Construction of benzyl isothiocyanate-loaded fish skin gelatin-luteolin compound emulsion delivery system, and its digestion and absorption characteristics

Zhiyu Liu,Songyi Lin,Wenwen Liu,Yoshimasa Nakamura,Yue Tang

BACKGROUND Fish skin gelatin (FSG) and luteolin (LUT) were used as composite emulsifiers, and benzyl isothiocyanate (BITC) was used as a model of nutrient delivery to construct a stable emulsion. The storage stability of the FSG–LUT emulsion and its effect on BITC release were investigated both in vitro and ex vivo . RESULTS LUT can quench FSG fluorophores statically and form a stable complex through hydrogen bonding and hydrophobic interactions. The FSG–LUT emulsion storage stability and embedding rate were higher than those of the FSG emulsion. The FSG–LUT emulsion microstructure was resistant to oral and gastric digestion, and the BITC retention rate and bioaccessibility were much higher than those of the FSG emulsion. Lastly, the ex vivo everted gut sac of rat intestine study demonstrated that BITC showed the highest absorption in the ileum, and the FSG–LUT emulsion absorbed BITC and sustained a controlled release in a specific position. CONCLUSION LUT could form stable complexes with FSG, which improved the stability and bioavailability of BITC in the FSG–LUT emulsion delivery system, and promoted further intestinal BITC absorption. © 2022 Society of Chemical Industry.

A Novel Indicator Based on Polyacrylamide Hydrogel and Bromocresol Green for Monitoring the Total Volatile Basic Nitrogen of Fish

Zhepeng Zhang,Haiqing Tang,Keyan Cai,Ruiping Liang,Li Tong,Changrong Ou

An intelligent indicator was developed by immobilizing bromocresol green (BCG) within the polyacrylamide (PAAm) hydrogel matrix to monitor the total volatile basic nitrogen (TVB-N) content of fish. The FTIR analysis indicated that BCG was effectively incorporated into the PAAm through the formation of intermolecular hydrogen bonds. A thermogravimetric analysis (TGA) showed that the PAAm/BCG indicator had a mere 0.0074% acrylamide monomer residue, meanwhile, the addition of BCG improved the thermal stability of the indicator. In vapor tests with various concentrations of trimethylamine, the indicator performed similarly at both 4 °C and 25 °C. The total color difference values (ΔE) exhibited a significant linear response to TVB-N levels ranging from 4.29 to 30.80 mg/100 g at 4 °C (R2= 0.98). Therefore, the PAAm/BCG indicator demonstrated stable and sensitive color changes based on pH variations and could be employed in smart packaging for real-time assessment of fish freshness.Keywords:TVB-N;polyacrylamide hydrogel;bromocresol green;colorimetric indicator;intelligent packaging

Dietary Betaine Attenuates High-Carbohydrate-Diet-Induced Oxidative Stress, Endoplasmic Reticulum Stress, and Apoptosis in Mandarin Fish (Siniperca chuatsi)

Hongyan Li,Yanzhi Zeng,Xinyu Zheng,Guangjun Wang,Jingjing Tian,Wangbao Gong,Yun Xia,Kai Zhang,Zhifei Li,Wenping Xie,Jun Xie,Ermeng Yu

To investigate the impact of betaine on high-carbohydrate-diet-induced oxidative stress and endoplasmic reticulum (ER) stress, mandarin fish (Siniperca chuatsi) (23.73 ± 0.05 g) were fed with control (NC), betaine (BET), high carbohydrate (HC), and high carbohydrate + betaine (HC + BET) diets for 8 weeks. The results showed that betaine significantly promoted the growth of mandarin fish irrespective of the dietary carbohydrate levels. The HC diet induced oxidative stress, as evidenced by significantly elevated MDA levels. The HC diet significantly stimulated the mRNA levels of genes involved in ER stress (ire1,perk,atf6,xbp1,eif2α,atf4,chop), autophagy (ulk1,becn1,lc3b), and apoptosis (bax). However, betaine mitigated HC-diet-induced oxidative stress by modulating antioxidant enzymes and alleviated ER stress by regulating the mRNA of genes in the PERK-eIF2a-ATF4 pathway. Additionally, betaine significantly reduced the mRNA levels ofbecn1andbax, along with the apoptosis rate, indicating a mitigating effect on autophagy and apoptosis. Overall, dietary betaine improved growth, attenuated HC-diet-induced oxidative stress and ER stress, and ultimately alleviated apoptosis in mandarin fish. These findings provide evidence for the use of betaine in aquafeeds to counter disruptive effects due to diets containing high carbohydrate levels. Keywords:mandarin fish;high carbohydrate diet;betaine;oxidative stress;ER stress

