Nanoparticles based on chitosan hydrochloride (CSH)-hyaluronic acid (HA)-PEG were prepared for delivering curcumin (CUR) (CUR-PNPs) to brain tumor. CUR-PNPs of 245.9 nm and spherical morphology were obtained at optimized CSH/HA/PEG20000/CUR ratios with negative charge of about −27.2 mV and EE of approximately 93.3%. Cytotoxicity studies showed that CUR-PNPs improved drug’s anticancer activity in rat glioma cells (C6). The cellular uptake mechanism study showed active targeting of CUR-PNPs into C6 cells by HA mediated endocytosis . Clathrin-coated pit mediated endocytosis, clathrin-mediated endocytosis and macropincytosis were also identified as the entry pathways of PNPs into C6 cells. Pharmacokinetics of preparations in rats after i.v. administration further proved the superiority of CUR-PNPs (3.98 times greater than the area under the curve of CUR solution). In conclusion, CUR-PNPs might be a promising carrier for the therapy of brain tumors.

Although PEGylation allows a drug delivery vehicle to have prolonged blood circulation time, it faces the problem of reduced cellular uptake. Removal of the polyethylene glycol (PEG)-shell at the appropriate time through tumor-microenvironment triggers could be a feasible solution to this problem. Here, paclitaxel (PTX)-loaded mixed micelles (PTX-mM) self-assembled from stearate-modified hyaluronic acid (SHA), mPEG-b-poly(β-amino ester) (mPEG-b-PAE), and ethylene acetyl-b-poly(β-amino ester) (EA-b-PAE) were developed. In the preparation of PTX-mM, SHA micelles were coated with EA-b-PAE followed by coloading of PTX and mPEG-b-PAE. PTX-mM were capable of extracellular pH-triggered PEG-detachment and poly(β-amino ester) (PAE)-mediated endosomal escape. When the pH was changed from pH 7.4 to pH 6.8, the particle size of PTX-mM significantly decreased from 97.5 ± 4.4 to 71.5 ± 2.3 nm. It also resulted in rapid and complete release of mPEG-b-PAE from PTX-mM as monitored using quartz crystal microbalance (QCM) technology. PTX-mM capable of PEG detachment provided significant enhancement of PTX accumulation in SKOV-3 cells compared to PEG nondetachable PTX-mM. Interestingly, intracellular transport studies using confocal laser scanning microscopy (CLSM) showed that EA-b-PAE could promote the escape of micelles from endolysosomes. The half-maximal inhibitory concentration (IC50) of PTX-mM against SKOV-3 cells was 5.7 μg/mL, and PTX-mM containing 20 μg/mL of PTX induced apoptosis in 53.0% of the cell population. PTX-mM exhibited a highly prolonged elimination half-life (t1/2, 2.83 ± 0.37 h) and improved area under the curve (AUC, 7724.82 ± 1190.75 ng/mL/h) than the PTX-loaded SHA micelles (PTX-M). Furthermore, PTX-mM showed the highest tumor inhibition rate (64.9%) and the longest survival time (53 days) against the SKOV-3 ovarian cancer xenograft models among all formulations. Taken together, the results suggested that PTX-mM have potential as an efficient anticancer formulation in treatment of ovarian cancer.

Nano-structured lipid carriers (NLCs) have been widely used for oral drug delivery due to their remarkable biocompatibility and biodegradability. In addition to formulation and particle size, particle surface properties are important for overcoming gastrointestinal (GI) barriers to oral drug delivery by lipid-based nanoparticles. In this study, we describe polyethylene glycol (PEG)-based mucus-penetrating NLCs and we report the effect of surface hydrophilic properties on their oral delivery across the GI mucus and intestinal epithelium cells in vivo and in vitro. The solvent evaporation method was used to prepare PEG-modified NLCs (pNLCs) with three different levels of polyethylene glycol (100) monostearate (S100) as the hydrophilic coating material. The stability and cytotoxicity of pNLCs, as well as their uptake by Caco-2 cells and transport across Caco-2 monolayers and mucus-secreting Caco-2/HT29 coculture monolayers, were investigated to confirm the optimal PEG level for oral delivery. In situ single-pass perfusion experiments, in vivo imaging and in vivo oral pharmacokinetic studies were also performed in rodents to evaluate the oral absorption of pNLCs with different hydrophilic surface properties. Our results showed that the hydration layer thickness of the pNLCs increased with the increasing S100 content. NLC containing 20% S100 (pNLC-20%) showed the highest uptake by Caco-2 cells and the most effective permeation through Caco-2/HT29 coculture monolayers compared with unmodified NLC, pNLC-40% and pNLC-80%. In addition, pNLC-20% was the most stable during the process of oral administration and uptake into the blood circulation, as judged by fluorescence resonance energy transfer (FRET). Moreover, in situ single-pass perfusion experiments, ligated intestinal loops tests, in vivo imaging and pharmacokinetics studies also demonstrated that pNLC-20% most rapidly penetrated the intestinal epithelium and had the highest AUC and Cmax of all the NLCs. In conclusion, optimization of surface hydrophilic properties is essential for enhancing the absorption of drugs delivered by oral administration of lipid-based nanocarriers.

