Design of donor–acceptor conjugated polymers based on diketopyrrolopyrrole for NIR-II multifunctional imaging

Mengyu Cao,Xuezhen Ma,Chang Wang,Wentao Zou,Fang Wang,Bing Yu,Hailin Cong,Youqing Shen

Diketopyrrolopyrrole (DPP) is an excellent photosensitizer and photothermal agent with the advantages of good planarity, strong electron affinity, high electron mobility, easy purification, easy structural modification and high molar absorption coefficient. It is regarded as one of the ideal choices for the design and synthesis of efficient organic photovoltaic materials. Therefore, two kinds of donor–acceptor (D–A) conjugated polymers were designed and synthesized with DPP as the acceptor, and their optical properties and applications in the near-infrared region were studied. The quantum yield (QY) of PBDT-DPP is 0.46%, and the highest temperature reached within 10 minutes after irradiation with a 660 nm laser is 60 °C. Another polymer, EDOT-DPP, has a QY of 0.48%, and its semiconductor polymer nanoparticle aqueous solution can reach 60 °C within 12 minutes under laser irradiation, achieving photothermal treatment of nude mice tumors. Both polymer NPs have good biocompatibility and promising applications in bioimaging and photothermal therapy.

Activatable NIR Fluorescence Probe for Epinephrine Detection and Bioimaging Based on Anionic Heptamethine Cyanine

Chen Luo,Youjia Chen,Jinxuan Gu,Huihui Cai,Huijin Lin,Zhigang Jin,Chaobiao Huang

Epinephrine (EP) is an essential catecholamine in the human body. Currently, most EP detection methods are not suitable for in vivo detection due to material limitations. An organic small molecule fluorescent probe based on a chemical cascade reaction for the detection of EP was designed. Anionic heptamethine cyanine dye was selected as a fluorescent dye because of its NIR fluorescence emission with excellent biocompatibility. The secondary amine of EP nucleophilically attacks the carbonate of the probe with its stronger nucleophilicity and further undergoes intramolecular nucleophilic cyclization to release the fluorophore. Other substances containing only primary amines or no β-OH lack reaction competitiveness due to their weaker nucleophilicity or inability to undergo further cyclization. The fluorescence recovery of the probe was linearly related to the EP concentration of 2–75 μmol/L. The detection limit was 0.4 μmol/L. The recovery rate was 94.78–111.32%. Finally, we successfully achieved bioimaging of EP in living cells and EP analogue in nematodes.

A smart capecitabine imprinted nanocontainer with the dual-responsive performance to pH/NIR light

Chensi Zeng,Yanfang Gong,Chenxi Qi,Xianghua Zeng,Ni Tan

Capecitabine (CAP) is a kind of prodrug that can be metabolized into 5-fluorouracil (5-FU) in vivo, so it is usually classified as the antimetabolite fluoropyrimidine deoxynucleoside analog. In this paper, CAP, upconversion nanoparticles (UCNPs), 3,5-dicarboxy-4-methacrylate-based azobenzene (MAPDIA)/acrylic acid-functionalized gelatin, N,N′-methylene bisacrylamide (MBA), and ammonium persulfate (APS) were used as template, light trigger, bifunctional monomers, cross-linking agent and initiator, respectively. By using the free-radical polymerization and the molecular imprinting technique, the potential drug delivery nanocontainer CAP-MIP with the dual-responsive performance to pH/near infrared light (NIR) was successfully prepared. Furthermore, the characterization of CAP-MIP was also carried out by the modern instrumental analysis such as FTIR, XRD, TGA, BET and so on. The stimulus-responsive release experiment in vitro indicated the maximum cumulative release rate of CAP changed from 1.71 % to 89.54 % when the pH value in medium varied from 1.2 to 8.3, and the equilibrium release time point also greatly prolonged to 420 min from 40 min, respectively. When irradiated by 980 nm monochromatic NIR light in the same solution of pH 6.8, the maximum release rate of the sample could reach 96.21 % at 180 min, which respectively corresponded to the data of 41.84 % at 420 min under the condition of the absence of additional NIR light. In addition, CAP-MIP also had the excellent specific adsorption for CAP with the selectivity factor of 2.904 and the maximum CAP-loaded capacity of 7 mg•g −1 . The above information denoted the target material should be of some potential in drug delivery system application.

