Antibiotic-loaded chitosan-gelatin scaffolds for infected seawater immersion wound healing

Qi Fang,Zexin Yao,Longbao Feng,Tian Liu,Shikun Wei,Pengcheng Xu,Rui Guo,Biao Cheng,Xinlu Wang

Skin damaged during sea battles is vulnerable to seawater immersion and bacterial infection. Scaffolds with effective biological function are highly desired for treatment of naval combat wound injuries. Herein, we prepared composite scaffolds of CS/GEL/GMs-CIP. The chitosan (CS) and gelatin (GEL) were cross-linked by genipin as matrix, and then gelatin microspheres loading ciprofloxacin hydrochloride (GMs-CIP) were add. From in vitro characterization results, CS/GEL/GMs-CIP had high water absorption ability, proper porosity, satisfactory fracture resistance, and flexibility. Furthermore, CS/GEL/GMs-CIP composite scaffold had excellent biocompatibility. Antibacterial experiments confirmed that CS/GEL/GMs-CIP had a significant inhibitory effect on E. coli , S. aureus and P. aeruginosa . The in vivo wound healing was evaluated using animal wound infection model of seawater immersion, and it was observed that the prepared composite scaffolds accelerated wound healing, reepithelialization, collagen deposition. Further analysis of wound tissue indicated that the expression of anti-inflammatory factor (TGF-β1) was up-regulated, but the serum endotoxin levels and expression of pro-inflammatory factor (TNF-a, IL-6, and IL-1β) were down-regulated. In summary, we believe that CS/GEL/GMs-CIP composite scaffold may serve as a promising multifunctional dressing for healing with open trauma wound infections and wound with seawater immersion.

Fabrication of antiseptic, conductive and robust polyvinyl alcohol/chitosan composite hydrogels

Luo ChunHui,Zhao Yufei,Sun Xinxin,Luo FaLiang

Fabricating robust and multi-functional hydrogels is of great importance and challenge. In this work, chitosan (CS) and polyvinyl alcohol (PVA) were used to design antiseptic, conductive and robust hydrogels by a two-step method. Chemical structures of gels, the degree of crystallinity, the state of water in hydrogels, as well as their microstructures were characterized via a combination of FT-IR, XRD, DSC and SEM. Segment lengths of cross-linking points were calculated from elastic rubber theory. Their mechanical properties were evaluated on the electronic testing machine. It was shown that the tensile strength and elongation at break of single PVA hydrogel were only 200 kPa and 135%, respectively, due to the heterogenous structure with pore sizes between 1.5 ~ 8.2 μm. By introducing CS into PVA matrix followed with soaking in a saturated NaCl solution, the network became homogeneous with a pore size of 0.5 ~ 1.1 μm. Moreover, free water changed to bond water, and frictions between polymer chains increased because of hydrophobic associations and entanglements of CS segments. As a result, the tensile stress and strain increased to 3800 kPa and 270%, respectively. The gel also exhibited antiseptic property, electrical conductivity and swelling-resistant properties. The strength after reaching swell equilibrium was 3400 kPa, much higher than most gels at swollen states. This gel might find applications in bionic cartilage, sensors, food preservation and wearable devices.

Rongli Zhang,Hui Hao,Cuige Zhang,Renchun Yang,Mingyue Sun,Ching-Ping Wong,Ying Xu

A kind of bioadhesive hydrocaffeic acid modified chitosan colloidal particles (HCA-CS/TPP CPs) containing synthetic catecholamine groups was prepared via amidation reaction and ionic gelation. The formation process and property of the HCA-CS/TPP CPs were characterized by ultraviolet visible spectrophotometry and Zeta PALS instruments. The prepared HCA-CS/TPP CPs were then used as particulate emulsifier to stabilize Pickering emulsions. The stability of the Pickering emulsion droplet was characterized by digital camera and optical microscope. The results revealed that HCA-CS/TPP CPs exhibited good emulsifying property at a wide pH value range, which will endow the potential application of the Pickering emulsions stabilized by HCA-CS/TPP CPs in various fields.

Preparation and physicochemical properties of antioxidant chitosan ascorbate/methylcellulose composite films

Wenqiang Tan,Jingjing Zhang,Xiang Zhao,Qing Li,Fang Dong,Zhanyong Guo

Polysaccharide-based biodegradable films have been considered as the promising candidates for food packaging industry instead of petroleum-based packaging materials. Here, we reported a class of edible composite films based on chitosan ascorbate and methylcellulose prepared by mixing different ratios (1,0, 4:1, 2:1, 1:1, 1:2, 1:4, and 0:1) of the biopolymers using the casting technique. Their physicochemical properties as well as the DPPH radical scavenging ability and reducing power were investigated. All physicochemical properties and antioxidant activities were significantly affected by the chitosan ascorbate/methylcellulose ratio in the matrix. The increases in tensile strength and elongation at break values, maximum decomposition temperatures, whitish index, compactness, moisture content, and a reduction in water vapor permeability were observed as the proportion of methylcellulose increased in the matrix. But the composite films containing a greater proportion of chitosan ascorbate exhibited the better barrier properties against UV–vis light and the stronger DPPH radical scavenging effect and reducing power. The chitosan ascorbate/methylcellulose composite films with interesting physicochemical properties and strong antioxidant action showed the potential value as biodegradable and edible biomaterials for food packaging.

