Novel dam-like effect based on piezoelectric energy conversion for drug sustained release of drug-loaded TiO2 @ BaTiO3 coaxial nanotube coating

Cong Wu,Xinghui Wei,Kang Zhao,Junyi Jiang,Mengzhen Jiao,Junhua Cheng,Zhen Tang,Zheng Guo,Yufei Tang

Nanotube coatings on titanium surfaces have favorable biocompatibility and structures and are widely used in local drug delivery. However, when a drug freely diffuses out of a nanotube, it leads to an explosive release, making the duration of action extremely short. In this study, a high concentration of vancomycin hydrochloride region similar to a dam was formed on a nanotube coating surface by exploiting the attraction of the electric charge generated by the energy conversion of the piezoelectric effect to vancomycin hydrochloride. The “dam” not only reduces the diffusion rate of vancomycin hydrochloride inside the nanotube into the dam and the diffusion rate of the dam to the outside but also forms a high concentration of vancomycin hydrochloride action region. The free diffusion rate of vancomycin hydrochloride loaded on the TiO 2 @ BaTiO 3 coaxial nanotubes was reduced by the piezoelectric effect, and the cumulative release of vancomycin hydrochloride for 7 days is reduced by 54.8%. The polarized vancomycin-containing coaxial nanotube coating has a long-lasting antibacterial effect on Staphylococcus aureus . The TiO 2 @ BaTiO 3 coaxial nanotubes loaded with vancomycin hydrochloride coating with piezoelectric properties has potential application value in controlled drug delivery.

Fe3O4 Magnetic Cores Coated with Metal–Organic Framework Shells as Collectable Composite Nanoparticle Vehicles for Sustained Release of the Pesticide Imidacloprid

Wenyan Meng,Yiyun Gao,Zhenfen Tian,Weijie Xu,Jiagao Cheng,Shengke Li,Aihua Zou

A magnetic metal–organic framework nanocomposite as a collectable pesticide delivery vehicle was developed through the incorporation of UiO-66 with polydopamine-coated Fe3O4 (Fe3O4@PDA@UiO-66). The obtained Fe3O4@PDA@UiO-66 nanocomposite was verified by X-ray diffraction, Fourier transform infrared spectroscopy, transmission electron microscopy, and zeta potential test. The magnetic collectability of Fe3O4@PDA@UiO-66 was demonstrated through a magnetic hysteresis loop study (∼2 emu/g) and magnetic collection experiment (up to a 96% collection rate). Imidacloprid (IMI) was employed as a model pesticide to be loaded into the Fe3O4@PDA@UiO-66 nanocomposite, with 15.87% drug loading. IMI was found to exhibit sustained-release behavior after being loaded into the delivery vehicle, and magnetic Fe3O4@PDA@UiO-66 can be readily collected after IMI release, which is beneficial to constrain the hazardous effects of the vehicles and the residual pesticide on the ecosystem. The insecticidal activity of IMI@Fe3O4@PDA@UiO-66 toward Aphis craccivora Koch was found to be superior to that of free IMI, and the efficacy was comparable to that of the commercial 70% IMI water dispersible granule, indicating its potential as a magnetic collectable green carrier for pesticides in agriculture.

Valorization of lignin through zein Co-assembly into green nanopesticides for photostable, sustained-release abamectin delivery with insecticidal efficacy and biosafety

