The biochemical composition of sweat is closely related to the human physiological state, which provides a favorable window for the monitoring of human health status, especially for the athlete. Herein, an ultra-simple strategy based on the surface-enhanced Raman scattering (SERS) technique for sweat analysis is established. Metal–phenolic network (MPN), an outstanding organic-inorganic hybrid material, is adopted as the reductant and platform for the in situ formation of Au-MPN, which displays excellent SERS activity with the limit of detection to 10−15M for 4-mercaptobenzoic acid (4-MBA). As an ultrasensitive SERS sensor, Au-MPN is capable of discriminating the molecular fingerprints of sweat components acquired from a volunteer after exercise, such as urea, uric acid, lactic acid, and amino acid. For pH sensing, Au-MPN/4-MBA efficiently presents the pH values of the volunteer’s sweat, which can indicate the electrolyte metabolism during exercise. This MPN-based SERS sensing strategy unlocks a new route for the real-time physiological monitoring of human health.Keywords:surface-enhanced Raman scattering;metal–phenolic network;sweat;molecular fingerprint;pH

Hyaluronidase expression is known to be upregulated in various pathological conditions. A method based on a combination of ratiometric surface-enhanced Raman scattering (SERS) and magnetic separation is described for the determination of hyaluronidase (HAase) activity. Gold nanospheres (AuNPs) functionalized by 4-mercaptophenylboronic acid (4-MPBA) form stable cyclic esters with diol on hyaluronic acid (HA) by the boronic acid group, while Fe 3 O 4 @ [email protected] modified with mercaptoethylamine (MEA) was used as a capture substrate to bind to the carboxyl group on the surface of HA, forming the “ [email protected] @HA/Fe 3 O 4 @ [email protected] @MEA” “core-satellite” structure. When HAase is present, HA is enzymatically disrupted, resulting in the destruction of the “core-satellite” structure, the SERS signal of 4-MPBA is subsequently weakened. The gold shell in the substrate protects the 5,5′-Dithio bis-(2-nitrobenzoic acid) (DTNB) from the external environment, which makes it become an ideal internal standard (IS) molecule for subsequent calibration. Under optimal conditions, the I 1075 /I 1324 varied in the range of 10 −3 - 10 U‧mL −1 HAase activity, with a limit of detection (LOD) of 0.32 mU‧mL −1 ,below the level of HAase in normal human body fluids. This method has been successfully applied to the determination of HAase activity in urine and is expected to provide a new method in disease detection, especially in the non-invasive detection of bladder cancer.

Multiplexed detection of Parkinson’s disease (PD) biomarkers is of great significance for early diagnosis and personalized treatment. In this study, we fabricated a robust surface-enhanced Raman scattering-enabled lab-on-a-chip (LoC-SERS) platform for simultaneous quantification of α-synuclein, phosphorylated tau protein 181, osteopontin, and osteocalcin. Herein, the antibody–DNA conjugate was designed to introduce the catalytic hairpin self-assembly (CHA) amplification into the protein detection. Au nano-stars (AuNSs) modified with Raman reporter molecules and hairpin-structure DNA 1 were applied as the SERS nanotags. Au-coated silicon nanocone array (Au/SiNCA) fabricated based on the maskless plasma etching-prepared high-density Si nanocone array (SiNCA) and surface ion sputtering was used as the capture substrate after the modification of hairpin-structure DNA 2. Benefitting from the antibody–DNA conjugate-induced CHA amplification, numerous AuNSs can be connected to the Au/SiNCA surface, which significantly amplify the plasmonic coupling effect for ultrasensitive SERS detection, and the limit of detection was less than the pg/mL level. The application of highly uniform Au/SiNCA and antibody–DNA conjugate endows the LoC-SERS platform excellent analytical performance, including superior reproducibility, satisfactory universality, and high sensitivity. In addition, a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice model was established, and satisfactory results were obtained in real sample analysis with the LoC-SERS platform, which may be enlightening for exploiting protein biomarkers in PD monitoring.

Acetamiprid poses a serious threat to human beings. In this work, the highly transparent fluorescent quantitative PCR sealing membranes (PCR-M) developed in our previous work which had attractive characteristics such as sensitivity, stability and reproducibility, and its capability of extracting analytes quickly, was used to detect acetamiprid. The detection limit of 1 nmol/L was obtained by direct detection of CN group in acetamiprid. In order to improve the selectivity and sensitivity, acetamiprid aptamer was introduced. After the interaction of aptamer with acetamiprid the SERS signal of adenine at 730 cm −1 increased. The detection limit of indirect detection was 10 -8 mol/L. Aptamer advanced Ag-PCR-M-based Surface enhanced Raman spectroscopy (SERS) method for indirect SERS detection which significantly improves the detection sensitivity and greatly increases the range of signal response value. This will be conducive to the detection of actual samples and expand the practical application.

