Antibacterial chitosan composite films with food-inspired carbon spheres immobilized AgNPs

Zhengguo Wu,Shuwei Tang,Weijie Deng,Jiwen Luo,Xiaoying Wang

The cumulative toxicity of AgNPs has limited their application in food packaging. As such, the quest for AgNPs should focus on controlling their release to reduce the cumulative toxicity. Here, two kinds of green hydrothermal carbonized methods were used to treat sulfhydryl-modified chitosan to obtain two kinds of carbon spheres/AgNPs (Glutinous rice sesameballs-like AgNPs-SMCS and dragon fruit-like SMCS-Ag), which exhibited good stability and high immobilization efficiency for AgNPs, and the release of total Ag from AgNPs-SMCS and SMCS-Ag were only about 5.63% and 3.59% after 14 days, respectively. Subsequently, they were added into chitosan separately to prepare chitosan-based films. Two carbon spheres/AgNPs regulated the microstructure of chitosan-based films because of the electrostatic interaction and the micro-nanometer filling behavior, thus further immobilized the AgNPs. Importantly, the films presented good antibacterial activity and excellent safety. These results will provide a theoretical basis for the green and safe design of AgNPs antibacterial agent.

ROS-Eliminating Carboxymethyl Chitosan Hydrogel to Enhance Burn Wound-Healing Efficacy

Yang Cheng,Chen Yuhui,Huang Hai,Fan Shicai,Yang Chengliang,Wang Liping,Li Wenqiang,Niu Wenxin,Liao Jianwen

Overexpression of reactive oxygen species (ROS) can lead to chronic inflammation, which limits skin wound healing. Therefore, it is of great significance to develop materials that can locally control the adverse reactions caused by excessive ROS. In this research, an ROS-sensitive hydrogel with strong free radical scavenging ability was prepared by introducing the thione (Tk) group into carboxymethyl chitosan (CMCTS) hydrogel. CMCTS hydrogel was cross-linked by NH2-Tk-NH2 agent and loaded curcumin (Cur), which possessed favorable nontoxicity, water absorption, mechanical property, biodegradability, drug release behavior, the M2 phenotype, and inflammatory factor regulating the capacity of macrophages. It is worth noting that Cur@CMCTS-Tk hydrogel can significantly inhibit oxidative damage of human fibroblasts in the H2O2-induced microenvironment and protect their viability by reducing the production of intracellular ROS. In vivo, ROS-removing hydrogel effectively accelerated the process of wound healing and possessed good regenerative properties, including hair follicle formation, promotion of new blood vessel formation, and highly orderly arrangement of collagen fibers in the full-thickness skin burn defect rat model. Hence, we expect that the Cur@CMCTS-Tk hydrogel could be used for wound treatment and tissue regeneration due to the ability to scavenge excess ROS.

Removal of Phloridzin by Chitosan-Modified Biochar Prepared from Apple Branches

Zhiting Ma,Wenyan Du,Zhubing Yan,Xuesen Chen,Yanfang Wang,Zhiquan Mao

Phloridzin is a phenolic acid associated with the genus Malus (Mill.) that causes apple replant disease. The removal of phloridzin is of great significance to alleviate this condition, and hence biochar was investigated for this purpose. The goal of this study was to improve the adsorption capacity of biochar for phloridzin by chemical modification. Biochar from apple branches and biochar modified with chitosan to were characterized by elemental analysis, Brunauer Emmett Teller analysis, Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscopy. Batch adsorption experiments were employed to select the optimal biochar dosage and adsorption time. The results showed that biochar treated for 24 h at 6 g L−1 achieved best adsorption properties and that the chitosan-modified biochar provided a higher adsorption capacity than the unmodified material. The influence of pH upon the adsorption was studied and the results were fitted with isotherm and kinetic equations. The results showed that adsorption process corresponds to a Langmuir isotherm and pseudo-second-order kinetics. The qmax of the biochar was increased from 15.93 to 28.96 mg g−1 by modification with chitosan. The pH was shown to play an important role in the adsorption process. The adsorption of phloridzin to biochar was demonstrated to be a single-layer multiple component adsorption process based on chemisorption. This study provides a theoretical basis to alleviate apple replant disease.

