Grape seed extract (GSE) displays strong antioxidant activity, but its instability creates barriers to its applications. Herein, three HP-β-CD/GSE inclusion complexes with host–guest ratios of 1:0.5, 1:1, and 1:2 were successfully prepared by co-precipitation method to improve stability. Successful embedding of GSE in the HP-β-CD cavity was confirmed by fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM) analyses. The Autodock Tools 1.5.6 was used to simulate the three-dimensional supramolecular structure of the inclusion complex of 2-hydroxypropyl-β-cyclodextrin and grape seed extract (HP-β-CD/GSE) by molecular docking. The MALDI-TOF-MS technology and chemical database Pubchem, and structural database PDB were combined to reconstitute the three-dimensional structure of target protein. The binding mode of the HP-β-CD/GSE inclusion complex to target protein was studied at the molecular level, and the antioxidant ability of the resulting HP-β-CD/GSE inclusion complexes was investigated by measuring 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging. The effects of HP-β-CD/GSE on myofibrillar protein from lamb tripe were also investigated under oxidative conditions. The positions and interactions of the binding sites of HP-β-CD/GSE inclusion complexes and target protein receptors were simulated by molecular docking. The results showed that HP-β-CD/GSE inclusion complexes were successfully prepared, optimally at a molar ratio of 1:2. At low (5 μmol/g) to medium (105 μmol/g) concentrations, HP-β-CD/GSE inclusion complexes decreased the carbonyl content, hydrophobicity, and protein aggregation of myofibrillar protein from lamb tripe, and increased the sulphydryl content. Furthermore, high concentration (155 μmol/g) of HP-β-CD/GSE inclusion complexes promoted protein oxidation. Keywords:grape seed extract;inclusion complex;hydroxypropyl cyclodextrin;antioxidant;myofibrillar protein;oxidation

A hydrophilic and anti-fouling coating layer was constructed on a polyvinylidene fluoride (PVDF) microfiltration membrane by a novel surface modification method. The pristine membrane was firstly coated by (3-chloropropyl) trimethoxysilane/polyethyleneimine and tannic acid. Then, the metal ion was induced on the coating layer to coordinate with tannic acid and polyethyleneimine, forming a more stable and hydrophilic coating on the surface. The membrane’s surface morphology and chemical element analysis showed that the Tannic acid/ polyethyleneimine (TA/PEI) coating layer was denser and had more stability after the addition of metal ions, and this may be due to the coordination bond formed between the TA/PEI coating and metal ions. The results of the water contact angle and pure water flux measurements showed that the hydrophilicity and wettability of the modified membranes were improved obviously after introducing the metal ion layers. The anti-fouling performance and stability of the modified membrane were also characterized by the underwater oil contact angle (OCA), the separation efficiency, and the contact angle variation value for before and after the rinsing experiment. The modified membrane showed obvious stability and antifouling. Moreover, the retention rate of some composite membranes could reach 99.6%.Keywords:surface modification;metal ion;anti-fouling;retention rate

