Accelerating effects of biochar for pyrite-catalyzed Fenton-like oxidation of herbicide 2,4-D

Xiaoxiao Zhu,Jianfa Li,Bin Xie,Dongqing Feng,Yimin Li

Biochar was mixed with natural pyrite to catalyze the Fenton-like oxidation of herbicide 2,4-dichlorophenoxyacetic acid (2,4-D), so as to accelerate the degradation reaction. The accelerating effects of two biochars with different adsorption capacities were compared. The biochar’s functions on accelerating the oxidation were investigated by using radical scavengers, electron spin resonance (ESR), and the chemical probe method. The results indicated that the rate constant ( k obs ) for the degradation reaction of 2,4-D was enhanced 1.98–2.39 times after the addition of 0.1 g/L of biochar in the pyrite (1.5 g/L) catalyzed system. However, the biochar with the higher adsorption capacity did not outperform the other biochar in accelerating the reaction. Biochar did not influence the Fe lixiviation from pyrite; instead, it accelerated the Fenton oxidation of 2,4-D catalyzed by the dissolved Fe 2+ . Hydroxyl radicals ( OH) were proved to be the major reactive species contributing to the oxidation of 2,4-D, and the addition of biochar increased the cumulative production of OH 2.72 times that in the pyrite-catalyzed system. The mechanism is related to the transformation of O 2 − to OH by biochar, in which the quinone-like structure acts as the electron shuttle. Based on the identified intermediates and products by gas chromatography-mass spectrometry (GC–MS), high performance liquid chromatography (HPLC), and ion chromatography (IC), as well as the released Cl − , a reaction pathway was summarized for mineralization of 2,4-D. Furthermore, the degradation of 2,4-dichlorophenol (2,4-DCP) was accelerated by biochar as well, indicating the wide feasibility of biochar for accelerating the Fenton-like oxidation of organic pollutants.

6′'-O-acetylsaikosaponin D targets STAT3-mediated transcriptional remodeling to induce ferroptosis and apoptosis

Xuehui Li,Yunxiao Qiao,Xinyi Xu,Jinyu Zhu,Chuang Xie,Shan Lin,Lingfan Xu,Fuwen Yuan

Background Natural products from traditional medicinal plants represent an important source of anti-cancer agents and lead compounds for drug development. Saikosaponin D (SSD), a triterpenoid saponin isolated from Radix Bupleuri, exhibits anti-tumor activity but is limited by a narrow therapeutic window and cytotoxicity. Objective This study aimed to evaluate the anti-tumor activity of 6′'-O-acetylsaikosaponin D (6′'-O-acSSD), a naturally occurring acetylated derivative of Saikosaponin D that is identified in Radix Bupleuri , and to elucidate its underlying molecular mechanisms. Methods The anti-cancer effects of 6′'-O-acSSD were assessed using multiple cancer cell models, three-dimensional tumor spheroids, and patient-derived tumor organoids. Transcriptomic and bioinformatic analyses were performed to identify regulated cell death pathways. Molecular docking, molecular dynamics simulation, cellular thermal shift assays, western blotting, and CUT&Tag profiling were used to identify molecular targets and define transcriptional reprogramming. Results 6′'-O-acSSD markedly suppressed cancer cell proliferation, migration, and invasion and elicited a transcriptional program distinct from that induced by SSD. Transcriptomic and biochemical analyses revealed preferential activation of ferroptosis-associated signatures, including enhanced lipid peroxidation. Mechanistically, 6′'-O-acSSD engaged STAT3, inhibited its phosphorylation, and extensively remodeled STAT3 chromatin occupancy. Functional analyses showed that STAT3 inhibition was required for the full anti-tumor activity of 6′'-O-acSSD. Notably, blockade of ferroptosis only partially rescued cell viability, whereas STAT3 targeting additionally triggered apoptosis-associated transcriptional and biochemical changes. The anti-tumor effects of 6′'-O-acSSD were further validated in patient-derived tumor organoids. Conclusion 6′'-O-acSSD exerts potent anti-tumor effects by targeting STAT3-dependent transcriptional programs and inducing coordinated ferroptosis and apoptosis. These findings highlight the therapeutic potential of structurally optimized natural products for anti-cancer treatment.

