In order to further improve the photocatalytic performance of the SiO2/TiO2composite under visible light irradiation, curcumin-doped SiO2/TiO2nanocomposites were synthesized via directly incorporating it into the structure of SiO2/TiO2during the synthesis using an inexpensive and readily available natural pigment (curcumin) as doping agent. The physicochemical properties of SiO2/TiO2nanocomposites were characterized in detail by X-ray diffraction, transmission electron microscopy, Fourier transform-infrared spectroscopy, N2adsorption–desorption isotherms, X-ray photoelectron spectroscopy, and UV-vis diffuse reflectance spectroscopy. The results indicate that all SiO2/TiO2nanocomposites exhibited an anatase phase with a typical mesoporous structure. It was found that the dope of curcumin in the SiO2/TiO2composite could decrease the crystal size, slightly improve the specific surface areas, significantly enhance the visible light absorption, and effectively narrow the band gap energy from 3.04 to 10(eV). Compared with bare SiO2/TiO2, the curcumin-doped SiO2/TiO2resulted in enhanced photocatalytic reduction activity for Cr(VI) under visible light irradiation, and the CTS (12) sample with the appropriate content of curcumin of 12 wt % shows the photocatalytic yield reaching 100% within 2.5 hours, which is larger than CT (12) without silica. This could be attributed to the curcumin doping and the synergetic effects of SiO2and TiO2in SiO2/TiO2nanocomposites.Keywords:curcumin pigment;SiO2/TiO2nanocomposites;visible photocatalyst;reduction of Cr (VI)

Background A novel nanoemulsion (CU/FU-LN) was developed as an oral 5-fluorouracil and curcumin co-delivery system for synergistic efficacy against liver cancer. Methods MTT assay, confocal laser scanning microscope, and H&E staining were utilized to establish the efficacy and safety of CU/FU-LN.Results The AUC(0-t) of CU/FU-LN was 8.85-fold and 8.59-fold greater than those of CU and FU, respectively. The IC50 of CU/FU-LN was 4.6-fold and 4.9-fold lower than those of FU and CU in HepG2 cells, respectively. In vivo anti-tumor trials, the tumor inhibition rate was significantly elevated by CU/FU-LN (49.29%), compared 24.84% and 4.72% for FU and CU, respectively. Ki-67 immunohistochemical analysis revealed that CU/FU-LN had an obvious anti-proliferation effect. The IC50 of CU/FU-LN in L02 cells was 1.51-fold and 2.60-fold higher than those of CU and FU, respectively. Certain vital organs in the mice of the CU/FU-LN group showed markedly fewer lesions than those of the CU, FU, and CU+FU groups. The CU/FU-LN treatment caused no significant change in mouse body weight relative to the control group (P > 0.05). Conclusions We successfully prepared a promising co-delivery platform for the synergistic treatment of liver cancer and it has a comparatively enhanced efficacy and mitigated toxicity.

The simultaneous therapy of tumor recurrence and bone defects resulting from surgical resection of osteosarcoma is still a challenge in the clinic. Combination therapy based on a localized drug-delivery system shows great promise in the treatment of osteosarcoma. Herein, bifunctional polydopamine (PDA)-modified curcumin (CM)-loaded silk fibroin (SF) composite (SF/CM-PDA) nanofibrous scaffolds, which combined photothermal therapy with chemotherapy to synergistically enhance osteosarcoma therapy, were prepared by PDA coating of the SF/CM nanofibrous scaffolds fabricated by supercritical carbon dioxide (SC-CO2) technology. The PDA coating improved hydrophilicity and mechanical strength of the SF/CM scaffolds. The SF/CM-PDA scaffolds present good photothermal conversion capacity and excellent photostability. The low pH and near-infrared (NIR) irradiation could effectively accelerate release of CM in the SF/CM-PDA scaffolds. The in vitro anticancer results indicated that the biocompatible SF/CM-PDA scaffolds had a long-term, stable, and superior anticancer effect compared to pure CM. Furthermore, the SF/CM-PDA scaffolds significantly increased the growth inhibition of osteosarcoma MG-63 cells under NIR irradiation (808 nm and 1.3 W/cm2). Besides, the SF/CM-PDA scaffolds could enhance osteoblast MC3T3-E1 cell proliferation in vitro when the mass ratio of CM was 0.05–0.5%. This work has therefore demonstrated that the bifunctional SF/CM-PDA scaffolds provide a competitive strategy for local osteosarcoma therapy and bone regeneration.

