Curcumin-Loaded Silver-Based Metal–Organic Frameworks: Efficient Antibacterial and Antioxidant Properties against Escherichia coli and Staphylococcus aureus for Promoting Infected Wound Healing

Jinyan Du,Jinrui Hou,Shiji Liu,Xinyue Wu,Liangde Hu,Wenjiang Xu,Shujuan Zhuo

Slow or noticeably delayed wound healing is frequently intimately linked to bacterial infection and excessive reactive oxygen species (ROS), while the inappropriate usage of antibiotics fuels the rise of bacterial resistance. The innovative materials are desperately needed to eliminate bacteria and effectively accelerate wound healing. In this work, a curcumin-loaded silver-based metal–organic framework (Cur/Ag-MOF) composite nanomaterial was developed, which exhibited good antimicrobial activity, biocompatibility, and drug resistance. Meanwhile, surface-loaded curcumin can effectively eliminate excess free radicals and promote wound healing due to its antioxidative and ROS scavenging properties. Additionally, it was discovered that the application of Cur/Ag-MOF to the site of skin trauma significantly sped up the process of wound closure in mice used as subjects. These findings highlighted its great potential for treating bacterial infection-induced skin injuries and aiding the healing and reconstruction of skin tissues.

A colorimetric sandwich assay based on magnetic separation of Brevinin-1BW-functionalized magnetic beads and porcine IgG for the detection of Staphylococcus aureus

He Dongxia,Liu Qi,Wang Lei,Han Qinqin,Zhang Jinyang,Li Chao,Song Yuzhu

A novel strategy integrating Brevinin-1BW (BW)-functionalized magnetic bead-based separation with a colorimetric immunoassay was developed to concentrate and enrich Staphylococcus aureus ( S. aureus ) from sample matrices using magnetic beads functionalized with antimicrobial peptide BW (MBs-BW) as carriers. To ensure the selectivity of the method, horseradish peroxidase (HRP)-labeled porcine IgG (HRP-porcine IgG) was used as a molecular recognition reagent and signal amplification probe. MBs-BW/ S. aureus /HRP-porcine IgG probe sandwich complex was successfully obtained. HRP chromogenic substrate 3,3′,5,5′-tetramethylbenzidine (TMB) was used to realize the chromogenic reaction. Finally, S. aureus was quantitatively analyzed based on the color signal generated. Under the optimal conditions, the assay showed a response in the concentration range of 1.0 × 10 2 –1.0 × 10 7  CFU/mL of S. aureus , and the detection limit was as low as 60 CFU/mL. This method does not require complicated sample pretreatment, and the operation process is fast and simple, which can directly complete the whole process of bacterial enrichment and detection. The recovery of this strategy in different types of spiked samples reached 86.8–97.8%, and the analytical results of this strategy were highly consistent with those of the conventional plate counting method. This strategy provides a new way for the rapid detection of S. aureus in complex samples. Graphical

Antimicrobial mechanism of alkyl gallates against Escherichia coli and Staphylococcus aureus and its combined effect with electrospun nanofibers on Chinese Taihu icefish preservation

Yu-gang Shi,Run-run Zhang,Chen-min Zhu,Ming-feng Xu,Qing Gu,Rammile Ettelaie,Shan Lin,Yi-fan Wang,Xin-yi Leng

The objective of this study was to investigate the antibacterial activity and potential mechanism of alkyl gallates against Escherichia coli and Staphylococcus aureus . Results show that the length of the alkyl chain plays a pivotal role in eliciting the activity and octyl gallate (OG) exerted excellent bactericidal activity through a multiple bactericidal mechanism. OG functions against both bacteria through damaging bacterial cell wall integrity, permeating into cells and then interacting with DNA, as well as disturbing the activity of the respiratory electron transport chain to induce a high-level toxic ROS (hydroxyl radicals) generation and up-regulation of the ROS genes. Also, electrospun nanofibers with OG have unique superiorities for maintaining the freshness of the icefish (4 °C). This research not only provides a more in-depth understanding of the interaction between OG and microorganisms but also highlights the great promise of using OG as a safe multi-functionalized food additive for food preservations.