Macroporous zwitterionic composite cryogel based on chitosan oligosaccharide for antifungal application

Peng Dong,Xiulin Shu,Ruqun Peng,Shunying Lu,Xiaobao Xie,Qingshan Shi

Chitosan oligosaccharide (COS), a time-dependent antimicrobial carbohydrate, is found antifungal active with a short duration of action due to excessive solubility. We attempted to address this issue by employing a hydrogel as a COS carrier. In this research, macroporous zwitterionic composite cryogels composed of COS and poly( N -methacryl arginine) (PMarg) were fabricated, serving as long-term antifungal dressings. Firstly, Marg was synthesized and characterized by Fourier transform infrared spectroscopy (FT-IR), 1 H and 13 C nuclear magnetic resonance (NMR), and high-resolution mass spectrometry (HRMS). Then, the COS/PMarg cryogels were prepared by redox initiation cryopolymerization. The macroporous morphology of the cryogels was confirmed by scanning electron microscope (SEM) with pore size varying from 20.86 to 50.87 μm. FTIR indicated that hydrogen bonding formed between COS and PMarg, and the interaction elevated thermal stability of the cryogels as evidenced by thermal-gravimetric analysis (TGA). Swelling capacity, mechanical properties, and COS release behavior of the COS/PMarg cryogels were investigated. With the release of COS, the antifouling activity of the cryogel increased. Antimicrobial tests indicated the COS/PMarg cryogel could effectively inhibit the proliferation of Candida albicans . It demonstrated that the macroporous zwitterionic COS/PMarg composite cryogel might be a potential antifungal dressing with sequential “sterilization-release” capacity.

Enhanced cellular compatibility of chitosan/collagen multilayers LBL modified nanofibrous mats

Fangfang Dai,Jia Yu,Mengqin Yuan,Zhimin Deng,Yanqing Wang,Yaqi Fan,Hongbing Deng,Yanxiang Cheng

Developing novel, lightweight and biocompatible mesh for pelvic organ prolapse, a gynecological disease, has attracted the global interest of biomedical researchers. In our study, polycaprolactone and nylon 6 mats with excellent mechanical properties were fabricated via co-electrospinning. Positively charged chitosan with antimicrobial properties and negatively charged collagen with good biocompatibility were then assembled on the nanofiber mats via layer by layer (LBL) technology. SEM and mechanical property tests showed that LBL deposition increased the nanofibers’ diameter and elastic modulus. To evaluate the successful deposition process, the zeta potential, The Fourier-transform infrared, X-ray diffraction, and X-ray photoelectron spectroscopy analyses were conducted to systematically study the mats’ chemical characterization. The results indicated that the mats’ composition and structure significantly changed after LBL modification. Cell biology experiments demonstrated that the LBL-structured mats can facilitate cell growth, proliferation, and adhesion. Antibacterial tests confirmed their excellent antibacterial activity. Chitosan/Collagen-coated Polycaprolactone/Nylon 6 mats demonstrate considerable potential as implant materials for pelvic reestablishment in pelvic organ prolapse patients.

Cationic chitosan-modified silica nanoparticles for oral delivery of protein vaccine

Xi Wu,Muhammad Asim Farooq,Tiantian Li,Tianjiao Geng,Perpeuta Takunda Kutoka,Bo Wang

Mesoporous silica nanoparticles coated with Chitosan are exploited here as a potential carrier for oral vaccine delivery. Bovine serum albumin (BSA) was used as a protein antigen model to reveal the carrier property. Chitosan-coated BSA-loaded silica NPs had particle size 345 ± 60 nm with a cationic surface charge of 18.28 ± 0.71 mV. The encapsulation efficiency, drug loading was 25.34 ± 0.76 and 20.21 ± 0.48%, respectively. Transmission electron microscopy investigation showed the spherical shape of NPs, also confirmed surface coating around modified nanoparticles (NPs), and nitrogen absorption/desorption isotherm confirmed mesostructured inside the NPs. Fourier transform infrared spectroscopy did not show any physiochemical interactions between excipients and formulations. The structural stability of antigen after release from NPs was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis, and chitosan-coated silica NPs exhibited a slow-release pattern. The results of in vivo experiments presented that chitosan-mesoporous silica NPs could induce a robust immune response in mice, indicating that chitosan-mesoporous silica NPs might be used as a promising carrier for oral vaccine delivery.

