Femtosecond laser synthesis and processing of colloids offer a platform for novel nanomaterial synthesis and applications, advancing side by side with high-end material innovations. In this work, RE2(Ce0.2Zr0.2Hf0.2Sn0.2Ti0.2)2O7 (RE = Sm, Eu, Er), one kind of A2B2O7-type (A = rare earth cation, B = tetravalent metallic cation) high-entropy oxide ceramic is chosen as the ablation target for colloid synthesis by femtosecond laser ablation in ethanol. Both surface structural and colloidal morphologies are analyzed to unveil the effects of photothermal (melting) and photomechanical (cracking) phenomena on colloid synthesis. High-resolution transmission electron microscopy (HRTEM) and selected area electron diffraction (SAED) jointly show that the synthesized nanomaterials in different sizes and shapes are mainly in the amorphous state, irrespective of the pristine samples’ composition. Energy-dispersive X-ray spectroscopy (EDS) mapping of as-prepared colloids indicates the emergence of elemental segregation, mainly occurring to small particles (mainly <10 nm); whereas for large particles, multiple elements are homogeneous, reflecting the high entropy characteristic of the samples. The discovery of a tailed nanostructure with elongated void embedment indicates special spallation dynamics containing void freezing or bubble capturing. Our findings may help to better understand the ablation dynamics relative to ceramics’ colloid formation. RE2(Ce0.2Zr0.2Hf0.2Sn0.2Ti0.2)2O7 (RE = Sm, Eu, Er) are most frequently considered to be good candidates for next-generation thermal barrier coatings in the film form; downsizing the ceramic dimensions into nanoscale via laser synthesis in liquids may expand its widespread applications in various fields such as optics, photonics, environment, catalysis, and biomedicine.

Betulinic acid (BA), a naturally occurring lupane-type triterpenoid, possesses a wide range of potential activities against different types of cancer. However, the molecular mechanisms involved in anti-cervical cancer about BA were rarely investigated. Herein, the role of BA in cervical cancer suppression by ROS-mediated endoplasmic reticulum stress (ERS) and autophagy was deeply discussed. The findings revealed that BA activated Keap1/Nrf2 pathway and triggered mitochondria-dependent apoptosis due to ROS production. Furthermore, BA increased the intracellular Ca 2+ levels, inhibited the expression of Beclin1 and promoted the expression of GRP78, LC3-II, and p62 associated with ERS and autophagy. Besides, BA initiated the formation of autophagosomes and inhibited autophagic flux by the co-administration of BA with 3-methyladenine (3-MA) and chloroquine (CQ), respectively. The in vivo experiment manifested that hydroxychloroquine (HCQ) enhanced the apoptosis induced by BA. For the first time, we demonstrated that BA could initiate early autophagy, inhibit autophagy flux, and induce protective autophagy in HeLa cells. Thus, BA could be a potential chemotherapy drug for cervical cancer, and inhibition of autophagy could enhance the anti-tumor effect of BA. However, the interactions of signaling factors between ERS-mediated and autophagy-mediated apoptosis deserve further attention. Graphical abstract

Diabetic retinopathy (DR) is a neurovascular complication of diabetes. As a crucial player in the retinal physiology, Müller cells are affected in DR, impairments of Müller cell function lead to retinal malfunctions. Therefore, searching for approaches to mitigate diabetes-induced injury in Müller cells is imperative for delaying DR. Peroxiredoxin 4 (PRDX4), an important endoplasmic reticulum (ER)-resident antioxidant, was explored in this study for its potential protective role against DR. Streptozotocin (STZ)-induced mouse model of diabetes and high glucose (HG)-induced Müller cells were utilized to assess the impact of PRDX4. Compared to wild-type mice, PRDX4 knockout exacerbated retinal neurodegeneration, reactive gliosis, cell apoptosis, endoplasmic reticulum (ER) stress, oxidative stress, and mitochondrial dysfunction in diabetic retinas. Knockdown of PRDX4 aggravated high glucose (HG)-induced reactive gliosis, apoptosis, ER stress, oxidative stress, and mitochondrial dysfunction in Müller cells. Conversely, PRDX4 overexpression in Müller cells protected against HG-induced cell damage. Mechanistically, PRDX4 promoted the degradation of DPP4, which is associated with DR in type 1 diabetics, thereby alleviating HG-stimulated Müller cell abnormalities. Our study indicated that PRDX4 is a crucial protective regulator in DR progression via destabilization of DPP4 protein and suggested enhancement of PRDX4 level may represent a promising approach for treating DR.

