Rapid screening and detection of XOD inhibitors from S. tamariscina by ultrafiltration LC-PDA–ESI-MS combined with HPCCC

Wang Jing,Liu Shu,Ma Bing,Chen Lina,Song Fengrui,Liu Zhiqiang,Liu Chun-ming

Xanthine oxidase (XOD) catalyzes the metabolism of hypoxanthine and xanthine to uric acid, the overproduction of which could cause hyperuricemia, a risk factor for gout. Inhibition of XOD is a major treatment for gout, and biflavonoids have been found to act as XOD-inhibitory compounds. In this study, ultrafiltration liquid chromatography with photodiode-array detection coupled to electrospray-ionization tandem mass spectrometry (UF-LC-PDA–ESI-MS) was used to screen and identify XOD inhibitors from S. tamariscina . High-performance counter-current chromatography (HPCCC) was used to separate and isolate the active constituents of these XOD inhibitors. Furthermore, ultrahigh-performance liquid chromatography (UPLC) and triple-quadrupole mass spectrometry (TQ-MS) was used to determine the XOD-inhibitory activity of the obtained XOD inhibitors, and enzyme kinetics was performed with Lineweaver–Burk (LB) plots using xanthine as the substrate. As a result, two compounds in S. tamariscina were screened as XOD inhibitors: 65.31 mg amentoflavone and 0.76 mg robustaflavone were isolated from approximately 2.5 g  S. tamariscina by use of HPCCC. The purities of the two compounds obtained were over 98 % and 95 %, respectively, as determined by high-performance liquid chromatography (HPLC). Lineweaver–Burk plot analysis indicated that amentoflavone and robustaflavone were non-competitive inhibitors of XOD, and the IC 50 values of amentoflavone and robustaflavone for XOD inhibition were 16.26 μg mL −1 (30.22 μmol L −1 ) and 11.98 μg mL −1 (22.27 μmol L −1 ), respectively. The IC 50 value of allopurinol, used as the standard, was 7.49 μg mL −1 (46.23 μmol L −1 ). The results reveal that the method for systematic screening, identification, and isolation of bioactive components in S. tamariscina and for detecting their inhibitory activity using ultrafiltration LC–ESI-MS, HPCCC, and UPLC–TQ-MS is feasible and efficient, and could be expected to extend to screening and separation of other enzyme inhibitors. ᅟ

Yirong Jiang,Shuqin Mao,Weisu Huang,Baiyi Lu,Zengxuan Cai,Fei Zhou,Maiquan Li,Tiantian Lou,Yajing Zhao

Variations of phenylethanoid glycoside profiles and antioxidant activities in Osmanthus fragrans flowers through the digestive tract were evaluated by a simulated digestion model and UPLC/PDA/MS. Major phenylethanoid glycosides and phenolic acids, namely, salidroside, acteoside, isoacteoside, chlorogenic acid, and caffeic acid, were identified in four cultivars of O. fragrans flowers, and the concentration of acteoside was the highest, being up to 71.79 mg/g dry weight. After simulated digestion, total phenylethanoid glycoside contents and antioxidant activities were significantly decreased. Acteoside was identified as decomposing into caffeic acid, whereas salidroside was found to be stable during simulated digestion. According to Pearson’s correlation analysis, acteoside contents showed good correlations with antioxidant activities during simulated digestion (R2 = 0.994, P < 0.01). In conclusion, acteoside was the major contributor to the antioxidant activity of O. fragrans flowers, and salidroside was considered as the major antioxidant compound of O. fragrans flowers in vivo.

A Sequential Therapeutic Strategy Based on Aptamer/Polymer-Functionalized PDA/Ag for Precise Acinetobacter Baumannii Imaging and Pneumonia Treatment

Yun Li,Zhuorui Dong,Yingfei Ma,Jian Zhang,Yanan Wang,Dandan Cui,Kaifang Wang,Baojie Du,Runzhi Li,Zhifei Wang,Ruixue Wang,Bing Cao,Ruiping Zhang

