Self-healing, pH/ROS-dual responsive chitosan-phenylboronic acid hydrogel coating for intelligent mango preservation

Yangchun Ge,Zhuochao Shen,Yuying Xie,Jiangkuan Tang,Haohua Zhang,Yupeng Qiu,Xuqiang Peng,Lanlan Zhai,Zhongyu Li

This study reports the rational design and fabrication of an intelligent, self-healing hydrogel tailored for fruit preservation via the dynamic cross-linking of chitosan (CS) with a synthesized phenylboronic acid-based crosslinker (DMSA-PBA). The strategic introduction of dynamic boronate ester bonds as the key crosslinking nodes is pivotal, granting the material not only rapid gelation (<60 s) and autonomous self-healing capabilities but also dual responsiveness to pH and reactive oxygen species (ROS). The resulting hydrogel network exhibits a suite of exceptional properties: superior mechanical robustness, a high swelling ratio (∼1000%), and outstanding water vapor barrier performance. Furthermore, the inherent synergistic antibacterial activity, stemming from the chitosan and the DMSA-PBA components, was confirmed. When evaluated as a protective coating on mangoes, this multifunctional hydrogel significantly delayed quality deterioration during ambient storage, effectively reducing weight loss and decay incidence while maintaining firmness and acidity. This work underscores the immense potential of dynamic covalent chemistry in designing advanced coatings for sustainable food preservation, presenting a feasible and eco-friendly strategy grounded in a well-defined material design.

Chitosan-Based Double Network Hydrogel with Strong Adhesion and ROS Scavenging for Accelerating Wound Healing

Jingjing Ning,Jie Hu,Qing Li,Chen Xu,Su Chen

Hydrogel is increasingly appreciated as a reliable dressing for wound repair and regeneration due to the established favorable microenvironment for healing. Thus, hydrogel with multiple synergistic functions are of particular interest in biomedicine. Herein, we develop a double network hydrogel with low swelling property, outstanding adhesive capacity, good biocompatibility, excellent anti-bacterial and ROS scavenging capacity, which shows potential for accelerating wound healing. Quaternized chitosan serves as the first network to endow the hydrogel with anti-bacterial capacity (75.18% for E. coli and 93% for S. aureus ). Zwitterionic monomer is copolymerized with acrylic acid to form the second network, which improves the mechanical properties and reduces the swelling behavior (ultralow swelling ratio of 2.42%). Tannic acid and dihydrocaffeic acid contribute excellent tissue adhesion (adhesion strengths of 124.4 kPa to pig skin) and antioxidant capacity (ROS scavenging rate of 96%). Based on these advantages, the hydrogel greatly facilitates the wound closure, as well as promotes epithelialization, collagen deposition and neovascularization.

Oridonin targets PRDX1 to promote apoptosis by inducing ROS-mediated ER stress and modulating autophagy

Ziyang Ding,Sisi Wang,Junhui Chen,Yali Song,Zhou Zhu,Haibo Tong,Ashok Iyaswamy,Peng Chen,Xu Wei,Wei Zhang,Jigang Wang,Chuanbin Yang,Yulin Feng

Oridonin is a bioactive diterpenoid derived from the widely used traditional Chinese medicinal herb Rabdosia rubescens , exhibits broad-spectrum anti-cancer activity, with several derivatives currently in clinical trials. However, the molecular mechanism underlying its anticancer effects, especially its direct target proteins, remain to be fully elucidated. Here, we found that Oridonin promoted intracellular reactive oxygen species (ROS) accumulation, which in turn induced endoplasmic reticulum (ER) stress-mediated apoptosis. Moreover, ER stress was instrumental in inducing autophagy after Oridonin treatment, while blockade of autophagy further exacerbated Oridonin-induced cytotoxicity. Notably, using activity-based protein profiling (ABPP), we identified the anti-oxidant enzyme Peroxiredoxin 1 (PRDX1) as a key direct covalent target of Oridonin. By binding to Cysteine 173 of PRDX1, Oridonin increased intracellular ROS levels. Furthermore, PRDX1 over-expression mitigated, whereas PRDX1 knockdown potentiated, Oridonin-induced ROS accumulation, autophagy, and subsequently apoptosis. Overall, our results indicate that PRDX1 is a direct covalent binding target mediating Oridonin-induced apoptosis. These findings not only provide fresh insights into the core mechanism of Oridonin-induced cytotoxicity, but also highlight PRDX1 as a potential therapeutic target for renal cancer drug development.

