Plain Language Summary Background Intracerebral hemorrhage (ICH) induces secondary brain injury, driven in part by oxidative stress caused by hemin, a toxic hemoglobin breakdown product. Cortical astrocytes, critical for maintaining redox homeostasis, are vulnerable to hemin-induced oxidative damage, exacerbating neuronal injury. Induced pluripotent stem cell–derived neural progenitor cells (iPSC-NPCs) have shown therapeutic potential in brain injury models by promoting neural repair, but their ability to protect astrocytes against hemin toxicity remains unexplored. We hypothesized that iPSC-NPCs coculture mitigates hemin-induced oxidative stress in astrocytes by activating the nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) antioxidant pathway. Materials and methods Astrocytes were exposed to hemin with or without iPSC-NPC coculture. We assessed cell viability, reactive oxygen species (ROS) accumulation, apoptosis, and the role of the Nrf2/HO-1 pathway using small-interfering RNA. Results iPSC-NPC coculture significantly reduced hemin-induced oxidative damage by promoting Nrf2 nuclear translocation and upregulating HO-1, thereby decreasing ROS and apoptosis. Silencing Nrf2 abolished these protective effects. Conclusion Our findings demonstrate that iPSC-NPCs protect astrocytes from hemin toxicity via the Nrf2/HO-1 pathway, suggesting a novel therapeutic strategy for ICH-induced oxidative injury. Plain Language Summary Intracerebral hemorrhage (ICH) can cause brain damage due to oxidative stress from hemin, a toxic breakdown product of hemoglobin. This study explored whether induced pluripotent stem cell-derived neural progenitor cells (iPSC-NPCs) can protect astrocytes, which are crucial for brain health, from hemin-induced damage. Researchers found that when astrocytes were cocultured with iPSC-NPCs, oxidative stress and cell death were reduced. This protective effect was linked to the activation of the Nrf2/HO-1 antioxidant pathway. Silencing Nrf2 negated the benefits, highlighting the pathway's importance. These findings suggest iPSC-NPCs could be a promising treatment for oxidative injury in ICH. Text is machine generated and may contain inaccuracies. FAQ

Cinnamic acid, isolated from cinnamon bark, is a natural product with excellent bioactivity, and it effectively binds with cyclohexanedione to form novel 4-hydroxyphenylpyruvate dioxygenase (HPPD) inhibitors. According to the active sub-structure combination principle, a series of novel 3-hydroxy-2-cinnamoyl-2-en-1-one derivatives were designed and synthesized. The title compounds were characterized by infrared, 1H NMR, 13C NMR, and HRMS. The in vitro inhibitory activity of AtHPPD verified that compound II-13 showed the most activity with a half-maximal inhibitory concentration (IC50) value of 0.180 μM, which was superior to that of mesotrione (0.206 μM) in vitro. The preliminary herbicidal activity tests demonstrated that some compounds had good herbicidal activity especially compound II-13 at a concentration of 150 g ai/ha. The binding mode of AtHPPD through molecular docking indicated that two oxygens of compounds II-13 formed bidentate interactions with metal ions, and the benzene ring formed π-π accumulation effects with Phe-381 and Phe-424. The results of molecular dynamics simulations showed that compound II-13 exhibited a more stable binding ability with AtHPPD than mesotrione. This study provided insights into the development of natural and efficient herbicides in the future.

The density, viscosity, and refractive index of a pure asymmetrical dicationic ionic liquid, 1-(1-methypiperidinium-1-yl) pentane-(1-pyridinium) bis(trifluoromethane)sulfonamide, and its binary mixtures with acetonitrile were investigated at temperatures from 293.15 K to 323.15 K over the entire range of composition at ambient pressure. From the experimental data, the excess molar volumes, dynamic viscosity deviations, and refractive index deviations were, respectively, calculated and correlated with Redlich-Kister-type polynomials. The temperature dependence of viscosity for pure ionic liquid was described by modified Vogel–Tammann-Fulcher (VTF) equation. The enthalpy and entropy of activation for viscous flow were obtained. The values of molar refraction and polarizability indicated that the dominant interactions between ionic liquids and acetonitrile were dipole-dipole molecular interactions. These thermodynamic properties were discussed in terms of nature of molecular interactions between the ionic liquid and acetonitrile.

