Polyphenol-enhanced wet adhesive hydrogel with synergistic mechanical activation and ROS scavenging for accelerating diabetic wound healing

Min You,Yaxin Guo,Hui Yu,Haiyan Yin,Xinlei Shi,Ziqing Tang,Jia Yang,Gang Qin,Jianliang Shen,Qiang Chen

Diabetic wound healing is severely affected by the vicious cycle of wound exposure and impaired repair ability. Here, a polyphenol-enhanced wet adhesion hydrogel with the synergistic effect of mechanical activation and reactive oxygen species (ROS) scavenging was developed to promote the healing of infected diabetic wounds. The hydrogel was copolymerized with acrylic acid (AAc), 1-vinylimidazole (VI), and tannic acid (TA) in dimethyl sulfoxide, followed by solvent exchange with water to obtain a strong toughness and wet adhesion hydrogel (PAAcVI-TA hydrogel). The introduction of TA not only enhanced the toughness and wet tissue adhesive strength of the PAAcVI-TA hydrogel but also conferred outstanding ROS scavenging performance, inflammatory inhibition, and vascular growth promotion. More importantly, the PAAcVI-TA hydrogel could transmit mechanical control force to wet skin through robust adhesion and mechanically contract diabetic wounds. After stretching, the tough PAAcVI-TA hydrogel presented excellent resilience, which allowed for long-term contraction of the diabetic wound, reduced bacterial infection, promoted granulation tissue formation and collagen deposition, and accelerated re-epithelialization. As a result, the PAAcVI-TA hydrogel, which combined mechanical activation with ROS scavenging, might provide a new approach to the effective treatment of diabetic wounds and other forms of chronic wounds.

Polyphenol-Assisted Biomineralization of Metal–Organic Framework Nanoparticles for Precision Delivery of Therapeutic Proteins to Cancer Cells

Tianli Luo,Qizhen Zheng,Ji Liu,Rui Yao,Ming Wang

The delivery of proteins into the cytosol holds great promise for cell signaling manipulation and the development of precision medicine. However, this potency is challenged by achieving targeted and controlled delivery, specifically within diseased cells. In this study, we introduce a versatile and effective method for the precision delivery of therapeutic proteins to cancer cells by designing polyphenol-assisted biomineralization of zeolite imidazole framework-8 (ZIF-8). We demonstrate that by leveraging the strong noncovalent binding affinity of epigallocatechin gallate (EGCG) with both proteins and ZIF-8, our approach significantly enhances the biomineralization of ZIF-8, which in turn improves the efficiency of protein encapsulation and intracellular delivery. Moreover, the incorporation of EGCG within ZIF-8 enables controlled degradation of the nanoparticles and the selective release of the encapsulated proteins in cancer cells. This selective release is triggered by the oxidation of EGCG in response to the high levels of reactive oxygen species (ROS) found within cancer cells that destabilize the EGCG/ZIF-8 nanoparticles. We have further demonstrated the ability of EGCG/ZIF-8 to deliver a wide range of proteins into cancer cells, including bacterial virulence protein, to rewire cell signaling and prohibit tumor cell growth in a mouse xenograft model. Our strategy and findings underscore the potential of designing the EGCG/ZIF-8 interface for specific and controlled protein delivery for targeted cancer therapy.

Polyphenol-hyaluronic acid-based hydrogel remodels the wound microenvironment and eliminates bacterial infection for accelerating wound healing

Tao Liu,Hu-Chen Cao,Ru Wang,Qiang Yang,Shuang Wei,Pei Pan,Hui Shi

The wound microenvironment, often characterized by alkaline pH and severe hypoxia, presents significant challenges to the healing of bacterial-infected wounds. While considerable research has focused on improving wound healing through effective bacterial elimination using advanced therapeutic approaches, the importance of regulating the wound microenvironment has received less emphasis. In this work, we developed a biocompatible hydrogel, HTFC, by incorporating CaO 2 nanoparticles (CaO 2 NPs) into a gel formed by tannic acid (TA), hyaluronic acid (HA), and Fe 3+ . The HA and TA in HTFC hydrogel help to create a slightly acidic microenvironment, facilitating the decomposition of CaO 2 NPs to release H 2 O 2 for chemodynamic therapy (CDT). The reduction properties of TA promote the recycling of Fe 3+ /Fe 2+ , enhancing CDT efficacy and partially converting H 2 O 2 to O 2 , thereby alleviating hypoxia. Additionally, Fe III TA complexes in HTFC enhance CDT through photothermal therapy (PTT)-induced improvement of the Fenton reaction. This multifunctional hydrogel, with its synergistic effects of PTT and CDT, along with its ability to remodel the wound microenvironment from hypoxic and alkaline to normoxic and acidic, accelerates the bacterial-infected wound healing process.

