arginine [nitric oxide (NO) donor] into a calcium ion cross-linked sodium alginate matrix. The hydrogel has a relatively high swelling capacity and good ability to scavenge reactive oxygen species (ROS), which are beneficial for absorbing wound exudate and reducing the inflammatory response of wounds. Under irradiation with a low-power NIR-II laser (1064 nm, 1 W cm–2), Nb2C-Arg/Gel achieves rapid photothermal heating (48.6 °C in 1 min), while thermally enhances arginine decomposition-triggered on-demand NO release. This synergistic mechanism provides superior antimicrobial efficacy against Staphylococcus aureus (97.65%) and methicillin-resistant S. aureus (90.05%). In vivo studies on MRSA-infected murine wounds demonstrate that the residual area on day 10 was 2.9%, which accelerated healing compared to that of the control group (17.8%), accompanied by reduced inflammation and enhanced angiogenesis. The excellent biocompatibility of hydrogel is validated by low hemolysis (<5%) and promoted fibroblast proliferation (125% cell viability). Combining with photothermal, NO-mediated antibacterial action, and ROS scavenging to alleviate oxidative stress, this antibiotic-free hydrogel dressing has great potential for clinical application for the treatment of refractory-infected wounds.

Developing high-quality NIR-II fluorophores (emission in 1000–1700 nm) for in vivo imaging is of great significance. Benzothiadiazole-core fluorophores are an important class of NIR-II dyes, yet ongoing limitations such as aggregation-caused quenching in aqueous milieu and non-activatable response are still major obstacles for their biological applications. Here, we devise an activatable nanoprobe to address these limitations. A molecular probe named BTPE-NO 2 is synthesized by linking a benzothiadiazole core with two tetraphenylene groups serving as hydrophobic molecular rotors, followed by incorporating two nitrophenyloxoacetamide units at both ends of the core as recognition moieties and fluorescence quenchers. An FDA-approved amphiphilic polymer Pluronic F127 is then employed to encapsulate the molecular BTPE-NO 2 to render the nanoprobe BTPE-NO 2 @F127. The pathological levels of H 2 O 2 in the disease sites cleave the nitrophenyloxoacetamide groups and activate the probe, thereby generating strong fluorescent emission (950~1200 nm) and ultrasound signal for multi-mode imaging of inflammatory diseases. The nanoprobe can therefore function as a robust tool for detecting and imaging the disease sites with NIR-II fluorescent and multispectral optoacoustic tomography (MSOT) imaging. Moreover, the three-dimensional MSOT images can be obtained for visualizing and locating the disease foci.

Transition metal selenide nanodots (NDs) represent distinctive antitumor agents for cancer treatment, but their non-biodegradability may bring serious adverse effects and potential long-term toxicity to internal tissues/organs, which substantially hinder their further clinical translations. In this work, the construction of a multifunctional theranostic nanosystem based on degradable and excretable ultrasmall Rh 3 Se 8 NDs by a general bovine serum albumin-templated strategy is reported. The constructed Rh 3 Se 8 NDs exhibit distinctively high photothermal-conversion efficiency (57.5%) in the second near-infrared biowindow, making them highly applicable for photoacoustic imaging and photonic hyperthermia at the desired wavelength. Rh 3 Se 8 NDs exhibit a high tumor growth inhibition rate (98.1%) on 4T1 breast tumor-bearing mice due to the desirable photonic hyperthermia performances. Especially, the fabricated Rh 3 Se 8 NDs feature the large X-ray attenuation coefficients of the Rh component for contrast-enhanced X-ray computed tomography imaging. Importantly, the prominent biodegradability of Rh 3 Se 8 NDs enables their quick excretion out of the body for potentially avoiding inflammation and mitigating long-term toxicity. Therefore, this work highlights the construction of proof-of-concept biodegradable and excretable ultrasmall inorganic theranostic nanosystems for multiple bioimaging-guided cancer nanotherapeutics, guaranteeing the further clinical translations of inorganic nanoparticles in biomedicine.