Biodegradable multi-blocked polyurethane (PU) based micelles with a hydrophilic PEG corona were extensively studied for anti-cancer drug delivery systems. The hydrophilic PEG segment usually incorporated as a soft segment or as an end capping reagent, which has difficulty forming a dense PEG coating with a brush like conformation due to the low mobility of the PEG domains at the soft segment and the low amount of the PEG at the end of PU chains. In the present study, biodegradable pH sensitive polyurethane micelles with a dense brush like coating of PEG were prepared by a new kind of PEG grafted polyurethanes (PEG-g-PU) which were synthesized using PEGylated diethanolamine (MPEG-DEAM) as a chain extender. The high mobility of pendant MPEG in PEG-g-PU results in the formation of a dense and brush like PEG corona on the PEG-g-PU micelles. Meanwhile the MPEG attached on the hard segment will transfer the diethanolamine (DEAM) to the surface of nanoparticles during the self-assembly process and the DEAM render the particles with positive charges which potentially enhances cellular uptake and endosomal escape. DLS, TEM and AFM showed that a dense PEG domain was formed on the surface of PEG-g-PU micelles while no obvious PEG microdomain was observed for the other two kinds of micelles. FTIR and DSC results demonstrated the enhanced microphase separation of PEG-g-PU micelles compared with PEG-g-PU bulk materials and the other two contrast PU micelles, i.e. PEG-b-PU and PEG-c-PU. Paclitaxel (PTX) was chosen as a model hydrophobic drug to evaluate the loading and pH-triggered release of the PU micelles. The enhanced cytotoxicity of PTX-loaded PEG-g-PU-3 micelles against H460 cancer cells reveals that they are more potent for intracellular delivery of PTX as compared to PEG-b-PU-3 and PEG-c-PU-3 micelles.

Biomimetic synthesis represents a cutting-edge topic in chemistry/materials science. Herein, we demonstrate poly(ethylene glycol) (PEG) short side-chain-assisted monomer complex coacervation and reaction-induced polyion complex compartmentalization that lead to oxygen-tolerant polymerization-induced electrostatic self-assembly (PIESA). This is achieved by the one-pot synthesis of a PEGylated anionic polyelectrolyte and heterogeneous iterative polymerization of a cationic monomer under ecofriendly ambient, in-air aqueous photo-RAFT conditions. Simultaneous reversible all-segment-participating ternary complex coacervation and Coulombic interdomain interactions lead to coacervate nanoreactors that are capable of immediate initiation and fast reversible addition–fragmentation chain transfer reactions. Approximately 2 nm monomer complex nanoclusters act as building blocks to drive liquid–liquid phase separation. Polymerization induces hierarchical self-assembly in a droplet nucleation–fusion–fission mechanism together with PEG-crowded polyion complex compartmentalization, using nanoclusters as building blocks, mechanistically similar to liquid–liquid phase separation through supramolecular polymerization. Consequently, protein-like, one-component multicompartment coacervate nanoreactors with oxygen-tolerant well-controlled fast reactions are achieved. This work provides important implications for the efficient precise synthesis of biomimetic coacervate nanodevices of increasing complexity.