Polyphenol-Modified Mg−Zn Layered Hydroxide-Contained Microneedle Patch Enhance Mucosal Repair by Remolding Diabetic Oral Microenvironment

Haibo Liu,Zhongyi Sun,Mengting Yin,Jing Ru,Xiaohao Liu,Ziyan Chen,Yating Liu,Wang Sun,Longpo Zheng,Xinyu Zhao,Feng Chen

Diabetic oral ulcers pose a significant challenge in healing due to persistent inflammation. Despite local therapeutic interventions remaining the primary mode of treatment, the dynamic nature of the oral cavity, characterized by continuous muscular activity and salivary secretion, poses barriers to sustained drug retention and thereby limits therapeutic efficacy. To address this issue, an approach has been devised that aims to facilitate transdermal delivery of bioactive components to promote the healing of diabetic oral ulcers. A multifunctional soluble microneedle (MN) patch is prepared using γ-polyglutamic acid (γ-PGA), which is loaded with quercetin (Qu)-modified Mg−Zn layered hydroxide salt (LHS) nanosheets (LHSQ) with anti-inflammatory, antimicrobial and pro-angiogenic bioactivities to combat the diabetic oral ulcers. These findings demonstrate that the LHSQ-loaded MN (LHSQ-MN) patches are capable of effectively penetrating the oral mucosa. In a rat model of diabetic oral ulcers, the application of LHSQ-MN patches is found to facilitate the establishment of regeneration sites, regulate the inflammatory microenvironment of damaged tissues, exhibit potent antibacterial effects, expedite the reconstitution of the mucosal epithelium, and accelerate ulcer healing through the promotion of angiogenesis. These outcomes highlight the potential of LHSQ-MN patches as a promising therapeutic strategy for the management of oral ulcers in diabetic patients.

Preparation and structural characterization of epoxidized soybean oils-based pressure sensitive adhesive grafted with tea polyphenol palmitate

Jun Li,Yongyan Kuang,Wenlong Li,Panpan Xu,Dan Peng,Penghui Zhou,Yanlan Bi

Vegetable oils-based pressure sensitive adhesives (PSAs) are green and sustainable but face unsatisfactory adhesion strengths and are prone to aging during storage and application due to the existence of residual double bonds and massive ester bonds. Nine common antioxidants (tea polyphenol palmitate (TPP), caffeic acid, ferulic acid, gallic acid, butylated hydroxytoluene, tertiary butylhydroquinone, butylated hydroxyanisole, propyl gallate, and tea polyphenols) were grafted into epoxidized soybean oils-PSA (ESO-PSA) system to enhance antiaging properties and adhesion strengths. Results showed ESO-PSAs grafted with caffeic acid, tertiary butylhydroquinone, butylated hydroxyanisole, propyl gallate, tea polyphenols, or TPP didn't occur failure with TPP having best performance. The optimal conditions were ESO reacted with 0.9 % TPP, 70 % rosin ester, and 7.0 % phosphoric acid at 50 °C for 5 min, under which peel strength and loop tack increased to 2.460 N/cm and 1.66 N, respectively, but peel strength residue reduced to 138.09 %, compared with control (0.407 N/cm, 0.43 N, and 1669.99 %). Differential scanning calorimetry and thermogravimetric results showed TPP grafting increased the glass transition temperature of ESO-PSA slightly but improved its thermal stability significantly. Fourier transform infrared spectroscopy and 1 H nuclear magnetic resonance results showed TPP, phosphoric acid, and rosin ester all partially participated in the covalently crosslinking polymerization of ESO-PSAs and the rest existed in the network structures in the free form.

Xueting Zhao,Tingyuan Wang,Ruoxi Wang,Youyou Lan,Yuanyuan Jiang,Jiefeng Pan

Solar-driven interfacial evaporation, as an emerging solar thermal utilization technology for water production, provides an opportunity for alleviate water-energy nexus. Superwetting membranes, as famous interface materials , combine hierarchical micro-nano architectures and open-porous structures that enable high-efficient solar-driven interfacial evaporation. Herein, the design and synthesis of photothermal membranes with polyphenol-mediated nanostructured coating by integrating the tannic acid (TA)-polyethyleneimine (PEI) complexation and the oxidation-mediated TA-Fe II complexation is reported. The assembled TA-PEI/Fe II/III coating features hierarchical raspberry-like architectures derived from the TA-PEI nanoconjugates and the oxidized metal-organic hybrid networks with black appearance, which enabled the broaden solar absorption, excellent photothermal conversion and fast water diffusion of the photothermal membranes. Driven by sunlight illumination, the photothermal membranes are capable to achieve evaporation rate of 2.01 kg·m −2 ·h −1 and solar-to-vapor conversion efficiency of 94.6 %, which competes well with other photothermal membranes. The superposition of TA-PEI/Fe II/III coating over open-porous membrane substrates synchronously promoted thermal localization and facilitated water-to-steam generation. The reliable durability and salt resistance of the photothermal membranes are also demonstrated. Furthermore, photothermal membranes enables clean water production from seawater and sewage, demonstrating >99.9 % salt and oil pollutant removal . The universality of the polyphenol-mediated coating strategy offers powerful opportunities to engineer photothermal membranes for water purification application.

