Calycosin‑7‑O‑β‑D‑glucoside (CG), a major active ingredient of Astragali Radix, exerts neuroprotective effects against cerebral ischemia; however, whether the effects of CG are associated with mitochondrial protection remains unclear. The present study explored the role of CG in improving mitochondrial function in a HT22 cell model of oxygen‑glucose deprivation/reperfusion (OGD/R). The Cell Counting Kit‑8 assay, flow cytometry, immunofluorescence and western blotting were performed to investigate the effects of CG on mitochondrial function. The results demonstrated that mitochondrial function was restored after treatment with CG, as indicated by reduced mitochondrial reactive oxygen species levels, increased mitochondrial membrane potential and improved mitochondrial morphology. Overactivated mitophagy was revealed to be inhibited by the regulation of proteins involved in fission [phosphorylated‑dynamin‑related protein 1 (Drp1) and Drp1] and mitophagy (LC3, p62 and translocase of outer mitochondrial membrane 20), and mitochondrial biogenesis was demonstrated to be enhanced by increased levels of sirtuin 1 (SIRT1) and peroxisome proliferator‑activated receptor γ coactivator‑1α (PGC‑1α). In addition, neuronal apoptosis was ameliorated by CG, as determined by a decreased rate of apoptosis, and levels of caspase‑3 and Bcl‑2/Bax. In conclusion, the present study demonstrated that CG may alleviate OGD/R‑induced injury by upregulating SIRT1 and PGC‑1α protein expression, and reducing excessive mitochondrial fission and overactivation of mitophagy.

7, 8-Dihydroxyflavone (7, 8-DHF) is an effective drug for the treatment of Parkinson’s disease. However, its application is heavily limited by its poor water solubility. To improve its hydrophilicity, biocompatible poly (ethylene glycol)acrylate (PEG-A) was grafted onto 7, 8-DHF via in situ reversible addition-fragmentation chain transfer (RAFT) polymerization, forming a kind of a drug/polymer composite, DHF/PPEG-A. It was found that the solubility of DHF/PPEG-A was significantly improved by 78 times compared to the unmodified precursor. Impressively, both the cytotoxicity and the drug efficacy of the synthesized DHF/PPEG-A just slightly decreased compared with the unmodified 7, 8-DHF. Moreover, the drug efficacy of DHF/PPEG-A can be regained by the hydrolysis in the presence of the esterase D due to the decomposition to 7, 8-DHF in body. This dramatical improvement of the water solubility of DHF/PPEG-A and the insignificant compromise of its drug cytotoxicity could envision extensive practical clinical administration of 7, 8-DHF for enhanced drug efficacy.

A fixed time point on the curve of the absorbance variation with time determined in the hydroxyl polyaluminum solution by 7-iodine-8-hydroxyquinoline-5-sulfonic acid-timed spectrometry is usually used to quantify the mononuclear aluminum species in the hydroxyl polyaluminum solution. The right choice of the determining time of mononuclear Al must be established on the basis of the sufficient reaction between chromogenic reagent and the hydroxyl polyaluminum solution. But this question has never been noted, which resulted in the confusion of application and research of this assay. In this work, the effect of variation of the determining time on the accurate determination of mononuclear Al in the hydroxyl polyaluminum solution was first studied. The experimental results showed that 60 s was the best under the appropriate dosage of 7-iodine-8-hydroxyquinoline-5-sulfonic acid. The reasonableness of 60 s was further verified by 27Al nuclear magnetic resonance spectrometer and the absorbance–time curve obtained from the pure mononuclear Al solution with pH < 2. The above conclusion was further used to analyze the results reported in literature. The mononuclear Al determined at 60, >60, and <60 s in the literature were comparable to the corresponding results of this study.

Nearly monodispersed spherical silver nanoparticles (Ag NPs) were synthesized by using tannic acid (TA) as both reductant and stabilizer in a 30 °C water bath. The size of the as-prepared Ag NPs could be tuned in a range of 7–66 nm by changing the molar ratio of TA to silver nitrate and pH of the reaction solutions. UV–vis spectra, TEM observations, and temporal evolution of the monomer concentrations for the reactions carried out at different experimental conditions showed that the improved size distribution and size tunability of the Ag NPs were mainly attributed to the use of TA, which could promote the balance of nucleation and growth processes of the NPs effectively. The size of the Ag NPs was extendable up to 200 nm in one-pot fashion by the multi-injection approach. The size-dependent surface-enhanced Raman scattering (SERS) activity of the as-prepared Ag NPs was evaluated, and the NPs with size around 100 nm were identified to show a maximum enhanced factor of 3.6 × 105. Moreover, the as-prepared TA-coated Ag NPs presented excellent colloidal stability compared to the conventional citrate-coated ones.

