Infection associated with implants and devices is a problem in medicine, especially when bacterial colonization leads to the formation of biofilms on the implant surface. Here, we report an effective strategy to develop dual-functional coatings on titanium implants with antibacterial and antifouling properties to combat biofilm infections. Coating monomers with antibacterial and antifouling blocks were prepared by conjugating curcumin-derived curcumin carboxylic acid (Cur-COOH) with different chain lengths of allyl polyethylene glycol (APEG; MW 1200 or 2400). The coating monomer (APEG 1200/2400 -Cur) was grafted onto the surface of the titanium implant through azobisisobutyronitrile (ABIN)-induced radical polymerization to form dual-functional coatings with antibacterial and antifouling properties. Scanning electron microscopy & energy dispersive spectroscopy (SEM & EDS), contact angle (CA), and X-ray spectroscopy (XPS) confirmed successful grafting of coupling compounds. The coatings exhibited excellent antibacterial activity against Gram-negative bacteria ( Escherichia coli ) and Gram-positive bacteria ( Staphylococcus aureus ) and prevented biofilm formation. The coatings also reduced the adsorption of proteins and the adhesion of platelets, indicating their excellent antifouling properties. Notably, the coatings also exhibited favorable biocompatibility with mammalian erythrocytes and pre-osteoblasts. In summary, this dual-functional coating showed remarkable potential for application in preventing and treating infections caused by biofilms.

Diabetic wounds suffer from bacterial infections, reactive oxygen species, and inflammatory reactions. Curcumin (Cur) is a natural diketone with antibacterial, anti-inflammatory, antioxidant activities, but poor water solubility and toxicity at high local concentration. Herein, inspired by the oxygen transport process of red blood cells via the dynamic interaction of hemoglobin and oxygen, phenylboronic acid (PBA)-anchored hydrogel (GOHA-PBA) is utilized to achieve active enrichment and smart release of Cur, which can reverse the inflammatory microenvironment and promote wound healing. First, PBA-modified gelatin (Gel-PBA) and oxidized hyaluronic acid (OHA) crosslink and form GOHA-PBA by borate ester reaction and Schiff base reaction. GOHA-PBA possesses excellent curcumin loading efficiency with 130 000 times than that in water and forms Cur-laden GOHA-PBA (GOHA-Cur). GOHA-Cur has injectability, self-healing, and self-adaptive properties, which are conducive to surgical procedure. GOHA-Cur owns reversible adhesion properties, facilitating it be peeled off and remove harmful substances, including excess glucose, acids, bacteria, and ROS at the wounds. GOHA-Cur can pH and glucose responsively release Cur; hence, it shows excellent antibacterial, antioxidant, anti-inflammatory effects in vitro. Animal experiments show that GOHA-Cur can inhibit inflammation and promote wound regeneration. This study can provide a valuable concept for Cur delivery and diabetic wound healing.

In this study, hyaluronic acid (HA) and curcumin (CCM) are used as functional molecular skeletons to prepare a reticular polymer HA-CCM* via the free radical polymerization reaction. The results showed that HA-CCM* formed nanomicelles through self-assembly in the aqueous medium. These nanomicelles were seen as star-shaped micelles and the particle diameter distribution range of 220–280 nm, which are used to carry doxorubicin (DOX) with a high drug loading and an entrapment efficiency. The experiment of drug release in vitro indicated that HA-CCM* micelles showed significant acid sensitivity. The anticancer activity of DOX-loaded by HA-CCM* micelles (HA-CCM* + DOX) was quantified by MTT assay , indicating that the cytotoxicity of HA-CCM* + DOX to human cervical cancer cell line (HeLa) and human renal cell adenocarcinoma cell line (786-O) was significantly improved and to normal human renal epithelial cell line (293A) was obviously reduced. DOX from HA-CCM* + DOX micelles can enter into the cells and resulted in cell death. The multifunctional HA-CCM* micelles have good prospects in the field of drug delivery. Overall, these results confirmed the potential of these nanomicelles for carrying DOX in the improved cancer chemotherapy.