Zhaohui Liang,Ze Zhou,Liheng Chen,Xueqing Qiu

Industrial alkali lignin is an abundant aromatic biological macromolecule with intrinsic UV shielding ability, but its structural heterogeneity and aqueous self-aggregation limit its use as a stable carrier matrix. Herein, industrial alkali lignin was valorized through zein regulated co-assembly to construct surfactant free hybrid biological macromolecular nanocapsules (LZNP@Abm) for abamectin (Abm) delivery. The composite macromolecular matrix afforded excellent colloidal stability and a loading capacity of 79.22%, and LZNP@Abm retained 74.94% Abm after 72 h of UV irradiation, effectively mitigating photolytic degradation. By integrating lignin derived UV shielding with zein mediated interfacial stabilization and pH and temperature responsive gating, LZNP@Abm suppressed release, with only 34.18% release after 180 h under weakly acidic leaf surface and soil conditions, while accelerating release at pH 9 and 35 °C, conditions relevant to alkaline and warm lepidopteran larval midguts. Molecular dynamics and density functional theory analyses identified key zein residues, including asparagine, serine, and proline, that enhanced Abm binding through cooperative hydrogen bonding and van der Waals interactions. Improved foliar wettability and liquid retention further promoted leaf deposition and reduced potential non-target pesticide loss. LZNP@Abm achieved 5.7–23.5-fold reductions in LC 50 against Spodoptera frugiperda and complete mortality of S. litura , restored seed germination to 85.56%, and maintained earthworm survival at 83.33 to 96.67%. This work demonstrates a mechanism informed biopolymer carrier strategy for converting industrial lignin into a functional pesticide delivery platform with photoprotection, controlled release, and improved biosafety. Construction of composite nanoparticles of β-cyclodextrin and corn starch encapsulating 5-hydroxytryptophan for sustained release and sleep improvement in vivo

Lin Gao,Luping Gu,Wentian Li,Gang Wang,Jianxin Zhao,Wei Chen,Peijun Tian

5-Hydroxytryptophan (5-HTP) is a functional food ingredient widely recognized for its sleep-enhancing properties, but it has a short elimination half-life in vivo , resulting in suboptimal pharmacokinetic performance. In this study, we developed a composite nanoparticles system based on β-cyclodextrin and corn starch using the anti-solvent precipitation method for the encapsulation and sustained release of 5-HTP. The nanoparticles were characterized using laser particle size analysis, Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and evaluation of their digestive properties in simulated gastrointestinal fluids. We further evaluated their effects on the pharmacokinetic profile and distribution of 5-HTP in rats, including serum, gut, and brain, demonstrating a significant extension of the drug elimination half-life and improved bioavailability. In an insomnia mouse model, administration of the composite nanoparticles system significantly alleviated behavioral abnormalities and circadian rhythm disorder induced by sleep deprivation. This study provides a solid theoretical basis and technical support for the efficient application of 5-HTP in functional foods.

Cisplatin-directed coordination-crosslinking nanogels with thermo/pH-sensitive triblock polymers: improvement on chemotherapic efficacy via sustained release and drug retention

Hao Zhao,Jiabao Xu,Jiangshan Wan,Shinan Geng,Han Li,Xiaole Peng,Qianwen Fu,Ming He,Yanbing Zhao,Xiangliang Yang

To realize the sustained release and long-term intratumoural retention of water-soluble cisplatin, thermo/pH-sensitive cisplatin-directed coordination-crosslinking nanogels (Pt-PNA) were developed via the coordination bonds of Pt–carboxyl groups. As the coordination ratio (CR) of the Pt–carboxyl bonds increased from 5% to 35%, the sizes of the Pt-PNA nanogels decreased from 999 nm to 167 nm, and their zeta potentials increased from −35 mV to −13 mV. Only through a simple mixing of cisplatin and PNAs, the entrapment efficiencies (EEs) of the Pt-PNA nanogels reached near 100% (>90%), and the drug-loading amounts (DLs) of cisplatin could achieve up to 25.5 ± 0.1%. For water-soluble cisplatin, Pt-PNA nanogels exhibited a sustained release for as long as 5 days. The thermo/pH-sensitive sol–gel phase-transition behaviour of the Pt-PNA nanogels were investigated via inverting-vial and rheological methods. Platinum elemental analysis indicated that the Pt-PNA nanogels showed a much stronger ability of cisplatin retention in tumours than free cisplatin. The platinum content in a tumour treated by the Pt-PNA nanogels was far higher than that by free cisplatin: 200.7 ± 63.6 μg vs. 82.7 ± 26.8 μg at the 1st day, or 118.9 ± 35.2 μg vs. 18.5 ± 9.4 μg at the 14th day. The evaluation of the in vivo antitumour efficacy indicated that only after a single dose of Pt-PNA nanogels, the tumour volume continuously decreased to 0.73 ± 0.07 times that of the original tumour volume (OTV) for 14 days; however, it rapidly increased by 3.37 ± 0.82, 8.01 ± 0.53 and 9.25 ± 1.85 times that of the OTV with the same dose of free cisplatin, PNA, and NS, respectively. Some preliminary evaluations of the biocompatibility indicated that the toxic side effects of cisplatin could be greatly improved via cisplatin-directed coordination-crosslinking with PNA. As a result, Pt-PNA nanogels could likely become a promising versatile strategy for improving antitumour efficacy and reducing the toxicity and size effects of platinum-based drugs, and they could also be developed as promising nanomedicines for regional chemotherapy.