As a hydrophilic polyether molecule, polyethylene glycol has environmentally friendly, degradable, and compatibility characteristics, gradually becoming a potential choice for green lubrication. Two novel multifunctional protic ionic liquids (ILs) were synthesized, and an ILs-PEG200 lubrication system was constructed with a strong hydrogen bonding network structure. The determination of rheological properties and boundary lubrication states inspired us to consider the interaction between polar additives and systems. Surprisingly, an extremely stable run-in period was obtained by the high efficiency adsorption contributed from high-polarity ILs at the sliding interface in the initial shear stage. Meanwhile, the ILs-PEG200 lubrication system also has superior load carrying and antishear properties, as well as remarkable lubrication stability even under very high loads, showing certain potential for engineering applications. In addition, the lubrication mechanism was obtained by exploring the interaction among ILs, PEG system, and metal surface, and the results revealed that tribo-oxidation reactions, physical/chemical adsorption, and tribochemical reactions occurred on the metal asperity during shear. Therefore, a tribofilm containing metal phosphates and sulfides is generated through the occurrence of tribochemical reaction and the synergistic effect of the base system, metal interface, and functional additives. The protic ILs lubrication additives in this work can significantly enhance the tribological performance of poly(ethylene glycol) systems under harsh conditions, providing a theoretical basis for its development in the engineering field.

Background Melanoma is an aggressive skin tumor with limited therapeutic options due to rapid proliferation, early metastasis, and poor prognosis. Baicalin (BA), a natural flavonoid, shows promise in inducing ferroptosis and apoptosis but faces challenges of poor solubility and bioavailability. To address these issues, we developed a multifunctional drug delivery system: manganese-doped ZIF-8 nanoparticles (ZIF(Mn)) loaded with BA and modified with folic acid (FA) and polyethylene glycol (PEG). FA targets melanoma cells by exploiting folate receptor overexpression, while PEG enhances biocompatibility and systemic circulation. Manganese enables magnetic resonance (MR) imaging for real-time, non-invasive therapy monitoring. Methods BA-loaded ZIF(Mn)/FA-PEG nanoparticles were synthesized via a one-pot method, enabling drug encapsulation, Mn²+ incorporation, and surface modification. The nanoparticles were comprehensively characterized (particle size, Zeta potential, FTIR, and XRD). Cytotoxicity and cellular uptake were evaluated in B16-F10 melanoma cells, and in vivo experiments in C57BL/6J mice investigated MR imaging capability, antitumor efficacy, and biosafety. Results BA@ZIF(Mn)/FA-PEG nanoparticles demonstrated excellent stability, a BA loading capacity of 33.50 ± 0.04%, and pH-responsive release, with accelerated drug release under acidic tumor conditions. Mn²+ provided strong T1-weighted MR imaging contrast. Cellular and animal studies showed enhanced uptake, reduced premature drug release, and improved compatibility. Mechanistically, the nanoparticles induced significant ferroptosis and apoptosis in melanoma cells, leading to potent antitumor effects. Conclusion The BA@ZIF(Mn)/FA-PEG nanoplatform effectively integrates targeted delivery, imaging guidance, and dual ferroptosis-apoptosis induction, offering a promising strategy for improving melanoma treatment outcomes.

Hybrid quasi-solid polymer electrolytes (QSPEs) were obtained by adding modified polyethylene glycol (mPEG) prepolymer into porous polyvinyl butyral (PVB). The prepared QSPEs have good luminous clarity (transmittance >70% in the visible region), high ionic conductivity in the level of 10 −5 S cm −1 . On the other hand, bond stress of the QSPE reaches 0.57 fold increase with introduction of 12 wt % PVB, attributing to functional groups in PVB. Moreover, an electrochromic device (ECD) was fabricated using PProDOT-Me 2 and Li-Ti doped NiO as working and counter electrode respectively, and hybrid QSPE as electrolyte. Compared with ECD based on mPEG electrolyte, the ECD shows 43.81% of optical modulation (ΔT) at 585 nm, and maintains 84% of the initial ΔT value after 20,000 cycles. These outstanding comprehensive performances demonstrate the QSPE has extensive prospects for electrochromic applications.

In this article, ZIF-8 nanoparticles were synthesized in water and directly added to a polyethyleneglycol (PEG) aqueous solution (water-based mixing process) to prepare ZIF-8/PEG mixed matrix membranes (MMMs) for pervaporation desulfurization. The structure and properties of ZIF-8 particles were characterized by Fourier Transform-Infrared Spectroscopy (FT-IR), scanning electron microscopy (SEM), X-ray diffraction (XRD), and BET analysis. The surface composition and structural morphology of all ZIF-8/PEG MMMs were systematically investigated by FT-IR and SEM. The effect of ZIF-8 content on the pervaporation performance of ZIF-8/PEG membranes was investigated. Versus pure PEG membranes, both the total flux and the sulfur enrichment factor of the ZIF-8/PEG MMMs were remarkably increased. This phenomenon is attributed to the good compatibility between PEG and ZIF-8 particles and the high thiophene selective sorption of ZIF-8 particles (confirmed by molecular simulation). Optimal pervaporation performance was achieved at a 4 wt% loading of ZIF-8 particles with the flux of 1.96 kg/(m 2 h) and the sulfur enrichment factor of 8.93. These increased by 397.46% and 18.75% versus pristine PEG membrane, respectively. In addition, the effect of operation temperature and feed sulfur content on membrane performance were also systematically investigated.