Background Synovial fluid components, especially lipids, can trigger oxidation of ultrahigh-molecular-weight polyethylene (UHMWPE) artificial joint components in vivo. The use of antioxidants such as vitamin E effectively diminishes the oxidative cascade by capturing free radicals and reducing the oxidation potential of UHMWPE implants. Using a thermo-oxidative aging method, we recently found that tea polyphenols can enhance the oxidation resistance of irradiated UHMWPE in comparison with commercial vitamin E. However, it is yet unknown whether tea polyphenols can reduce lipid-induced oxidation. Questions/purposes We explored whether tea polyphenol-stabilized UHMWPE would exhibit (1) lower squalene absorption; (2) stronger oxidation resistance; and (3) lower content of free radicals than vitamin E-stabilized UHMWPE under a physiologically-motivated in vitro accelerated-aging model. Methods Tea polyphenol (lipid-soluble epigallocatechin gallate [ ls EGCG]) and vitamin E were blended with UHMWPE powders followed by compression molding and electron beam irradiation at 100 and 150 kGy. Small cubes (n = 3, 60 mg, 4 × 4 × 4 mm 3 ) cut from the blocks were doped in squalene at 60°, 80°, 100°, and 120° C for 2 hours. Gravimetric change of the cubes after squalene immersion was measured to assess absorption. Thin films (n = 3, ∼60 μm) were also microtomed from the blocks and were doped at 120° C for 24 hours. Oxidation induction time (n = 3, 5 mg of material from the cubes) and incipient oxidation temperature (n = 3, thin films) were obtained to determine the oxidation stability. Signal intensity of the free radicals, obtained by electron spin resonance spectroscopy, was used to qualitatively rank the antioxidant ability of vitamin E and ls EGCG. Results Squalene absorption was comparable between ls EGCG/UHMWPE and vitamin E/UHMWPE at a given temperature and radiation dose. The oxidation induction time of 100 kGy-irradiated UHMWPE was increased with ls EGCG compared with vitamin E except at 120° C. For example, the oxidation induction time value of 100 kGy-irradiated ls EGCG/UHMWPE immersed at 60 o C was 25.3 minutes (24.2–27.8 minutes), which was 8.3 minutes longer than that of 100 kGy-irradiated vitamin E/UHMWPE which was 17.0 minutes (15.0–17.1 minutes) (p = 0.040). After squalene immersion at 120° C, the incipient oxidation temperature of 100 and 150 kGy irradiated ls EGCG/UHMWPE was 234° C (227–240° C) and 227° C (225–229° C), which was higher than vitamin E-stabilized counterparts with value of 217° C (214–229° C; p = 0.095) and 216° C (207–218° C; p = 0.040), respectively. The electron spin resonance signal of 150 kGy irradiated ls EGCG/UHMWPE was qualitatively weaker than that of 150 kGy irradiated vitamin E/UHMWPE. Conclusions ls EGCG-stabilized UHMWPE demonstrated higher oxidation resistance than vitamin E-stabilized UHMWPE after squalene immersion, likely because ls EGCG donates more protons to eliminate macroradicals than vitamin E. Clinical Relevance Our in vitro findings provide support that ls EGCG may be effective in protecting against oxidation that may be associated with synovial fluid-associated oxidation of highly crosslinked UHMWPE joint replacement components.

In this study, tea polyphenols (TPs) was first extracted from tea waste by superheated steam (SS) pretreated ultrasonic-assisted hydrothermal extraction (UAH). The optimized strategy presented extracts with the extraction yield up to 21.19% with a significantly higher antioxidant ability, compared with the one without SS pretreatment. Further investigation proved that the SS suppressed the polyphenol oxidase activity of the TPs extract. The ability to scavenge the free radicals were compared in mouse liver mitochondria. Mitochondrial swelling, mitochondrial membrane potential (MMP), cardiolipin peroxidation, and respiratory chain complex (RCC) I–V activities were also evaluated as the index of the mitochondrial oxidative damage. The study supports evidence that the TPs extract exhibited significant protection against oxidative damage on mitochondrial. Furthermore, the effect of TPs on antioxidant ability in zebrafish embryo was evaluated. After TPs pretreatment for 1 day, zebrafish embryos showed a significantly higher survival rate as well as heart rate when facing the oxidative stress. Practical applications Polyphenols from tea leaves have been viewed as an antioxidant additive in food, mainly due to the ability of scavenging free radicals and reactive oxygen species. The results of this study suggest that the SS pretreatment could be used as an efficient method to extract TPs from the tea waste for the prevention of oxidative damage in the mouse liver mitochondria and zebrafish embryos.

Improvement of activity and property in polyphenol oxidases (PPOs) by protein engineering may contribute to synthesis of theaflavin (TF1) in industrial processes. Here, the potential noncatalytic residues of PPO from Rosa chinensis (RcPPO) were selected by sequence alignment, structural analysis and surface residues prediction and the mutants were gained by semi-rational site-directed mutagenesis. Mutants L173F, A174K and N334I possessed 10.48, 9.74 and 7.13 times increases respectively in preference for diphenols over monophenols. Enzymatic activity, half-life at 40 and 50 °C of the most active mutant RcPPO-FKI increased to 2.67, 2.62 and 3.37 times respectively in comparison with wild type, implying that activity and thermostability of RcPPO through mutation were improved. The improvement of activity and thermostability in the mutant should be ascribed to more hydrogen bonds formed by the conserved catalytic histidine residues with substrate, reduced fluctuation of the loop regions and increased hydrogen network of the mutational amino acids, revealed by molecular dynamics simutation and molecular docking. Moreover, the yield of TF1 was improved by catalysis of mutant RcPPO-FKI with higher synthetic efficiency of TF1 in comparison with some reported catalysts and as a promising alternative to commercial mushroom tyrosinase. PPOs with enhanced performances provide the robust candidate for industrial application.

