Biomolecular insights into the inhibition of heavy metals on reductive dechlorination of 2,4,6-trichlorophenol in Pseudomonas sp. CP-1

Di Cao,Xueqi Chen,Jun Nan,Aijie Wang,Zhiling Li

Influences of heavy metal exposure to the organohalide respiration process and the related molecular mechanism remain poorly understood. In this study, a non-obligate organohalide respiring bacterium, Pseudomonas sp. strain CP-1, was isolated and its molecular response to the five types of commonly existed heavy metal ions were thoroughly investigated. All types of heavy metal ions posed inhibitory effects on 2,4,6-trichlorophenol dechlorination activity and cell growth with the varied degree. Exposure to Cu (II) showed the most serious inhibitive effects on dechlorination even at the lowest concentration of 0.05 mg/L, while the inhibition by As (V) was the least with the removal kinetic constant k decreased to 0.05 under 50 mg/L. Further, multi-omics analysis found compared with Cu (II), As (V) exposure led to the insignificant downregulation of a variety of biosynthesis processes, which would be one possible account for the less inhibited activity. More importantly, the inhibited mechanisms on the organohalide respiration catabolism of strain CP-1 were firstly revealed. Cu (II) stress severely downregulated NADH generation during TCA cycle and electron donation of organohalide respiration process, which might decrease the reducing power required for organohalide respiration. While both Cu (II) and As (Ⅴ) inhibited substrate level phosphorylation during TCA cycle, as well as electron transfer and ATP generation during organohalide respiration. Meanwhile, CprA-2 was confirmed as the responsible reductive dehalogenase in charge of 2,4,6-TCP dechlorination, and transcriptional and proteomic studies confirmed the directly inhibited gene transcription and expression of CprA-2. The in-depth reveal of inhibitory effects and mechanism gave theoretical supports for alleviating heavy metal inhibition on organohalide respiration activity in groundwater co-contaminated with organohalides and heavy metals.

Impact of Natural Microorganisms on the Removal of COD and the Cells Activity of the Chlorella sp. in Wastewater

Qingnan Sun,Xiaoping Zhang,Xin Zhang

In the treatment of wastewater containing only chemical oxygen demand (COD) byChlorellasp., the cell activity and proliferation ability ofChlorellasp. decreased with the culture time, which affected the removal of COD in wastewater. To solve these problems, theChlorellasp.–natural microorganism symbiosis system was prepared. The system was used to explore how natural microorganisms affect the cell activity and the proliferation ability ofChlorellasp. in wastewater. In the treatment of COD byChlorellasp., the removal rate of COD decreased from 45.47% to 28.88%, with a decrease in the cell activity and proliferation ability ofChlorellasp. In theChlorellasp.–natural microorganism symbiotic system, the removal rate of COD reached 45.75%. With the introduction of natural microorganisms, the circulation of CO2and O2betweenChlorellasp. and natural microorganisms promoted photosynthesis and respiration, which enhanced the cell activity ofChlorellasp. Under the condition that the dosage of natural microorganisms was between 1% and 6%, the concentration ofChlorellasp. was close to the logarithmic growth phase, which maintained the proliferation ability ofChlorellasp. At the same time, the natural microorganisms grew and proliferated in wastewater containing only COD through preying onChlorellasp.Keywords:Chlorellasp.;natural microorganisms;symbiosis system;cell activity;predation

Degradation and detoxification of azo dyes with recombinant ligninolytic enzymes from Aspergillus sp. with secretory overexpression in Pichia pastoris

Siqi Liu,Xiaolin Xu,Yanshun Kang,Yingtian Xiao,Huan Liu

Ligninolytic enzymes, including laccase (Lac), manganese peroxidase (MnP) and lignin peroxidase (LiP), have attracted much attention in the degradation of contaminants. Genes of Lac (1827 bp), MnP (1134 bp) and LiP (1119 bp) were cloned from Aspergillus sp. TS-A, and the recombinant Lac (69 kDa), MnP (45 kDa) and LiP (35 kDa) were secretory expressed in Pichia pastoris GS115, with enzyme activities of 34, 135.12 and 103.13 U l−1, respectively. Dyes of different structures were treated via the recombinant ligninolytic enzymes under the optimal degradation conditions, and the result showed that the decolourization rate of Lac on Congo red (CR) in 5 s was 45.5%. Fourier-transform infrared spectroscopy, gas chromatography–mass spectrometry analysis and toxicity tests further proved that the ligninolytic enzymes could destroy the dyes, both those with one or more azo bonds, and the degradation products were non-toxic. Moreover, the combined ligninolytic enzymes could degrade CR more completely compared with the individual enzyme. Remarkably, besides azo dyes, ligninolytic enzymes could also degrade triphenylmethane and anthracene dyes. This suggests that ligninolytic enzymes from Aspergillus sp. TS-A have the potential for application in the treatment of contaminants.

