Inhibition of d-ribose-induced glycation of metalloprotein human myoglobin by bioactive quinones

Jing-Jing Liu,Si-Dian Miao,Zhan-Yi Wang,Ya-Han Zhu,Bin-Bin Jiang,Shuang-Shuang Long,Shu-Qin Gao,Ying-Wu Lin

The anti-glycation effects and mechanisms of eight quinone-related compounds, primarily sourced from medicinal and edible plants, were evaluated in this study. In vitro glycation experiments were conducted to study the structural perturbation induced by d -ribose on human myoglobin (HMb) in the presence of the tested compounds. Spectroscopic and electrophoretic analyses indicated that among the four benzoquinone-related substances, thymoquinone and its structurally closest analogue 2,5-dimethyl-1,4-benzoquinone, exhibited significant inhibitory effects on the d -ribose-induced glycation, thereby maintaining protein structural stability, reducing advanced glycation end products (AGEs) formation, and attenuating the covalent protein modification by d -ribose. In contrast, 1,4-benzoquinone and hydroquinone (a reduction product of 1,4-benzoquinone) promoted HMb denaturation through the Michael addition mechanism. Of the two naphthoquinones, juglone and plumbagin, showed anti-glycation effects, whereas the anthraquinones emodin and chrysophanol showed no significant effects. Nano-HPLC-MS/MS analysis further showed that thymoquinone was more effective in preventing glycation than juglone and plumbagin. This study investigates the mechanisms underlying glycation inhibition by quinones, revealing that the presence of dual β -substituted carbons on the benzoquinone ring is critical for stabilizing protein conformations and suppressing glycation. It provides a preliminary mechanistic basis and identifies candidate molecules for potential applications of quinones in retarding glycation, such as in managing diabetic complications or inhibiting glycation reactions during food processing.

A methyl-β-D-galactopyranoside breath probe for early detection of hepatocellular carcinoma

Fuping Wang,Xinyi Peng,Zhengpeng Zhong,Mingchen He,Qinyan Lu,Yifan Li,Huaiyang Liu,Wang Li,Guobao Chen,Bo Zhou,Zhongmin Chen

A non-isotopic methanol-releasing probe detects HCC early via breath. • Selectively cleaved by β-galactosidase in HCC for tumor-specific signal. • A low-cost dual-sensor array enables real-time exhaled methanol detection. • Distinguishes early-stage tumor-bearing mice from controls (< 2 mm). • Offers a portable, affordable screening tool for primary healthcare. Early detection is crucial for improving therapeutic outcomes in hepatocellular carcinoma (HCC), yet the widespread implementation of low-cost, accessible screening for high-risk populations remains a significant challenge. To address this issue, we investigated the β-galactosidase–methanol signaling axis using a hydrophilic, cell-membrane-impermeable probe, methyl-β-D-galactopyranoside (MGP). This probe is selectively hydrolyzed by β-galactosidase, which is overexpressed in the HCC microenvironment, releasing methanol—an easily volatile, non-isotopic, and metabolically inert volatile organic compounds (VOCs). Methanol levels in exhaled breath were quantified using a custom-designed sensor array. Through in vitro enzymatic assays, cellular studies, and animal experiments, we demonstrated that methanol concentrations in tumor-bearing mice (with tumor diameters <2 mm) were significantly elevated compared to healthy controls, and these levels positively correlated with tumor volume. Our findings highlight the potential of Me-β-Gal in addressing critical barriers to early HCC detection. With further validation, this approach could potentially improve clinical management for high-risk populations, including patients with liver cirrhosis.

Thermostable D-Allulose 3-Epimerase for Long-Term Food-Compatible Continuous Production Systems

Jiawei Cui,Yan Li,Ming Yan

D-allulose is a rare sugar with promising applications in food and health industries, owing to its low caloric value and multiple health benefits. In this study, we systematically investigated a thermostable D-allulose 3-epimerase (TcDAEase) fromThermogemmatispora carboxidivoransfor food-compatible continuous production. The enzyme exhibited remarkable thermostability, with over 70% activity retained at 80 °C, and showed broad pH tolerance across the range of 8.0 to 13.0. Notably, TcDAEase exhibited high catalytic activity toward D-allulose and D-fructose even without the addition of metal ions. Moreover, food-grade Mg2+was identified as enhancing enzyme activity by 14.3%, thus ensuring compliance with Generally Recognized as Safe (GRAS) standards for food applications. To improve industrial applicability, the enzyme was immobilized using a chitosan-diatomaceous earth (DE) matrix via three-step adsorption–crosslinking–embedding strategy. The immobilized TcDAEase achieved 48.7% ± 2.4% activity recovery and retained 90.3% ± 1.5% activity over seven reaction cycles. Furthermore, continuous production of D-allulose was realized in a packed-bed reactor, operating stably at 60 °C, pH 8.0 and 5 mM Mg2+for 150 days, producing 756 kg of D-allulose with a conversion yield exceeding 89.7% of the theoretical maximum. Overall, this study provides a feasible strategy for the continuous and efficient production of high-value-added D-allulose in the food industry.

