Casein (CN) is a major allergen in bovine milk, and reactive species generated by cold plasma have been shown to modulate its conformation and antigenicity. The effects of scavenging major reactive oxygen species (ROS) from cold argon plasma on CN modification and antigenicity were investigated in this study. Notably, scavenging hydroxyl radicals (·OH) partially reversed the plasma-induced reduction in IgE-binding capacity, with the value recovering from 24.37% to 80.44%. Structural analyses indicated that ·OH predominantly drives CN degradation and insoluble aggregation. Moreover, ·OH and singlet oxygen ( 1 O 2 ) induced irreversible oxidation and conformational rearrangement, while hydrogen peroxide (H 2 O 2 ) caused mild, partially reversible oxidation. This study highlights irreversible oxidative damage by ROS and offers insights into antigenicity regulation during cold plasma treatment.

Intrahepatic cholangiocarcinoma (ICC) is a highly aggressive liver malignancy with rising global incidence and poor prognosis. Despite recent advances in treatment strategies, effective therapeutic options for ICC remain limited. Solamargine (SM), a natural steroidal alkaloid, has demonstrated anticancer activity, yet its mechanisms in ICC are not fully understood. This study shows that SM inhibits ICC progression by inducing both apoptosis and ferroptosis in vitro and in vivo. SM significantly suppressed proliferation, migration, and invasion of ICC cells (HUCCT-1 and HCCC-9810), with minimal cytotoxicity toward normal biliary epithelial cells (HIBEC). Mechanistically, SM induced mitochondrial dysfunction, ROS accumulation, and lipid peroxidation, leading to apoptosis and ferroptosis. Transcriptome sequencing and gene set enrichment analysis (GSEA) revealed that SM treatment activated apoptosis- and ferroptosis-related pathways, including MAPK signaling. Western blot analysis and pharmacological inhibition assays confirmed that p38 MAPK activation was essential for SM-induced cell death, with ROS acting as an upstream activator. In vivo, SM significantly inhibited tumor growth in both orthotopic and subcutaneous ICC mouse models and showed better tolerability than gemcitabine. Tumor tissue analysis further confirmed increased markers of apoptosis and ferroptosis, along with p38 MAPK activation. These findings suggest that SM exerts dual antitumor effects in ICC, which are associated with the induction of apoptosis and ferroptosis and may involve the ROS/p38 MAPK signaling axis, highlighting its potential as a therapeutic candidate for ICC.

Sonodynamic therapy (SDT) for deep-seated tumors is limited by tumor microenvironment (TME) barriers. We developed a hyaluronic acid (HA)-modified mesoporous calcium carbonate nanoplatform (HA/CaCO 3 @Ce6) to synergistically enhance calcium overload and SDT. The CD44-targeted nanoplatform demonstrated pH-responsive degradation in acidic TME, resulting in the release of Ca 2+ and chlorin e6 (Ce6). The released Ca 2+ induced mitochondrial calcium overload, causing 71% collapse in membrane potential and 1.6-fold increase in reactive oxygen species (ROS) generation, establishing a “Ca 2+ -ROS positive feedback loop.” This synergy triggered robust immunogenic cell death (ICD), enhancing CRT exposure by 94.2%, HMGB1 release by 46.2%, and ATP decrease by 74.5%. In vivo , it achieved 90.9% tumor inhibition and 80% 60-day survival rate, alleviated tumor hypoxia, and inhibited tumor proliferation and angiogenesis. This “ion-interference SDT” paradigm overcomes the limitations of SDT through self-amplified ROS cycle and provides an effective strategy for treating deep-seated solid tumor with significant clinical translation potential.

Polycystic ovary syndrome (PCOS) involves oxidative stress-driven ovarian dysfunction and remains difficult to treat due to drug side effects and poor target engagement. We engineered a reactive oxygen species (ROS)-responsive, macrophage-membrane-camouflaged quercetin nano-therapy (MM@PCD@QNPs) to enhance ovarian delivery and mitigate toxicity. The core comprises a pinacol phenylboronate-dextran conjugate that encapsulates quercetin and undergoes ROS-triggered release; a surface M0 macrophage membrane confers immune evasion and lesion tropism via retained proteins (e.g., CD11b and CD47-SIRPα). MM@PCD@QNPs displayed nanoscale dimensions and stability (133.63 ± 14.60 nm; −33.13 ± 1.52 mV) and released drug under elevated ROS. In DHT-injured granulosa cells and a DHEA-induced PCOS mouse model, the formulation promoted granulosa cell proliferation, suppressed apoptosis, reduced ROS, and preferentially accumulated in ovaries, with negligible in vitro and in vivo toxicity. Transcriptomics and validation implicate activation of the MAPK7-Nrf2-NQO1 axis as a principal mechanism; pharmacologic MAPK7 inhibition abrogated therapeutic effects. By coupling ROS-triggered release with macrophage-mimetic targeting, MM@PCD@QNPs overcome quercetin's low bioavailability and off-target exposure and provide a safe, effective nanoplatform for PCOS therapy.

