The development of therapeutic strategies capable of achieving functional cure remains an unmet need in chronic hepatitis B management. Class A capsid assembly modulators (CAM-As) emerge as a promising treatment option. CAM-As not only directly disrupt the normal assembly of capsids, but some of which have also been reported to activate the innate immune response in animal models with unclear mechanisms. GLS4 is one of CAM-As with great potential. In this study, we investigate its capacity to activate immune response both in vitro and in vivo, as well as the underlying mechanisms involved. GLS4 activates the RIG-I-mediated interferon signaling pathway in both HBV-expressing hepatocellular carcinoma cell lines and HBV carrier mouse models, as indicated by RNA-seq. Besides, combination treatment with GLS4 and ritonavir elevates the frequencies of both peripheral blood IFNγ + NK cells and liver-resident IFNγ + CD8+ T cells in the pAAV/HBV1.2 hydrodynamic injection (HDI) model. In conclusion, our study reveals a previously unknown mechanism by which GLS4 activates the interferon signaling pathway in HBV-expressing hepatocytes. Furthermore, GLS4 partially restores innate and adaptive immunity in vivo. This signifies a potentially effective strategy for achieving a functional cure for HBV infection.

A new Rhodamine B-based fluorescent probe (RBO) is successfully designed and synthesized, which is a higher selective and sensitive chemosensor for Cu 2+ than other ions. Under physiological conditions (pH = 7.0), the non emission RBO displays a rapid fluorescence increase together with a color change after addition of Cu 2+ and the detection limit is down to 28 nM, which can clearly illustrate the distribution of Cu 2+ with the help of laser scanning confocal microscope in plant tissues. Eventually, it confirmed that the Cu 2+ accumulates mostly in the vascular cylinder and very less in the epidermal cells of maize roots, which is important to understand how the plants take up, transport and store in the Cu 2+ .

Enterovirus infections are life-threatening viral infections which occur mainly among children and are possible causes of viral outbreak. Until now, treatment and management of infections caused by members of the genus Enterovirus largely depended on supportive care, and no antiviral medications are currently approved for the treatment of most of these infections. The urgency of discovering new therapeutic options for the treatment of enterovirus infection is increasing. In the present study, we identified that trans-2-hexenoic acid (THA), a natural product from a dietary source, possesses antiviral activity against coxsackievirus B (CVB) and enterovirus A71 (EV-A71) in a dose-dependent manner. We found that THA possesses antiviral activity at 50% effective concentrations (EC50) of 2.9 μM and 3.21 μM against CVB3 and EV-A71 infections, respectively. The time of addition assay revealed that THA inhibits both CVB3 and EV-A71 replication at the entry stage of infection. Additional results from this study further suggest that THA inhibits viral replication by blocking viral entry. Given that THA has received approval as a food additive, treatment of enterovirus infections with THA might be a safe therapeutic option or could pave the way for semisynthetic manufacturing of more antiviral drugs in the future.

In this study, one well-known CHM residue (Atropa belladonna L., ABL) was used to prepare biochar capable of adsorbing rhodamine B (RhB) with an ultrahigh surface area for the first time. Three micropore-rich ABL biochars including ABL@ZnCl2 (1866 m2/g), ABL@H3PO4 (1488 m2/g), and ABL@KOH (590 m2/g) were obtained using the one-step carbonization method with activation agents (ZnCl2, H3PO4, and KOH) via chemical activation and carbonization at 500 °C, and their adsorption performance for RhB was systematically studied with adsorption kinetics, isotherms, and thermodynamics. Through pore diffusion, π–π interaction, and hydrogen bonding, ABL biochar had excellent adsorption performance for RhB. Moreover, when C0 was 200 mg/L, biochar dosage was 1 g/L, and the contact time was 120 min; the maximum RhB adsorption capacity and removal efficiency on ABL@ZnCl2 and ABL@H3PO4 were 190.63 mg/g, 95% and 184.70 mg/g, 92%, respectively, indicating that it was feasible to prepare biochar from the ABL residue for RhB adsorption. The theoretical maximum adsorption capacities of ABL@ZnCl2 and ABL@H3PO4 for RhB were 263.19 mg/g and 309.11 mg/g at 25 °C, respectively. Furthermore, the prepared biochar showed good economic applicability, with pay back of USD 972/t (ABL@ZnCl2) and USD 987/t (ABL@H3PO4), respectively. More importantly, even after five cycles, ABL@H3PO4 biochar still showed great RhB removal efficiency, suggesting that it had a good application prospect and provided a new method for the resource utilization of traditional CHM residues. Additionally, pore diffusion, π–π interactions, and hydrogen bonding all play roles in the physical adsorption of RhB on ABL biochar. π–π interactions dominated in the early stage of RhB adsorption on ABL@H3PO4, while pore diffusion played a crucial role in the whole adsorption process on both adsorbents.

