Bridging traditional Chinese medicine theory with advanced MOFs delivery for synergistic drug-resistant infected-chronic wound therapy

Chen Chen,Na Zhang,Fructueux Modeste Amona,Xiaolei Han,Qi Tang,Lanxin Geng,Guangfu Liao,Jie Zhang,Tushuai Li

Chronic wounds infected with multidrug-resistant bacteria pose a significant challenge to global health, as traditional monotherapies often fail to eradicate pathogens and restore the damaged wound microenvironment. Engineering innovative nanocomposites that integrate synergistic therapeutics and controlled release is a pivotal strategy for advanced wound management. Here, we developed a novel multi-drug metal-organic framework (MOF) nanocomposite, BBR + FA@UiO, by covalently grafting ferulic acid (FA) onto UiO-66-NH 2 and physically loading berberine (BBR). Drawing from "Jun–Chen–Zuo–[ Shi ]" hierarchy of Traditional Chinese Medicine (TCM), this design used the MOF as a robust porous carrier “[ Shi ]” for the antibacterial drug "Jun" (BBR) and antioxidant drug "Chen/Zuo" (FA). BBR + FA@UiO exhibits potent, synergistic antibacterial and antibiofilm activity against critical clinical pathogens like MRSA and Pseudomonas aeruginosa, through a pH/ROS-responsive release mechanism in the infected site. Functionally, BBR + FA@UiO activates the Nrf2/Keap1 pathway in macrophages, thereby mitigating oxidative stress and enhancing mitochondrial function. Consequently, proliferation and migration of Human Umbilical Vein Endothelial Cells (HUVECs) are restored through promoting angiogenesis. In vivo, BBR + FA@UiO significantly accelerates healing by eradicating bacteria, enhancing collagen deposition, and improving vascularization, driven by the coordinated activation of the Nrf2/HO-1 antioxidant axis and pro-regenerative Wnt/β-catenin and TGF-β signaling pathways. Demonstrating excellent biocompatibility and no systemic toxicity, this work establishes BBR + FA@UiO as a new therapeutic approach that merges TCM with advanced MOFs for comprehensive wound care.

Microwave assisted extraction of chlorogenic acid from sunflower seed meal using natural deep eutectic solvent based on COSMO-RS design

Boshen Wang,Jihong Fu,Jun Tang,Haipeng Zhang,Junli Yu,Qichao Wu,Mengdie Li

Sunflower seed meal is a rich source of chlorogenic acid (CGA). In this work, the conductor-like screening model for realistic solvation (COSMO-RS) was used to assist in identifying the optimal natural deep eutectic solvents (NADESs) for the extraction of CGA from the sunflower seed meal. A total of 20 hydrogen bond donors (HBDs) and 10 hydrogen bond acceptors (HBAs) at different molar ratios resulting in 600 NADESs were screened via the COSMO-RS model by calculating the infinite dilution activity coefficients of CGA. The results showed that the solvent composed of choline chloride (ChCl) and urea at a molar ratio of 2:1 had the highest extraction rate of 25.92 mg g −1 for CGA. Additionally, ChCl-Urea (2:1) was chosen as the solvent in response surface methodology (RSM) to optimize the microwave assisted NADES extraction parameters, including liquid-solid ratio, microwave power, temperature and irradiation time, yielding a CGA content of 26.39 mg g −1 . Furthermore, the antioxidant activities of the CGA extract using NADES were determined, which exhibited better DPPH, ABTS radicals scavenging abilities and ferric reducing antioxidant power (FRAP). Therefore, COSMO-RS and RSM can serve as effective techniques for screening NADESs and optimizing extraction processes, thereby expanding the application potential of NADES in extracting various bioactive compounds.