Radiotherapy for head and neck cancer frequently induces severe radiation-induced oral mucositis (RIOM), in which excessive reactive oxygen species (ROS), defective macrophage efferocytosis, and persistent inflammation form a self-amplifying pathological loop that delays mucosal repair. Current symptomatic treatments are insufficient to simultaneously suppress oxidative stress and restore immune-mediated clearance of damaged cells. Here, we developed a detachable microneedle patch with ROS-responsive tips (PTC MN) for localized in situ macrophage programming within RIOM lesions. The PTC MN was fabricated from a PPBA-TA-PVA hydrogel matrix, which combines intrinsic ROS-scavenging capacity with oxidative stress triggered degradation, and loaded with engineered hybrid nanovesicles (HLENs-CAR) carrying a plasmid encoding CAR-ectoCRT-IL-4. This design enabled local delivery of gene-loaded nanovesicles, macrophage-targeted genetic programming, and sustained retention of the therapeutic payload in the injured mucosa. In a murine RIOM model, PTC MN accelerated mucosal epithelial regeneration, reduced oxidative stress and inflammatory infiltration, enhanced reparative macrophage responses, and attenuated fibrosis-associated tissue remodeling. Functionally, the platform enhanced macrophage efferocytosis, promoted IL-4-associated reparative polarization, and shifted the lesion microenvironment toward inflammation resolution and tissue regeneration. Collectively, this study establishes a smart in situ immunomodulation strategy that integrates ROS-responsive microneedle delivery with CAR-macrophage programming, providing a potential therapeutic paradigm for refractory mucosal injury.

Thermoelectric materials enabled thermoelectric-reactive oxygen species (ROS) conversion under temperature gradients, offering a promising strategy for suppressing postoperative recurrence of bone tumors and associated bacterial infections. However, their clinical translation was hindered by the inability to achieve spatiotemporally controlled ROS release within bone tissue. To address this, a magnetothermal-thermoelectric heterojunction (Bi 2 Te 3 -MnFe 2 O 4 ) was constructed by in situ growth of manganese ferrite (MnFe 2 O 4 ) on bismuth telluride (Bi 2 Te 3 ) nanosheets, and was then incorporated into poly-L-lactic acid (PLLA) scaffold using additive manufacturing. Under an alternating magnetic field, the magnetothermal effect of MnFe 2 O 4 induced a localized temperature increase, which activated the thermoelectric effect of Bi 2 Te 3 . This process drove the separation of electron-hole pairs and catalyzed ROS generation to antitumor and antibacterial. Furthermore, the multivalent metal ions (Mn 2+ /Mn 3+ and Fe 2+ /Fe 3+ ) in MnFe 2 O 4 mediated Fenton-like reactions, further enhancing ROS production and depleting glutathione via chemodynamic therapy. The magnetothermal heating also accelerated the kinetics of these processes, forming a synergistic therapeutic outcome. Experimental results demonstrated that the ROS produced by the scaffold induced mitochondrial dysfunction and apoptosis in tumor cells, and effectively killed bacteria by causing intracellular content leakage. This study integrated the magnetothermal, thermoelectric, and chemodynamic effects into a bone scaffold, providing an innovative strategy for comprehensive postoperative treatment of bone tumors. Reactive oxygen species (ROS) are significantly elevated in tumor cells, providing an intrinsic trigger for selective drug activation and tumor-specific therapy. However, commonly used ROS-responsive linkers, such as thioketals and boronates, often suffer from limited sensitivity or poor physiological stability, and the development of new ROS-responsive motifs remains scarce. Herein, we develop a hydrazide-derived diazaborine (DAB) scaffold and establish it as a general ROS-responsive linker for small-molecule prodrugs, combining physiological stability with efficient ROS responsiveness. Based on this platform, we construct a DAB-derived ROS-responsive copper complex small-molecule prodrug, DDABD@Cu, for tumor-specific therapy. DDABD@Cu integrates a DAB-caged diethyldithiocarbamate (DTC) moiety with an inert copper donor, in which the DAB group effectively blocks the copper-chelating activity of DTC, enabling inert co-delivery of both components. Upon ROS activation, DTC is released to initiate copper transfer, generating cytotoxic Cu(DTC) 2 in situ to induce proteotoxic stress via ubiquitinated protein accumulation while concomitantly producing quinone methide (QM) to deplete glutathione (GSH) and amplify oxidative stress. This cascade improves antitumor efficacy against non-small cell lung cancer while limiting off-target toxicity. Overall, this work provides a feasible strategy for designing ROS-responsive and copper complex prodrugs.