A core-shell structured S-1@ZSM-5 was prepared by a seed-induced method, and its physicochemical properties and catalytic performance in methanol aromatization were studied based on the growth control of ZSM-5 shell. Results revealed that the ZSM-5 shell can be well coated on the surface of the silicalite-1 (S-1) seed under a low temperature crystallization, which induced the formation of worm-like morphology with a controllable shell thickness of 30 nm. Importantly, the weak acid sites dominated by Lewis acid sites can be constructed based on the control of growth degree. For methanol aromatization, the weak acid sites not only ensured the aromatization reaction, but also inhibited the isomerization and alkylation of para-xylene. The selectivity of aromatics and para-xylene in xylene reached 25.96% and 47.53%, respectively. The thin shell promoted the rapid removal of the coke precursor, which gave the catalyst a relatively long lifetime. Interestingly, on such a shell with weak acid sites, the limited sites can be easily covered by coke deposition along with the reaction progress, and thus the para-xylene selectivity can be gradually increased to 73.81%. Additionally, protective desilication was implemented for core-shell catalysts, and the aromatic selectivity was decreased while the catalytic lifetime and para-xylene selectivity remained unchanged. This reflected the inherent catalytic stability for methanol aromatization due to the thin shell and weak acidity. This work provides a new preparation strategy for ZSM-5 with high para-xylene selectivity and catalytic lifetime based on the direct synthesis of suitable acid sites and the diffusion channels.

Myeloperoxidase (MPO) plays critical role in the pathology of cerebral ischemia–reperfusion (I/R) injury via producing hypochlorous acid (HOCl) and inducing oxidative modification of proteins. High-mobility group box 1 (HMGB1) oxidation, particularly disulfide HMGB1 formation, facilitates the secretion and release of HMGB1 and activates neuroinflammation, aggravating cerebral I/R injury. However, the cellular sources of MPO/HOCl in ischemic brain injury are unclear yet. Whether HOCl could promote HMGB1 secretion and release remains unknown. In the present study, we investigated the roles of microglia-derived MPO/HOCl in mediating HMGB1 translocation and secretion, and aggravating the brain damage and blood-brain barrier (BBB) disruption in cerebral I/R injury. In vitro, under the co-culture conditions with microglia BV cells but not the single culture conditions, oxygen–glucose deprivation/reoxygenation (OGD/R) significantly increased MPO/HOCl expression in PC12 cells. After the cells were exposed to OGD/R, MPO-containing exosomes derived from BV2 cells were released and transferred to PC12 cells, increasing MPO/HOCl in the PC12 cells. The HOCl promoted disulfide HMGB1 translocation and secretion and aggravated OGD/R-induced apoptosis. In vivo, SD rats were subjected to 2 h of middle cerebral artery occlusion (MCAO) plus different periods of reperfusion. Increased MPO/HOCl production was observed at the reperfusion stage, accomplished with enlarged infarct volume, aggravated BBB disruption and neurological dysfunctions. Treatment of MPO inhibitor 4-aminobenzoic acid hydrazide (4-ABAH) and HOCl scavenger taurine reversed those changes. HOCl was colocalized with cytoplasm transferred HMGB1, which was blocked by taurine in rat I/R-injured brain. We finally performed a clinical investigation and found that plasma HOCl concentration was positively correlated with infarct volume and neurological deficit scores in ischemic stroke patients. Taken together, we conclude that ischemia/hypoxia could activate microglia to release MPO-containing exosomes that transfer MPO to adjacent cells for HOCl production; Subsequently, the production of HOCl could mediate the translocation and secretion of disulfide HMGB1 that aggravates cerebral I/R injury. Furthermore, plasma HOCl level could be a novel biomarker for indexing brain damage in ischemic stroke patients.

To recover isopropanol from its aqueous solution, alkanols (1-pentanol, 1-hexanol and 1-heptanol) were selected as the solvents to separate water and isopropanol. Liquid-liquid equilibrium data for water + isopropanol + 1-pentanol/1-hexanol/1-heptanol ternary systems were measured at 298.15 K under 101.3 kPa. The mass conservation of the experimental data was checked, and the reliability of the tie-lie data were evaluated by the Bachman, Hand and Othmer-Tobias equations. The extraction performance of the three solvents was assessed through the separation factor and partition coefficient, and compared with those for other reported solvents {esters, ketones, ethers, alkanes (or alkenes), ionic liquids and deep eutectic solvents}, indicating that 1-heptanol has better extraction performance for isopropanol compared with other two solvents. The NRTL and UNIQUAC models were used to correlate the data to obtain regression parameters. The mixed surface analysis based on Gibbs energy topology verified that the binary interaction parameters are consistent with the measured data. Furthermore, the LLE binodal curve for the researched ternary systems were predicted through the UNIFAC and COSMO-UNIFAC models.

