The development of a multifunctional single molecule phototherapeutic agent with excellent fluorescence imaging, photothermal therapy and photodynamic therapy at the same time is still a challenging task, which mainly arises from the low absorbance of the molecule, and the complexity of energy dissipation and molecular design. Herein, four donor–acceptor (D–A) compounds were synthesized by linking triphenylamine (TPA), thiophene/thieno[3,2-b]thiophene and different cyano acceptor structures. In this design, we propose a molecular design strategy to redshift absorption and increase the molar extinction coefficient (ε) by enhancing electron-withdrawing acceptors and enlarging the π-conjugation plane unit. Due to the twisted structure of TPA, these compounds exhibit aggregation-induced emission (AIE) characteristics. Notably, these AIEgens have long emission wavelengths, excellent photostability, biocompatibility, photothermal stability and singlet oxygen (1O2) generation performance. Among them, the photothermal conversion efficiency of a compound (named TCF-SS-TPA NPs) can reach 84.5%. Cellular internalization and therapy showed that TCF-SS-TPA NPs have good biocompatibility, excellent cell bioimaging and cancer phototherapy capabilities in vitro. This study will stimulate the molecular design of multifunctional phototherapeutics to realize effective synergistic cancer therapy.

Mitochondrial membrane potential (Δ Ψ m)-targeting molecular probes play an essential role in diagnosing and treating diseases. Recently, the second near-infrared window (NIR-II, 1000–1700 nm) fluorescent imaging has been actively studied as an attractive imaging modality. However, small molecule Δ Ψ m-targeting NIR-II probes are rarely reported, especially for highly efficient imaging and treatment of tumors. Herein, a small molecule probe named TQPTPP is designed and synthesized by conjugation of a novel D-A type dye TQT1009 with Δ Ψ m-targeting molecule alkyl triphenylphosphine (TPP) through a polyethylene glycol-8 (PEG 8 ) linker. The conventional ICG dye is also coupled with TPP through PEG 8 to produce ICGTPP as a comparison. TQPTPP showed a fluorescence quantum yield of 0.041% and excellent photothermal conversion efficiency (61.4%). It can be self-assembled into nanoparticles and still preserve Δ Ψ m-targeting capability. Tumor imaging is further performed, and results showed a long tumor retention time of TQPTPP (maximum tumor signal on day five and signal-to-noise ratio up to nine). As a comparison, ICGTPP remained a single molecule with Δ Ψ m-targeting capability. But it has shorter tumor retention and lower photostability. These results suggested the novel D-A small molecule ΔΨ m-targeting NIR-II probe TQPTPP provided a new tool for diagnosing and treating tumors.

Near-infrared (NIR) light-triggered photocatalysis antibacterial therapy has attracted significant attention. Herein, we designed a straddle-type heterostructure film of ZnS/Ag 2 S nanorod on the titanium implant, which showed excellent NIR photocatalytic ability. Under NIR, the electrons of Ag 2 S cascaded jump to ZnS conduction band (CB), and the electron of ZnS was half-jumped to Ag 2 S valence band (VB). The VB of Ag 2 S acted as a consumption platform for the holes of Ag 2 S and the electrons of ZnS, improving photocatalytic efficiency. To prove the feasibility of this mechanism, another straddle-type heterostructure of ZnS/Bi 2 S 3 with similar energy band structure was fabricated, and its NIR photocatalytic ability was also distinctly enhanced. Furthermore, the ZnS/Ag 2 S heterostructure films rapidly removed bacteria in vitro and in vivo under the NIR irradiation. This work innovatively discovered NIR photocatalytic effect of the straddle-type heterostructure, which complemented the design of heterostructures in photocatalysis.

Near-infrared responsive photothermal therapy is an emerging and non-invasive cancer treatment. However, the inability of real-time imaging tracking and monitoring for the photothermal agents (PTAs) severely limits the accuracy of treatment. In this study, a series of visual xGd-WO 3 @PEG PTAs were synthesized by a one-pot solvothermal method. The effect of different Gd doping amounts on the structure, morphology, and photothermal performance has been investigated in detail. Under the near-infrared laser irradiation, the samples exhibited satisfied photothermal properties of hyperthermia in a short time. The photothermal conversion efficiency and stability of the xGd-WO 3 @PEG (x = 7 %) were further investigated. Subsequently, the near-infrared thermal and CT imaging performance was studied. The in vitro cell viability on LO2 normal and 4T1 cancer cells have also been conducted. The results showed the xGd-WO 3 @PEG nanomaterials have excellent potential in imaging-guided photothermal cancer therapy.