A Dual-Dynamic Crosslinked Polysaccharide-Based Hydrogel Loaded with Exosomes for Promoting Diabetic Wound Healing

Ding Lin,Zhenhao Li,Jianying Hao,Xiaobo Xu,Xiuqiang Li,Yuan Feng,Xiaochen Lu,Fanglian Yao,Hong Zhang,Junjie Li

Diabetic wounds are often accompanied by severe inflammation, which is unfavorable for vascular growth and wound repair. Therefore, promoting the healing of diabetic wounds is of great significance. In this study, carboxymethyl chitosan (CMCS) was grafted with 4-formylphenylboronic acid (FPBA) and then crosslinked with oxidized sodium alginate (OAlg) to form a dual-dynamic covalent hydrogel (CPOA) based on borate ester bond and Schiff base bonds. Mesenchymal stem cells’ exosomes (Exos) were incorporated into the CPOA to construct CPOA@Exos for diabetic wound healing. Owing to the dual-dynamic covalent crosslinking network, the CPOA hydrogel showed good injectability and self-healing ability. In addition, the hydrogel displayed reactive oxygen species (ROS) responsive properties, enabling both scavenging of multiple free radicals and on-demand release of Exos in the ROS-rich wound microenvironment. A diabetic wound model was established on C57 mice, and treatment with CPOA@Exos demonstrated that it could promote the polarization of macrophages toward the M2 phenotype, enhance cellular proliferation in the wounded area, and thereby accelerate the healing of diabetic wounds. In conclusion, this study provides a new hydrogel wound dressing that can inhibit inflammation for the management of diabetic wounds.

Polydopamine-sealed mesoporous silica nanocarriers loaded with organic UV filters for effective full-spectrum UV protection

Zixin Kang,Kehan Du,Zhuo-Ran Yang,Shunlan Luo,Xiaoping Zeng,Dawei Wang,Hongyang Zhao,Xiaowen Ou,Qianqian Liu,Hao Jiang,Jiangyu Wu

Ultraviolet radiation induces skin carcinogenesis through DNA damage and oxidative stress, driving demand for high-efficacy sunscreens. Current organic filters (e.g., octyl methoxycinnamate, OMC) suffer from narrow spectral coverage and phototoxicity. To address these limitations, we engineered OMC-loaded hollow mesoporous silica nanoparticles with polydopamine coating (OMC@HMSN@PDA). This nanoplatform achieved: (i) synergistic broad-spectrum protection, the sun protection factor (SPF) and UVA protection factor (UVAPF) up to 54 ± 0.74, 26.8 ± 6 with the concentration of 6 wt% OMC respectively, exceeding OMC@HMSN by 10.4-fold in UVB and 11.2-fold in UVA attenuation; (ii) 93.1 ± 1.2 % of DPPH and 98.5 ± 2.5 % of ABTS radical scavenging at 10 µg mL −1 , and 94.6 ± 0.03 % intracellular reactive oxygen species (ROS) suppression; and (iii) prevention of the payload leakage and photo-instability of OMC, resolution of OMC’s phototoxicity with above 80 % cell viability in vitro . In vivo studies demonstrated prevention of UV-induced epidermal hyperplasia and lower inflammation. This technology establishes a promising approach for photoprotection integrating UV filtering, antioxidant activity, and enhanced safety.

Simple preparation and purification of citrus pectic oligosaccharides: structural analysis and antioxidant potential

Su Xiang,Zhao Zhe,Guo Kaige,Cao Yunli,Liu Huipeng,Li Zhipeng

Pectic oligosaccharides (POS) exhibit promising bioactive properties, primarily attributed to their simple structure, low viscosity, high solubility, and low molecular weight (Mw). In this study, a simple method based on the Fenton reaction and ethanol precipitation was used to prepare and isolate citrus pectic oligosaccharides (CPOS). The structural characteristics of the resulting CPOS were analyzed using FTIR, SEM, NMR, and UPLC-Q-TOF-MS, and their antioxidant activities were evaluated. Structural characterization revealed that the obtained CPOS consisted mainly of galacturonic acid (GalA). Further analysis via NMR and UPLC-Q-TOF-MS confirmed that CPOS with a degree of polymerization (DP) up to 13 was rich in GalA and galactose (Gal). Antioxidant activity assays demonstrated that CPOS exhibited strong DPPH (91.57% at 2 mg/mL) and ABTS (87.14% at 1 mg/mL) scavenging rates. This study indicated that a simple method based on Fenton reaction and ethanol precipitation can be used to produce CPOS with antioxidant activity, providing a technical reference for the industrialized low-cost production of POS and the development of functional food ingredients.