Antibacterial treatment reduces residual bacterial burden in periodontitis, but rapid bacterial killing and lysis can also increase exposure to danger signals such as bacterial DNA (bDNA), which can contribute to cGAS–STING-associated innate immune activation. To address this bactericidal stress-associated inflammation, we constructed a ROS-responsive coordination nanosystem, CPLEA, in which the STING inhibitor C-176 was incorporated during Ag⁺-mediated co-assembly of epicatechin and the sulfur-containing PEG ligand methoxy poly(ethylene glycol)–dihydrolipoic acid (mPEG-DHLA), thereby integrating antibacterial activity with the regulation of post-bactericidal STING-associated inflammation. Without compromising bacterial killing, CPLEA promoted C-176 release under oxidative conditions, and the release profile showed temporal overlap with bacterial killing and bDNA exposure. CPLEA attenuated the inflammatory state of macrophages and relieved the suppression of hPDLSC osteogenic differentiation by macrophage-conditioned medium. Transcriptomic analysis indicated downregulation of cytosolic DNA sensing, NF-κB, IL-17, and osteoclast differentiation programs. In a ligature-induced mouse model of periodontitis, CPLEA attenuated local inflammation and TRAP-positive osteoclast accumulation and reduced alveolar bone loss. Collectively, these findings support bactericidal stress-associated STING signaling as a relevant target during local antibacterial treatment of periodontitis and position CPLEA as a feasible materials-based approach for this purpose. Statement of significance Periodontitis treatment often requires additional antimicrobial control after mechanical debridement, yet rapid bacterial killing and lysis can increase the exposure of bacteria-derived danger signals and thereby sustain host inflammatory responses. This study develops an oxidation-responsive coordination nanoplatform that integrates antibacterial activity with STING-targeted immunomodulation. By incorporating the STING inhibitor C-176 into an Ag⁺-mediated coordination system, CPLEA enables oxidation-promoted C-176 release within a temporal window overlapping bacterial killing and bacterial DNA exposure. The study further demonstrates that early C-176 intervention more effectively regulates STING-associated inflammatory signaling than delayed intervention. CPLEA attenuated macrophage inflammatory responses, alleviated inflammation-associated suppression of periodontal ligament stem cell osteogenic differentiation, and reduced inflammation, osteoclast accumulation, and alveolar bone loss in experimental periodontitis. These findings highlight the importance of immunomodulator availability during the early phase of bactericidal treatment and provide a biomaterials strategy for integrating residual bacterial control with local host modulation. Hydrogel is increasingly appreciated as a reliable dressing for wound repair and regeneration due to the established favorable microenvironment for healing. Thus, hydrogel with multiple synergistic functions are of particular interest in biomedicine. Herein, we develop a double network hydrogel with low swelling property, outstanding adhesive capacity, good biocompatibility, excellent anti-bacterial and ROS scavenging capacity, which shows potential for accelerating wound healing. Quaternized chitosan serves as the first network to endow the hydrogel with anti-bacterial capacity (75.18% for E. coli and 93% for S. aureus ). Zwitterionic monomer is copolymerized with acrylic acid to form the second network, which improves the mechanical properties and reduces the swelling behavior (ultralow swelling ratio of 2.42%). Tannic acid and dihydrocaffeic acid contribute excellent tissue adhesion (adhesion strengths of 124.4 kPa to pig skin) and antioxidant capacity (ROS scavenging rate of 96%). Based on these advantages, the hydrogel greatly facilitates the wound closure, as well as promotes epithelialization, collagen deposition and neovascularization.

