Ethanopharmacological relevance Potentilla kleiniana Wight et Arn is a wide-spread wild plant in the mountainous areas in southern China. The whole herb has been used as a traditional herbal medicine to treat fever, arthritis , malaria, insect and snake bites , hepatitis, and traumatic injury. In vitro studies have reported the antibacterial activity use of the plant in traditional medicinal systems. Aim of the study The aim of this study was to investigate the inhibitory activity of total flavonoid from Potentilla kleiniana Wight et Arn (TFP) on methicillin-resistant Staphylococcus aureus (MRSA) in planktonic state and biofilm state. Materials and methods Antibacterial activities of TFP on planktonic MRSA were determined by agar diffusion method, microtiter plate assay and time-kill curve assay. Electrical conductivity, membrane permeability, membrane potential and autoaggregation were analyzed to study TFP effects on planktonic MRSA growth. Crystal violet (CV) staining and confocal laser scanning microscopy (CLSM) were analyzed to study TFP effects on aggregation and maturation of MRSA biofilm. After TFP treatment, extracellular polymeric substances (EPS) production were examined. Morphological changes in planktonic and MRSA biofilm following TFP treatment were determined with scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Moreover, α-Toxin protein expression and adhesion-related gene expression were also determined. Results The minimum inhibitory concentration (MIC) of TFP against MRSA was 20 μg/mL. The agar diffusion method and time-kill curve assay results indicated that TFP inhibited planktonic MRSA growth. TFP treatment significantly inhibited planktonic MRSA growth by inhibiting autoaggregation, α-hemolysin activity, α-Toxin protein expression, but increasing electrolyte leakage, membrane permeability and membrane potential and impacting cell structure. Moreover, TFP treatment significantly inhibited aggregation and maturation on MRSA biofilm by decreasing surface hydrophobicity , EPS production and adhesion-related gene expression. Conclusion The results of this trial provide scientific experimental data on the traditional use of Potentilla Kleiniana Wight et Arn for traumatic injury treatment and further demonstrate the potential of TFP to be developed as a novel anti-biofilm drug.

Biofilms have been related to the persistence of infections on medical implants, and these cannot be eradicated because of the resistance of biofilm structures. Therefore, a biocompatible phototherapeutic system is developed composed of MoS 2 , IR780 photosensitizer, and arginine–glycine–aspartic acid–cysteine (RGDC) to safely eradicate biofilms on titanium implants within 20 min. The magnetron-sputtered MoS 2 film possesses excellent photothermal properties, and IR780 can produce reactive oxygen species (ROS) with the irradiation of near-infrared (NIR, λ = 700–1100 nm) light. Consequently, the combination of photothermal therapy (PTT) and photodynamic therapy (PDT), assisted by glutathione oxidation accelerated by NIR light, can provide synergistic and rapid killing of bacteria, i.e., 98.99 ± 0.42% eradication ratio against a Staphylococcus aureus biofilm in vivo within 20 min, which is much greater than that of PTT or PDT alone. With the assistance of ROS, the permeability of damaged bacterial membranes increases, and the damaged bacterial membranes become more sensitive to heat, thus accelerating the leakage of proteins from the bacteria. In addition, RGDC can provide excellent biosafety and osteoconductivity, which is confirmed by in vivo animal experiments.

