Conventional cancer treatments often result in tissue loss, incomplete tumor excision, infections, and systemic toxicity, severely impacting patient recovery. These complications frequently involve excessive reactive oxygen species (ROS) generation and persistent inflammation, pathological features also characteristic of burn wounds and postoperative tumor sites. To address this, an injectable ROS/pH-responsive hydrogel integrating localized tumor therapy and wound healing is developed. The hydrogel is fabricated via Schiff base crosslinking of a thioketal-based ROS-cleavable linker with aldehyde-functionalized hyaluronic acid, enabling degradation under oxidative and acidic conditions. The hydrogel demonstrates mechanical robustness, excellent biocompatibility, and stimulus-triggered, on-demand drug release. In a breast tumor model, the designed hydrogel facilitates controlled co-delivery of a chemotherapeutic agent 5-fluorouracil (5-FU) and a photosensitizer precursor 5-aminolevulinic acid (ALA), and ensures precise, sustained drug release, achieving synergistic chemo-photodynamic therapy and superior tumor inhibition. Given the pathological similarities between tumor resection wounds and burn injuries, a second-degree burn model is employed to assess the hydrogel's regenerative performance under ROS-rich, inflammation-prone conditions. The hydrogel enables localized antioxidant drug N-acetyl-L-cysteine (NAC) delivery and non-mechanical, on-demand removal. These findings highlight the potential of this multifunctional hydrogel as a minimally invasive, dual-purpose platform for managing cancer-associated tissue injuries and oxidative stress-mediated wound healing.

Abdominal adhesions are a common complication following abdominal surgery, often resulting in inflammation, pain, and dyskinesia, which significantly impair treatment outcomes and hinder patient recovery. Hydrogel-based materials have been extensively investigated to prevent postoperative adhesions due to their excellent biocompatibility, biodegradability, moisture retention, and drug-loading capacity. However, conventional hydrogels still suffer from insufficient anti-inflammatory and anti-adhesion properties, underscoring the need for more advanced biomaterials to address these challenges. In this study, a reactive oxygen species (ROS)-responsive hydrogel (CS-Linker-HA) was developed by crosslinking chitosan (CS) and hyaluronic acid (HA) through an ROS-sensitive linker, serving as a controlled drug delivery system. Additionally, for the first time, honeysuckle-derived extracellular vesicles engineered with puerarin (HEVs-P) were incorporated into the hydrogel (CS-Linker-HA@HEVs-P) to enhance its anti-inflammatory and anti-adhesion properties. The results indicated that this multifunctional hydrogel not only exhibited effectively ROS scavenging ability and anti-swelling properties but also possessed the ROS-triggered controlled release of HEVs-P, the synergism of which would further inhibit postoperative adhesions, promote the healing of abdominal and cecal injury in both murine and rabbit models. Therefore, the therapeutic strategy combined the multifunctional hydrogels with puerarin-engineered honeysuckle extracellular vesicles, providing a promising approach in peritoneal surgery aimed at preventing postoperative adhesions and promoting postoperative recovery.

Sonodynamic therapy (SDT) is a promising noninvasive cancer treatment with deep tissue penetration, but its therapeutic efficacy is limited by insufficient reactive oxygen species (ROS) production. Herein, we developed copper-based sonosensitizer (CCG MPNs) by coordinating copper ions (Cu2+) with chlorin e6 (Ce6) and gallic acid (GA) for dual-amplified ROS-mediated tumor therapy and cuproptosis induction. CCG MPNs dissociate in the high-glutathione (GSH) tumor microenvironment, releasing Cu ions, Ce6, and GA. GA rapidly depletes GSH, enhancing tumor cell sensitivity to ROS and cuproptosis. Additionally, the released Cu+ ions generate cytotoxic hydroxyl radicals via a highly efficient Fenton reaction and ultrasound (US) activation of Ce6 produces singlet oxygen, amplifying ROS-mediated apoptosis by evoke chemodynamic therapy (CDT) and SDT. Moreover, the yielded ROS would cut off the ATP supply, resulting in a decrease in the expression of the copper transporter protein ATP7A. Large accumulations of intracellular Cu+ ions can be more readily induce cuproptosis by promoting lipoylated protein aggregation. In vitro and in vivo anti-tumor studies showed that CCG MPNs group under US treatment exhibited better anti-tumor performance. This multimodal approach overcomes the limitations of the tumor microenvironment, offering a synergistic strategy to enhance tumor therapy efficiency through combined ROS generation and cuproptosis activation.