Considering the vital role of sulfide ions (S 2- ) in food safety, environmental pollution and public health, accurate and efficient detection of S 2- is of great importance. In this study, Er-MOFs with the structure of four-pointed star was successfully designed and synthesized using Er(III) as the metal center and TCPP as the ligand. It is found that host-guest effects occur between Er-MOFs and S 2- , blocking the ligand-to-metal charge transfer (LMCT) effect and O-H vibration. This progress leads to the fluorescence signal-on of Er-MOFs and the production of a brown color. In view of this, a Er-MOFs-based sensor was constructed for rapid and on-site monitoring of S 2- in water, which shows the triple-signal outputs including fluorescence, absorbance and smartphone-based RGB analysis with high sensitivity and selectivity. More importantly, the successful measurement of S 2- in real water samples with satisfactory recoveries from 96.26 % to 104.03 %, demonstrating the practicality and effectiveness of Er-MOFs for S 2- recognition. Overall, this assay exhibits the promising applications for S 2- measurement in the fields of environmental pollution and food safety.

The homogeneously transparent hybrid gels (Ln–N–P-gel, Ln = Sm, Yb, Nd, Er, Pr, Ho, Tm) containing lanthanide complexes (by using dinaphthoylmethane (dnm) as the primary ligand and 1,10-phenanthroline (phen) as the synergic ligand) via the in situ approach have been prepared. The dnm ligand was synthesized and adopted with the aim to sensitize the lanthanide ions with visible light. The properties of Ln–N–P-gels were investigated by Fourier-transform infrared (FT-IR), diffuse reflectance (DR), scanning electron micrographs (SEM), fluorescence spectra and thermogravimetric analyses. The results reveal that the lanthanide complexes are successfully in situ synthesized in the corresponding Ln–N–P-gels. Under the excitation of visible-light, the Ln–N–P-gels showed the respective visible (Sm3+) and near-infrared (Sm3+, Yb3+, Nd3+, Er3+, Pr3+, Ho3+, Tm3+) luminescence. The near-infrared (NIR) luminescence of Sm3+, Pr3+, Ho3+, and Tm3+ complexes sensitized by visible-light had been rarely reported previously, which is highly desired for biomedical and photonics applications. Based on the relevant energy levels, a model was proposed to explain the visible-NIR luminescence of the Ln–N–P-gels based on the visible-light excitation.

Tunable photoluminescence (PL) has wide applications in optical waveguides and communication. Since the PL intensity is strongly dependent on the crystal field symmetry, curvature-dependent strain-induced PL modulation is expected in flexible antiferroelectric (AFE) thin film materials. Here, flexible Er-doped Pb 0.98 La 0.02 Zr 0.95 Ti 0.05 O 3 (PLZT) AFE thin films were prepared via the sol-gel method. A giant and reversible PL intensity modulation (482%) was achieved during the bending process. Based on density functional theory (DFT) calculations and Raman spectra measurements, the relationship amongst the curvature-dependent strains, crystal structure, and PL modulation was discussed. The induced strain leads to lattice distortion and even a phase transition, both of which contribute to the reduction in crystal symmetry and thus significantly enhance the PL intensity.

Background Anterior gradient-2 (AGR2) is a proto-oncogene involved in tumorigenesis and cancer progression. AGR2, predominantly localized in the endoplasmic reticulum (ER), is also a secreted protein detected in the extracellular compartment in multiple cancers. However, the biological functions of intracellular and extracellular AGR2 remain to be elucidated. Methods Based on the biochemical structure of AGR2 protein, PANC-1 pancreatic cancer cells stably expressing ER-resident or secreted AGR2 were generated by a lentivirus-mediated stable overexpression system. The capacities of cell proliferation, migration, invasion and survival were assessed in PANC-1 stable cells. Moreover, EGFR expression and activation were determined to explore the possible mechanism of AGR2 roles in pancreatic cancer tumorigenesis. Results It was discovered that secreted AGR2, but not ER-resident AGR2, promotes cell proliferation, migration and invasion of PANC-1 cells. Moreover, the data indicated that both the ER-resident and the secreted AGR2 enhance the survival capacity of PANC-1 cells after tunicamycin-induced ER stress and gemcitabine treatment. However, EGFR expression and activation were not found to be involved in AGR2-dependent oncogenic phenotypes in PANC-1 cells. Conclusions Secreted AGR2 is predominantly involved in cell proliferation, migration and invasion in PANC-1 pancreatic cancer cells. Both secreted and ER-resident AGR2 contribute to the survival of PANC-1 cells under the challenging conditions. These findings provide insight into how different localizations of AGR2 have contributed to pancreatic cancer growth, metastasis, and drug sensitivity.