Ethnopharmacological relevance Jie-Geng-Tang (JGT), a famous traditional Chinese medicine prescription, consists of Platycodonis Radix and Glycyrrhizae Radix et Rhizoma . According to traditional medicinal theory, JGT exerts various effects, including apocenosis, detoxifying, moisturizing the lung and relieving sore throat. It is often used to treat throat inflammation and lung diseases. Aim of the study To determine the protective effect of JGT on Staphylococcus aureus ( S. aureus )-induced acute lung injury (ALI) in mice and to identify the compounds in the prescription that may be responsible for antibacterial activity . Materials and methods The protective effect of JGT was assessed using S. aureus -induced ALI mice (i.g., 2.7 g/kg/day). Bacterial burden, pathological morphology, cytokine levels of TNF-α, IL-1β, KC, and MIP-2 were evaluated in the lung and bronchoalveolar lavage fluid at 24 h post-infection, respectively. Twenty three compounds in the prescription were evaluated for their minimum inhibitory concentration (MIC) in vitro by means of microbroth dilution method against S. aureus . The antibacterial effects in vitro of licochalcone A and isoliquiritigenin were also investigated by transmission electron microscopy . In vivo antibacterial activities of licochalcone A and isoliquiritigenin were evaluated by survival rates, bacterial burden, and pathological morphology of lung tissues on S. aureus -induced ALI in mice (i.p., 160 mg/kg/day). Results Pretreatment with JGT significantly improved the pathological morphology of lung tissues on S. aureus -induced ALI in mice, accompanied with the reduced bacterial burden in the lungs and inhibited expression of inflammatory cytokine levels at 24 h post-infection. Five compounds, namely licochalcone A, licoisoflavone B, glyasperin A, isoliquiritigenin, and licochalcone B from Jie-Geng-Tang displayed good antibacterial activities against S. aureus (MIC < 128 μg/mL). Furthermore, applications of licochalcone A and isoliquiritigenin resulted in the increased survival rates, reduced bacterial burden in the lungs, and improved pathological morphology of lung tissues in S. aureus infected mice. Conclusion The study demonstrated that Jie-Geng-Tang presented protective role of acute lung injury, which supported its traditional use for the treatment of lung diseases. Licochalcone A, isoliquiritigenin, licoisoflavone B, glyasperin A, and licochalcone B might contribute to the antibacterial activity of JGT on S. aureus -induced acute lung injury. The anti- S. aureus activity of licoisoflavone B, glyasperin A, and licochalcone B in vitro , as well as the anti- S. aureus activity of licochalcone A in vivo , were first reported in this study.

Timely and accurate anti-infective therapy significantly benefits treatment of various life-threatening bacterial infectious diseases. Unfortunately, owing to the lack of selectivity for recognizing bacterial species, most of the reported phenotypic antimicrobial susceptibility testing protocols suffer from time-consuming bacterial isolation and identification. In this work, a highly-selective molecular recognition-based sandwich fluorimetric assay was developed to rapidly detect Staphylococcus aureus ( S. aureus ), and furtherly to assess its susceptibility to antibiotics. Immobilized porcine IgG was utilized to capture S. aureus through the selective interaction between Fc fragment of porcine IgG and protein A in the cell wall of S. aureus . Fluorescein isothiocyanate-labeled teicoplanin was adopted as the signal tracer utilizing the binding behavior between teicoplanin and d -Ala- d -Ala moieties in the peptidoglycans of Gram-positive bacterial cell wall. S. aureus could be detected within a wide linear rang of 1.0 × 10 3 to 1.0 × 10 7 CFU mL −1 . Subsequently, this protocol utilizing porcine IgG and teicoplanin as the indicator molecules was utilized to assess the antimicrobial susceptibility of S. aureus after the bacteria were treated with antibiotics. The minimum inhibitory concentrations of penicillin, cefoxitin, clindamycin, trimethoprim/sulfamethoxazole and erythromycin were estimated to be >0.25,<4, <0.25, <2/38 and <0.5 μg mL −1 , respectively. The whole process for antimicrobial susceptibility testing can be completed within 4 h since it avoided extra time-consuming isolation and identification of bacterial species.

Despite the development of various drugs and materials, bacterial infections remain a serious problem for wound healing. Most current materials cannot simultaneously provide antibacterial effects, reusability, and biocompatibility, and participate in stimulating cellular behaviour to promote bacterial wound healing. Herein, multifunctional nitrogen-rich carbon-coated bismuth/cobalt nanoparticles ( [email protected] @CN) with foam-like structures synthesised by a simple two-step method showed remarkably improved magnetic and photoconversion effects. These are promising candidates as photodynamic inactivation materials. Within 20 min, >99.999% Staphylococcus aureus ( S. aureus ) were killed by the composite due to its strong ability to generate reactive oxygen species (ROS) and thermal energy under simulated visible light. Additionally, [email protected] @CN also displayed high stability: its antibacterial efficiency underwent rapid photodynamic inactivation over five cycles. In vitro results revealed that ROS and photothermal conversion of materials can seriously damage bacterial cell membranes and inhibit the virulence factors produced by bacteria, which ultimately leads to the death of S. aureus . Moreover, in vivo assessment of wound healing showed that the material was very effective for bacteria-accompanied wound healing and did not cause detectable damage to the major organs. Therefore, the [email protected] @CN composite has great potential as a safe multimodal treatment system for active photodynamic inactivation and wound healing.

