Parkinson's disease (PD) is a neurodegenerative disorder with a lack of effective treatment options. mangiferin, a bioactive compound derived from mango, has been shown to possess strong neuroprotective properties. In this study, we investigated the neuroprotective effects of mangiferin on PD and its underlying mechanisms using both in vitro and in vivo models of 6-OHDA-induced PD. Additionally, we conducted molecular docking experiments to evaluate the interaction between mangiferin and AKR1C3 and β-catenin. Our results demonstrated that treatment with mangiferin significantly attenuated 6-OHDA-induced cell damage in PC12 cells, reducing intracellular oxidative stress, improving mitochondrial membrane potential, and restoring the expression of tyrosine hydroxylase (TH), a characteristic protein of dopaminergic neurons. Furthermore, mangiferin reduced the accumulation of α-synuclein and inhibited the expression of AKR1C3, thereby activating the Wnt/β-catenin signaling pathway. In vivo studies revealed that mangiferin improved motor dysfunction in 6-OHDA-induced PD mice. Molecular docking analysis confirmed the interaction between mangiferin and AKR1C3 and β-catenin. These findings indicate that mangiferin exerts significant neuroprotective effects in 6-OHDA-induced PD by inhibiting AKR1C3 and activating the Wnt/β-catenin signaling pathway. Therefore, mangiferin may emerge as an innovative therapeutic strategy in the comprehensive treatment regimen of PD patients, providing them with better clinical outcomes and quality of life.

In this Article, we present a facile microwave-assisted synthesis route for the preparation of water-soluble and high-quality CuInS2/ZnS nanocrystals (NCs) with glutathione as the stabilizer. The as-prepared CuInS2/ZnS NCs exhibited small particle sizes (∼3.3 nm), long photoluminescence lifetimes, and color-tunable properties ranging from the visible to the near-infrared by varying the initial ratio of Cu/In in the precursors. The low-toxicity, highly luminescent and biocompatible CuInS2/ZnS NCs were applied to cell imaging, showing that they could be used as promising fluorescent probes. Furthermore, the CuInS2/ZnS NCs were used as the signal labels for a fluoroimmunoassay of the biomarker IL-6, showing their great potential for use as reliable point-of-care diagnostics for biomarkers of cancer and other diseases.

Herein, a novel electrochemical biosensor has been proposed for highly sensitive and simultaneous determination of ascorbic acid (AA), dopamine (DA), and uric acid (UA) based on 4-sulfocalix[6]arene–functionalized carbon nanotubes (CNTs/SCX6) decorated with AuPt bimetallic nanoparticles. Ultraviolet–visible (UV–Vis) spectrophotometry, transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS) mapping, X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) have been applied to characterize the morphology and structure of CNTs/SCX6-AuPt. Cyclic voltammetry (CV), differential pulse voltammetry (DPV), and amperometric i–t curve reveal that CNTs/SCX6-AuPt–modified glassy carbon electrode (GCE) possesses excellent electrooxidation activity toward AA, DA, and UA. The obtained detection ranges of CNTs/SCX6-AuPt for simultaneous detection of AA, DA, and UA are 0.1–2 mM, 1.5–20 μM, and 2–40 μM with corresponding detection limits of 5.4 μM, 0.08 μM, and 0.37 μM, respectively. The high electrocatalytic activity and rapid electronic transfer ability of CNTs/SCX6-AuPt result in high selectivity, stability, reproducibility, and repeatability. The constructed electrochemical sensor has been used to simultaneously determine AA, DA, and UA in urine samples with good recoveries, which may become a promising method for trace analysis of biological molecules in the complex physiological environment.

Sensitive and accurate detection of interleukin 6 (IL-6) is crucial for the early diagnosis of cerebral infarction to improve patient survival rates. However, the low-abundance of IL-6 in cerebral infarction presents a significant challenge in developing effective diagnosis method. Herein, we studied and analyzed the strong fluorescence property of 4-aminophenol phosphate (APP) and developed an enzyme-linked immunosorbent assay (ELISA) for IL-6 detection. The detection was based on the integration of optical signal change induced by alkaline phosphatase (ALP)-catalyzed APP hydrolysis and ALP-mediated ELISA. The generated colorimetric signal of 4-aminophenol, APP hydrolysis product, was used for ELISA of IL-6 with a detection limit of 0.1 ng/mL, and the visual detection of IL-6 was achieved. The changes in APP fluorescence have a good linear relationship with the logarithm of IL-6 concentration in the range of 0.005 ng/mL to 5.0 ng/mL, with a detection limit of 0.001 ng/mL, which was 100 times lower than that of conventional pNPP-based ELISA. Furthermore, the constructed ELISA effectively distinguished between samples from patients with cerebral infarction and volunteers with non-cerebral infarction, and the severity of symptoms was well distinguished based on IL-6 measurement. The dual-mode ELISA demonstrated high feasibility of low-abundance biomarker detection and displayed good potential for accurate in vitro diagnosis.