Photosensitizer (PS)-mediated photodynamic therapy (PDT) has attracted more and more attention as an alternative to traditional antibiotic therapy. Nevertheless, the limitations of traditional photosensitizers seriously hinder their practical application, as a result, the methods to improve the antibacterial properties of traditional photosensitizers have become a hot topic in the field of photomedicine. Herein, a compound nano-PS system has been constructed with synergistic photodynamic and photothermal (PTT) antibacterial effects, triggered by a dual-wavelength illumination. Fluorescent carbon dots (CDs) were synthesized and employed as carriers for the delivery of curcumin (Cur) to obtain CDs/Cur. Upon combined near-infrared and 405 nm visible dual-wavelength irradiation, CDs/Cur could simultaneously generate ROS and a moderate temperature increase, triggering synergistic antibacterial effects against both Gram-positive and Gram-negative bacteria. The results of scanning electron microscopy and fluorescence confocal imaging showed that the combined effect of CDs/Cur with PDT and PTT caused more serious damage to the cell membrane. In addition, CDs/Cur exhibited low cytotoxicity and negligible hemolytic activity, showing great biocompatibility. Therefore, the construction of CDs/Cur by employing CDs as photosensitizer delivery carriers provides a strategy for the improvement of the antibacterial effect of the photosensitizer and the design of next-generation antibacterial agents in photomedicine.

Curcumin is a naturally occurring polyphenolic compound that is commonly used in both medicine and food additives, but its low aqueous solubility and poor bioavailability hinder further clinical applications. For assessing the effect of the glycosylation of curcumin on its aqueous solubility, two glycosyltransferase genes (BsGT1andBsGT2) were cloned from the genome of the strainBacillus subtilisATCC 6633 and over-expressed inEscherichia coli. Then, the two glycosyltransferases were purified, and their glycosylation capacity toward curcumin and its two analogues was verified. The results showed that bothBsGT1andBsGT2could convert curcumin and its two analogues into their glucosidic derivatives. Then, the structures of the derivatives were characterized as curcumin 4′-O-β-D-glucoside and two new curcumin analogue monoglucosides namely, curcumoid-O-α-D-glucoside (2a) and 3-pentadienone-O-α-D-glucoside (3a) by nuclear magnetic resonance (NMR) spectroscopy. Subsequently, the dissolvability of curcumin 4′-O-β-D-glucoside was measured to be 18.78 mg/L, while its aglycone could not be determined. Furthermore, the optimal catalyzing conditions and kinetic parameters ofBsGT1andBsGT2toward curcumin were determined, which showed that the Kcat value ofBsGT1was about 2.6-fold higher than that ofBsGT2, indicating that curcumin is more favored forBsGT2. Our findings effectively apply the enzymatic approach to obtain glucoside derivatives with enhanced solubility. Keywords:curcumin 4′-O-β-D-glucoside;glycosyltransferases;Bacillus subtilisATCC 6633

A redox prodrug consisting of camptothecin-conjugating PEG through the diselenide bond was synthesized, and the resultant amphiphilic prodrug was then used to load the curcumin to form the co-delivery system for the synergetic B16 melanoma cells inhibition. The co-delivery system showed a good response to glutataione, and the prodrug could be degraded and the two loaded drugs were then released. Both camptothecin and curcumin could induce the obvious cytotoxicity to B16 melanoma cells. Particularly, the co-delivery strategy showed a synergetic inhibition effect to B16 melanoma cells through an in vitro assay. In vivo assay also showed that the co-delivery system could inhibit B16 tumor growth and showed much better inhibition effect than that of only camptothecin or curcumin used. Moreover, as an injectable drug delivery system, the biocompatibility including the blood compatibility of the co-delivery system was confirmed, suggesting that the redox co-delivery system have a potential application in B16 tumor therapy.