Curcumin-Loaded Tetrahedral Framework Nucleic Acids Alleviate Pulmonary Hypertension by Regulating Multiple Synergistic Signaling Pathways

Xiaomeng Yue,Jie Lan,Lin Yang,Liujie Zhao,Xing Li,Zejie Liu,Meng Li,Ruxia Liu,Dong Li,Shao-Kai Sun,Jiaojiao Yu

Pulmonary hypertension (PH) is a severe pulmonary vascular disease that ultimately leads to right heart failure, which involves multiple core mechanisms, including oxidative stress, inflammation, and pathological cellular proliferation. However, current therapies are limited in simultaneously modulating multiple core mechanisms, leading to poor clinical outcomes and high mortality. Herein, we introduce tetrahedral framework nucleic acids-Curcumin (tFNAs-Cur) as a novel therapeutic for PH, targeting multiple synergistic pathways including TGF β1, NF-κB, and PI3K-Akt. The proposed tFNAs-Cur featured the advantages of high drug loading capacity, good water solubility, strong stability, and excellent biocompatibility. Cellular experiments demonstrate that tFNAs-Cur is efficiently internalized by pulmonary artery smooth muscle cells, alleviates oxidative stress, and inhibits their proliferation and migration. In the monocrotaline-induced PH rat model, tFNAs-Cur significantly improves pulmonary artery acceleration time, reduces vascular wall thickness, and medial pulmonary artery diameter. Cardiac assessments show improved exercise capacity, reduced Fulton index, and alleviated right ventricular fibrosis. Further transcriptomic and biochemical analyses reveal that the therapeutic effects of tFNAs-Cur were mediated via the ROS/TGF β1 antioxidant, NF-κB anti-inflammatory, and PI3K-Akt anti-proliferative pathways. The proposed tFNAs-Cur provides an efficient and safe method with potential for clinical translation that simultaneously utilizes multiple synergistic mechanisms for the treatment of PH.

Engineered Targeted Ce-Based MOF Nanozymes for ROS Scavenging and Inflammatory Reprogramming in Chronic Pancreatitis

Yongkang Lai,Yongliang Ouyang,Xiaojing Yin,Tao Yu,Jianhua Wan,Xueyang Li,Yi Hu,Xu Shu,Huan Wang

Chronic pancreatitis (CP) is a lifelong progressive fibrotic inflammatory disorder for which no effective cure is currently available. Persistent and recurrent inflammatory stimulation induced by reactive oxygen species (ROS) is a key driver of pancreatic fibrogenesis, making oxidative stress a promising therapeutic target to halt disease progression. In this study, we developed a nanosystem, HC@CeMOF, consisting of a small-sized cerium-based metal–organic framework (CeMOF) core loaded with curcumin and coated with hyaluronic acid (HA), enabling precise targeting of inflamed pancreatic tissue. HC@CeMOF exhibits a small-sized particle size along with favorable cellular and biological safety profiles. Once administered in vivo , the nanosystem exploits the specific binding affinity of HA to CD44 receptors on macrophages to selectively accumulate at inflamed pancreatic sites. Subsequently, the cerium-based nanozyme efficiently scavenges ROS through the reversible redox cycling between Ce 3+ and Ce 4+ , while the slow release of curcumin further suppresses the NF-κB signaling pathway and modulates inflammatory cytokine levels, thereby achieving synergistic anti-inflammatory and antioxidant effects. Collectively, these mechanisms substantially attenuate CP progression. This targeted ROS-scavenging and anti-inflammatory strategy holds promise as an alternative therapeutic approach for chronic pancreatitis.

Photo-controlled spatiotemporal sequential release of MXenes/NO gas from bilayer (clean-cure) hydrogel promotes healing of MRSA biofilm-infected diabetic ulcer wounds

Ying-Tong Ye,Xi Xiao,Hong-Ying Xia,Jie Li,Zhen-Zhen Gong,Ai-Zheng Chen,Shi-Bin Wang,Ranjith Kumar Kankala

