A smart drug delivery system responsive to pH/enzyme stimuli based on hydrophobic modified sodium alginate

Xue Gao,Zhe Yu,Bingxin Liu,Jixu Yang,Xiaoyan Yang,Yueqin Yu

A biocompatible dual-stimuli responsive drug delivery for hydrophobic drugs based on self-assembled polymeric micelles was presented. Hydrophobic modified alginate (HMA), used dodecyl glycidyl ether as a hydrophobic group, could form self-assembled micelles in aqueous solution above the critical micelle concentration. Doxorubicin (DOX) was chosen as a model drug and successfully loaded in HMA micelles. The percentage of drug loading efficiency of the HMA was determined as 65.4% by using fluorescence measurement. The pH-triggered and enzyme-triggered release behavior of DOX from DOX-HMA micelles indicated that the release of DOX was accelerated in acidic medium and that the release rate was dramatically enhanced in the presence of Alpha-L-fucosidase. It was found that almost 80% DOX was released from the DOX-HMA micelles in the presence of AFU at pH 5.5. Moreover, in vitro drug release studies exhibited that the DOX-HMA was effectively taken up by the cancer cells, and the blank micelles showed low toxicity toward the cancer cells, while the drug loaded micelles had high toxicity, even than the free drugs. These results demonstrate that HMA may serve as a general platform for therapeutic delivery.

Enhancing soil aggregation and acetamiprid adsorption by ecofriendly polysaccharides hydrogel based on Ca2+- amphiphilic sodium alginate

Siqi Zhang,Furui He,Xiuqin Fang,Xinyu Zhao,Yuanyuan Liu,Gaobo Yu,Yang Zhou,Yuhong Feng,Jiacheng Li

Soil aggregation plays an important role in agricultural production activities. However, the structure of soil aggregation is destroyed by the natural environment and unreasonable farming management, resulting in the loss of water, fertilizers and pesticides in soil. At present, hydrogels have been widely reported to promote the formation of soil aggregation. In this paper, amphiphilic calcium alginate (ASA/Ca 2+ ) was applied to promote the formation of soil aggregation and enhance pesticide retention. Initially, an ASA was obtained through the one-pot Ugi condensation (a four-component green chemical reaction). Then, ASA/Ca 2+ hydrogel is prepared by Ca 2+ cross-linking. The formation of soil aggregation was determined through the Turbiscan Lab Expert stability analyzer, Confocal Laser Scanning Microscope (CLSM), and Transmission Electron Microscope (TEM). And the effect of soil aggregation on acetamiprid environmental behavior was investigated by adsorption kinetics, adsorption isotherms, and leaching. The results shown that the three-dimensional network structure of ASA/Ca 2+ hydrogel can promote the formation of soil aggregation. Aggregate durability index (ADI) was 0.55 in the presence of ASA/Ca 2+ hydrogel, indicating that amphiphilic hydrogel can enhance the stability of soil aggregation. The adsorbing capacity of acetamiprid was 1.58 times higher than pure soil, and the release of acetamiprid only about 20% in the presence of ASA/Ca 2+ hydrogel. These results would be helpful for the formation of soil aggregation and pesticides adsorption on soil aggregation. Thus, ASA/Ca 2+ hydrogel is likely to improve soil quality, simultaneously it can minimize the mobility of pesticides in the agricultural system.

Fabrication and characterization of bamboo shoot cellulose/sodium alginate composite aerogels for sustained release of curcumin

Aiping Zhang,Yucheng Zou,Yuhang Xi,Peng Wang,Yue Zhang,Liangru Wu,Hui Zhang

The feasibility of using unmodified bamboo shoot cellulose (BSC) to produce composite aerogels with sodium alginate (SA) in a fast and green way for sustained release of curcumin was explored for the first time, in which calcium ion-induced SA cross-linking could effectively retain the structural stability of aerogel skeleton. The aerogels were characterized by scanning electron microscopy, Fourier transform infrared spectroscopy and differential scanning calorimetry . The encapsulation and release of curcumin from aerogels were studied while the antioxidant activity of encapsulated curcumin was investigated. Curcumin was evenly encapsulated in the composite aerogels and showed a sustained release behavior, followed the first-order rate expression. Interpenetrating network structures were built between BSC and SA mainly through hydrogen bonding, which could be further reinforced by the cross-linking of CaCO 3 on the SA matrix. The original characteristics of BSC in the composite aerogels were well retained. The thermal stability and mechanical properties of the composite aerogels were improved by Ca 2+ -induced cross-linking, while the uncross-linked composite aerogels exhibited better encapsulation efficiency and in vitro antioxidant activity. Overall, this study was the first to use cellulose from bamboo shoot to develop aerogels for drug delivery purposes. The cellulose/alginate composite aerogels were promising to be used as biocompatible carriers for drug and nutraceutical delivery.

