NIR induced self-healing electrical conductivity polyurethane/graphene nanocomposites based on Diels−Alder reaction

Changhong Lin,Dekun Sheng,Xiangdong Liu,Shaobin Xu,Fance Ji,Li Dong,Yan Zhou,Yuming Yang

In this work, a crosslinker terminal maleimide groups (EDM) has been synthesized and was used in reactions with the pendant furan groups of polyurethane (PUE) to prepare the self-healing electrically conductive composites based on Diels-Alder reaction, and Ag nanowires (AgNWs) as the conductive network inlaid in the surface layer of the polymer matrix layer. Inspired by the graphene-based materials for its versatility in both material and chemical properties, and we present a new synthetic strategy for the preparation of functional graphene nanosheets with DA group that can be employed in the self-healing polyurethane system. To our best knowledge, it hasn't been reported yet the structure graphene oxide materials (rmGO) in any systems. The rmGO served as fillers to reinforce the composites and would participated in the DA reactions. The excellent NIR light-induced self-healing property of prepared composites was realized through the photo-thermal effect of rmGO. The composite is capable of both structural and electrical healing via NIR irradiation. The injured samples can be locally repaired with high precision and efficiency without an obvious influence on those uninjured parts. The underlying polymer matrix can reform via the dynamic dissociation/recombination of Diels-Alder bonds to bring the separated areas of the AgNWs layer into contact to restore the conductivity. The NIR light-induced self-healing electrically conductive composites have wide potential for many fields, e.g., soft robots, flexible devices, and health-monitoring.

Construction of Hierarchical Polymer Brushes on Upconversion Nanoparticles via NIR-Light-Initiated RAFT Polymerization

Zhongxi Xie,Xiaoran Deng,Bei Liu,Shanshan Huang,Pingan Ma,Zhiyao Hou,Ziyong Cheng,Jun Lin,Shifang Luan

Photoinduced reversible addition–fragmentation chain transfer (RAFT) polymerization generally adopts high-energy ultraviolet (UV) or blue light. In combination with photoredox catalyst, the excitation light wavelength was extended to the visible and even near-infrared (NIR) region for photoinduced electron transfer RAFT polymerization. In this report, we introduce for the first time a surface NIR-light-initiated RAFT polymerization on upconversion nanoparticles (UCNPs) without adding any photocatalyst and construct a functional inorganic core/polymer shell nanohybrid for application in cancer theranostics. The multilayer core–shell UCNPs (NaYF4:Yb/Tm@NaYbF4:Gd@NaNdF4:Yb@NaYF4), with surface anchorings of chain transfer agents, can serve as efficient NIR-to-UV light transducers for initiating the RAFT polymerization. A hierarchical double block copolymer brush, consisting of poly(acrylic acid) (PAA) and poly(oligo(ethylene oxide)methacrylate-co-2-(2-methoxy-ethoxy)ethyl methacrylate) (PEG for short), was grafted from the surface in sequence. The targeting arginine–glycine–aspartic (RGD) peptide was modified at the end of the copolymer through the trithiolcarbonate end group. After loading of doxorubicin, the UCNPs@PAA-b-PEG-RGD exhibited an enhanced U87MG cancer cell uptake efficiency and cytotoxicity. Besides, the unique upconversion luminescence of the nanohybrids was used for the autofluoresence-free cell imaging and labeling. Therefore, our strategy verified that UCNPs could efficiently activate RAFT polymerization by NIR photoirradiation and construct the complex nanohybrids, exhibiting prospective biomedical applications due to the low phototoxicity and deep penetration of NIR light.

An NIR-responsive hydrogel loaded with polydeoxyribonucleotide nano-vectors for enhanced chronic wound healing

Yanzhen Sun,Yao Li,Xiaokang Ding,Pan Xu,Xiaodong Jing,Hailin Cong,Hao Hu,Bing Yu,Fu-Jian Xu

Chronic diabetic wounds are difficult to treat due to imbalanced inflammatory responses, high blood glucose levels, and bacterial infections. Novel therapeutic approaches based on nucleic acid analogues have been proposed, with unique advantages in improving angiogenesis, increasing collagen synthesis, and exerting anti-inflammatory effects. However, the inherent electronegativity of nucleic acids makes them less susceptible to cellular uptake. In this paper, a kind of near infrared (NIR)-responsive nanocomposite hydrogel loaded with nucleic acid vectors was proposed for promoting wound healing. The redox system composed of molybdenum disulphide nanosheets (MoS 2 NSs) initiated the copolymerization of quaternized chitosan containing double bonds and N -isopropylacrylamide (NIPAAm) to form the matrix. In addition, MoS 2 NSs with photothermal conversion performance endow the nanocomposite hydrogel to have NIR-response property and act as physical crosslinking points in the matrix. Polydeoxyribonucleotides (PDRN), which have the effect of promoting wound healing, were made into nucleic acid vectors, and loaded into the NIR-responsive hydrogel. MoS 2 NSs can convert NIR irradiation into heat, causing phase transitions of temperature-sensitive segments that trigger volume contraction of the hydrogel to extrude the nucleic acid vector. Promoting angiogenesis, slowing inflammation, and guiding tissue regeneration were demonstrated in the diabetic wound model treated with the NIR-responsive nanocomposite hydrogel.