Effect of Cholinium Amino Acids Ionic Liquids As Cosolvents on the Bioconversion of Phytosterols by Mycobacterium sp. Resting Cells

Xia Xiao,Jing-Kai He,Yi-Xin Guan,Shan-Jing Yao

Androst-4-ene-3,17-dione (AD) is an important intermediate of steroid drugs, and the microbial side-chain cleavage of sterols to produce AD by Mycobacterium sp. has attracted much attention. Unfortunately, a low yield of this reaction due to the extremely poor solubility of sterols in aqueous medium is a critical problem. Ionic liquid (IL) as a promising alternative to organic solvents is deemed to increase the solubility of hydrophobic substrates; however, the tolerance of microorganisms to ILs is limited and biocompatible ILs are desperately needed. In this work, cholinium amino acids ([Ch][AA]) ILs were first introduced as cosolvents for the whole-cell bioconversion of phytosterols to produce AD. The physicochemical properties of these novel ILs were investigated on the basis of their structures, and the solubility of phytosterols and AD in [Ch][AA] ILs aqueous solution was determined. The results indicated that moderate cell membrane permeability was obtained by [Ch][AA] ILs, and [Ch][Asp], [Ch][Glu], and [Ch][Gln] showed proper biocompatibility to Mycobacterium sp. MB 3683 cells among 18 kinds of [Ch][AA] ILs. The highest production of AD was achieved in 1% (v/v) [Ch][Asp], which was about twice as high as that in the buffer. Therefore, [Ch][AA] ILs are potential cosolvents in steroid biotransformation to promote medium engineering for a sustainable environment.

A sustainable solution to plastics pollution: An eco-friendly bioplastic film production from high-salt contained Spirulina sp. residues

Chaofan Zhang,Chengyu Wang,Guoliang Cao,Dawei Wang,Shih-Hsin Ho

Plastic products have become a major contaminant in environmental ecology due to their recalcitrant biodegradation, poor management and risky disposal. Therefore, much research attention has been paid to developing the biodegradable bio-based plastics. However, many of the substitute bioplastics derived from agricultural materials may present a potential threat to food security and eco-systems. Herein, we propose a sustainable, eco-friendly and simple procedure to convert the hazardous high-salt contained microalgal residues into bioplastic film. With 35 % poly (vinyl alcohol) (PVA) assistance, the composite bioplastic films achieved 22 MPa tensile strength under alkali condition and 77 % elongation at break under acidic condition. The average maximum contact angle of 94.4° confirmed a desirable water resistance potential. The synthesis mechanism demonstrated that the inorganic salts existed in microalgal residues could act as the filler in shape of sheets under alkali condition or as the cross linker under acidic condition, significantly enhancing the practical feasibility. This work demonstrates a promising biodegradable bioplastics formed from sustainable eco-friendly waste reutilization process, providing a new insight for fundamentally reducing the plastics pollution.

Impact of UV irradiation on Chlorella sp. damage and disinfection byproducts formation during subsequent chlorination of algal organic matter

Feilong Dong,Qiufeng Lin,Jing Deng,Tuqiao Zhang,Cong Li,Xuedong Zai

The frequent occurrence of algal blooms in surface water has attracted more and more attention, which caused many water quality problems, including disinfection byproducts (DBPs). Algal organic matter (AOM) including intracellular organic matter (IOM) and extracellular organic matter (EOM), was a well-known precursor to DBPs formation in drinking water. This study evaluated the effect of ultraviolet (UV) irradiation on the cell integrity, IOM release and DBPs formation during subsequent chlorination of Chlorella sp. Results showed the damage rates of algal cells increased to 40.1% after the high UV irradiation of 528 mJ/cm 2 , which contributed to the release of IOM. In addition, UV irradiation was effective in reducing the formation of haloacetic acids (HAAs) both in AOM and IOM, but promoted the formation of nitrogenous DBPs (N-DBPs) from AOM in subsequent chlorination. Furthermore, neutral pH exerted a positive effect on the formation of DBPs. UV irradiation decreased the bromine substitution factor (BSF) value of AOM at a high bromide level. The BSF values increased with increasing of the concentration of bromide. Moreover, more amino acids and low molecular weight precursors were produced after UV irradiation in filtered supernatant, which contributed to the formation of N-DBPs with algal chlorination. Overall, this information demonstrated pre-oxidation of UV irradiation could be used to treat the algal-rich drinking water.