Preparation and characterization of arginine-modified chitosan/hydroxypropyl methylcellose antibacterial film

Jinxing Song,Huafeng Feng,Mengqi Wu,Linxiao Chen,Wei Xia,Wenqing Zhang

Chitosan and its derivatives are widely used in medical, cosmetic and food fields. In this study, chitosan-N-arginine (CSA) was synthesized and characterized by Fourier-transform infrared (FT-IR), 1 H NMR, gel permeation chromatography (GPC), thermogravimetric analysis (TGA) and scanning electron microscopy (SEM). A novel antibacterial composite film consisting of CSA, hydroxypropyl methylcellose (HPMC) and glycerol was then prepared. The transparent and homogeneous film presented good compatibility between CSA and HPMC, confirmed by SEM. The thickness of the film was about 44.8 μm and its moisture content was 23.0%. Antimicrobial evaluation of CSA/HPMC film showed 9.0 mm bacteriostatic diameter zone against E. coli and 10.5 mm one against S. aureus . The film exhibited cell biocompatibility and promoted proliferation with L929 cell cytotoxicity test . Both antibacterial and cytotoxic results showed that the CSA/HPMC film was a promising material for medication, cosmetics and food preservation applications.

Janus Self-Propelled Chitosan-Based Hydrogel Spheres for Rapid Bleeding Control

Qiao Yu,Baihai Su,Weifeng Zhao,Changsheng Zhao

Uncontrolled hemorrhage is a major cause of potentially preventable death in civilian trauma nowadays. Considerable concern has been given to the development of efficient hemostats with high blood absorption, self-propelled property, and Ca 2+ release ability, for irregularly shaped and noncompressible hemorrhage. Herein, Janus self-propelled chitosan-based hydrogel with CaCO 3 (J-CMH@CaCO 3 ) is developed by partial ionic crosslinking of carboxylated chitosan (CCS) and Ca 2+ , gravity settlement, and photopolymerization, followed by removing the shell of CCS. The obtained J-CMH@CaCO 3 is further used as a hemostat powered by the internal CaCO 3 and coordinated protonated tranexamic acid (J-CMH@CaCO 3 /T). Bubbles are generated and detached to provide the driving force, accompanied by the release of Ca 2+ . The two aspects work in synergy to accelerate clot formation, endowing the J-CMH@CaCO 3 /T with excellent hemostatic efficiency. The J-CMH@CaCO 3 /T presents high blood absorption, favorable blood-clotting ability, desired erythrocyte and platelet aggregation, and acceptable hemocompatibility and cytocompatibility. In rodent and rabbit bleeding models, the J-CMH@CaCO 3 /T exhibits the most effective hemostasis to the best knowledge of the authors, wherein the hemorrhage is rapidly halted within 39 s. It is believed that the J-CMH@CaCO 3 /T with self-propelled property opens up a new avenue to design high-performance hemostats for clinical application.

Loading and releasing behavior of carboxymethyl cellulose and chitosan complex beads

Ali A. Altam,Liping Zhu,Dafaalla Babiker,Hajo Yagoub,Shuguang Yang

The complex beads of carboxymethyl cellulose (CMC) and chitosan (CHI) that are fabricated by interfacial complexation have a core-shell structure composed of a thin skin layer and a 3D entangled internal structure, which endows the beads with adsorption possibilities toward anionic and cationic molecules via electrostatic interaction. However, the loading and releasing behavior of such a system is essential and fundamental for their application yet has not been studied systematically. In this work, the influences of beads physical state, adsorbent dosage, agitation speed, solution pH and temperature on the loading behavior were investigated. The CMC@CHI beads demonstrated different loading efficiency to three typical dyes of methylene blue (MB), acid blue-113 (AB), and methyl orange (MO) with different charge status. The adsorption rate followed an order of MB ˃ AB ˃ MO, giving 97.4%, 73.3% and 20.2%, respectively. The releasing of three dyes from CMC@CHI beads also exhibited different rates, and MB release was found to be prolonged compared to AB and MO. The loading mechanism of dyes was further investigated via X-ray photoelectron spectroscopy. These CMC@CHI beads may have possible applications in mixed dye removal as well as encapsulating systems of drugs or sensitive chemicals with different charges.

