Isotachophoresis (ITP) coupled to zone electrophoresis (ZE), either free zone electrophoresis (FZE) or gel electrophoresis (GE), carried out mainly in capillaries and, although less frequently, also in microchips, is a powerful preconcentration and separation technique which has been successfully used for the study of many low molecular-weight analytes. However, this analytical technique has been scarcely applied for proteins separation. In this work, an on-chip transient ITP coupled to free zone electrophoresis (t-ITP-MFZE) mode with LIF detection is developed for the preconcentration and separation of the proteins α-lactalbumin and β-lactoglobulin. Firstly, for LIF detection, the proteins were off-chip fluorescently labeled with the fluorogenic reagent Chromeo P503. Then, several separation parameters in t-ITP-MFZE mode such as leading electrolyte, terminating electrolyte, separation voltage, and injection time were optimized to achieve the maximum sensitivity while maintaining an adequate resolution between α-lactalbumin and β-lactoglobulin in a single column configuration t-ITP. Using the optimized electrolytes (50 mM imidazole/HCl pH = 8 as leading electrolyte and 100 mM imidazole/12 mM HEPES pH = 8 as terminating electrolyte) separation of both proteins was achieved in less than 4 min with peak resolution of 1.5. The LODs were 55 nM and 380 nM, for α-lactalbumin and β-lactoglobulin, respectively, which are adequate for some food allergenicity studies. Finally, comparison of the optimized t-ITP-MFZE method to the equivalent MFZE method, carried out also in microchips but without the isotachophoretic preconcentration step, provided preconcentration indexes for both proteins around 10.

Isolating and in situ profiling the heterogeneous molecular phenotype of circulating tumor cells are of great significance for clinical cancer diagnosis and personalized therapy. Herein, an on-chip strategy is proposed that combines size-based microfluidic cell isolation with multiple spectrally orthogonal surface-enhanced Raman spectroscopy (SERS) analysis for in situ profiling of cell membrane proteins and identification of cancer subpopulations. With the developed microfluidic chip, tumor cells are sieved from blood on the basis of size discrepancy. To enable multiplex phenotypic analysis, three kinds of spectrally orthogonal SERS aptamer nanovectors are designed, providing individual cells with composite spectral signatures in accordance with surface protein expression. Next, to statistically demultiplex the complex SERS signature and profile the cellular proteomic phenotype, a revised classic least square algorithm is employed to obtain the 3D phenotypic information at single-cell resolution. Combined with categorization algorithm partial least square discriminate analysis, cells from different human breast cancer subtypes can be reliably classified with high sensitivity and selectivity. The results demonstrate that this platform can identify cancer subtypes with the spectral information correlated to the clinically relevant surface receptors, which holds great potential for clinical cancer diagnosis and precision medicine.

Traces of mercury ions in environmental water can harm humans and animals. Paper-based visual detection methods have been widely developed for the rapid detection of mercury ions; however, existing methods are not sensitive enough to be used in real environments. Here, we developed a novel, simple and effective visual fluorescent sensing paper-based chip for the ultrasensitive detection of mercury ions in environmental water. CdTe-quantum-dots-modified silica nanospheres were firmly absorbed by and anchored to the fiber interspaces on the paper’s surface to effectively avoid the unevenness caused by liquid evaporation. The fluorescence of quantum dots emitted at 525 nm can be selectively and efficiently quenched with mercury ions, and the ultrasensitive visual fluorescence sensing results attained using this principle can be captured using a smartphone camera. This method has a detection limit of 2.83 µg/L and a fast response time (90 s). We successfully achieved the trace spiking detection of seawater (from three regions), lake water, river water and tap water with recoveries in the range of 96.8–105.4% using this method. This method is effective, low-cost, user-friendly and has good prospects for commercial application. Additionally, the work is expected to be utilized in the automated big data collection of large numbers of environmental samples.