RNA splicing is fundamental to cellular function, yet systematic investigation of its complex regulation has been limited by existing methods. Here, we present SPLiCR-seq ( SPL icing regulator i dentification through CR ISPR screening), a high-throughput CRISPR screening platform that enables direct measurement of RNA splicing outcomes for pooled genetic perturbations, overcoming limitations of traditional fluorescence-based approaches. Applying SPLiCR-seq to investigate XBP1 splicing during the unfolded protein response (UPR), we conduct targeted and genome-wide screens across diverse cellular contexts, revealing both common and cell-type specific regulators. Notably, we identify GADD34 ( PPP1R15A ) as a novel modulator of IRE1α-XBP1 signaling, demonstrating that it directly interacts with IRE1α and functions independently of its canonical role in eIF2α dephosphorylation. Pharmacological inhibition of GADD34 using Sephin1 effectively suppressed XBP1 splicing and alleviated CAR-T cell exhaustion in an ex vivo model, leading to enhanced tumor-killing capacity across multiple cancer models. This work not only establishes a powerful new tool for systematically studying RNA splicing regulation but also uncovers a promising therapeutic strategy for improving CAR-T cell immunotherapy through modulation of the IRE1α-XBP1 pathway. Systematic investigation of RNA splicing regulation remains challenging. Here, authors present SPLiCR-seq, a CRISPR platform enabling direct measurement of splicing outcomes for pooled perturbations. They identify GADD34 as a novel IRE1α-XBP1 regulator, with inhibition enhancing CAR-T cell activity.

ER-Fc-MOF@CF monolith integrates PE capture and Fenton-like oxidation. • Carbonization tunes conductivity, defects, and MOF anchoring on CF. • CF-900 achieves 91.14% PE removal under flow-through conditions. • Captured PE undergoes localized ROS-mediated surface oxidation. • Monolithic design reduces powder catalyst loss and secondary release. Microplastic remediation requires efficient particle capture and interfacial oxidation because dispersed polymers have limited contact with short-lived reactive oxygen species (ROS) in bulk solution. Here, an electroresponsive ferrocene metal–organic framework (ER-Fc-MOF) was grown in situ on thermally engineered foam carbon (CF) to construct ER-Fc-MOF@CF monoliths for polyethylene microplastic capture and subsequent electro-Fenton-like treatment. Carbonization temperature regulated the scaffold architecture, surface chemistry, defect structure, electrical resistivity, and ER-Fc-MOF coverage. Among the investigated samples, CF-900 retained accessible hierarchical porosity while exhibiting the lowest resistivity and densest ER-Fc-MOF coverage. The optimized ER-Fc-MOF@CF-900 achieved 91.14% flow-through removal from a 100 mg·L −1 polyethylene (PE) suspension. Zeta-potential, FTIR, and XPS analyses suggested pH-dependent electrostatic attraction and localized polar interactions involving oxygen-containing groups, while the interconnected monolith facilitated particle interception and retention. The low-resistance CF-900 scaffold supported electron transport and H 2 O 2 activation at ER-Fc-MOF domains. SEM, EDS, FTIR, and XPS showed progressive PE surface oxidation, including roughening, oxygen enrichment, and increased C O, C O, and O–C=O signals, while EPR identified ·OH, ·O 2 − , and 1 O 2 . After 36 h, dissolved organic carbon (DOC) reached 67.21 mg·L −1 in the PE-containing system versus 8.80 mg·L −1 in the PE-free control, supporting PE-associated dissolved organic carbon release. This work establishes a self-supporting MOF–carbon platform coupling PE capture, flow-through interception, and interfacial oxidative transformation. Diabetic wounds are characterized by persistent inflammation, impaired angiogenesis, and delayed tissue regeneration, yet effective strategies to regulate the pathological immune microenvironment remain limited. Here, integrated single-cell transcriptomics and multi-omics analyses revealed significant enrichment of endoplasmic reticulum stress (ERS)-related signaling in pro-inflammatory macrophages within diabetic wounds, suggesting a potential mechanism driving chronic inflammation. To target this process, we developed an injectable dynamic hydrogel (GPOK-2@EVs) incorporating Artemisia argyi-derived extracellular vesicles (EVs) for localized immunomodulatory therapy. The hydrogel was formed through dual dynamic crosslinking between phenylboronic acid-modified gelatin and oxidized konjac glucomannan, providing self-healing capability, injectability, tissue adhesion, and pH/ROS-responsive degradation for sustained EV release. In vitro experiments demonstrated that GPOK-2@EVs exhibited excellent biocompatibility and effectively reprogrammed macrophages toward an anti-inflammatory phenotype, accompanied by reduced inflammatory cytokine secretion, alleviated ERS activation, decreased ROS accumulation, and improved mitochondrial integrity. GPOK-2@EVs significantly enhanced endothelial angiogenic activities, including cytoskeletal remodeling, tube formation, and migration. In a diabetic mouse wound model, GPOK-2@EVs accelerated wound closure, promoted granulation tissue formation and collagen deposition, enhanced vascularization, and suppressed inflammatory and ERS responses. These findings demonstrate that EV-loaded dynamic hydrogels can modulate the inflammatory microenvironment and promote diabetic wound regeneration, providing a promising biomaterial strategy for chronic wound therapy. Graphical GPOK-2@EVs hydrogel promotes wound healing through ROS-responsive EV release, macrophage modulation, inflammatory cytokine reduction, and enhanced angiogenesis.

