The combination of scallop male gonads hydrolysates (SMGHs) and κ -carrageenan (KC) was applied for curcumin (Cur) embedment and preservation, the corresponding Cur retention rate as well as gelation and microstructural properties were investigated during storage subjected to various pH (8 and 6) and temperature (25 °C and 4 °C). SMGHs-based gels could well protect Cur from degradation during 4 °C storage, with retention rate ranging from 95% to 97% after 30 day storage. At 25 °C, Cur could still be well preserved in SMGHs-based gels under pH 6 with Cur retention rate above 90%, while pH 8 led to rapid Cur degradation to around 75%. In addition, at pH 8 and 25 °C, the gel strength of SMGHs-Cur deteriorated significantly while evidently enhanced in SMGHs/KC-Cur due to synergistic combination within SMGHs and KC in couple with dehydration effect. Moreover, the water migration and distribution of SMGHs-based Cur-loaded vehicles were relatively stable at 4 °C while revealed apparent deterioration at 25 °C during storage. Furthermore, the microstructures of SMGHs/KC-Cur gradually transformed from homogeneous network with dense and smooth pores to tattered network with large and fragmentary pores, especially at 25 °C, promoting more Cur degradation. This study demonstrated that SMGHs/KC-Cur exhibited better than SMGHs-Cur in Cur preservation, potentially performing as excellent and sustainable protein hydrolysate/polysaccharide soft materials in food and biomedical fields.

Hollow mesoporous carbon nanospheres with accessible interior cavity, short diffusion length, and abundant mesopore channels can favor guest encapsulation and facilitate mass diffusion and transfer. However, there still remains great challenge in controllable synthesizing mesoporous carbons with uniform hollow architectures and relatively large mesopores by a straightforward method. Herein, a self-generating dual template synergetic assembly strategy was developed to construct hollow structured nitrogen-doped mesoporous carbon nanospheres (NMCNs) for the first time. Differentiating from conventional emulsion template methods, this synthesis involved the generation of nanoemulsion, which was driven by the interaction between oleic acid and triethanolamine based on the newborn soap effect. The process from alkalic to acidic environment induced the polymerization of the 3D cross-linking framework and established coulomb forces for dual template synergetic assembly. Significantly, the inner-cavity size of NMCNs could be facilely changed by simply varying the addition of oleic acid. Furthermore, varying the ethanol/water volume ratio would regulate the surface roughness of the NMCNs by directing the polymerization mode. This method innovatively coupled the dual template strategy and emulsion polymerization system, providing new insight into the construction of hollow mesoporous materials. Impressively, the hollow NMCNs featured dual pore size (5.2, 8.6 nm), high surface area (≈ 723.48 m 2 /g), and high nitrogen content (12.24 wt%), which delivered enhanced release rate and in vitro bioactivity of insoluble drug curcumin.

In this study, in order to minimize the degradation of curcumin, Pickering emulsion with embedded curcumin (PCNCs/EGCG-Cur) was prepared and stabilized by pineapple peel cellulose nanocrystals (PCNCs) and ( −)-Epigallocatechin-3-gallate (EGCG). The effect of EGCG on the stability and bioaccessibility of the embedded curcumin in Pickering emulsion was assessed. FTIR, XRD and zeta-potential analysis proved the construction of PCNCs/EGCG structure and CLSM showed typical oil-in-water system of PCNCs/EGCG-Cur. Furthermore, the addition of EGCG affected contact angles of the oil–water interface. 30-day storage period showed that the retained curcumin was about 70.35 ± 1.06% in PCNCs/EGCG(0.04%)-Cur, while the retention rate was 47.15 ± 0.98% in the control. PCNCs/EGCG(0.04%)-Cur was the most stable under the conditions of thermal and ultraviolet light treatments with the maximum retention rates of curcumin were 92.20 ± 1.05% and 93.56 ± 1.37%, respectively. Moreover, EGCG was found to improve the antioxidant activity of the prepared emulsions. Simulated digestion exhibited PCNCs/EGCG(0.04%)-Cur could improve the bioaccessibility of both EGCG (24.99 ± 3.29%) and curcumin (35.97 ± 1.26%). These results are conducive to the further application of EGCG in the emulsified food stabilized with cellulose nanocrystals.