Size-Dependent Biodistribution of Fluorescent Furano-Allocolchicinoid-Chitosan Formulations in Mice

Iuliia Gracheva,Maria Konovalova,Dmitrii Aronov,Ekaterina Moiseeva,Alexey Fedorov,Elena Svirshchevskaya

The aim of this study was to compare the biodistribution in mice of functionalized rhodamine B (Rh) labeled colchicine derivative furano-allocolchicinoid (AC,6) either conjugated to 40 kDa chitosan (AC-Chi,8) or encapsulated into chitosan nanoparticles (AC-NPs). AC-NPs were formed by ionotropic gelation and were 400–450 nm in diameter as estimated in mice by dynamic light scattering and confocal microscopy. AC-Chi and AC-NPs preserved the specific colchicine activity in vitro. AC preparations were once IV injected into C75BL/6 mice; muscles, spleen, kidney, liver, lungs, blood cells and serum were collected at 30 min, 2, 5, 10, and 20 h post injection. To analyze the distribution of the furano-allocolchicinoid preparations in body liquids and tissues, Rh was measured directly in sera or extracted by acidic ethanol from tissue homogenates. Preliminary Rh extraction rate was estimated in vitro in tissue homogenates and was around 25–30% from total quantity added. After in vivo injection, AC-NPs were accumulated more in liver and spleen, while less in kidney and lungs in comparison with free AC and AC-Chi. Therefore, incorporation of colchicine derivatives as well as other hydrophobic substances into nano/micro sized carriers may help redistribute the drug to different organs and, possibly, improve antitumor accumulation.Keywords:furano-allocolchicinoid;chitosan;nano/micro particles;biodistribution;Wnt-1 breast tumor

Semiconductive, Flexible MnO2 NWs/Chitosan Hydrogels for Efficient Solar Steam Generation

Muhammad Sultan Irshad,Xianbao Wang,Misbah Sehar Abbasi,Naila Arshad,Zihe Chen,Zhenzhen Guo,Li Yu,Jingwen Qian,Jun You,Tao Mei

Solar-driven steam generation is anticipated as one of the most promising and inventive technologies to address the primitive issues of water shortage. Although extensive attempts have been made to develop highly efficient solar steam generators, hindrances are faced to integrate all desired functions in a single evaporating system. Herein, we designed semiconductive in situ-polymerized MnO2 nanowires/chitosan (SPM-CH) hydrogels as flexible, built-in, vertically aligned, macropore-based water channels (∼0.5 μm pore size) for enhanced solar water generation (17.02 kg m–2 in 1 day). The nonradiative relaxation-dependent defect engineering of SPM-CH hydrogel promotes more lattice vibrations, and its polymeric network endorses the formation of enhanced intermediate water clusters for vapor generation. The self-floating and salt-resistant device possesses an excellent evaporation rate (1.78 kg m–2 h–1) during a single sunny day along with efficient solar energy conversion efficiency (90.6%) under 1 sun intensity, good solar absorption (94%), and good compressing flexibility (42% compressive strain). Moreover, COMSOL Multiphysics simulations of SPM-CH hydrogels under experimental conditions reveal its superior centralized heat accumulation within the top-interface matrix. The single-step execution for a freshwater supply purified from various contaminations including industrial wastewater and oil-emulsified water shows its potential as a reusable device toward real-life applications.

Facile synthesis and antibacterial activity of geraniol conjugated chitosan oligosaccharide derivatives

Ran Bi,Lin Yue,Sobia Niazi,Imran Mahmood Khan,Dan Sun,Bin Wang,Zhouping Wang,Qixing Jiang,Wenshui Xia

A new chitosan oligosaccharide derivative (COS-N-Ger), based on geraniol (Ger) modificated onto the NH 2 position of chitosan oligosaccharide (COS) via a facile method, was prepared and employed to evaluate in vitro antibacterial activity. The structures of COS-N-Ger derivatives were confirmed by FT-IR, 1 H, 13 C NMR, and elemental analysis. The characterization results showed successful synthesis of derivatives and the degrees of substitution (DS) of COS-N-Ger1-3 were from 0.260 to 0.283 with the yields up to 78 %. The in vitro antibacterial activity evaluation results presented a significant inhibition effect of COS-N-Ger1-3 derivatives on Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) as compared to COS. Moreover, their antibacterial activities were dose-dependent and more sensitive to S. aureus than E. coli . The results provide reliable theoretical supports for exploring the application of COS derivatives in the food industry as new potential antibacterial agents.