Acinetobacter baumannii ( A. baumannii ) is a leading cause of pneumonia. Conventional antibiotics can increase the risk of drug resistance and non-specifically harm microflora, disrupting their balance. In addition to pathogenic bacteria, lung inflammation also damages tissue. This study develops an aptamer/polymer-functionalized mesoporous polydopamine/silver nanocomposite (Apt/ODEA-PDA/Ag-ICG) to specifically target A. baumannii and sequentially exhibit antibacterial/antioxidative properties for pneumonia treatment. After synthesizing the polyzwitterion (ODEA), the polymer/aptamer is conjugated with the PDA/Ag core, and ICG is further labeled to fabricate the nanocomposite. Upon intratracheal instillation, nanocomposites penetrate the mucus barrier with the help of polyzwitterions. The nanocomposite precisely targets A. baumannii through specific recognition by the aptamer, without affecting other microflora. Moreover, ICG is conjugated and distributed disorderedly within the polymer. ICG near the PDA/Ag core contributes to high NIR-I absorbance and good photothermal ability. ICG further from the PDA/Ag acts as a fluorescent molecule for specific A. baumannii imaging. Furthermore, PDA/Ag efficiently exhibits synergistic antibacterial activity owing to the acidic environment/heat-accelerated Ag + release and photothermal effect. Importantly, alongside Ag + release, the antioxidant ability of PDA scavenges reactive oxygen species and alleviates inflammation, thereby accelerating pneumonia treatment. In summary, nanocomposites provide an alternative approach for designing advanced antibacterial systems.

Le Wang,Jianguang Liu,Yan Shen,Yanli Yin,Zifu Ni,Jun Xi,Yuansen Hu,Qipeng Yuan

The preparation of a novel composite carrier of polydopamine-modified carbon fiber/polyurethane foam (PDA-CF/PUF) was proposed to improve cell immobilization and the fermentation of xylitol, which is an important food sweetener and multifunctional food additive.Candida tropicaliswas immobilized on the composite carrier by adsorption and covalent binding. The properties and immobilization mechanism of the composite carrier and its effect on immobilized cells were investigated. It showed that the modification of PDA enhanced the loading of CF on the PUF surface and the adhesion of cells on the composite carrier surface. Also, the biocompatibility of carriers to cells was improved. In addition, the introduction of PDA increased the active groups on the surface of the carrier, enhanced the hydrophilicity, promoted the cells immobilization, and increased the xylitol yield. It was also found that expression of the related gene XYL1 in cells was significantly increased after the immobilization of the PDA-CF/PUF composite carrier during the fermentation. The PDA-CF/PUF was an immobilized carrier with the excellent biocompatibility and immobilization performance, which has great development potential in the industrial production of xylitol.

Multifunctional PVDF/CeO2@PDA nanofiber textiles with piezoelectric and piezo-phototronic properties for self-powered piezoelectric sensor and photodetector

Shuhong Huang,Xiaohui Zhao,Chenlong Hao,Zhixin Ma,Hui Wang,Aijia Zhang,Wanzhihan Zhang,Ling Li,Wenming Zhang

In recent years, multifunctional sensors have attracted significant attention due to their ability to detect different types of signals. However, it still remains a challenge to develop flexible materials with multiple sensing functions. Herein, we propose a strategy to fabricate multifunctional flexible textiles (PCP) that can apply piezoelectric and piezo-phototronic effects to self-powered piezoelectric sensor and photodetector (PD) by utilizing the synergistic effect of polydopamine-coated cerium oxide (CeO 2 @PDA) nanoparticles and poly (vinylidene fluoride) (PVDF). The CeO 2 @PDA nanoparticles served as local nucleating agents and photosensitizers for the PVDF, enhancing the polar crystalline phase and inducing piezoelectric effect, semiconductor, and optical process coupling in PCP textiles. The piezoelectric sensors consisting of PCP textiles produced extremely high sensitivity (1–10 kPa: 1.27 V/kPa; 10–50 kPa: 0.085 V/kPa; 50–100 kPa: 0.02 V/kPa) and significant electrical output (25.5 V and 0.37 μA). Besides, the piezoelectric sensor had extensive applications including pulse wave measurement, limb motion detection, and active voice recognition. It's especially noteworthy that the PD based on PCP textiles is sensitive for the detecting of sunlight illumination as confirmed by response time (t On  = 2 s and t Off  = 4.6 s). Meanwhile, the fabricated self-powered PD showed significant improvement in output current, confirming the realization of the piezo-phototronic effect in PCP textiles. In this work, the low-cost and high-efficiency multifunctional sensing materials were successfully prepared, providing a novel approach for the next generation of self-powered piezoelectric sensors and photodetectors.