BiFeO 3 nanoflakes (BFO NFs) were synthesized via a facile hydrothermal route. The crystal structure, morphology, specific surface area and optical properties of the flakes were characterized by X-Ray Diffraction (XRD), Brunner–Emmet–Teller measurements (BET) and UV–vis Diffuse Reflectance Spectrum (UV–vis DRS). The results showed that the thickness and diameter of BFO NFs are 100–150 nm and 1.0–1.2 μm, respectively. Moreover, the results of UV–vis DRS indicated that BFO NFs exhibits good response to the visible light and the bandgap was calculated to be 2.2 eV according to the Kubelka–Munk theory. The photo-Fenton performance of BFO NFs was obviously enhanced due to the piezoelectric effect for the degradation efficiency of Rhodamine B (Rh B) (10 mg L −1 ) within 60 min and greatly elevated from 72 to 97%. The intermediate active species of ·O 2− and ·OH were detected by radical capture experiments and Electron Spin Resonance (ESR). The possible catalytic mechanism was proposed based on the above experiments results. In previous researches about photo-Fenton reaction, the piezoelectric properties of BFO NFs were rarely mentioned. This study represents a potential approach toward waste water treatment.

Since hepatitis B virus (HBV) infection can cause chronic viral hepatitis, increase the risk of liver fibrosis and cirrhosis, and ultimately result in hepatoma, it is of great significance to explore convenient, rapid, and sensitive diagnostic strategies for HBV DNA detection. In this study, we developed a novel and sensitive fluorescent biosensor for HBV detection based on the sensing system consisting of CRISPR-Cas12a enzymes and metal nanoclusters as the luminescent nanoprobes, involving gold nanoclusters (AuNCs), silver nanoclusters (AgNCs), and copper nanoclusters (CuNCs). The DNA probes could effectively be applied as the template for the preparation of fluorescent metal nanoclusters. In the presence of the HBV target, the trans-cleavage capability of Cas12a was able to be initiated to degrade DNA probes, which brought about the inhibition of metal nanoclusters formation, thereby giving minimal fluorescence signals. In comparison with AuNCs and AgNCs, the CuNCs-based nanosensor exhibited much higher sensitivity. In addition, the fluorescence response of CuNCs to HBV detection was significantly faster, which could be completed within 25 min without the requirements of sophisticated equipment or harsh reaction conditions. Moreover, the practical application of this analysis method was also demonstrated to detect HBV DNA target in human serum samples with satisfactory recovery, which was promising for simple, rapid, sensitive, and label-free HBV DNA bioanalysis .

We explored the biological role of long non-coding RNA (lncRNA) MAPKAPK5_AS1 (MAAS) and the mechanism of its differential expression in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). Differentially expressed lncRNAs in HBV-related HCC were determined using bioinformatics analysis. Gain-of-function experiments were conducted to evaluate the effect of MAAS on cell proliferation. A xenograft model was established for in vivo experiments. Dual-luciferase reporter assays, chromatin immunoprecipitation, co-immunoprecipitation, and methylated RNA immunoprecipitation were performed to elucidate the underlying molecular mechanisms. MAAS was upregulated in HBV-related HCC cancerous tissues and its high expression was closely related to the poor survival probability of patients. Functional assays revealed that MAAS overexpression facilitated the proliferation of HBV + HCC cells in vitro and in vivo. Mechanistically, MAAS promoted the MYC proto-oncogene (c-Myc)-induced transcriptional activation of cyclin-dependent kinase 4 (CDK4), CDK6, and S-phase kinase associated protein 2 via stabilizing c-Myc protein, thereby facilitating G1/S transition. The latter contributed to the paradoxical proliferation of HBV + HCC cells. Although MAAS was upregulated in HBV-related HCC cancerous tissues, it was highly expressed in M2 macrophages, a major phenotype of tumor-associated macrophages in HBV-related HCC, instead of in HBV + HCC cells. HBeAg, an HBV-associated antigen, further elevated the MAAS level in M2 macrophages by enhancing the methyltransferase-like 3-mediated N6-methyladenosine modification of MAAS. The increased MAAS in the M2 macrophages was then transferred to HBV + HCC cells through the M2 macrophage-derived exosomes, promoting cell proliferation. Our findings show that HBV + HCC cell-secreted HBeAg upregulates MAAS expression in M2 macrophages by affecting its m 6 A modification. The upregulated MAAS is then transferred to HBV + HCC cells via exosomes, facilitating the proliferation of HBV + HCC cells by targeting c-Myc.

TiO 2 based photocatalyst with sufficient reusability for the degradation of water pollutants remains a challenge. Here, we report a composite chitosan-based aerogel containing TiO 2 nanoparticles , multiwalled carbon nanotubes and layered silicate rectorite with sufficient mechanical strength for Rhodamine B degradation. The aerogels with homogeneous oriented lamellar structure were successfully prepared via a unidirectional freeze-casting technique. As-prepared aerogels showed specific surface area of 84.59 m 2 /g. The addition of rectorite and carbon nanotubes accelerated the photodegradation and rectorite significantly enhanced the overall mechanical performance. Rapid degradation of rhodamine B (95% in 100 min) was observed on aerogels with 2 wt% and 4 wt% of rectorite. After 3 cycles, 75% degradation of Rhodamine B was achieved with CTCR4, confirming its reusability. Thus, the composite chitosan aerogels show high photocatalytic degradation efficiency towards Rhodamine B during cycle use.