Real-time tracking drug release behavior is fundamentally important for avoiding adverse effects or unsuccessful treatment in personalize medical treatment. However, the development of a non-invasive drug reporting platform still remains challenging. Herein the design of a novel synthetic magnetic resonance imaging (MRI) agent for drug release tracking (SMART) is reported, which integrates photothermal core and paramagnetic ion/drug loading shell with a thermal valve in a hybrid structure. Through near-infrared (NIR)-II photothermal effect originating from inner Au-Cu 9 S 5 nanohybrid core, burst release of drugs loaded in the mesoporous silica shell is achieved. The concomitant use of a phase change material not only prevents premature drug release, but also regulates heating effect, keeping local temperature below 45 °C, enabling synergistic chemotherapy and mild hyperthermia in vitro and in vivo. Furthermore, the drug release from SMART facilitates proton accessibility to the paramagnetic ions anchored inside mesopores channels, enhancing longitudinal T 1 relaxation rate and displaying positive signal correlation to the amount of released drug, thus allowing non-invasive real-time monitoring of drug release event. The current study highlights the potential of designed MRI nanophores such as SMART for real-time and in-situ monitoring of drug delivery for precision theranostic applications.

Silver chalcogenide (Ag2X; X = S, Se, Te) quantum dots (QDs) have facilitated the biomedical applications of QDs due to their low toxicity and tunable emission reaching the second near-infrared window (NIR-II window, 1000–1700 nm). The photoluminescence intensity of Ag2S QDs and Ag2Se QDs is considerably enhanced via Pb doping. However, the developed Pb-doped Ag2S QDs lacked in vivo verification, and the toxicity of the Pb-doped Ag2X QDs was only preliminarily assessed. Although QDs containing Pb have been widely introduced into the field of biomedicine, there are still concerns regarding toxicity associated with Pb. In the present study, we prepared polyethylene glycol (PEG)-modified Pb-doped Ag2S QDs with better biocompatibility and tested their in vivo bioimaging and toxicity in comparison with Ag2S QDs modified with the same PEG under identical concentrations. Results indicated that the PEG-modified Pb-doped Ag2S QDs significantly enhanced the brightness of in vivo bioimaging and spatial resolution for vessels inside the tumor without increasing toxicity, assessed based on cell viability post incubation for 96 h, intracellular ROS level, DNA damage, early apoptosis and mitochondrial injury. Our study demonstrated that the PEG-modified Pb-doped Ag2S QDs could serve as an advanced fluorescent probe for various biomedical applications.

Near-infrared phosphor-converted light-emitting diodes (NIR pc-LEDs) offer numerous advantages, including compact size, tunable emission spectra, energy efficiency, and high integration potential. These features make them highly promising for various applications, such as night vision monitoring, food safety inspection, biomedical imaging, and theragnostics. All-inorganic halide double-perovskite materials, known for their large absorption cross section, excellent defect tolerance, and long carrier diffusion radius, serve as unique matrices for constructing near-infrared fluorescent materials. In this study, we successfully prepared the all-inorganic metal halide double-perovskite Cs2NaYCl6:Cr3+ using a grinding–sintering method. A small fraction of the [YCl6] octahedra within the host material’s lattice was substituted with Cr3+ ions, resulting in the creation of the Cs2NaYCl6:Cr3+ phosphor. When excited with λ = 310 nm UV light, the phosphor exhibited a broad emission range spanning from 800 to 1400 nm, covering the NIR-I and NIR-II regions. It had a broad bandwidth emission of 185 nm and achieved a fluorescence quantum yield of 20.2%. The unique broadband emission of the phosphor originates from the weak crystal field environment provided by the Cs2NaYCl6 host matrix, which enhances the luminescence properties of the Cr3+ ions. To create NIR pc-LEDs, the phosphor was encapsulated onto a commercially available UV LED chip operating at 310 nm. The potential application of these NIR pc-LEDs in night vision imaging was successfully validated.