The increasing prevalence of drug-resistant bacteria and the ecological burden of synthetic antimicrobials demand sustainable biomaterials with superior antibacterial properties. Natural lignin, an amorphous polymer, presents considerable potential as a structural building block for sustainable biomaterial. Herein, inspired by antimicrobial photodynamic inactivation, kraft lignin was first reacted with formaldehyde and ethylenediamine to synthesize aminated lignin, the aminated lignin was further reacted with acetic anhydride, resulting in modified lignin. Thereafter, the modified lignin functions as a carrier, encapsulating the hydrophobic photosensitizer chlorin e6 to improve its dispersibility and aqueous solubility. Chitosan was further utilized to coat the nanoparticles, improving bacterial uptake, ultimately obtaining the desired final nanospheres (CS-ML-Ce6). Upon laser irradiation, CS-ML-Ce6 achieved >99.99 % bactericidal efficiency against methicillin-resistant Staphylococcus aureus . Additionally, it demonstrated a minimum inhibitory concentration of modified lignin of 67.71 μg/mL when exposed to light. It could result in membrane disruption and intracellular component leaking, while bacterial adsorption increased to 47.36 %. The dead/live rate of CS-ML-Ce6 against drug-resistant pathogens was almost 8-fold to modified lignin control. Concurrently, hemolysis assays with rabbit red blood cells confirmed exceptional biocompatibility, with hemolysis rates maintained below 5 % even at 6 × MIC concentrations. Considerable reactive oxygen species scavenging effectiveness at both the solution and cellular levels was also observed of CS-ML- Ce6, this dual-function approach uniquely combines light-activated bactericidal properties and dark-phase antioxidant repair. Utilizing lignin, an industrial byproduct, provides an efficient and promising antimicrobial alternative to prevent overuse of antibiotics and has significant potential for human healthcare.

Alcohol, one of the most widely popular and consumed beverages worldwide, often leads to acute alcoholic liver injury (AALI) when excessively consumed within a short timeframe. It can further induce alcoholic hepatitis, posing a serious risk to human health and public health safety. Pyroptosis, a typical mode of programmed cell death, is closely associated with the body's inflammatory response and plays a central role in AALI. However, the role of yogurt in acute alcohol-induced pyroptosis and inflammation is unclear. In this study, we found that yogurt and its metabolite d -lactate effectively mitigate alcohol-induced liver damage in mice, reducing hepatocyte pyroptosis and attenuating the inflammatory response. For the specific mechanism, further studies have shown that yogurt may inhibit autophagy through its substance d -lactate, leading to the accumulation of P62 protein in cells. This accumulation subsequently triggers the activation of Nrf2 through nonclassical pathways. Nrf2 translocates to the nucleus to play its antioxidant role, thereby reducing intracellular reactive oxygen species levels, inhibiting the activation of the alcohol-induced NLRP3 inflammasome, and ultimately alleviating pyroptosis and inflammation. Overall, this study reveals the potential protective effect of yogurt and its metabolite d -lactate in alleviating AALI, offering new insights and scientific basis for yogurt as a dietary strategy to improve AALI.

Accurate monitoring of ROS levels is essential for oxidative stress (OS) management, but their low concentration and instability pose significant detection challenges. Here, we developed a highly sensitive and selective electrochemical biosensor for real-time, micro-volume ROS detection in leukemia cells. The sensor was fabricated using ultrathin g-C₃N₄-rGO (CN-rGO) nanosheet synthesized through a calcination and hydrothermal approach. These nanosheets were deposited onto a screen-printed carbon electrode (SPE) and further modified with silver nanoparticles (AgNPs) to construct a CN-rGO@AgNPs/SPE biosensor. The porous nanostructure significantly enhanced the catalytic surface area and electron transfer efficiency, achieving high sensitivities towards H₂O₂ and O₂·⁻ detection. Using a window titration platform, the sensor detected ROS in 0.1 mL lysates of leukemia cell lines (U-937, THP-1, and MOLM-13) with concentrations of 7.38 ± 4.81 μM, 11.74 ± 0.985 μM, 19.84 ± 3.73 μM, respectively. Furthermore, the platform allowed precise monitoring of doxorubicin (DOX)-induced OS, providing valuable insights for guiding leukemia chemotherapy. Additionally, Lipopolysaccharide (LPS) stimulation experiments demonstrated the sensor's exceptional real-time performance, enabling dynamic detection of H₂O₂ release kinetics and ROS generation rates at the cellular level. Combining high sensitivity, low detection limits, and micro-volume analysis capabilities, this sensor holds great potential for clinical auxiliary diagnosis and drug efficacy assessment in leukemia research.