The inadequate quantity of hydrogen peroxide (H2O2) in cancer cells promptly results in the constrained success of chemodynamic therapy (CDT). Significant efforts made throughout the years, nevertheless, researchers are still facing the great challenge of designing a CDT agent and securing H2O2 supply within the tumor cell. In this study, taking advantage of H2O2 level maintenance mechanism in cancer cells, a nanozyme-based bimetallic metal-organic frameworks (MOFs) tandem reactor is fabricated to elevate intracellular H2O2 levels, thereby enhancing CDT. In addition, under near-infrared excitation, the upconversion nanoparticles (UCNPs) loaded into the MOFs can perform photocatalysis and generate hydrogen, which increases cellular susceptibility to radicals induced from H2O2, inhibits cancer cell energy, causes DNA damages and induces tumor cell apoptosis, thus improving CDT therapeutic efficacy synergistically. The proposed nanozyme-based bimetallic MOFs-mediated CDT and UCNPs-mediated hydrogen therapy act as combined therapy with high efficacy and low toxicity.

The colon tumor microenvironment has a high concentration of H 2 S and glutathione, which is highly immunosuppressive and adverse to multiple therapeutic methodologies such as ferroptosis. Here, an engineered microbial nanohybrid based on Escherichia coli ( E. coli ) and Cu 2 O nanoparticles to specific colon tumor therapy and immunosuppression reversion is reported. The as-prepared E. coli @Cu 2 O hybrid can accumulate in tumor sites upon intravenous injection, and Cu 2 O nanoparticles convert to Cu x S by consuming the endogenous H 2 S, which exhibits strong photothermal conversion at near-infrared II (NIR II) biological window. Furthermore, E. coli @Cu 2 O is able to induce cellular ferroptosis and cuproptosis through inactivation of glutathione peroxidase 4 and aggregation of dihydrolipoamide S-acetyltransferase, respectively. Photothermal-enhanced ferroptosis/cuproptosis achieved by E. coli @Cu 2 O reverses the immunosuppression of colon tumors by triggering dendritic cell maturation (about 30%) and T cell activation (about 50% CD8 + T cells). Concerted with immune checkpoint blockade, the engineered microbial nanohybrid can inhibit the growth of abscopal tumors upon NIR illumination. Overall, the designed microbial nanohybrid can achieve tumor-specific photothermal-enhanced ferroptosis/cuproptosis and immunosuppression reversion, showing promise in precise tumor therapy in future clinical translation.

Theranostic nanoplatforms integrating simultaneously photodynamic therapy (PDT) and photothermal therapy (PTT) exhibit intrinsic advantages in tumor therapy due to distinct mechanisms of action. However, it is challenging to achieve PDT and PTT under single near-infrared (NIR) laser irradiation with a nanoplatform utilizing conventional organic photodynamic agent and inorganic photothermal agent owing to the difference in inherent excitation wavelengths. Particularly, the single NIR light (660 nm)-triggered PTT and PDT nanoplatform, constructed from chlorin e6 (Ce6) and copper sulfide (CuS) nanoparticles (NPs), has never been reported. Herein, we, for the first time, designed and established a dual-modal phototherapeutic nanoplatform that achieved both PTT and PDT under single NIR laser (660 nm) irradiation for Ce6 and CuS NPs with the strategy of core–shell structured CuS@Carbon integrated with Ce6. Introducing of carbon shell not only endows small CuS NPs with excellent tumor accumulation, but also significantly strengthens the photothermal performance of CuS NPs, realizing efficient photothermal performance under 660 nm laser irradiation. Moreover, Ce6 in carbon shell endowed the nanoplatform with photodynamic effect under 660 nm laser irradiation. The as-prepared Ce6/CuS@Carbon nanoplatform thus achieved dual-modal phototherapy under single NIR laser irradiation, significantly inhibiting tumor growth with minimal adverse effects and superior biosafety.

Considering the key roles of macrophages in tissue repair and immune therapy, designing smart biomaterials able to harness macrophage phenotypes on demand during the healing process has become a promising strategy. Here, a novel “sandwich” cell culture platform with near-infrared (NIR) responsive dynamic stiffness was fabricated to polarize bone marrow-derived macrophages (BMDMs) in situ for revealing the relationship between the macrophage phenotype and substrate stiffness dynamically. Under NIR irradiation, calcium ions (Ca2+) diffused through the middle layer of the IR780-mixed phase change material (PCM) due to the photothermal effect of IR780, resulting in an increase of hydrogel stiffness in situ by the crosslinking of the upper layer of the hyaluronic acid-sodium alginate hydrogel (MA-HA&SA). The up-regulation of inducible nitric oxide synthase (iNOS) and tumor necrosis factor-α (TNF-α) was quantified by immunostaining and enzyme-linked immune sorbent assay (ELISA), respectively, indicating the transformation of macrophages from the anti-inflammatory to pro-inflammatory phenotype under dynamic stiffness. The nuclear Yes-associated-protein (YAP) ratio positively correlated with the shift of the macrophage phenotype. The modulation of macrophage phenotypes by stiffness-rise without the stimuli of cytokines offers an effective and noninvasive strategy to manipulate immune reactions to achieve optimized healing or therapeutic outcomes.