The development of efficient, economic, reliable, and accurate monitoring of hypochlorite (ClO–) in food matrices is in great demand for food safety assessment, particularly during its massive use against the COVID-19 epidemic. Here, we prepared an aggregation-induced emission (AIE) fluorophore tetraphenylethylene (TPE)-incorporated curcumin-based hybrid ratiometric fluorescence nanoprobe (Curcumin/TPE@HyNPs) through amphiphilic phospholipid polymer-powered nanoprecipitation, which exhibited a fast, highly sensitive, and selective response to the residual ClO– in real food matrices. Because of the inner filter effect (IFE) from curcumin toward TPE inside the nanoprobe, the bright fluorescence of TPE aggregation at ∼437 nm was effectively quenched, along with an enhanced fluorescence of curcumin at ∼478 nm. Once there was a ClO– residue in food matrices, ClO– triggered the oxidation of o-methoxyphenol inside curcumin and led to the almost complete absorption collapse, thereby terminating curcumin fluorescence at ∼478 nm and the IFE process. Accordingly, the fluorescence of TPE at ∼437 nm was recovered. In this case, a ratiometric fluorescent response of Curcumin/TPE@HyNPs toward the residual ClO– in food matrices (e.g., milk) was proposed with a low detection limit of 0.353 μM and a rapid response time of 140.0 s. Notably, the phospholipid polymer as the protection layer effectively reduced/evaded the nonspecific binding of signal reporters inside the nanoprobe, facilitating it to directly monitor the residual ClO– in real food matrices. This work provided a novel approach to utilize the unconventional AIE luminophors for constructing the efficient and reliable early warning mechanisms toward various food contaminants.

The present study focused on the development of Cur-loaded SOHA nanogels (Cur-SHNGs) to enhance the topical administration of Cur. The physiochemical properties of Cur-SHNGs were characterized. Results showed that the morphology of the Cur-SHNGs was spherical, the average size was 171.37 nm with a zeta potential of −13.23 mV. Skin permeation experiments were carried out using the diffusion cell systems. It was found that the skin retention of Cur-SHNGs was significantly improved since it showed the best retention value (0.66 ± 0.17 μg/cm2). In addition, the hematoxylin and eosin staining showed that the Cur-SHNGs improved transdermal drug delivery by altering the skin microstructure. Fluorescence imaging indicated that Cur-SHNGs could effectively deliver the drug to the deeper layers of the skin. Additionally, Cur-SHNGs showed significant analgesic and anti-inflammatory activity with no skin irritation. Taken together, Cur-SHNGs could be effectively used for the topical delivery of therapeutic drugs.

Excessive free fatty acid- (FFA-) induced endothelial lipotoxicity is involved in the pathogenesis of atherosclerosis. Endoplasmic reticulum (ER) stress is mechanistically related to endothelial lipotoxicity. Lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) is the major oxidatively modified low-density lipoprotein (OxLDL) receptor in endothelial cells and is highly abundant in atherosclerotic lesions. Curcumin reduces the LOX-1 expression; however, the mechanism underlying this effect remains unknown. In the current study, we explored whether curcumin ameliorates palmitic acid- (PA-) induced endothelial lipotoxicity and LOX-1 upregulation by reducing ER stress in human umbilical vein endothelial cells (HUVECs). We built endothelial lipotoxicity in vitro and found that LOX-1 was upregulated after PA stimulation, during which ER stress played an important role. Next, we observed that curcumin substantially alleviated PA-induced lipotoxicity by restoring cell viability, increasing angiogenesis, and decreasing lipid deposition. Furthermore, LOX-1 upregulation in HUVECs was blocked by curcumin, possibly via ER stress suppression. Overall, our findings demonstrated that curcumin alleviates endothelial lipotoxicity and LOX-1 upregulation, and ER stress inhibition may play a critical role in this effect.