Chitosan-Based Thermo-Sensitive Hydrogel Loading Oyster Peptides for Hemostasis Application

Dongying Zhang,Zhang Hu,Lingyu Zhang,Sitong Lu,Fengyan Liang,Sidong Li

Uncontrolled massive hemorrhage is one of the principal causes of death in trauma emergencies. By using catechol-modified chitosan (CS-C) as the matrix material and β glycerol phosphate (β-GP) as a thermo-sensitive agent, chitosan-based thermo-sensitive hydrogel loading oyster peptides (CS-C/OP/β-GP) were prepared at physiological temperature. The hemostatic performance of CS-C/OP/β-GP hydrogel was tested in vivo and in vitro, and its biological safety was evaluated. The results showed that the in vitro coagulation time and blood coagulation index of CS-C/OP/β-GP hydrogel were better than those of a commercial gelatin sponge. Notably, compared with the gelatin sponge, CS-C/OP/β-GP hydrogel showed that the platelet adhesion and erythrocyte adsorption rates were 38.98% and 95.87% higher, respectively. Additionally, the hemostasis time in mouse liver injury was shortened by 19.5%, and the mass of blood loss in the mouse tail amputation model was reduced by 18.9%. The safety evaluation results demonstrated that CS-C/OP/β-GP had no cytotoxicity to L929 cells, and the hemolysis rates were less than 5% within 1 mg/mL, suggesting good biocompatibility. In conclusion, our results indicate that CS-C/OP/β-GP is expected to be a promising dressing in the field of medical hemostasis. Keywords:chitosan;oyster peptides;hydrogel;hemostasis

The preparation of multifunction chitosan adhesive hydrogel by “one- step” method

Ruixue Liu,Yingbo Li,Jichao Chen,Xiaojing Zhang,Zhibin Niu,Yadong Sun

Chitosan adhesive hydrogels have attracted interest because of their unique properties. However, it is still a challenge to use simple and unmodified chitosan to synthesize multifunctional hydrogels with adhesive, self-healing and strain sensing properties. Here, a multifunctional hydrogel of Ca2+-chitosan/polyacrylamide(Ca2+- CS/PAAm)with adhesive self-healing and strain sensing properties was carried out by one-step preparation method. The hydrogel has the ability to adhere to a variety of materials, wherein the adhesion strength to rubber surface is 48.4 kPa. The hydrogel has excellent strain sensing capability on finger bending or cyclic stretching, the sensitivity factor is 0.41 ± 0.015. The cut hydrogel healed completely at room temperature within 4 h, and the healing hydrogel can be stretched to 5 times of its original length. This makes Ca2+-CS/PAAm hydrogel to be great potential in the application of strain sensors, wound dressing and skin adhesives.

Intra-articular Injection of Chitosan-Based Supramolecular Hydrogel for Osteoarthritis Treatment

Mou Donggang,Yu Qunying,Zhang Jimei,Zhou Jianping,Li Xinmin,Zhuang Weiyi,Yang Xuming

Background: Pain and cartilage destruction caused by osteoarthritis (OA) is a major challenge in clinical treatment. Traditional intra-articular injection of hyaluronic acid (HA) can relieve the disease, but limited by the difficulty of long-term maintenance of efficacy. Methods: In this study, an injectable and self-healing hydrogel was synthesized by in situ crosslinking of N -carboxyethyl chitosan ( N -chitosan), adipic acid dihydrazide (ADH), and hyaluronic acid–aldehyde (HA-ALD). Results: This supramolecular hydrogel sustains good biocompatibility for chondrocytes. Intra-articular injection of this novel hydrogel can significantly alleviate the local inflammation microenvironment in knee joints, through inhibiting the inflammatory cytokines (such as TNF-α, IL-1β, IL-6 and IL-17) in the synovial fluid and cartilage at 2- and even 12-weeks post-injection. Histological and behavioral test indicated that hydrogel injection protected cartilage destruction and relieved pain in OA rats, in comparison to HA injection. Conclusion: This kind of novel hydrogel, which is superior to the traditional HA injection, reveals a great potential for the treatment of OA.