Sensitive colorimetric detection of heparin using reverse modulation of peroxidase- and oxidase-mimetic activities in Fe3O4@PDA@MnO2 nanocomposites

Tengteng Wang,Lian Xue,Chao Di,Peng Ma,Xiaojun Feng,Peng Chen,Wei Du,Bi-Feng Liu

Heparin, a widely studied glycosaminoglycan, plays crucial roles in the regulation of various physiological and pathological processes. Therefore, it's important to develop highly selective and sensitive methods for convenient monitoring of heparin levels in biological systems. We report the design and synthesis of Fe 3 O 4 @PDA@MnO 2 nanoparticles (FPM-NPs), which exhibit dual enzymatic activities, enabling quantitative detection of heparin. The FPM-NPs feature a unique tri-layer spherical shell structure, possessing both peroxidase-like and oxidase-like activities, and catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence or absence of H 2 O 2 . Remarkably, upon co-incubated with heparin, the oxidase activity of FPM-NPs decreases, while the peroxidase activity increases. By leveraging these dual enzymatic properties of FPM-NPs, a highly sensitive and specific colorimetric detection of heparin is achieved, with a detection limit reaching 6.51 nM and a good linear response to quantify heparin ranging 10–800 nM. Additionally, the developed FPM-NPs are successfully applied to measure heparin in fetal bovine serum samples. We also extend this detection method to a paper-based chip, enabling portable detection of heparin through grayscale analysis of mobile phone photographs. The multi-nanozyme-based heparin detection approach provides a new perspective for future research on expanding the application of nanocomposite materials in biomedical detection and analysis.

Hongyong Lin,Tengyang Liao,Yaoyao Sang,Jin Mao,Yu Liu,Bin Zhang,Di Sun,Wei Jiang

Highly accurate and sensitive detection of Vibrio parahaemolyticus is crucial for ensuring seafood safety and preventing the transmission of foodborne diseases. Herein, a dual-recognition strategy-driven colorimetric biosensor for the detection of V. parahaemolyticus was developed. This biosensor was based on the use of PEG-mediated cefepime-functionalized magnetic nanoparticles (Cefe-PEG-MNPs) and aptamer-modified Fe-doped polydopamine (Fe@PDA) nanozymes. The magnetic platform Cefe-PEG-MNPs was introduced to enhance the enrichment efficiency of V. parahaemolyticus , achieving a capture efficiency exceeding 90%. The Fe@PDA nanozyme exhibited notable peroxidase mimetic enzyme activity. The colorimetric biosensor developed for V. parahaemolyticus detection demonstrated good sensitivity, with a detection range spanning from 2.1 × 10 1 to 2.1 × 10 6  CFU/mL, and a limit of detection of 21 CFU/mL. Moreover, the biosensor demonstrated remarkable specificity, achieving good recovery rates from 95.80% to 103.80% in Penaeus vannamei with a relative standard deviation of less than 1.38%. The newly developed colorimetric biosensor offers a remarkable opportunity for the accurate and sensitive detection of V . parahaemolyticus in seafood.

Qiuyan Bi,Shiai Xu

A positively charged PI/PDA/PEI nanofiltration membrane was prepared in this study by crosslinking between polyethyleneimine (PEI) and poly(dopamine) (PDA) on the polyimide (PI) support membrane for separating Mg 2+ and Li + from MgCl 2 /LiCl mixed solution, and its chemical structure, surface properties, and separation performance were investigated. The results indicate that the PI/PDA/PEI membrane has a thin positively charged layer and shows excellent rejections against many inorganic salts [MgCl 2 (93.1%) > MgSO 4 (86.5%) > Na 2 SO 4 (74.7%) > LiCl (37.6%)]. More importantly, the membrane exhibits long-term stability and good separation of Mg 2+ and Li + , and the Mg/Li ratio of the permeate of brine obtained from West Taijnar salt lake can be reduced from 65.3 to 5.0 at pH 6.5 by filtration.

Fabrication of complex hydrogels based on pullulan, nanocellulose and carbon dots using photo-Fenton process to eliminate doxycycline contamination

Meng Liu,Wenzhuo Lv,Fan Zeng,Canhui Wang,Xin Guo,Bo Zhou,Yiqiang Wu

Overcoming the inherent activity–stability trade-off in photo-Fenton systems requires advanced material architectures that integrate multifunctional synergies. In this study, we developed a multifunctional hydrogel, termed PCCF, fabricated by confining Fe 3 O 4 @polydopamine (PDA) nanoparticles within a three-dimensional (3D) pullulan–nanocellulose matrix specifically tailored for efficient doxycycline (DOX) elimination. PCCF exhibited a hierarchically engineered structure based on three synergistic strategies: (1) a pullulan (Pu)–cellulose nanofibrils (CNF)@carbon dots (CD) co-network with a hierarchically porous architecture (surface area: 230.6 m 2 /g), enabling ultrahigh adsorption capacity (Q m  = 272.9 mg/g); (2) PDA-mediated nanoparticle immobilization to achieve spatial uniformity and reduce catalyst leaching (< 5 ppb Fe after five cycles); and (3) an optically transparent and mechanically resilient hydrogel framework (94.2 % transmittance at 500 nm, tensile strength: 41.7 kPa, elongation at break: 84.5 %). These integrated features synergistically improved the photo-Fenton degradation rate constant of PCCF by 3.03-fold compared to Fe 3 O 4 nanoparticles, while maintaining 90.4 % catalytic efficiency after five cycles. This work establishes a Pu hydrogel-based structural paradigm that harmonizes catalytic activity with long-term stability, offering a versatile blueprint for high-performance environmental remediation systems.

