Dielectric barrier discharge plasma-modified chitosan flocculant and its flocculation performance

Quanfa Zhao,Haixia Wu,Wang Shen,Xiao Han,Bin Zheng,Yong Wang

The flocculation performance of chitosan can be enhanced by grafting modification to overcome its disadvantages of poor water solubility. In this study, chitosan was modified by dielectric barrier discharge plasma and polymerized with acrylamide and aluminum chloride to synthesize a new chitosan-based flocculant, namely, chitosan–acrylamide–aluminum chloride (CA-PAC). After optimizing the synthesis conditions of CA-PAC, the best conditions were as follows: discharge time of 3 min, discharge power of 50 W, polymerization temperature of 60 °C, polymerization time of 3 h, total monomer concentration of 100 g/L, and m(AlCl 3 ):m(CA) ratio of 2:1. Characterization was performed through SEM, XPS , FTIR , XRD, TG and 1 H NMR. Results showed that the preparation of CA-PAC was successful. The influences of flocculant dosage, pH, and stirring intensity on flocculation efficiency were investigated. The removal efficiency of turbidity was 94.1 %. The investigation of the flocculation mechanism revealed that CA-PAC mainly relied on charge neutralization or the synergic action of electric neutralization, bridging, and roll–sweep under acidic and neutral conditions, but it depended on the joint action of adsorption bridging and net sweeping under alkaline conditions. This study provides new ideas for the preparation and development of modified chitosan and broadens its application in water treatment.

Chitosan based aerogel fibers for piezoelectric and moisture electric energy harvesting

Yutong Song,Rui Zhang,Meijie Qu,Ruitong Zheng,Qingqing Zhao,Ping Tang,Yuezhen Bin,Hai Wang

Taking advantages of renewable resourced materials in the preparation of multifunctional aerogel fibers which integrate thermal insulation , piezoelectric and moisture electric generation could be a promising way of relieving pressure on both energy loss and generation. A series of chitosan-konjac glucomannan (CS-KGM) aerogel fibers was obtained by means of wet-spinning method followed by freeze-drying. The CS-KGM aerogel fibers with submicron porous morphology exhibit excellent thermal insulation performance. Single layer of CS-KGM aerogel fiber with only a thickness of 0.6 mm shows a thermal insulation temperature difference of 23 °C. A stable piezoelectric output voltage of 0.35 V can be generated from a single CS-KGM aerogel fiber during a 500 press-release cycle attributing to the intrinsic piezoelectricity of CS and the porous structure. Single aerogel fibers were coated with multi-walled carbon nanotubes as a way to prepare conductive aerogels. Meanwhile, the moisture electric output voltage can be as high as 160 mV and last for >8 h at 60% relative humidity for CS-KGM single aerogel fiber assisted by a thin layer of carbon nanotube coating. Overall, CS-KGM aerogel fibers prepared in this work has good compatibility and potential applications in thermal management and energy harvesting from low density resource, which provides a new strategy for the preparation of sustainable and multifunctional wearable materials.

Development of mussel-inspired chitosan-derived edible coating for fruit preservation

Chaomei Zhou,Jinrong Bai,Fantao Zhang,Rongya Zhang,Xiaolei Zhang,Kai Zhong,Bin Yan

Fruit rotting at the postharvest stage severely limits their marketing supply chains and shelf-life. Thus, developing a green and cost-effective approach to extend the shelf-life of perishable foods is highly desired. In this study, inspired by the mussel-adhesion strategy, a multifunctional fruit coating material has been developed using a quaternized catechol-functionalized chitosan (CQ-CS) grafted with 2, 3-epoxypropyl trimethyl ammonium chloride and 3, 4-dihydroxy benzaldehyde . The as-prepared CQ-CS coating exhibited excellent mechanical properties, universal surface adhesion abilities, antimicrobial and antioxidant capacities without any potential toxicity effects. Using strawberry and banana as model fruits, we showed that the CQ-CS coating could effectively maintain the fruit's firmness and color, decrease the weight loss rate, and prevent microbial growth , thus finally extending their shelf- life when compared to uncoated samples, indicating the universal application of the as-prepared CQ-CS coating. These findings demonstrated that this novel conformal coating of CQ-CS has great potential for fruit preservation in the food industry.