Low-concentration molecular detection is of great value in chemical analysis, environmental monitoring, biological analysis, and other trace detection fields. Surface-enhanced Raman scattering (SERS) is widely used in the analysis of low-concentration molecules with great potential for biological detection due to nondestructive measurement. However, detection uniformity and repeatability are challenges in the application of SERS, especially as regards ultrasensitive detection. Here, a AgNPs@LIG SERS chip with excellent sensitivity and uniformity was fabricated by electroplating Ag nanoparticles (AgNPs) on a laser-induced graphene (LIG) surface prepared on a polyimide (PI) film, which realized large-scale production with controllable shape and area. Such an SERS chip features a limit of detection (LOD) of 10–14 M, an enhancement factor (EF) of 3 × 1010, and fine uniformity (∼9.6% relative standard deviation) for rhodamine 6G, benefiting from excellent local electromagnetic field enhancement generated by the gap of AgNPs and excellent performance of porous LIG. Finally, the chip was applied to the detection of melamine with the LOD of 10–9 M, which confirmed the potential application value of the SERS chip for pollutant analysis. In addition, the preparation of the composite SERS chip is simple for mass production, which provides great potential for the development of ultrasensitive SERS biosensors for various biological and chemical analyses.

Bisphenol A (BPA) is an endocrine disrupting chemical having estrogenic properties that can enter the body through leaking from food containers or the food chain. An upconversion nanoparticle-based aptasensor integrated with microfluidic chip was simply designed to achieve the determination of BPA in foodstuff. Core-shell structured upconversion nanoparticles with 6.41-fold luminescence enhancement were used as fluorescence donors and gold nanoparticles as fluorescence acceptors to construct fluorescence resonance energy transfer (FRET) system. The BPA aptamer serves as a link between the building of the FRET system and the BPA-specific recognition component. The fishbone-structured microfluidic chip only needs 0.5 μL sample solution and 4 min to determine the presence of BPA. The proposed method has good linearity for BPA in the range of 0.02–100 ng/mL with calibration curve of y = 0.0918 x + 0.6477 and limit of detection of 0.014 ng/mL. It is tremendously competitive to perform on-chip analysis that enables microsampling and rapid determination.

Enrofloxacin, a fluoroquinolone widely used in animal husbandry, presents environmental and human health hazards due to its stability and incomplete hydrolysis leading to residue accumulation. To address this concern, a highly sensitive aptamer biosensor utilizing a localized surface plasmon resonance (LSPR) sensing chip and microfluidic technology was developed for rapid enrofloxacin residue detection. AuNPs were prepared by the seed method and the AuNPs-Apt complexes were immobilized on the chip by the sulfhydryl groups modified on the end of the aptamer. The properties and morphologies of the sensing chip and AuNPs-Apt complexes were characterized by Fourier transform infrared spectroscopy (FTIR), UV-Vis spectrophotometer, and scanning electron microscope (SEM), respectively. The sensing chip was able to detect enrofloxacin in the range of 0.01–100 ng/mL with good linearity, and the relationship between the response of the sensing chip and the concentration was Δλ (nm) = 1.288log ConENR (ng/mL) + 5.245 (R2= 0.99), with the limit of detection being 0.001 ng/mL. The anti-interference, repeatability, and selectivity of this sensing chip were studied in detail. Compared with other sensors, this novel aptamer biosensor based on AuNPs-Apt complexes is expected to achieve simple, stable, and economical application in the field of enrofloxacin detection.