Paraquat is a highly effective herbicide and widely used in agricultural production. However, paraquat residue is harmful for human health and can cause irreversible hazard. Thus, it is crucial for monitoring of paraquat residues. In this paper, an efficient SERS platform based on cactus-inspired nanoparticles is proposed for sensitive detection of paraquat. The cactus-liked nanoparticles obtained from one-pot stepwise reduction method possess multiple spiny structures and can produce abundant hot spots, resulting in remarkable SERS performance. SEM, TEM, UV–vis and Raman tests were conducted to characterize and optimize the morphology of cactus-liked nanoparticles under different preparation conditions. The synthesis mechanism and corresponding parameters influence mechanism of cactus-liked nanoparticles were explored in detail. Optimized substrate exhibited a high sensitivity with the detectable concentration of crystal violet (CV) down to 10 -9 M and an excellent reproducibility proved by SERS mapping. Furthermore, it behaved good linear relationship with a correlation coefficient (R 2 ) of 96.89% between Raman intensities and concentrations of paraquat, which indicates the SERS substrate prepared with cactus-liked nanoparticles could offer a great potential for identification of paraquat.

The focusing field effect excited by the cavity mode has a positive coupling effect with the metal localized surface plasmon resonance (LSPR) effect, which can stimulate a stronger local electromagnetic field. Therefore, we combined the self-organizing process for component and array manufacturing with imprinting technology to construct a cheap and reproducible flexible polyvinyl alcohol (PVA) nanocavity array decorating with the silver nanoparticles (Ag NPs). The distribution of the local electromagnetic field was simulated theoretically, and the surface-enhanced Raman scattering (SERS) performance of the substrate was evaluated experimentally. The substrate shows excellent mechanical stability in bending experiments. It was proved theoretically and experimentally that the substrate still provides a stable signal when the excited light is incident from different angles. This flexible substrate can achieve low-cost, highly sensitive, uniform and conducive SERS detection, especially in situ detection, which shows a promising application prospect in food safety and biomedicine.Keywords:surface-enhanced Raman spectroscopy;localized surface plasmon resonance;flexible SERS substrate;cavity

Ofloxacin is one kind of quinolone antibiotic drugs, the abuse of ofloxacin in livestock and aquaculture may bring bacterial resistance and healthy problem of people. The illegally feeding cattle with ofloxacin will help it keep health, but the sedimentation of ofloxacin could bring problem in food safety. The accurate, simple and instant monitoring ofloxacin from beef by portable sensor was of vital issue in food quality. A simple and reliable method was proposed for instant and quantitative detecting ofloxacin in beef, in which the thin-layer chromatography (TLC) -surface-enhanced Raman scattering (SERS) spectroscopy was in tandem with machine learning analysis base one principal component analysis-back propagation neural network (PCA-BPNN). The TLC plate was composed with diatomite, that was function as the stationary phase to separate ofloxacin from beef. The real beef juice was directly casted onto the diatomite plate for separating and detecting. The directly monitor ofloxacin from beef was achieved and the sensitivity down to 0.01 ppm. The PCA-BPNN was used as reliable model for quantitative predict the concentration of ofloxacin, that shown superior accuracy compared with the traditional model. The results verify that the diatomite plate TLC-SERS combined with machine-learning analysis is an effective, simple and accurate technique for detecting and quantifying antibiotic drug in meat stuff to improve the food safety.

Colorectal cancer (CRC) remains a significant contributor to the global mortality rate, and a single biomarker cannot meet the specificity required for CRC screening. To this end, we developed a multiplexed, pump-free surface-enhanced Raman scattering (SERS) microfluidic chip (LoC-SERS) using a one-step recognition release mechanism; the aptamer-functionalized novel Au nanocrown array (AuNCA) was used as the detection element embedded in the detection zone of the platform for rapid and specific detection of protein markers in multiple samples simultaneously. Here, the corresponding aptamer specifically captured the protein marker, causing the complementary strand of the aptamer carrying the Raman signal molecule to be shed, reducing the SERS signal. Based on this platform, sensitive and specific detection of the target can be accomplished within 15 min with detection limits of 0.031 pg/mL (hnRNP A1) and 0.057 pg/mL (S100P). Meanwhile, the platform was consistent with ELISA results when used to test clinical. By substituting different aptamers, this platform can provide a new solution for the rapid and sensitive detection of protein markers, which has promising applications in future disease detection.