A novel strategy for ultrasensitive detection and effective inactivation of Staphylococcus aureus based on Fe3O4-QCS-PEI-Cu-aptamer and ladder-branch HCR

Ren Dongxia,Wang Lin,Wei Hua,Mu Shijie,Yang Longfei

Bacterial infection is one of the leading causes of human death globally and is considered to be an urgent public health challenge. It is important to accurately and rapidly detect and kill pathogenic microorganisms. Therefore, a multifunctional platform for sensitive detection and efficient elimination of pathogenic bacteria should be developed to reduce the threat of bacteria. In this study, an integrated strategy based on Fe 3 O 4 -quaternary ammonium chitosan-polyetherimide-Cu-aptamer (Fe 3 O 4 -QCS-PEI-Cu-aptamer) and ladder-branch hybridization chain reaction (HCR) was proposed for the detection and inactivation of Staphylococcus aureus ( S. aureus ). The aptamer-complementary chain (aptamer-CP) conjugated to the surface of Fe 3 O 4 -QCS-PEI-Cu induces CP release in the presence of S. aureus , further initiating the ladder-branch HCR reaction and achieving signal amplification. Under optimal conditions, the developed sensor demonstrated a limit of detection (LOD) of 3 CFU/mL for S. aureus and exhibited high selectivity against several other pathogenic bacteria. Moreover, the Fe 3 O 4 -QCS-PEI-Cu-aptasensor showed potent antibacterial properties under near infrared (NIR) irradiation, achieving an antimicrobial efficiency of 99.946%. This strategy synergistically combined the high-sensitivity molecular detection with the antibacterial activity of novel nanomaterials activated by near infrared light, providing a new insight into the prevention of bacterial infections. Biocompatible formulation of a hydrophobic antimicrobial peptide L30 through nanotechnology principles and its potential role in mouse pneumonia model infected with Staphylococcus aureus

Ruihua Li,Huiping Gao,Ruiling Zhang,Beibei Zhang,Xueqin Wang,Xinhui Zhang,Ruifang Li

Hydrophobic antimicrobial peptide L30, a potential antibiotic candidate, has poor water solubility and hemolytic activity. Herein, a biocompatible nano-formulation composed of liposomes and dendritic mesoporous silica encapsulation (LDMSNs@L30) was constructed for L30 to solve the limits for its clinical development. The characterization, antimicrobial activity and therapeutic effect of LDMSNs@L30 on Staphylococcus aureus 9 ( cfr + ) infected mice models were investigated. LDMSNs@L30 displayed a smooth, spherical, and monodisperse nanoparticle with a hydrodynamic diameter of 177.40 nm, an encapsulation rate of 56.13%, a loading efficiency of 32.26%, a release rate of 66.5%, and effective slow-release of L30. Compared with free L30, the formulation could significantly increase the solubility of L30 in PBS with the maximum concentration from 8 μg/mL to 2.25 mg/mL and decrease the hemolytic activity of hydrophobic peptide L30 with the HC 5 from 65.36 μg/mL to more than 500 μg/mL. The nano delivery system LDMSNs@L30 also exhibited higher therapeutic effects on mice models infected with S. aureus 9 ( cfr + ) than those of free L30 after 7 days of treatment by reducing the lung inflammation and the inflammatory cytokines levels in plasma, showing better health score and pulmonary pathological improvement. Our research suggests that nano-formulation can be expected to be a promising strategy for peptide drugs in therapeutic applications.

Ultrasensitive microfluidic immunosensor with stir bar enrichment for point-of-care test of Staphylococcus aureus in foods triggered by DNAzyme-assisted click reaction

Jiaqi Wang,Lei Tan,Wenchao Bi,Haoyu Shen,Dengfeng Li,Zhenzhong Yu,Ning Gan

This work demonstrated an ultrasensitive and simple microfluidic immunosensor for point-of-care test of Staphylococcus aureus ( S. aureus ) based on the stir bar enrichment and DNAzyme-assisted click reaction. Initially, S. aureus was enriched by the 4-mercaptophenylboronic acid-functionalized stir bar. The yolk antibody (immunoglobulin Y) and copper-labeled polydopamine nanoparticles were then specifically conjugated with the captured target. The Cu(II) was released under acidic conditions and effectively catalyzed the copper-catalyzed azide-alkyne cycloaddition (CuAAC) between the alkyne group-labeled DNAzyme and the streptavidin–biotin-azido with the assistance of DNAzyme. Finally, the DNAzyme-streptavidin complexes were detected by microfluidic chips to quantify S. aureus . Under optimum conditions, this immunosensor showed good detection performances toward S. aureus within 10 to 2.5 × 10 4 CFU/mL with a limit of detection of 3 CFU/mL. Moreover, the satisfying detection results of real samples of animal origin also implied that this immunosensor owned great potential in practical applications.