The activation of Type III heterojunctions through inserting plasmonic metal nanoparticles (NP) between the semiconductor components as a bridge for the selective capture and interband transfer of the photothermal-induced charges and constructing a robust Type B heterojunction photothermocatalyst shows the potential to overcome the trade-off between light response range and redox potentials of the mainstream Type II, Z-scheme, and S-scheme heterojunctions. A series of Type B heterojunctions CuWO4/Ag/GdCrO3, WO3/Ag/GdCrO3, and Bi2WO6–xF2x/Ag/GdCrO3 are constructed through introducing a plasmonic Ag NP bridge, and the relation between the required plasmonic intensity of the bridge and energy barrier of the parent heterojunctions is correlated. The as-developed Type B heterojunction WO3/Ag/GdCrO3 exhibits a 73 times higher photothermocatalytic toluene degradation rate and a 44 times higher photothermocatalytic CO yielding rate from CO2 reduction than those of parent Type III WO3/GdCrO3, and ranges among the best of state-of-the-art photocatalytic or photothermocatalytic performance in both VOC oxidation and CO2 reduction.

Introduction and objectives The activation of hepatic stellate cells (HSCs) is the main cause of liver fibrosis. The beneficial effects of fibroblast growth factor (FGF) 19 on liver fibrosis were recently reported. The S. miltiorrhiza as well as S. miltiorrhiza derived bioactive chemical components has shown prominent antifibrotic effects in liver fibrosis but the mechanism is still not fully understood. We aimed to investigate the bioactive compounds derived from S. miltiorrhiza which exerts antifibrotic effects in HSCs via regulating FGF19. Materials and methods FGF19 level in culture media was determined by enzyme-linked immunosorbent assay. Cell proliferation was measured by Cell Counting Kit-8 assay. Further, mRNA and protein expressions were assessed by quantitative polymerase chain reaction and western blotting, respectively. Knocking down of FGF receptor 4 (FGFR4) by transfection with siRNA was used to confirm the role of FGF19/FGFR4 signaling. Results Using the human HSC cell line LX-2, we screened several natural products and found that bioactive compounds isolated from Salvia miltiorrhiza , particularly salvianolic acid B, strongly upregulated FGF19 secretion by LX-2 cells. We further showed that salvianolic acid B inhibited lipopolysaccharide (LPS)-induced HSC proliferation and activation. LPS treatment may also reduce the mRNA and protein levels of FGF19 and its receptor FGFR4. Salvianolic acid B treatment restored the impaired expressions of FGF19 and FGFR4. Finally, FGFR4 knockdown abolished the antifibrotic effects of salvianolic acid B in the LPS-induced HSC activation model. Conclusions Salvianolic acid B prevented LPS-induced HSC proliferation and activation by enhancing antifibrotic FGF19/FGFR4 signaling.

The electrochemical CO2 reduction reaction (CO2RR) on Pd-based electrodes to dissolved formate and/or gaseous CO is largely dependent on potential and the electrode material, yet there is a lack of molecular-level insights into this dependence. Herein, in situ attenuated total reflection surface enhanced infrared absorption spectroscopy (ATR-SEIRAS) in conjunction with differential electrochemical mass spectrometry (DEMS), gas chromatography (GC), and nuclear magnetic resonance (NMR) measurements is applied to investigate the CO2RR on Pd and Pd-B film electrodes, providing a direct observation of the role of surface CO as well as the B-doping effect at varied potentials. Comprehensive spectrometric results reveal that at lower overpotentials, CO gradually accumulates on both Pd electrode surfaces poisoning the dominant formate pathway, while at higher overpotentials, surface CO forms facilely with linearly bonded CO (the minor surface CO species) acting as an active precursor and bridge-bonded CO (the major surface CO species) as a spectator toward the gaseous CO product. Moreover, B-doping in Pd hinders CO formation and promotes formate production on the Pd-B electrode for the CO2RR as compared to that on the pristine Pd electrode at all of the overpotentials under investigation.

A novel electrochemiluminescence (ECL) system consisting of Eosin B (EB) and K 2 S 2 O 8 was studied and found to generate ECL following the oxidation or reduction of Eosin B and K 2 S 2 O 8 . The type of buffer solution, type of co-reactant, pH value, and K 2 S 2 O 8 concentration were optimized through experiments to make the reaction system reach the best value. In addition, we found that Cr 3+ has a great quenching effect on the EB reaction system. Through the comparison of ECL and CV under different conditions, DPV curve, UV spectrum and ECL spectrum experiments, we focused on the quenching mechanism of Cr 3+ on the system and made reasonable guesses. This method demonstrates the potential application value of ECL in the field of environmental monitoring and reveals the potential value of organic molecular ECL luminophores.