Rewiring Pyroptosis to Potentiate Cancer Immunotherapy via a Gasdermin D Agonist Bypassing Caspase‐3

Dan Zhao,Lingling Zhang,Yue Wen,Lianghui Cheng,Sidan Tian,Fanling Meng,Liang Luo

Pyroptosis holds great promise for evoking robust anti‐tumor immunity, but many pyroptosis induction strategies rely on caspase‐3 (Casp‐3) activation and face challenges of the immunosuppressive nature of apoptosis and the frequent silencing of gasdermin E (GSDME), a substrate of Casp‐3, in tumors. Here, we report a strategy to rewire the pyroptotic pathway by bypassing Casp‐3 to directly activate gasdermin D (GSDMD). We identify a small‐molecule agonist, (E)‐2,3‐diiodobut‐2‐ene‐1,4‐diol (DIBDO), which undergoes deiodination to release iodide ions upon activation by the tumor‐abundant nucleophile glutathione, thereby catalytically generating singlet oxygen and molecular iodine. This cascade induces oxidative damage that downregulates key mediators of caspase‐9 and caspase‐8 pathways, thereby down‐regulating Casp‐3 activation. This mechanism selectively triggers GSDMD‐mediated pyroptosis, provoking robust immunogenic cell death and inflammatory cytokine release to reconfigure the tumor immune microenvironment. In murine tumor models, DIBDO enhances tumor infiltration of cytotoxic T cells and synergizes with checkpoint blockade therapy to suppress both primary and distal tumors. It can also serve as an in situ or exogenous vaccine to elicit potent and durable antitumor immunity. This work presents a paradigm‐shifting approach to cancer immunotherapy by decoupling pyroptosis from Casp‐3 dependence, offering a promising avenue to expand the scope of immunogenic cell death‐based treatments. This work introduces a small‐molecule agonist, DIBDO, that bypasses Casp‐3 to directly trigger GSDMD‐mediated pyroptosis in tumors. Activated by glutathione, DIBDO generates singlet oxygen and molecular iodine, blocking Casp‐3 pathways and inducing immunogenic cell death. This reconfigures the tumor microenvironment, transforms the tumor cells into in situ or exogenous vaccine, offering a novel paradigm for cancer immunotherapy.

Multi-signal gate-array OFET platform for reliable D-Dimer quantification in complex biofluids

Jing Zhang,Qi-Ting Wang,Tian-Tian Song,Bin Liu,Shan-Shan Cheng

Real-time, high-sensitivity biosensors are essential for personalized healthcare, enabling early disease diagnosis and continuous monitoring. Organic field-effect transistor (OFET) sensors offer label-free detection, rapid response, and low cost, but noise interference and inherent architectural limitations hinder their use in high-throughput, multi-signal analysis. To address these issues, this study presents a gate-array-based OFET biosensor that improves detection sensitivity, stability, and analytical throughput. The integrated gate-array architecture and signal modulation strategies enable rapid acquisition of multiple transfer curves for efficient concentration analysis of unknown samples. To validate the approach, the sensor detects the thrombotic biomarker D-Dimer with an ultra-low limit of 0.354 ng/mL in PBS, well below clinical thresholds, and performs reliably in human serum and patient samples. This work introduces a robust platform for high-throughput, multi-target detection in complex biological environments, advancing next-generation biomedical diagnostics and personalized healthcare.