The simultaneous therapy of tumor recurrence and bone defects resulting from surgical resection of osteosarcoma is still a challenge in the clinic. Combination therapy based on a localized drug-delivery system shows great promise in the treatment of osteosarcoma. Herein, bifunctional polydopamine (PDA)-modified curcumin (CM)-loaded silk fibroin (SF) composite (SF/CM-PDA) nanofibrous scaffolds, which combined photothermal therapy with chemotherapy to synergistically enhance osteosarcoma therapy, were prepared by PDA coating of the SF/CM nanofibrous scaffolds fabricated by supercritical carbon dioxide (SC-CO2) technology. The PDA coating improved hydrophilicity and mechanical strength of the SF/CM scaffolds. The SF/CM-PDA scaffolds present good photothermal conversion capacity and excellent photostability. The low pH and near-infrared (NIR) irradiation could effectively accelerate release of CM in the SF/CM-PDA scaffolds. The in vitro anticancer results indicated that the biocompatible SF/CM-PDA scaffolds had a long-term, stable, and superior anticancer effect compared to pure CM. Furthermore, the SF/CM-PDA scaffolds significantly increased the growth inhibition of osteosarcoma MG-63 cells under NIR irradiation (808 nm and 1.3 W/cm2). Besides, the SF/CM-PDA scaffolds could enhance osteoblast MC3T3-E1 cell proliferation in vitro when the mass ratio of CM was 0.05–0.5%. This work has therefore demonstrated that the bifunctional SF/CM-PDA scaffolds provide a competitive strategy for local osteosarcoma therapy and bone regeneration.

Photosensitizer (PS)-mediated photodynamic therapy (PDT) has attracted more and more attention as an alternative to traditional antibiotic therapy. Nevertheless, the limitations of traditional photosensitizers seriously hinder their practical application, as a result, the methods to improve the antibacterial properties of traditional photosensitizers have become a hot topic in the field of photomedicine. Herein, a compound nano-PS system has been constructed with synergistic photodynamic and photothermal (PTT) antibacterial effects, triggered by a dual-wavelength illumination. Fluorescent carbon dots (CDs) were synthesized and employed as carriers for the delivery of curcumin (Cur) to obtain CDs/Cur. Upon combined near-infrared and 405 nm visible dual-wavelength irradiation, CDs/Cur could simultaneously generate ROS and a moderate temperature increase, triggering synergistic antibacterial effects against both Gram-positive and Gram-negative bacteria. The results of scanning electron microscopy and fluorescence confocal imaging showed that the combined effect of CDs/Cur with PDT and PTT caused more serious damage to the cell membrane. In addition, CDs/Cur exhibited low cytotoxicity and negligible hemolytic activity, showing great biocompatibility. Therefore, the construction of CDs/Cur by employing CDs as photosensitizer delivery carriers provides a strategy for the improvement of the antibacterial effect of the photosensitizer and the design of next-generation antibacterial agents in photomedicine.

Background A novel nanoemulsion (CU/FU-LN) was developed as an oral 5-fluorouracil and curcumin co-delivery system for synergistic efficacy against liver cancer. Methods MTT assay, confocal laser scanning microscope, and H&E staining were utilized to establish the efficacy and safety of CU/FU-LN.Results The AUC(0-t) of CU/FU-LN was 8.85-fold and 8.59-fold greater than those of CU and FU, respectively. The IC50 of CU/FU-LN was 4.6-fold and 4.9-fold lower than those of FU and CU in HepG2 cells, respectively. In vivo anti-tumor trials, the tumor inhibition rate was significantly elevated by CU/FU-LN (49.29%), compared 24.84% and 4.72% for FU and CU, respectively. Ki-67 immunohistochemical analysis revealed that CU/FU-LN had an obvious anti-proliferation effect. The IC50 of CU/FU-LN in L02 cells was 1.51-fold and 2.60-fold higher than those of CU and FU, respectively. Certain vital organs in the mice of the CU/FU-LN group showed markedly fewer lesions than those of the CU, FU, and CU+FU groups. The CU/FU-LN treatment caused no significant change in mouse body weight relative to the control group (P > 0.05). Conclusions We successfully prepared a promising co-delivery platform for the synergistic treatment of liver cancer and it has a comparatively enhanced efficacy and mitigated toxicity.