Colorimetric/Fluorescent Sensing of Metal Ions and Advanced Molecular Information Safety Based on Fish Scale-Derived Carbon–Gold Nanocomposites

Ying Wu,Qing Yu Liu,Zhen Qi Bu,Min Xia Quan,Jie Zhou,Zhi Xin Xie,Jiao Yang Lu,Wei Tao Huang

It is valuable to develop simple and environmentally friendly methods for converting biowaste into functional nanomaterials with multiple applications. However, there are few reports on the facile preparation of multifunctional carbon–metal nanocomposites based on biowastes and their application from sensing to information technology. Herein, we used fish scale-derived carbon nanoparticles (FSCN) as reducing agents to directly and facilely prepare multifunctional carbon–gold nanocomposites (FSCN-AuNPs) in one step. Our prepared FSCN-AuNPs have multisignal response capabilities and can be used for colorimetric and fluorescence quantitative sensing of Pb2+ and Cu2+ with detection limits of 86.29 nM (for Pb2+) and 53.74 nM (for Cu2+), respectively. Additionally, FSCN-AuNPs can detect Pb2+ in milk and serum, as well as Cu2+ in water and serum. The multisignal selective responses of FSCN-AuNPs can be converted into binary strings by setting thresholds, which were further applied to encrypt and hide two types of information (like text and two-dimensional pattern information). Overall, this research not only promotes the preparation and comprehensive application of multifunctional carbon–metal nanocomposites but also accelerates the cross-integration of molecular science, such as the integration of molecular sensing and molecular information science.

A screening for optimal selenium enrichment additives for selenium-enriched fish production: Application of a HPLC-ICP-MS method

Chao Zhu,Qimin Liu,Yang Wang,Xiaolin Wang,Yuxuan Ma,Fangxia Yang,Wuzi Dong,Hong Ji

The production of selenium-enriched fish contributes to alleviating selenium deficiency for humans. In this study, selenium nanoparticles (SeNPs) comparable in bioavailability to selenomethionine (SeMet), increased SeMet content in O. macrolepis ( Onychostoma macrolepis ) muscle. Additionally, dietary SeNPs significantly enhanced selenocysteine (SeCys 2 ) and methylselenocysteine (MeSeCys) levels in O. macrolepis muscle. The effect of SeNPs on selenium speciation in grass carp muscle was consistent with O. macrolepis results. SeCys 2 and MeSeCys showed antioxidant capacity in HEK293T cells, indicating enhanced health benefits of Se-enriched fish produced using SeNPs. Furthermore, the addition of 0.3 mg/kg SeNPs significantly improved the flesh quality of O. macrolepis by reducing the content of crude fat and heavy metals, as well as increasing the levels of crude protein, eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA), and the ratio of n-3/n-6 polyunsaturated fatty acids (PUFAs). Therefore, selenium-enriched fish produced from SeNPs is a good source for improving human dietary selenium intake.

Synthesis of lipoic acid ferulate and evaluation of its ability to preserve fish oil from oxidation during accelerated storage

Zhiyong Xue,Juan Liu,Qing Li,Yuanyuan Yao,Yalin Yang,Chao Ran,Zhen Zhang,Zhigang Zhou

Lipoic acid ferulate (LAF) was synthesized and its anti-free radical ability in vitro was determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonicacid) (ABTS) assays. Protective effects of LAF in stabilizing fish oil were tested, compared to antioxidants such as lipoic acid, ferulic acid and tert -butylhydroxyquinone (TBHQ) by measuring peroxide values, thiobarbituric acid reactants, p -anisidine values, nuclear magnetic resonance (NMR) spectra and gas chromatography–mass spectrometry (GC–MS) spectra of fish oil during accelerated storage (12 days, 80 °C). The inhibitory effect of these antioxidants on fish oil oxidation followed the order TBHQ ≧ LAF > ferulic acid > lipoic acid. In addition, the omega-3 polyunsaturated fatty acids were the first to be oxidized. The formation of oxidation products followed a first-order kinetic model, and the addition of LAF effectively reduced the reaction rate constants. Therefore, LAF can effectively slow down the formation of oxidative products and prolong the shelf life of fish oil.

