Drug-induced liver injury (DILI) has aroused wide concern. Finding new markers or indicators as well as detoxification molecules for DILI is of great significance and good application prospect, which can help develop effective preclinical screening methodology and corresponding treatment protocols. Herein, in this article, DILI caused by antidepressant drugs of duloxetine and fluoxetine and its remission were evaluated by a two-photon fluorescent probe, RPC-1, through discriminating and imaging HClO and H2S simultaneously. By being applied both in vitro and in vivo, RPC-1 revealed slight up-regulation of HClO and negligible liver damage after administration of either of the two drugs. In contrast, an apparent up-regulation of HClO and obvious liver damage was observed after combined administration of the drugs. Meanwhile, the pretreatment of N-acetyl cysteine (NAC) resulted in the increasing of endogenous H2S level, which contributed to the remission of DILI. The histological analysis and serological test both gave good consistency with the imaging results. These findings demonstrate that HClO may be an appropriate indicator of DILI, and H2S plays an important role in the antidotal effect of NAC. We envision that RPC-1 can be used as a powerful tool to predict clinical DILI and study the effect of antidote, as well as explore the molecular mechanisms involved.

H 2 S gas sensors with facile preparation, low detection limits, and high selectivity are crucial for environmental and human health monitoring. However, it is difficult to maintain a high response of H 2 S gas sensors under high humidity in practical applications. To face this dilemma, a layer-by-layer growth method is applied to in situ prepare a nanostructured Co(CO 3 ) 0.5 (OH)·0.11H 2 O/WO 3 coated by a hydrophobic hierarchical ZIF-67 as the H 2 S sensor. This novel composite exhibits excellent humidity immunity without sacrificing the excellent sensitivity and selectivity of H 2 S. At a low operating temperature of 90 °C, a remarkable response value of 1052.3 to 100 ppm H 2 S has been achieved, which is 779 and 9.36 times higher than that of pure WO 3 and Co(CO 3 ) 0.5 (OH)·0.11H 2 O/WO 3 , respectively. More importantly, an 82.2% relative response value remains at a high humidity of 75%RH. The sensing mechanisms are investigated using gas chromatography-mass spectrometry (GC-MS), which revealed that the reaction products are H 2 O and SO 2 . The high humidity immunity and fast response of the Co(CO 3 ) 0.5 (OH)·0.11H 2 O@ZIF-67/WO 3 demonstrate the layer-by-layer in situ synthesis method holds the potential application for the development of high-performance WO 3 -based H 2 S sensors.

Scope Allicin, a product of enzymatic reaction when garlic is injured, plays an important role in maintaining glucose homeostasis in mammals. However, the effect of allicin on glucose homeostasis in the state of insulin resistance remains to be elucidated. This study investigates the effect of allicin on glucose metabolism using different muscle fibers in a chicken model. Methods and results Day-old male Arbor Acres broilers are randomly divided into three groups and fed a basal diet supplemented with 0, 150, or 300 mg kg −1 allicin for 42 days. Results show that allicin improves the zootechnical performance of broilers at the finishing stage. The glucose loading test (2 g kg −1 body mass) indicates the regulatory role of allicin on glucose homeostasis. In vitro results demonstrate allicin increases glutathione (GSH) level and the expression of cystathionine γ lyase (CSE), leading to endogenous hydrogen sulfide (H 2 S) production in M. pectoralis major (PM) muscle-derived myotubes. Allicin stimulates adenosine monophosphate-activated protein kinase (AMPK) S-sulfhydration and AMPK phosphorylation to promote glucose uptake, which is suppressed in the presence of d , l -propargylglycine (PAG, a CSE inhibitor). Conclusion This study demonstrates that allicin induces AMPK S-sulfhydration and AMPK phosphorylation to promote glucose uptake via the CSE/H 2 S system in a muscle fiber-dependent manner.

To further enhance the performance of MOFs-based hydrogen sulfide (H 2 S) detection probes, we proposed a novel strategy for constructing for H 2 S detection. We have designed and synthesized a new reactive coumarin-based ligand (NTA) and directly prepared the MOFs-based fluorescent probe UiO-66-NTA using the new ligand. This approach overcame the issues of low grafting rates and uneven reactive sites associated with post synthetic modification (PSM) strategy prepared MOFs-based fluorescent probes. This construction strategy endowed MOFs with a greater number of active sites on their surfaces for H 2 S detection, thereby achieving superior sensing performance for H 2 S, which included an ultra-fast response time (5 s) and a low detection limit (0.063 μM). The rigid topological structure of the MOFs minimizes π-π interactions between fluorophores, conferring aggregation-induced emission (AIE) characteristics to the probe. To enhance the practical utility of the probe, a smartphone-assisted test strip sensing system was developed and successfully applied to the detection of real water samples. Additionally, the probe was integrated into the sodium alginate (SA) hydrogel to create a portable sensor UiO-66-NTA@SA for assessing the freshness of meat by detecting H 2 S.