Iron-gallic acid chelate nanoparticles (Fe-GA NPs) have emerged as promising Fenton catalysts and drug carriers in oncology. However, their therapeutic efficacy remains constrained by tumor microenvironment (TME) limitations-- suboptimal pH and insufficient endogenous hydrogen peroxide. To overcome these barriers, we engineered an ATP-responsive core-shell nanoarchitecture (GOx@Fe-GA) integrating glucose oxidase (GOx) with Fe-GA coordination networks. Upon encountering elevated ATP concentrations in tumor cells, the nanosystem undergoes programmed disassembly: released GOx depletes glucose to induce metabolic starvation while generating substantial H2O2 and acidifying the TME, thereby creating ideal conditions for Fe-GA-mediated Fenton reactions. Simultaneously, Fe-GA acts as a photothermal agent under near-infrared irradiation, achieving mild hyperthermia that synergizes with reactive oxygen species (ROS) to overcome thermotolerance by disrupting heat shock protein (HSP70) defenses. In vitro and in vivo studies demonstrated potent tumor suppression with minimal systemic toxicity, establishing GOx@Fe-GA as a self-enhancing therapeutic platform where tumor-specific ATP triggers cascading therapeutic amplification through ROS storm, metabolic deprivation, and photothermal sensitization.

Reactive oxygen species (ROS) are critical mediators of soil biogeochemical processes. While the production of ROS with biochar (BC) in the rhizosphere has not been explored. We demonstrate that BC and Fe-modified biochar (FeBC), prepared at 400°C and 600°C, influence ROS generation in paddy soil containing biodegradable (polybutylene succinate: PBS) and conventional (polystyrene) microplastics (MPs). FeBC, particularly FeBC prepared at 600°C, enhance ROS (O 2 •− , H 2 O 2 , • OH) production in paddy soil. The results of rhizosphere-focused pot experiment simulating field-like flooded conditions highlight that FeBC significantly increased O 2 •− , H 2 O 2 , • OH concentrations, especially with PBS (25.63, 27.69, 35.82 µM/kg, respectively) surpassing control by 3.1–3.6-folds. Iron plaque formation, water-soluble phenols, and microbial activity further influenced ROS dynamics and accelerated MP degradation via oxidative fragmentation (SEM surface fissures), dissolved organic carbon release (484 mg/kg), and SUVA 254 decline (4.6 folds). Microbial shifts toward Actinobacteria (37–43 %) and Firmicutes (25–28 %) correlated with ROS dynamics, linking biotic-abiotic degradation pathways. This work reveals the novel role of FeBC in rhizosphere ROS generation and the environmental behavior of MPs in BC-treated agricultural systems.