Integrated volatolomics and metabolomics analysis reveals the characteristic flavor formation in Chouguiyu, a traditional fermented mandarin fish of China

Jia-Nan Chen,Yu-Ying Zhang,Xu-Hui Huang,Meng Dong,Xiu-Ping Dong,Da-Yong Zhou,Bei-Wei Zhu,Lei Qin

Volatolomics and metabolomics were performed to explore the generation mechanism of the characteristic flavor of mandarin fish during fermentation. This study revealed a novel finding that umami-tasting amino acids, succinic acid, and peptides increased, while taste-presenting nucleotides decreased after fermentation. The results showed that 19 key aroma compounds were identified. The most nitrogenous compounds were produced after fermentation, the total concentration of which was >5 mg/kg. A high odor activity value of 443 was established for stinky indole. PLS-DA showed that sn -glycero-3-phosphocholine, hypoxanthine, creatine, and trimethylamine N -oxide were the key metabolites associated with the key volatiles. Umami-tasting amino acids could contribute to the characteristic taste. Metabolic pathway analysis revealed that tryptophan metabolism, trimethylamine metabolism, and monoterpenoid biosynthesis were the potential generation pathways of indole, trimethylamine, and terpenoids, respectively. Collectively, the results provide thoughts for targeted controlling the flavor of fermented mandarin fish.

Controllable synthesis of flower-like AuNFs@ZIF-67 core-shell nanocomposites for ultrasensitive SERS detection of histamine in fish

Shan Xu,Peifang Chen,Xichi Lin,Imran Mahmood Khan,Xiaoyuan Ma,Zhouping Wang

Histamine is a significant biomarker to assess the freshness of fish products. In this study, a novel MOF-based SERS sensor for histamine determination was synthesized by wrapping PVP-capped Au nanoflowers with a ZIF-67 shell (Au [email protected] ). The highly branched Au NFs core exhibited a strong electromagnetic field enhancement effect and provided an ultra-sensitive SERS fingerprint spectrum, while ZIF-67 shell was the contributor to enrich the target and stabilize the substrate. The morphology of the core-shell structures can be easily controlled by the concentrations of the capping agent PVP and MOF precursor Co ion . Consequently, 4-MBA pre-grafted on the optimized SERS substrate can act as the Raman internal standard (IS) to eliminate signal fluctuations through standardizing all spectra against its peak at 1074 cm −1 . Moreover, as the specific receptor for histamine molecules, 4-MBA helped reach the low detection sensitivity, where the SERS intensity ratio, I 1172 /I 1074 presented a good linear relationship towards the histamine concentrations (10 −3 -10 −7  M) with the LOD of 0.87 × 10 −7  M (R 2  = 0.9930). Furthermore, the application in monitoring fish spoilage process demonstrated the feasibility and reliability of the developed sensor. This work provided a facile strategy to construct MOF-based SERS substrate as a potential platform for the shelf-life prediction of fish products.

Colorimetric multi-channel sensing of metal ions and advanced molecular information protection based on fish scale-derived carbon nanoparticles

Ying Wu,Qing Yu Liu,Zhen Qi Bu,Min Xia Quan,Jiao Yang Lu,Wei Tao Huang

Some nanosystems based on carbon nanomaterials have been used for fluorescent chemical/biosensing, elementary information processing, and textual coding. However, little attention has been paid to utilizing biowaste-derived carbon nanomaterials for colorimetric multi-channel sensing and advanced molecular information protection (including text and pattern information). Herein, fish scale-derived carbon nanoparticles (FSCN) were prepared and used for colorimetric detection of metal ions, encoding, encrypting and hiding text- and pattern-based information. The morphology and composition of FSCN were analyzed by TEM, XRD, FTIR, and XPS, and it was found that the FSCN-based multi-channel colorimetric sensing system can detect Cr 6+ (detection limit of 56.59 nM and 13.32 nM) and Fe 3+ (detection limit of 81.55 nM) through the changes of absorption intensity at different wavelengths (272, 370, and 310 nm). Moreover, the selective responses of FSCN to 20 kinds of metal ions can be abstracted into a series of binary strings, which can encode, hide, and encrypt traditional text-based and even two-dimensional pattern-based information. The preparation of carbon nanomaterials derived from waste fish scales can stimulate other researcheres’ enthusiasm for the development and utilization of wastes and promoting resource recycling. Inspired by this work, more researches will continue to explore the world of molecular information technology.

