Dielectric barrier discharge plasma-modified chitosan flocculant and its flocculation performance

Quanfa Zhao,Haixia Wu,Wang Shen,Xiao Han,Bin Zheng,Yong Wang

The flocculation performance of chitosan can be enhanced by grafting modification to overcome its disadvantages of poor water solubility. In this study, chitosan was modified by dielectric barrier discharge plasma and polymerized with acrylamide and aluminum chloride to synthesize a new chitosan-based flocculant, namely, chitosan–acrylamide–aluminum chloride (CA-PAC). After optimizing the synthesis conditions of CA-PAC, the best conditions were as follows: discharge time of 3 min, discharge power of 50 W, polymerization temperature of 60 °C, polymerization time of 3 h, total monomer concentration of 100 g/L, and m(AlCl 3 ):m(CA) ratio of 2:1. Characterization was performed through SEM, XPS , FTIR , XRD, TG and 1 H NMR. Results showed that the preparation of CA-PAC was successful. The influences of flocculant dosage, pH, and stirring intensity on flocculation efficiency were investigated. The removal efficiency of turbidity was 94.1 %. The investigation of the flocculation mechanism revealed that CA-PAC mainly relied on charge neutralization or the synergic action of electric neutralization, bridging, and roll–sweep under acidic and neutral conditions, but it depended on the joint action of adsorption bridging and net sweeping under alkaline conditions. This study provides new ideas for the preparation and development of modified chitosan and broadens its application in water treatment.

Water-in-oil Pickering emulsions stabilized by phytosterol/chitosan complex particles

Chenglei Wang,Hang Jiang,Yunxing Li

The development of colloidal particles for the preparation of water-in-oil (w/o) Pickering emulsions is highly desirable for food, pharmaceuticals, and cosmetics. However, the lack of bio-based hydrophobic particulate stabilizers makes it remain a great challenge. Herein a water-in-cyclopentasiloxane (w/o) Pickering emulsion was prepared using phytosterol/chitosan (PS/CS) complex particles as the stabilizer. Adsorption of CS onto the surface of PS particles via electrostatic interaction changed the particle wettability and interfacial activity, leading both PS particles and CS to be absorbed at the w/o interface. By varying the pH and ionic strength of aqueous phase, the stabilizing mechanism was verified and the results suggested that the formation of PS/CS complex particles was critical in the preparation of w/o Pickering emulsions with small and uniform droplets. The PS/CS complex particles can emulsify a broad range of water volume fraction and even be used to prepare a high internal phase w/o Pickering emulsion. Meanwhile, the as-prepared w/o Pickering emulsions had good long-term stability, especially at ambient or lower temperature. Finally, we demonstrated that the prepared w/o Pickering emulsion could effectively protect encapsulated epigallocatechin gallate from heat and pH shock.

A PEGylated Chitosan as Gel Polymer Electrolyte for Lithium Ion Batteries

Anqi Wang,Yue Tu,Sijie Wang,Hongbing Zhang,Feng Yu,Yong Chen,De Li

Due to their safety and sustainability, polysaccharides such as cellulose and chitosan have great potential to be the matrix of gel polymer electrolytes (GPE) for lithium-based batteries. However, they easily form hydrogels due to the large numbers of hydrophilic hydroxyl or amino functional groups within their macromolecules. Therefore, a polysaccharide-based amphiphilic gel, or organogel, is urgently necessary to satisfy the anhydrous requirement of lithium ion batteries. In this study, a PEGylated chitosan was initially designed using a chemical grafting method to make an GPE for lithium ion batteries. The significantly improved affinity of PEGylated chitosan to organic liquid electrolyte makes chitosan as a GPE for lithium ion batteries possible. A reasonable ionic conductivity (1.12 × 10−3S cm−1) and high lithium ion transport number (0.816) at room temperature were obtained by replacing commercial battery separator with PEG-grafted chitosan gel film. The assembled Li/GPE/LiFePO4coin cell also displayed a high initial discharge capacity of 150.8 mA h g−1. The PEGylated chitosan-based GPE exhibits great potential in the field of energy storage. Keywords:PEGylated chitosan;gel polymer electrolyte;lithium ion batteries

Preparation, characterisation and antibacterial properties of thermoplastic chitosan/nano ZnO composites