An endoplasmic reticulum-targeted NIR fluorescent nanoprobe for early diagnosis of glaucoma in vivo

Yu Lan,Haixia Zhang,Ruichen Xie,Kecheng Li,Feiran Xie,Peihong Xiao,Li Jiang

Glaucoma is a leading cause of irreversible blindness worldwide and characterized by progressive loss of retinal ganglion cells (RGCs). As glaucoma progresses asymptomatically and the lack of specific biomarkers or noninvasive imaging tools during the early stage, it is often discovered at a relatively late stage without cure. Development of an effective tool for early diagnosis remains a necessity. As well known, endoplasmic reticulum (ER) stress and subsequent accumulation of reactive oxygen species (ROS) play a vital role in the development of glaucoma, with peroxynitrite (ONOO − ) as one of the most abundant cytotoxic ROS. In this work, we constructed a novel near-infrared (NIR) fluorescent nanoprobe named ER-ONOO-NPs. It specifically and sensitively targets ER-associated ONOO − with low biotoxicity in cultured RGCs. Systematically applied to the NMDA-induced mouse model of glaucoma, ER-ONOO-NPs performs well for real-time dynamic monitoring of ONOO − fluctuations at the Ganglion cell layer of mice prior to RGCs apoptosis. Thus, our results suggest that the ER-targeted NIR fluorescent nanoprobe could be a noninvasive imaging tool for early diagnosis of glaucoma in high-risk population.

APE1-Activated and NIR-II Photothermal-Enhanced Chemodynamic Therapy Guided by Amplified Fluorescence Imaging

Xiaofeng Bi,Jinyue Feng,Xiyuan Feng,Dianpeng Li,Yumin Wang,Shulin Zhao,Liangliang Zhang

The development of intelligent nanotheranostic technology that integrates diagnostic and therapeutic functions holds great promise for personalized nanomedicine. However, most of the nanotheranostic agents exhibit “always-on” properties and do not involve an amplification step, which may largely limit imaging contrast and restrict therapeutic efficacy. Herein, we construct a novel nanotheranostic platform (Hemin/DHPs/PDA@CuS nanocomposite) by assembling DNA hairpin probes (DHPs) and hemin on the surface of PDA@CuS nanosheets that enables amplified fluorescence imaging and activatable chemodynamic therapy (CDT) of tumors. The cancer-relevant APE1 triggers nucleic acid amplification with DHPs to generate activatable and amplified fluorescence signals for discriminating cancer cells from normal cells. Meanwhile, excessive G-quadruplex/hemin-based DNAzyme are also activated, and they function as Fenton-like catalysts to catalyze the production of highly toxic hydroxyl radicals (•OH) for CDT. Moreover, owing to the excellent photothermal conversion efficiency in the near-infrared-II (NIR-II) window, the PDA@CuS not only improves the catalytic performance of CDT but also furnishes PTT. A remarkable antitumor therapeutic effect is demonstrated both in vitro and in vivo. Therefore, the Hemin/DHPs/PDA@CuS nanocomposite is expected to provide a promising avenue for precise imaging-guided antitumor therapy.

Au/CeO2 Nanozyme Scaffold Boosts Electron and Hydrogen Transfer for NIR-Enhanced Chemodynamic Therapy

Qi Zhong,Kangdong Wang,Gao Pan,Shuping Peng,Cijun Shuai

CeO2 nanozymes have demonstrated the potential to enhance biological scaffolds with chemodynamic therapy. However, their catalytic efficacy is limited by the slow conversion of Ce4+ to Ce3+ and the lack of substrates like H2O2 and H+. To address these challenges, we adopted a dual-pronged strategy that utilized the plasmonic resonance of Au nanoparticles and their glucose-oxidase mimicry to boost electron and hydrogen transfer. Specifically, we integrated Au/CeO2 nanozymes into poly-l-lactic acid scaffolds via selective laser sintering. This conversion of Ce3+ to Ce4+ in the scaffolds enhanced the reduction of H2O2 to a hydroxyl radical, inducing oxidative stress in tumor cells. The Au nanoparticles played a crucial role in boosting the Ce3+/Ce4+ catalytic cycle by providing both the energy and the catalytic substrates. They recycled Ce4+ back to Ce3+ by exploiting plasmonic-induced hot electrons and catalyzed glucose oxidation to supply H2O2 and H+. Our nanoscale and atomic-scale simulations confirmed that the Au/CeO2 hybrid structure utilized near-field coupling to amplify the plasmonic resonance and the Au–O–Ce bridge reduced the electron transfer barrier. Consequently, the Au/CeO2 scaffold decreased the activation energy from 22.57 to 9.92 kJ/mol. These findings highlight the significant promise of the Au/CeO2 nanozyme scaffold for NIR-enhanced chemodynamic therapy.