The chemical composition and antimicrobial mechanism of action of black pepper chloroform extract (BPCE) were investigated, as well as the potential antibacterial activities of BPCE against Escherichia coli and Staphylococcus aureus. The results showed that 1H-Cycloprop[e]azulen-7-ol, decahydro-1,1,7-trimethyl-4-methylene-, [1ar-(1aα,4aα,7β,7a,β,7bα.)]- (8.39%) and 2-methylene-4,8,8-trimethyl-4-vinyl-bicyclo[5.2.0]nonane (6.92%) were identified as the two primary components of BPCE. The release of intracellular transaminases from bacteria after being incubated with BPCE revealed that the bacterial cell walls and membranes were degraded and that protein synthesis was inhibited to some extent. The inhibition of bacterial Na+/K+-ATPase activity upon the addition of BPCE also indicated an enhanced permeability of bacterial cell membranes. Moreover, an analysis of hexokinase and pyruvate kinase activities showed that BPCE affected the metabolic rate of glycolysis and disrupted the normal metabolism of bacteria. This phenomenon was supported by an observed accumulation of lactic acid (LA) in the treated bacterial cells. Overall, our results indicated that BPCE damaged bacterial cell walls and membranes, which was followed by a disruption of bacterial cell respiration.

The key factor to control the incidence rate of diseases caused by bacteria is rapid detection and early diagnosis. Herein, we proposed a new electrochemical bacterial sensor by coupling DNA walking and rolling circle amplification (RCA) reaction-assisted “silver-link” crossing electrode. Staphylococcus aureus ( S. aureus ) was detected using this proof-of concept strategy. Aptamer/DNA walker and auxiliary sequence (AS)/RCA reaction probe (RP) duplexes were modified on the electrode surface . The binding of S. aureus with its aptamer caused the disintegration of aptamer/DNA walker and released DNA walker. With the help of Exo III, DNA walker moved along the electrode surface and AS in AS/RP duplex was continuously digested to release RP. By introducing phi29 DNA polymerase, RCA reaction was performed using RP as the reaction primer to form long single-strand RCA extension products between the electrodes. The “silver-link” crossing electrode was formed by metallization of “gene-link”, significant conductivity was thus acquired for bacteria detection . The limit of detection (LOD) was 10 CFU/mL and detection time was 2 h. The proposed sensor has high efficiency, good stability and low background signal, human serum and milk samples were successfully detected, which emerged a promising potential in the food monitoring and clinical diagnosis.

Background: In the field of antibacterial, nanomaterials are favored by researchers because of their unique advantages. Medicinal plants, especially traditional Chinese medicine, are considered to be an important source of new chemicals with potential therapeutic effects, as well as
an important source for the discovery of new antibiotics. MRSA is endangering people's lives as a kind of multidrug-resistant Staphylococcus aureus, which is resistant to tetracycline, amoxicillin, norfloxacin and other first-line antibiotics. It is a hotspot to find good anti-drug-resistant
bacteriae, nature-originated nanomaterials with good biocompatibility. Objective: We reported the formation of phytochemical nanoparticles (NPs) by the self-assembly of berberine (BBR) and 3,4,5-methoxycinnamic acid (3,4,5-TCA) from Chinese herb medicine, which had good antibacterial
activity against MRSA. Methods and Results: We found that NPs had good antibacterial activity against MRSA; especially, its antibacterial activity was better than first-line amoxicillin, norfloxacin and its self-assembling precursors on MRSA. When the concentration reached 0.1 μmol/mL,
the inhibition rate of NPs reached 94.62%, which was higher than that of BBR and the other two antibiotics (p < 0.001). It was observed by Field-Emission Scanning Electron Microscopy (FESEM) that NPs could directly adhere to the bacterial surface, which might be an important aspect of the
antibacterial activity of NPs. Meanwhile, we further analyzed that the self-assembly was formed by hydrogen bonds and π-π stacking through Ultraviolet-Visible (UV-vis), Fourier Transform Infrared Spectroscopy (FTIR), hydrogen Nuclear Magnetic Spectrum (1H NMR), and powder X-ray Diffraction
(pXRD). NPs’ morphology was observed by FESEM and TEM. The particle size and surface charge were characterized by Dynamic Light Scattering (DLS); and the surface charge was -31.6 mv, which proved that the synthesized NPs were stable. Conclusion: We successfully constructed a naturally
self-assembled nanoparticle, originating from traditional Chinese medicine, which had a good antibacterial activity for MRSA. It is a promising way to obtain natural nanoparticles from medicinal plants and apply them to antibacterial therapy.