Despite the success of various intelligent materials in meeting the multiple needs of diabetic ulcer wounds (DUW) healing, it is often challenging to consider the entire healing process, from early antibacterial and anti-inflammatory effects to late matrix remodeling and angiogenesis characteristics, by releasing multiple active ingredients in stages and on demand. Herein, we designed a versatile bilayer (GCM/Clean-GDA/Cure) hydrogel dressing with light-controlled, spatiotemporal sequential release of MXene nanosheets (MX NSs)/nitric oxide (NO) gas for the healing of methicillin-resistant Staphylococcus aureus (MRSA) biofilm-infected DUWs. Initially, the lower GCM layer comprised carboxymethyl chitosan-gelatin and cerium oxide (CeO 2 )-decorated MX NSs (CO-MX), and the upper GDA layer consisted of dopamine-modified methacrylated gelatin (GelMA-DA) with L-arginine (L-Arg) as a precursor. Under NIR irradiation, the GCM hydrogel releases photothermal-induced CO-MX NSs as self-regenerating antioxidants, exhibiting 98.25 % inhibition against MRSA and 96.37 % inhibition against Pneumonia aeruginosa. The abundant oxygen vacancies and reversible Ce(III)/Ce(IV) redox pairs of the released Ce species from CO-MX NSs could attenuate inflammation and promote early macrophage polarization. The light-assisted gel–sol thermal transition of the lower GCM layer further exposes the upper GDA layer, providing a conducive microenvironment for angiogenesis and tissue remodeling. L-Arg in the GDA layer could enable light-controlled in situ release of NO gas activated by elevated reactive oxygen species (ROS), reaching 7.71 μM within 10 min of exposure to NIR light. Thus, the NIR-assisted bilayer treatment group achieved a 97.74 % wound closure rate. Finally, the transcriptomic analysis validated the activation of regenerative pathways and suppression of excessive inflammatory and metabolic stress signals. Together, the photo-controlled sequential release of MX NSs and NO gas from the bilayer could expand the application of light-responsive nanomaterials, providing an innovative therapeutic modality for in situ treatment of diabetic wounds infected by highly drug-resistant MRSA biofilms.

Poly(Citric Acid) Bidirectional Regulator: Coordinating Iron Homeostasis to Suppress Stress Responses and Boosting Mitochondrial Bioenergetics for Enhanced Nerve Repair

Xinyue Liang,Yuchen Song,Xianzhen Dong,Junwei Su,Yuanfang Huo,Junwei Yang,Zhiqiang Li,Hao Zhang,Mengdi Liu,Aixi Yu,Honglian Dai

Due to the complex regenerative microenvironment, peripheral nerve repair poses significant challenges in clinical treatment. Severe injuries can lead to dysregulated iron homeostasis, and excessive iron produces reactive oxygen species (ROS) through the Fenton reaction. The subsequent oxidative stress further leads to mitochondrial and endoplasmic reticulum (ER) stress, which impedes nerve regeneration. In addition, insufficient vascular remodeling also limits the repair of damaged nerves. Herein, a poly(citric acid) (PCA)-loaded gelatin-lipoic acid (Gel-LA) microgel hydrogel-filled oriented electrospun fiber conduit was developed, which guides axonal alignment by topographical cues, promotes cellular infiltration and nutrient transport through a microgel cascade pore structure, and regulates the regenerative microenvironment via the ferrous ion (Fe 2+ ) chelation effect of PCA. The results demonstrate that this conduit effectively reduces the levels of Fe 2+ at the injury site, thereby alleviating mitochondrial and ER stress while promoting energy metabolism, vascular reconstruction, and nerve regeneration. This study highlights the potential of PCA in modulating the microenvironment of nerve injuries and provides new insights for developing tissue-engineered scaffolds.

Covalent cross-linking of whey protein isolate-chlorogenic acid nanoparticles mediated by laccase: Structural, antioxidant and functional characteristics