Rare earth-doped nanomaterials can achieve diverse upconversion luminescence by changing doping ions, presenting them as promising candidates for rapid temperature measurement. However, most upconversion luminescent phosphors have a thermal quenching effect, which greatly weakens the luminous intensity of the material and impairs thermometry performances. Here, we studied the synthesis of Bi 4 Ti 3 O 12 nanosheets tridoped with Er 3+ , Yb 3+ , and Nd 3+ ions using molten salt method. Notably, this luminescent material demonstrates both thermal quenching and thermal enhancement effects. The unusual phenomenon results in a high sensitivity of the optical temperature sensor based on the inter-ion energy level between Er 3+ and Nd 3+ . At 523 K, this sensitivity reaches 5.94×10 −2 K −1 , surpassing that of sensors relying on the thermal coupling of only Er 3+ ions by nearly ninefold. Our findings suggest a promising strategy to augment the temperature sensing sensitivity of luminescent materials, paving the way for potential applications in precise temperature monitoring.

6-Methoxydihydrosanguinarine is a natural alkaloid derived from medicinal plants that exhibits significant antitumor activity, making it a promising candidate for cancer therapy. However, the exact molecular mechanisms underlying its effects require further investigation. In this study, we investigated the cytotoxicity and underlying mechanisms of 6-Methoxydihydrosanguinarine in human non-small cell lung cancer (NSCLC) cells. Our findings reveal that reactive oxygen species (ROS) accumulation is the key driver of its antitumor activity. Mechanistically, 6-Methoxydihydrosanguinarine activates the JNK signaling pathway and induces endoplasmic reticulum (ER) stress, both of which can be reversed by the ROS scavenger N-acetylcysteine (NAC). Interestingly, 6-Methoxydihydrosanguinarine also activates autophagy, and inhibition of autophagy reverses the JNK and ER stress pathway activation induced by 6-Methoxydihydrosanguinarine. Notably, 6-Methoxydihydrosanguinarine synergistically enhances cisplatin-induced NSCLC cell death, and this synergistic effect is abolished by NAC, highlighting the critical role of ROS accumulation in their combined efficacy. This study systematically elucidates the molecular mechanisms of 6-Methoxydihydrosanguinarine against NSCLC, revealing that, in addition to the JNK and autophagy pathways, ER stress also mediates its antitumor effects. Moreover, our data establish a rationale for exploring 6-Methoxydihydrosanguinarine in NSCLC therapy and highlight its combination with cisplatin as a potentially effective strategy.
