Methicillin-resistant Staphylococcus aureus (MRSA) remains a major pathogen in ventilator-associated pneumonia and wound infections. To address the limitations of traditional antibiotics, we developed a novel iridium-based single-atom catalyst (Ir/CN SAC) anchored on a nitrogen-doped carbon matrix. Engineered for ultra-low metal loading and maximal active site exposure, this catalyst integrates robust photothermal and catalytic functionalities. Under second near-infrared (NIR-II, 1270 nm) irradiation, the Ir/CN SAC efficiently converts light to heat and catalytically generates reactive oxygen species (ROS), achieving a potent photothermal-catalytic synergistic effect. This dual-action mechanism enabled rapid bacterial eradication in vitro and significantly accelerated wound healing and lung tissue repair in MRSA-infected in vivo models. Transcriptomic analyses revealed downregulation of pro-inflammatory pathways, shedding light on the immunomodulatory roles of the treatment. Notably, the Ir/CN SAC exhibited negligible toxicity and enhanced peroxidase-mimicking activity via thermal activation. Collectively, the Ir/CN SAC presents a promising strategy for treating MRSA infections in wounds and the lungs via a synergistic treatment model.

Atherosclerosis (AS) is a risk factor for various cardiovascular diseases (CVD), and early diagnosis and plaque removal are challenges in the management of AS and CVD. To address these challenges, we designed a tungsten‑doped polydopamine nanoparticle (W-PDA NPs). The near-infrared (NIR) optical absorption and enzymatic properties of W-PDA were tuned by adjusting the ratio of W 6+ to PDA. It was found that the W-PDA NPs contain a mixture of W 6+ and W 5+ , which generated large amounts of free electrons, thus conferring both second near-infrared (NIR-II) light absorption capability and superoxide dismutase (SOD)- and catalase (CAT)-like activities. Plaque-targeting was conferred by appending an anti-osteopontin antibody via coordination between carboxyl moieties of the antibody with W 6+ ions on the W-PDA surface. The NIR-II light absorption ability can be utilized for photoacoustic diagnosis of AS, while the SOD- and CAT-like activities facilitate the conversion of superoxide into H 2 O 2 and subsequently O 2 , thereby clearing reactive oxygen species and mitigating inflammation, enabling therapy of AS. Thus, the proposed theranostic agent not only can dynamically report the progression of AS but also can effectively treat AS plaques. This strategy holds broad application prospects in the early and accurate diagnosis and treatment of AS.