The constitutive activation of signal transducer and activator of transcription 3(STAT3) is associated with aggressive development and metastasis in colorectal cancer (CRC), but STAT3-targeting drugs remain elusive in clinic. Here, structure-based strategy was used to remodel the natural compound cryptotanshinone into a more effective STAT3 inhibitor LYW-6. Using the Biolayer Interferometry assay, we observed that LYW-6 exhibited specific interactions with STAT3(KD = 6.6 ± 0.7 μM). Western blot analysis and electrophoretic mobility shift assays (EMSA) showed that LYW-6 inhibited the phosphorylation of STAT3 tyrosine 705 (Tyr-705) and had slight effects on STAT1 and STAT5 phosphorylation. Western blot analysis on the upstream kinases of STAT3 confirmed that the inhibitory mechanism on p-STAT3 was independent of upstream kinases. Further investigation demonstrated that LYW-6 downregulated the expression of downstream oncogenes to inhibit cell viability, cell cycle development, and potently increased cell apoptosis in human CRC cells. The invasion and metastasis linked signaling was also blocked by LYW-6 treatment. LYW-6 was found to reduce the metastasis foci in lung on tail-lung metastasis models. In addition, it was observed that LYW-6 markedly diminished STAT3 phosphorylation in tumor tissue and significantly inhibited tumor growth on xenograft models. Tumor development on chemically-induced colorectal cancer model also significantly inhibited by LYW-6 treatment. These findings provided adequate evidence that STAT3 inhibitor LYW-6 might be a potential candidate agent for CRC treatment.

An integrated multi-functional additive was fabricated by successively grafting melamine (MEL) and phytic acid (PhA) on multiwalled carbon-nanotubes (MWNCTs), and was then applied in PA6 to improve the flame retardancy and light aging resistance of the composite. The limit oxygen index of PA6 composite containing 7 wt% PhA-MEL-MWCNTs was increased to 26.4 from 21.0. The smoke and CO release were significantly reduced by 48% and 88% respectively, and the severe melt dripping of PA6 in burning was eliminated. It is proved that PhA-MEL-MWCNTs can absorb ultraviolet (UV) radiation, and hence significantly reduces the mechanical property loss of the PA6 composite after UV aging. The tensile strength of the aged PA6/7 wt%PhA-MEL-MWCNTs composite sample only decreased by 18.1%, which was significantly lower than the loss rate of the control aged PA6 sample (62.5%). This protocol provides a new opportunity for fabricating long-life flame retardant polyamide composites.

6-Gingerol, an active herbal ingredient of ginger has various bioactivities such as anti-neurodegenerative disease, anti-inflammatory and so on. The aim of the present study was to enhance the oral bioavailability and brain distribution of 6-Gingerol via polymeric micelles. A polymeric micelles drug delivery system of 6-Gingerol consisting of D-α-Tocopheryl polyethylene glycol 1000 succinate (TPGS) and Poly (ethylene glycol)-poly (ε-caprolactone) (PEG-PCL) was prepared via solvent injection method. The developed 6-Gingerol-loaded TPGS/PEG-PCL micelles (6-GTPMs) were characterized based on particle size, polydispersity index (PDI), zeta potential, encapsulation efficiency (EE), drug loading (DL) and in vitro release profile. The pharmacokinetics and tissue distribution studies were also evaluated. The nanoformulation produced a particle size of 73.24 ± 2.84 nm with acceptable PDI (0.129 ± 0.03), zeta potential (−2.74 ± 0.92 mV), DL (4.64%) and EE (79.68%). The in vitro release profile showed that the 6-GTPMs enhanced the solubility of 6-Gingerol, while the pharmaceutical analysis in rats indicated that 6-GTPMs significantly improved the oral bioavailability of 6-Gingerol (about 3 folds) in circulation. The 6-GTPMs exhibited remarkable brain targetability in the tissue distribution analysis. Collectively, a 6-Gingerol polymeric micelle with enhanced oral bioavailability coupled with excellent brain distribution was successfully developed.

Abstract ZrO 2 ceramic fibers have gained widespread study for the high temperature stability, low thermal conductivity and high mechanical properties. While for oxide fibers, grain growth is one of the limiting factors for long-term high temperature applications. In the present study, polycrystalline ZrO 2 –6 mol % Y 2 O 3 ceramic fibers with the addition of ~0–5 mol % La 2 O 3 to refine the grain size were fabricated by the dry-spinning method. The XRD and Raman spectral results showed that the pyrochlore La 2 Zr 2 O 7 phase was formed as a second phase in the tetragonal zirconia fiber matrix. The formation of a second phase was an effective way to hinder grain growth for the competition phase effect and solid solution mechanism. With the addition of La 2 O 3 , the grain size decreased and concentrated in a smaller range of 50–250 nm compared with the ZrO 2 –6 mol % Y 2 O 3 matrix at 1400°C. Furthermore, the thermal conductivity of the fibers decreased with the additive of La 2 O 3 less than 3 mol %. The present system is a promising candidate for high temperature applications.