The emergence of multidrug-resistant (MDR) bacteria represented by MRSA (Methicillin-resistant Staphylococcus aureus ) poses a great challenge to current anti-infection treatment. It is critical to develop efficient MRSA anti-bacteria drugs and explore simple therapeutic strategies with low MDR risk. Herein, we synthesized high-valent (Ag II /Ag III ) water-soluble porphyrins (cationic AgTMPyP and anionic AgTMPPS) and investigated their direct bactericidal property for MRSA without photoactivation in vitro and in vivo. The cationic porphyrin AgTMPyP exhibits well oxidase-like activity and has 100 % sterilizing rate at 8 μmol/L concentration. Besides, AgTMPyP can effectively destroy biofilms in vitro , mediate the polarization of macrophages from M1 to M2, and promote wound healing in vivo . Combined with DFT calculation, the related antibacterial mechanism is further discussed. High-valent silverporphyrins can maintain stable in water for at least 200 days. The moment they encounter MRSA, high-valent silver ions from AgTMPyP can be immediately released from the porphyrin ring and attack the MRSA with efficient sterilization. Together with the hemolysis, blood routine and blood biochemistry tests, it is proved that AgTMPyP can have great prospects in the direct treatment of bacterial infections in skin diseases in the future. Statement of significance The emergence of multidrug-resistant (MDR) bacteria represented by MRSA poses a great challenge to current anti-infection treatment. It has become critical to develop efficient MRSA anti-bacteria drugs and explore simple therapeutic strategies with low MDR risk. We synthesized high-valent (Ag II /Ag III ) water-soluble silver porphyrins (AgTMPyP and AgTMPPS), which can be stable for long periods in aqueous solutions. AgTMPyP can directly and efficiently kill bacteria and destroy biofilms without photoactivation in vitro and in vivo . Combined with DFT calculation, the related antibacterial mechanism is further discussed. AgTMPyP is a superior antimicrobial agent with good biocompatibility and it can have great prospects in the direct treatment of bacterial infections and wound healing in the future.

Bacteriophage-based biosensors have the potential for rapid, convenient, and sensitive detection of Staphylococcus aureus in food products; however, their effectiveness is often limited by the host range of the bacteriophage. This study presents a new colorimetric biosensor using the S. aureus phage SapYZU04 exhibiting a broad host range, and a MnFeO nanozyme for the effective detection of S. aureus in food products. Notably, SapYZU04 exhibited excellent features, such as a high titre (1.0 × 10 10 PFU/mL), a short latency period (5 min), a large burst size (210 PFU/cell), a wide host spectrum (98.95%, 94/95), and good stability at pH 3.0–12.0. The biosensor, referred to as SapYZU04@MnFeO was constructed by immobilising SapYZU04 on the MnFeO nanozyme via electrostatic interactions. SapYZU04@MnFeO demonstrated a broad detection range of bacterial concentrations, particularly within the range of 3 × 10 2 to 3 × 10 9 CFU/mL, with a low detection limit of 65 CFU/mL and rapid detection time (14 min). Furthermore, it exhibited high accuracy in detecting and quantifying live S. aureus with recovery rates ranging from 92.21% to 105.56%. The chromogenic system demonstrated high selectivity for S. aureus and was unaffected by other interfering microorganisms or food preservatives, highlighting its specificity and interference-resistance. Whole-genome sequencing of SapYZU04 suggested that the selectivity, sensitivity, and specificity of SapYZU04@MnFeO could be attributed to the tail fibre proteins of the phage. In conclusion, the SapYZU04@MnFeO biosensor is a highly effective tool for identifying viable S. aureus strains in the food industry.