A CD44-targeted, NIR-II-responsive nanoplatform (CuS@ZIF-8/HA NPs) was developed for TNBC therapy. • Acidic TME triggers ZIF-8 decomposition, releasing Cu 2+ for sustained mitochondrial copper overload. • Cu + drives lipoylated DLAT oligomerization, the core event of cuproptosis. • NIR-II photothermal therapy (55.6 °C) synergizes with cuproptosis for enhanced tumor ablation. • In vivo : pronounced tumor suppression without relapse or toxicity. Triple-negative breast cancer (TNBC) lacks targetable receptors and rapidly expels cytotoxic copper via ATPase copper–transporting alpha/beta (ATP7A/B) pumps, rendering conventional cuproptosis inducers ineffective. We report an acid-activatable, CD44-directed nanoplatform (CuS@ZIF-8/HA NPs) that converts NIR-II irradiation into a synchronized mitochondrial “copper flood–heat shock” event. In the acidic tumor microenvironment, ZIF-8 decomposes and releases Cu 2+ , which is reduced to cytotoxic Cu + , triggering oligomerization of lipoylated dihydrolipoamide S-acetyltransferase (DLAT), the core driver of cuproptosis. Concomitant ferredoxin 1 (FDX1) protein degradation serves as a downstream biochemical marker of cuproptotic injury. The photothermal effect (55.6 °C at 1 W cm −2 ) simultaneously accelerates copper release, amplifies oxidative damage, and ablates cells by hyperthermia. In 4 T1 tumor mice model, eight i.v. doses (10 mg kg −1 ) plus NIR-II light achieved profound tumor suppression without relapse or systemic toxicity, outperforming either monotherapy. Thus, CuS@ZIF-8/HA NPs turns NIR-II light into a spatially confined cuproptotic-photothermal double hit, offering a safe, precise, and translatable strategy against therapy-refractory TNBC. en This link goes to a English section zh This link goes to a Chinese section Diabetic wound infections are a common complication of diabetes, which severely impact patients' quality of life. Effective treatment of diabetic wound infections remains one major challenge in the clinic, partially due to the formation of bacterial biofilm and antibiotic resistance. It is imperative to develop non-antibiotic-dependent strategies to efficiently eradicate biofilm infections in diabetic wounds. In this work, an innovative nanospray (CDs-HM) was successfully prepared by linking one acoustic sensitizer (hematoporphyrin monomethyl ether, HMME) to CDs fabricated with hemin, nickel(II) chloride, and polymer ethylene imine. CDs-HM exhibited catalase-enhanced sonodynamic properties and photothermal-enhanced chemodynamic properties. In vitro experiments and transcriptomic analysis demonstrated that CDs-HM successfully killed the bacteria and destroyed bacterial biofilm by disrupting bacterial cell membrane integrity, inducing oxidative stress, and inhibiting ATP production through synergistic photothermal therapy/chemodynamic therapy/sonodynamic therapy (PTT/CDT/SDT) effects. In the diabetic wound infection mouse model, CDs-HM remarkably eradicated MRSA biofilm, reduced inflammation levels, and promoted angiogenesis/collagen deposition so as to accelerate wound healing. It is noteworthy that CDs-HM displayed superior bacteria killing and wound healing effects compared with conventional vancomycin and nanosilver dressing interventions. The practicality and efficiency of CDs-HM endowed it with broad clinical translation prospects. It paved the way for developing novel strategies for combating diabetic wound infections. 摘要 en This link goes to a English section zh This link goes to a Chinese section 糖尿病伤口感染是糖尿病的常见并发症,严重影响患者的生活质量。

As the primary malignant tumor in the brain, glioblastoma exhibits a high mortality due to the challenges for complete treatment by conventional therapeutic methods. It is of great importance to develop innovative therapeutic agents and methods for treatment of glioblastoma. In this work, the imaging and therapy of glioblastoma are reported by using dye sensitized core-shell NaYF 4 :Yb/Tm@NaYF 4 :Nd nanoparticles with strong up/down-conversion luminescence, of which the ultraviolet up-conversion emissions at 348 and 365 nm are significantly enhanced by nearly 28 times and used to control the release of SO 2 from 5-Amino-1,3-dihydrobenzo[c]thiophene 2,2-dioxide prodrug for gas therapy, and the second near-infrared (NIR-II) down conversion emission at 1340 nm is increased five times and applied for imaging. It is revealed that the released SO 2 molecules not only cause oxidative stress damage of tumor cells, but also induce their pro-death autophagy by down-regulating the expression of p62 and up-regulating the ratio of LC3-II/LC3-I, ultimately inhibiting tumor growth. The work demonstrates the great potential of rare earth nano-platform with functions of NIR-II imaging and photo-controlled gas therapy in the diagnosis and treatment of orthotopic glioblastoma.

On-demand delivery of drug to deep tumor tissues with long-term tumor retention has directly influence on therapeutic efficiency and prognosis of patients, which still remains challenge in cancer therapy. Here we introduce metallo-supramolecular nanoflowers through multi-drugs and metal coordination effect for near-infrared (NIR) or acidic-triggered multi-drugs release, long-term accumulation at tumor tissues and NIR-II fluorescence imaging-guided photo-chemotherapy. The nanoflowers (refer to ICG⊃EDOX) are constructed by (−)-epigallocatechin-3-gallate (EGCG), Cu 2+ ions, indocyanine green (ICG) and doxorubicin hydrochloride (DOX) through coordination effect, π-π stacking and hydrophobic force. Due to strong intermolecular interaction, the ICG⊃EDOX is superior stable in physiological conditions, while releases DOX/ICG on-demand upon the acidic/NIR laser irradiation. Moreover, ICG⊃EDOX could penetrate deep tumor tissues with ultralong tumor retention (>8 d) by microvesicle (MVs)-mediated intercellular interaction. Irradiated by 808 nm laser, the higher NIR-II fluorescence signal from ICG⊃EDOX in tumors can effectively guide local phototherapy, and the synergistic therapeutic efficiency of nanoflowers with completely elimination of tumors is achieved in comparison with DOX or DOX/ICG treated 4T1-bearing mice . Overall, the excellent therapeutic effect of nanoflowers may hold great promise for the clinic translation.