The aim of this work is to develop curcumin-loaded hollow mesoporous silica microspheres (HMSMs@curcumin) to improve the poor oral bioavailability of curcumin. Hollow mesoporous silica microspheres (HMSMs) were synthesized in facile route using a hard template. HMSMs and HMSMs@curcumin were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), nitrogen adsorption/desorption measurements, differential scanning calorimetry (DSC), Fourier transform infrared (FTIR), and X-ray diffraction (XRD). In addition, to demonstrate the potential application of the HMSMs@curcumin, cytotoxicity, in vitro release behavior and in vivo pharmacokinetics of curcumin loaded in these HMSMs were investigated by using of Caco-2 cells and Sprague-Dawley (SD) rats, respectively. These mono-dispersed HMSMs exhibited high drug loading ratio and encapsulation efficiency due to the mesoporous shell and hollow core. The excellent characteristics of HMSMs such as mono-dispersed morphology, smooth surface, uniform, ordered and size-narrowing mesopores resulted in a good in vitro release profile of curcumin from HMSMs@curcumin. Moreover, an impressive improvement in the oral absorption of curcumin and prolonged systemic circulation time were achieved in the in vivo animal studies. In addition, the good biocompatibility of developed HMSMs with Caco-2 cells was confirmed based on the in vitro cytotoxicity assay. In conclusion, this system demonstrated a great potential for efficient delivery of curcumin in vitro and in vivo, suggesting a good prospect for its application in clinic for therapeutic drug delivery in future.

Curcumin (Cur) and its nanomedicine were researched in Caco-2 cells to investigate the internalization and subcellular transport across Caco-2 cell monolayer membrane. Cur loaded polymer micelle (PM), nanoemulsion (NE) and liposome (Lipo) were prepared to explore the transport process incorporating endocytosis, intracellular trafficking and transcytosis in Caco-2 cells. Firstly, nanocarriers could contribute to the cellular uptake and improve the stability of Cur within the Caco-2 cells. The co-mediation of clathrin , lipid raft/caveolae and/or macropinocytosis were involved in the internalization of Cur nanopreparations, indicating the diversity and partial energy-dependency of the endocytosis. Secondly, the endocytosed nanocarriers with Cur successively transported from apical early endosome to late endosome with the extension of incubation time . Furthermore, the intracellular location of Cur nanomedicines in lysosome and endoplasmic reticulum (ER) were detected and illustrated the actual transport of Cur nanomedicine to lysosome and ER. Finally, nanocarriers could enhance the transcytosis of Cur, involving the energy-dependent patterns. Especially, Cur-PM exhibited more advantages in regulating and coordinating the entry and the exit of Cur. The study indicated that the coexistence of multiple transport pathways had the potency to promote nanocarriers transcytosis and provided a rational dosage design for oral Cur.

In this study, hyaluronic acid (HA) and curcumin (CCM) are used as functional molecular skeletons to prepare a reticular polymer HA-CCM* via the free radical polymerization reaction. The results showed that HA-CCM* formed nanomicelles through self-assembly in the aqueous medium. These nanomicelles were seen as star-shaped micelles and the particle diameter distribution range of 220–280 nm, which are used to carry doxorubicin (DOX) with a high drug loading and an entrapment efficiency. The experiment of drug release in vitro indicated that HA-CCM* micelles showed significant acid sensitivity. The anticancer activity of DOX-loaded by HA-CCM* micelles (HA-CCM* + DOX) was quantified by MTT assay , indicating that the cytotoxicity of HA-CCM* + DOX to human cervical cancer cell line (HeLa) and human renal cell adenocarcinoma cell line (786-O) was significantly improved and to normal human renal epithelial cell line (293A) was obviously reduced. DOX from HA-CCM* + DOX micelles can enter into the cells and resulted in cell death. The multifunctional HA-CCM* micelles have good prospects in the field of drug delivery. Overall, these results confirmed the potential of these nanomicelles for carrying DOX in the improved cancer chemotherapy.

The development of high drug loading and sustained release nanoparticles by simple and economical methods is significant for the treatment of diseases. Amorphous calcium carbonate (ACC) is an ideal drug delivery system because of its excellent pH responsiveness, biocompatibility and biodegradability. In this work, ACC nanoparticles were successfully precipitated in ethanol–calcium solutions using ammonium carbonate as the internal source of CO 2 , which exhibited a relatively narrow size distribution in range of 10–200 nm. The microscopic morphology, amorphous characters, porous structure and thermal behavior of resultants were investigated by electron microscopy, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller and thermogravimetric analysis, respectively. In vitro drug release assay showed that ACC nanoparticles had high loading capacity of curcumin (Cur) and favorable drug release properties. Furthermore, ACC-Cur showed excellent abilities to scavenge free radicals, protect Cur stability and damage A549 cells, providing a broadened space for the applications of this material in the fields of biomedical or food.