A redox prodrug consisting of camptothecin-conjugating PEG through the diselenide bond was synthesized, and the resultant amphiphilic prodrug was then used to load the curcumin to form the co-delivery system for the synergetic B16 melanoma cells inhibition. The co-delivery system showed a good response to glutataione, and the prodrug could be degraded and the two loaded drugs were then released. Both camptothecin and curcumin could induce the obvious cytotoxicity to B16 melanoma cells. Particularly, the co-delivery strategy showed a synergetic inhibition effect to B16 melanoma cells through an in vitro assay. In vivo assay also showed that the co-delivery system could inhibit B16 tumor growth and showed much better inhibition effect than that of only camptothecin or curcumin used. Moreover, as an injectable drug delivery system, the biocompatibility including the blood compatibility of the co-delivery system was confirmed, suggesting that the redox co-delivery system have a potential application in B16 tumor therapy.

Curcumin (Cur) and its nanomedicine were researched in Caco-2 cells to investigate the internalization and subcellular transport across Caco-2 cell monolayer membrane. Cur loaded polymer micelle (PM), nanoemulsion (NE) and liposome (Lipo) were prepared to explore the transport process incorporating endocytosis, intracellular trafficking and transcytosis in Caco-2 cells. Firstly, nanocarriers could contribute to the cellular uptake and improve the stability of Cur within the Caco-2 cells. The co-mediation of clathrin , lipid raft/caveolae and/or macropinocytosis were involved in the internalization of Cur nanopreparations, indicating the diversity and partial energy-dependency of the endocytosis. Secondly, the endocytosed nanocarriers with Cur successively transported from apical early endosome to late endosome with the extension of incubation time . Furthermore, the intracellular location of Cur nanomedicines in lysosome and endoplasmic reticulum (ER) were detected and illustrated the actual transport of Cur nanomedicine to lysosome and ER. Finally, nanocarriers could enhance the transcytosis of Cur, involving the energy-dependent patterns. Especially, Cur-PM exhibited more advantages in regulating and coordinating the entry and the exit of Cur. The study indicated that the coexistence of multiple transport pathways had the potency to promote nanocarriers transcytosis and provided a rational dosage design for oral Cur.

The aim of this work is to develop curcumin-loaded hollow mesoporous silica microspheres (HMSMs@curcumin) to improve the poor oral bioavailability of curcumin. Hollow mesoporous silica microspheres (HMSMs) were synthesized in facile route using a hard template. HMSMs and HMSMs@curcumin were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), nitrogen adsorption/desorption measurements, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR), and X-ray diffraction (XRD). In addition, to demonstrate the potential application of the HMSMs@curcumin, cytotoxicity, in vitro release behavior and in vivo pharmacokinetics of curcumin loaded in these HMSMs were investigated by using of Caco-2 cells and Sprague-Dawley (SD) rats, respectively. These mono-dispersed HMSMs exhibited high drug loading ratio and encapsulation efficiency due to the mesoporous shell and hollow core. The excellent characteristics of HMSMs such as mono-dispersed morphology, smooth surface, uniform, ordered and size-narrowing mesopores resulted in a good in vitro release profile of curcumin from HMSMs@curcumin. Moreover, an impressive improvement in the oral absorption of curcumin and prolonged systemic circulation time were achieved in the in vivo animal studies. In addition, the good biocompatibility of developed HMSMs with Caco-2 cells was confirmed based on the in vitro cytotoxicity assay. In conclusion, this system demonstrated a great potential for efficient delivery of curcumin in vitro and in vivo, suggesting a good prospect for its application in clinic for therapeutic drug delivery in future.