Characterisation of a colourimetric biosensor SapYZUM13@Mn3O4-NH2 reveals the mechanisms underlying its rapid and sensitive detection of viable Staphylococcus aureus in food

Yeling Han,Wenyuan Zhou,Yuhong Wu,Aiping Deng,Lei Yuan,Yajun Gao,Huaxiang Li,Zhirong Wang,Bo Wang,Guoqiang Zhu,Zhenquan Yang

In this study, a colourimetric biosensor based on bacteriophage SapYZUM13 and an aminated Mn 3 O 4 (Mn 3 O 4 -NH 2 ) nanozyme was constructed and evaluated for its ability to detect Staphylococcus aureus in food. The biosensor had a detection time of 20 min, with a detection limit of 2 × 10 1  CFU/mL and recovery rate of 92.42–106.96%, indicating its high reliability and accuracy in detecting the food pathogen . Mechanistically, SapYZUM13@Mn 3 O 4 -NH 2 exhibited oxidase-mimicking capability, producing O 2 •– free radicals which oxidise 3,3′,5,5′-tetramethylbenzidine (TMB) to yield blue-coloured oxTMB. In the presence of S. aureus , the oxidase activity decreased remarkably owing to shielding of the nanozyme active sites. Moreover, SapYZUM13@Mn 3 O 4 -NH 2 could detect viable S. aureus from various sources, likely because of the special receptor-binding proteins of SapYZUM13 adsorbing to the wall teichoic acids on the S. aureus cell surface. Thus, SapYZUM13@Mn 3 O 4 -NH 2 has broad application prospects for the detection of viable S. aureus in various foods.

The Combined Strategy of Baicalin and Oxacillin Sodium Against Methicillin-Resistant Staphylococcus aureus: Biofilm Inhibition, Virulence Attenuation and In Vivo Anti-Infection Efficacy

Chao Ning,Jiale Zhou,Zhiyun Yu,Yuxuan Yang,Mengna Kang,Zhiyao Dong,Yantong Sun,Xin Meng,Haiyong Guo