Reactive oxygen species (ROS), generated by sonosensitizers, play a pivotal role in tumor cell apoptosis during sonodynamic therapy (SDT), particularly for tumors located deep within tissues. Nevertheless, conventional sonosensitizers present limitations including inadequate ROS generation, insufficient tumor-specific accumulation, and associated adverse effects, significantly restricting their clinical applicability. To address these limitations, novel drug-free multifunctional nanoparticles (designated HGMP NPs) were synthesized. These NPs consist of mesoporous polydopamine (MPDA)-loaded protoporphyrin IX (PpIX), further surface-modified with glucose oxidase (GOx) and hyaluronic acid (HA), to achieve integrated photothermal, sonodynamic, and starvation-based tumor therapy. Upon exposure to near-infrared (NIR) irradiation (808 nm) combined with ultrasound (US), HGMP NPs exhibited pronounced synergistic anticancer effects. Specifically, the photothermal effect triggered by NIR irradiation effectively enhanced local oxygen supply within tumor sites, thus significantly augmenting ROS production and improving the therapeutic outcomes of SDT. Concurrently, GOx-mediated glucose depletion induced tumor starvation and produced hydrogen peroxide (H 2 O 2 ), further exacerbating oxidative stress within the tumor microenvironment. Transcriptomic analysis revealed that ROS and TNF signaling pathways represented key mechanisms underlying tumor elimination by this multimodal synergistic strategy. Real-time PCR analysis and ELISA assays further validated activation of the TNF signaling pathway. Importantly, this study first confirmed the high biocompatibility and biosafety of HGMP NPs via serum metabolomics, demonstrating no detectable systemic metabolic perturbations. Collectively, the prepared HGMP NPs provide a rational paradigm for synergistic anticancer therapy. These findings highlight the potential of HGMP NPs as an exceptionally safe and effective nanoplatform for cancer treatment, offering valuable insights into future developments in cancer nanomedicine. Mesenchymal stem cell (MSC)-based cell sheet engineering holds great promise for periodontal tissue engineering and restoring damaged tissues. However, achieving optimal regenerative outcomes remains a challenge. This study introduces a novel approach that integrates melatonin-coated nanofiber fragments (mNFs) into dental follicle stem cell sheets (mNF-CSs) to enhance their regenerative potential. The mNF-CSs were engineered using electrospun poly-L-lactic acid (PLLA) nanofiber sheets fragmented into mNFs and coated with melatonin, a potent antioxidant and osteogenic agent. The mNF-CSs exhibited a homogenous morphology with sustained melatonin release over 21 days, providing a prolonged antioxidative effect critical for tissue regeneration. In vitro experiments demonstrated that mNF-CSs significantly reduced intracellular reactive oxygen species (ROS) levels and upregulated antioxidative proteins such as SOD1 and SOD2, resulting in improved cell viability, maintenance of stemness, and enhanced osteogenic differentiation potential of dental follicle stem cells (DFCs). In vivo studies using both a nude mouse and a rat periodontal defect model demonstrated that mNF-CSs promoted alveolar bone repair and periodontal tissue regeneration, with enhanced retention of MSCs at the defect site and increased expression of periodontal tissue-related markers. Transcriptome analysis revealed the upregulation of genes associated with biomineralization, odontogenesis, and autophagy pathways in mNF-CSs, suggesting their ability to modulate crucial cellular processes for tissue regeneration. Overall, mNF-CSs offer a promising strategy for enhancing periodontal tissue engineering applications, providing a platform for improving bone grafts and regenerative medicine strategies in the context of periodontal diseases.

The acute lung injury (ALI) is an inflammatory disorder associated with cytokine storm, which activates various reactive oxygen species (ROS) signaling pathways and causes severe complications in patients as currently seen in coronavirus disease 2019 (COVID-19). There is an urgent need for medication of the inflammatory lung environment and effective delivery of drugs to lung to reduce the burden of high doses of medications and attenuate inflammatory cells and pathways. Herein, we prepared dexamethasone-loaded ROS-responsive polymer nanoparticles ( [email protected] NPs) by a modified emulsion approach, which achieved high loading content of DEX (11.61 %). DEX was released faster from the [email protected] NPs in a ROS environment, which could scavenge excessive ROS efficiently both in vitro and in vivo . The PFTU NPs and [email protected] NPs showed no hemolysis and cytotoxicity. Free DEX, PFTU NPs and [email protected] NPs shifted M1 macrophages to M2 macrophages in RAW264.7 cells, and showed anti-inflammatory modulation to A549 cells in vitro . The [email protected] NPs treatment significantly reduced the increased total protein concentration in BALF of ALI mice. The delivery of [email protected] NPs decreased the proportion of neutrophils significantly, mitigated the cell apoptosis remarkably compared to the other groups, reduced M1 macrophages and increased M2 macrophages in vivo . Moreover, the [email protected] NPs had the strongest ability to suppress the expression of NLRP3, Caspase1, and IL-1β. Therefore, the [email protected] NPs could efficiently suppress inflammatory cells, ROS signaling pathways, and cell apoptosis to ameliorate LPS-induced ALI. Statement of Significance The acute lung injury (ALI) is an inflammatory disorder associated with cytokine storm, which activates various reactive oxygen species (ROS) signaling pathways and causes severe complications in patients. There is an urgent need for medication of the inflammatory lung environment and effective delivery of drugs to modulate the inflammatory disorder and suppress the expression of ROS and inflammatory cytokines. The inhaled [email protected] NPs prepared through a modified nanoemulsification method suppressed the activation of NLRP3, induced the polarization of macrophage phenotype from M1 to M2, and thereby reduced the neutrophil infiltration, inhibited the release of proteins and inflammatory mediators, and thus decreased the acute lung injury in vivo .
