SERS Fingerprinting of β2-Agonists for Anti-Doping Based on Au-COF Substrate

Siqing Liu,Liping Chen,Xingju Li,Ruxi Lin,Yunxin Zhang,Xiaojun Luo

Background β 2 −agonists, also known as β 2 −adrenoceptor agonists, are secreted by the adrenal medulla of the animal body and have a characteristic catecholamine structure. When inhaled, the drug rapidly relaxes the smooth muscles of the airways in asthma patients, improves ventilation, and temporarily increases alertness during exercise and helps relieve fatigue. However, the use of β 2 −agonists to improve physical performance in a short period of time can lead to extreme dependence and irritation, and there are still many unknown hazards. They are now on the World Anti−Doping Organization (WADA) prohibited list. Results Surface−enhanced Raman spectroscopy (SERS) can directly detect the characteristic peaks associated with β 2 −agonist compounds in a fast and sensitive manner. This study is the first to utilize a direct Raman detection technique, achieving successful acquisition of the fingerprint spectra of three target analytes (vilanterol, fenoterol, bambuterol) as well as the establishment of a corresponding fingerprint spectral analysis method. To further improve its performance, in this study, gold nanoparticles were synthesized via redox methods and integrated with covalent organic framework (COF) to construct a novel highly sensitive detection platform for β 2 −agonists. The Au−COF composite exhibits high adsorption performance and can significantly enhance the SERS signal of the target, thereby achieving low detection limits, including 9.25 × 10 −6 g·mL −1 for bambuterol, 9.76 × 10 −6 g·mL −1 for vilanterol, and 7.54 × 10 −6 g·mL −1 for fenoterol, which makes it promising for improving the overall sensitivity of β 2 −agonist detection. Significance This study develops a novel analytical method utilizing Au−COF composites for highly sensitive SERS detection. It pioneers the application of this strategy in the qualitative and quantitative analysis of trace β2-agonists. The research not only significantly expands the scope of Au−COF composites but also provides a technologically promising approach with substantial practical value for ensuring medical safety through clinical medication monitoring and for detecting banned substances in sports.

Rapid detection of thiram using a dynamic ratiometric in-tube SPME-SERS strategy

Yuxuan Xin,Zixiao Yang,Rubing Liu,Yinan Gai,Wei Wang,Yongchao Lai

A dynamic ratiometric in-tube SPME-SERS platform was built for thiram detection. • Forced convection shortened adsorption equilibrium from ∼90 min to ∼300 s. • SCN − internal standard enabled ratiometric correction with RSD < 10%. • Direct analysis of environmental water achieved recoveries of 90.9%–108.5%. Surface-enhanced Raman spectroscopy (SERS) is a powerful tool for pesticide detection but is hindered in field applications by slow analyte mass transport and quantitative irreproducibility. Herein, we report an in-tube solid-phase microextraction-SERS (in-tube SPME-SERS) strategy that simultaneously overcomes these bottlenecks. A silver-nanoplate-decorated copper wire was synthesized via galvanic replacement, functionalized with thiocyanate (SCN − ) as a covalently bonded internal standard (IS), and assembled into a fused-silica capillary. Hydrodynamic flow compresses the diffusion boundary layer, accelerating the extraction equilibrium from ∼90 min (static mode) to ∼300 s. The SCN − internal standard (2136 cm −1 ) enables ratiometric correction ( I 1380 /I 2136 ) for thiram quantification, effectively compensating for substrate heterogeneity and instrumental drift. Mechanistic studies reveal a competitive adsorption process where thiram partially displaces SCN − , ensuring precise analyte-IS co-localization. This method exhibits excellent analytical performance for thiram, including a wide linear range (20–1000 nM), a low limit of detection (33.8 nM), and high reproducibility (RSD < 10%). Furthermore, direct analysis of surface and groundwater samples without pretreatment yields satisfactory recoveries (90.9%–108.5%). With robust 28-day stability, this ratiometric in-tube SPME-SERS platform provides a reliable, kit-based solution for on-site environmental monitoring. Assembly Nanoparticle Arrays Decorated with Ultrathin AAO Nanopores for Highly Sensitive SERS Substrate