The storage quality of fresh-cut peach fruit is compromised by weight loss and chromatic aberration. In this experiment, taurine at 30 mg L−1was applied to fresh-cut peaches to evaluate its effect on storage quality. Changes in weight loss, chromatic aberration, antioxidant system, endogenous nitric oxide (NO) metabolism, endogenous hydrogen sulfide (H2S) metabolism, and phenylpropanoid metabolism were studied. The results indicated that 30 mg L−1taurine effectively improved the ability to scavenge ROS, including •OH, O2•−, MDA, and H2O2, by inhibiting the decrease in the activities of SOD and CAT, enhancing POD activity, reducing PPO activity, and maintaining the AsA-GSH cycle. Moreover, taurine treatment increased the activities of 4CL and PAL and retarded the decrease in the activities of SKDH and C4H, thereby improving the accumulation of total phenols, flavonoids, and lignin. These findings showed that taurine mitigated oxidative damage in fresh-cut peaches by enhancing the antioxidant defense system. Exogenous taurine consistently enhanced NOS-like activity while decreasing NR activity in the early storage phase, and elevated the contents of L-Arginine, nitrite, and endogenous NO. Taurine treatment up-regulated the activities of SAT, L-CD, and OAS-TL, thus promoting endogenous H2S content in fresh-cut peaches. Taurine alleviated weight loss and chromatic aberration in fresh-cut peaches during storage by enhancing the antioxidant system and modulating NO, H2S and phenylpropanoid metabolism.

The clinical translation of cancer vaccines remains limited by the inability to effectively coordinate innate and adaptive immunity. Leveraging the metabolism pattern of the immune system, herein, we report a glucose metabolism-modulatory nanobiohybrid vaccine with superior antitumor immunity via orchestrating cross-presentation-enhanced adaptive immunity and proinflammatory signaling-mediated innate immunity. This nanobiohybrid vaccine is constructed by a nanoscale hydrogen-bonded organic framework (HOF) co-encapsulating glucose oxidase (GOx), and a model antigen, ovalbumin (OVA) (denoted OVA/GOx@HOF). OVA/GOx@HOF enables a GOx-mediated enzyme reaction that consumes glucose and produces H 2 O 2 within dendritic cells (DCs) in controlled kinetics, which disrupts intracellular glucose metabolism and causes redox imbalance but minimally impacts cell viability. These effects induce autophagic machinery in DCs that promotes antigen cross-presentation, thereby boosting cytotoxic T cell response. Meanwhile, through RNA sequencing, we found that OVA/GOx@HOF stimulates innate immunity through an NF-κB signaling-mediated and endogenous hydrogen sulfide (H 2 S)-enhanced NLRP3 inflammasome pathway, leading to significant DC maturation. In a melanoma tumor model, this nanobiohybrid vaccine effectively inhibits tumor growth and metastasis, and can be applied to encapsulate neoantigens to generate a potent personalized cancer vaccine. Our findings suggested the remarkable potential of enzyme-driven nanobiohybrid materials as a metabolism-modulatory platform for cancer vaccines.

The structure and morphology of gas-sensitive materials were extremely important to performances of gas sensor. In this work, ZnO nano-crystalline (ZnO NCs) was synthesized on foam-like GaN (F-GaN) by sol-gel method as gas-sensitive materials of H 2 S gas sensor. When ZnO NCs/F-GaN composite structure was used as gas- sensitive materials for detection of H 2 S with the concentration of 50 ppm at 220 °C. Importantly, rapid response and recovery time was 78 and 31 s, respectively. Enhancement of the sensing response was mainly attributed to the combination between ZnO NCs and F-GaN. In addition, a multiphase catalytic mechanism was proposed to description the catalytic role of F-GaN in the gas sensing procedure. These results further broadened the application of as-prepared metal oxide semiconductor based GaN in the gas sensing field.

The lengthy processes and cumbersome operations for the separate removal of COS and H 2 S have severely limited the development of blast furnace gas recycling process. CuO exhibits excellent H 2 S oxidation performance, but weak COS hydrolysis performance; in contrast, ZrO 2 shows the opposite trend. However, the functional combination of CuO and ZrO 2 has never been used for simultaneous removal of COS and H 2 S. In this study, a Cu/Zr co-doped K@TiO 2 bifunctional catalyst was tailored to simultaneously remove COS and H 2 S. The combined COS and H 2 S capacity of this catalyst reaches 340.5 mg/g at a low temperature of 60 °C, which is much higher than those of other reported bifunctional catalysts. The microcrystal structure formed by Cu/Zr co-doping increases the specific surface area of the catalyst and promotes the adsorption of COS/H 2 S. Specifically, Zr doping increases the content of moderate basic sites and significantly improves the hydrolysis efficiency of COS. Moreover, Cu doping induces lattice distortion of TiO 2 , while CuO captures 4 s valence electrons of K to form Cu 2 O. These two processes lead to the release of lattice oxygen from TiO 2 and CuO, generating oxygen vacancies and ·O 2 − , which promotes H 2 S oxidation. By proposing a simple route for the bifunctionalization of a desulfurization catalyst, this study drives innovation in both the theory and application of simultaneous removal of COS and H 2 S.