Simple SummaryMethicillin-resistantStaphylococcus aureus(MRSA) infections present significant therapeutic challenges and constitute a major public health hazard. While the synergistic application of phytochemicals alongside conventional antibiotics offers a promising strategy to suppress bacterial growth with a reduced risk of triggering resistance, the precise mechanisms underlying this antibacterial effect and its in vivo efficacy remain poorly understood. Our study demonstrates that co-administration of baicalin and oxacillin sodium facilitates the intracellular buildup of reactive oxygen species (ROS). Furthermore, this combination reduces the levels of extracellular polymeric substances (EPSs) and total protein within biofilms, suppresses the metabolic functions of biofilm cells, and hinders the production of key virulence factors, specifically lipase and staphyloxanthin. These observed anti-biofilm and anti-virulence effects are likely attributable to the downregulation ofsarAgene expression induced by the baicalin-oxacillin sodium mixture. Building on these findings, we further assessed the in vivo anti-infective potential of this combined therapy. The results indicated that this combined therapy significantly decreased the infiltration of inflammatory cells within the peritoneal cavity of murine models, suppressed the production of pro-inflammatory mediators, and reduced bacterial burdens in tissues. Furthermore, it facilitated the restoration of pathological tissue injuries and exhibited protective properties for both hepatic and renal functions. In conclusion, the synergistic application of baicalin and sodium oxacillin effectively impedes MRSA biofilm development and virulence factor synthesis, thereby attenuating pathogenicity in a mouse model of peritonitis induced by MRSA USA300 strain. These findings validate the anti-infective efficacy of integrating plant-derived compounds with conventional antibiotics, offering novel experimental support for clinical strategies aimed at managing peritonitis caused by drug-resistant pathogens.Over the past few years, considerable scholarly interest has been directed toward the synergistic application of natural compounds alongside conventional antibiotics to address infections stemming from multidrug-resistant (MDR) pathogens. The primary objective of this research is to investigate the efficacy of baicalin (BA), an extract obtained fromScutellaria baicalensis, when used in conjunction with oxacillin sodium (OXS). Specifically, the study evaluates their combined impact on biofilm formation and toxicity reduction in methicillin-resistantStaphylococcus aureus(MRSA) strain USA300. Furthermore, a murine peritonitis model induced by MRSA USA300 was developed to determine the therapeutic potential of this combination therapy against infection. Experimental data indicate that the co-administration of BA and OXS does not induce hemolysis in vitro. In comparison to treatments involving either BA or OXS alone, the combined regimen significantly enhances the accumulation of intracellular reactive oxygen species (ROS) within MRSA USA300. Additionally, this synergistic approach suppresses the production of extracellular polymeric substances (EPSs), decreases the overall protein content within the biofilm matrix, and impairs the metabolic functions of biofilm cells. The investigation also revealed that the synergistic application of BA and OXS intensifies the suppression of key virulence determinants, specifically lipase activity and staphyloxanthin production, while simultaneously downregulating the transcription ofsarA(a global regulator of virulence). These findings substantiate the anti-virulence efficacy of the BA-OXS combination. In a murine model of peritonitis established using MRSA USA300, the healthy mice group, the MRSA USA300 group, the BA group, the OXS group, the combined group of BA and OXS, and the VAN group were set up, with eight mice in each group. The results showed that the combined therapy significantly mitigated body weight reduction, decreased the circulating counts of inflammatory cells, including leukocytes and lymphocytes, and suppressed the secretion of pro-inflammatory mediators such as TNF-α, IL-6, and IL-1β, thereby demonstrating potent anti-inflammatory properties. Furthermore, the co-administration of BA and OXS reduced bacterial burden in the abdominal organs of infected mice and alleviated associated histopathological injuries. Importantly, the treatment regimen exhibited no hepatorenal toxicity in the peritonitis mice, effectively maintaining normal levels. In the plasma of mice suffering from peritonitis, the concentration of malondialdehyde (MDA), a marker of oxidative stress, was reduced, while the activities of catalase (CAT) and superoxide dismutase (SOD) were elevated. This modulation contributes to anti-infective effects. These findings offer a theoretical foundation for subsequent investigations into the synergistic application of natural compounds and conventional antibiotics against MRSA.

Near-infrared light and pH-responsive hyaluronic acid-enveloped ZIF-8 nanoparticles for the treatment of pneumonia caused by methicillin-resistant Staphylococcus aureus

Yujie Xu,Xiaoli Zhang,Shuyi Liang,Congmin Niu,Jiayi Guo,Chunbo Lu

There is a serious threat to public health posed by the rapid spread of methicillin-resistant Staphylococcus aureus (MRSA). Photodynamic sterilization is the most effective method of combating bacterial infection, especially multidrug-resistant bacterial infection. In this study, zeolitic imidazolate framework-8 (ZIF-8) nanoparticles, one of the most attractive metal–organic framework material, were used to encapsulate the indocyanine green (ICG, a potent PS); after that, hyaluronic acid (HA) coating was applied to the nanoparticle to create a host cell-tracking nanoplatform (ICG@ZIF-8/HA). Upon near-infrared light irradiation, ICG@ZIF-8/HA exhibit excellent efficacy in producing reactive oxygen species (ROS) and killing multidrug-resistant bacteria in vitro . After the tail vein injection, ICG@ZIF-8/HA can precisely deliver to lung infection sites, and display an effective PDT-based elimination of multidrug-resistant bacteria in mouse lung infection models. These results suggest that the ICG@ZIF-8/HA nanoplatform may exhibit potent curative effects on infectious diseases.