Involvement of trimethylamine N-oxide in major depressive disorder via astrocytic d-Serine dysregulation

Liwang Lin,Longyu Li,Shiao Ren,Liping Liu,Ying Liu,Wenlei Zhang,Zengliang Gao,Xiaoting Ni,Haoxuan Li,Xinsheng Duan,Sijing Tao,Tianyu Zhao,Xin Hai

Trimethylamine N-oxide (TMAO), a co-metabolite of the gut microbiota and host, has been implicated in the pathogenesis of various neuropsychiatric disorders. However, its role in major depressive disorder (MDD) remains poorly understood. This study aims to verify the relationship between TMAO and MDD and elucidate the underlying mechanisms. Plasma TMAO concentrations were quantified by HPLC-MS/MS and compared between MDD patients and healthy controls. To explore the effects of TMAO on depressive-like behavior and cerebral d -serine metabolism, mice were administered TMAO via dietary supplementation. Finally, astrocytes were treated with TMAO to examine its impact on d -serine metabolism at the cellular level. Here, we found that plasma TMAO levels were significantly elevated in MDD patients and positively correlated with both HAMD-17 and HAMA-14 scores. Consistently, mice fed TMAO for seven weeks exhibited pronounced depressive-like behaviors. While TMAO exposure did not induce apparent astrocytic damage, it markedly promoted d -serine secretion and caused notable neuronal injury. Mechanistically, TMAO activated the AMPK/SIRT-1 signaling pathway in astrocytes, resulting in upregulated serine racemase expression. Furthermore, both exogenous d -serine and conditioned medium from TMAO-treated astrocytes triggered neuronal apoptosis. Collectively, these findings demonstrate that TMAO contributes to MDD pathogenesis by activating astrocytic AMPK/SIRT-1 signaling to enhance d -serine production, subsequently inducing neuronal apoptosis. TMAO may thus represent a promising therapeutic target for MDD.

High-Entropy Metal Sulfides with Enhanced d-Orbital Coupling for Boosted Sodium Storage Performance

Shengqin Guan,Xinyu Liu,Xingbo Yu,Jianlong Wang,Baoen Xu,Kaixi Li,Taotao Guan

Abstract The advanced high-entropy configuration approach is employed to stabilize the structural integrity of transition metal sulfides during cycling by suppressing volume expansion. This configuration simultaneously preserves abundant active sites and maintains high electronic conductivity. However, the sodium storage mechanism in high-entropy materials remains inadequately understood. In this study, a solvothermal method is employed to synthesize a unique class of (FeCoNiCuW)S2 high-entropy metal sulfides (HEMSs), which exhibit distinct d-orbital coupling characteristics. Density functional theory (DFT) calculations reveal that stronger d-orbital coupling among the metals leads to higher electron density near the Fermi energy (EF), resulting in an upward shift of the entire d-band center relative to EF, thus enhancing the adsorption of the Na2S intermediate. As a result, (FeCoNiCuW)S2 demonstrates superior rate capability, achieving a specific capacity of 518.6 mA·h·g–1 at 5.0 A·g–1. Moreover, ex situ characterization confirms a reversible sodium storage mechanism, further demonstrating that the high-entropy effect stabilizes the conversion reaction reversibility upon prolonged cycling. This work highlights structurally stable HEMSs as promising anode materials for sodium-ion batteries.

Ultra-sensitive optical waveguide sensor with D–π–A chromophore for fluoride ion detection