Photothermal immunotherapy has shown great potential for efficient cancer treatment. However, the immunosuppressive tumor microenvironment forms a heavy barrier for photothermal-induced anti-tumor immunity by inhibiting dendritic cells (DCs) maturation and cytotoxic T cell response. Moreover, the lack of reliable spatiotemporal imaging modalities makes photothermal immunotherapy difficult to guide tumor ablation and monitor the therapeutic outcomes in real time. Herein, we designed a theranostic thermosensitive liposome (PLDD) as a versatile nanoplatform to boost adaptive anti-tumor immunity of photothermal immunotherapy and achieve multiple bioimaging modalities in a real-time manner. The PLDD contains two major functional components: a multifunctional photothermal agent (DTTB) and an immune potentiator STING pathway agonist (DMXAA). Upon irradiation, the heat generated by DTTB induced the immunogenic cell death (ICD) of the tumor and dissociated the structure of thermosensitive liposome to release DMXAA, which ultimately activated the STING pathway and promoted the ICD-induced immune response by increasing DC cells maturation and T cell recruitment. Meanwhile, the DTTB in PLDD displayed excellent second near-infrared (NIR-II) fluorescence and photoacoustic (PA) dual-modal imaging, which provided omnibearing information on the tumor and guided the subsequent therapeutic operation. Therefore, this versatile PLDD with light-triggered promotion of anti-tumor immunity and multiple spatiotemporal imaging profiles holds great potential for the future development of cancer immunotherapy.

Fluorescence and magnetic resonance imaging (FL/MRI) has received much attention due to their complementary characteristics. However, the simultaneous enhancement of fluorescence and MR signals plus the efficacy of the treatment is still a major challenge. To tackle this difficulty, we put forward a strategy of aggregation and de-aggregation based on luminogens with aggregation-induced emission (AIE) features. NIR II photosensitizer 4,4′-(6,7-diphenyl-[1,2,5]thiadiazolo[3,4-g]quinoxaline-4,9-diyl)bis(N,N-diphenylaniline) (TQ-TPA) with type I reactive oxygen species (ROS) generation ability and amphiphilic 2TPE-Gd were synthesized, and both of them showed the AIE feature. During the construction of nanomicelles, hydrophobic TQ-TPA spontaneously aggregated into the core of the nanomicelles formed by 1,2-distearoyl-sn-glycerol-3-phosphoethanolamine-N-methoxy(polyethylene glycol)-2000 (DSPE-PEG). Meanwhile, aggregates of 2TPE (tetraphenylethylene)-Gd in aqueous solution could be de-aggregated and inserted into the interface, and formed TGdTT nanomicelles (NMs). Based on this strategy and the AIE feature, the TGdTT NMs exhibited strong NIR II fluorescence and type I ROS generation ability, and enhanced T1 relaxivity (r1). Moreover, in vitro, in vivo, and pharmacokinetics results demonstrated that these nanomicelles have good biosafety and a long blood circulation time. Finally, they allowed successful realization of complementary MR/NIR II fluorescence dual-modal imaging-guided photodynamic therapy (PDT) to inhibit tumor growth. This work demonstrated that the aggregation and de-aggregation strategy of AIEgens in core–shell nanomicelles is very attractive for constructing multifunctional theranostic probes.

