Highly sensitive sensing and quantitative detection of sulfate ion with a SERS chip-based on boric acid’s Lewis effect

Gao Yueying,Wan Yuqi,Liu Lin,Liu Jinghan,Li Jingkun,Li Yu,Ozaki Yukihiro,Pi Fuwei

Based on the Lewis acid’s coordination principle, a surface-enhanced Raman scattering (SERS) chip strategy had been developed for the ultrasensitive quantitation of SO 4 2− . Through the immobilization of silver nanoparticles (Ag NPs) and the construction of the boric acid-based sensing unit, the chip system displayed outstanding merits on the direct sensing of SO 4 2− , e.g., simple operation, ultra-high sensitivity, reproducibility, excellent selectivity and specificity. Moreover, an accurate evaluation was obtained by ratiometric calculations on characteristic peaks (1382 and 1070 cm −1 ) for quantitative detection of SO 4 2− . The detection limit was down to 10 nM. Tap water, beer, and mineral water samples were tested, and high recoveries were achieved (97.12–110.12%). Besides, such SERS chip also displayed strong applicability for the evaluation of SO 3 2− . Therefore, this SERS chip provided a promising idea for the quantification of trace amounts of SO 4 2− and SO 3 2− in the fields of food safety and environmental monitoring. Graphical abstract

Flexible fabrication of lipophilic-hydrophilic micromotors by off-chip photopolymerization of three-phase immiscible flow induced Janus droplet templates

Kailiang Zhang,Yukun Ren,Tianyi Jiang,Hongyuan Jiang

Self-propelled microparticles are promising for lots of applications ranging from analytical detection to water treatment . Herein, we present an effective approach to fabricate lipophilic-hydrophilic micromotors via the photocuring of three-phase immiscible flow induced droplet templates. In the microfluidic system, two immiscible inner fluids, the lipophilic 1, 6-Hexanediol diacrylate (HDDA), and the hydrophilic poly (ethylene glycol) diacrylate (PEGDA), are simultaneously injected into a theta-shaped cylindrical capillary from two separate inlets, and they are emulsified into Janus drops when encountering the outer immiscible silicone oil. Because of the immiscible feature of droplet templates, off-chip photopolymerization strategy has been used, which can significantly decrease the blocking chance of microdevice. And also, the lipophilic-hydrophilic structure of droplets is convenient for the loading of cargos with different characteristics. More importantly, the size and configuration of droplet templates can be flexibly regulated by changing the flow rates of three different phases. Accordingly, multifunctional micromotors can be fabricated by adding different nanoparticles and materials into the HDDA or PEGDA phase first and then photocuring the droplets. Taking the bubble-propelled micromotors for example, we prepare microswimmers by loading Ag, TiO 2 and Fe 3 O 4 nanoparticles into the PEGDA phase. The swimming behaviors of micromotors in H 2 O 2 solution are systematically investigated, finding that the proportion of PEGDA phase and the concentration of H 2 O 2 both positively affect the moving speed. Furthermore, the applicability of motor particles on water treatment is successfully demonstrated by using neutral red solution as the model pollutant. And the micromotors can be recycled using magnets after the catalytic degradation process. Therefore, this micromotor generation technique and this kind of micromotor can be attractive for many applications.

A reversible valve-assisted chip coupling with integrated sample treatment and CRISPR/Cas12a for visual detection of Vibrio parahaemolyticus

Hui Wu,Yanju Chen,Qunqing Yang,Cheng Peng,Xiaofu Wang,Mengyao Zhang,Siwenjie Qian,Junfeng Xu,Jian Wu

Vibrio parahaemolyticus ( V. parahaemolyticus ) is regarded as a major cause of seafood-associated illnesses, which has aroused widespread public concern. Here, a rapid and convenient detection method for V. parahaemolyticus detection was established by a reversible valve-assisted chip coupling with CRISPR/Cas12a. With optimized lysis buffer, loop mediated isothermal amplification (LAMP) reagents and CRISPR reagents, the whole detection process from sampling to results could be finished within 50 min. The structure of chip was simple and the cost was low. By relying on three reversible rotary valves and the rotation direction-dependent Coriolis pseudo force, the flow order of liquid and the direction of liquid flow could be precisely controlled. The LAMP amplicons were specifically and sensitively identified by CRISPR/Cas12a. Positive amplification would produce green fluorescent signal while negative amplification generated no fluorescent signal, which could be clearly distinguished by the naked eye. With 600 μL of samples processed, the limit of detection (LOD) for both pure cultured V. parahaemolyticus or spiked shrimp samples could achieve 30 copies/reaction. These illustrated the established method displayed great feasibility for real samples detection. In the future, the chip could also combine with other amplification reactions, like PCR or recombinase polymerase amplification reaction (RPA), to conduct detection by changing the corresponding lyophilized amplification reagents. Overall, the proposed detection platform displays great potential for food safety analysis and clinical diagnostics, especially in resource-limited areas.