Gasdermin D permeabilization of mitochondrial inner and outer membranes accelerates and enhances pyroptosis

Rui Miao,Cong Jiang,Winston Y. Chang,Haiwei Zhang,Jinsu An,Felicia Ho,Pengcheng Chen,Han Zhang,Caroline Junqueira,Dulguun Amgalan,Felix G. Liang,Junbing Zhang,Charles L. Evavold,Iva Hafner-Bratkovič,Zhibin Zhang,Pietro Fontana,Shiyu Xia,Markus Waldeck-Weiermair,Youdong Pan,Thomas Michel,Liron Bar-Peled,Hao Wu,Jonathan C. Kagan,Richard N. Kitsis,Peng Zhang,Xing Liu,Judy Lieberman

Gasdermin D (GSDMD)-activated inflammatory cell death (pyroptosis) causes mitochondrial damage, but its underlying mechanism and functional consequences are largely unknown. Here, we show that the N-terminal pore-forming GSDMD fragment (GSDMD-NT) rapidly damaged both inner and outer mitochondrial membranes (OMMs) leading to reduced mitochondrial numbers, mitophagy, ROS, loss of transmembrane potential, attenuated oxidative phosphorylation (OXPHOS), and release of mitochondrial proteins and DNA from the matrix and intermembrane space. Mitochondrial damage occurred as soon as GSDMD was cleaved prior to plasma membrane damage. Mitochondrial damage was independent of the B-cell lymphoma 2 family and depended on GSDMD-NT binding to cardiolipin. Canonical and noncanonical inflammasome activation of mitochondrial damage, pyroptosis, and inflammatory cytokine release were suppressed by genetic ablation of cardiolipin synthase (Crls1) or the scramblase (Plscr3) that transfers cardiolipin to the OMM. Phospholipid scramblase-3 (PLSCR3) deficiency in a tumor compromised pyroptosis-triggered anti-tumor immunity. Thus, mitochondrial damage plays a critical role in pyroptosis.

Dietary D-xylose promotes intestinal health by inducing phage production in Escherichia coli

Hu Jie,Wu Yifan,Kang Luyuan,Liu Yisi,Ye Hao,Wang Ran,Zhao Jinbiao,Zhang Guolong,Li Xilong,Wang Junjun,Han Dandan

Elimination of specific enteropathogenic microorganisms is critical to gut health. However, the complexity of the gut community makes it challenging to target specific bacterial organisms. Accumulating evidence suggests that various foods can change the abundance of intestinal bacteria by modulating prophage induction. By using pathogenic Escherichia coli ( E. coli ) ATCC 25922 as a model in this research, we explored the potential of dietary modulation of prophage induction and subsequent bacterial survival. Among a panel of sugars tested in vitro, D -xylose was shown to efficiently induce prophages in E. coli ATCC 25922, which depends, in part, upon the production of D -lactic acid. In an enteric mouse model, prophage induction was found to be further enhanced in response to propionic acid. Dietary D -xylose increased the proportion of Clostridia which converted D -lactic acid to propionic acid. Intestinal propionic acid levels were diminished, following either oral gavage with the dehydrogenase gene ( ldhA )-deficient E. coli ATCC 25922 or depletion of intestinal Clostridia by administration of streptomycin. D -Xylose metabolism and exposure to propionic acid triggered E. coli ATCC 25922 SOS response that promoted prophage induction. E. coli ATCC 25922 mutant of RecA, a key component of SOS system, exhibited decreased phage production. These findings suggest the potential of using dietary components that can induce prophages as antimicrobial alternatives for disease control and prevention by targeted elimination of harmful gut bacteria.

Protective effect of thymoquinone on glycation of human myoglobin induced by d-ribose

Jing-Jing Liu,Zhan-Yi Wang,Bin-Bin Jiang,Shu-Qin Gao,Ying-Wu Lin

Nonenzymatic glycation and the subsequent accumulation of advanced glycation end-products (AGEs) in proteins are factors underlying long-term pathogenesis in diabetes. The study of protein glycation is crucial for elucidating their relationship with diabetes mellitus and related disorders. This study explores the interaction between d -ribose and human myoglobin (HMb), as well as the protective effect of thymoquinone (TQ) on glycation. A time-dependent in-vitro glycation study was performed to investigate the mechanism of d -ribose-induced structural interference of HMb in the absence and presence of TQ. Spectroscopic and proteomic analysis indicated that the presence of TQ significantly reduced the total amount of AGEs while maintaining structural characteristics of HMb. 14 glycated sites on HMb were further identified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) after incubation with d -ribose for 12 h, predominantly interacting with lysine residues. TQ was found to disrupt this interaction, reducing the glycated sites from 14 to 12 sites and the percentage of glycated peptides from 26.50 % to 12.97 %. Additionally, there was a significant decrease in the degree of glycation at the same sites. In summary, our findings suggest that TQ has the potential to act as an anti-glycation agent and provide a comprehensive understanding underlying the inhibition mechanism of glycation.