Sequential and sustained release of SDF-1 and BMP-2 from silk fibroin-nanohydroxyapatite scaffold for the enhancement of bone regeneration

Xiaofeng Shen,Yanxia Zhang,Yong Gu,Yun Xu,Yong Liu,Bin Li,Liang Chen

In this study, a cell-free bone tissue engineering system based on a silk fibroin (SF)/nano-hydroxyapatite (nHAp) scaffold was developed, in which two bioactive molecules , stromal cell derived factor-1 (SDF-1) and bone morphogenetic protein-2 (BMP-2), were embedded and released in a sequential and controlled manner to facilitate cell recruitment and bone formation, respectively. BMP-2 was initially loaded into SF microspheres , and these BMP-2 containing microspheres were subsequently encapsulated into the SF/nHAp scaffolds, which were successively functionalized with SDF-1 via physical adsorption. The results indicated rapid initial release of SDF-1 during the first few days, followed by slow and sustained release of BMP-2 for as long as three weeks. The composite scaffold significantly promoted the recruitment of bone marrow mesenchymal stem cells (BMSCs) and osteogenic differentiation of them in vitro . Further, the in vivo studies using D-Luciferin-labeled BMSCs indicated that implantation of this composite scaffold markedly promoted the recruitment of BMSCs to the implanted sites. Enhanced bone regeneration was identified at 12 weeks' post-implantation. Taken together, our findings suggested that the sequential and sustained release of SDF-1 and BMP-2 from the SF/nHAp scaffolds resulted in a synergistic effect on bone regeneration. Such a composite system , therefore, shows promising potential for cell-free bone tissue engineering applications .

The sustained-release behavior and in vitro and in vivo transfection of pEGFP-loaded core-shell-structured chitosan-based composite particles

Yun Wang,Fu-xing Lin,Yu Zhao,Mo-zhen Wang,Xue-wu Ge,Zheng-xing Gong,Dan-dan Bao,Yu-fang Gu

Novel submicron core-shell-structured chitosan-based composite particles encapsulated with enhanced green fluorescent protein plasmids ( pEGFP ) were prepared by complex coacervation method. The core was pEGFP -loaded thiolated N-alkylated chitosan (TACS) and the shell was pH- and temperature-responsive hydroxybutyl chitosan (HBC). pEGFP -loaded TACS-HBC composite particles were spherical, and had a mean diameter of approximately 120 nm, as measured by transmission electron microscopy and particle size analyzer. pEGFP showed sustained release in vitro for >15 days. Furthermore, in vitro transfection in human embryonic kidney 293T and human cervix epithelial cells, and in vivo transfection in mice skeletal muscle of loaded pEGFP , were investigated. Results showed that the expression of loaded pEGFP , both in vitro and in vivo, was slow but could be sustained over a long period. pEGFP expression in mice skeletal muscle was sustained for >60 days. This work indicates that these submicron core-shell-structured chitosan-based composite particles could potentially be used as a gene vector for in vivo controlled gene transfection.

