Coconut milk allergenicity: Insight into reducing the affinity of coconut globulin to immunoglobulin E by atmospheric cold plasma

Yang Chen,Yile Chen,Tian Li,Jiamei Wang,Weimin Zhang

Atmospheric cold plasma (ACP) presents a promising method for the sterilization of coconut milk and exhibits a modifying effect on coconut globulin (CG), the primary allergen in coconut milk. This study investigated the potential role of ACP treatment in mitigating the allergenic properties of coconut milk by examining changes in protein structure. ACP treatment induced structural alterations in CG, disrupting binding sites with immunoglobulin E (IgE). Consequently, this led to a reduction in the affinity between CG and IgE, evidenced by a decrease in K a from 2.17 × 10 4 /M to 0.64 × 10 4 /M, thereby diminishing IgE-mediated allergic reactions. The findings from allergenic and cellular models further corroborated that ACP treatment decreased the allergenicity of CG by 55.18%, while inhibiting degranulation and the release of allergic mediators. This study presents an innovative methodology for producing hypoallergenic coconut milk, thereby expanding the applicability of ACP technology within the food industry.

Cucurbitacin E Alleviates Colonic Barrier Function Impairment and Inflammation Response and Improves Microbial Composition on Experimental Colitis Models

Fengxia Zhan,Wei Song,Yong Fan,Fangjian Wang,Qian Wang

Purpose Cucurbitacins, which are found in a variety of medicinal plants, vegetables and fruits, were known for their diverse pharmacological and biological activities, including anticancer, anti-oxidative and anti-inflammatory effects. Cucurbitacin E, one of the major cucurbitacins, was recently proved to inhibit inflammatory response. Methods To explore the therapeutic effects of cucurbitacin E on colitis and the underlying mechanisms, male mice drunk water containing 2.5% dextran sulfate sodium (DSS) to establish colitis model and administrated with cucurbitacin E during and after DSS treatment. The disease activity index was scored and colonic histological damage was observed. Intestinal tight junction and inflammatory response were determined. 16S rRNA and transcriptome sequencing were performed to analyze gut microbiota composition and gene expression, respectively. Results We found that cucurbitacin E alleviated DSS-induced body weight loss and impaired colonic morphology. Cucurbitacin E decreased the expression of inflammatory cytokines and cell apoptosis, and maintained barrier function. Additionally, cucurbitacin E retrieved DSS-induced alterations in the bacterial community composition. Furthermore, a variety of differentially expressed genes (DEGs) caused by cucurbitacin E were enriched in several pathways including the NFκB and TNF signaling pathways as well as in Th17 cell differentiation. There was a close relationship between DEGs and bacteria such as Escherichia-Shigella and Muribaculaceae. Conclusion Our results revealed that cucurbitacin E may exert protective effects on colitis via modulating inflammatory response, microbiota composition and host gene expression. Our study supports the therapeutic potential of cucurbitacin E in colitis and indicates that gut microbes are potentially therapeutic targets.

Coral bionic e-skin for motion monitoring and intelligent recognition of underwater communication commands assisted by deep learning

Yudong Song,Yang Zou,Qingquan Zhao,Xinjian Shi,Shengyan Yin,Zhenning Liu,Hang Sun

Continuous and dependable underwater motion monitoring and communication are crucial for ensuring the safety of swimming, divers and underwater workers. We have developed a coral-inspired piezoresistive electronic skin (Melamine sponge-poly(pyrrole)-polydimethylsiloxane, MS-ppy-PDMS e-skin) for motion monitoring and underwater communication. The microstructure of the coral-like poly(pyrrole) and the PDMS acting as “limestone” make MS-ppy-PDMS e-skin superhydrophobic, stable (up to 80,000 cycles and acid and alkali resistance), and sensitive over a wide pressure sensing range (1–2,000 kPa). Moreover, MS-ppy-PDMS e-skin exhibits remarkable breathability, photothermal antibacterial efficacy, and biocompatibility, thereby increasing the comfort of wear while maintaining its outstanding sensing functionality. MS-ppy-PDMS e-skin exhibited dependable electrical signals output in motion monitoring tests on land and underwater. Most crucially, the combination of underwater communication commands captured by MS-ppy-PDMS e-skin with a multilayer perceptron (MLP) neural network allows for rapid and precise analysis and recognition, achieving a 99.38% accuracy rate in identifying underwater communication commands. This enables the realization of intelligent perception function, and is expected to further advance the development of human brain-like cognitive systems. Overall, the coral bionic MS-ppy-PDMS e-skin demonstrates tremendous application potential in the fields of motion monitoring and underwater communication.