Curcumin is a naturally occurring polyphenolic compound that is commonly used in both medicine and food additives, but its low aqueous solubility and poor bioavailability hinder further clinical applications. For assessing the effect of the glycosylation of curcumin on its aqueous solubility, two glycosyltransferase genes (BsGT1andBsGT2) were cloned from the genome of the strainBacillus subtilisATCC 6633 and over-expressed inEscherichia coli. Then, the two glycosyltransferases were purified, and their glycosylation capacity toward curcumin and its two analogues was verified. The results showed that bothBsGT1andBsGT2could convert curcumin and its two analogues into their glucosidic derivatives. Then, the structures of the derivatives were characterized as curcumin 4′-O-β-D-glucoside and two new curcumin analogue monoglucosides namely, curcumoid-O-α-D-glucoside (2a) and 3-pentadienone-O-α-D-glucoside (3a) by nuclear magnetic resonance (NMR) spectroscopy. Subsequently, the dissolvability of curcumin 4′-O-β-D-glucoside was measured to be 18.78 mg/L, while its aglycone could not be determined. Furthermore, the optimal catalyzing conditions and kinetic parameters ofBsGT1andBsGT2toward curcumin were determined, which showed that the Kcat value ofBsGT1was about 2.6-fold higher than that ofBsGT2, indicating that curcumin is more favored forBsGT2. Our findings effectively apply the enzymatic approach to obtain glucoside derivatives with enhanced solubility. Keywords:curcumin 4′-O-β-D-glucoside;glycosyltransferases;Bacillus subtilisATCC 6633

The development of high drug loading and sustained release nanoparticles by simple and economical methods is significant for the treatment of diseases. Amorphous calcium carbonate (ACC) is an ideal drug delivery system because of its excellent pH responsiveness, biocompatibility and biodegradability. In this work, ACC nanoparticles were successfully precipitated in ethanol–calcium solutions using ammonium carbonate as the internal source of CO 2 , which exhibited a relatively narrow size distribution in range of 10–200 nm. The microscopic morphology, amorphous characters, porous structure and thermal behavior of resultants were investigated by electron microscopy, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller and thermogravimetric analysis, respectively. In vitro drug release assay showed that ACC nanoparticles had high loading capacity of curcumin (Cur) and favorable drug release properties. Furthermore, ACC-Cur showed excellent abilities to scavenge free radicals, protect Cur stability and damage A549 cells, providing a broadened space for the applications of this material in the fields of biomedical or food.

Alzheimer’s disease is a neurodegenerative disorder mainly characterized by β-amyloid deposit and tau hyperphosphorylation with no curative treatments. Curcumin (Cur) has been proved to have potential use in Alzheimer’s disease with its anti-amyloid, anti-inflammatory, and anti-oxidant properties, etc. However, its hydrophobicity and low bioavailability hinder its application. In this paper, we designed a novel brain-target nanoparticle, poly(lactide-co-glycolide)-block-poly(ethylene glycol) (PLGA-PEG) conjugated with B6 peptide and was loaded with Cur (PLGA-PEG-B6/Cur) and administered it into HT22 cells and APP/PS1 Al transgenic mice. The in vitro assays including dynamic light scattering (DLS), flow cytometry (FCM), red blood cell (RBC) lysis, and thromboelastography (TEG) analysis indicated that this nanoparticle could narrow the diameter of Cur, increase its cellular uptake and possess good blood compatibility. The results from Morris water maze proved that PLGA-PEG-B6/Cur could tremendously improve the spatial learning and memory capability of APP/PS1 mice, compared with native Cur. The ex vivo assays including Bielschowsky silver staining, immunostaining, and western blotting demonstrated that PLGA-PEG-B6/Cur could reduce hippocampal β-amyloid formation and deposit and tau hyperphosphorylation. Thus, we suggested that PLGA-PEG-B6/Cur nanoparticles would be of potential and promising use for the treatment of Alzheimer’s disease.

