Modification of microporous bionanocomposite films with visible light-activated photocatalytic antimicrobial TNT-CuO nanoparticles for active fruit packaging

Huan Zhang,Shuting Huang,Yali Zhao,Hong Sabrina Tian,Mengshi Lin,Yunfei Xie,Zhilong Yu

Active packaging technologies are evolving to enhance the preservation of fresh produce by fighting against microbial contamination and controlling internal packaging atmospheres. This study introduced an active fruit packaging called MT film, created by modifying a microporous polyvinyl alcohol/chitosan/cellulose nanocrystal bionanocomposite film with CuO-doped titania nanotubes. The MT film, with an average micropore size of 2.4 μm, displayed excellent mechanical properties and hydrophobicity due to its crosslinked structure. When exposed to visible light, the MT film could produce reactive oxygen species, effectively inhibiting the growth of Staphylococcus aureus and Escherichia coli O157:H7. Moreover, experiments on blueberry preservation demonstrated that the MT film could remove excess CO 2 and maintain a higher O 2 level. Under visible light, this film significantly reduced total viable count (4.6 ± 0.2 log CFU/g) and mold colony count (2.6 ± 0.1 log CFU/g), with Bacillus and Ascochyta being the primary inhibited genera. These findings highlight the potential of MT film in utilizing visible light to prevent microbial growth on blueberries and regulating the gas exchange of food packaging. MT film holds promise as an active packaging solution to improve the quality and shelf life of fresh produce while reducing food losses in the supply chain.

Microwave-Assisted “One-Pot” Acidolysis and Extraction for the Rapid Determination of Mancozeb in Fruit and Vegetable Samples

Qiaoxia Tian,Hongxing Li,Lixia Chen,Bingjun Han

Mancozeb is an extensively consumed fungicide, which often leaves high residue levels on agricultural products. The conventional method for detecting mancozeb involves a time-consuming process using gas chromatography (GC) after a 2-hour water-bath acidolysis, resulting in low efficiency and recovery rates. This study developed a rapid method for detecting mancozeb in fruits and vegetables using microwave-assisted acidolysis and extraction coupled with GC analysis. Mancozeb underwent “one-pot” acidolysis to generate CS 2 gas and was subsequently extracted from samples using microwave treatment, requiring only 50 seconds of pretreatment time. The average recoveries of mancozeb ranged from 81% to 112%. The limit of detection and limit of quantification were 0.003 and 0.01 mg kg −1 , respectively. The scanning electron microscope imaging showed that strong cell crumpling after microwave treatment improved the acidolysis rate significantly, where the acidolysis rate was 91.8% for mancozeb. In addition, this method is rapid, simple, and precise for detecting residues of mancozeb and other dithiocarbamate fungicides.

Modification of hollow microporous organic network with polyethyleneimine for efficient enrichment of phenolic acids from fruit juice samples

Shuo-Wen Gao,Na Li,Yuan-Yuan Cui,Cheng-Xiong Yang

Owning to the hydrophobic characteristics of microporous organic networks (MONs), their utilizations still largely limited in non- and weak-polar analytes. To expend their applications, here we reported the synthesis of a novel hollowed H-MON-PEI 1800 –2 composite via sacrifice template method and subsequent modification with polyethyleneimine (PEI) for efficient solid phase extraction of polar and ionic phenolic acid (PAs) from fruit juice samples. H-MON-PEI 1800 –2 exhibits large surface area, rapid extraction kinetics, remarkable chemical and thermal stabilities, and provides synergistic electrostatic, π-π, hydrogen bonding, and hydrophobic interaction sites for PAs. The developed method owns low limit of detection, wide linear range, large enrichment factors, and good reusability. The recoveries of H-MON-PEI 1800 –2 for PAs are 1–3 orders of magnitude higher than those of commercial adsorbents like activated carbon, C18 and Oasis HLB. This work highlights the prospects of functional H-MONs for enriching polar and ionic targets from complex sample matrices.