SUMMARY The microbial community residing on the phyllosphere is influenced by many factors, including the host plant's genotype as well as its secondary metabolites. Anthocyanins are a group of flavonoids renowned for their antioxidative properties and are widely distributed across plant tissues. However, the potential impact of anthocyanins on plant-associated microbial communities remains unknown. In the model legume Medicago truncatula , we isolated a mutant named purple leaves ( pl ) that produces purple leaves at a young stage due to over-accumulated anthocyanins. Through sequencing 16S rRNA amplicons of phyllosphere microbes in the pl mutant, we show that anthocyanins significantly enhance the abundance of endophytic lactic acid bacteria within plant leaves. Further in vitro study revealed that anthocyanins derived from pl can significantly promote the growth of lactic acid bacteria under anaerobic conditions. The accumulated anthocyanins in pl leaves reduced reactive oxygen species (ROS), thereby creating a favorable environment for the growth of facultative anaerobic lactic acid bacteria and resultantly increasing the abundance of phyllosphere lactic acid bacteria. Our findings elucidate the role of anthocyanins in modulating the community structure of phyllosphere microbiota in M. truncatula and provide new insights into the relationship between plant secondary metabolites and phyllosphere microbiota.

Photodynamic therapy (PDT) is a promising cancer treatment approach that relies on the localized generation of reactive oxygen species (ROS) to eliminate cancer cells. In particular, the nanophotonic approach based on upconversion nanoparticles (UCNPs) offers a key advantage by enabling the use of near-infrared (NIR) light, which enhances light penetration into tissue and expands clinical applicability of PDT. Real-time monitoring of ROS generation and degradation during the PDT process offers distinct advantages over conventional endpoint assays for elucidating PDT mechanisms, optimizing photosensitizer (PS) formulations and refining treatment protocols. In this study, we not only distinguish and quantify the relative contribution of NaYF 4 :Yb 3+ ,Tm 3+ UC nano-antennas, Rose Bengal (RB) PS, NIR activation laser, and culture medium in UCNP-based PDT for the first time via real-time ROS analysis using dissolved oxygen (DO) data which cannot be achieved by endpoint assays but also introduce new and insightful concepts such as medium activation time (FWHM), maximum PL lifetime change (Δτ max ), and time to reach the maximum PL lifetime change ( τ max). This is realized by implementation of a 3D-printed optofluidic dissolved oxygen (DO) sensor for indirect analysis of ROS dynamics which infer from changes in the sensor's photoluminescence (PL) lifetime (τ). Thus, performance and optimum concentrations of NaYF 4 :Yb 3+ ,Tm 3+ UCNPs and RB PS are first determined via MTT assays using A375 melanoma cells, and subsequent in-vitro PDT tests using a 980 nm laser. Quantitative analyses show that, UCNPs, RB, and the cell culture medium contribute approximately 25 %, 26 %, and 4 % to the total Δτ respectively. The maximum performance occurs when all components are present and activated, resulting in the highest ROS level with the longest activation time. Interestingly, even laser excitation of the medium alone or UCNPs without PS results in partial ROS generation. These findings provide valuable insights for optimizing UCNP-based PDT drugs for cancer treatment.

Sclerotinia stem rot (SSR), caused by Sclerotinia sclerotiorum , significantly reduces soybean yields. Conventional SSR-based pesticide application presents environmental and health hazards, whereas ROS-responsive nanocarrier systems demonstrate superior efficacy as a sustainable alternative. In this study, PYD@OxbCD NPs, encapsulating Pydiflumetofen (PYD), were synthesized and showed ROS-triggered release and strong in vitro activity against SSR. This targeted release mechanism reduces ecological risk by minimizing pesticide exposure to non-target organisms. PYD@OxbCD delivers PYD more precisely to infected soybean tissues, outperforming traditional Pydiflumetofen suspension concentrate (SC) in antifungal efficacy. It also upregulates defense-related genes and reduces disease incidence and severity in soybean plants. PYD@OxbCD also mitigates the phytotoxic effects of PYD SC on seedling growth. Overall, PYD@OxbCD shows strong potential as an effective nanoplatform for managing SSR in soybeans, enhancing disease control and pesticide utilization for sustainable agriculture.