Jinkai Wang,Yingzhe Wang,Yingchao Zhang,Xiangzhuo Han,Zhanchen Cui,Zuosen Shi

We present a polymer optical waveguide sensor for fluoride ion (F − ) detection utilizing D–π–A dipolar molecules functionalized with tert-butyl dimethyl siloxane (TBS) groups. The sensing mechanism relies on F − -induced cleavage of Si O bonds through a desilylation reaction, which triggers a corresponding fluorescence quenching at 607 nm. By employing a 532 nm excitation laser and exploiting evanescent field interactions at waveguide interfaces, we developed an optical sensing platform that enables both qualitative identification and quantitative measurement of trace F − concentrations through real-time fluorescence monitoring. The sensor demonstrates exceptional performance with an ultra-low detection limit of 2.25 ppb and rapid response characteristics (≈3 s). The device architecture permits configuration into multiple parallel detection channels, making it particularly promising for high-efficiency environmental monitoring and biomedical applications requiring simultaneous multi-sample analysis. Furthermore, the sensor exhibits excellent anti-interference capability and specific detection performance, demonstrating reliable detection characteristics both in the presence of other interfering substances and in practical sample applications. The combination of superior sensitivity, rapid response, and scalable design positions this platform as a significant advancement in optical chemical sensing technology

Synthesis and luminescent properties of the bipyridine derivative with a D-A-A structure

Hanfei Xiang,Yonghe Zhang,Zhefeng Wang,Jie Sun,Zhengcheng Zhang,Zengrui Xing,Peng Zhang,Zhenghua Ju

A donor-acceptor1-acceptor2 (D-A1-A2) pyridine derivative (P263) was designed and synthesized. The theoretical calculations showed that the mixed character of the locally excited (LE) and intramolecular charge transfer (ICT) in the T n states could be beneficial to achieve high spin-orbit coupling (SOC) values, thereby promoting efficient intersystem crossing (ISC). The fluorescence emission spectra of P263 exhibited an obvious solvent-dependent emission behavior, which was attributed to the effective ICT through the introduction of dual acceptor units. In addition, the protonation/deprotonation processes of the nitrogen atoms in the pyridine units induced the alterations in the emission spectra, rendering P263 a fluorescent sensor for proton ( H + ) detection. Moreover, the pyridine units could form the hydrogen-bonding interactions with the poly(vinyl alcohol) (PVA) matrix, which was favorable for the suppression of the nonradiative decays and the stabilization of the triplet excitons. The obtained P263@PVA film through doping P263 into PVA showed room-temperature phosphorescence (RTP) properties with a long lifetime of 535 ms and the quantum yields of 3.62 %.

Chalcogenapyrylium-engineered D-π-A probe for enhanced optoacoustic imaging of amyloid-β plaques

Jian Li,Tingting Wang,Xun Zhang,Xinhui Su,Nian Liu

High-resolution optoacoustic visualization of amyloid-β (Aβ) plaques facilitates assessment of Alzheimer's disease (AD)’s states and related pathological mechanisms. However, it still lacks effective Aβ-probes due to suboptimal optoacoustic properties. In this study, we proposed the use of N, N-dimethylaniline as the Aβ recognition unit and chalcogenapyrylium as the acceptor to construct a novel D-π-A typed probe (DMA-Te). This design not only enhances the near-infrared optoacoustic signal but also maintains this enhancement upon Aβ fibrils binding through the preservation of twisted intramolecular charge transfer state. Furthermore, the stringent structure of DMA-Te exhibits high blood-brain barrier permeability and Aβ-binding capacity. Leveraging these advantages, dual-wavelength optoacoustic microscopy can effectively visualize Aβ plaques and diminished blood vessels in the AD brain. Therefore, this study presents a promising optoacoustic probe for the detailed depiction of AD progression.

Yanan Wang,Teng Wang,Boran Li,Yitong Lin,Qi Zhang,Fei Chang,Yutong Han,Wenxuan Li,Hao Zhang,Xiaoyun He,Huilian Che,Kunlun Huang,Nan Cheng

Pd@Pt nanozymes are recognized for their extensive specific surface area and enzyme-like catalytic capabilities. However, the role of Pd@Pt nanoparticles in managing metabolic disorders such as diabetes and its related disorders remains poorly understood. This study endeavors to explore the regulatory function of Pd@Pt nanoparticles in metabolic balance, elucidating the associated mechanisms. We demonstrate that Pd@Pt inhibits the production of reactive oxygen species and modulates oxidative stress through its superoxide-dismutase-like enzyme activity in palmitic-acid-induced insulin resistance cell model. Furthermore, Pd@Pt was found to enhance glucose uptake, reduce hyperglycemia, and improve glucose tolerance and insulin sensitivity, attributable to its anti-inflammatory effects. Additionally, the effectiveness of the multifunctional Pd@Pt-based microneedle in expediting the healing of diabetic wounds was confirmed by suppressing key inflammatory signaling pathways. Collectively, our findings highlight the potential of Pd@Pt nanoparticles as powerful anti-inflammatory agents, offering promising therapeutic options for type 2 diabetes and its associated complications.