Rapid and flexible detection or accurate recognition of trace drugs is of great importance in cracking down on drug crimes, but it remains to be expected. Here, a solid chip is presented for the efficient detection and recognition of trace opioids (typically morphine) in aqueous solutions based on surface-enhanced Raman spectroscopy (SERS). Firstly, a Au-coated Si nano-cone array (Au-SNCA) is designed and fabricated via Si-based organic colloidal template etching and Au deposition. This Au-SNCA shows three-dimensional nanostructure with high densities of nanotips and deep nanogaps as well as high structural consistency, which exhibits strong SERS activity to morphine and outstanding stability. Then, such Au-SNCA is used as solid SERS chip to detect morphine in aqueous solutions. It has been demonstrated that using such solid chip, trace morphine in solutions could be recognized and detected within 1 min, and the detection limit is 10 -5 mg/mL (∼10 ppb), showing rapid and sensitive detection , which is much better than the previous reports. Meanwhile, the Au-SNCA chip also can be utilized to detect trace morphine in tap water and reservoir water, the recoveries range from 90.4% to 102.4%. Such excellent SERS performance of this Au-SNCA chip is attributed to its special structure which enhances not only local electromagnetic field but also molecular adsorption . The experimental results about the effects of immersion time and concentration show that the adsorption behavior of morphine molecules on such Au-SNCA chip can be explained by the pseudo-second-order kinetic model and Freundlich adsorption mode. Moreover, the Au-SNCA chip is also suitable for the identification of morphine homologues and the broad-spectrum detection of various common drugs. This study presents a practical solid chip and a simple approach for the efficient SERS detection and recognition of trace drugs in solutions. This is of significance to on-site detect drugs in forensic science.

Thermoplastic polyimide (TPI) has been extensively applied in electronic industry due to improved processibility, superior mechanical and dielectric properties. However, TPI is still restricted by the limited thermal stability and insufficient UV absorption for the application of temporary bonding and debonding (TBDB) in ultra-thin chip package. In this work, the rigid biphenyl group, twisted non-coplanar structure, rigid bulky side group and flexible ether bond are introduced to obtain a series of novel quaternary copolymerized TPIs with superior comprehensive performance. The results show that the as-prepared TPI possesses excellent solubility in NMP , DMF, DMAc and DMSO , and thermoplasticity ( E ’ drop > 10 3 MPa). Besides, the temperature of 5% weight loss and glass transition temperature could achieve as high as 548.1 °C and 335.4 °C, respectively. Furthermore, the tensile strength of TPI-4 is 128.7 MPa and elongation at break is about 14.2%. Moreover, the TPI-4 also exhibits superior adhesion on silicon wafer (adhesion strength >5 B by hundred grid test) and wonderful absorption at 355 nm UV laser (99.7%). At last, the application in TBDB process has also been proved with a laser energy of 400 mJ/cm 2 within 60 s, showing superior efficiency and low-cost. All these results prove their application in TBDB of ultra-thin chip package for 5G era and so forth.

Surface-enhanced Raman scattering (SERS) with its outstanding performance in ultra-sensitivity has been applied in contaminants monitoring. Herein, a facile strategy of integrating oriented layered MOF nanoarray with controlled electrodeposition of gold nanoparticles via semi-sacrificial template method was proposed to fabricate a versatile self-supporting MOF chip ( [email protected] ) for efficient SERS detection of food contaminants. The S-MOF was ingeniously formed by directly using flexible Ni sheet as both metal source and scaffold, which in favour of greatly improving intrinsic electrical conductivity during electrodeposition process as well as endowing prepared chip with uniform distribution of gold nanoparticles and high stability. Furthermore, amounts of SERS “hot spots” in the oriented layered [email protected] can yield strong electromagnetic field for Raman amplification with superior reproducibility. By virtue of good flexibility and pre-concentration effect of [email protected] , tartrazine (TA), chloramphenicol (CP), imidacloprid (IDP) and crystal violet (CV) in food samples were sensitively detected using three collected approaches (drip coating, soaking and wiping) with low limits of detection (1.0 × 10 −9 M for TA, 8.0 × 10 −11 M for CP, 6.4 × 10 −11 M for IDP and 5.0 × 10 −12 M for CV), wide linear detection ranges (1.0 × 10 −8 – 1.0 × 10 −3 M for TA, 1.0 × 10 −10 – 1.0 × 10 −6 M for CP, 1.0 × 10 −10 – 1.0 × 10 −7 M for IDP and 1.0 × 10 −11 – 1.0 × 10 −7 M for CV) and satisfactory recoveries (96.2 – 125.1%). This method provides a novel strategy to develop an advanced SERS platform for efficient food contaminants monitoring.