Dong Jun,Li Chenlu,Wang Yan,Cao Yi,Fan Yimeng,Han Qingyan,Gao Wei,Wang Yongkai,Qi Jianxia

Surface-enhanced Raman scattering (SERS) is widely employed because it offers quick, microscopic, and traceless detection. This study used high-voltage and low-voltage ultrasonic oscillation to embed gold-silver nanoparticles (Au-Ag NPs) into the pores of chemically modified ultrathin anodic alumina (AAO) films, resulting in a highly sensitive three-dimensional SERS substrate. We improved the substrate’s stability and Raman activity by adjusting the particle alloy ratio. For the substrate in this alloy ratio, the Raman signal of probe molecules (Rh6G) adsorbed on the substrate surface is enhanced. the enhancement factor (EF) was as high as 1.40 × 10 7 , the relative standard deviation (RSD) of 10.6%, and the concentration of Rh6G shows a linear relationship with Raman intensity, with a linear correlation coefficient of 0.961. In addition, we evaluated the substrate’s detection effect on thiram molecules. It has been proven that this structure has good practicality and high sensitivity as a Raman enhanced substrate.

Four-Color SERS Monitoring of Size-dependent Nanoparticle Delivery in the Same Tumor

Chang Liu,Jianguo Dong,Zedong Zhang,Kexin Fu,Dekun Wang,Xue Mi,Shijing Yue,Xiaoyue Tan,Yuying Zhang

The physicochemical properties of nanoparticles (NPs) significantly influence their deposition at the disease site, ultimately impacting the overall therapeutic efficacy; however, precisely assessing the effects of various factors on NP accumulation within a single cell/tumor tissue is challenging due to the lack of appropriate labeling techniques. Surface-enhanced Raman spectroscopy (SERS) tag is a powerful encoding method that has recently been intensively employed for immunodetection of biomarkers. Herein, we introduce a multiplexed SERS tracking approach for systematic investigation of size-dependent accumulation and distribution of NPs within the same tumor. Four-sized (34, 60, 108, and 147 nm) NPs encoded with different SERS “colors” were fabricated, mixed, and incubated with monolayer tumor cells, multicellular tumor spheroids, or injected into mouse models bearing xenograft solid tumors in a single dose. Multicolor SERS detection of the specimens revealed that NP accumulation in tumor cells, tumor spheroids, and solid tumors was in the order of 34 nm > 60 nm > 108 nm > 147 nm, 60 nm > 34 nm > 108 nm > 147 nm, and 34 nm > 147 nm > 108 nm > 60 nm, respectively. Inductively coupled plasma mass spectroscopy determination performed in parallel samples were in alignment with the four-color SERS probing results, demonstrating the effectiveness of this multiplexed evaluation assay. Furthermore, in combination with fluorescence labeling of specific biomolecules, this method can be applied for the colocalization of different NPs in various pathological structures and provide additional information for analysis of the possible mechanisms.

Metal-organic framework template-guided electrochemical lithography on substrates for SERS sensing applications

Lu Youyou,Zhang Xuan,Zhao Liyan,Liu Hong,Yan Mi,Zhang Xiaochen,Mochizuki Kenji,Yang Shikuan

The templating method holds great promise for fabricating surface nanopatterns. To enhance the manufacturing capabilities of complex surface nanopatterns, it is important to explore new modes of the templates beyond their conventional masking and molding modes. Here, we employed the metal-organic framework (MOF) microparticles assembled monolayer films as templates for metal electrodeposition and revealed a previously unidentified guiding growth mode enabling the precise growth of metallic films exclusively underneath the MOF microparticles. The guiding growth mode was induced by the fast ion transportation within the nanochannels of the MOF templates. The MOF template could be repeatedly used, allowing for the creation of identical metallic surface nanopatterns for multiple times on different substrates. The MOF template-guided electrochemical growth mode provided a robust route towards cost-effective fabrication of complex metallic surface nanopatterns with promising applications in metamaterials, plasmonics, and surface-enhanced Raman spectroscopy (SERS) sensing fields.