Yaoxin Zhang,Lixia Huang,Taoxing Dong,Bin Li,Yu Zhang

This study prepares thermoplastic chitosan/nano ZnO (CZ) composites by thermomechanical treatment in an internal mixer. Dilute hydrochloric acid aqueous solution is used as the destructuring agent of chitosan and solvent of nano ZnO. Fourier transform infrared spectroscopy and scanning electron microscopy confirm the chitosan–ZnO interaction. The crystallinity and UV transmittance of the composites decreases, while their thermal stability increases due to the addition of nano ZnO. The antibacterial activity of the CZ composites is examined in terms of their inhibitory and bactericidal effects against Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. Improved antibacterial properties are obtained for the CZ composites, compared to pure chitosan sheet. Gram-positive bacteria (S. aureus and B. subtilis) are more sensitive to the antibacterial activity of the CZ composites than Gram-negative bacteria (E. coli). This work provides a simple method to prepare chitosan-based materials and obtains CZ composites with improved antibacterial activities, which can be applied in agriculture and food safety.

Enzymatically crosslinked hydrogel based on tyramine modified gelatin and sialylated chitosan

Peng Ding,Qianqian Wei,Ning Tian,Xiaoyue Ding,Ling Wang,Bin Wang,Oseweuba Valentine Okoro,Amin Shavandi,Lei Nie

The enzymatically crosslinked hydrogel could replicate the cellular microenvironment for biomedical applications. In the present study, to improve the cytocompatibility of chitosan (CS), sialic acid (SA) was introduced to CS to synthesize sialylated CS (CS-SA), and the tyramine (TA) was grafted to gelatin (G) to obtain TA modified gelatin (G-TA). The successful synthesis of CS-SA and G-TA was confirmed using 1H NMR and UV–Vis absorption spectra. The interpenetrating polymer networks G-TA/CS-SA (GC) hydrogel was then fabricated via blending G-TA and CS-SA solutions and crosslinked using horseradish peroxidase. The storage modulus (G') of the fabricated GC hydrogels with different ratios of G-TA/CS-SA greatly varied during the formation and strain of hydrogels. With the increase of CS-SA concentration from 0% to 2%, the storage modulus of GC hydrogels was also observed to decrease from 1500 Pa to 101 Pa; the water uptake capacity of GC hydrogels increased from 1000% to 4500%. Additionally, the cell counting kit-8 and fluorescent images demonstrated the excellent cytocompatibility of GC hydrogels after culturing with NIH 3T3 cells. The obtained results indicated that the fabricated GC hydrogels might have potential in biomedical fields, such as wound dressing.

Loading and releasing behavior of carboxymethyl cellulose and chitosan complex beads

Ali A. Altam,Liping Zhu,Dafaalla Babiker,Hajo Yagoub,Shuguang Yang

The complex beads of carboxymethyl cellulose (CMC) and chitosan (CHI) that are fabricated by interfacial complexation have a core-shell structure composed of a thin skin layer and a 3D entangled internal structure, which endows the beads with adsorption possibilities toward anionic and cationic molecules via electrostatic interaction. However, the loading and releasing behavior of such a system is essential and fundamental for their application yet has not been studied systematically. In this work, the influences of beads physical state, adsorbent dosage, agitation speed, solution pH and temperature on the loading behavior were investigated. The CMC@CHI beads demonstrated different loading efficiency to three typical dyes of methylene blue (MB), acid blue-113 (AB), and methyl orange (MO) with different charge status. The adsorption rate followed an order of MB ˃ AB ˃ MO, giving 97.4%, 73.3% and 20.2%, respectively. The releasing of three dyes from CMC@CHI beads also exhibited different rates, and MB release was found to be prolonged compared to AB and MO. The loading mechanism of dyes was further investigated via X-ray photoelectron spectroscopy. These CMC@CHI beads may have possible applications in mixed dye removal as well as encapsulating systems of drugs or sensitive chemicals with different charges.