Green and Efficient Extraction Approach for Polyphenol Recovery from Lotus Seedpods (Receptaculum Nelumbinis): Gas-Assisted Combined with Glycerol

Nina Bao,Marwan M. A. Rashed,Bianling Jiang,Kefeng Zhai,Zisheng Luo

In this paper, the gas-assisted combined with glycerol extraction (GAGE) for polyphenol recovery from lotus seedpods (LSPs) was modeled and optimized. Box–Behnken design was applied to optimize the total polyphenol content (TPC) of LSP along with enhancing antioxidant activities using response surface methodology based on the TPC extraction yield (%), which was affected by glycerol concentration, time, temperature, and glycerol-to-solid ratio. The optimal conditions for the LSP extract were glycerol-to-solid ratio, 42 mL/g; time, 50 min; concentration of glycerol, 45%; and temperature, 70 °C. Ultra-high-pressure liquid chromatography integrated with triple-time-of-flight mass spectrophotometry (UPLC-Triple-TOF/MS) analysis revealed nine biologically active polyphenols. Furthermore, Fourier-transform infrared spectroscopy and scanning electron microscopy results demonstrated the effect and influence during extraction. The findings suggested that GAGE is a potential, green, and high-efficiency alternative that could be used to recover polyphenols from plant source byproducts.

Development and application of a high-performance underwater adhesive based on natural polyphenol/IPDI-modified epoxidized soybean oil

Jiali Kong,Yuanyuan Tu,Jiyong Wu,Shuyan Yan,Runkai Wang

Achieving high-strength, durable adhesion in extreme humid or submerged environments has long been a challenge in materials science. Traditional adhesives failed to meet the requirements for rapid wetting, strong interfacial locking, and long-term stability due to issues like hydration layer interference, hydrolysis instability, and curing difficulties. This study proposed a novel ternary design using polyphenol/IPDI-modified epoxidized soybean oil (ESO) to develop a polyphenol-based soybean oil adhesive modified by gallic acid, tea polyphenols, and tannic acid (GTSO), as well as its IPDI-introduced crosslinked derivative (GTSO-I), for underwater adhesion. This system offers environmentally friendly properties, strong interfacial compatibility, high adhesion strength, rapid curing, and long-term stability. The system incorporates three natural polyphenols—gallic acid (GA), tea polyphenols (TP), and tannic acid (TA)—to form synergistic hydrogen bonds, coordination bonds, and hydrophobic interactions at the underwater interface. These polyphenols enable a ring-opening reaction with the epoxy moiety of epoxidized soybean oil (ESO), creating a chemical framework that provides strong adhesion to wet substrates. IPDI was then introduced to form a dense crosslinked network, enhancing curing speed and stability. Experimental results showed that GTSO-I achieved a shear strength of 4.50 MPa on steel substrates, surpassing most reported underwater adhesives. After 30 days of immersion, it retained over 93 % of its initial adhesion strength and maintained a shear strength of over 3.2 MPa across a broad pH range (1-13) and in 2.0 mol/L NaCl solution. The adhesive showed rapid curing underwater and was compatible with various substrates, demonstrating potential for applications in pipeline sealing, hull repair, and biomedicine.

A novel smart pH-responsive nanocomposite based on cerium-polyphenol -amine self-assembly for anticorrosion waterborne epoxy coatings

Peng Xu,Xinwei Wang,Tianguan Wang,Xinyu Zhao,Jingsha Tan,Wenfeng Ge,Bo Zhang,Guozhe Meng

To overcome the limited protective performance of waterborne epoxy (WEP) coatings, a novel scalable cerium-polyphenol-amine ternary system (Ce-TA-PEI, CTP) was synthesized via a simple dynamic supramolecular self-assembly strategy. The material effectively anchored corrosion-inhibiting cerium ions within its network through cerium-phenolic coordination, achieving an exceptionally high cerium loading capacity of 20.3 wt%. The polyphenol–amine crosslinked network of the CTP nanocontainer can form chemical bonds with the epoxy matrix, ensuring good interfacial compatibility and significantly reducing the intrinsic defects of the coating. Consequently, the dry and wet adhesion strengths of the coating increased to 3.13 MPa and 2.99 MPa, respectively. After 60 days of immersion in 3.5 wt% NaCl solution, the | Z | 0.01Hz value of CTP2/WEP remained as high as 1.21 × 10 9  Ω·cm 2 , more than an order of magnitude greater than that of pure WEP (1.05 × 10 8  Ω·cm 2 ). In addition, UV–Vis analysis confirmed that CTP is pH-responsive and can sustain continuous cerium release for up to 144 h. This behavior arises from changes in coordination strength between cerium ions and phenolic hydroxyl groups, together with variations in electrostatic interactions within the CTP framework under different pH conditions, which promote rapid cerium release in acidic environments. When incorporated into WEP coatings, this “smart” nanocontainer responded dynamically to localized acidic corrosion at coating defects. It not only enhanced the physical barrier properties of the coating but also imparted active self-healing functionality. This work therefore provides a new strategy for advancing environmentally friendly WEP coatings toward broad engineering applications.