Co-culture of Cronobacter sakazakii and Staphylococcus aureus: Explore the influence of mixed biofilm formation and regulation of Cronobacter sakazakii biofilm formation genes

Danliangmin Song,Ai Jia,Xuehe Qi,Kai Dong,Shiyu Liu,Chaoxin Man,Xinyan Yang,Yujun Jiang

Bacterial biofilm is a protective matrix composed of metabolites secreted by bacteria that envelop bacteria. By forming a biofilm, bacteria can considerably improve their environmental tolerance. In food-related processing environment, different types of microorganisms are often present in biofilms. The main contaminating strain in the powdered infant formula (PIF) processing environment, Cronobacter sakazakii and Staphylococcus aureus continues to pollute the PIF processing environment after biofilm production. This study selected Cronobacter sakazakii with a weak biofilm-forming ability as one of the test organisms. The coexistence of Cronobacter sakazakii and Staphylococcus aureus on the surface of production equipment was simulated to analyze the interaction. Biofilm formation in the co-culture group was significantly higher than the others. In-depth study of the effect of Staphylococcus aureus on the biofilm formation genes of Cronobacter sakazakii . Results show two bacteria can coexist on the surface of a metal device, forming a more compact hybrid biofilm structure. Under co-culture conditions, S. aureus increased bcsA and fliD expression in Cronobacter sakazakii , whereas decreased bcsC expression. Signaling molecules produced by Staphylococcus aureus (Autoinducer 2) significantly promoted the biofilm formation of Cronobacter sakazakii at the concentration of 0–500 ng/mL (0.099–0.177) and up-regulated the expression of bcsA , filD and flhD genes.

Exploring the antibacterial mechanism of essential oils by membrane permeability, apoptosis and biofilm formation combination with proteomics analysis against methicillin-resistant staphylococcus aureus

Cailin Tang,Jiali Chen,Lixia Zhang,Rongfei Zhang,Sichen Zhang,Shaoxia Ye,Zhimin Zhao,Depo Yang

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the important causes of food poisoning and infectious diseases worldwide, it can produce a large number of virulence factors, enhance the colonization ability of the host so that it can quickly colonize and spread on the surface of the objects. Essential oil (EO) is one of the natural products with antimicrobial properties, can be used as an important source of antibacterial agent discovery, and has a broad development prospect. However, the unclear mechanisms of antibacterial action have become an obstacle to its further development and use. Hence, the objective of the present study was to reveal the antibacterial mechanism of EO from Amomum villosum Lour ( A villosum Lour) against MRSA using label-free quantitative proteomics, investigate the effect of EO on the bacterial proteome, enzymatic activities and leakage of bacterial intracellular biomacromolecule. Proteomic analysis of MRSA in the presence of EO found that a total of 144 differential expressed proteins (DEPs) between the control and treatment group, in which 42 proteins were distinctly up-regulated and 102 proteins were down-regulated. Besides, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis, determination of cell membrane permeability and apoptosis, scanning electron microscopy (SEM) observations, bacterial surface hydrophobicity, and biofilm formation measurement were performed. Collectively, the above results indicated that the cell membrane damage by EO leads to the loss of membrane integrity and causes leakage of intracellular macromolecular substances, inhibition of protein, and biofilm synthesis. These findings manifested that EO exerts antibacterial effect by multiple avenues and expands our understanding of the antibacterial mechanism, it has potential application value in food preservative and pharmaceutical industries.

Ultrasensitive immunoassay of Staphylococcus aureus based on colorimetric and fluorescent responses of 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole to l-cysteine

Wenfang Deng,Chang Cheng,Hui Yang,Hui Wang,Yueming Tan,Qingji Xie,Ming Ma,Shouzhuo Yao

We report here a sensitive method for immunoassay of Staphylococcus aureus ( S. aureus ) based on colorimetric and fluorescent responses of 4-chloro-7-nitrobenzo-2-oxa-1,3-diazole (NBD-Cl) to l -cysteine (Cys). S. aureus cells were separated from samples by immunomagnetic nanoparticles and labeled with Cys-encapsulated liposome ( [email protected] ) immuno-nanocapsules. After mangnetic separation, the [email protected] immuno-nanocapsules linked to bacterial cells were destructed by Tween-20, and then the released Cys molecules reacted with NBD-Cl to generate colorimetric and fluorescent signals. Due to the amplification effect of liposome nanocapsules , the high capture efficiency of immunomagnetic nanoparticles, and the high responses of NBD-Cl to Cys, the detection limits for colorimetric and fluorescent detection of S. aureus were as low as 10 and 1 CFU mL −1 , respectively. The proposed method was successfully used to detect S. aureus in milk samples. This work has provided a novel NBD-Cl based colorimetric and fluorescent platform for immunoassay.