Bingjie Xue,Jing Zhang,Sijia Yan,Yanhui Yang,Wupeng Ge,Lili Zhao

To enhance the stability and bioavailability of chlorogenic acid (CHA), laccase (LAC)-catalyzed cross-linking was employed to prepare whey protein isolate (WPI)-CHA nanocomposites. The complexes exhibited sensitivity to variation in pH and temperature. This study investigated the effects of pH (3.0–7.0) and temperature (15–45 °C) on the structural, antioxidant, and functional properties of WPI-CHA (WC) and WPI-LAC-CHA (WLC) complexes. Under the optimal catalytic conditions for LAC (pH 5.0, 35 °C), WLC complexes exhibited the highest structural stability, characterized by the smallest particle size (133 ± 3.93 nm), highest surface hydrophobicity (822.79 ± 1.74), and lowest free amino content (209.99 ± 1.09 mM/mL). The 52 % fluorescence quenching, amide I red shift and >180 kDa aggregates formation confirmed tertiary structure unfolding and covalent cross-linking, indicative of efficient protein-polyphenol conjugation. WLC complexes exhibited optimal radical scavenging activity at pH 5.0 and 35 °C, with DPPH and ABTS scavenging rates of 78.33 ± 5.11 (μmoL TE/g) and 83.99 ± 1.92 %, respectively. Following in vitro gastrointestinal digestion, WLC complexes retained superior antioxidant capacity (48.5 ± 2.21 μmoL TE/g for DPPH and 70.5 ± 1.70 % for ABTS) compared to free CHA and WC complexes. WLC complexes exhibited significantly greater UV protective efficiency of CHA, functional characteristics (solubility, EAI, ESI, FC and FS) compared to WC. These findings highlighted LAC-catalyzed cross-linking as an effective strategy to fabricate high-performance WPI-CHA nanocomposites, which might be applied in nano-carriers and packaging materials in the food industry.

Synergistic citric acid cross-linked chitosan/sodium alginate films with porous starch-encapsulated thymol for sustainable pork preservation

Zhou Huang,Ming Zhou,Baijun Ma,Hao Luo,Yucheng Jiao,Shi Chen

To overcome the challenges of volatile active agents and the limited functionality of biopolymer-based packaging, a novel synergistic strategy was devised by constructing a biomimetic “organelle-cytoplasm-cell wall” system within a chitosan/sodium alginate (CS/SA) matrix. This was achieved by simultaneously incorporating citric acid as a covalent crosslinker and porous starch (PS) pre-loaded with thymol (THY) as a sustained-release organelle. The synergistic interplay between the robust cross-linked network and the PS-THY capsules endowed the composite film (CS-c-SA/PS-THY) with exceptional comprehensive properties: a 42.23% enhancement in tensile strength, an order-of-magnitude reduction in water vapor permeability (7.11 g/(m 2 ·24 h)), and a 23 °C increase in thermal decomposition temperature. Crucially, the film achieved sustained and controlled THY release that obeyed Fickian diffusion, which underpinned its potent, long-lasting antioxidant (75.82% DPPH scavenging) and antibacterial (inhibition zones >18 mm) efficacy. In practical pork preservation, the CS-c-SA/PS-THY significantly outperformed the control and the CS-c-SA/THY, effectively inhibiting microbial growth and quality deterioration to extend the shelf life by at least 4 days. This work provides a scalable and fundamental strategy for developing high-performance, sustainable active packaging.

Novel bio-based active packaging film with colloidosome-encapsulated clove essential oil

Yichen Yang,Li Yang,Haiming Cheng

To mitigate the thermal instability of essential oils, we engineered robust colloidosomes via the cross-linking of soy protein isolate nanoparticles (SPI NPs) using poly(hexamethylene diisocyanate) (PHDI) to encapsulate clove essential oil (CEO). Subsequent to a systematic screening of filler loadings, these colloidosomes were incorporated into a gelatin-κ-carrageenan (GK) matrix. Scanning electron microscopy (SEM), X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FTIR) confirmed that PHDI initiated nucleophilic addition, constructing a rigid shell while facilitating secondary interfacial grafting. This covalent interlocking ensured uniform dispersion and eliminated phase separation defects. In particular, the 40 wt% PHDI treatment yielded complete shell integrity, effectively shielding the CEO from thermal degradation. Accordingly, the optimized GK-CE-40 film featured a densified network structure, translating to superior UV/water vapor barrier properties, high tensile strength (13.41 ± 1.56 MPa), and sustained antioxidant/antibacterial activity (51.28% DPPH scavenging; 20 mm inhibition against E. coli ). Preservation trials on cherry tomatoes demonstrated significant retention of firmness and appearance over 10 days. These results validate interfacial cross-linking as a potent strategy to resolve the stability-dispersibility trade-off in bio-based packaging.