NIR- and UV-dual responsive amphiphilic copolymer micelles with light-dissociable PAG-side groups

Wang Youpeng,Li Guo,Cheng Ruidong,Zhang Xuan,Jiang Jinqiang

Amphiphilic random copolymer micelle of P(NVP- co -NHPM) that responds to both UV and NIR light was designed and investigated. N -hydroxyphthalimide methacrylate — bearing a photoacid generator functional group was synthesized and used to construct the hydrophobic segment of P(NVP- co -NHPM); the N–O bond can be photocleaved when exposing the polymer aqueous solution to 310 nm UV light irradiation, inducing the micellar dissociation and delivery of loaded substances. Dynamic light scattering (DLS), atomic force microscope (AFM), and transmission electron microscopy (TEM) were used to characterize the micellar dissociation and controlled release processes. Moreover, the upconverting nanoparticles (NaLuF 4 :Gd/Yb/Tm) that can effectively absorb NIR light and convert it into visible and UV light were coloaded into polymer micelles to activate the micellar dissociation and the delivery of loaded cargoes via 980 nm NIR light.

NIR-responsive polypeptide copolymer upconversion composite nanoparticles for triggered drug release and enhanced cytotoxicity

Gang Liu,Na Liu,Linzhu Zhou,Yue Su,Chang-Ming Dong

Near infrared (NIR) light-responsive polymeric nanomedicines might achieve spatiotemporal, pulsatile, and on-demand drug release profiles and are appealing as a noninvasive technology for various clinical therapies. Polypeptide copolymer composite nanoparticles with different amounts of upconversion nanoparticles (UCNPs) were successfully fabricated in aqueous solution, and the NIR sensitivity of the composite nanoparticles was characterized by time-resolved UV-vis spectroscopy, on-line dynamic light scattering, and transmission electron microscopy. The composite nanoparticles disassembled with the help of UCNPs converting NIR light into UV light, and the NIR-responsive time can be tuned by both the loaded amount of UCNPs and the polypeptide chain length. The composite nanoparticles loaded with the anticancer drug doxorubicin (DOX) can release DOX in a controllable and/or pulsatile manner, and the drug release profile can be manipulated by NIR light. The DOX-loaded composite nanoparticles can be quickly internalized by HeLa cells and then release DOX inside the cells, as evidenced by flow cytometry and confocal laser scanning microscopy. After 5 or 10 min of NIR irradiation, the half maximal inhibitory concentration (IC50) for the DOX-loaded composite nanoparticles dropped to 5.08 μg DOX equiv. mL−1 or 2.95 μg DOX equiv. mL−1, respectively, compared to 8.26 μg DOX equiv. mL−1 for the non-irradiated sample, demonstrating a tunable NIR-triggered cytotoxicity. This work provides a versatile platform for the fabrication of NIR-responsive polypeptide copolymer nanomedicines with the potential for on-demand drug delivery and cancer therapy.

pH- and NIR light responsive nanocarriers for combination treatment of chemotherapy and photodynamic therapy

Sheng Wang,Weitao Yang,Jing Cui,Xue Li,Yan Dou,Lin Su,Jin Chang,Hanjie Wang,Xiaodong Li,Bingbo Zhang

The side effects of antitumor drugs and low treatment efficacy are two major challenges of current chemotherapy. To address these issues, we developed a new kind of smart nanocarriers that combine pH-responsive chemotherapy and near-infrared (NIR) light triggered photodynamic therapy. These nanocarriers were based on upconversion nanoparticle (UCN)-loaded folate-conjugated polymeric lipid vesicles (UFPLVs). Merocyanine 540 (MC540), as a photosensitizer, was loaded in the UFPLVs; doxorubicin hydrochloride (DOX), as an antitumor drug, was conjugated to the surfaces of UFPLVs by pH-sensitive hydrazone bonds. The newly developed MC540&DOX–UFPLVs had a nanosized structure with targeting ligand modification, so they had the potential to accumulate into tumor sites via a combination of passive and active targeting effects. An in vitro singlet oxygen test showed that the nanocarriers can generate cytotoxic singlet oxygen successfully under the irradiation of NIR light. In vitro DOX release profiles demonstrated that the nanocarriers can achieve a pH-triggered drug release. It has been demonstrated by a cellular uptake study that the nanocarriers can efficiently deliver drugs and photosensitizers into tumor cells. In vitro and in vivo combination treatments evidenced the high antitumor effects of MC540&DOX–UFPLVs under NIR light irradiation. These results suggest that the MC540&DOX–UFPLVs may be promising nanocarriers for tumor combination therapy applications.