Bacterial infections caused by biofilms are severe clinical problems, resulting in high drug resistance by limiting the penetration of antibiotics. Herein, a near-infrared (NIR)-activated chem/photodynamic/photothermal combined therapeutic agent is proposed by loading fluorescein isothiocyanate (FITC), ultrasmall copper sulfide nanoparticles (Cu2–xSNPs), and ε-polylysine (PLL) onto mesoporous silica nanoparticles (MSNs) through a layer-by-layer self-assembly approach. FITC-doped MSNs are prepared to monitor the permeability and accumulation of nanocomposites into biofilms. MSNs can also act as hosts for the synthesis of ultrasmall Cu2–xSNPs, which has effective photodynamic and photothermal ablation against bacteria under NIR light irradiation. Moreover, biodegradable PLL introduced can not only enhance adhesion toward the bacterial surface to increase the effectiveness of phototherapy but also damage bacteria through electrostatic interaction. As a result, the prepared nanocomposites could not only penetrate biofilms but also ablate biofilms through combined chem/photodynamic/photothermal effects under NIR light irradiation. Furthermore, the nanocomposites could treat bacterial infections in vivo with negligible tissue toxicity. Overall, the finely designed nanocomposites are anticipated to display promising applications in imaging-guided chem/photodynamic/photothermal combined therapy for bacterial infections.

Photodynamic therapy (PDT) is an appealing treatment for oncologic skin diseases owing to its high spatiotemporally selectivity and non-invasive features. However, its application in melanoma has been restricted by limited penetration depth of ultraviolet (UV)-visible (vis) light, melanin-optical filtration and weak immunogenicity. To circumvent the incomplete tumor damage and insufficient anti-tumor immunity, we prepared a mesoporous coated upconverting nanoparticles (NaYF 4 :Yb, [email protected] 4 @mSiO 2 @lipsome)-based platform by co-loading chlorin e6 (Ce6) and buthionine sulfoximine (BSO). This platform converts near-infrared (NIR) light to UV-vis light which enables the destruction of deeply seated nidus, amplification of ferroptotic and apoptotic cell death, as well as an increase in the anti-tumor immunity. Under NIR laser irradiation, NaYF 4 :Yb, [email protected] 4 @mSiO 2 @lipsome nanoparticles embedded with Ce6 were designed to ease burden in tumor sites via elevating levels of hydroxyl radicals and the induction of apoptosis. Meanwhile, glutathione depletion was induced through BSO co-administration and direct thiol oxidation, followed by the inactivation of lipid peroxide repair enzyme and initiation of ferroptosis. This combinational strategy resulted in an amplification in intratumoral oxidative stress, which was accompanied with the strong cytotoxicity in vitro and in vivo . The aggravated anti-tumor performance then brought about the exposure of damage-associated molecular patterns, followed by promoted maturation of dendritic cells and effector function of tumor-infiltrating lymphocytes. Taken together, this ferroptosis-apoptosis combined photodynamic strategy provides insights in designing nanomedicines for immunotherapy in melanoma.

Cesium-doped tungsten bronze CsxWO3 (CWO) is an ideal near infrared (NIR) shielding material for solar filters. However, the NIR shielding ability of CWO-dispersed films easily deteriorates in hot humid environments, which severely hinders the commercial application of CWO. In this paper, UV/NIR shielding nanocomposite films were prepared by dispersing core–shell structured CWO@polydopamine (CWO@PDA) in a poly(vinyl alcohol) matrix. Because of the strong ultraviolet light absorption ability of PDA, it can shield ultraviolet light, which is generally detrimental to our health. The prepared nanocomposite films can efficiently shield 88.3% UV and 85.5% NIR radiation even though they show relatively high transparency in the visible range. Importantly, the good protection of the continuous PDA shells played an important role in enhancing the stability of CWO nanoparticles. The nanocomposite films also exhibit excellent stability in hot humid environments. Therefore, core–shell structured CWO@PDA nanoparticles have great potential as a novel UV/NIR shielding material for the development of efficient energy-saving windows.

Microbial contamination of water represents a great threat to the public health that has attracted worldwide attention. In this work, polypyrrole magnetic nanoparticles (Fe 3 O 4 @PPy NPs) with sterilization properties were fabricated. More specifically, the Fe 3 O 4 @PPy NPs obtained via aqueous dispersion polymerization and an in situ chemical oxidative polymerization exhibited a cationic surface and high photothermal conversion efficiency. More than 50% of bacteria adsorption can be achieved at a dosage of 100 μg/mL Fe 3 O 4 @PPy NPs under magnetic field, and high photothermal sterilization efficacy (~100%) can be obtained upon NIR exposure at the same dosage for 10 min. Noteworthy, the Fe 3 O 4 @PPy NPs can be recycled by magnetism and reused without affecting their photothermal sterilization capability. This study clearly provides experimental evidence of the great potential of Fe 3 O 4 @PPy NPs as stable and reusable nanocomposite materials for bacteria adsorption and photothermal sterilization performance. The application of Fe 3 O 4 @PPy NPs can realize enviromental-friendly bacterial contaminated water treatment as well as provide stratgies for synergistical antibacterial materials design.