An injectable multifunctional thermo-sensitive chitosan-based hydrogel for periodontitis therapy

Shiqing Ma,Xuemei Lu,Xinying Yu,Yaqi Du,Shendan Xu,Minting Li,Cheng Peng,Zihao Liu,Jiayin Deng

Bacteria are recognized as the driving factors of periodontitis. However, excessive reactive oxygen species (ROS) can harm periodontal tissue while also causing an uncontrolled inflammatory response. Hence, eliminating excessive ROS and blocking ROS-induced abnormal inflammatory response by antioxidants are achieving remarkable results in periodontitis therapy. Moreover, influenced by the deep and irregular periodontal pockets, injectable thermo-sensitive chitosan-based hydrogels have attracted a lot of attention. This study aimed to formulate an antibacterial and antioxidant therapeutic regimen by incorporating antimicrobial peptides (Nal-P-113) and/or antioxidants (polydopamine nanoparticles, PDNPs) into chitosan-based hydrogels. The hydrogel was characterized in vitro and finally examined in rats using the experimental periodontitis model. The release kinetics showed that the hydrogel could stably release Nal-P-113 and PDNPs for up to 13 days. The scavenging activity of the hydrogel against DPPH was about 80 % and the antibacterial ratio against Streptococcus gordonii ( S. gordonii ), Fusobacterium nucleatum ( F. nucleatum ) and Porphyromonas gingivalis ( P. gingivalis ) was about 99 %. Importantly, it was examined that the hydrogel had the ability to prevent periodontal tissue damage. Thus, chitosan-based hydrogels may provide a basis for designing multifunctional local drug delivery biomaterials for the treatment of periodontitis.

A new chitosan-based thermosensitive nanoplatform for combined photothermal and chemotherapy

Yanyan Zhang,Gareth R. Williams,Jiadong Lou,Wanting Li,Cuiwei Bai,Tong Wang,Shiwei Niu,Chun Feng,Li-Min Zhu

Targeting the delivery of anti-cancer drugs to a tumor site is essential for effective treatment and to ensure minimal damage to healthy cells and tissues. In this work, a chitosan-based nanoplatform was constructed for combined photothermal therapy and chemotherapy of breast cancer. The pH-sensitive and biocompatible biopolymer chitosan (CS) was grafted with N -vinylcaprolactam (NVCL) and modified with biotin (Bio), imparting it with temperature sensitive property and also the ability for active targeting. The polymer self-assembled to give nanoparticles (NPs) loaded with indocyanine green (ICG) and doxorubicin (DOX). When the NPs are exposed to near-infrared (NIR) laser irradiation, ICG converts the light to heat, inducing a significant phase transition in the NPs and facilitating the release of the drug cargo. In addition, the solubility of chitosan is increased in the slightly acidic microenvironment of the tumor site, which also promotes drug release. A detailed analysis of the NPs both in vitro and in vivo showed that the carrier system is biocompatible, while the drug-loaded NPs are selectively taken up by cancer cells. Particularly when augmented with NIR irradiation, this leads to potent cell death in vitro and also in an in vivo murine xenograft model of breast cancer.

Preparation and performance of chitosan/cyclodextrin-g-glutamic acid thermosensitive hydrogel

Hui Yun Zhou,Jia Nan Tong,Li Jun Ren,Pei Yan Hao,Hui Jie Zheng,Xu Ming Guo,Ya Wei Chen,Jun Bo Li,Hyun Jin Park

Chitosan hydrogel had been widely used in cell culture and tissue engineering, but its long gel time, weak mechanical strength, and weak antibacterial performance limited its application. In this study, the cyclodextrin grafted glutamic acid , a water-soluble cyclodextrin derivatives , was prepared by glutamic acid (Glu) and mono-(6-OTs)-β-CD under alkaline conditions and confirmed by FTIR and 1 H NMR. Then chitosan (CS) and cyclodextrin grafted glutamic acid (CD-g-Glu) were used to prepare chitosan/cyclodextrin derivatives thermosensitive hydrogel (CS/CD-g-Glu) to improve the properties of chitosan-based thermosensitive hydrogels. The CS/CD-g-Glu hydrogel had temperature sensitivity and stable three-dimensional network structure. In addition, the hydrogel loaded with berberine hydrochloride (BBH) showed optimal sustained-release effect, and the cumulative release rate was 57% in 24 h. More importantly, the CS/CD-g-Glu hydrogel loaded with BBH showed prefect antibacterial activity against E. coli and S. aureus . Therefore, the CS/CD-g-Glu hydrogel has great promising applications in drug delivery, tissue engineering, biotechnological, and biomedical fields.