Enhanced-permeability delivery system for hydroxyl radical-responsive NIR-II fluorescence-monitored thrombolytic therapy

Nan Lv,Shaodong Zhai,Jun Xiong,Nan Hu,Xiang Guo,Zhida Liu,Ruiping Zhang

Pathological thrombus can cause serious acute diseases that present a significant threat to human health, such as myocardial infarction and stroke. Challenges remain in achieving effective thrombolysis and real-time monitoring of therapeutic effects while minimizing side effects. Herein,a multifunctional nanoplatform (TG-OPDEA@UK/MnO 2 -H1080) with enhanced thrombus-permeability was developed to monitor the therapeutic effect of antioxidant-thrombolysis by hydroxyl radical-responsive NIR-II fluorescence imaging. The polyzwitterion poly (oxidized N,N-Diethylaminoethyl methacrylate- co -n-butyl methacrylate) (OPDEA) was prepared as the matrix of nanoparticles to simultaneously loading urokinase (UK) and MnO 2 QDs, as well as NIR-II fluorescent molecule, H-1080. Subsequently, the fibrin targeted peptide CREKA was modified on the surface of the nanoparticles. OPDEA exhibits efficient loading capacity while endowing nanoparticles with the ability to effectively increased penetration depth of UK by 94.1 % into the thrombus, for extensive thrombolysis and fluorescence monitoring. The loaded UK exhibited good thrombolytic effect and greatly reduced the risk of bleeding by 82.6 %. TG-OPDEA@UK/MnO 2 -H1080 showed good thrombolytic efficacy and specific thrombus monitoring in the mouse carotid artery thrombosis model induced by ferric chloride (FeCl 3 ). This work prepares a nanoplatform for thrombolytic therapy and real-time efficacy assessment based on an independent externally forced thrombus penetration delivery strategy.

De Novo siRNA nanoplatform with NIR-II imaging for treatment of carotid atherosclerotic plaques

Jialu Gao,Hua Zhu,Huanhuan Cai,Qihang Ding,Xiaoxing Xiong,Tong Zhang,Bo Chen,Wumei Wang,Xinpeng Ju,Jianying Huang,Xiaodong Zeng,Jong Seung Kim,Xuechuan Hong

Reducing foam cell formation and accumulation within plaques is a critical strategy for treating atherosclerosis. Although current lipid-lowering therapies have significantly improved the management of atherosclerosis, the risk of cardiovascular events remains. Thus, it is necessary to explore next-generation therapies to reduce foam cell formation, inflammation, and oxidative stress. To address these challenges, we present an innovative near-infrared II (NIR-II) fluorescent dye coupled with a lesion-targeting peptide, forming a molecular self-assembly nanoplatform for effective in vitro small interfering RNA (siRNA) delivery. This platform downregulates Epsin1/2, a family of ubiquitin-binding endocytic adaptors, upregulates the expressions of ABCG1 and LRP-1 proteins, and markedly reduces foam cell formation. Additionally, this system enables real-time tracking of plaque accumulation, therapeutic response, and efficacy through in vivo noninvasive NIR-II imaging. Systemic siRNA delivery significantly suppresses lipid accumulation, plaque size, and necrotic core area in the carotid plaque model of ApoE −/− mice. These results suggest that this multi-functional nanoplatform could become an effective tool for NIR-II image-guided siRNA delivery for personalized treatment of atherosclerosis.

Extending visual range of bacteria with upconversion nanoparticles and constructing NIR-responsive bio-microrobots

Wei Xu,Zhen Liu,Jing Wang,Kai Jin,Lulu Yue,Lin Yu,Luqi Niu,Qingqing Dou,Jinliang Liu,Yuzhe Zhang,Xiaohui Zhu,Yihan Wu

The motility of bacteria is crucial for navigating competitive environments and is closely linked to physiological activities essential for their survival, such as biofilm development. Precise regulation of bacterial motility enhances our understanding of these complex processes. While optogenetic tools have been used to control and investigate bacterial motility, the excitation light in most existing systems are limited to the visible light spectrum. Here, we introduce a new type of bio-microrobot comprising genetically engineered E. coli cells and orthogonally emissive upconversion nanoparticles that can respond to both 980 nm and 808 nm NIR light. This system allows toggling of bacterial states between tumbling and swimming via simply alternating the NIR light between different wavelengths. It is believed that the use of NIR light with deeper tissue penetration suggests potential applications for these bio-microrobots in areas like targeted drug delivery.