Mesoporous silica nanoparticles (MSNs) are promising solid supports for controlled anticancer drug delivery. Herein, we report biocompatible layer-by-layer (LbL) coated MSNs (LbL-MSNs) that are designed and crafted to release encapsulated anticancer drugs, e.g., doxorubicin hydrochloride (DOX), by changing the pH or by adding competitive agents. The LbL coating process comprises bis-aminated poly(glycerol methacrylate)s (BA-PGOHMAs) and cucurbit[7]uril (CB[7]), where CB[7] serves as a molecular bridge holding two different bis-aminated polymeric layers together by means of host–guest interactions. This integrated nanosystem is tuned to respond under specific acidic conditions or by adding adamantaneamine hydrochloride (AH), attributed to the competitive binding of hydronium ions or AH to CB[7] with BA-PGOHMAs. These LbL-MSN hybrids possess excellent biostability, negligible premature drug leakage at pH 7.4, and exceptional stimuli-responsive drug release performance. The pore sizes of the MSNs and bis-aminated compounds (different carbon numbers) of BA-PGOHMAs have been optimized to provide effective integrated nanosystems for the loading and release of DOX. Significantly, the operating pH for the controlled release of DOX matches the acidifying endosomal compartments of HeLa cancer cells, suggesting that these hybrid nanosystems are good candidates for autonomous anticancer drug nanocarriers actuated by intracellular pH changes without any invasive external stimuli. The successful cellular uptake and release of cargo, e.g., propidium iodide (PI), in human breast cancer cell line MDA-231 from PI-loaded LbL-MSNs have been confirmed by confocal laser scanning microscopy (CLSM), while the cytotoxicities of DOX-loaded LbL-MSNs have been quantified by the Cell Counting Kit-8 (CCK-8) viability assay against HeLa cell lines and fibroblast L929 cell lines. The uptake of DOX-loaded LbL-MSNs by macrophages can be efficiently reduced by adding biocompatible hydrophilic poly(ethylene glycol) or CB[7] without destroying the capping. In vivo tumor-growth inhibition experiments with BALB/c nude mice demonstrated a highly efficient tumor-growth inhibition rate of DOX-loaded LbL-MSNs, suggesting that the novel type of LbL-MSN materials hold great potentials in anticancer drug delivery.

The environmental impact caused by azo dyes such as acid orange 7 (AO7) is currently a major concern, mainly due to their toxicity, potential carcinogenic nature and difficult degradation. This research described the heterogeneous reactions of persulfate with milli-ZVI (1 mm), micro-ZVI (150 μm) and nano-ZVI (50 nm) under various experimental conditions. The results demonstrated that the combination of persulfate with ZVI was efficient in promoting the degradation of AO7 with the pseudo first-order and second-order kinetics reaction, and different ZVI types exhibited respective level of ability to activate persulfate to degrade acid orange 7, which was ranged as nano-ZVI > micro-ZVI > milli-ZVI. However, the presence of dissolved SiO 2 decreased the AO7 degradation ratio and prolonged the persulfate half-life. The iron corrosion products coating on the ZVI after reaction was comprised of α-Fe 2 O 3 , Fe 3 O 4 and α-FeOOH with some Fe 8 O 8 (OH) 6 SO 4 , which was identified by Raman spectroscopy, X-ray diffraction (XRD) and Fourier transform infrared (FTIR). With the presence of SiO 2 , surface complexes of ≡FeH 3 SiO 4 or ≡ Fe H 2 SiO 4 - for binding of silicates were formed. These observations indicated that the dissolved SiO 2 in the ZVI activated persulfate system affected the corrosion of iron, altered the surface area with the adsorption of SiO 2 , and appeared to influence the decomposition of persulfate and AO7.