Injectable multifunctional carboxymethyl chitosan/hyaluronic acid hydrogel for drug delivery systems

Fengjiao Zhang,Sufeng Zhang,Rui Lin,Shuyuan Cui,Xiaokai Jing,Sergiu Coseri

Injectable hydrogels with notable mechanical properties and self-healing ability are promising carriers for use as a drug delivery system. Here, adipic acid dihydrazide (ADH) and calcium ions (Ca 2+ ) were introduced into quaternary ammonium carboxymethyl chitosan and aldehyde-modified hyaluronic acid hydrogels (QCS + OHA). The hydrogels were synthesized through the interaction of the Schiff bases (imine bonds, acylhydrazone bonds) and coordination bonds via a facile one-step approach. The gelation time (∼54 s) ensured excellent injectability. The QCS + OHA + ADH + Ca 2+ hydrogel had notable mechanical properties (compressive stress up to 896.30 KPa), good self-healing ability (up to 94 %), good pH responsiveness, and excellent antibacterial properties . In addition, the QCS + OHA + ADH + Ca 2+ hydrogel had a high drug loading capacity (121.3 mg/g) and sustained drug release behaviour (≥120 h). The results of cytotoxicity tests showed a high cell proliferation rate (up to 98 %) and good cytocompatibility. In summary, this work presents an injectable and self-healing pH-responsive hydrogel that can be used as a carrier for drug delivery systems.

Biocompatibility and osteogenic potential of choline phosphate chitosan-coated biodegradable Zn1Mg

Chen Zhong,Haoran Zhu,Yinying Sheng,Jin Wo,Deqiang You,Guodong Sun,Zhentao Yu,Wei Li,Xiaojian Wang

Zinc alloys have demonstrated considerable potentials as implant materials for biodegradable vascular and orthopedic applications. However, the high initial release of Zn 2+ can trigger intense immune responses that impede tissue healing. To address this challenge and enhance the osteogenic capacity of zinc alloys, the surface of Zn1Mg was subjected to CO 2 plasma modification (Zn1Mg-PP) followed by grafting with choline phosphate chitosan (Zn1Mg-PP-PCCs). This study aims to investigate the in vitro and in vivo biocompatibility of the surface-modified Zn1Mg. The effect of the surface modification on the inflammatory response and osteogenic repair process was investigated. Compared with unmodified Zn1Mg, the degradation rate of Zn1Mg-PP-PCCs was significantly decreased, avoiding the cytotoxicity triggered by the release of large amounts of Zn 2+ . Moreover, PCCs significantly enhanced the cell-material adhesion, promoted the proliferation of osteoblasts (MC3T3-E1) and upregulated the expression of key osteogenic factors in vitro . Notably, the in vivo experiments revealed that the surface modification of Zn1Mg suppressed inhibited the expression of inflammatory cytokines, promoting the secretion of anti-inflammatory factors, thereby reducing inflammation and promoting bone tissue repair. Furthermore, histological analysis of tissue sections exhibited strong integration between the material and the bone, along with well-defined new bone formation and reduced osteoclast aggregation on the surface. This was attributed to the improved immune microenvironment by PCCs, which promoted osteogenic differentiation of osteoblasts. These findings highlight that the preparation of PCCs coatings on zinc alloy surfaces effectively inhibited ion release and modulated the immune environment to promote bone tissue repair. Statement of significance Surface modification of biodegradable Zn alloys facilitates the suppression of intense immune responses caused by excessive ion release concentrations from implants. We modified the surface of Zn1Mg with choline phosphate chitosan (PCCs) and investigated the effects of surface modification on the inflammatory response and osteogenic repair process. In vitro results showed that the PCCs coating effectively reduced the degradation rate of Zn1Mg to avoid cytotoxicity caused by high Zn 2+ concentration, favoring the proliferation of osteoblasts. In addition, in vivo results indicated that Zn1Mg-PP-PCCs attenuated inflammation to promote bone repair by modulating the release of inflammation-related factors. The surface-modified Zn1Mg implants demonstrated strong osseointegration, indicating that the PCCs coating effectively modulated the immune microenvironment and promoted bone healing.