Toxic effects and metabolic fate of carbamazepine in diatom Navicula sp. as influenced by humic acid and nitrogen species

Tengda Ding,Kunde Lin,Bo Yang,Menting Yang,Juying Li

Carbamazepine (CAB) is a commonly detected pharmaceutical in the surface waters. In the present study, we evaluated the effect of humic acid (HA) and nitrogen species on the toxicity of CAB in a typical diatom Navicula sp., and its metabolic fate. The Navicula sp. showed a high sensitivity to CAB (72 h EC 50 = 0.179 mg L −1 ). The addition of HA (< 30 mg L −1 ) and ammonium could promote the growth of Navicula sp. during 6 d of exposure to CAB. CAB underwent rapid dissipation in the algal culture. The degradation of CAB in Navicula sp. cultures was significantly increased by the addition of HA and higher HA contents caused more degradation of CAB. Degradation and accumulation of CAB in Navicula sp. were significantly inhibited by ammonium. Five metabolites were found and possible degradation pathways of CAB in the algal cell were proposed. Methoxylation, demethylation, glucuronidation and hydroxylation contributed to CAB transformation in the algal cell.

Enantioselective Resolution of (±)-1-Phenylethyl Acetate by Using the Whole Cells of Deep-sea Bacterium Bacillus sp. DL-2

Dong Lu,Xu Yongkai,Zhang Yun,Sun Aijun,Hu Yunfeng

Bacillus sp. DL-2 was isolated from the deep sea of the Western Pacific and further utilized as novel biocatalysts to efficiently asymmetrically hydrolyze (±)-1-phenylethyl acetate. After the optimization of hydrolytic reactions, chiral chemicals ( R )-1-phenylethanol and ( S )-1-phenylethyl acetate were obtained with high optical purities (96% and 99.8%, respectively). Our research is about the asymmeric hydrolysis of (±)-1-phenylethyl acetate using whole-cell biocatalysts. In addition, the optical purity of ( S )-1-phenylethyl acetate generated through the kinetic resolution of (±)-1-phenylethyl acetate using the whole-cells of Bacillus sp. DL-2 was the highest report so far. Using the whole cells of deep sea bacterium Bacillus sp. DL-2 as the biocatalysts is an environmentally friendly method and will play critical roles in industrial asymmetric synthesis.

Soil inoculation with Burkholderia sp. LD-11 has positive effect on water-use efficiency in inbred lines of maize

Fan Xianwei,Hu Haiyang,Huang Guiyuan,Huang Feiyan,Li Youzhi,Palta Jairo

Background and aims Plant-growth promoting rhizobacteria (PGPR) can promote plant performance under water deficit, but the physiology and biochemistry of the promoting process induced by PGPR under different water deficits is not well known in maize ( Zea mays L). Methods A glasshouse study was conducted to determine the effects of Burkholderia sp. LD-11 on morphophysiological traits for plant growth and homeostasis between reactive oxygen species (ROS) and antioxidant enzymes under five regimens in two maize inbred lines. Results Soil inoculation with Burkholderia sp. LD-11 promoted biomass accumulation and improved instantaneous water-use efficiency (WUEi), regardless of the soil water availability. It also triggered production of indole-3-acetic acid (IAA), decreasing the accumulation of abscisic acid (ABA) induced by the water deficit, alleviated ROS accumulation, and resulted in a reduction in lipid peroxidation induced by the water deficits. Soil inoculation also enhanced the tolerance to water deficit through reducing stomatal aperture by increasing the sensitivity of stomatal conductance ( g s ) to small changes in ABA concentration in the leaves. Conclusions Soil inoculation with Burkholderia sp. LD-11 enhanced root systems and WUEi, offering a potential avenue for improving maize tolerance to water deficit.