Staphylococcus aureus ( S. aureus ), a prevalent foodborne pathogen, poses a significant threat to public health. Rapid, specific, and sensitive on-site pathogen detection methods are crucial for food safety. In this study, a ternary MXene/WS 2 /Au NPs composite was synthesized via a solvothermal and in situ growth method. This composite serves dual purposes as a photoactive material for photoelectrochemical (PEC) aptasensing and as an effective antibacterial catalyst. A PEC aptasensor was developed using MXene/WS 2 /Au NPs as the substrate and a ferrocene-modified aptamer as the recognition probe, enabling quantitative detection of S. aureus . The sensor exhibits a wide linear range (1.0 × 10–1.0 × 10 7 CFU/mL) and an ultra-low detection limit (3.0 CFU/mL). Additionally, the nanocomposite demonstrates strong peroxidase-like activity, efficiently catalyzing H 2 O 2 to generate ·OH, offering broad-spectrum antibacterial properties for pathogen control in food safety monitoring.

Staphylococcus aureus ( S. aureus )–driven senescence of bovine mammary epithelial cells is a key determinant of mammary gland health, yet its molecular basis remains poorly defined. Sirtuin 5 (SIRT5), a mitochondria-localized desuccinylase, may play an important regulatory role in this process. This study aimed to elucidate the mechanisms by which S. aureus drives cellular senescence and to define the contribution of the SIRT5–mitochondrial axis to delaying senescence. We found pronounced oxidative stress and cellular senescence in mammary tissues from cows with S. aureus mastitis, accompanied by marked downregulation of SIRT5. In an S. aureus -infected epithelial cell model, infection induced mitochondrial stress characterized by excessive mitochondrial fragmentation, loss of membrane potential, and increased mitochondrial superoxide, along with oxidative damage and cellular senescence. Mechanistically, S. aureus toxins and the toxin-induced inflammatory response cooperatively drove mitochondrial stress, which in turn increased intracellular bacterial burden and exacerbated cell death. During infection, SIRT5 protein abundance was significantly reduced. Mass spectrometry and co-immunoprecipitation analyses indicated that infection upregulated the ubiquitin-conjugating enzyme ubiquitin C (UBC), enhanced its interaction with SIRT5, and promoted ubiquitin-mediated degradation of SIRT5. Loss of SIRT5 increased succinylation of dynamin-related protein 1 (DRP1), inhibited its ubiquitin-mediated degradation, and led to its excessive accumulation on the outer mitochondrial membrane, thereby promoting excessive mitochondrial fission. Functionally, SIRT5 overexpression markedly alleviated mitochondrial stress, oxidative damage, and senescence phenotypes. When mitochondrial fission was forcibly enhanced, the cytoprotective effect of SIRT5 was substantially weakened, confirming that SIRT5 acts through a pathway dependent on mitochondrial integrity. Collectively, S. aureus infection releases toxins and induces inflammatory injury, during which UBC-mediated SIRT5 degradation activates DRP1-dependent mitochondrial hyper-fragmentation, aggravating mitochondrial stress, oxidative stress, and mammary epithelial cell senescence. These findings identify SIRT5 as a critical regulator of redox and mitochondrial homeostasis in mammary epithelial cells and a potential therapeutic target for mitigating oxidative damage associated with bovine mastitis.
Staphylococcus aureus ( S. aureus ) is a leading cause of bovine mastitis, an intramammary infection that reduces milk yield, increases antibiotic use, and results in substantial economic loss. One way this bacterium damages the udder is by driving bovine mammary epithelial cells into premature senescence, but how infection triggers this process has not been fully understood. Here, we show that S. aureus infection induces an inflammatory response via its secreted toxins, which in turn provokes mitochondrial stress. We identify the mitochondrial enzyme Sirtuin 5 (SIRT5) as a key defender in this context. During infection, the host ubiquitin-conjugating enzyme ubiquitin C (UBC) targets SIRT5 for degradation. Loss of SIRT5 leads to accumulation of the mitochondrial fission protein dynamin-related protein 1 (DRP1), causing excessive mitochondrial fragmentation, enhanced oxidative damage, increased intracellular bacterial burden, and aggravated cellular senescence. Restoring SIRT5 helps to preserve mitochondrial integrity and reduces oxidative injury and senescence; however, this protection is weakened when mitochondrial fission is forcibly maintained at a high level. Together, our findings reveal that the UBC–SIRT5–DRP1 pathway links S. aureus infection to mitochondrial damage and cellular senescence, and they highlight SIRT5 as a potential therapeutic target to protect the bovine mammary gland (S1 Fig).