Shouyu Wu,Jiemei Ou,Shuyi Wang,Aiping Zheng,Fan Yang,Zidan Zhou,Xuanxuan Lu

The use of Pickering emulsions for biocatalysis is gaining increased attention. However, the extensive application is greatly limited due to the enzyme inactivation . Herein, a biocatalytic Pickering emulsion with high-performance utilizing cellulose nanocrystals immobilized lipases (CNCs-Lps) particles as stabilizer is advanced and applied for the synthesis of Vitamin E nicotinate . CNCs-Lps display high activity and reusability due to the construction of biocatalytic microreactor in the O/W emulsion system . The yield of vitamin E nicotinate ester reached up to 83 %. More importantly, the CNCs-Lps can be reused due to the similar principles to microreactors in Pickering emulsions . Reusability test showed that the CNCs-Lps could be recovered from the emulsion system by centrifugation and the yield of vitamin E nicotinate retains 78 % of initial value after five cycles, demonstrating overwhelming advantage than the fair counterpart with free lipases .

A Prototype of Graphene E-Nose for Exhaled Breath Detection and Label-Free Diagnosis of Helicobacter Pylori Infection

Xuemei Liu,Qiaofen Chen,Shiyuan Xu,Jiaying Wu,Jingwen Zhao,Zhengfu He,Aiwu Pan,Jianmin Wu

Helicobacter pylori (HP), a common microanaerobic bacteria that lives in the human mouth and stomach, is reported to infect ≈50% of the global population. The current diagnostic methods for HP are either invasive, time-consuming, or harmful. Therefore, a noninvasive and label-free HP diagnostic method needs to be developed urgently. Herein, reduced graphene oxide (rGO) is composited with different metal-based materials to construct a graphene-based electronic nose (e-nose), which exhibits excellent sensitivity and cross-reactive response to several gases in exhaled breath (EB). Principal component analysis (PCA) shows that four typical types of gases in EB can be well discriminated. Additionally, the potential of the e-nose in label-free detection of HP infection is demonstrated through the measurement and analysis of EB samples. Furthermore, a prototype of an e-nose device is designed and constructed for automatic EB detection and HP diagnosis. The accuracy of the prototype machine integrated with the graphene-based e-nose can reach 92% and 91% in the training and validation sets, respectively. These results demonstrate that the highly sensitive graphene-based e-nose has great potential for the label-free diagnosis of HP and may become a novel tool for non-invasive disease screening and diagnosis.

Suppression of photoreactivation of E. coli by excimer far-UV light (222 nm) via damage to multiple targets

Zi-Bo Jing,Wen-Long Wang,Yu-Jia Nong,Lu Peng,Zi-Chen Yang,Bei Ye,Min-Yong Lee,Qian-Yuan Wu

Low-pressure mercury lamps emitting at 254 nm (UV254) are used widely for disinfection. However, subsequent exposure to visible light results in photoreactivation of treated bacteria. This study employed a krypton chloride excimer lamp emitting at 222 nm (UV222) to inactivate E. coli . UV222 and UV254 treatment had similar E. coli -inactivation kinetics. Upon subsequent irradiation with visible light, E. coli inactivated by UV254 was reactivated from 2.71-log to 4.75-log, whereas E. coli inactivated by UV222 showed negligible photoreactivation. UV222 treatment irreversibly broke DNA strands in the bacterium, whereas UV254 treatment primarily formed nucleobase dimers. Additionally, UV222 treatment caused cell membrane damage, resulting in wizened, pitted cells and permeability changes. The damage to the cell membrane was mainly due to the photolysis of proteins and lipids by UV222. Furthermore, the photolysis of proteins by UV222 destroyed enzymes, which blocked photoreactivation and dark repair. The multiple damages can be further evidenced by 4.0–61.1 times higher quantum yield in the photolysis of nucleobases and amino acids for UV222 than UV254. This study demonstrates that UV222 treatment damages multiple sites in bacteria, leading to their inactivation. Employing UV222 treatment as an alternative to UV254 could be viable for inhibiting microorganism photoreactivation in water and wastewater.