Gadolinium-doped mesoporous calcium silicate/chitosan scaffolds enhanced bone regeneration ability

Fang Liao,Xiao-Yuan Peng,Fan Yang,Qin-Fei Ke,Zhen-Hong Zhu,Ya-Ping Guo

Chitosan (CTS) and mesoporous calcium silicate (MCS) have been developed for bone defect healing; however, their bone regeneration capacity still does not satisfy the patients with bone diseases. Gadolinium (Gd) is accumulated in human bones, and plays a beneficial role in regulating cell performance and bone regeneration. We firstly constructed Gd-doped MCS/CTS (Gd-MCS/CTS) scaffolds by a lyophilization technology. The interconnected arrangement of CTS films lead to forming macropores by using ice crystals as templates during the lyophilization procedure, and the Gd-MCS nanoparticles dispersed uniformly on the macropore walls. The biocompatible chemical components and hierarchical pores facilitated the attachment and spreading of rat bone marrow-derived mesenchymal stem cells (rBMSCs). Interestingly, the Gd dopants in the scaffolds effectively activated the Wnt/β-catenin signaling pathway, resulting in excellent cell proliferation and osteogenic differentiation capacities. The osteogenic-related genes such as alkaline phosphatase (ALP), runt-related transcription factor 2 (Runx2) and collagen type1 (COL-1) were remarkably up-regulated by Gd-MCS scaffolds as compared with MCS scaffolds, and their expression levels increased in a positive correlation with Gd doping amounts. Moreover, in vivo rat cranial defect tests further confirmed that Gd-MCS/CTS scaffolds significantly stimulated collagen deposition and new bone formation. The exciting finding suggested the beneficial effects of Gd 3+ ions on osteogenic differentiation and new bone regeneration, and Gd-MCS/CTS scaffolds can be employed as a novel platform for bone defect healing.

Dual ionically crosslinked chitosan–based injectable hydrogel as drug delivery system

Zhao Jing,Cui Lu,Wang Xiaoran,Deng Chunyu

Thermosensitive hydrogels based on chitosan play an important role in the field of tissue engineering and drug delivery system, with excellent properties such as biocompatible, biodegradable, and minimally invasive. In this work, a dual ionically crosslinked injectable hydrogel was composed of polyelectrolyte complex (PEC) and sodium hydrogen carbonate (SHC) as non-toxic gel agent. The PEC was prepared by ionic interaction between positively charged chitosan and negatively charged thermosensitive polymer poly(NIPAm-co-AMPS). The injectable hydrogel with rapid gelation was obtained by optimizing feed ratio, SHC amount, concentration, and temperature. The swelling, temperature sensitive, rheological, and cytotoxic properties of the hydrogels were evaluated. The potential of hydrogels applied in drug delivery system was studied by measuring the release property in vitro, taking aspirin as drug model. The results showed that the hydrogel exhibited a sol–gel transition at about 37 ℃, when the volume ratio of CS (20 mg/mL), PNAM (4.0 mg/mL), and SHC (0.4 M) was 1:0.2:0.5; the hydrogel with good mechanical strength and gelation time of 1–3 min in the physiological conditions was obtained. The hydrogels prepared with high SHC concentration or more gel agents showed shorter gelation time and better thermostability. The hydrogel presented a 1-day sustained release of aspirin with no significant burst release in vitro. The release properties demonstrated that the hydrogels released aspirin in a sustained manner, about 87% in 24 h. L929 cells had good survival and proliferative rates within CS-PNAM/SHC hydrogel after 24 h, showing excellent biocompatibility. Therefore, the injectable thermos-responsive hydrogels formed by CS-PNAM/SHC and loaded with aspirin turned into a smart drug delivery system, showing great potential for controlled release.

Amyloid-Like Lysozyme@Chitosan Composite for the Separation of Organic Pollutants

Qingmin Yang,Jintao Ma,Xueqian Wu,Siyu Zhao,Chenyue Dong,Zhuoyue Wu,Yuhan Lin,Chunyan Guo,Xi Li,Chao Sun,Jiaojiao Cao,Linlin Zhu,Renyuan Song,Facui Yang