Saikosaponin-d regulates angiogenesis in idiopathic pulmonary fibrosis through angiopoietin/Tie-2 pathway

Yan Wu,Jun Zhang,Xintian Wang,Yuncong Xu,Jinxu Zheng

Objective Idiopathic pulmonary fibrosis (IPF) is considered as a chronic interstitial lung disease with underlying mechanism of IPF remaining unclear, while there are no definitive treatment options. In recent years, scientists have gradually paid attention to the influence of angiogenesis on IPF. Because IPF is a progressive with microvascular remodeling disorder, scientists have postulated that angiogenesis may also be one of the initiating and contributing factors of the disease. Bupleurum is a common natural Chinese herbal medicine with antibacterial, anti-inflammatory, anti-tumor and other pharmacological effects. As the most important active monomer of Bupleurum, Saikosaponin-d (SSd) is a new discovery with anti-pulmonary fibrosis (PF) activity. This study attempts to investigate the role of SSd in the interference of PF through regulation of angiogenesis in IPF through Angiopoietin (Angpt) /Tie receptor 2 (Tie2) pathway. Methods Randomly, we allocated C57BL/6 mice into four groups (n = 20 in each group). Afterwards, establishment of IPF model was accomplished via intratracheal administration of bleomycin (BLM, 5 mg/kg), while corresponding drug intervention was given accordingly. On 3rd, 7th, 14th and 28th days after modeling, we performed histopathological examination through staining. Meanwhile, immunohistochemistry (IHC) of PF and the expression of related factors were observed, while Ang/Tie2 pathway was assessed by ELISA with the effect of SSd on angiogenesis related proteins in IPF being explored with IHC and Western Blot technique. Results Our results showed that SSd could reduce inflammation and PF levels in lung tissue of experimental mice, while levels of angiogenesis-related factors, namely Tie-2, Ang-1 and ANGPT2 (Ang-2), fibrosis- associated factors like Alpha-smooth muscle actin (α-SMA), collagen-I and hydroxyproline in SSd and dexamethasone (DXM) mice were significantly reduced at each time point compared to BLM (p < 0.01). Additionally, we discovered substantial decreased expressions of Ang-1, Ang-2, Tie-2, α-SMA and collagen-I at protein level in SSd and DXM mice at each time point compared to BLM (p < 0.05). Besides, insignificant differences were observed between SSd and DXM groups (p > 0.05). Conclusion This study has demonstrated that SSd could down-regulate the expression of ANG-1, Ang-2 and Tie2 in the Ang/Tie2 pathway, and may reduce lung inflammation and PF in BLM-induced mice via inhibition of angiogenesis.

O/D emulsions stabilized by quaternary ammonium gemini surfactants together with alumina nanoparticles

Peng Wei,Faming Wu,Qinghua Zhang,Zhirong Liu

Oil-in-dispersion (O/D) emulsions are the focus in many fields due to their new microstructures and new functions. Herein, O/D emulsions were successfully prepared using series of cationic gemini surfactants 12-s-12 (s = 2, 3, and 6) and alumina nanoparticles. The microstructures and type of emulsions were identified using optical microscopy, zeta potential and contact angle methods. Gemini surfactants with two head groups carry more charges than that of conventional surfactants when adsorbed at the oil–water interface. The O/D emulsions can be stabilized at lower surfactant concentrations compared with the conventional single-headed surfactants. In the presence of 0.1 wt% alumina nanoparticles, gemini surfactants 12-s-12 (s = 2, 3, and 6) can stabilize emulsions at the concentration of 3 × 10 −4 mM (1.84 × 10 −5 wt%), 6 × 10 −4 mM (3.76 × 10 −5 wt%) and 1 × 10 −3 mM (6.68 × 10 −5 wt%), respectively. Addition of excessive organic salts such as sodium salicylate and sodium p-methylbenzenesulfonate shielded the head group charges of gemini surfactants, leading to gemini surfactants to behave as nonionic surfactants Adsorption of surfactants at the alumina nanoparticles occurred, resulting in the transformation from the O/D emulsions to Pickering emulsions. This work shows the advantages of preparing O/D emulsions using gemini surfactants and also provides a new methods of emulsion type transformations by adding excessive oppositely charged organic salts.