A Diazaborine-Based ROS Switch Enables Copper Transfer for Tumor-Specific Prodrug Activation and Therapy

Huang Zeqian,Ding Yaqing,Wang Shengzhi,Lin Zijun,Wang Conghan,Zheng Dong,Zhang Tao,Zhao Chunshun

Reactive oxygen species (ROS) are significantly elevated in tumor cells, providing an intrinsic trigger for selective drug activation and tumor-specific therapy. However, commonly used ROS-responsive linkers, such as thioketals and boronates, often suffer from limited sensitivity or poor physiological stability, and the development of new ROS-responsive motifs remains scarce. Herein, we develop a hydrazide-derived diazaborine (DAB) scaffold and establish it as a general ROS-responsive linker for small-molecule prodrugs, combining physiological stability with efficient ROS responsiveness. Based on this platform, we construct a DAB-derived ROS-responsive copper complex small-molecule prodrug, DDABD@Cu, for tumor-specific therapy. DDABD@Cu integrates a DAB-caged diethyldithiocarbamate (DTC) moiety with an inert copper donor, in which the DAB group effectively blocks the copper-chelating activity of DTC, enabling inert co-delivery of both components. Upon ROS activation, DTC is released to initiate copper transfer, generating cytotoxic Cu(DTC) 2 in situ to induce proteotoxic stress via ubiquitinated protein accumulation while concomitantly producing quinone methide (QM) to deplete glutathione (GSH) and amplify oxidative stress. This cascade improves antitumor efficacy against non-small cell lung cancer while limiting off-target toxicity. Overall, this work provides a feasible strategy for designing ROS-responsive and copper complex prodrugs.

A ROS-Responsive Coordination Nanoplatform Integrates Antibacterial Activity with STING-Activating Immunomodulation to Attenuate Periodontitis

Qiuyu Wang,Qiwei Chen,Di Li,Xueke Jia,Jie Ren,Hezhen Xie,Fei Hu,Xiaoxue Wang,Xueyang Zhang

Antibacterial treatment reduces residual bacterial burden in periodontitis, but rapid bacterial killing and lysis can also increase exposure to danger signals such as bacterial DNA (bDNA), which can contribute to cGAS–STING-associated innate immune activation. To address this bactericidal stress-associated inflammation, we constructed a ROS-responsive coordination nanosystem, CPLEA, in which the STING inhibitor C-176 was incorporated during Ag⁺-mediated co-assembly of epicatechin and the sulfur-containing PEG ligand methoxy poly(ethylene glycol)–dihydrolipoic acid (mPEG-DHLA), thereby integrating antibacterial activity with the regulation of post-bactericidal STING-associated inflammation. Without compromising bacterial killing, CPLEA promoted C-176 release under oxidative conditions, and the release profile showed temporal overlap with bacterial killing and bDNA exposure. CPLEA attenuated the inflammatory state of macrophages and relieved the suppression of hPDLSC osteogenic differentiation by macrophage-conditioned medium. Transcriptomic analysis indicated downregulation of cytosolic DNA sensing, NF-κB, IL-17, and osteoclast differentiation programs. In a ligature-induced mouse model of periodontitis, CPLEA attenuated local inflammation and TRAP-positive osteoclast accumulation and reduced alveolar bone loss. Collectively, these findings support bactericidal stress-associated STING signaling as a relevant target during local antibacterial treatment of periodontitis and position CPLEA as a feasible materials-based approach for this purpose. Statement of significance Periodontitis treatment often requires additional antimicrobial control after mechanical debridement, yet rapid bacterial killing and lysis can increase the exposure of bacteria-derived danger signals and thereby sustain host inflammatory responses. This study develops an oxidation-responsive coordination nanoplatform that integrates antibacterial activity with STING-targeted immunomodulation. By incorporating the STING inhibitor C-176 into an Ag⁺-mediated coordination system, CPLEA enables oxidation-promoted C-176 release within a temporal window overlapping bacterial killing and bacterial DNA exposure. The study further demonstrates that early C-176 intervention more effectively regulates STING-associated inflammatory signaling than delayed intervention. CPLEA attenuated macrophage inflammatory responses, alleviated inflammation-associated suppression of periodontal ligament stem cell osteogenic differentiation, and reduced inflammation, osteoclast accumulation, and alveolar bone loss in experimental periodontitis. These findings highlight the importance of immunomodulator availability during the early phase of bactericidal treatment and provide a biomaterials strategy for integrating residual bacterial control with local host modulation. ROS- and Iron Ion-Responsive Gelatin Modulates the Pathological Microenvironment to Enhance Myocardial Infarction Treatment