Chemically renewable SERS sensor for the inspection of H2O2 residue in food stuff

Xin Tian,Yun Qin,Yuning Jiang,Xiaoyu Guo,Ying Wen,Haifeng Yang

Hydrogen peroxide (H 2 O 2 ) residue in foodstuffs will bring great harm to human health. We immobilize the composite of the reduced polyaniline (PANI R ) modified gold nanoparticles on the surface of ITO (ITO/AuNPs/PANI R ) to develop surface-enhanced Raman scattering (SERS) sensor for H 2 O 2. detection. The principle is that PANI R is oxidized by H 2 O 2 to generate a new SERS peak at 1460 cm −1 for realizing quantitative analysis of H 2 O 2 . Fe 2+ -Fenton reaction is introduced to catalytically react with H 2 O 2 to hydroxyl radical, which speeds up the oxidation of PANI R . Before SERS detection, acidic treatment could guarantee the reduced state of PANI R in composite. Limit of detection of ITO/AuNPs/PANI R -based SERS assay for H 2 O 2 is down to 1.78 × 10 −12 mol/L and a good linear relationship from 1 × 10 −10 to 3.16 × 10 −7 mol/L is achieved. Furthermore, the SERS sensor could be regenerated by acidic treatment. As a scenario, the renewable SERS sensor is utilized to monitor H 2 O 2 residues in food and environmental samples.

Au nanorod assembly for sensitive SERS detection of airway inflammatory factors in sputum

An-qi Yang,Wentong Zheng,Xiaoyang Chen,Jiayin Wang,Shuang Zhou,Hongzhi Gao

In this paper, we demonstrate a surface-enhanced Raman spectroscopy (SERS) biosensor based on the self-assembly of gold nanorods (AuNRs) for the specific detection of airway inflammatory factors in diluted sputum. The AuNR surface was modified with an antibody that was able to specifically recognize an airway inflammatory factor, interleukin-5 (IL-5), so that a end-to-end self-assembly system could be obtained, resulting in an order of magnitude amplification of the Raman signal and greatly improved sensitivity. Meanwhile, the outer layer of the biosensor was coated with silicon dioxide, which improved the stability of the system and facilitated its future applications. When the detected concentration was in the range of 0.1–50 pg/mL, the SERS signal generated by the sensor showed a good linear relationship with the IL-5 concentration. Moreover, it had satisfactory performance in diluted sputum and clinical subjects with asthma, which could achieve sensitive detection of the airway inflammatory factor IL-5. Overall, the developed biosensor based on the SERS effect exhibited the advantages of rapid and sensitive detecting performance, which is suitable for monitoring airway inflammatory factors in sputum.

LoC-SERS platform for rapid and sensitive detection of colorectal cancer protein biomarkers

Xiaowei Cao,Zhengqing Liu,Xiaogang Qin,Yuexing Gu,Yong Huang,Yayun Qian,Zhenguang Wang,Hongbo Li,Qunshan Zhu,Wei Wei

Colorectal cancer (CRC) remains a significant contributor to the global mortality rate, and a single biomarker cannot meet the specificity required for CRC screening. To this end, we developed a multiplexed, pump-free surface-enhanced Raman scattering (SERS) microfluidic chip (LoC-SERS) using a one-step recognition release mechanism; the aptamer-functionalized novel Au nanocrown array (AuNCA) was used as the detection element embedded in the detection zone of the platform for rapid and specific detection of protein markers in multiple samples simultaneously. Here, the corresponding aptamer specifically captured the protein marker, causing the complementary strand of the aptamer carrying the Raman signal molecule to be shed, reducing the SERS signal. Based on this platform, sensitive and specific detection of the target can be accomplished within 15 min with detection limits of 0.031 pg/mL (hnRNP A1) and 0.057 pg/mL (S100P). Meanwhile, the platform was consistent with ELISA results when used to test clinical. By substituting different aptamers, this platform can provide a new solution for the rapid and sensitive detection of protein markers, which has promising applications in future disease detection.