Multifunctional antibacterial-anti-inflammatory-osteogenic performance of ZIF-8/PDA composite scaffold for osteoporotic bone regeneration via intelligent quercetin delivery

Henigul osman,Xiaohui Tang,Dong Zhang,Bo Liu,Jie Gao,Xin Chen,Tao Jiang,Haobo Pan,Yingbo Wang

To address the dual challenges of controlled drug delivery and microenvironment modulation in repairing osteoporotic bone defects, we developed a multifunctional poly(l-lactic acid) (PLLA)-based composite fibrous scaffold (PLLA/PDA/ZIF-8@Qu). The scaffold was fabricated via electrospinning, followed by in situ polydopamine (PDA) polymerization to robustly immobilize ZIF-8 nanocarriers loaded with quercetin (Qu) on the fiber surface. The mussel-inspired PDA coating served as an adhesive interlayer between ZIF-8 and PLLA. Additionally, it provided near-infrared (NIR) responsiveness and surface functionalization capability. Under osteoporotic acidic conditions (pH 5.0–6.0), the scaffold exhibited pH-triggered release behavior, ZIF-8 degradation accelerated Zn 2+ release, and cumulative Qu release reached 90 %. Zn 2+ contributed to antibacterial activity, while the PDA coating enabled NIR-driven photothermal/photodynamic synergy, achieving 100 % inhibition against both E. coliand S. aureus. Quercetin (Qu) exerted antioxidant effects via the Nrf2/ARE pathway and promoted macrophage polarization toward the M2 phenotype, thereby ameliorating the local inflammatory microenvironment. In vitro , the scaffold enhanced osteogenic differentiation of bone marrow mesenchymal stem cells, as indicated by a 2.3-fold increase in alkaline phosphatase activity, a 1.85-fold increase in mineralized nodule formation, and upregulated Runx2 expression. In a rat femoral defect model, the scaffold group at 8 weeks post-implantation showed increased bone mineral density (BMD) from 0.454 to 0.935 g/cm 3 and trabecular number (Tb.N) from 0.694 to 0.923 mm −1 , with the defect largely filled by newly formed bone. Collectively, these results demonstrate that the PDA-enabled, ZIF-8/Qu-integrated PLLA scaffold couples pH-responsive release with antibacterial, anti-inflammatory, and pro-osteogenic functions, supporting its potential as a bone repair material responsive to pathological microenvironments.

The interface bonding strengthening enhancement of Al-Li alloy powders with dual-layer coating of composite fluorosilane polymers and PDA

Huaqiang Zhu,Shuo Wang,Zhanjun Guo,Hongdi Fu,Xinyi Han,Haiyang Wang,Yonghao Zhao,Haiyue Yu,Yong Yang,Siyuan Tang,Yiheng Li,Yanrui Hu,Tinglu Song,Meishuai Zou

Al-Li alloys are promising fuels for composite solid propellants due to their high energy density and favorable combustion characteristics. However, their application is limited by poor storage stability and inadequate interfacial adhesion. This study presents a novel dual-layer synergistic coating strategy using fluorinated alkylsilane (FAS) and polydopamine (PDA) to improve the interfacial stability of Al-Li alloys and the mechanical performance of Al-Li alloy-based propellants. The Sequential chemical deposition of FAS hydrophobic layer and PDA adhesive layer onto Al-5Li alloy particles was successfully accomplished and uniformly covered. The interfacial adhesion and wettability are increased by introducing catechol groups, which addresses the problems caused by FAS coating. Thermal analysis revealed negligible weight loss (≤0.191 %) from organic coating decomposition, indicating minimal impact on energy density. Mechanical testing showed that the fracture stress of propellants containing Al-5Li@FP particles was significantly improved to 1.593 MPa, and even surpassed those with pure aluminum, which was 1.263 MPa. Combustion experiments further showed a significant enhancement in the burning intensity of the modified alloy powders. This study achieves synergistic improvement of stability and interfacial adhesion in Al-Li alloys via a rationally designed PDA and FAS bilayer interface. Thereby offering an innovative pathway for developing high-performance solid propellants.