Ti3C2Tx MXene-Loaded 3D Substrate toward On-Chip Multi-Gas Sensing with Surface-Enhanced Raman Spectroscopy (SERS) Barcode Readout

Kuo Yang,Kai Zhu,Yuanzhe Wang,Ziting Qian,Yizhi Zhang,Zhaoyan Yang,Zhuyuan Wang,Lei Wu,Shenfei Zong,Yiping Cui

Gas sensors lie at the heart of various fields ranging from medical to environmental sciences, and the demand of gas sensors is instantly expanding. However, in the face of complex gas samples, how to maintain high sensitivity while performing multiplex detection still puzzles the researchers. Here, by introducing Ti3C2Tx MXene into a microfluidic gas sensor with a three-dimensional (3D) transferable SERS substrate, a powerful gas sensor having both multiplex detecting ability and high sensitivity is demonstrated. The employ of MXene endows the sensor with a universal high adsorption efficiency for various gases while the generation of in situ gas vortices in the sophisticated nanomicro structure extends the molecule residence time in SERS-active area, both leading to the increased sensitivity. In the proof-of-concept experiment, a limit of detection (LOD) of 10–50 ppb was achieved for three typical volatile organic compounds (VOCs) according to the intrinsic SERS signals of gas molecules. Besides, the well-designed periodic 3D structure solves the general repeatability problem of SERS substrates. In addition, the detailed composition of gas mixture was revealed using classic least-square analysis (CLS) with an average accuracy of 90.6%. Further, a chromatic barcode was developed based on the results of CLS to read out the complex composition of samples visually.

Self-assembly of super-hydrophobic PMMA microspheres on silicon wafer as MALDI-MS chip for rapid quantification of peptides

Shuzhen Dou,Ning Li,Xueyun Wang,Qunyan Zhu,Zhongshun Wang,Nan Lu

Rapid quantification of peptides by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is a challenge. Herein, we prepared a super-hydrophobic poly(methyl methacrylate) (PMMA) structured chip for quantifying peptides without internal standard by improving the detection reproducibility and sensitivity. The chip was formed by PMMA microspheres through a self-assembly method. As a good adsorbent of peptides, PMMA microspheres make peptides distributed uniformly on the chip, resulting in the elimination of the coffee ring effect and the homogeneous co-crystallization of peptides/matrix. The relative standard deviations (∼ 10 %) and coefficients of determination (∼ 0.99) are satisfactory when detecting 3 standard peptides, which show that the chip is applicable for rapid quantification of peptides. The limit of detection is improved by 1000-fold on this chip compared with conventional steel target because of the enrichment of the super-hydrophobicity. It can be concluded that the detection reproducibility and sensitivity are significantly improved by the super-hydrophobic structured-PMMA chip. The applicability of the chip for practical samples is demonstrated by quantifying bacitracin A in milk extract and bradykinin1-7 in serum with good linearities. The structured-PMMA chips exhibit competitive advantages for quantitative analysis of peptides, which have a great potential for proteomic research.

High-performance homogeneous carboxymethylcellulose-stabilized Au@Ag NRs-CMC surface-enhanced Raman scattering chip for thiram detection in fruits

Bingxue Hu,Da-Wen Sun,Hongbin Pu,Zhibin Huang

Cellulose material holds considerable promise for effective surface-enhanced Raman scattering (SERS) substrate construction due to its extensive availability, chemically modifying capacity, ease of manufacture, high flexibility and low optical activity. A large-area, high-sensitivity, stable and uniform Au@Ag nanorods (NRs)-CMC substrate was successfully developed via electrostatic repulsion by using negatively-charged core–shell Au@Ag NRs as SERS active plasmonic nanomaterial, combined with negatively-charged carboxymethylcellulose (CMC) hydrogel for nanoparticles stabilization, homodisperse and protection. The obtained Au@Ag NRs-CMC substrate showed excellent sensitivity for the detection of thiram residues in fruits containing low and abundant pigment interferents, such as apples and blueberries, with detection limits of 58 and 78 ppb, respectively. Additionally, it retained more than 80% SERS performance after storage for 9 months under ambient conditions, demonstrating its great potential in facilitating the commercialization of cellulose-based SERS technology for cost-effective detection of food contaminants with advantages of facile preparation procedure, uniformity, reproducibility and long-term stability.