Chitosan with enhanced deprotonation for accelerated thermosensitive gelation with β-glycerophosphate

Yang Liu,Chuang Lang,Yaping Ding,Siyu Sun,Guangwei Sun

Chitosan (CS)/β-sodium glycerophosphate (β-GP) gels have low temperature sensitivity, which limits their biomedical application. The protonation and deprotonation of the CS amino groups are regulated by pH. It is possible to accelerate thermosensitive gelation by increasing the pH of the CS solution to maximize the degree of CS amino deprotonation and enhance hydrophobic interactions. In this study, a CS solution with a higher pH (HpHCS; pH 6.19, close to pKa 6.2) was obtained by means of the consumption of hydrogen ions using excessive chitosan. Raman spectroscopy showed that strong hydrogen bonds might exist in HpHCS. Gelation was accelerated significantly at 37 °C using HpHCS due to the decreased sol–gel transition temperature. Support vector machine and t-distributed stochastic neighbor embedding analyses confirmed that the gel derived from HpHCS was significantly different from that of the other groups. In addition, the HpHCS gel exhibited a low degradation rate and low cytotoxicity. Furthermore, a mathematical equation was established to predict the gelation time of HpHCS/β-GP at 37 °C. A better concentration of HpHCS for in vivo injection was obtained due to the shorter gelation time and lower viscosity. We found that human mesenchymal stem cells survived and proliferated in vitro in the HpHCS/β-GP hydrogel. Therefore, this study provides an optimized HpHCS solution with enhanced deprotonation for accelerated thermosensitive gelation with β-GP.

Prussian blue composite microswimmer based on alginate-chitosan for biofilm removal

Xiaoli Zhang,Qingli Qu,Anquan Yang,Jing Wang,Weixia Cheng,Aying Zhou,Ranhua Xiong,Chaobo Huang

Bacterial infections pose a serious threat to public health, causing worldwide morbidity and about 80 % of bacterial infections are related to biofilm. Removing biofilm without antibiotics remains an interdisciplinary challenge. To solve this problem, we presented a dual-power driven antibiofilm system Prussian blue composite microswimmers based on alginate-chitosan, which designed into an asymmetric structure to achieve self-driven in the fuel solution and magnetic field. Prussian blue embedded in the microswimmers given it the ability to convert light and heat, catalyze Fenton reaction, and produce bubbles and reactive oxygen species. Moreover, with the addition of Fe 3 O 4 , the microswimmers could move in group under external magnetic field. The composite microswimmers displayed excellent antibacterial activity against S. aureus biofilm with an efficiency as high as 86.94 %. It is worth mentioning that the microswimmers were fabricated with device-simple and low-cost gas-shearing method. This system integrating physical destruction, chemical damage such chemodynamic therapy and photothermal therapy, and finally kill the plankton bacteria embedded in biofilm. This approach may cause an autonomous, multifunctional antibiofilm platform to promote the present most areas with harmful biofilm difficult to locate the surface for removal.

Phenylboronic acid-functionalized chitosan for enabling transmucosal delivery for cancer therapy

Wenqi Jiang,Mingkang Liang,Qifang Lei,Dashi Deng,Chenchen Li,Yuqing Li,Simin Yuan,Kai Wu,Guangzhi Li,Song Wu

Mucosa administration and transmucosal polymeric carriers demonstrate specific advantages in locoregional therapy, which, however, suffer from unsatisfactory transmucosal delivery efficiency and limited hydrophobic or bio-macromolecular drug loading. Herein, we demonstrate several phenylboronic acid-modified chitosan derivatives to screen out a self-assembly nano-system. The phenylboronic acid unit synthesized in chitosan skeleton (CS) afforded donor–acceptor coordination with amidogen of pirarubicin (THP) and aPD-1 [1] , [2] , [3] ,the drug loading content was increased from 18.2 ± 0.4% to 37.0 ± 1.0% compared with CS. Besides, the phenylboronic acid modification provided an additional transmucosal advantage for cationic CS via non-covalent binding with mucosal glycosaminoglycans and enhanced the penetration ability of drugs through the endocytosis, Golgi-endoplasmic reticulum and exocytosis pathway. Furthermore, the encapsulated agents can be selectively released in response to the high reactive oxygen species concentration in tumor tissues, which combined with enhanced transmucosal capacity extremely contribute to improving the therapeutic efficacy and biosafety of [email protected] and [email protected] NPs in bladder cancer perfusion therapy as well as lung cancer inhalation treatment. Even more exciting is that in addition to THP and aPD-1, our platform could be easily extended to other chemotherapeutics and bio-macromolecular agents with electron-donating groups, rendering a simple and robust strategy for enabling highly efficient mucosal administration-based cancer therapy.