Xiaoyu Liang,Xuanling Li,Xue Fu,Kaijing Liu,Changduo Wang,Ni Zhu,Chao Liu,Chunxiao Qi,Jing Yang

Myocardial infarction (MI) generates a pathological microenvironment characterized by ischemia and hypoxia, and this increases mortality and morbidity worldwide. We fabricated p-hydroxyphenylpropionic acid-modified gelatin (GTN), a dual-responsive system capable of in situ gelation in the presence of hydrogen peroxide (H 2 O 2 ) or iron ions. This feature enables GTN to cross-link in the infarcted myocardium, where excessive reactive oxygen species (ROS) and iron ions trigger ferroptosis. The successful modification of gelatin was verified, along with its time- and concentration-dependent responsiveness to H 2 O 2 and iron ions in vitro. We further assessed the biocompatibility, ROS-scavenging activity, and cytoprotective effects of GTN in hostile MI microenvironments. Death receptor 5 fusion protein (DR5) was incorporated into the hydrogel to construct DR5@GTN, aiming to alleviate cardiomyocyte apoptosis. In vivo experiments demonstrated that GTN effectively targeted and accumulated in the infarcted region via transglutaminase-mediated recognition, thereby remodeling the pathological microenvironment. DR5@GTN significantly restored cardiac functions, increased the left ventricular ejection fraction, and reduced the infarct size. DR5@GTN attenuated myocardial fibrosis, promoted angiogenesis, suppressed cardiomyocyte apoptosis, decreased ROS levels, and inhibited myocardial hypertrophy. Collectively, DR5@GTN regulated the “ROS–iron–apoptosis” axis with favorable in situ gelation properties, and this represents a promising strategy for myocardial repair by ameliorating the harsh microenvironment following MI. Statement of Significance ● Adaptively responds to and eliminates ROS and iron ions at the infarcted region, matching the pathological characteristics of the MI microenvironment. ● Fosters angiogenesis in the infarcted region, mediates extracellular matrix remodeling, and synergistically attenuates cardiomyocyte death. ● Represents the first attempt to validate the feasibility of in situ gelation therapy for myocardial infarction via a minimally invasive strategy. Sialylated Macrophage Nano-Decoys Mitigate Inflammatory-ROS Microenvironment and Reprogram Endothelial Function in Myocardial Infarction

Xinman Hu,Chengbin He,Ping Zeng,Liwen Zhang,Beiduo Wang,Shifen Li,Yonglei Zhao,Wenxing Liu,Hongjie Hu,Changyou Gao

Myocardial infarction (MI) induces the infiltration of abundant immune cells, such as macrophages, and thereby elevated inflammation characterized by elevated reactive oxygen species (ROS), which leads to dysfunction of myocardial microvessels and exacerbates myocardial ischemia and necrosis. Intravenously injected drugs hardly remain in the MI region of the heart, whereas hydrogels or cardiac patches need a complicated thoracotomy. In this study, a ROS-scavenging polyurethane nano-decoys (NDs) modified with N-acetylneuraminic acid (sialic acid, SA) (PTSA) is developed for therapy of MI via convenient intravenous injection. Due to the high affinity of SA to macrophages, PTSA is better camouflaged to avoid fast immune clearance, allowing long-term retention in the damaged myocardium. The loaded nicorandil (NIC) could be faster released in response to ROS environment, which in turn scavenged ROS and alleviated the inflammation. The NIC@PTSA effectively inhibited abnormal mitochondrial function and apoptosis of cardiomyocytes in vitro. Under the synergistic effect of ROS-scavenging and NIC release, the function of endothelial cells is reprogrammed, promoting the process of vascularization. Treatment of NIC@PTSA in vivo significantly reduced oxidative stress, promoted angiogenesis, and inhibited adverse ventricular remodeling. These multifunctional NDs provide an effective strategy for MI therapy, especially from the viewpoint of realistic application.