Bi-model detection of sulfonamide antibiotics using a microfluidic chip-lateral flow immunoassay based on liposome-modified PCN-222

Haizhi Huang,Hefei Wang,Keyu Du,Xiaoping Yu,Xuping Shentu

Lateral flow immunoassay (LFIA) is an affordable and rapid technique for detecting veterinary drugs such as sulfonamides (SAs). However, balancing accuracy, sensitivity, and convenience remains a challenge. To address this, a microfluidic chip-based LFIA utilizing a zirconium-based organic framework, PCN-222@liposome, was developed to simultaneously detect four Sas: sulfamethizole, sulfamethoxazole, sulfamethoxydiazine, and sulfathiazole, with both colorimetric and fluorescent dual-signal detection. In this study, PCN-222 was modified with liposomes to enhance its biocompatibility and water solubility. Characterization through scanning electron microscopy, transmission electron microscopy, powder X-ray diffraction, and Fourier-transform infrared spectroscopy, confirmed the successful incorporation of liposomes with PCN-222. Under optimized conditions, the PCN-222@liposome-LFIA demonstrated high sensitivity, with detection limits ranging from 0.01 to 0.05 μg/L using fluorescent signals—10 times more sensitive than conventional gold nanoparticle-based LFIA. Finally, the PCN-222@liposome-LFIA integrated with the microfluidic chip successfully detected the four SAs in chicken, shrimp, and milk samples, with recoveries ranging from 89.65 % to 118.78 %. This study presents a simple, sensitive, and efficient method for the rapid detection of veterinary drug residues.

A recyclable PDMS microfluidic surface-enhanced Raman scattering Cu/AgNP chip for the analysis of sulfadiazine in aquatic products

Jianying Huang,Ling Xia,Xiaohua Xiao,Gongke Li

A copper/polydimethylsiloxane (Cu/PDMS) microfluidic chip was designed to synthesize SERS substrates through a displacement reaction between silver and copper in microfluidic channels to form a microfluidic SERS nanoparticles (AgNPs)/Cu chip. This SERS substrate fabrication method doesn’t require complex manufacturing processes or expensive equipment, and it can be achieved at room temperature in just 4 min. Meanwhile, the AgNPs grown on copper foil could offer ample hot spots, effectively boosting Raman signals of target molecules, and the enhancement factor (EF) was 5.12 × 107 for methylene blue, 4.08 × 107 for crystal violet, and 4.81 × 105 for sulfadiazine. The chip could be reused at least 9 times with consistent signal performance. Combined with the concentration gradient microchannel network design, we could establish the standard curve for samples through one injection on a single chip. A rapid SERS method for detecting sulfadiazine in aquatic products was developed. The linear range for sulfadiazine analysis was 75.0–1000 μg L−1 with a limit of detection of 28.4 μg L−1. Sulfadiazine was found in aquatic products (43.8 μg kg−1), with recovery rates ranging from 82.7% to 110%. The result was validated using high-performance liquid chromatography, showing a relative error of 9.3%. The entire detection procedure could be completed in just 20 min to detect sulfadiazine. These results demonstrate that the developed SERS method is fast, accurate, and reliable for analysis of sulfadiazine in aquatic products.

Bi-content micro-collagen chip provides contractility-based biomechanical readout for phenotypic drug screening with expanded and profiled targets

Hui Zhao,Lyu Zhou,Qiang Zhang,Xiaoying Zhou,Yonghui Zhang,Huijun Chen,Yanan Du

Phenotypic screening has regained momentum in the pharmaceutical industry owing to its success over target-based screening. Most phenotypic screening relies on nonspecific biochemical readouts regarding cellular viability, which hampers the discovery of novel drug mechanisms of action (MOAs). Here we present a Contractility-based bi-Content micro-Collagen Chip (3CChip), which establishes cellular contractility as a biomechanics-related phenotype for drug screening. Bi-content analysis of cell contractility (imaged by iPhone) and viability suggests that the label-free contractility-based analysis exhibits superior sensitivity to compounds targeting contractile elements (e.g. focal adhesion, cytoskeleton), resulting in a enlarged target pool for drug assessment. Six typical readout patterns of drug response are summarized according to the relative positions of the contraction/viability curves, and drug targets are profiled into three categories (biomechanical, biochemical and housekeeping) by 3CChip, which will benefit subsequent target identification. The simple-to-use and effective 3CChip offers a robust platform for micro-tissue-based functional screening and may lead to a new era of mechanism-informed phenotypic drug discovery.