A feasible strategy of on-demand drug delivery for the treatment of dermal inflammation under low-pH conditions is proposed, employing zeolitic imidazolate framework-8 (ZIF-8) as a pH-responsive nanoparticle and curcumin (CCM) as a model drug. To overcome the low bioavailability of topically treated drug, a microneedle (MN) form is used to incorporate CCM and ZIF-8. Taking advantage of the fact that ZIF-8 degrades under acidic conditions, CCM is embedded in porous ZIF-8 nanoparticles such that CCM is released when ZIF-8 comes into contact with an acidic dermal fluid at the inflammation site, and this CCM-encapsulated ZIF-8 (CCMZIF) is incorporated into water-dissolvable poly(vinyl pyrrolidone) MN. The ZIF-8 shows a high loading capacity (∼40.5%) of CCM through chemical bonding and physical adsorption. From in vitro tests with both a buffered solution and porcine skin, CCM from the CCMZIF MN is released in a higher amount at pH 5.0 than at pH 7.4, demonstrating the capability of the pH-responsive release of the drug when needed at inflammatory sites. The analytical investigation conducted here reveals that an acidic environment triggers the structural degradation of ZIF-8, allowing the release of the chemically bonded CCM. Cytotoxicity and stability tests demonstrate the good biocompatibility and bioavailability of ZIF-8. This study highlights the analytical discussion of the encapsulation and release mechanism of CCM in a ZIF-8-implemented MN drug delivery platform. The results demonstrate an advanced on-demand therapeutic strategy for skin disorder treatment.

Summary Antimicrobial photodynamic therapy (aPDT) is an effective method in the field of sterilisation. Singlet oxygen ( 1 O 2 ) is an important substance in photodynamic sterilisation that needs to be detected. It involves energy transfer from an excited photosensitiser to surrounding oxygen molecules to produce cytotoxic 1 O 2 species, a process termed as type II reaction. Traditional photodynamic sterilisation for fresh produce generally involves only a single photosensitiser and uses nanoemulsions coating to make the photosensitiser work better. This allows the emulsion to produce only a limited amount of reactive oxygen species (ROS) in the presence of light, such that the bactericidal effect is limited. We have innovatively compounded two photosensitisers and stored them as nanoemulsions. The composite photosensitiser produces relatively higher ROS and the nanoemulsions have good stability as well as rheological properties, and the photosensitisers are well coated in the emulsions. In summary, nanoemulsions based on composite photosensitisers showed synergistic activity in controlling the growth of Staphylococcus aureus , which provides a novel approach for raw sterilisation to maintain food safety.

The combination of scallop male gonads hydrolysates (SMGHs) and κ -carrageenan (KC) was applied for curcumin (Cur) embedment and preservation, the corresponding Cur retention rate as well as gelation and microstructural properties were investigated during storage subjected to various pH (8 and 6) and temperature (25 °C and 4 °C). SMGHs-based gels could well protect Cur from degradation during 4 °C storage, with retention rate ranging from 95% to 97% after 30 day storage. At 25 °C, Cur could still be well preserved in SMGHs-based gels under pH 6 with Cur retention rate above 90%, while pH 8 led to rapid Cur degradation to around 75%. In addition, at pH 8 and 25 °C, the gel strength of SMGHs-Cur deteriorated significantly while evidently enhanced in SMGHs/KC-Cur due to synergistic combination within SMGHs and KC in couple with dehydration effect. Moreover, the water migration and distribution of SMGHs-based Cur-loaded vehicles were relatively stable at 4 °C while revealed apparent deterioration at 25 °C during storage. Furthermore, the microstructures of SMGHs/KC-Cur gradually transformed from homogeneous network with dense and smooth pores to tattered network with large and fragmentary pores, especially at 25 °C, promoting more Cur degradation. This study demonstrated that SMGHs/KC-Cur exhibited better than SMGHs-Cur in Cur preservation, potentially performing as excellent and sustainable protein hydrolysate/polysaccharide soft materials in food and biomedical fields.