Selectin E upregulation upon forkhead box P1 loss augments neutrophil infiltration and brain damage in hypertensive intracerebral hemorrhage

Naizhu Wang,Jingkun Wu,Zhaofei Song,Hongbin Wang,Yan Zhao

Hypertension in the brain may lead to hypertensive intracerebral hemorrhage (HICH), a devastating disease. This study, grounded on bioinformatics insights, aims to investigate the functions of Selectin E (SELE) and forkhead box P1 (FOXP1) in the progression of HICH. Increased SELE expression was detected in the striatum of a mouse model of HICH generated via angiotensin II and L-NAME treatments. Knockdown of SELE alleviated hemorrhage, neutrophil infiltration, and blood–brain barrier (BBB) rupture in the mouse brain. FOXP1, poorly expressed in the HICH mice, was found to repress SELE transcription by binding to its promoter region. Overexpression of FOXP1 resulted in analogous alleviating effects in the HICH mice; however, the effects were abrogated by the additional SELE overexpression. In vitro , human brain microvascular endothelial cells (HBMECs) were treated with thrombin to generate a cellular model of ICH, followed by co-culture with HL-60 cell-derived neutrophils. The FOXP1 overexpression reduced the adhesion of neutrophils, and it alleviated HBMEC apoptosis and permeability while enhancing angiogenesis. Still, these effects were counteracted by the SELE upregulation. In conclusion, this study demonstrates that SELE upregulation upon FOXP1 loss is associated with neutrophil infiltration and BBB rupture in HICH.

Vitamin E nanoemulsion enhances porcine sperm quality and reproductive performance via antioxidant enhancement and fatty acid metabolism regulation

Chaoyue Cao,Xue Qin,Yiwen Sha,Haoqi Xiao,Zhenheng Lai,Yi Zheng,Rui Cai,Weijun Pang

Vitamin E (VE) is widely used as an antioxidant in boar semen preservation. Its low water solubility and limited bioavailability, however, compromise its functional efficacy during storage. To address these limitations, we developed a lecithin-stabilized vitamin E nanoemulsion (NE) to enhance its dispersibility and antioxidant efficacy. Semen from Duroc boars was diluted in Beltsville thawing solution (BTS) containing either 0.6 mM NE, 4 mM free VE, or no additive, and stored at 17 °C. Compared to VE and control groups, NE significantly enhanced sperm motility, preserved the structural integrity of the plasma and acrosomal membranes, and maintained mitochondrial function, while reducing oxidative stress and DNA fragmentation over a 7d storage period ( P  < 0.01). Untargeted metabolomics revealed that NE supplementation regulated key pathways including unsaturated fatty acid metabolism and glutathione-related antioxidation. Artificial insemination (AI) trials involving 637 sows confirmed that NE-treated semen improved total and healthy piglet numbers per litter ( P  < 0.05), without adverse effects. These findings suggest that NE provides an effective and practical strategy for enhancing boar semen preservation and reproductive outcomes in porcine production.

Electrolyte-Dependent Stability of Sulfonated Lignin Phenolic Resin Nanoplastics in Aquatic Systems: e-DLVO Theory and Molecular Modeling

Dan Zhao,Shunkai Zhang,Weili Yang,Cheng Xue,Guanglite Shen,Wenhui Zhang,Huixia Feng,Nali Chen,Lin Tan

Graphical Through the calculation based on the e-DLVO theory, investigate the aggregation situation of sulfonated phenolic resin in diverse aquatic environments. Plastic waste is widely recognized as a global environmental threat. Nanoplastics (NPs), derived from the degradation of macroplastics and microplastics, pose potential risks to human health. The aggregation behavior of NPs in aquatic systems is primarily governed by electrolyte concentration and pH. In this study, the aggregation behavior of NPs, represented by sulfonated lignin phenolic resin (SLPFR), was systematically investigated in aqueous solutions containing a series of monovalent and divalent electrolytes. Aggregation kinetics experiments, adsorption studies, material characterization, and extended-Derjaguin–Landau–Verwey–Overbeek (e-DLVO) theoretical calculations demonstrated that electrostatic interaction is the dominant force governing the aggregation of SLPFR NPs. Different electrolyte types alter the surface charge of NPs, consequently increasing the electrostatic repulsion and steric hindrance between particles. These findings provide significant insights for better assessing the aggregation of NPs in diverse aquatic environments and their associated environmental impacts and risks.