Cryogel-templated oleogels (CTO) were fabricated via a facile polyphenol crosslinking strategy, where apple polyphenol was utilized to crosslink the gelatin/egg white protein conjugates without forming hydrogels. After freeze-drying, cryogel templates were obtained and used to construct CTO by oil absorption. Apple polyphenol crosslinking improved the emulsion-related properties with appearance changes on samples, and infrared spectroscopy further confirmed the interactions between proteins and apple polyphenol. The crosslinked cryogels presented porous microstructures (porosity of over 96 %), enhanced thermal/mechanical stabilities, and could absorb a high content of oil (14.41 g/g) with a considerable oil holding capacity (90.98 %). Apple polyphenol crosslinking also influenced the rheological performances of CTO, where the highly crosslinked samples owned the best thixotropic recovery of 85.88 %. Moreover, after the rapid oxidation of oleogels, the generation of oxidation products was effectively inhibited by crosslinking (POV: 0.48 nmol/g, and TBARS: 0.53 mg/L). The polyphenol crosslinking strategy successfully involved egg white protein and gelatin to fabricate CTO with desired physical/chemical properties. Apple polyphenol acted as both a crosslinker and an antioxidant, which provided a good reference for fabricating pure protein-based CTO.

The emergence of antibiotic resistance represents a significant and growing threat to global public health. The development of novel and antibiotic-free antibacterial strategies is therefore highly importance. In this context, chemodynamic therapy (CDT), which is based on Fenton or Fenton-like chemistry, is a promising strategy. CDT utilizes Fe 2+ to convert H 2 O 2 into highly reactive hydroxyl radicals (•OH), which can oxidize biomolecules to achieve antibacterial effects. The antibacterial efficacy of CDT is contingent upon both the catalyst and substrate. We developed a straightforward, convenient and economical approach to fabricate Fe-caffeic acid metal–polyphenol networks (Fe-CA MPNs) by mixing CA and Fe 3+ . The Fe-CA MPNs are acid-sensitive and self-enhanced CDT agents with remarkable antibacterial efficacy against both Staphylococcus aureus ( S. aureus ) and methicillin-resistant S. aureus (MRSA). Antibacterial efficacy was evaluated via a variety of methods, including the zone of inhibition, spread plate, turbidimetric, and live/dead bacterial staining tests. Specifically, the inhibition rates of Fe-CA MPNs for S. aureus and MRSA were 96.33 ± 0.33 % and 96.33 ± 0.15 %, respectively. CaO 2 @Fe-CA nanocomposites with H 2 O 2 self-supply properties were synthesized by depositing Fe-CA MPNs on the surface of CaO 2 nanoparticles which function as a sustained-release source of H 2 O 2 . The antibacterial effect of the CaO 2 @Fe-CA nanocomposites was comparable to that of Fe-CA MPNs combined with H 2 O 2 . Both Fe-CA MPNs and CaO 2 @Fe-CA possess favorable in vitro biosafety profiles. This work will facilitate the development of innovative antibiotic-free CDT antibacterial strategies based on •OH.

The effects of defatting and dephenolization treatments on the structural, physico-chemical and technological properties of passion fruit epicarp flour (PFEF) were evaluated in this research. The FT-IR, SEM and XRD results illustrated that defatted and defree phenolics did not change the basic chemical structure of PFEF, while debound phenolics treatment hydrolyzed the ester bonds between dietary fiber and polyphenols and hydrolyzed the amorphous region of dietary fiber, resulting in self-assembly into a larger aggregate with a compact texture. All three kinds of treatment could improve the flowability of passion fruit epicarp flour to some extent. In addition, dephenolization treatment brought about an obvious change in the technological properties of PFEF, and removal of free polyphenols or bound polyphenols contributed to two completely different effects on partial indicators. These properties may be important for their value-added utilization.

Wound infections present a significant clinical challenge, as the antibiotics commonly utilized in treating infected wounds confront the pressing issue of bacterial resistance. Consequently, it is imperative to develop antibacterial materials that can diminish reliance on antibiotics for effective management of such infections. In this study, we introduce a novel metal-polyphenol nanoplatform (designated as ICG@Fe-Qu) that synergistically integrates photothermal therapy (PTT), photodynamic therapy (PDT), chemodynamic therapy (CDT), and ferroptosis for the treatment of infected wounds. ICG@Fe-Qu is constructed through one-step assembly of quercetin (Qu), Fe 2+ and indocyanine green (ICG) in an aqueous solution. Upon exposure to near-infrared (NIR) laser irradiation, ICG@Fe-Qu generates localized heat for PTT, which disrupts bacterial membranes and facilitates iron ion penetration into bacterial interiors. Concurrently, ICG produces reactive oxygen species (ROS) under NIR irradiation for PDT applications. Combined with Fe-mediated Fenton reaction to generate hydroxyl radicals for CDT, substantial ROS is generated to induce lipid peroxidation ultimately leading to bacterial ferroptosis. Importantly, both in vivo and in vitro experiments demonstrate robust bactericidal activity of ICG@Fe-Qu against Staphylococcus aureus and Escherichia coli , along with efficient eradication of bacterial biofilms. Furthermore, ICG@Fe-Qu enhances fibroblast migration and accelerates wound healing processes, indicating its promising potential for treating infected wounds in clinics.