Characterization of extracellular chitinase from Chitinibacter sp. GC72 and its application in GlcNAc production from crayfish shell enzymatic degradation

Cong Gao,Alei Zhang,Kequan Chen,Zhikui Hao,Junmao Tong,Pingkai Ouyang

In this study, a novel chitinase-producing bacterium Chitinibacter sp. GC72 was isolated and investigated for N -acetyl-D-glucosamine production from crayfish shell enzymatic degradation. A dimeric chitinase was purified, which had an optimal activity at a pH of 6.8 and 40   °C. The metal ions Al 3+ , Cu 2+ , and Zn 2+ inhibited the chitinase activity, whereas Ca 2+ , Mn 2+ , and Mg 2+ promoted the activity. The chitinase was active on p -NP-GlcNAc with apparent K m value of 152.83   μmol/L and V m value of 49.12   μmol/L   min at 37   °C. Based on the hydrolysate formed, the chitinase was characterized as an exo-hydrolytic N -acetyl glucosaminidase. Furthermore, combined treatment of ultra-micro grinding and ultrasonic with direct enzymatic hydrolysis of crayfish shell for GlcNAc production was investigated, which resulted in 15.2   g of GlcNAc production from 100   g of crayfish shell following chitinase degradation for 36   h.

Steam explosion-ionic liquid pretreatments on wetland lignocellulosic biomasses of Phragmites (sp.) and Thalia dealbata for Bio H2 conversion

Hongyun Peng,Lingling Gao,Mengjiao Li,Yibin Shen,Qiongqiu Qian,Xia Li

Bio H2 conversion from wetland lignocellulosic biomass is one of the promising alternatives to fossil fuels. Both Phragmites (sp.) and Thalia dealbata are holocellulose-rich and lignin-rich wetland plant biomasses. Scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) demonstrated alteration of lignocellulosic structures due to hemicelluloses removal by steam explosion (SE) pretreatment, and further disruption of cellulose crystallinity after treatment with SE followed by [Bmim]Cl (SE-[Bmim]Cl). Thermogravimetry/differential thermogravimetry (TG/DTG) revealed an increase in amorphous cellulose and partial delignification in lignocellulosic structures as a consequence of SE-[Bmim]Cl. The pretreatment of both SE and SE-[Bmim]Cl yielded lignocellulosic substrates with improved properties in terms of their conversion into glucose and bio H2. Five-to-ten folds (by SE) and ten-to-twenty folds (by SE-[Bmim]Cl) more glucose was released from the lignocellulosic substrates of both plants than those of contrast samples. Compared to Phragmites (sp.), the greater destruction in lignocellulosic structure of Thalia dealbata as a result of SE and SE-[Bmim]Cl, increased the accessible surface area and disrupted the cellulose crystallinity much more, thus making it more efficient for bio H2 conversion. The bio H2 of 1.97 ± 0.14 mmol H2 g−1 dry weight (DW) was yielded after sludge anaerobic fermentation of Thalia dealbata treated with SE, and it increased to 4.79 ± 0.86 mmol H2 g−1 DW after SE-[Bmim]Cl treatment. For Phragmites (sp.) it was 1.45 ± 0.42 and 2.75 ± 0.76 mmol H2 g−1 DW after SE and SE-[Bmim]Cl pretreatment, respectively. Therefore, SE-[Bmim]Cl pretreatment can be developed for efficiently enhancing bio H2 conversion from wetland plant Thalia dealbata.

Enhancement of gaseous chlorobenzene biodegradation and power generation in a microbial fuel cell by bifunctional Acinetobacter sp. HY-99C

Haoyang Liu,Yang Yu,Shengtao Jiang,Haimin Sun,Weixi Zhang,Jianmeng Chen,Dongzhi Chen

The efficient biodegradation of volatile chlorinated hydrocarbons using microbial fuel cells (MFCs) offers a feasible approach for purifying waste gas and alleviating energy crises. However, power generation is limited by poor pollutant biodegradation and slow electron transfer . The bifunctional bacterium Acinetobacter sp. HY-99C was screened and used to improve the performance of a conventional MFC. The inoculation of strain HY-99C into the conventional MFC promoted the formation of a compact biofilm with high metabolic activity and an enriched bifunctional genus ( Acinetobacter ), which resulted in the accelerated decomposition of chlorinated aromatic compounds into biodegradable organic acids. This led to efficient chlorobenzene removal and power generation from the MFC, with a chlorobenzene elimination capacity of 70.8 g m −3 h −1 and power density of 89.6 mW m −2 , which are improved over those of previously reported MFCs. This study provides novel insights into enhancing pollutant removal and power generation in MFCs.