Green controllable synthesis of Au–Ag alloy nanoparticles using Chinese wolfberry fruit extract and their tunable photocatalytic activity

Li Sun,Yuechao Yin,Pengcheng Lv,Wenxian Su,Lixin Zhang

The controllable synthesis of Au–Ag alloy nanoparticles (NPs) without any hazardous or expensive chemical agent has been a challenge. This paper reports on the Chinese wolfberry fruit extract-assisted synthesis of Au–Ag alloy NPs with homogeneous composition, uniform alloy microstructure and a small average size less than 15 nm. The composition and the surface plasmon resonance (SPR) wavelength of Au–Ag alloy NPs could be linearly adjusted by the synthesis time in one-pot. Au–Ag alloy NPs were characterized by means of various techniques, and then their synthesis mechanism was discussed. The obtained Au–Ag alloy NPs were used as photocatalysts for the reduction reactions of 2-, 3-, and 4-nitrophenol and exhibited superior photocatalytic activity and stability. The rate constants of the above photocatalytic reactions could be linearly tuned by the composition of the Au–Ag alloy NPs and also by their addition amount, displaying a new method to control the rate constant. Furthermore, based on the controllable synthesis, the rate constant could even be predetermined just through setting the synthesis parameters of the Au–Ag alloy NPs.

Dynamic Changes of Active Components and Volatile Organic Compounds in Rosa roxburghii Fruit during the Process of Maturity

Su Xu,Junyi Deng,Siyao Wu,Qiang Fei,Dong Lin,Haijiang Chen,Guangcan Tao,Lingshuai Meng,Yan Hu,Fengwei Ma

Rosa roxburghii (R. roxburghii), native to the southwest provinces of China, is a fruit crop of important economic value in Guizhou Province. However, the changes in fruit quality and flavor duringR. roxburghiifruit ripening have remained unknown. Here, this study investigated the changes of seven active components and volatile organic compounds (VOCs) during the ripening of theR. roxburghiifruit at five different ripening stages including 45, 65, 75, 90, and 105 days after anthesis. The results indicated that during the ripening process, the levels of total acid, vitamin C, and soluble sugar significantly increased (p< 0.05), while the levels of total flavonoids, superoxide dismutase (SOD), and soluble tannin significantly decreased (p< 0.05). Additionally, the content of total phenol exhibited a trend of first decreasing significantly and then increasing significantly (p< 0.05). A total of 145 VOCs were detected by HS-SPME-GC-MS at five mature stages, primarily consisting of aldehydes, alcohols, esters, and alkenes. AsR. roxburghiimatured, both the diversity and total quantity of VOCs in the fruit increased, with a notable rise in the contents of acids, ketones, and alkenes. By calculating the ROAV values of these VOCs, 53 key substances were identified, which included aromas such as fruit, citrus, green, caramel, grass, flower, sweet, soap, wood, and fat notes. The aromas of citrus, caramel, sweet, and wood were predominantly concentrated in the later stages ofR. roxburghiifruit ripening. Cluster heatmap analysis revealed distinct distribution patterns of VOCs across five different maturity stages, serving as characteristic chemical fingerprints for each stage. Notably, stages IV and V were primarily characterized by a dominance of alkenes. OPLS-DA analysis categorized the ripening process ofR. roxburghiifruit into three segments: the first segment encompassed the initial three stages (I, II, and III), the second segment corresponded to the fourth stage (IV), and the third segment pertained to the fifth stage (V). Following the variable importance in projection (VIP) > 1 criterion, a total of 30 key differential VOCs were identified across the five stages, predominantly comprising ester compounds, which significantly influenced the aroma profiles ofR. roxburghiifruit. By integrating the VIP > 1 and ROAV > 1 criteria, 21 differential VOCs were further identified as key contributors to the aroma changes inR. roxburghiifruit during the ripening process. This study provided data on the changes in quality and aroma ofR. roxburghiifruit during ripening and laid the foundation for the investigation of the mechanism of compound accumulation during ripening.