Macrophage-targeted cerium-tannic acid metal-polyphenol framework nanoenzyme promotes in situ heart valves regeneration through adaptive immunomodulation

Peng Song,Yunlong Wu,Xing Chen,Yafei Liu,Mengna Dong,Jiawei Shi,Xichi Wang,Nianguo Dong,Weihua Qiao,Qin Wang

Constructing in situ tissue engineered heart valves based on xenogeneic decellularized heart valves (DHVs) is a promising strategy for heart valve regeneration. However, the inflammation triggered by foreign body responses results in maladaptive matrix remodeling and compromised mechanical support. Given the critical role of macrophages (M ø s) in regulating several homeostasis to relieve xenogeneic rejection and promote tissue regeneration, folic acid modified cerium ions-tannic acid metal-polyphenol framework nanoparticles (FCT NPs) have been synthesized via a green coordination method and then loaded onto thiolated DHVs to reprogram macrophage phenotype. FCT NPs, with multiple enzyme-mimicking activity, biodegradability and biocompatibility, moderately scavenge various reactive oxygen species in M1 M ø s, reprogramming them to increase the M2 phenotype. This reduces inflammatory factors levels and promotes secretion of anti-inflammatory and pro-regenerative cytokines, enabling elimination of inflammation and promotion of adaptive matrix remodeling. In vitro studies show that FCT-loaded DHVs (FCT@DHVs) exhibit excellent immunomodulatory capability, mechanical properties, hemocompatibility, and cytocompatibility. Rat implantation models reveal that FCT@DHVs achieve re-endothelialization and adaptive matrix remodeling via immunomodulation. They also exhibit excellent hemodynamics, hemocompatibility, histocompatibility, anti-calcification and mechanical support. Notably, M2 M ø s numbers decrease with scaffold degradation, indicating self-adaptive immunomodulation. This strategy offers a promising approach for in situ heart valve regeneration based on xenogeneic DHVs.

Summary Here, we present a protocol for controllable biomimetic mineralization at the nano-scale, simulating natural ion-enriched sedimentary mineralization. We describe steps for treatment of metal-organic frameworks with polyphenol-mediated stabilized mineralized precursor solution. We then detail their use as templates to assemble metal-phenolic frameworks (MPFs) with mineralized layers. Furthermore, we demonstrate the therapeutic benefits of MPF delivery by hydrogel to the full-thickness skin defect model in rats. One-step rapid co-deposition of oxidant induced mussel-polyphenol coating on PVDF substrate for separating oily water

Yaping Zhang,Yujia Tong,Lijian Shi,Changqing Miao,Weixing Li

Inspired by mussels and plant polyphenols, surface-coated membranes for oil-in-water emulsion separation have attracted extensive attention recently. Here, a one-step rapid co-deposition strategy was proposed to fabricate membranes for separating oily water by oxidation reaction of tannic acid and polydopamine. The polyphenol cross-linked network was formed and quickly co-deposited on the surface of polyvinylidene difluoride (PVDF) membrane. The developed hydrophilic membrane was able to efficiently separate different oil-in-water emulsions. FTIR-ATR, XPS and SEM characterization methods were used to investigate the chemical composition and surface morphology of the modified membranes. Water contact angle, underwater oil contact angle, emulsion permeability, and oil rejection were measured. The experimental results showed that the permeability of pure water and the isooctane-in-water emulsion using PVDF-0.1/0.4/0.2 membrane were 15,713 L∙m −2 ∙h −1 ∙bar −1 and 14,552 L∙m −2 ∙h −1 ∙bar −1 , respectively. The oil rejection reached 99.3%. After five cycles, the initial permeability and the oil rejection were 12,849 L∙m −2 ∙h −1 ∙bar −1 and 98.6%, respectively. The developed PVDF-0.1/0.4/0.2 membrane still maintained excellent underwater oleophobicity after being treated in acid, base and salt solutions for 30 days. Furthermore, the time for membrane fabrication was shortened to only 1 h. The proposed membrane modification strategy would have potential application prospects in the efficient separation of oily wastewater.