Enteric Polymer Based on pH-Responsive Aliphatic Polycarbonate Functionalized with Vitamin E To Facilitate Oral Delivery of Tacrolimus

Menglin Wang,Jin Sun,Yinglei Zhai,He Lian,Cong Luo,Lin Li,Yuqian Du,Dong Zhang,Wenya Ding,Shuhong Qiu,Yuhai Liu,Longfa Kou,Xiangfei Han,Rongwu Xiang,Yongjun Wang,Zhonggui He

To improve the bioavailability of orally administered drugs, we synthesized a pH-sensitive polymer (poly(ethylene glycol)–poly(2-methyl-2-carboxyl-propylene carbonate)–vitamin E, mPEG–PCC–VE) attempting to integrate the advantages of enteric coating and P-glycoprotein (P-gp) inhibition. The aliphatic polycarbonate chain was functionalized with carboxyl groups and vitamin E via postpolymerization modification. Optimized by comparison and central composite design, mPEG113–PCC32–VE4 exhibited low critical micelle concentration of 1.7 × 10–6 mg/mL and high drug loading ability for tacrolimus (21.2% ± 2.7%, w/w). The pH-responsive profile was demonstrated by pH-dependent swelling and in vitro drug release. Less than 4.0% tacrolimus was released under simulated gastric fluid after 2.5 h, whereas an immediate release was observed under simulated intestinal fluid. The mPEG113–PCC32–VE4 micelles significantly increased the absorption of P-gp substrate tacrolimus in the whole intestine. The oral bioavailability of tacrolimus micelles was 6-fold higher than that of tacrolimus solution in rats. This enteric polymer therefore has the potential to become a useful nanoscale carrier for oral delivery of drugs.

Effect of Hyperbaric Oxygen Treatment on Lipid Metabolism and Neurovascular Microenvironment in an Apolipoprotein E Knockout Mouse Model

Guoying Dong,Yuxiao Liu,Huijun Liu,Chen Qiao,Xia Chen,Linxiao Wang

Background:Dyslipidemia during midlife represents a significant risk factor for neuropathological alterations associated with cognitive decline. Given the currently incurable nature of dementia, implementation of preventive strategies and early therapeutic interventions prior to disease progression are paramount. Emerging evidence suggests that hyperbaric oxygen (HBO) therapy exhibits neuroprotective properties in various neurological conditions. However, whether HBO treatment modulates lipid metabolism dysregulation and subsequent neurodegeneration remains unanswered. This investigation aimed to elucidate the therapeutic potential of HBO treatment in ameliorating cerebral dysfunction and metabolic perturbations using apolipoprotein E (ApoE)-deficient (ApoE-/-) mice.Methods:ApoE-/- mice received HBO treatment for 10 consecutive days, and then behavioral assessment tests were performed. Serum and brain tissue were collected to measure oxidative stress levels and inflammatory factors. Results:Compared with ApoE-/- group, cognitive declines was significantly reversed in mice of the ApoE-/-+HBO mice. The blood lipid profiles of ApoE-/- mice were also improved after HBO treatment, accompanied by a reduction in body weight. Moreover, HBO treatment was found to ameliorates neuronal injury and amyloid-β deposition in the hippocampus of ApoE-/- mice. Further studies have revealed that the benefits of HBO treatment occurred through the reduction of inflammatory factors and attenuation of oxidative stress. Conclusions:These findings indicate that HBO treatment effectively improves the intracerebral microenvironment of ApoE-/- mice, providing a novel regulatory mechanism of protection against dyslipidemia-associated brain deficits by HBO treatment.