Development of a Dual-Readout Multicolor Immunoassay for the Rapid Analysis of Isocarbophos in Vegetable and Fruit Samples

Zijian Chen,Wei-Xuan Huang,Hongwu Wang,Meiling Zhang,Kai Chen,Hao Deng

Multicolor immunoassay is a powerful tool for rapid analysis without the use of bulky instruments owing to various color conversions, which is suitable for on-site visual analysis for pesticides. Herein, this study developed a multicolor immunoassay for the rapid detection of isocarbophos. After competitive immunoassay, the secondary antibody (GAM-ALP) catalyzed ascorbyl-2-phosphate (AAP) into ascorbic acid (AA). The AA can reduce K3[Fe(CN)6] into K4[Fe(CN)6]. The latter can react with Fe3+to form Prussian blue; otherwise, the orange AAP-Fe3+complex was generated. Therefore, the multicolor immunoassay achieved a color conversion of orange–green–blue in response to isocarbophos, allowing for rapid semiquantitative analysis by the naked eye. After parameter optimization, the multicolor immunoassay was developed depending on the ratiometric absorbance between the Prussian blue and AAP-Fe3+complex. Moreover, a smartphone was used to measure the RGB value of the color conversion for the development of portable visual, quantitative analysis. Both the absorbance-based and RGB-based multicolor immunoassays showed good accuracy and practicability in the recovery test. This study provided a common approach for the development of dual-readout multicolor immunoassay, which can be used for on-site rapid screening by quantitative or visual semiquantitative analysis.

Differential energy pathways are required for rapid long-term memory formation in the oriental fruit fly, Bactrocera dorsalis

Jinxin Yu,Jiayi He,Xuefeng Zhang,Chuxiao Lin,Shiyan Liu,Xin Gong,Xinnian Zeng,Jiali Liu

To ensure the reliability of learned information, most insects require multiple intervals of experience before storing the information as Long-term memory (LTM), and this requirement has been validated in insects from the behavioral to the molecular level. Recent studies have shown that some insects can form LTM after a single experience, although the mechanisms underlying one-trial LTM formation are not well understood. Therefore, understanding the mechanisms underlying rapid learning and subsequent preference formation in insects is crucial. Here we show that the agricultural pest Bactrocera dorsalis can rapidly form LTM, which is dependent on protein synthesis, and that the formation of LTM requires high energy support at the cost of reduced survival. Furthermore, based on a liquid chromatography-mass spectrometry (LC-MS) metabolomics approach, we found that LTM-related processes are sequentially coupled to two processes for energy generation, the TCA cycle and oxidative phosphorylation. This was further confirmed by blocking these energy generation processes. Our results provide a theoretical basis for the development of behavioral modulators in oriental fruit flies that target energy generation intermediate metabolites, as well as a new perspective on the rapid formation of LTM in insects.

A combination of cuminaldehyde with citral- / trans-2-hexen-1-al alleviates green mold disease in citrus fruit

Reymick Okwong Oketch,Duan Bin,Feng Zhao,Zhang Yong Hua,Ouyang Qiuli,Kaaya Archileo N.,Tao Nengguo

Management of Penicillium digitatum requires use of highly effective fungicides that are not prone to drug resistance and are safe to humans and the environment. This study examined the fungicidal potential of combinations of cuminaldehyde (CA) + trans-2-hexen-1-al (TH) and CA + Citral. Both CA + TH and CA + Citral exhibited enhanced fungitoxicity against P. digitatum than CA, citral or TH alone. The minimum inhibitory concentration (MIC) of CA + Citral consisted of 0.13 ml/L CA and 0.3 ml/L citral, representing a synergistic interaction. MIC of CA + TH consisted of 0.12 ml/L CA and 0.15 ml/L TH, representing an additive interaction. The minimum fungicidal concentration (MFC) of CA + Citral and of CA + TH consisted of 0.26 ml/L CA and 0.6 ml/L citral, and 0.18 ml/L CA and 0.24 ml/L TH, respectively. 10×MFC CA + Citral in Tween-80 (WCI) delayed onset of hyphal growth in fruits inoculated with P. digitatum to day 5 compared to day 4 in the 10×MFC CA + TH in Tween-80 (WCT) treatment, and day 3 for the control samples. Daily green mold disease incidence and disease severity were much lower in WCI and WCT treatments than in the 10×MFC CA in Tween-80 (WCA) treatment and in control samples. Our results show that WCI was much fungitoxic against P. digitatum than WCT and WCA, its effect being comparable to that of imazalil. Additionally, at 25 ± 2 °C and 85–90% relative humidity, both WCI and WCT markedly preserved fruit quality by day 6 of treatment. WCI and WCT are thus, safe, cheap and highly effective alternatives to chemical fungicides being used to control postharvest green mold in citrus fruits.