1-Hexene epoxidation is difficult to achieve for the low electron cloud density of long-chain terminal olefin double bonds structure; the development of efficient and stable catalysts is the key to achieving the highly efficient epoxidation of 1-hexene. The titanosilicate/H2O2 system has been the most promising and environmentally friendly epoxidation route in recent research, and the WOx@HTS-1 nanoparticles were successfully prepared by an etching–impregnation–recrystallization strategy in this work. Unique structures of the WOx@HTS-1 nanoparticles could achieve the effects of “killing three birds with one stone” for 1-hexene epoxidation. I. Hierarchical structure and “hollow nests” could significantly enhance the accessibility of the active sites, shorten the transmission path, and improve the diffusion efficiency of the reactants and products. II. Titanium active centers and metal oxide active sites had significant and efficient synergistic catalytic effects on 1-hexene epoxidation. III. Fine WOx particles were highly dispersed on the hollow nests of WOx@HTS-1 nanoparticles, which prevented the sintering and poisoning of WOx clusters during the reaction process and ensured excellent stability of the catalysts by the restricted-domain effect. Attractively, 1-hexene conversion and 1,2-epoxyhexane selectivity were more than 90 and 96% with WOx@HTS-1 nanoparticles and H2O2, respectively, which provided many opportunities for the directional design of nanoparticles and their application in emerging catalytic fields.

Destruction of the blood-brain barrier is a critical component of epilepsy pathology. Several studies have demonstrated that sphingosine 1-phosphate receptor 1 contributes to the modulation of vascular integrity. However, its effect on blood-brain barrier permeability in epileptic mice remains unclear. In this study, we prepared pilocarpine -induced status epilepticus models and pentylenetetrazol -induced epilepsy models in C57BL/6 mice. S1P1 expression was increased in the hippocampus after status epilepticus, whereas tight junction protein expression was decreased in epileptic mice compared with controls. Intraperitoneal injection of SEW2871, a specific agonist of sphingosine-1-phosphate receptor 1, decreased the level of tight junction protein in the hippocampus of epileptic mice, increased blood-brain barrier leakage, and aggravated the severity of seizures compared with the control. W146, a specific antagonist of sphingosine-1-phosphate receptor 1, increased the level of tight junction protein, attenuated blood-brain barrier disruption, and reduced seizure severity compared with the control. Furthermore, sphingosine 1-phosphate receptor 1 promoted the generation of interleukin-1β and tumor necrosis factor-α and caused astrocytosis. Disruption of tight junction protein and blood-brain barrier integrity by sphingosine 1-phosphate receptor 1 was reversed by minocycline, a neuroinflammation inhibitor. Behavioral tests revealed that sphingosine 1-phosphate receptor 1 exacerbated epilepsy -associated depression-like behaviors. Additionally, specific knockdown of astrocytic S1P1 inhibited neuroinflammatory responses and attenuated blood-brain barrier leakage, seizure severity, and epilepsy -associated depression-like behaviors. Taken together, our results suggest that astrocytic sphingosine 1-phosphate receptor 1 exacerbates blood-brain barrier disruption in the epileptic brain by promoting neuroinflammation .