Ethnopharmacological relevance Viral pneumonia is the leading cause of death after SARS-CoV-2 infection. Despite effective at early stage, long-term treatment with glucocorticoids can lead to a variety of adverse effects and limited benefits. The Chinese traditional herb Pogostemonis Herba is the aerial part of Pogostemon Cablin (Blanco) Benth., which has potent antiviral, antibacterial, anti-inflammatory, and anticancer effects. It was used widely for treating various throat and respiratory diseases, including COVID-19, viral infection, cough, allergic asthma, acute lung injury and lung cancer. Aim of the study To investigate the antiviral and anti-inflammatory effects of chemical compounds from Pogostemonis Herba in SARS-CoV-2-infected hACE2-overexpressing mouse macrophage RAW264.7 cells and hACE2 transgenic mice. Materials and methods The hACE2-overexpressing RAW264.7 cells were exposed with SARS-CoV-2. The cell viability was detected by CCK8 assay and cell apoptotic rate was by flow cytometric assay. The expressions of macrophage M1 phenotype markers (TNF-α and IL-6) and M2 markers (IL-10 and Arg-1) as well as the viral loads were detected by qPCR. The mice were inoculated intranasally with SARS-CoV-2 omicron variant to induce viral pneumonia. The levels of macrophages, neutrophils, and T cells in the lung tissues of infected mice were analyzed by full spectrum flow cytometry. The expressions of key proteins were detected by western blot assay. Results Diosmetin-7-O-β-D-glucopyranoside (DG) presented the strongest anti-SARS-CoV-2 activity. Intervention with DG at the concentrations of 1.25-50 μM not only reduced the viral replication, cell apoptosis, and the productions of inflammatory cytokines (IL-6 and TNF-α) in SARS-CoV-2-infected RAW264.7 cells, but also reversed macrophage polarity from M1 to M2 phenotype. Furthermore, treatment with DG (25-100 mg/kg) alleviated acute lung injury, and reduced macrophage infiltration in SARS-COV-2-infected mice. Mechanistically, DG inhibited SARS-COV-2 gene expression and HK3 translation via targeting YTHDF1, resulting in the inactivation of glycolysis-mediated NF-κB pathway. Conclusions DG exerted the potent antiviral and anti-inflammatory activities. It reduced pneumonia in SARS-COV-2-infected mice via inhibiting the viral replication and accelerating M2 macrophage polarization via targeting YTHDF1, indicating its potential for COVID-19 treatment.

A hollow porous molecularly imprinted polymer (HPMIP) is described for use in dispersive solid phase extraction of macrolide antibiotics (MACs). The HPMIP was prepared by using spiramycin as the template, methacrylic acid as the functional monomer, and mesoporous MCM-41 (Mobile Composition of Matter No. 41; with a size of about 100 nm) as a sacrificial support. The sorbent was characterized by Fourier transform infrared spectrometry, transmission electron microscopy, nitrogen adsorption and thermo-gravimetric analysis. Several parameters affecting the extraction efficiency were optimized. The material has a large surface area (359 m 2 ·g −1 ), and most recognition sites are located on the surface of the HPMIPs. This results in high binding capacity (120 μmol·g −1 ) and fast binding (20 min) in comparison to either MCM-41-core surface MIPs or solid MIPs. The method was applied to the extraction of the MACs azithromycin, spiramycin, tilmicosin, tylosin, clarithromycin, roxithromycin and josamycin from spiked honey. The recoveries, determined by HPLC-MS/MS, ranged from 88.0% to 117% at the three spiking levels tested (1, 5 and 20 μg·kg −1 ). Intra-day and inter-day assay precision at three spiking levels are <10.7% (for n = 6) and 12.6% ( n = 3), respectively. The limits of detection are between 3 and 17 ng·kg −1 . This indicates the superiority of the method in selective extraction of macrolides even from complex matrices.
Hollow porous molecularly imprinted polymers using spiramycin as the template are shown to be viable dispersive solid phase extraction adsorbents for selective enrichment of macrolide antibiotics in honey products prior to their quantitation by HPLC-MS/MS.