Rheumatoid arthritis (RA) is currently one of the most common systemic autoimmune disorders, with no clinically effective theranostics. Therapeutic manipulation of the gasotransmitter hydrogen sulfide (H 2 S) has exhibited promising potential as an innovative strategy in RA, whereas limited with non-targeting, low solubility, and systemic toxicity. In this work, based on the physicochemical and structural advantages of stimulus-responsive nanotechnology, we develop multifunctional glutathione (GSH)-responsive H 2 S nanogenerators (BDMA NGs), achieving activatable precise second near-infrared fluorescence (NIR-II FL)/photoacoustic (PA) bimodal imaging-guided gas/chemotherapy synergistic therapy. Contributed to the suitable size and excellent biocompatibility, the BDMA NGs accumulate passively in the RA region and then feature a responsive deformation reaction with GSH to release functional small molecules: H 2 S and methotrexate (MTX). The H 2 S undergoes a mineralization process with Ag + ions in situ, resulting in the rapid formation of Ag 2 S to “turn on” the NIR-II FL/PA signals. Remarkably, the effective generation of H 2 S downregulates hypoxia-inducible factor-1 α (HIF-1 α ) and promotes nuclear factor erythroid 2-related factor 2 (Nrf2) nuclear translocation, thereby restoring homeostasis in the RA microenvironment and facilitating the reprogramming of pro-inflammatory macrophages (M1) into anti-inflammatory phenotype (M2). Moreover, the controlled release of MTX, clinically approved drug for RA, synergistically promotes repolarization to further suppress local inflammation. Overall, the H 2 S-based nanogenerators BDMA NGs present a sophisticated and versatile strategy for implementing stimuli-responsive imaging and M1-to-M2 repolarization, demonstrating significant potential in the theranostics of RA and clinical transformation.

The development of phosphor-converted light-emitting diodes (pc-LEDs) in the second near-infrared window (NIR-II, 1000–1700 nm) represents an important, newly emerging, and dynamic field in NIR spectroscopy. Unfortunately, lacking efficient NIR-II phosphors that can be excited by commercial blue LED chips impedes NIR spectroscopy applications. Herein, a donor–acceptor strategy is developed by introducing Cr 3+ sensitizers to greatly enhance the blue-light excitation efficiency of NIR-II-emitting MgO:Cr 3+ ,Ni 2+ phosphor. Consequently, a high-efficiency blue-light-excitable MgO:Cr 3+ ,Ni 2+ phosphor that gives a NIR-II emission at 1335 nm with a full width at half-maximum (FWHM) of 235 nm, is demonstrated. The product has a high internal quantum efficiency of 92.7 % and excellent thermal stability, maintaining 83.0 % of the room temperature emission intensity at 150 °C. Excitingly, the fabricated NIR-II pc-LED device shows a high NIR-II optical power (27.4 mW@350 mA). The performances of the achieved NIR-II pc-LED are almost the best results until now. Additionally, multifunctional applications including nondestructive detection and anti-counterfeiting of the NIR-II light source are demonstrated. These results are pretty crucial for the further development of NIR-II spectroscopy and imaging technology.

Elevating photothermal conversion efficiency (PCE) is the key to enhance curative efficacy during photothermal therapy. Nevertheless, current functionalized or synthetic methods for preparing photothermal agents with high PCE are too complicated, greatly hindering the development of photothermal therapy. In this study, we developed a tumor-targeting flower-like nanocomposite termed mesoporous platinum nanoparticles@Cu 2- x Se-AS1411 (MPNPs@Cu 2- x Se-AS1411, denoted as MPCSA) with remarkable PCE for high-resolution photoacoustic (PA) imaging guided photothermal/chemodynamic therapy for tumor ablation. We discovered that MPNPs served as carriers for delivering PA contrast agents. Cu 2- x Se nanosheets emerged as promising near-infrared-II (NIR-II) PA imaging and photothermal/chemodynamic therapy agents owing to their strong localized surface plasmon resonance property in the NIR-II region. The amine-modified AS1411 aptamer, which is connected to the surface of the flower-like MPNPs@Cu 2- x Se can target the nucleolar protein on the surface of 4T1 cells, achieving the accumulation of MPCSA at the tumor site in mice. Notably, these MPCSA nano-flowers exhibited an excellent PCE up to 82.33 % under 1064 nm laser irradiation, resulting in highly efficient cancer cell ablation and greatly hindering in vivo tumor growth. This study has paved the way for imaging-guided accurate cancer diagnosis and targeted cancer therapy.