Kirkendall effect enhanced sustained-release Fe-C composites for long-term reductive dechlorination of trichloroethylene: Interfacial reactions and slow-release degradation

Daibing Hou,Xuedan Cui,Yiming Tang,Meng Liu,Hantong Qie,Pengjie Zhao,Wenpeng Leng,Nan Luo,Huilong Luo,Wenxia Wei,Aijun Lin,Tianwen Zheng

Rebounding and lagging during the process of remediating contaminated sites are challenging problems. A potential solution is developing slow-release materials that could assist in the long-term remediation of contaminated sites. This study synthesised a slow-release Fe-C composite (CSBC-nZVI@β-CD), which exhibits a synergistic effect of encapsulated layers and nanocracks, using a liquid-phase reduction method. Furthermore, the reduction and dechlorination mechanism of trichloroethene (TCE) was systematically investigated. The introduction of β-cyclodextrin (β-CD) intensified the Kirkendall effect, forming nano zero-valent iron (nZVI) with abundant radial nanocracks and a litchi-like appearance on the biochar surface. Additionally, β-CD created an encapsulation layer measuring 5–7 nm on the surface of nZVI, improving its electron transport capacity and hydrophobic properties. The CSBC-nZVI@β-CD nanocomposite successfully removed 99 % of TCE in solution within 2 h and accomplished a degradation efficiency of 98.9 % within 15 d. The synergistic effect of the β-CD encapsulating layer and nanocracks facilitated the reactivity and electron-retarding capacity, resulting in a broad pH tolerance and anti-interference ability. Density-functional theory (DFT) calculations and experimental methods were used to study the mechanism of TCE reduction dechlorination; the unique structure of β-CD tended to bind with Fe as well as adsorb TCE, thereby facilitating electron transfer and the enrichment of TCE at the reaction interface. In addition, the sustained-release Fe-C composites reduced the bonding energy required during the TCE degradation reaction and promoted the complete reaction of the product. Overall, CSBC-nZVI@β-CD exhibited significant potential for the long-term remediation of groundwater.

Hydrogen bond-mediated reversible BHT/PVA nanocarriers integrating sustained release kinetics and small-size synergy for potentiated pesticide stability and longevity

Hongfa Duan,Xiaochi Liu,Lulu Bian,Zhongyi Li,Ning Li,Muhammad Umair Sial,Zhe Sun,Haiqiang Gao,Beixing Li,Daxia Zhang,Feng Liu

The preparation of pesticide-loaded microcapsules using biodegradable polymers and the balance between their sustained-release performance and small-size effects represent an effective strategy to enhance pesticide utilization efficiency. This study devised a one-step sol-gel self-assembly approach to fabricate abamectin-loaded microcapsules using biodegradable polyvinyl alcohol (PVA) and butylated hydroxytoluene (BHT) as raw materials. The results demonstrated that the reaction through hydrogen bonding by PVA and BHT at the o/w interface could generate shells and encapsulate pesticides to make microcapsules. Increasing PVA usage could reduce the particle size and enhance shell stability. The addition of BHT also improved the shell structure stability and encapsulation efficiency, whereas 3 %–7 % concentrations impeded the formation of the shell structure. Abamectin, which has a self-polymerizing ability, also increases microcapsule stability. A 54 °C temperature increase was maintained for 24 h to induce emulsion separation. Subsequent cooling and agitation prompts the reconstitution of pesticide-loaded microcapsules. The synergistic effect of the water-responsive mechanism, high encapsulation efficiency, and thicker shells endows nano-formulations with superior sustained-release performance compared to micro-scale formulations. The photostability of abamectin nanocapsules is significantly enhanced by 68.26 % compared to conventional suspensions, and the content of active ingredients remains at 50.54 % after 60 h of release under light conditions. Owing to their nanoscale sustained-release characteristics, their biological safety is improved by 36.43 %. The encapsulated Aba@BHT-PVA nano-system, leveraging excellent rainwater erosion resistance and leaf surface affinity, significantly prolongs its leaf retention time. Synergizing with the small-size effect, it demonstrates substantial enhancement in both the control efficacy and persistence against Plutella xylostella . Owing to its unique properties and easy regulation, this fabrication technique has great application value in the future.