Efficient conversion of low reactive CO 2 within a short residence time of flow chemistry presents a challenge. We have tackled this issue by developing 3D microfluidic chips (MFCs) with high-density, small-diameter, and large-volume for CO 2 gas–liquid reactions using femtosecond laser technology. Our innovative design effectively shortens reaction times while satisfying the required temperature and pressure conditions. One of the key features is the integration of the 3D Barker's transform structure, which provides an efficient gas–liquid contact area for mass and heat transfer, even at low Reynolds numbers. As a proof of concept, we synthesize 2,4,5-trifluorobenzoic acid and various other fluorinated carboxylic acids in 3D MFCs using Grignard reagents and CO 2 . Impressively, the Grignard reagents can be generated in situ and utilized at room temperature, and the CO 2 carboxylation reaction with 3D MFCs takes only 40 s with a yield of 97 %. Our findings highlight the feasibility of this multifunctional, efficient, and fast CO 2 flow conversion process, providing a valuable tool to produce value-added chemicals via 3D MFCs techniques.

Pyocyanin is a specific virulence factor and quorum sensing (QS) signal molecule of Pseudomonas aeruginosa , and plays an important role in biofilm formation. In this paper, a biosensor chip was proposed and fabricated for the long term cultivation of Pseudomonas aeruginosa biofilms and in-situ monitoring of pyocyanin. The biosensor chip consisted of two parts, a top polydimethylsiloxane (PDMS) layer with micro-channels and micro-chambers for providing an adequate microenvironment for Pseudomonas aeruginosa biofilm growth, and a bottom microelectrode integrated on a glass substrate for monitoring the QS virulence factor and Pseudomonas aeruginosa biofilms. The concentration of pyocyanin was monitored by AuNPs-modified microelectrodes by differential pulse voltammetry (DPV) on the designed chip. It was demonstrated that the change in pyocyanin concentration was closely related to the developing process of the Pseudomonas aeruginosa biofilms. In addition, the effect of curcumin, a Chinese herb QS inhibitor, on pyocyanin and biofilm production was investigated and analyzed. The results indicate that curcumin inhibited the growth of Pseudomonas aeruginosa biofilms and reduced the production of pyocyanin. The designed biosensor chip provides a promising microenvironment for the quantitative monitoring of pyocyanin in the formation of biofilms and acts as a platform for investigating the mechanism behind anti-bacterial agents.

Rapid development of miniature, portable and highly integrated electronics increases the demand for micro-sized power sources and on-chip energy storage units; micro-supercapacitors with in-plane electrode finger arrays are developed for this purpose. This study fabricates flexible, solid-state, interdigital micro-supercapacitors with MoO 3-x nanorods as cathode fingers and glucose-derived activated carbon nanospheres as anode fingers via a facile and economic screen-printing technique. A highly concentrated sodium alginate bio-hydrogel is used as the non-toxic and biocompatible electrolyte, which provides wider electrochemical stability window (∼2.8 V) than that of conventional aqueous electrolytes (1.23 V). The fabricated asymmetric micro-supercapacitors present outstanding electrochemical performances, for example, high areal capacitance of 47.20 mF cm −2 , superior energy density of 21.20 μWh cm −2 (47.11 mWh cm −3 ) at a power density of 0.18 mW cm −2 (0.40 W cm −3 ), and excellent capacitance retention of 95% after 10,000 cycles. The device also exhibits good mechanical stability and can be integrated into any printed circuits, demonstrating its potential application for highly customized power systems in the Internet of Things and wearable/implantable on-chip electronics with high safety requirement.