Diverse binding of important anions in 1-D tricopper anion coordination polymer (ACP) architectures

Yu-Peng Zhou,Zhang-Wen Wei,Zhuo-Jia Lin,Hui-Tao Ling,Zhengang Guo,Mei Zhang,Chi-Keung Lam,Bao-Hui Ye,Hsiu-Yi Chao

With the idea of supramolecular hosts encircling monoanions, a series of urea-containing trinuclear cationic copper complexes were designed, prepared and structurally analyzed. Copper(I) acetylides [Cu3(μ-dppm)3(μ3-η1-CCC6H4-4-NHC(O)NHC6H4-4-R)2]+ (dppm = bis(diphenylphosphino)methane; R = NO2 (1), CF3 (2), Cl (3), H (4), tBu (5), and OCH3 (6)) were prepared with F−, BF4−, or NO3− present as the anion. Eight 1-D anion coordination polymers (ACPs), 1·F, 2·F, 3·BF4, 4·F, 5·F, 5·BF4, 5·NO3 and 6·0.5F·0.5BF4, were clearly determined by X-ray diffraction. Diversity in the polymeric architecture is generated by both the choice of R-group used and anion present. 2·F, 3·BF4, 4·F, and 5·F show linear 1-D structures; 5·BF4 and 5·NO3 feature single-stranded helical structures; 1·F and 6·0.5F·0.5BF4 exhibit meso-helical structures. Solid state UV-vis absorption and luminescence properties are also discussed.

Development of Saikosaponin D Liposome Nanocarrier with Increased Hepatoprotective Effect Against Alcoholic Hepatitis Mice

Yu Xiao,Pan Jielu,Shen Nan,Zhang Haiyan,Zou Lu,Miao Hongyu,Xing Lianjun

The mortality rate of ethanol induced liver disease has substantially raised to alert level with an increasing use of alcohol, but development of definite hepatoprotective drug is still challenging. The efficacy of Saikosaponin D, one of the natural herbal medicine has been studied in different diseases. Nonetheless, its clinical application is restricted by poor bioavailability, stability and solubility. This study sought to develop a Saikosaponin D loaded liposome via thin film hydration method. The surface morphology, encapsulation efficiency and drug loading capacity were detected with transmission electron microscopy and HPLC, in vitro dissolution was via dialysis method, but efficacy and safety evaluation was through pharmacokinetics, while the assessment of hepatoprotective activity on alcohol induced acute hepatitis mice models was conducted. The optimized liposomes showed significant greater therapeutic effect on liver, through decreased serum levels of alanine transaminase (ALT) and aspartate transaminase (AST), malondialdehyde (MDA), tumor necrosis factor alpha (TNF-α), total cholesterol (TC) and triglyceride (TG) in liver homogenate. In contrast, levels of glutathione peroxidase (GSH-Px) and total superoxide dismutase (T-SOD) were increased significantly. Pathological study exhibited remarkable alteration of hepatitis liver architecture to almost normal state after administration of Saikosaponin D liposome. The increased hepatoprotective effect of Saikosaponin D liposome was observed during the attenuation of alcoholic hepatitis in mice, which might be ascribable to the anti-oxidative and anti-inflammatory properties of the drug. This study provides a theoretical basis for developing advanced system of Saikosaponin D delivery for the promotion of the therapeutic effects of the liposome against various kinds of diseases.