In situ and dynamic SERS monitoring of glutathione levels during cellular ferroptosis metabolism

Xu Lixing,Du Xing,Liu Tianqing,Sun Dan

Ferroptosis is a non-apoptotic cell death regulated by iron-dependent lipid peroxidation. Glutathione (GSH), a key antioxidant against oxidative damage, is involved in one of the most important metabolic pathways of ferroptosis. Herein, an excellent plasmonic nanoprobe was developed for highly sensitive, in situ, dynamic real-time monitoring of intracellular GSH levels during ferroptosis. A nanoprobe was prepared by functionalizing gold nanoparticles (AuNPs) with the probe molecule crystal violet (CV). The fluctuation in the SERS signal intensity of CV via the competitive displacement reaction can be used to detect GSH. The advantages of the plasmonic nanoprobe including low-cost production techniques, outstanding stability and biocompatibility, high specificity and sensitivity towards GSH with a detection limit of 0.05 μM. It enables real-time dynamic monitoring of GSH levels in living cells during erastin-induced ferroptosis. This approach is expected to provide important theoretical support for elucidating the GSH-related ferroptosis metabolic mechanism and advancing our understanding of ferroptosis-based cancer therapy. Graphical abstract Overview of the workflow of sensing principle for highly sensitive, in situ and dynamic tracking of intracellular GSH levels during drug-triggered ferroptosis process.

Highly sensitive SERS cytosensor based on catalytic hairpin assembly-mediated zipper-like structures

Zhengying Peng,Youyuan Chen,Pengkun Yin,Fan He,Daibing Luo,Yixiang Duan,Qingwen Fan,Zhimei Wei,Qingyu Lin

Circulating tumor cells (CTCs) are cancer biomarkers for the diagnosis at early stage. However, it’s still a great challenge to develop the cytosensor for CTCs detection due to its ultralow counts in human peripheral blood. Herein, a highly sensitive cytosensor based on surface-enhanced Raman scattering (SERS) was developed. The tumor cells were first identified by a strand replacement reaction (SDR) that released the initiator of catalytic hairpin assembly (CHA) reaction. Subsequently, the initiator opened the residue hairpin DNA outside the silver coated gold nanoparticles ( [email protected] NPs). The double strand DNA (dsDNA) CHA products drove the [email protected] NPs towards the same side of dsDNA to form a zipper-like structure. This structure brought about hotspots area for SERS quantitative analysis of A549 tumor cells. The designed cytosensor exhibited good linearity ranging from 10 to 500 cells/mL with a calculated limit of detection (LOD) of 8 cells/mL. A SERS cytosensor with high sensitivity and good reproducibility was obtained in this work. In addition, this research presents a new method for rapid tumor cell detection in biological samples for cancer diagnosis.

LoC-SERS Platform Integrated with the Signal Amplification Strategy toward Parkinson’s Disease Diagnosis

Xiaowei Cao,Shengjie Ge,Miao Chen,Haiyan Mao,Ying Wang

Multiplexed detection of Parkinson’s disease (PD) biomarkers is of great significance for early diagnosis and personalized treatment. In this study, we fabricated a robust surface-enhanced Raman scattering-enabled lab-on-a-chip (LoC-SERS) platform for simultaneous quantification of α-synuclein, phosphorylated tau protein 181, osteopontin, and osteocalcin. Herein, the antibody–DNA conjugate was designed to introduce the catalytic hairpin self-assembly (CHA) amplification into the protein detection. Au nano-stars (AuNSs) modified with Raman reporter molecules and hairpin-structure DNA 1 were applied as the SERS nanotags. Au-coated silicon nanocone array (Au/SiNCA) fabricated based on the maskless plasma etching-prepared high-density Si nanocone array (SiNCA) and surface ion sputtering was used as the capture substrate after the modification of hairpin-structure DNA 2. Benefitting from the antibody–DNA conjugate-induced CHA amplification, numerous AuNSs can be connected to the Au/SiNCA surface, which significantly amplify the plasmonic coupling effect for ultrasensitive SERS detection, and the limit of detection was less than the pg/mL level. The application of highly uniform Au/SiNCA and antibody–DNA conjugate endows the LoC-SERS platform excellent analytical performance, including superior reproducibility, satisfactory universality, and high sensitivity. In addition, a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice model was established, and satisfactory results were obtained in real sample analysis with the LoC-SERS platform, which may be enlightening for exploiting protein biomarkers in PD monitoring.