Enzyme-assisted extraction and liquid chromatography-inductively coupled plasma mass spectrometry for the determination of arsenic species in fish

Fa Zhao,Yanming Liu,Xiqi Zhang,Rui Dong,Wenjiang Yu,Yanfang Liu,Zhimou Guo,Xinmiao Liang,Jianhua Zhu

A sensitive, simple and rapid method for the simultaneous determination of eleven arsenic species has been developed. A high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) technique was used for the analysis of eleven arsenic species in less than 17 min. Different extraction solutions were explored and the recovery results using water and aqueous acidic solvents, aqueous organic solvents and enzymes showed that 40 mg protease with 0.75 mL 0.5% hydrochloric acid ( v/v ) as the extraction agent gave the best experimental results. Species separation with ammonium carbonate solution as the mobile phase was conducted on an anion-exchange chromatographic column using gradient elution. The column temperature was 20 °C and kinetic energy discrimination (KED) was employed to eliminate spectral interference. The use of KED mode effectively removed interference from 75 ArCl. The detection limit (L D ) of the method was in the range of 0.11–0.59 μg kg −1 . Repeatability values obtained for spiked real fish samples were in the range of 1.1%–7.6%. Accuracy was calculated based on the analysis of spiked real fish samples at five concentration levels. Obtained recoveries were 91%–106%. The validated method was used in a pilot study to analyze real samples of fish, the organic arsenic especially AsB was the major arsenic specie present in the analyzed samples, only trace amount of inorganic arsenic were detected. The analytical method should improve the assessment of human exposure associated with arsenic intake from fish.

Effect of preparation factors and storage temperature on fish oil-loaded crosslinked gelatin nanoparticle pickering emulsions in liquid forms

Mengzhen Ding,Ting Zhang,Huan Zhang,Ningping Tao,Xichang Wang,Jian Zhong

Fish oils are rich in omega-3 unsaturated fatty acids and have many important physiological functions and potential disease prevention abilities. It is necessary to develop novel dosage form and formulation such as food emulsions to encapsulate fish oils for protection, increase water solubility, and isolate the fishy taste. In this work, we mainly explore the effects of preparation factors and storage temperature on fish oil-loaded crosslinked nanoparticle Pickering emulsions in liquid form. Firstly, we prepared the crosslinked gelatin nanoparticles by a two-step desolvation method. Then we prepared fish oil-loaded gelatin nanoparticle Pickering emulsions by a homogenization processes. Subsequently, we explored the effects of preparation and storage factors (gelatin nanoparticle solution pH, homogenizing time, homogenizing speed, and storage temperature). Our results confirmed the successful preparation of crosslinked gelatin nanoparticles and fish oils-loaded gelatin nanoparticle Pickering emulsions. The Pickering emulsions mainly consist of three types of droplets with different sizes. The droplet sizes linearly decreased with the increase of gelatin nanoparticle solution pH and homogenizing time, whereas exponentially decreased with the increase of homogenizing speed. Storage temperatures had obviously different effects on the storage of the fish oil-loaded crosslinked gelatin nanoparticle Pickering emulsions. This work provides a way to encapsulate fish oils in emulsions and provides potential promising application prospectives in food beverages. It would be also beneficial to basic understanding of the formation of gelatin nanoparticle-based Pickering emulsions.

Species-specific bioaccumulation and correlated health risk of arsenic compounds in freshwater fish from a typical mine-impacted river

Yuyu Jia,Lin Wang,Shan Li,Junfei Cao,Zhaoguang Yang

Arsenic (As) speciation and bioaccumulation in fish muscle tissues have been intensively investigated in marine ecosystem. However, little is known about these in freshwater fish. In this study, freshwater fish including 120 specimens and 8 species were collected from the Xiang River, a typical mine-impacted river in China. Six As species including arsenite (AsIII), arsenate (AsV), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenocholine (AsC) and arsenobetaine (AsB) were simultaneously separated and determined using HPLC-ICP-MS. The mean (±   SD) concentration of total As (tAs) in the dried fish muscle was 0.748   ±   0.651   mg·kg −   1 . AsB was found as the predominant As species in most of the studied fish samples, in accordance with the reports in marine fish. However, the diversity of inorganic/organic As proportion observed in the studied freshwater fish species was larger than that in marine fish species due to greater spatial variability of As contamination, mobilization and origination in the studied catchments. The percentage of AsB (AsB%) in fish muscle was irrelevant to tAs concentration, while the percentage of iAs (iAs%) decreased with tAs concentration in a hyperbolic pattern. This can be attributed to restricted assimilation and accumulation of toxic iAs with increasing tAs concentration in fish. Chronic non-carcinogenic and carcinogenic health risks were evaluated through Monte-Carlo simulation. The result indicated that consuming freshwater fish in the Xiang River could cause considerable carcinogenic risk to local inhabitants.
