Exploring the antibacterial mechanism of essential oils by membrane permeability, apoptosis and biofilm formation combination with proteomics analysis against methicillin-resistant staphylococcus aureus

Cailin Tang,Jiali Chen,Lixia Zhang,Rongfei Zhang,Sichen Zhang,Shaoxia Ye,Zhimin Zhao,Depo Yang

Methicillin-resistant Staphylococcus aureus (MRSA) is one of the important causes of food poisoning and infectious diseases worldwide, it can produce a large number of virulence factors, enhance the colonization ability of the host so that it can quickly colonize and spread on the surface of the objects. Essential oil (EO) is one of the natural products with antimicrobial properties, can be used as an important source of antibacterial agent discovery, and has a broad development prospect. However, the unclear mechanisms of antibacterial action have become an obstacle to its further development and use. Hence, the objective of the present study was to reveal the antibacterial mechanism of EO from Amomum villosum Lour ( A villosum Lour) against MRSA using label-free quantitative proteomics, investigate the effect of EO on the bacterial proteome, enzymatic activities and leakage of bacterial intracellular biomacromolecule. Proteomic analysis of MRSA in the presence of EO found that a total of 144 differential expressed proteins (DEPs) between the control and treatment group, in which 42 proteins were distinctly up-regulated and 102 proteins were down-regulated. Besides, sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analysis, determination of cell membrane permeability and apoptosis, scanning electron microscopy (SEM) observations, bacterial surface hydrophobicity, and biofilm formation measurement were performed. Collectively, the above results indicated that the cell membrane damage by EO leads to the loss of membrane integrity and causes leakage of intracellular macromolecular substances, inhibition of protein, and biofilm synthesis. These findings manifested that EO exerts antibacterial effect by multiple avenues and expands our understanding of the antibacterial mechanism, it has potential application value in food preservative and pharmaceutical industries.

Simultaneous electrochemical determination of Staphylococcus aureus and Listeria monocytogenes based on Ti3C2Tx MXene nanoribbons/copper nanoparticles coupled with aptamer-gated probe/UiO-66 framework

Xiaohua Jiang,Zhiwen Lv,Changquan Rao,Xiaowen Chen

Foodborne pathogenic bacteria pose severe threats to human health and often coexist within the same environment. Thus, developing a sensitive and reliable method for their simultaneous detection is critically important. In this study, we first synthesized Ti 3 C 2 T x MXene nanoribbons/copper nanoparticles (Ti 3 C 2 T x NR/Cu) via a simple self-reduction method to modify electrodes for signal amplification. Concurrently, we constructed UiO-66 MOFs encapsulated with distinct probes and gated by target-specific aptamers (Apts) as bio-gatekeepers. We then proposed an innovative electrochemical strategy for the simultaneous detection of two model pathogens: Staphylococcus aureus ( S. aureus ) and Listeria monocytogenes (LM). The Ti 3 C 2 T x NR/Cu nanohybrid synergistically enhanced the current signals of the probes by leveraging the combined advantages of Ti 3 C 2 T x NR and Cu nanoparticles. For S. aureus detection, methylene blue (MB) and S. aureus -specific aptamer (S-Apt) were employed as the probe and bio-gatekeeper, respectively, while ferrocene (Fc) and LM-specific aptamer (L-Apt) were used for LM. Upon target binding, the Apts dissociated from the UiO-66 MOFs, releasing the encapsulated probes (MB or Fc). The resulting current signals, measured at the Ti 3 C 2 T x NR/Cu electrode, enabled the quantitative and simultaneous detection of both pathogens. This platform's modular design allows easy adaptation for detecting other bacteria by simply replacing the aptamers, offering broad potential applications in controlling food safety.

Oligonucleotide-mediated the oxidase-mimicking activity of Mn3O4 nanoparticles as a novel colorimetric aptasensor for ultrasensitive and selective detection of Staphylococcus aureus in food

Shuangju Zhu,Yue Tang,Bin Shi,Wenying Zou,Xueli Wang,Chunxiao Wang,Yuangen Wu

Colorimetric detection of pathogenic bacteria in food still faces many challenges. Herein, a novel label-free and ultrasensitive colorimetric aptasensor was firstly established for the detection of food-borne pathogenic like Staphylococcus aureus ( S. aureus ) based on oligonucleotide-mediated the oxidase-mimicking activity of octahedral Mn 3 O 4 nanoparticles (NPs). The study confirmed that the common oligonucleotide such as SA31 aptamer can adsorb on the surface of octahedral Mn 3 O 4 NPs, and significantly inhibit the oxidase-mimicking activity by blocking their electron transfer to 3,3′,5,5′-tetramethylbenzidine. In the presence of S. aureus , aptamers will preferentially bind to bacterial cell, resulting in the recovery of oxidase-mimicking activity. The naked eye can easily recognize the color change from green to yellow, with the increase concentration of S. aureus from 10 CFU·mL −1 to 2 × 10 5 CFU·mL −1 . The colorimetric aptasensor shows a low detection limit of 3 CFU·mL −1 (S/N = 3). This strategy has excellent selectivity whether the presence of other competitive bacteria, and has been successfully applied to the non-enrichment detection of S. aureus in milk and pork samples.