An improved ion chromatographic method including two elution procedures was proposed for the quantitative determination of atmospheric alkylamines in field atmospheric samples involving high levels of inorganic cations by using 18-crown-6 as mobile phase additive. When 18-crown-6 was added to the mobile phase, the retention times increased significantly for Na + , NH 4 + , K + and primary alkylamines but decreased for secondary and tertiary alkylamines due to the complexation with certain cations and interaction with both stationary and mobile phases of 18-crown-6. As a result, the separation of the cations was greatly promoted, which reduced the interference of peak distortion of overloaded inorganic cations on the quantitation of adjacent alkylamines. By using the presented method, five inorganic cations (Na + , NH 4 + , K + , Mg 2+ , Ca 2+ ) and six alkylamines (dimethylamine (DMAH + ), trimethylamine + diethylamine (TMAH + + DEAH + ), propylamine (MPAH + ), triethylamine (TEAH + ), ethanolamine (MEOAH + ) and triethanolamine (TEOAH + )) were effectively separated and determined, and the relative standard derivations (RSDs) of objective cations were all less than 1% for retention time and 3.1% for peak area (n = 9), respectively. The linearity was excellent for each cation (R 2 > 0.993) except for NH 4 + and TEOAH + showing a non-linear response (R 2 > 0.998 for theoretical non-linear fitting), and the detection limit of these cations were 0.03–1.19 ng. The proposed method was successfully used in the determination of both alkylamines and inorganic cations in ambient particulate matters and gaseous alkylamines in ceiling duct exhaust. The annual average concentrations of DMAH + , TMAH + + DEAH + and TEAH + were 15.56, 4.35 and 16.00 ng m −3 in PM 2.5 over Shanghai in 2013. The concentrations of gaseous DMA and TMA + DEA in ceiling duct exhaust reached a maximum of 940.0 and 112.7 μg m −3 , and were positively correlated with the human activity intensity, suggesting that human excreta emissions was a potential important source of atmospheric alkylamines in urban area of Shanghai.

The modification of 2-amino-6-chloropurine on polyamidoamine was performed to synthesize a derivative, AP-PAMAM, which was then employed as a carrier for p53 gene delivery to achieve anti-tumor efficacy. Using a human prostate tumor cell line PC-3 as a model, enhanced expression level of p53 gene could be detected by RT-PCR, qPCR and western blotting. Based on the successful p53 gene transfection, an obvious anti-proliferative effect could be obtained, which was identified to be associated with the simultaneous induction of cell apoptosis and cell cycle arrest at the G1 phase. The cell apoptosis was elucidated to be triggered by the activation of the mitochondria-dependent apoptotic signaling pathway. In addition, AP-PAMAM/p53 transfection could restrain the migration of tumor cells as demonstrated by wound healing and Transwell migration assays. Notably, AP-PAMAM-mediated p53 delivery exhibited a stronger inhibitory effect on both cell proliferation and migration than the PAMAM/p53 group. In summary, AP-PAMAM/p53 transfection provided a promising route to achieve the p53-based gene therapy.

In this paper, nitrogenous bases adenine (A), guanine (G), cytosine (C), and uracil (U), were used to tailor the hydrogen-bonded network of melamine cyanurate (MCA) and were noted as A-MCA, G-MCA, C-MCA, U-MCA, respectively. The results showed that the size, morphology and thermal behavior of MCA are regulated by base due to the formation of hydrogen bonds between them. These modified MCAs were used to improve the flame retardancy of polyamide 6 (PA6) which was evaluated by using the limiting oxygen index (LOI) and the vertical burning (UL-94) test, etc. The results showed that the PA6 sample containing 9 wt% C-MCA achieves the UL-94 V-0 rating and has a LOI value of 30.7%, while the other samples with equal amounts of MCA only pass the UL-94 V-2 rating. The flame retardant mechanism was studied, and the results reveal that C-MCA with smaller size has greater contact area with PA6 which helps to catalyze the decomposition of PA6; Thus the PA6/C-MCA releases more NH 3 , CO 2 etc., and produces more droplets than others, which dilutes combustible gas and takes away heat to make the samples extinguish.

To discover novel natural-product-based pesticidal agents, we prepared a series of oxime sulfonate derivatives of 2′(2′,6′)-(Di)chloropicropodophyllotoxins by structural modification of podophyllotoxin. Their structures were well-characterized by proton nuclear magnetic resonance (1H NMR), high-resolution mass spectrometry (HRMS), optical rotation, and melting point. Moreover, the key steric structure of compound 5f was unambiguously determined by single-crystal X-ray diffraction. Additionally, their insecticidal activity was evaluated at 1 mg/mL against the pre-third-instar larvae of oriental armyworm (Mythimna separata Walker), a typical lepidopteran pest. Among all derivatives, compounds 4c, 5c, and 5d exhibited more promising insecticidal activity, with the final mortality rates greater than 60%, when compared to their precursor podophyllotoxin and the positive control, toosendanin. It demonstrated that introduction of the chlorine atom at the C-2′ or C-2′,6′ position on the E ring of picropodophyllotoxin or oxime sulfonate derivatives of picropodophyllotoxin was important for the insecticidal activity and introduction of a halogen (e.g., fluorine, chlorine, or bromine) atom-substituted phenylsulfonyl group on the oxime fragment of 2′(2′,6′)-(di)chloropicropodophyllones could lead to more promising compounds.