Synthesis of gentamicin-grafted-chitosan with improved solubility and antibacterial activity

Tingting Yan,Chengpeng Li,Qianqian Ouyang,Dongying Zhang,Qingkun Zhong,Puwang Li,Sidong Li,Ziming Yang,Tao Wang,Qingxia Zhao

Due to superior properties, chitosan (CS) has been widely applied in health field. However its acid-solubility and poor antimicrobial activity compromise its safety and applications, respectively. To enhance its water-solubility and antimicrobial activity, gentamicin-grafted-chitosan (GT-CS) is synthesized for the first time, which is then confirmed by various characteristic methods and the graft ratio for gentamicin is about 22.1%. Particularly, it is the first study, disclosing there are degradation and crosslinking involved simultaneously during CS oxidation . Furthermore, GT-CS shows greatly improved water solubility (10.0w%). Compared to CS, the antibacterial activity of GT-CS is improved significantly, approaching the pure gentamicin. It should be noted that the antimicrobic mechanism may not be the target-specific interactions but the nonspecific interactions with the bacterial membrane as shown by scanning electron microscope. To sum up, GT-CS is a superior antimicrobial with good solubility.

Mesoporous silica nanoparticles carried on chitosan microspheres for traumatic bleeding control

Xun Sun,Yan Fang,Zonghao Tang,Zhengchao Wang,Xinqing Liu,Haiqing Liu

Chitosan has been made into various hemostats, but their hemostatic efficiency for controlling severe traumatic bleeding is still inadequate. The aim of this work is to make quick hemostats by incorporating mesoporous silica nanoparticles into chitosan. Porous chitosan-silica composite microspheres (CSMS-S) with high hemostatic efficacy were fabricated through a combination of the microemulsion , thermally induced phase separation, and surfactant templating method. A large number of mesoporous silica nanoparticles were formed on and within the CSMS-S microspheres, which had abundant surface and inner macropores. The synergetic two hemostatic mechanisms from chitosan and mesoporous silica nanoparticles let CSMS-S composite microspheres with proper amount of silica displayed better hemostatic potential than the single component porous chitosan microspheres (CSMS). Within a same time interval, the whole blood clotting kinetics showed that CSMS-S could form larger blood clots than CSMS. The hemostatic time of CSMS-S was down to 97 s from 114 s of CSMS in the rat liver laceration model. The cytotoxicity and histological analysis proved that CSMS-S was a safe hemostatic agent without noticeable adverse effects on tissues around the wound. Our results demonstrate that CSMS-K is a promising quick hemostatic agent for traumatic hemorrhaging control.

Preparation and evaluation of porous starch/chitosan composite cross-linking hemostatic

Yan Chen,JunQing Qian,ChangYan Zhao,LieChao Yang,Jing Ding,Hui Guo

Composite porous starch (PS) with chitosan (CS) was easily cross-linked by sodium trimetaphosphate (STMP) to obtain a hemostat called STMP/PS/CS (SPC) with ideal hemostatic effect. The water absorption ratio and swelling ratio of SPC reach up to 150.8%, 355.0% respectively and it was observed by scanning electron microscopy (SEM) that a large amount of CS was combined with PS, which indicated the composite effect was ideal. Synergistic hemostatic effect of SPC was indicated in blood cells evaluation experiment and the activation of platelets triggers a multistep cascade that produces a clot by adsorbing more blood cells on the surface of SPC. The average hemostasis time of SPC in tail amputation 1 cm, 2 cm and liver laceration was shortened by 29%, 42%, 37% respectively compared with blank control, and had better hemostatic effect than CS, PS in the hemostatic performance on the rat tail amputation and liver laceration. Besides, we found SPC displayed a superior adhesiveness in rat injuries.

Tetracycline hydrochloride loaded citric acid functionalized chitosan hydrogel for wound healing

Hao Chen,Biyun Li,Bei Feng,Hao Wang,Huihua Yuan,Zhiwei Xu

Citric modified chitosan (CC) hydrogel containing antibacterial drugs is developed by the freezing and thawing treatment method. The SEM image of the CC hydrogel revealed a porous structure. The rheological properties, porosity, swelling rate, water uptake, tensile properties and in vitro degradation were found to be tunable via CC concentration. To enhance antibacterial properties, tetracycline hydrochloride (TH) representing the drug model, was integrated into the CC hydrogel. The cumulative release of drug was also tunable via CC concentration. The drug loaded CC hydrogel showed enhanced antimicrobial activity against E. coli and S. aureus. In animal tests, it was found the TH loaded CC hydrogel accelerated the healing of the wounds created on rats. These results suggest that the drug loaded CC hydrogel has a promising future in wound healing as a wound dressing.