Layers of Electrostatically Self-assembled Composite Hydrogel Beads Based on Porous Starch and ZnO Filling for Intestinal Sustained Release of Quercetin

Shuqi Guo,Xueze Zhang,Shangxuan Li,Huifang Wang,Xianghua Gao,Baolong Niu,Wenfeng Li

Quercetin (Que) is a naturally occurring bioactive compound with various potential health benefits, but its low stability and low bioaccessibility limit its application in functional foods. Therefore, there is an urgent need to prepare a novel composite hydrogel bead with efficient and controlled release. Specifically, porous starch (PS) was first prepared by enzymatic hydrolysis to increase the specific surface area and thus enhance the adsorption rate of Que. Scanning electron microscopy confirmed the formation of the PS and the adsorption of Que. The maximum swelling properties of carboxymethylcellulose-porous starch composite hydrogel beads (CQPZ) were reduced by adding different amounts of zinc oxide to achieve slow release. The results showed that the higher the amount of zinc oxide, the stronger the slow-release performance, while the lower the drug loading capacity. To obtain composite hydrogel beads with both low swelling and high drug-carrying properties, bilayer gel beads (CQPZ/CS) were prepared by electrostatic self-assembly of carboxymethyl cellulose and chitosan. The experimental results showed that the drug loading was significantly increased by nearly 2-fold while enhancing the slow-release performance. In addition, the CQPZ/CS composite hydrogel beads enhanced the stability of Que under light and heat conditions, and the DPPH radical scavenging rate after UV irradiation was still greater than 75%, which was much higher than that of free quercetin (31.08%). In conclusion, the prepared hydrogel beads are a promising delivery system for bioactive compounds.

A double-network fish gelatin/sodium alginate composite hydrogel as a carrier for the sustained release of a soy-derived osteogenic peptide

Jinpeng Gong,Boya Liu,Tao Huang,Lu Wei,Yupeng Ma,Hao Chen,Jing Gan,Junbo Ge

Osteoporosis, a prevalent metabolic bone disease, poses a significant challenge for bone repair in aged populations. Soy-derived osteogenic peptide (SOP) holds promise but its clinical translation is hampered by rapid enzymatic degradation and poor oral bioavailability, representing a major challenge that necessitates the development of a protective delivery system. To address this, we developed a novel composite hydrogel based on fish gelatin and sodium alginate (SA/FG) as an oral delivery vehicle to overcome these limitations. Systematic characterization revealed that the SA/FG formulation offers distinct advantages: the incorporation of an optimal amount of SA significantly enhanced the mechanical strength and stability of the hydrogel, addressing the common weakness of single-network gelatin hydrogels, while maintaining a high water content (~90%) and a superior rehydration capacity (83.65% in distilled water). In a glucocorticoid-induced zebrafish osteoporosis model, the SOP-loaded SA/FG hydrogel significantly increased skull bone mass by 120.17% and improved larval swimming behavior, markedly outperforming the free SOP group. This study presents the first report of an SA/FG composite hydrogel for effective oral delivery of an osteogenic peptide. The system demonstrates great potential as a functional food or supplement for preventing and treating osteoporosis, offering an innovative strategy to enhance peptide stability and bioavailability through a biocompatible, double-network carrier.

Functionalization of mesoporous silica as an effective composite carrier for essential oils with improved sustained release behavior and long-term antibacterial performance

Ximing Zhong,Fan Gao,Hongjie Wei,Hongjun Zhou,Xinhua Zhou

In this work, a novel composite carrier system for loading essential oils was developed by using tetraethyl orthosilicate (TEOS) and (3-aminopropyl) triethoxysilane (APTES) as silica precursors and cetyl trimethyl ammonium bromide (CTAB) as a template, and the resultant aminated mesoporous silica was further chemically modified by polyacrylic acid (PAA). The obtained composite carriers exhibited a high loading capability toward tea tree oil (TTO), and they also significantly improved the release behavior of TTO due to the steric hindrance of silica mesopore and the polymer restriction. Besides, it was found that the release behavior followed the First-Order kinetic model, revealing that the release of TTO was driven by the concentration gradient. In addition, these composite carriers with essential oil-loaded demonstrated remarkable antibacterial performance against E. coli and S. aureus, and they could retain antibacterial performance even after 50 d. Moreover, the antibacterial mechanism was also elucidated with the assistance of nucleic acid and conductivity measurements. Therefore, this work provides a facile and environmentally friendly approach to preparing effective composite carriers for improving the sustained release of essential oils, and the long-term antibacterial performance of these essential oil-loaded composite carriers makes them tremendously potential for practical applications.