Cocrystallization between a BCS II class drug, carbamazepine , and polyphenolic compounds such as naringenin and naringin , is performed in the present study. New carbamazepine-naringenin (CBZ-NRG) cocrystal with 1: 1 molar ratio was obtained and characterized by using the methods of X-ray diffraction, nuclear magnetic resonance (solid state 13 C-NMRand liquid 1 H NMR), thermal and spectral analyses (FT-IR and solid state fluorescence). CBZ-NRG crystallizes in the P 2 1 /c space group, which belongs to monoclinic system. The molecular interactions were elaborated in the packing diagrams and confirmed by solid state 13 C NMR and FT-IR methods. It is scarce that CBZ-NRG cocrystal presents higher endotherm ( T onset = 272.7 °C) than both starting materials ( T onset = 188.6, 248.6 °C) using DSC analysis. The fluorescence values of CBZ-NRG cocrystal (179 A.U.) decreases significantly in comparison with CBZ (6121 A.U.). The in vitro/vivo assessment of NRG effect on CBZ was conducted in the present study. Compared with CBZ (97.2 ± 1.5 μg/mL) in water the solubility of CBZ-NRG cocrystal decreases to 7.2 ± 0.6 μg/mL, the same trends in pH = 1.2 and 6.8 buffer solution. The IDR values of CBZ and CBZ-NRG cocrystal are 0.042 and 0.016 mg/cm 2 /min respectively. Interestingly, CBZ has one absorption peak while the CBZ-NRG cocrystal has double absorption peaks in plasma concentration-time curves. The peak concentration ( C max = 491.3 ± 97.6 ng/mL) of CBZ-NRG cocrystal decreases with longer elimination half life ( t 1/2 = 8.5 ± 1.0 h) in comparison with CBZ ( C max = 5258.1 ± 904.2 ng/mL, t 1/2 = 0.7 ± 0.2 h). The obtained results suggest that CBZ-NRG cocrystal presents different in vitro/vivo performance in comparison with CBZ.

This study presents a novel approach in the realm of catalytic organic synthesis by integrating biomass catalytic conversion with organic synthesis techniques. UtilizingN-acetylglucosamine as the primary feedstock, the first phase of the research involves its catalytic transformation into 5-hydroxymethylfurfural (HMF). The subsequent phase employs a condensation reaction between HMF and 3,3-Dimethyl-2-butanone to synthesize a new compound, (E)-1-(5-(hydroxymethyl) furan-2-yl)-4,4-dimethylpent-1-en-3-one. This two-step process not only demonstrates the feasibility of converting biomass into valuable chemical precursors but also exemplifies the synthesis of novel compounds through green chemistry principles. The successful execution of this methodology offers fresh insights and opens new avenues for advancements in catalytic organic synthesis, emphasizing sustainability and efficiency.

Plate-like TS-1 zeolites offering a short diffusion pathway along the b- axis are highly desirable catalysts in the reaction of alkene epoxidation. Herein, we successfully developed a simple synthetic strategy toward reducing the thickness along the b -axis of TS-1 zeolite crystals, l -lysine adopted as a zeolite growth modifier was firstly proposed to prepare plate-like TS-1 zeolite. Meanwhile, through tuning the preliminary aging time, plate-like TS-1 zeolites with thickness in the range of 80–1000 nm can be controllable obtained. In this work, the synthesis parameters controlling the formation of plate-like TS-1 zeolites are studied, and the factors controlling the zeolite growth were well elucidated. Compared with the conventional TS-1 zeolite as well as the plate-like TS-1 synthesized in the fluoride-containing medium, the obtained plate-like TS-1 shows improved catalytic performances in catalyzing epoxidation of 1-hexene.

Tumor-associated macrophages (TAMs) are vital in the tumor microenvironment, contributing to immunosuppression and therapy tolerance. Despite their importance, the precise re-education of TAMs in vivo continues to present a formidable challenge. Moreover, the lack of real-time and efficient methods to comprehend the spatiotemporal kinetics of TAMs repolarization remains a significant hurdle, severely hampering the accurate assessment of treatment efficacy and prognosis. Herein, we designed a metal–organic frameworks (MOFs) based Caspase-1 nanoreporter (MCNR) that can deliver a TLR7/8 agonist to the TAMs and track time-sensitive Caspase-1 activity as a direct method to monitor the initiation of immune reprogramming. This nanosystem exhibits excellent TAMs targeting ability, enhanced tumor accumulation, and stimuli-responsive behavior. By inducing the reprogramming of TAMs, they were able to enhance T -cell infiltration in tumor tissue, resulting in inhibited tumor growth and improved survival in mice model. Moreover, MCNR also serves as an activatable photoacoustic and fluorescent dual-mode imaging agent through Caspase-1-mediated specific enzyme digestion. This feature enables non-invasive and real-time antitumor immune activation monitoring. Overall, our findings indicate that MCNR has the potential to be a valuable tool for tumor immune microenvironment remodeling and noninvasive quantitative detection and real-time monitoring of TAMs repolarization to immunotherapy in the early stage.