Adsorption of As(III), Pb(II), and Zn(II) from Wastewater by Sodium Alginate Modified Materials

Hongchuan Li,Rui Jin,Hongxiang Hu,Yusef Kianpoor Kalkhajeh,Yingying Zhao,Yue Gao,Borui Zhang

Sodium alginate (SA), polyvinyl oxide (PEO), and ceramic nanomaterials were used to prepare alginate composite gel. The present study examined the removal rate and adsorption capacity of alginate composite gel for removal of wastewater As(III), Pb(II), and Zn(II). Batch experiments were conducted to study the influence of experimental parameters such as pH and temperature, as well as the mechanism of As(III), Pb(II), and Zn(II) adsorption with the new adsorbent. The results showed the high efficiency of sodium alginate composite gel for removal of wastewater As(III), Pb(II), and Zn(II). Under the condition of the best liquid-solid ratio and the contact time, the removal rates of As(III), Pb(II), and Zn(II) were 67.42%, 95.31%, and 93.96%, respectively. The pseudo-second-order kinetic equation was superior to fit the adsorption kinetics process. The isothermal adsorption models of As(III) and Pb(II) fitted well with the Freundlich model, and Zn(II) fitted well with the Langmuir model. The results of SEM, EDS, XPS, and FTIR analyses revealed that the adsorption process occurred mainly via chemisorption. The results of the present study suggest that new adsorbents can be effectively utilized for As(III), Pb(II), and Zn(II) removal from water.

Dynamic regulable sodium alginate/poly(γ-glutamic acid) hybrid hydrogels promoted chondrogenic differentiation of stem cells

Penghui Wang,Yajie Pu,Yanhan Ren,Rong Yang,Wenjie Zhang,Xiaoyan Tan,Wenliang Xue,Shuai Liu,Shuang Li,Bo Chi

Traditional hydrogels often fail to match the dynamic interactions between mechanical and cellular behaviors exhibited by the natural cartilage extracellular matrix. In this research, we constructed a novel hybrid hydrogels system based on sodium alginate and polyglutamic acid. By controlling the grafting rate and concentration of polymer, the gelation time and mechanical strength can be adjusted between range of 8–28 s and 60–144 kPa. By adding microcrystalline cellulose into the system, so that the degradation time was prolonged (125%) and the swelling rate was reduced (470%). Additionally, the presence of hydrazone bonds gives the system some dynamic response characteristics, and the hydrogel exhibits excellent self healing and injectable ability. It was found that the system had positive cytocompatibility (80%), which accelerated regulatory gene expression in cartilage tissue. In conclusion, this injectable hydrogel with self-healing and customizable mechanical strength will have broad application prospects in future biomedical engineering.

Synthesis and assessment of sodium alginate-modified silk fibroin microspheres as potential hepatic arterial embolization agent

Guobao Chen,Runan Wei,Xiang Huang,Fuping Wang,Zhongmin Chen

Transcatheter arterial chemoembolization (TACE) is well known as an effective treatment for hepatocellular carcinoma (HCC). In the present study, a novel embolic agent of sodium alginate (SA)-modified silk fibroin (SF) microspheres was successfully prepared by emulsifying cross-linking method. The SA-modified SF microspheres were evaluated for the ability of embolization by investigating the morphology, particle size, swelling ratio, degradation, cytotoxicity, blood compatibility, and in vivo embolization. The results found that SA-modified SF microspheres had smooth surfaces and good sphericity. Swelling ratio of the microspheres can meet the requirements of arterial embolic agent and have pH and temperature sensitivity. Furthermore, hemolytic and anticoagulant studies have proved that the microspheres have good blood compatibility. Cytotoxicity tests indicated that the microspheres could promote the proliferation of fibroblasts and HUVEC. In vivo embolization evaluation of microspheres revealed that the arteries could be embolized by SA-modified SF microspheres in 3 weeks. The ability of drug loading and releasing of microspheres was proved by the controlled release profile of Adriamycin hydrochloride. The findings indicated that the SA-modified SF microspheres can be used as a potentially biodegradable arterial embolic agent for liver cancer therapy.