Glycyrrhiza uralensis Fisch. suppresses cell migration via ROS and JAK/STAT signalling pathways in Drosophila

Fangfei Zhou,Qingge Lu,Lingyu Kong,Sitong Wang,Haixia Zhang,Meng Zhao,Yue Hu,Fanwu Wu,Chenxi Wu

Background: Cancer is a global public health crisis and the leading cause of death among middle-aged and older individuals, with its incidence increasingly shifting toward younger populations. Approximately 90% of the patients succumb to advanced metastasis, and effective treatments remain elusive. The specific molecular mechanisms underlying cancer cell invasion and migration remain poorly understood, hindering the development of effective targeted therapies. Therefore, inhibiting or reversing cancer cell invasion and migration may be crucial for reducing mortality. Our previous research revealed that the five drugs (FD), derived from Xuefu Zhuyu Decoction (XFZYD), play a significant role in inhibiting cell migration.Hypothesis/purpose: This study aims to explore the main drug components of FD and investigate the underlying mechanism in inhibiting cell migration. Methods: We used the Drosophila ptc>scrib-IR cell migration model to investigate the effects of FD. FD was disassembled and analyzed using an orthogonal design. Drug extracts were prepared and administered to Drosophila larvae. We assessed the effects of FD on cell migration, reactive oxygen species (ROS) levels, and gene expression. Results: In FD disassembled recipes and orthogonal test design, a significant difference was observed in the intervention with or without Glycyrrhiza uralensis Fisch. (GUF) in migrating cell number (P < 0.01), which emerged as a more potent inhibitor of FD from XFZYD in cell migration. High-performance liquid chromatography revealed that GUF and its extract contained effective medicinal components, namely glycyrrhizic acid, liquiritin, liquiritigenin, and glycyrrhetinic acid. Moreover, GUF at 4.0 mg/mL displayed strong inhibitory effect in migrating cell number and distance when compared with model, XFZYD or FD. Excessive ROS can activate the JAK/STAT signaling pathway and promote the EMT process. GUF inhibited ptc>scrib-IR-induced cell migration by reducing ROS levels, JAK/STAT signalling, and the transcription of upd2, upd3, hop and socs36E. Finally, GUF rescued the altered expressions of the epithelial-mesenchymal transition (EMT)-related proteins, including matrix metalloproteinase 1 (MMP1), β-integrin and E-cadherin, triggered by cell migration. Conclusion: Our findings demonstrate that GUF may serve as a promising candidate for targeting advanced metastatic tumors by suppressing ROS-mediated JAK/STAT signaling and EMT.

Defective RuO2 Nanospheres Attenuate Osteoarthritis Progression via Suppressing the ROS/NLRP3/Caspase-1 Signaling Pathway

Jie Lv,Faheem Muhammad,Zheng Wang,Chenfeng Qiao,Xiang Gu,Yuan Liu,Weitong Li,Wenli Gong,Zhongyang Lv,Yuxiang Fei,Liqian Peng,Zhihao Lu,Nuo Xu,Chunqing Hu,Hanwen Zhang,Rui Wu,Xingquan Xu,Hui Wei,Ziying Sun,Dongquan Shi

Osteoarthritis (OA) is the most prevalent degenerative joint disorder, characterized by chronic inflammation, cartilage degeneration, and the formation of osteophytes, which leads to serious economic and social challenges. Previous studies have demonstrated that oxidative stress-driven inflammation plays a crucial role in the pathophysiological progression of OA. In this study, we presented defective RuO2 (d-RuO2) as an effective antioxidant for the treatment of OA. Unlike the crystalline RuO2, the amorphous hydrous RuO2 nanospheres (composed of self-assembled ultrasmall RuO2) displayed superior nanozymatic antioxidant activities. In vitro studies demonstrated that d-RuO2 significantly reduced intracellular levels of reactive oxygen species (ROS), and decreased the expression of key inflammatory markers including inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX2), tumor necrosis factor-α (TNF-α), and interleukin-1β (IL-1β), indicating its anti-inflammatory effects. In vivo experiments showed that d-RuO2 effectively relieved pain, improved physical activity, and mitigated synovitis, cartilage degeneration, and bone remodeling induced by destabilization of the medial meniscus (DMM). Furthermore, the mechanistic investigations indicated that d-RuO2 attenuated the progression of OA by suppressing the ROS/NLRP3/Caspase-1 signaling pathway. In conclusion, we presented d-RuO2 as an efficient ROS scavenger, providing a potential therapeutic strategy for OA.