Pectin/zinc alginate films containing anthocyanins from dragon fruit peel as intelligent pH indicators for shrimp freshness monitor

Chang Liu,Ning Li,Xinyu An,Xu Li,Zhiming Liu

The novel incorporation of dragon fruit peel extract (DE), rich in anthocyanins, Zn 2+ (from Zinc Alginate) and pectin was applied to create active and intelligent food packaging composite films. These films were characterized for their microstructure and properties. Various levels of anthocyanin extracts (1 %, 3 %, and 5 %) were evaluated for their impact on the films' physical and functional properties, incorporating microstructure, mechanical strength, barrier properties, pH sensitivity, and bacteriostatic effectiveness. The films exhibited a significant antibacterial rate of up to 99.99 % against common foodborne pathogens, enhanced flame retardancy with an enhancement of 32.7 %, and a broad pH sensitivity range, indicating their adaptability to various conditions. The results demonstrated that the prepared indicator film achieved a 50 % reduction in water vapor permeability. Additionally, the mechanical properties were enhanced, with only a slight decrease of 12.2 % in tensile strength and 14.0 % in elongation at break. In tests monitoring shrimp freshness, pectin/ZA/DE films showed notable color changes correlating with shrimp quality. These specific values highlight the pectin/ZA/DE films' potential for real-world applications, suggesting that they have potential applications as smart packaging materials in the food industry.

The screened triterpene acids from Loquat fruit by CysLTR1-immobilized column could serve as alternative anti-inflammatory agents

Yang Tian,Yan YiFan,Xu YiYing,Chen YiLong,Zhi DaLong,Zhang Yan,Xiao ChaoNi

Background Loquat (Eriobotrya japonica Lindl.) fruit has been used as a heath food and folk medicine for preventing and treating the respiratory tract inflammation diseases, however, the active compounds responsible for anti-inflammation effects remain unclear. Methods Cysteinyl leukotriene receptor-1 (CysLTR1) has become an important target protein for developing therapeutics to manage the inflammation-associated diseases. Herein, a rapid approach was established by immobilizing CysLTR1 onto microspheres for the column packing, and it was applied in screening the active compounds from Loquat fruit. Results The screened compounds targeting CysLTR1 were identified as corosolic acid, ursolic acid and oleanolic acid in combination with Mass Spectrum. The high-dose combination of the three ones exhibited the stronger anti-inflammatory activity than triterpene acid alone and the Loquat extract, which can significantly decrease the counts of inflammatory cells and inhibit the production of pro-inflammatory mediators as well as increase the secretion of anti-inflammatory mediator in the LPS-inflamed mice. It was proved that three triterpene acids exhibited anti-inflammatory activities by the attenuation of CysLTR1 expression. Conclusion sOur results indicated that CysLTR1-immobilized column was a rapid and effective tool to seek out the target-specific components from herbal extract, and the screened triterpene acids could serve as alternative anti-inflammatory agents to Loquat fruit.