Nanocellulose-mediated conductive hydrogels with NIR photoresponse and fatigue resistance for multifunctional wearable sensors

Chenyu Sang,Shaowei Wang,Xiaoyue Jin,Xiaoyu Cheng,Huining Xiao,Yiying Yue,Jingquan Han

The rapid development of hydrogels has garnered significant attention in health monitoring and human motion sensing. However, the synthesis of multifunctional conductive hydrogels with excellent strain/pressure sensing and photoresponsiveness remains a challenge. Herein, the conductive hydrogels (BPTP) with excellent mechanical properties, fatigue resistance and photoresponsive behavior composed of polyacrylamide (PAM) matrix, 2,2,6,6-tetramethylpiperidin-1-yloxy-oxidized cellulose nanofibers (TOCNs) reinforcement and polydopamine-modified black phosphorus (BP@PDA) photosensitizer are prepared through a facile free-radical polymerization approach. The PDA adhered to the BP surface by π-π stacking promotes the optical properties of BP while also preventing BP oxidation from water. Through hydrogen bonding interactions, TOCNs improve the homogeneous dispersion of BP@PDA nanosheets and the mechanical toughness of BPTP. Benefiting from the synergistic effect of PDA and TOCNs, the conductive BPTP integrates superior mechanical performances, excellent photoelectric response and photothermal conversion capability. The BPTP-based sensor with high cycling stability demonstrates superior strain sensitivity ( GF  = 6.0) and pressure sensing capability ( S  = 0.13 kPa −1 ) to monitor various human activities. Therefore, this work delivers an alternative construction strategy for generating high-performance conductive hydrogels as multifunctional wearable sensors.

NIR-activated endoplasmic reticulum stress amplifier to regulate phagocytosis for macrophage-mediated cancer immunotherapy

Yaru Zhang,Wenyu Xie,Jiamin Luo,Rui Gao,Zhimin Gao,Xinyi Wang,Yongxin Huang,Chen Lin,Meiling Tan,Zhiyao Hou

Effectively activating macrophages to engulf tumor cells has attracted intensive attention in cancer immunotherapy. However, tumor cells can upregulate anti-phagocytic signals (such as CD47 molecules) to bind with signal regulatory protein-α (SIRPα), ultimately providing a “don’t eat me” signal to evade phagocytosis. Blockading CD47 alone is insufficient to stimulate macrophages phagocytosis due to the absence of endogenous activating signals (“eat me” signal). Herein, a mesoporous silica (MSN) modified by Cu 9 S 5 nanoparticle with mimicking enzyme activities were synthesized and utilized to electrostatically load the anti-CD47 antibodies (aCD47), generating MCC nanozymes, to improve macrophage-mediated cancer immunotherapy. MCC nanozymes can target tumor cells by specifically binding to the cell-surface CD47, disabling the “don’t eat me” signal. Meanwhile, under near infrared light (1064 nm) stimulation, MCC nanozymes exhibit the enhanced glutathione oxidase-like and peroxidase-like enzymatic activities by the photothermal effect, which lead the reactive oxygen species (ROS) generation to induce endoplasmic reticulum (ER) stress amplification, resulting the exposure of calreticulin (CALR) for providing the endogenous “eat-me” signals to macrophages. In a 4T1 breast cancer model, even under mild photothermal condition, MCC nanozymes effectively inhibit tumor growth and metastasis. Collectively, this dual-regulatory strategy presents a promising approach for macrophage-mediated cancer immunotherapy.

Xue-Mei Dong,Chen Chen,Yin-Xiang Li,Hong-Chao Sun,Bin Liu,Zi-Fan Li,Kai-Li Wang,Zi-Xi He,Meng-Na Yu,Wei Huang,Ju-Qing Liu

Neuromorphic vision sensors capable of multispectral perception and efficient recognition are highly desirable for bioretina emulation, but their realization is challenging. Here, we present a cocrystal strategy for preparing an organic nanowire retinamorphic vision sensor with UV–vis–NIR perception and fast recognition. By leveraging molecular-scale donor–acceptor interpenetration and charge-transfer interfaces, the cocrystal nanowire device exhibits ultrawide photoperception ranging from 350 to 1050 nm, fast photoresponse of 150 ms, high specific detectivity of 8.2 × 1012 Jones, and responsivity of 15 A W–1, as well as retina-like photosynaptic plasticity behaviors. Utilizing the sensor nerve and convolutional neural network, the architecture achieves 90% accuracy in recognizing colorful images. The cocrystal design offers an effective method for constructing nanowire photosynases with high performance in artificial visual systems.