Formation of helical alginate microfibers using different G/M ratios of sodium alginate based on microfluidics

Rui Liu,Bin Kong,Yun Chen,Xueping Liu,Shengli Mi

The helical structure is one of the most intriguing structures in nature; such an ingenious structure can be found from macromolecules to macroscopic areas, and the fabrication of helical structures has attracted research interest from mechanisms to applications. However, the fabrication of helical microfiber still remains a big challenge. In this study, a coaxial microfluidic device was made to continuously and controllably fabricate helical hydrogel microfibers with flexible shape by simply adjusting the flowrates. Various experimental conditions that can influence the diameter (d), pitch (p) and amplitude (A) of these formed coiled fibers were discussed. For the first time, the effects of G/M ratio of the alginates on the microfiber coiling phenomenon were examined. Additionally, magnetically responsive helical fibers were synthesized, and it is envisioned that hydrogel helical structure microfibers are highly versatile in different area, such as micro sensors, soft actuators and medical applications.

Preparation and characterisation of a novel silk fibroin/hyaluronic acid/sodium alginate scaffold for skin repair

Wei Yang,Hongjie Xu,Yong Lan,Qiyu Zhu,Yu Liu,Shaoshan Huang,Shengjun Shi,Andrei Hancharou,Bing Tang,Rui Guo

To mimic the natural structure of tissue extracellular matrix, a novel silk fibroin (SF)/hyaluronic acid (HA)/sodium alginate (SA) composite scaffold (92% in porosity) was prepared by freeze-drying. Fourier-transform infrared spectroscopy and Raman spectra indicated interactions among SF, HA, and SA molecules. Scanning electron microscopy showed that the prepared SF/HA/SA scaffold had soft, elastic characteristics, with an average pore diameter of 93 μm. Mechanical property, thermogravimetric analyses and degradation results indicated that the SF/HA/SA scaffold had good physical stability in body fluid and mechanical movement-related environments. Cell proliferation, morphological, and live-dead analyses showed that NIH-3T3 fibroblast cells were better able to attach, grow, and proliferate on the SF/HA/SA scaffold compared with SF, SF/HA, and SF/SA scaffolds. We evaluated the wound healing effects in a rat full-thickness burn model. The hematoxylin-eosin (H&E) and Masson's trichrome staining results from SF/HA/SA scaffold showed that improved re-epithelialization, enhanced extracellular matrix remodeling. Our findings showed that the prepared SF/HA/SA scaffold can provide a potential way as a wound dressing for skin repair.

Effect of Sodium Alginate on the Morphology and Properties of High Energy Insensitive Explosive TKX-50

Wenbo Dong,Shusen Chen,Shaohua Jin,Minglei Chen,Bo Yan,Yu Chen

Dihydroxylammonium 5,5′-bistetrazole-1,1′-diolate (TKX-50) is an energetic ion salt with high energy and low mechanical sensitivity. However, its poor morphology and low binding ability with conventional binders have significantly limited its application. In the present work, we reported a novel strategy to improve the morphology and surface properties of TKX-50 using sodium alginate (SA) as the modifier by the anti-solvent method. The results of imaging characterization indicated that SA greatly affected both crystal habit and morphology of TKX-50, resulting in modified TKX-50 granulars with low aspect ratios. The formation mechanism was explored by the molecular dynamics simulation. The XPS and SEM characterizations also revealed that SA adhered onto TKX-50 surface to form a coating layer. The non-isothermal kinetic parameters were calculated from DSC results. Our work has provided a new strategy to improve the morphology and surface properties of TKX-50 for its practical applications.