A series of controllable thermal and pH dual-responsive copolymeric hydrogels (PMA) were prepared by a one-pot reaction with poly(ethylene glycol) methyl ether acrylate (PEGA), 2-methoxyethyl acrylate (MEA) and acroleic acid (AA). The hydrogels exhibited good mechanical properties and a sensitive response to pH and temperature. Besides, the Lower Critical Solution Temperature (LCST) of the hydrogels can be adjusted from 37 °C to 58 °C by changing the content of AA. The hydrogels also showed excellent selective adsorption properties. The maximum adsorption quantity of organic cationic dye brilliant green and methylene blue were 0.49 mg mg−1 and 0.42 mg mg−1 respectively, much better than previous reports. Furthermore, using the thermal and pH responsibility, the PMA hydrogels can release the adsorbed molecules with control. Nearly 95% of carriers could be released at pH 4.01 and 65 °C over 8 h. The regeneration ability makes the materials easy to reuse many times. Due to these properties, these dual-responsive hydrogels have great potential applications in various fields for adsorption, drug delivery, release and tissue engineering.

Purpose To enhance therapeutic efficacy and prevent phlebitis caused by Asulacrine (ASL) precipitation post intravenous injection, ASL-loaded hybrid micelles with size below 40 nm were developed to improve drug retention and tumor penetration. Methods ASL-micelles were prepared using different weight ratios of 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-polyethyleneglycol-2000 (DSPE-PEG 2000 ) and D-α-tocopheryl polyethylene glycol 1000 succinate (TPGS) polymers. Stability of micelles was optimized in terms of critical micelle concentration (CMC) and drug release properties. The encapsulation efficiency (EE) and drug loading were determined using an established dialysis-mathematic fitting method. Multicellular spheroids (MCTS) penetration and cytotoxicity were investigated on MCF-7 cell line. Pharmacokinetics of ASL-micelles was evaluated in rats with ASL-solution as control. Results The ASL-micelles prepared with DSPE-PEG 2000 and TPGS (1:1, w /w) exhibited small size (~18.5 nm), higher EE (~94.12%), better sustained in vitro drug release with lower CMC which may be ascribed to the interaction between drug and carriers. Compared to free ASL, ASL-micelles showed better MCTS penetration capacity and more potent cytotoxicity. Pharmacokinetic studies demonstrated that the half-life and AUC values of ASL-micelles were approximately 1.37-fold and 3.49-fold greater than that of free ASL. Conclusions The optimized DSPE-PEG 2000 /TPGS micelles could serve as a promising vehicle to improve drug retention and penetration in tumor.

Deep eutectic solvents (DES) were prepared using urea (U) and acrylamide (AM) as hydrogen bond donors (HBD) and choline chloride (ChCl) as hydrogen bond acceptor (HBA), and polyethylene glycol (PEG) was selected as a filler and uniformly dispersed in DES to prepare PEG/P(U-AM-ChCl) composite hydrogels by light polymerization. The composite hydrogels were characterized by scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The effects of the content of PEG on the swelling properties, mechanical properties and fatigue resistance of the composite hydrogels were investigated. The results showed that the compressive strength and fatigue strength of the composite hydrogels were gradually enhanced with the increase of the PEG content in the composite hydrogels, in which the maximum compressive strength of the hydrogels with 1 wt% PEG added was increased by 1.86 times. The composite hydrogel had excellent swelling properties, and the equilibrium swelling degree of the hydrogel with 1 wt% PEG added reached 10.15. Meanwhile, the PEG/P(U-AM-ChCl) composite hydrogel had excellent self-healing properties, and the self-healing rate of the composite hydrogel with a PFG content of 1 wt% could reach 91.93% after 48 hours of healing. This study provides a convenient and efficient method to prepare composite hydrogels with superior swelling properties and self-healing properties.