A high-performance ultraviolet (UV) nonlinear optical (NLO) material requires three essential characteristics: a short UV absorption edge, a strong secondharmonic generation (SHG) response, and favorable NLO coefficients. In this study, a novel rare-earth borate compound with the formula K 7 MgY 2 (B 5 O 10 ) 3 was successfully synthesized using a high-temperature solution method combined with chemical cosubstitution. The crystal adopts a non-centrosymmetric (NCS) structure (space group R32) and exhibits an SHG intensity 1.3 times that of potassium dihydrogen phosphate (KDP) under 1064 nm laser irradiation. Its UV cutoff edge is determined to be 290 nm, demonstrating exceptional potential for UV optical applications. Combined experimental and theoretical analyses reveal that the synergistic interplay between polarizable [B 5 O 10 ] groups and heterovalent K + /Mg 2+ /Y 3+ substitution underpins its superior NLO performance.

Antimicrobial peptide BmKn2-7, a rationally optimized derivative of the scorpion venom peptide BmKn2, shows strong potential as an antibiotic alternative for controlling Vibrio parahaemolyticus in aquaculture. This study evaluated its antibacterial mechanisms and in vivo efficacy in Litopenaeus vannamei through biochemical assays, transcriptomic profiling, and challenge experiments. BmKn2-7 exhibited potent in-vitro activity (MIC = 125 µg/mL) and disrupted bacterial homeostasis by increasing membrane permeability, inducing oxidative stress, and inhibiting P-type ATPase. RNA-seq analysis revealed extensive transcriptional reprogramming, including the downregulation of quorum-sensing, virulence, and β-lactam resistance–associated genes. In vivo, BmKn2-7 significantly improved shrimp survival following V. parahaemolyticus infection and selectively reduced Vibrio abundance in the gut without compromising overall microbiota diversity. Together, these findings demonstrate that BmKn2-7 acts through complementary bactericidal and anti-virulence pathways while promoting intestinal microbial resilience, supporting its potential as a promising antimicrobial candidate for reducing vibriosis risk in shrimp aquaculture.

Monodisperse polyethylene glycol (PEG) derivatives offer significant advantages over conventional polydisperse PEGs for biomedical applications due to their precisely defined molecular structures. This study establishes an eco-efficient, selective, and sensitive analytical assay integrating microscale solid-phase extraction (μ-SPE) with ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) to investigate the cellular uptake of HO-PEG 8 -OH polymers in MCF-7 cells. The method achieved greater than 91% recovery from 20 μL lysates using M-PEG 6 -OH as the internal standard, with validated linearity (10–1000 ng/mL, R > 0.997), accuracy (relative error < ± 7.49%), and precision (RSD < 7.50%). The novelty of the assay is the harmonization of analytical performance with green analytical chemistry principles. The validated method provides a robust platform for studying monodisperse PEG derivatives while addressing growing demands for sustainable analytical technologies. Green analytical chemistry metric assessments confirmed the environmental sustainability of the method. Cellular pharmacokinetic analysis revealed time-/concentration-dependent uptake kinetics of HO-PEG 8 -OH polymers in MCF-7 cells, showing 3.2-fold accumulation between 0.5 and 48 h exposure. These findings offer important insights for PEG-based drug delivery system optimization and establish a new standard for environmentally conscious bioanalytical method development. Graphical

Although antioxidants such as vitamin E (VE) and tea polyphenols have been employed to enhance the oxidation resistance of ultra-high molecular weight polyethylene (UHMWPE) for artificial joint applications, the long-term oxidative behavior of tea polyphenol-stabilized, crosslinked UHMWPE under real-time shelf aging remains insufficiently understood. In this study, the performance evolution of crosslinked UHMWPE containing VE, epigallocatechin gallate (EGCG), and lipid-soluble EGCG ( ls EGCG) was investigated after 7 years of real-time shelf storage. Notably, the EGCG-stabilized UHMWPE exhibited a markedly higher crosslink density (≥ 40% increase compared with the VE group) and superior tribological performance, with the coefficient of friction reduced by 16‒35%. Furthermore, it reduced the fraction of submicron wear particles (≤ 1 µm), lowering the potential risk of osteolytic inflammation. The enhanced stabilization efficacy of tea polyphenols, particularly EGCG, originates from their polyphenolic hydroxyl groups, which could effectively quench free radicals and preserve crosslink network during long-term storage. Overall, these findings demonstrate that tea polyphenols represent a durable and effective antioxidant strategy for UHMWPE, with significant potential to extend the clinical service life of joint implants.