For the therapy attenuating renal ischemia–reperfusion (IR) injury, a novel drug delivery system was urgently needed, which could precisely deliver drugs to the pathological renal tissue. Here, we have prepared new nanomaterials with a reactive oxygen species (ROS)-responsive hydrogen sulfide (H2S) donor and hyaluronic acid that targets CD44 receptor. The novel material was synthesized and characterized via related experiments. Then, rapamycin was loaded, which inhibited kidney damage. In the in vitro study, we found that the micelles had ROS-responsiveness, biocompatibility, and cell penetration. In addition, the experimental results showed that the intracellular H2S concentration after administration was threefold higher than that of the control group. The western blot assay revealed that they have anti-inflammatory effects via H2S donor blocking the NF-κB signaling pathway. Consequently, the rising CD44 receptor-targeting and ROS-sensitive H2S donor micelles would provide a promising way for renal IR injury. This work provides a strategy for improving ischemia/reperfusion injury for pharmaceuticals.

The biphasic amine solvent based on ionic liquid could effectively improve the efficiency of carbon dioxide capture and regeneration, greatly reduce the energy consumption of regeneration, and hold an excellent application prospect in the field of large-scale biogas upgrading. In this work, a biphasic mixed amine system consisting of Methyl monoethanolamine (MMEA), N-methyldiethanolamine (MDEA), and 1-butyl-3-methylimidazolium tetrafluoroborate (BF) was prepared for the removal of trace components and CO 2 during biogas upgrading and compared with traditional 30 wt% MEA and mixed amine solution of MMEA and MDEA for biogas upgrading. The results showed that H 2 S and NH 3 would chemically react with the biphasic solvent and thus facilitate the removal of trace components, and that the reacted solution would not affect its regeneration characteristics and could basically be recycled at a regeneration temperature of 125 °C. In addition, the phase separation efficiency of the biphasic agent was not affected by the introduction of trace components, and all reaction products except the ionic liquid were enriched in the rich phases and the carbon enrichment rate reached about 98%, which reduced regeneration energy consumption by 44.07% compared to 30 wt% MEA. Finally, the mechanism of phase separation of reaction products and ionic liquids was discussed in the context of quantum chemical calculations, and the importance of intermolecular van der Waals forces in the phase separation process was clarified, and the reasons for the appearance of phase separation phenomena were explained.

Hydrogen sulfide (H₂S) is a colorless compound with a characteristic rotten egg odor that serves as a significant indicator of pathological processes and food spoilage. In this study, we developed a novel probe SCN for H 2 S detection. When exposed to H 2 S, the Probe SCN selectively cleaves the oxygen-nitrile bond, resulting in a strong red fluorescence emission at 662 nm, which can be attributed to recovery from the intramolecular charge transfer (ICT) effect. This probe demonstrated outstanding features, including a substantial Stokes shift (236 nm) and a low detection limit of 0.41 μM. We successfully used the Probe SCN to detect H 2 S changes in common food samples such as eggs, fish, pork, and shrimp. Furthermore, the Probe SCN can also be used to detect H 2 S in living cells and mice. These findings confirmed the effectiveness of the SCN probe as a real-time tool for H 2 S detection and related imaging applications.

Neuropathic pain (NP) imposes a significant burden on individuals, manifesting as nociceptive anaphylaxis, hypersensitivity, and spontaneous pain. Previous studies have shown that traumatic stress in the nervous system can lead to excessive production of hydrogen sulfide (H 2 S) in the gut. As a toxic gas, it can damage the nervous system through the gut-brain axis. However, whether traumatic stress in the nervous system leading to excessive production of H 2 S in the gut can ultimately cause neuropathic pain through the gut-brain axis remains to be investigated. This study established a model of chronic constriction injury (CCI) in mice to determine its effects on gut H 2 S production, the associated damage via the gut-brain axis, the potential neuropathic pain, as well as the probable mechanism. A CCI mouse model was developed using a spinal nerve ligation approach. Subsequently, the mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were used to determine the mice’s pain thresholds. A variety of assays were performed, including immunofluorescence, western blotting, real-time quantitative Polymerase Chain Reaction (PCR), and membrane clamp whole-cell recordings. Mice subjected to CCI showed decreased MWT and TWL, decreased ZO-1 staining, decreased HuD staining, increased Glial fibrillary acidic protein (GFAP) staining, increased expression of tumor necrosis factor-alpha (TNF-α) protein and interleukin-6 (IL-6) protein, increased expression of NMDAR2B (NR2B) protein and NR2B mRNA, increased colocalization of vGlut2- and c-fos-positive cells, and a higher amplitude of evoked excitatory postsynaptic potential (EPSP) compared to Sham group. These changes were significantly reversed by H 2 S inhibitor treatment, and the specific NMDA receptor inhibitor MK-801 effectively restored the neurotoxicity of H 2 S. H 2 S is involved in CCI-induced neuropathic pain in mice, which might be mediated by the activation of the NMDA signaling pathway.