Anthocyanins promote the abundance of endophytic lactic acid bacteria by reducing ROS in Medicago truncatula

Junjie Liu,Yuanyuan Huang,Huan Du,Jing Tian,Fan Zhu,Jianguo Zhang,Qing Zhang,Xiaolin Wang,Liangfa Ge

SUMMARY The microbial community residing on the phyllosphere is influenced by many factors, including the host plant's genotype as well as its secondary metabolites. Anthocyanins are a group of flavonoids renowned for their antioxidative properties and are widely distributed across plant tissues. However, the potential impact of anthocyanins on plant-associated microbial communities remains unknown. In the model legume Medicago truncatula , we isolated a mutant named purple leaves ( pl ) that produces purple leaves at a young stage due to over-accumulated anthocyanins. Through sequencing 16S rRNA amplicons of phyllosphere microbes in the pl mutant, we show that anthocyanins significantly enhance the abundance of endophytic lactic acid bacteria within plant leaves. Further in vitro study revealed that anthocyanins derived from pl can significantly promote the growth of lactic acid bacteria under anaerobic conditions. The accumulated anthocyanins in pl leaves reduced reactive oxygen species (ROS), thereby creating a favorable environment for the growth of facultative anaerobic lactic acid bacteria and resultantly increasing the abundance of phyllosphere lactic acid bacteria. Our findings elucidate the role of anthocyanins in modulating the community structure of phyllosphere microbiota in M. truncatula and provide new insights into the relationship between plant secondary metabolites and phyllosphere microbiota.

Integrating autophagy inhibition and ROS clearance in biohybrid nanoparticles for low-temperature cancer photothermal therapy

Suleixin Yang,Ruie Chen,Peng Hua,Yi Wu,Meiwan Chen

Photothermal therapy (PTT) demands efficient cancer ablation at relative low temperatures and minimal thermal damage to normal tissues, but suffers from both the protective autophagy-related thermal resistance in cancer cell and reactive oxygen species (ROS)-induced damage to normal cells. Here, we screened out curcumin-Fe ultrasmall nanoparticles (Cur-Fe) that manifested efficient photothermal conversion efficiency (η = 43.38 %) and ROS scavenging ability. Additionally, CRISPR/Cas9 plasmids (pCas-ATG5/ATG7) were also constructed to safely and precisely knockdown the protective autophagy for thermal resistance alleviation. The core Cur-Fe/ATG@ TK PF (CFA@T), which was composed of anionic C ur- F e and pCas- A TGs, was encapsulated by cationic thioketal-crosslinked and fluorinated polyethyleneimine ( T K PF) via electrostatic interaction. Further, CFA@TC was formed by CFA@T coated with an acidic pH-responsive shell OH C -PEG-CHO via Schiff base. Attributed to its dual responsiveness to pH and ROS, CFA@TC exhibited efficient tumor targeting and uptake following intravenous injection. Upon irradiation with a 652 nm laser, CFA@TC demonstrated enhanced efficacy in eradicating cancer cells by inhibiting autophagy, while concurrently mitigating inflammatory responses through intracellular ROS scavenging in vivo and in vitro . Taken together, our study provides a proof-of-concept that CRISPR can be effective for autophagy inhibition, and its integration with ROS-induced inflammatory responses relief can further improve PTT.

A cyclodextrin/ROS-responsive Pydiflumetofen nanoparticle for enhanced fungicidal activity against Sclerotinia sclerotiorum in soybean

Qian Zhao,Liyan Zhang,Ziyu Wang,Weibo Han,Jianzhong Wu

Sclerotinia stem rot (SSR), caused by Sclerotinia sclerotiorum , significantly reduces soybean yields. Conventional SSR-based pesticide application presents environmental and health hazards, whereas ROS-responsive nanocarrier systems demonstrate superior efficacy as a sustainable alternative. In this study, PYD@OxbCD NPs, encapsulating Pydiflumetofen (PYD), were synthesized and showed ROS-triggered release and strong in vitro activity against SSR. This targeted release mechanism reduces ecological risk by minimizing pesticide exposure to non-target organisms. PYD@OxbCD delivers PYD more precisely to infected soybean tissues, outperforming traditional Pydiflumetofen suspension concentrate (SC) in antifungal efficacy. It also upregulates defense-related genes and reduces disease incidence and severity in soybean plants. PYD@OxbCD also mitigates the phytotoxic effects of PYD SC on seedling growth. Overall, PYD@OxbCD shows strong potential as an effective nanoplatform for managing SSR in soybeans, enhancing disease control and pesticide utilization for sustainable agriculture.