Electrochemical Chiral Recognizing Tryptophan Enantiomers Based on Chiral Metal-Organic Framework D-MOF

Hou Huipeng,Tang Shanshan,Wang Wei,Liu Miao,Liang Axin,Xie Bingteng,Yi Yue,Luo Aiqin

I n this work, an electrochemical chiral sensor of a nanowire-like chiral metal-organic framework/multiwalled carbon nanotube-chitosan/glassy carbon electrode( D -MOF/MWCNTs-CS/GCE) was proposed for the enantiorecognition of L / D -tryptophan( L / D -Trp). The asymmetrical spatial structure of D -MOF provides the feasibility for the enantiorecognition of Trp enantiomers. Moreover, differential pulse voltammetry(DPV) was carried out to be the detection method and the DPV peak potential difference(Δ E p ) between L -Trp and D -Trp was referred as the index of the enantiorecognition performance. Several parameters, such as mass ratios and drop-coated volume of MWCNTs-CS, drop-coated volume and concentration of D -MOF, pH and detection temperature of D -MOF/MWCNTs-CS/GCE were optimized for the largest Δ E p value. And the molecular dynamics(MD) simulation was used to elucidate the enantiorecognition mechanism. Furthermore, the proportions of D -Trp( D -Trp%) in Trp mixtures were detected in a good linear relationship with the DPV peak potentials( E p ), and the proposed electrochemical chiral sensor exhibited good reproducibility, stability and enantiorecognition ability. Additionally, the proposed electrochemical chiral sensor( D -MOF/MWCNTs-CS/GCE) has a good application prospect in the fields of biomedicine, clinical diagnosis, chemical production, pharmaceuticals safety and so on.

Chemoenzymatic Approach to (S)-1,2,3,4-Tetrahydroisoquinoline Carboxylic Acids Employing D-Amino Acid Oxidase

Shuyun Ju,Mingxin Qian,Gang Xu,Lirong Yang,Jianping Wu

Optically pure 1,2,3,4-tetrahydroisoquinoline carboxylic acids constitute an important class of building blocks for the synthesis of natural products and synthetic pharmaceuticals. However, redox deracemization of racemic 1,2,3,4-tetrahydroisoquinoline carboxylic acids as an attractive method is still challenging for the lack of suitable oxidoreductases. Herein, a D-amino acid oxidase from Fusarium solani M-0718 ( Fs DAAO) with broad substrate scope and excellent enantioselectivity was exploited through genome mining, and applied for the kinetic resolution of a number of racemic 1- and 3-carboxyl substituted tetrahydroisoquinolines to yield the corresponding ( S )-enantiomers with excellent enantiomeric excess ( ee ) values (up to >99%). By using Fs DAAO in combination with ammonia-borane in one pot, deracemization of these racemic carboxyl-substituted tetrahydroisoquinolines was achieved with conversions up to >98% and >99% ee . Preparative-scale deracemization of racemic 1,2,3,4-tetrahydroisoquinoline-1-carboxylic acid and 1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid was also demonstrated with good isolated yields (82% and 73%, respectively) and ee >99%. Our study provides an effective method for the synthesis of enantiomeric pure 1,2,3,4-tetrahydroisoquinoline carboxylic acids. This method is expected to provide access to chiral carboxyl-substituted 1,2,3,4-tetrahydroquinolines and 1,2,3,4-tetrahydro-ß-carbolines.

A 3-D-Silicon Nanowire FET Biosensor Based on a Novel Hybrid Process

Xiaofeng Gong,Rui Zhao,Xiaomei Yu

Single-silicon nanowire field-effect transistor (SiNW-FET) devices are powerful tools for the detection and quantification of biochemical molecules. However, traditional SiNW-FET-based biosensors demonstrate low output signals and high fluctuation noises, which results from the tiny scale of a single channel and electrical property deterioration caused by the dry etching process. To improve the performance of the SiNW-FET biosensors, here we developed a 3-D-SiNW-FET by using a hybrid process of deep reactive ion etching and isotropic wet etching with HF, HNO3 and HAc (HNA) mixed solution. The 3-D-SiNW-FET that included 5 × 7 nanowire channels with a wire diameter of approximately 300 nm was successfully fabricated with the novel hybrid process. Compared with the performance of single-and 2-D-SiNW-FETs, the 3-D-SiNW-FET showed a fairly good ability to stabilize and enhance the source-drain current by parallelizing the signals of the multiple channels. With the fabricated 3-D-SiNW-FET, immunoglobulin G was detected at a concentration as low as 5 fM.