A keratin/chitosan sponge with excellent hemostatic performance for uncontrolled bleeding

Rong-Rong Yan,Dai Xue,Chang Su,Yan Xu,Jin-Song Gong,Yan-Ling Liu,Min Jiang,Yan Geng,Guo-Zhong Lv,Zheng-Hong Xu,Jin-Song Shi

Uncontrolled bleeding leads to a higher fatality rate in the situation of surgery, traffic accidents and warfare. Traditional hemostatic materials such as bandages are not ideal for uncontrolled or incompressible bleeding. Therefore, it is of great significance to develop a new medical biomaterial with excellent rapid hemostatic effect. Keratin is a natural, biocompatible and biodegradable protein which contains amino acid sequences that induce cell adhesion . As a potential biomedical material , keratin has been developed and paid attention in tissue engineering fields such as promoting wound healing and nerve repair . Herein, a keratin/chitosan (K/C) sponge was prepared to achieve rapid hemostasis . The characterizations of K/C sponge were investigated, including SEM, TGA , liquid absorption and porosity, showing that the high porosity up to 90.12 ± 2.17 % resulted in an excellent blood absorption. The cytotoxicity test and implantation experiment proved that the K/C sponge was biocompatible and biodegradable. Moreover, the prepared K/C sponge showed better hemostatic performance than chitosan sponge (CS) and the commercially available gelatin sponge in both rat tail amputation and liver trauma bleeding models. Further experiments showed that K/C sponge plays a hemostatic role through the endogenous coagulation pathway, thus shortening the activated partial thromboplastin time (APTT) effectively. Therefore, this study provided a K/C sponge which can be served as a promising biomedical hemostatic material.

Structural characteristics and rheological properties of hydroxypropyl trimethyl ammonium chloride chitosan

Ling Ding,Zhao Liu,Shen Song,Mohamed Aamer Abubaker,Lele Chen,Jipeng Shi,Zengjie Fan,Ji Zhang

Hydroxypropyl trimethyl ammonium chloride chitosan (HACC) was synthesized by reacting chitosan with glycidyl trimethylammonium chloride. Atomic force microscopy showed that HACC exhibited disorderly coils in dilute solution and formed a three-dimensional network. Flow, thixotropy , and dynamic viscoelasticity tests were conducted using an MCR301 rheometer. The HACC solution was a non-Newtonian pseudoplastic fluid, and the shear behavior of different concentrations (2–6 %, w/v) was evaluated by the Williamson model fitting. Furthermore, the thixotropy was highly dependent on concentration changes: the high-concentration solution structure was difficult to recover in a short time. The dynamic viscoelasticity test indicated that the viscoelasticity of the HACC solution not only exhibited a viscous behavior similar to that of a fluid, but also exhibited elastic properties of weak gel. HACC exhibited high-strength solid-like gel characteristics at high temperature.

Depletion stabilization of emulsions based on bacterial cellulose/carboxymethyl chitosan complexes

Yamei Zhang,Shujuan Yang,Haiyun Tang,Sihui Wan,Wenqi Qin,Qu Zeng,Junhao Huang,Gaobo Yu,Yuhong Feng,Jiacheng Li

The regulation of the magnitude of the depletion effect is necessary for accurately predicting and explaining the emulsion stabilization mechanism. Herein, the bacterial cellulose/carboxymethyl chitosan (BC/CCS) complexes with tunable assembled behaviors were prepared and designed via electrostatic interaction . Specially, the emulsions stabilized by BC/CCS complexes exhibited excellent stability as compared with that stabilized by BC polymers alone. At pH 9.6, BC/CCS complexes in the continuous phase induced long-range depletion-stabilization effect to stabilize emulsions. Additionally, the magnitude of depletion effect of BC/CCS complexes could be improved by increasing BC concentration, and effectively stabilized emulsions. Furthermore, with the decrease to pH 7.0, the interfacial adsorption layers at the oil-water interface prevented oil droplets from agglomerating, but did not show better emulsion stability. These results clarified that the magnitude of the depletion effect could be controlled by altering BC-based complexes particles, which would be useful for the applications of emulsions in numerous fields.