A dynamic ratiometric in-tube SPME-SERS platform was built for thiram detection. • Forced convection shortened adsorption equilibrium from ∼90 min to ∼300 s. • SCN − internal standard enabled ratiometric correction with RSD < 10%. • Direct analysis of environmental water achieved recoveries of 90.9%–108.5%. Surface-enhanced Raman spectroscopy (SERS) is a powerful tool for pesticide detection but is hindered in field applications by slow analyte mass transport and quantitative irreproducibility. Herein, we report an in-tube solid-phase microextraction-SERS (in-tube SPME-SERS) strategy that simultaneously overcomes these bottlenecks. A silver-nanoplate-decorated copper wire was synthesized via galvanic replacement, functionalized with thiocyanate (SCN − ) as a covalently bonded internal standard (IS), and assembled into a fused-silica capillary. Hydrodynamic flow compresses the diffusion boundary layer, accelerating the extraction equilibrium from ∼90 min (static mode) to ∼300 s. The SCN − internal standard (2136 cm −1 ) enables ratiometric correction ( I 1380 /I 2136 ) for thiram quantification, effectively compensating for substrate heterogeneity and instrumental drift. Mechanistic studies reveal a competitive adsorption process where thiram partially displaces SCN − , ensuring precise analyte-IS co-localization. This method exhibits excellent analytical performance for thiram, including a wide linear range (20–1000 nM), a low limit of detection (33.8 nM), and high reproducibility (RSD < 10%). Furthermore, direct analysis of surface and groundwater samples without pretreatment yields satisfactory recoveries (90.9%–108.5%). With robust 28-day stability, this ratiometric in-tube SPME-SERS platform provides a reliable, kit-based solution for on-site environmental monitoring. Hydrogen peroxide (H 2 O 2 ) residue in foodstuffs will bring great harm to human health. We immobilize the composite of the reduced polyaniline (PANI R ) modified gold nanoparticles on the surface of ITO (ITO/AuNPs/PANI R ) to develop surface-enhanced Raman scattering (SERS) sensor for H 2 O 2. detection. The principle is that PANI R is oxidized by H 2 O 2 to generate a new SERS peak at 1460 cm −1 for realizing quantitative analysis of H 2 O 2 . Fe 2+ -Fenton reaction is introduced to catalytically react with H 2 O 2 to hydroxyl radical, which speeds up the oxidation of PANI R . Before SERS detection, acidic treatment could guarantee the reduced state of PANI R in composite. Limit of detection of ITO/AuNPs/PANI R -based SERS assay for H 2 O 2 is down to 1.78 × 10 −12 mol/L and a good linear relationship from 1 × 10 −10 to 3.16 × 10 −7 mol/L is achieved. Furthermore, the SERS sensor could be regenerated by acidic treatment. As a scenario, the renewable SERS sensor is utilized to monitor H 2 O 2 residues in food and environmental samples.

The templating method holds great promise for fabricating surface nanopatterns. To enhance the manufacturing capabilities of complex surface nanopatterns, it is important to explore new modes of the templates beyond their conventional masking and molding modes. Here, we employed the metal-organic framework (MOF) microparticles assembled monolayer films as templates for metal electrodeposition and revealed a previously unidentified guiding growth mode enabling the precise growth of metallic films exclusively underneath the MOF microparticles. The guiding growth mode was induced by the fast ion transportation within the nanochannels of the MOF templates. The MOF template could be repeatedly used, allowing for the creation of identical metallic surface nanopatterns for multiple times on different substrates. The MOF template-guided electrochemical growth mode provided a robust route towards cost-effective fabrication of complex metallic surface nanopatterns with promising applications in metamaterials, plasmonics, and surface-enhanced Raman spectroscopy (SERS) sensing fields.

Surface-enhanced Raman scattering (SERS) is widely employed because it offers quick, microscopic, and traceless detection. This study used high-voltage and low-voltage ultrasonic oscillation to embed gold-silver nanoparticles (Au-Ag NPs) into the pores of chemically modified ultrathin anodic alumina (AAO) films, resulting in a highly sensitive three-dimensional SERS substrate. We improved the substrate’s stability and Raman activity by adjusting the particle alloy ratio. For the substrate in this alloy ratio, the Raman signal of probe molecules (Rh6G) adsorbed on the substrate surface is enhanced. the enhancement factor (EF) was as high as 1.40 × 10 7 , the relative standard deviation (RSD) of 10.6%, and the concentration of Rh6G shows a linear relationship with Raman intensity, with a linear correlation coefficient of 0.961. In addition, we evaluated the substrate’s detection effect on thiram molecules. It has been proven that this structure has good practicality and high sensitivity as a Raman enhanced substrate.