Rapid determination of organophosphorus insecticides residue in fruit juice by a fluorescence sensor based on carbon quantum dots

Xianqi Wang,Jie Wang,Beibei Hu,Yuyue Zhang,Hongzhe Tian

Nitrogen doped carbon quantum dots (N-CQDs) were synthesized by a hydrothermal method. A fluorescence sensor for rapid determination of organophosphorus pesticides residue was developed using the N-CQDs based on acetylcholine enzyme inhibition. Cu 2+ effectively quenched the strong fluorescence of N-CQDs, which could be recovered due to the complexation between copper ions and thiocholine, the hydrolysis product of acetylthiocholine catalyzed by acetylcholinesterase. Organophosphorus pesticides inhibited the activity of acetylcholinesterase, resulting in fluorescence quenching of N-CQDs. Fluorescence inhibiting efficiency depends on the concentration of the analytes, and therefore, the sensor could be used for quantitative analysis of organophosphorus pesticides residue. The average recoveries of the analytes ranged from 85.33 % to 110.67 % at the spiked concentrations of 0.015 mg/L, 0.04 mg/L and 0.1 mg/L, and the limits of detection were 2.89 ∼6.40 ng/mL. The developed method had the advantages of simple operation, fast speed, low cost, expensive apparatus-free, and environmental friendliness, which could be used for rapid screening of organophosphorus pesticides residue in real samples.

Antifungal properties and mode of action of natamycin against Monilinia fructicola causing brown rot in Shengzhou nane fruit

Yiwei Mo,Qing Zeng,Jianping Ye,Wenlong Jin,Tianrong Guo

Brown rot caused by Monilinia fructicola ( M. fructicola ) severely affects Shengzhou nane ( Prunus salicina var. taoxingli ). Natamycin (NAT), a natural antimicrobial preservative, has demonstrated good efficacy in mitigating post-harvest decay and maintaining fruit quality. This study investigated antifungal efficacy and underlying mechanism of NAT against M. fructicola in vitro and in vivo settings. In vitro assay demonstrated that NAT exhibited potent antifungal activity against mycelial growth and spore germination of M. fructicola , with efficacy increasing in correlation with higher concentrations of NAT. Specifically, 20.0 mg L −1 NAT induced notable mycelial and spore deformation, characterized by twisting and breakage, while 25.0 mg L −1 NAT completely inhibited mycelial growth and spore germination. NAT exposure suppressed antioxidative enzyme activity, for example, 20 mg L −1 NAT decreased SOD activity by 61.49% of the control at 24 h, and downregulated related gene expression, leading to intracellular accumulation of ROS and oxidative stress. Furthermore, NAT disrupted the integrity of cellular membrane, resulting in leakage of intracellular contents. For instance, DNA leakage increased by 3.2-fold after 24 h. NAT exposure damaged mitochondrial structural and impaired its function, as indicated by the decreased adenosine triphosphate content, by 58.54% of the control, lower energy charge levels, and the reduced activities of malate dehydrogenase, succinate dehydrogenase, Ca 2+ /Mg 2+ -ATPase, and Na + /K + -ATPase by 79.67%, 77.57%, 81.34%, and 89.79% of the control, respectively, following a 24 h exposure. In vivo experiments indicated that NAT treatment resulted in 93.30% reduction in disease incidence at concentration of 200 mg L −1 , as well as a decrease in lesion diameter in Shenzhou nane fruit. Additionally, NAT enhanced the activities of antioxidative enzymes, including superoxide dismutase, catalase, and peroxidase a significant reduction in malondiadehyde levels. NAT also promoted the activity of phenylalanine ammonia-lyase, chitinase, β-1, 3-glucanase, and polyphenol oxidase. The alleviation of oxidative stress and enhancement of defense enzyme activity together increased fruit resistance to M. fructicola . Consequently, higher fruit quality was achieved during storage, thus, 200 mg L −1 NAT increased total soluble solids by 2.42 folds of the control. In conclusion, our findings demonstrated that NAT exhibited a dual-action effect against antibacterial M. fructicola , acting both as a direct antifungal agent and an inducer of host fruit's innate defense mechanism. These findings highlight the potential of NAT treatment as an effective strategy for controlling brown rot caused by M. fructicola in Shengzhou nane fruit