Lead-free rare-earth-based perovskites have received widespread attention for their unique optical properties, although achieving efficient broadband near-infrared (NIR) emission with these materials remains a challenge. Here the synthesis of a rare earth-based double perovskite (Cs 2 NaScCl 6 ) by an improved solid phase method is reported. The doping of Cr 3+ led to the formation of [CrCl 6 ] 3− octahedron, which exhibited a broadband NIR emission peaked at 950 nm and a half-peak width of 162 nm. It is worth noting that with the same actual Cr 3+ content, the luminous intensity of Cs 2 NaScCl 6 synthesized by the improved solid-phase synthesis is four times higher than the product synthesized by the hydrothermal method. an efficient Cl − -Cr 3+ charge transfer sensitization facilitated by localized electrons in [CrCl 6 ] 3− octahedron is the mechanism for the strong NIR emission of Cr 3+ is proposed. Calculations based on density functional theory and Bader charge analysis support the notion that electrons in [CrCl 6 ] 3− octahedrons are strongly localized in Cs 2 NaScCl 6 :Cr 3+ , which is conducive to the Cl − –Cr 3+ charge transfer process, resulting the internal quantum efficiency of 100% and external quantum yield as 42.3%. The highly efficient ultra-broadband NIR emission with excellent stability offers many opportunities for applications in the field of NIR night vision and bio-imaging.

Multidrug resistance (MDR) caused by overexpressed P-glycoprotein (P-gp) in cell membrane is the main barrier for clinical tumor chemotherapy. P-gp can pump the chemical drugs out of tumor cells depending on ATP-provided energy. Herein, a photothermal-driven intracellular magnesium ion (Mg 2+ ) regulation strategy is proposed to reverse drug resistance through constructing Mg 2+ - and doxorubicin (DOX, as model drug)-loaded bimetallic Pd@Pt nanostructure (DPd@PtM). Although DPd@PtM can deliver Mg 2+ into tumor cells through endocytosis, large amount of Mg 2+ releases outside cells. To this end, the photothermal effect of Pd@Pt nanostructure in the second near-infrared region is expected to activate the thermosensitive transient receptor potential cation channel subfamily V member 1 (TRPV1) channel for extracellular released Mg 2+ influx. Intercellular Mg 2+ accumulation suppresses tricarboxylic acid cycle to block intracellular adenosine triphosphate (ATP) production (cutoff energy supply for P-gp) and reduce O 2 consumption (downregulate P-gp expression), then inhibiting P-gp-mediated tumor MDR. Both in vitro and in vivo results demonstrate that DPd@PtM can open TRPV1 channel to elevate Mg 2+ level and then inhibit the P-gp activity to enhance intracellular DOX concentration for chemotherapy. It is believed that this photothermal-mediated tumor Mg 2+ regulation therapy based on reversing MDR is a promising strategy to kill cancer cells.

Quantitative detection of cancer cells using portable devices is promising for the development of simple, fast, and point-of-care cancer diagnostic techniques. However, how to further amplify the detection signal to improve the sensitivity and accuracy of detecting cancer cells by portable devices remains a challenge. To solve the problem, we, for the first time, synthesized folic-acid-conjugated Au nanoframes (FA-Au NFs) with amplification of pressure and temperature signals for highly sensitive and accurate detection of cancer cells by portable pressure meters and thermometers. The resulting Au NFs exhibit excellent near-infrared (NIR) photothermal performance and catalase activity, which can promote the decomposition of NH4HCO3 and H2O2 to generate corresponding gases (CO2, NH3, and O2), thereby synergistically amplifying pressure signals in a closed reaction vessel. At the same time, Au NFs with excellent peroxidase-like activity can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to produce TMB oxide (oxTMB) with a strong photothermal effect, thereby cooperating with Au NFs to amplify the photothermal signal. In the presence of cancer cells with overexpressing folate receptors (FRs), the molecular recognition signals between FA and FR can be converted into amplified pressure and temperature signals, which can be easily read by portable pressure meters and thermometers, respectively. The detection limits for cancer cells using pressure meters and thermometers are 6 and 5 cells/mL, respectively, which are better than other reported methods. Moreover, such Au NFs can improve tumor hypoxia by catalyzing the decomposition of H2O2 to produce O2 and perform photothermal therapy of cancer. Together, our work provides new insight into the application of Au NFs to develop a dual-signal sensing platform with amplification of pressure and temperature signals for portable and ultrasensitive detection of cancer cells as well as personalized cancer therapy.