κ-Carrageenan/Sodium alginate double-network hydrogel with enhanced mechanical properties, anti-swelling, and adsorption capacity

Fei Yu,Tianran Cui,Changfu Yang,Xiaohu Dai,Jie Ma

The abuse of antibiotics is becoming increasingly serious, particularly in offshore aquaculture areas. Double-network polymer gel has good prospects for environmental application for the removal of antibiotics. In this work, κ-Carrageenan/Sodium Alginate (κ-car/SA) double-network hydrogels were designed and synthesized with enhanced mechanical properties, anti-swelling, and adsorption capacity. It was found that the intermolecular interaction and viscosity tend to increase with the increasing concentration of κ-carrageenan. The swelling degree of the composite hydrogel in NaCl solution presented a decreasing trend with the increase of carrageenan. SA can effectively improve the mechanical properties of κ-carrageenan composite gel and enhance its compressive resistance and elasticity. Ciprofloxacin (CIP) was used as the model pollutant for testing the adsorption performance. The results show that the Langmuir-Freundlich isotherm model is more suitable for fitting the adsorption isotherm data of CIP on gel beads, which indicates that κ-car/SA hydrogels have heterogeneous surface and different binding sites. κ-car/SA composite double-network hydrogels exhibit excellent adsorption properties for CIP (229 mg/g). The optimal adsorption capacity of κ-car/SA composite hydrogels was obtained at pH 4, and the adsorption capacity of the hydrogels increased with increasing ion concentration . FTIR spectroscopy and the Zeta potential test analyses showed that the adsorption mechanism may be explained by hydrogen bonding and the electrostatic interactions between κ-car/SA composite hydrogels and CIP. The formation of the new double-network hydrogel provided good properties and development potential for the adsorption of antibiotic in water.

Fabrication of chitosan-coated konjac glucomannan/sodium alginate/graphene oxide microspheres with enhanced colon-targeted delivery

Yi Yuan,Xiaowei Xu,Jingni Gong,Ruojun Mu,Yuanzhao Li,Chunhua Wu,Jie Pang

Microspheres play an increasingly important role in the food and medicine industries. In this study, konjac glucomannan (KGM)/sodium alginate (SA)/graphene oxide (GO) solution was injected into CaCl 2 solution under high-voltage static electricity assistance to fabricate microspheres. Then, chitosan (CS) was coated on the surface of the microspheres to enhance their stability. SEM images confirmed that increasing voltage decreased the particle size of microspheres obviously. Furthermore, GO was beneficial in maintaining the full structure of freeze-dried microspheres, and the CS membrane improved the surface of the microspheres with no relatively obvious gully. Results indicated that KGM interacted with SA by hydrogen bond, and GO improved this interaction in microspheres. Furthermore, swelling tests showed that the microspheres exhibited different swelling properties in different media, and the CS membrane could improve the stability of microspheres in simulated intestinal fluid and simulated colon fluids. Moreover, GO could greatly improve the ciprofloxacin (CPFX) loading efficiency of microspheres, and achieving a sustained release effect of CPFX. Thus, CS-coated KGM/SA/GO microspheres showed great potential application in drug and/or nutrition factor colon-targeted delivery.

Bio-inspired and lanthanide-induced hierarchical sodium alginate/graphene oxide composite paper with enhanced physicochemical properties

Shunli Liu,Jun Ling,Kewen Li,Fang Yao,Olayinka Oderinde,Zhihong Zhang,Guodong Fu

This paper reports an artificial nacre-like composite paper based on sodium alginate (SA) and graphene oxide (GO) by lanthanide ions cross-linking. SA-coated GO were used as building “bricks and mortar” and self-assembled into aligned GO/SA composite hydrogel by the coordination of lanthanide ions. Subsequently, the hierarchical GO/SA composite paper was formed under evaporation. Four types of lanthanide ions (Nd 3+ , Ce 3+ , Gd 3+ , and Yb 3+ ) were used to fabricate hierarchical GO/SA composite papers. The lanthanides enhanced the mechanical properties of the GO/SA composite papers (strength of 255.8 ± 8.52 MPa and toughness of 4.83 ± 0.28 MJ m −3 ) and endowed the papers with high stability. Due to the anisotropic nature along parallel and perpendicular directions , the hierarchical GO/SA composite papers exhibited a specific thermal conductivity . These features greatly expand the application of the composite papers, which are believed to show competitive advantages in aerospace, electronic devices, and thermal interface materials .