There is an urgent demand for efficient adsorbents to remove persistent organic pollutants from wastewater. Unlike previous protein‑based composites used mainly for metal cation separation, this study develops a positively charged amyloid‑like lysozyme@chitosan composite (PTL@CS) by incorporating chitosan into lysozyme, specifically targeting anionic dyes such as Congo red (CR). PTL@CS exhibits a stable structure enriched with synergistic functional groups (e.g., carboxyl, hydroxyl, amino) and demonstrates amyloid-like aggregation, confirmed by thioflavin T (ThT)/8-Anilino-1-naphthalenesulfonic acid (ANS) fluorescence assays, Fourier transform infrared (FTIR), and laser scanning confocal microscopy (LSCM). The material achieves exceptional CR adsorption capacity (up to 801 mg·g − 1 ), fitting the pseudo-second-order kinetics and the Langmuir isotherm models, indicating chemisorption-dominated monolayer adsorption. PTL@CS maintains high adsorption efficiency under acidic to neutral pH conditions as well as in complex ionic environments, exhibits excellent reusability, and shows low cytotoxicity toward IMR‑90 and human umbilical vein endothelial cells (HUVEC). These attributes underscore its potential as a sustainable, selective, and biocompatible adsorbent for next-generation water purification. Graphical Amyloid-like lysozyme@chitosan composite (PTL@CS) efficiently removes organic pollutants from water without causing environmental pollution, offering high adsorption capacity and reusability.

Anti-pilling of Cashmere Knitted Fabric Based on Siloxane-Modified Chitosan

Tian Wanyun,Chen Luyi,Zhang Xue,Wu Yarong

In this study, chitosan was modified by two kinds of different siloxanes, γ-glycidoxypropyl-trimethoxysilane (GPTMS) and γ-glycidoxypropyl-dimethoxy- methylsilane (GPDMS), respectively, then applied for anti-pilling of cashmere knitted fabrics. The structure of the modified chitosan, the apparent morphology and the mechanical properties of the modified film were analyzed by FTIR, SEM and DMA, respectively. The hydrophilicity and swelling properties of the modified film were compared. The results showed that epoxy groups of siloxane reacted with amino groups of chitosan, and the –OR (R represents alkyl) groups of siloxane hydrolyzed and polycondensation conducted during the film-forming process. After modification, the mechanical properties of the film were more flexible, the hydrophilicity and swelling of the film was decreased. Cashmere knitted fabric was treated with the modified chitosan and glutathione was chosen as the model compound to explore the reaction between modified chitosan and cashmere fibers. The apparent morphology and elements distribution of the fibers were observed by SEM and EDS, respectively. The directional friction effect of the fibers was tested by fiber friction coefficient tester. The pilling grade, handle, whiteness and launderability of fabric were investigated. The results showed that the epoxy groups remaining in siloxane were reacted with the amino groups of cashmere fiber, which enhance the binding of the fiber and siloxane-modified chitosan, the siloxane-modified chitosan film formed on fibers in situ. The directional friction effect of the fiber significantly reduced, the tendency of fuzzes to the surface of the fabric was weakened and the probability of tangling of exposed fuzzings was reduced, the pilling grade of the fabric treated by modified chitosan reached level 4 and the fabrics had good launderability, the soft and smooth handle was still preserved. There was no obvious color difference of cashmere fabrics treated with siloxane-modified chitosan.

Evaluation of Chitosan-Oleuropein Nanoparticles on the Durability of Dentin Bonding

Shuya Zhao,Yunyang Zhang,Yun Chen,Xianghui Xing,Yu Wang,Guofeng Wu

Purpose To evaluate the effects of dentin pretreatment with chitosan-loaded oleuropein nanoparticles (CONPs) on the durability of resin–dentin bonding interfaces. Methods Eighty freshly extracted non-carious human third molars were randomly divided into four groups (n = 20 each): a de-ionized water (DW) group, a chitosan (CS) group, a chlorhexidine (CHX) group and a CONP group. The dentin in the DW, CS, CHX, and CONP groups were pretreated with de-ionized water, 1.0 mg/L CS solution, 2% chlorhexidine solution, and CONP suspension (prepared with 100 mg/L oleuropein), respectively, followed by the universal adhesive and resin composites. The bonded teeth of each group were randomly divided into two subgroups: an immediate subgroup and an aged subgroup. The bonded teeth of each group were then cut into the bonded beams. We measured their microtensile bond strength (μTBS), observed the characteristics of bonding interface by atomic force microscope, calculated the percentage of silver particles in a selected area for interfacial nanoleakage analysis, and evaluated the endogenous gelatinase activity within the bonding interface for in-situ zymogram analysis. Data were analyzed with two-way ANOVA and LSD multiple comparison test (P < 0.05).Results Regardless of after 24 h or after thermocycling, CONP exhibited better μTBS (P < 0.05) than the other three groups except that there was not a statistical significance (P > 0.05) in the CONP and CHX groups after 24 h. Besides, the CONP group presented significantly higher modulus of elasticity in the hybrid layers (P < 0.05), lower expression of nanoleakage (P < 0.05), and better inhibitory effect of matrix metalloproteinases than the other three groups before and after thermocycling. Conclusion Altogether, the CONPs had the potential to act as a dentin primer, which could effectively improve the dentin-resin binding durability.