Self-activated LiGa5O8 storage phosphor: The insights into photo/thermo/mechano-stimulated NIR Luminescence

Min Jia,Xiangyu Zhang,Xue Yang,Zehao Lin,Dingjun Jia,Yuqiang Wang,Sining Yun,Dangli Gao

Multi-mode storage phosphors with photo/thermo/mechano-stimulated luminescence (PSL/TSL/ML) hold great potential applications in many fields such as biological imaging, human-machine interface, robotic manipulation, and stress/temperature visualization sensing. However, the physical mechanisms underlying this 'self-sustaining' luminescence are still debated, which in turn hinders the development of materials. Here, we demonstrated that the intrinsic defects such as the VO•• and Oi′′ in the matrix play an important role in the electronic structure and various physical properties of LiGa5O8 storage phosphor by combining first-principles calculations and experimental methods. Specifically, the intrinsic defects lead to the smaller bulk (B) and shear (G) modulus, Young’s moduli (E), Poisson’s ratio (v), B/G, and the bigger elastic anisotropy index (AU) and hardness (H) in three LiGa5O8 defect models, which is extremely advantageous for PSL/TSL/ML. We also demonstrate that the ML process, that is different from PSL/TSL processes, are intimately linked to the activation threshold of charge carriers in traps. This threshold can be lowered under axial stress by bandgap narrowing via weakening the binding force of polyhedral units to bind electrons in the converting process between mechanical energy and light energy under stress load. This study provides not only direct evidence for potential variation in ML process, but also guidance for designing the storage phosphors via defect engineering.

NIR Light-Fuse Drug-Free Photothermal Armor-Piercing Microcapsule for Femoral Vein Thrombosis Therapy

Ning Yang,Zhicheng Qian,Renqiang Yuan,Weikun Li,Xin Tan,Zonghao Liu,Qianli Zhang,Liqin Ge,Ling Liu

Acute thrombosis and its complications are leading global causes of disability and death. Existing thrombolytic drugs, such as alteplase and urokinase (UK), carry a significant bleeding risk during clinical treatments. Thus, the development of a novel thrombolysis strategy is of utmost urgency. Based on the previous work, the hollow structure of microcapsules (MC) is fabricated. Subsequently, armor-piercing MC, known as Fucoidan/S-Nitrosoglutathione/Melanin@MC (FGM@MC) is obtained, using a layer-by-layer (LBL) self-assembly method. Utilizing near-infrared (NIR) light as a trigger, the FGM@MC demonstrated photothermal thrombolysis at the site of thrombus due to its stable and outstanding photothermal properties. Simultaneously, photothermal stimulation leads to the release of a significant amount of nitric oxide from the FGM@MC, resulting in cavitation effects for mechanical thrombolysis. In vivo experiments confirmed the stable release of nitric oxide under NIR light irradiation. Treatment of femoral vein thrombosis in rats revealed that the thrombolytic effectiveness of FGM@MC+NIR (53.71%) is comparable to that of UK (59.70%). Notably, FGM@MC does not interfere with the coagulation function of rats and exhibits a favorable safety profile. In conclusion, this study demonstrates that the drug-free armor-piercing microcapsule has significant potential in the treatment of thrombosis, offering a safe and effective alternative to traditional thrombolytic therapies.