Estimation of the porosity of a chitosan–carbon nanoparticle membrane fabricated on a chip: A solute transport-based study

Weiping Ding,Xingxia Zhu,Lili Zou,Jing Liu,Dongmin Wang,Cheng Liang,Chengpan Li,Liqun He

Porosity is an important parameter by which to evaluate membrane performance. In recent years, membranes formed in microfluidic chips using laminar flow-based interfacial reaction technology have gained extensive attention due to their potential in micro-separation applications; however, so far, there has not been a feasible method to determine the porosity because of the very tiny size of this type of membrane. Here, we proposed a solute transport-based method to estimate the porosity of a chitosan–carbon nanoparticle membrane fabricated in a microdevice. In this method, a counter-current microdialysis mode was designed to experimentally study urea transport across the membrane, and a fluorescein was added to the urea solution to facilitate the observation of the urea concentration profile. Then, a porosity-related mass transfer model was developed to theoretically simulate the urea transport. The membrane porosity was obtained by iterating the porosity distribution to minimize the difference between the theoretical and experimental results. Finally, the membrane porosity was experimentally validated through a creatinine clearance study. The averaged porosities of the on-chip chitosan–carbon nanoparticle membrane formed in 15, 20, and 25   min were 0.356   ±   0.050, 0.242   ±   0.018 and 0.235   ±   0.009, respectively. The method proposed here is of significance for characterizing the performance of on-chip fabricated membranes.

A microfluidic chip based on an ITO support modified with Ag-Au nanocomposites for SERS based determination of melamine

Wang Rong,Xu Yi,Wang Renjie,Wang Chunyan,Zhao Huazhou,Zheng Xiangquan,Liao Xing,Cheng Li

This article describes a microfluidic SERS chip-based rapid and high-throughput method for the determination of chemical and biological analytes, specifically of melamine. The chip consists of an indium tin oxide (ITO) glass plate modified with silver-gold nanocomposites (Ag-Au NCs) and a polydimethylsiloxane (PDMS) cover plate. The chip has five parallel microfluidic channels. The ITO glass provides electrical conductivity and has a smooth surface which makes it well suited for the deposition of the Ag-Au NC by electrodeposition and galvanic replacement. This technique allows good control of the morphology and composition of the composite. Under optimized conditions, the microfluidic SERS chip provides excellent sensitivity for 4-mercaptobenzoic acid (4-MBA). The detection limit is 0.1 nM and the SERS enhancement factor is 2.7×10 10 . The chip exhibits good stability over time and good reproducibility, with a RSD of 8.6 % between the 5 micro-channels. The chip was successfully applied to the determination of melamine in milk with a 10 nM detection limit and an analytical range from 10 nM to 0.1 mM. It is demonstrated that the microfluidic SERS chip developed here shows great promise for use in biochemical assays. A microfluidic SERS chip employing gold nanocomposites (Ag-Au NCs) as SERS-active substrate was fabricated. Its SERS enhancement factor is 2.7⋅10 10 . The chip was applied to determinate melamine in milk with detection limit of 10 nM.

Microfluidic chip for isolation of viable circulating tumor cells of hepatocellular carcinoma for their culture and drug sensitivity assay

Yu Zhang,Xiaofeng Zhang,Jinling Zhang,Bin Sun,Lulu Zheng,Jun Li,Sixiu Liu,Guodong Sui,Zhengfeng Yin

Circulating tumor cells (CTCs) have been proposed to be an active source of metastasis or recurrence of hepatocellular carcinoma (HCC). The enumeration and characterization of CTCs has important clinical significance in recurrence prediction and treatment monitoring in HCC patients. We previously developed a unique method to separate HCC CTCs based on the interaction of the asialoglycoprotein receptor (ASGPR) expressed on their membranes with its ligand. The current study applied the ligand-receptor binding assay to a CTC-chip in a microfluidic device. Efficient capture of HCC CTCs originates from the small dimensions of microfluidic channels and enhanced local topographic interactions between the microfluidic channel and extracellular extensions. With the optimized conditions, a capture yield reached > 85% for artificial CTC blood samples. Clinical utility of the system was further validated. CTCs were detected in all the examined 36 patients with HCC, with an average of 14 ± 10/2 mL. On the contrary, no CTCs were detected in healthy, benign liver disease or non-HCC cancer subjects. The current study also successfully demonstrated that the captured CTCs on our CTC-chip were readily released with ethylene diamine tetraacetic acid (EDTA); released CTCs remained alive and could be expanded to form a spheroid-like structure in a 3-dimensional cell culture assay; furthermore, sensitivity of released CTCs to chemotherapeutic agents (sorafenib or oxaliplatin) could be effectively tested utilizing this culture assay. In conclusion, the methodologies presented here offer great promise for accurate enumeration and easy release of captured CTCs, and released CTCs could be cultured for further functional studies.