Multifunctional theranostic nanoplatforms enabling precise targeting and controlled deep tissue therapy are vital. Herein, we present a self-assembled, defect-engineered, biomimetic nanoplatform (CDMBBTO), in which piezoelectric material and downconversion nanoparticles (DCNPs) are co-assembled within a polymer matrix, preventing interactions between both components, thereby allowing each to retain its intrinsic properties while collectively enabling dual second near infrared fluorescence (NIR-II FL)/magnetic resonance (MR) imaging and synergistic piezo/chemodynamic therapy (PZDT/CDT). Mn/Bi codoped piezoelectric BaTiO 3 (MBBTO) NPs exhibit a significantly enhanced piezoelectric coefficient (~5 fold higher than pristine BTO) owing to bandgap modulation induced by defect engineering. The incorporated Mn 2+ not only catalyzes Fenton like reactions for sustained tumor suppression but also provides strong T 2 weighted MR contrast. Self-assembled NIR-II emissive DCNPs enable deep tissue optical imaging, forming an integrated theranostic system. Cloaking of U87 glioma cell membranes imparts homologous tumor targeting capability. In vivo dual modal imaging reveals efficient tumor accumulation, with peak NIR-II FL and MR signal intensities observed 6 h post-injection. Upon ultrasound activation, CDMBBTO elicits potent tumor ablation through synergistic reactive oxygen species generation and immune modulation, characterized by macrophage polarization (M2 → M1) and upregulation of proinflammatory cytokines (TNF-α and IL-6). This work establishes CDMBBTO as a powerful nanoplatform for dual modal imaging-guided, immune potentiated PZDT/CDT toward effective glioblastoma treatment.

Acute kidney injury (AKI) is a highly morbid and lethal syndrome that is often clinically “silent” in early stages. Multimodal imaging, which integrates complementary structural and functional insights from various techniques, holds great potential to enhance diagnostic sensitivity and accuracy. However, developing a single-agent probe that effectively combines multiple imaging modalities for AKI remains a significant challenge. Herein, we show neodymium-enriched carbon dots (Nd-CDs) as a novel bimodal imaging probe for the early diagnosis of AKI by using computed tomography (CT) and the second near-infrared window (NIR-II) fluorescence. Nd-CDs with high metal doping level (17.9%) can be achieved on a gram-scale (2.1 g) through a “bottom-up” strategy. The ultra-small hydrodynamic size (<2 nm) endows Nd-CDs with rapid renal clearance. Benefiting from narrow-band NIR-II fluorescence emission at 1060 nm and strong X-ray attenuation, Nd-CDs provide high-spatial-resolution CT and highly sensitive fluorescence imaging in vivo . In a cisplatin-induced AKI rat model, Nd-CDs-based bimodal imaging reveals a pronounced delay in renal metabolism caused by tubular injury, which precedes abnormalities in blood biochemical parameters, enabling early diagnosis and progression monitoring of AKI. The proposed strategy provides new insights into the development of single-agent probes integrating multiple imaging modalities and opens up a new way for early diagnosis of AKI.

High quality visualization of live mammalian embryo is an important requirement for studying fetal development. Photoacoustic tomography (PAT) and optical coherence tomography (OCT) are two advanced imaging modalities that have been utilized for embryonic imaging separately. However, high contrast, multiscale, and deep tissue imaging of embryos in vivo remains challenging. Here, we demonstrate multimodal contrast-enhanced imaging of live embryos by gold nanostar-meditated near-infrared I (NIR-I) PAT and near-infrared II (NIR-II) OCT. We investigate intravenous and intravaginal administration of gold nanostars, and significant enhancement of signal, image contrast, and imaging depth are achieved for both PAT and OCT. Our findings may facilitate detailed studies of the morphological and functional changes during embryonic development.