NIR-light controlled local delivery of nitric oxide based on self-targeting carbon dots

Na Chen,Yan Bai,Yuan Fang Li,Lei Zhan,Cheng Zhi Huang

Nitric oxide (NO)-based gas therapy has received ever-increasing attention in treatment of many diseases, especially cancer. However, the selective organelle targeted delivery and controllable release of NO remains highly desirable. In this work, a sequentially targeting delivery nanoplatform has been constructed by encapsulating heat-responsive S -nitrosocysteine (the NO donor, SNC) conjugated mitochondria-targeting carbon dots (MitoCDs) and indocyanine green (ICG) within liposomes which are further modified with aptamer that can specifically bind to nucleolin overexpressed on the surface of several cancer cells. MitoCDs were prepared via one-step hydrothermal reaction using citric acid and m -diethylaminophenol as starting materials and used without any external targeting molecules. ICG molecules were employed as NIR-triggered photothermal agent followed by NO precisely controlled release in mitochondria. By combining gas and photothermal treatment, the nanocarrier produced a synergistic and superior killing effect on several cancer cell lines. And the cell growth inhibition of NCL-CSI@Lip on HEp-2 cells raised up to 72.39 %. Such an ‘all-in-one’ mitochondria-targeted NO nanocarrier may lead to new way for overcoming the problem of multidrug resistance in intracellular drug delivery.

Oligopeptide modified ICG/Cisplatin@CaCO3 for targeted osteosarcoma NIR-II imaging guided multimodal therapy

Li-chen Ji,Jia-qing Huang,Xu-gang Zhong,Qi-hong Sun,Yong Fan,Li Yin,Qiong Zhang,Yu Cai,Zeju He

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

NIR-triggered self-healing SiO2/melanin coatings with enhanced corrosion-resistant and mechanical robustness

Zhiqiang Fang,Yangkai Xiong,Lei Huang,Jipeng Li,Daxiong Hu,Qitong Mao,Hao Jiang,Xuewei Tang,Panpan Tang,Zheng Li,Guoqing Wang

Polymer coatings are inherently susceptible to deformation and irreversible damage when they are subjected to external forces during service. Self-healing coatings integrated with exceptional mechanical robustness and corrosion resistance are essential for industrial applications but have been deemed challenging. Here, we present a NIR-triggered self-healing coating by combining a toughness epoxy network with melanin-modified silica (MMS). Moreover, to investigate the impact of pore size and porosity of silica on the corrosion protection performance, three types of various silica particles are selected as building blocks. The melanin-modified fumed silica/epoxy coatings demonstrate superior elongation (275 %), outstanding tensile strength (5.31 MPa) and high toughness (7.37 MJ·m 3 ). Moreover, the introduction of the MMS can not only improve the anticorrosion performance (|Z| 0.01 Hz value of 1.24 × 10 10 Ω·cm 2 ) but also realize fast photo-thermal repair (60 s). Our strategy furnishes a universal layout prototype for the manufacture of robust self-healing coatings with a scalable and cost-effective preparation method.

NIR-triggered bacterial cellulose-based wound dressings for multiple synergistic therapy of infected wound

Meiyan Sun,Dongmei Li,Yan Xi,Xiaotong Qin,Yuting Liao,Xiaozhi Liu,Shiru Jia,Yanyan Xie,Cheng Zhong

Skin wounds are repaired by a complex series of events and overlapping phases in which bacterial infection and insufficient angiogenesis at the wound site delay the healing process. Thus, functional wound dressings with enhanced antibacterial activity and angiogenic capacity have attracted attention. Herein, bacterial cellulose (BC)-based dressings were successfully fabricated by functionalization with a polydopamine (PDA) coating and copper sulfide nanoparticles (CuS NPs). Under 808 nm laser illumination, the BC/PDA/CuS composite membranes exhibited outstanding adjustable photothermal and photodynamic activities as well as controlled Cu 2+ release, endowing the composite membranes with synergetic antibacterial activity. Specially, a bactericidal efficiency of 99.7 % and 88.0 % for Staphylococcus aureus and Escherichia coli was achieved after treatment with BC/PDA/CuS5 sample under NIR irradiation (0.8 W/cm 2 , 10 min), respectively. Moreover, the BC/PDA/CuS5 composite membrane could enhance the angiogenesis due to the released Cu 2+ . In vivo experiments revealed that the BC/PDA/CuS5 composite membrane dressing could accelerate the wound closure process of the full-thickness skin defects with S. aureus by synergistically reducing inflammation, enhancing collagen deposition, and promoting vascularization under NIR irradiation. Additionally, the BC/PDA/CuS5 composite membrane exhibited high biocompatibility and biosafety. This work offers a new strategy to prepare multifunctional BC-based dressing for clinical wound healing.