In this study, we used fucoidan (FD), a sulfated polysaccharide , to stabilize zein and obtain a composite nanosystem with better performance that can be used to encapsulate and deliver hydrophobic active substances. The composite nanoparticles of zein and FD with different mass ratios were prepared by the antisolvent precipitation method. The solution (zein/FD = 1:2) could maintain its stable solution state without precipitation under different environmental conditions (pH 2–8, heating at 80 °C for 120 min, salt concentration < 25 mmol/L and storage at 4 °C for 28 days). Subsequently, curcumin (Cur) was used as the model of hydrophobic active substances. When 2 mg Cur was added, the composite nanosystem had a high encapsulation efficiency (81.07%) and could reduce the release of Cur in simulated gastrointestinal digestion. Our results demonstrated that the composite nanosystem prepared by FD and zein, which are both natural biopolymers , has great potential in the encapsulation and delivery of hydrophobic active substances.

A simple approach was developed to formulate polysaccharide-based high internal phase emulsions (HIPEs) using complexes of sugar beet pectin (SBP), tannic acid (TA), and chitosan (CS) to stabilize the system. The ability of pectin to stabilize the HIPEs was significantly improved after it formed complexes with TA, which were likely held together by hydrogen bonding. The stability of the HIPEs was further improved by adding CS, which was attributed to the formation of a 3D biopolymer network around and between the oil droplets. The impact of TA concentration, CS concentration, and pH on the oil droplet size, microstructure, and rheology of the HIPEs was investigated. Compared with SBP/TA complexes, the SBP/TA/CS complexes facilitated the formation of stable HIPEs with a gel-like texture and good stability to centrifugation, dilution, thermal treatment, and ultraviolet light. HIPEs prepared from SBP/TA/CS complexes were shown to delay lipid digestion and improve curcumin bioaccessibility. These findings provide a useful approach for designing HIPE-based soft solids from natural ingredients that can be used in foods and other commercial products.

In order to further improve the photocatalytic performance of the SiO2/TiO2composite under visible light irradiation, curcumin-doped SiO2/TiO2nanocomposites were synthesized via directly incorporating it into the structure of SiO2/TiO2during the synthesis using an inexpensive and readily available natural pigment (curcumin) as doping agent. The physicochemical properties of SiO2/TiO2nanocomposites were characterized in detail by X-ray diffraction, transmission electron microscopy, Fourier transform-infrared spectroscopy, N2adsorption–desorption isotherms, X-ray photoelectron spectroscopy, and UV-vis diffuse reflectance spectroscopy. The results indicate that all SiO2/TiO2nanocomposites exhibited an anatase phase with a typical mesoporous structure. It was found that the dope of curcumin in the SiO2/TiO2composite could decrease the crystal size, slightly improve the specific surface areas, significantly enhance the visible light absorption, and effectively narrow the band gap energy from 3.04 to 10(eV). Compared with bare SiO2/TiO2, the curcumin-doped SiO2/TiO2resulted in enhanced photocatalytic reduction activity for Cr(VI) under visible light irradiation, and the CTS (12) sample with the appropriate content of curcumin of 12 wt % shows the photocatalytic yield reaching 100% within 2.5 hours, which is larger than CT (12) without silica. This could be attributed to the curcumin doping and the synergetic effects of SiO2and TiO2in SiO2/TiO2nanocomposites.Keywords:curcumin pigment;SiO2/TiO2nanocomposites;visible photocatalyst;reduction of Cr (VI)