The internal water migration within calcium silicate hydrate (C-S-H) in dry environments is considered to be the primary factor affecting the volume stability of cementitious materials. In this research, the in-situ polymerization product of plant polyphenol tannic acid (TA) was applied to modify C-S-H based on an organic-inorganic composite modification method. The chemical structure, microstructure, composition, dimensional changes and water migration characteristics of modified C-S-H were analyzed. Experimental results showed that TA improved the polymerization degree of siloxane chains in the C-S-H nanostructure, with a maximum improvement of 80.70%, and increased the interlayer spacing in the C-S-H structure, confirming the modification of C-S-H at the nanoscale, exhibited by 29 Si Nuclear Magnetic Resonance (NMR), X-ray diffraction (XRD) and Transmission Electron Microscopy (TEM). This modification by TA optimized the C-S-H nanostructure and microscopic pores, and increased the interlayer and gel pore water content, especially gel pore water increased by 56.53% compared to that of the unmodified C-S-H, revealed by nitrogen adsorption and 1 H LF-NMR. In the dry environment, the blocking and cladding effects of TA on C-S-H effectively inhibited the gel water loss and reduced the drying shrinkage, especially at low TA concentration. This research aims to improve the volume stability of C-S-H by in-situ polymerization product of plant polyphenol, which provides new insights into improving the volume stability of cementitious materials.

Sonodynamic therapy (SDT) is an efficient non-invasive strategy for treating breast cancer. However, the therapeutic efficacy of SDT is greatly limited by various defense mechanisms in the tumor microenvironment, particularly the overexpression of B-cell lymphoma-2 (Bcl-2). In this study, based on drug self-delivery systems, a carrier-free ultrasound-responsive polyphenol nanonetwork (GTC) was developed to enhance SDT by inhibiting Bcl-2. A one-pot method, involving the interaction of the polyphenolic Bcl-2 inhibitor gossypol (GOS), transferrin, and the sonosensitizer chlorin e6 (Ce6), was used to synthesize the GTC. The GTC was efficiently internalized by MDA-MB-231 and 4T1 cells through specific binding to transferrin receptors, and no external carriers were needed. After cellular internalization, GOS increased the lethality of Ce6-mediated SDT by reducing the expression of the Bcl-2 protein, which caused multiple toxic effects. RNA-seq analysis confirmed the transcriptomic alterations in oxidative stress and apoptotic pathways induced by the GTC nanosystem. In vivo studies revealed that GOS-assisted SDT not only eliminated tumors through sonodynamic effects and triggered immunogenic cell death but also enhanced sono-immunotherapy, thus effectively suppressing distant tumors and metastasis. This study might provide insights into carrier-free nanomedicines for SDT-based synergistic tumor therapy.

This study aims to enhance gelatin film performance in the food industry by incorporating polyphenol–titanium complexes (PTCs) as crosslinkers. PTCs introduce multiple linkages with gelatin, including coordination and hydrogen bonds, resulting in synergistic crosslinking effects. This leads to an increased hydrodynamic volume, particle size, and thermal stability of the gelatin films. Compared to films crosslinked solely by polyphenols or titanium, PTC-crosslinked gelatin films exhibit significant improvements. They show enhanced mechanical properties with a tensile strength that is 1.7 to 2.6 times higher than neat gelatin films. Moreover, these films effectively shield UV light (from 82% to 99%), providing better protection for light-sensitive food ingredients and preserving lutein content (from 74.2% to 78.1%) under light exposure. The incorporation of PTCs also improves film hydrophobicity, as indicated by water contact angles ranging from 115.3° to 131.9° and a water solubility ranging from 31.5% to 33.6%. Additionally, PTC-enhanced films demonstrate a superior antioxidant ability, with a prolonged polyphenol release (up to 18 days in immersed water) and a higher free radical scavenging ability (from 22% to 25.2%). Overall, the improved characteristics of gelatin films enabled by PTCs enhance their performance, making them suitable for various food packaging applications. Keywords:gelatin film;polyphenol–titanium complexes;synergistic effect