Dual-mode colorimetric and fluorescent detection of Staphylococcus aureus using a bacteriophage-modified nanozyme with tetra-(4-aminophenyl) ethylene as a dual-functional substrate

Donghang Yin,Jing Quan,Mengyuan Tan,Hui Yuan,Tianyi Chen,Lei Liu,Siyuan Chang,Tao Zhang,Rong Feng,Genxi Zhang,Xuechao Xu

Herein, a bacteriophage-modified nanozyme sensing platform was developed for dual-mode detection of Staphylococcus aureus in food samples. A peroxidase-like Fe/Ce metal-organic framework nanozyme was functionalized with bacteriophage SAYZU01 to obtain SAYZU01@Fe/Ce MOF, enabling specific recognition and capture of S. aureus . Tetra-(4-aminophenyl) ethylene (TPE-4A), an aggregation-induced emission-active molecule, was introduced as a dual-functional substrate to generate both colorimetric and fluorescent signals after catalytic oxidation. Compared with conventional TMB, TPE-4A provides complementary colorimetric and fluorescent readouts, allowing cross-checking of the sensing response. The captured bacteria inhibited the catalytic activity of SAYZU01@Fe/Ce MOF, resulting in concentration-dependent signal changes. Under optimized conditions, the platform achieved low detection limits of 1.48 × 10 1  CFU mL −1 for colorimetry and 1.27 × 10 1  CFU mL −1 for fluorometry, with a linear range of 2.43 × 10 1 –2.43 × 10 7  CFU mL −1 . This strategy provides a dual-mode approach for rapid pathogen detection in food safety monitoring. Download: Download high-res image (342KB) Download: Download full-size image

Overcoming Methicillin-Resistant Staphylococcus aureus Infections by Disrupting Membrane Integrity and Inducing DNA Degradation with A Dual-Mechanism Fluorescent Phenanthro[9,10-d]imidazole Molecule

Ting Xu,Xiaoting Yan,Tingting Wang,Liping Cui,Shangshang Qin,Ruige Yang,Hong Yao,Yong Guo

The rise of multidrug-resistant bacteria, especially methicillin-resistant Staphylococcus aureus (MRSA), poses a serious threat to human health, necessitating an urgent need to develop innovative antibacterials with multifunctions for this. Here, focusing on integrating multi-antimicrobial mechanisms with visual monitoring, we report a novel phenanthro[9,10-d]imidazole fluorogen 9b, outstanding for its antibacterial effects against S. aureus and various clinical MRSA isolates (minimum inhibitory concentration = 0.5 to 1 μg/ml) with rapid bactericidal activity, high membrane selectivity, and low susceptibility to drug resistance. Further investigation revealed its dual antimicrobial mechanisms of action by concurrently disrupting bacterial cell membranes and inducing bacterial DNA degradation to accelerate bacterial death. Ultraviolet–visible and fluorescence spectroscopy studies showed that the fluorescence intensity of 9b was dramatically enhanced when it interacted with S. aureus. Notably, in vivo studies demonstrated that 9b effectively treated both skin and thigh MRSA infections, showing a favorable safety profile and superior efficacy compared to the first-line antibiotic vancomycin. The “dual-mechanism” fluorescent phenanthro[9,10-d]imidazole fluorogen 9b holds promise as a candidate for developing innovative antibacterial agents that enable simultaneous real-time diagnosis and treatment.

Design of Ready-to-Use “Ball-in-Ball” Staphylococcus aureus Microsphere Based on Novel Cryoprotectant and Drop Freeze-Drying Technology: Effective Preservation and Application

Zile Wang,Dongdong Chen,Xiaomei Zheng,Yuqing Li,Shaoqian Jiang,Yanfei Chen,Jingjian Jia,Libo Yu,Tao Peng

Staphylococcus aureus(S. aureus) poses a significant threat to public health and safety, and enhancing the monitoring ofS. aureusin food is essential to curb and prevent foodborne transmission. In order to obtain strains for more convenient and rapid use in quality control or quantitative analysis, this study designed a ready-to-use “ball-in-ball” microsphere based on a novel cryoprotectant combined with drop freeze-drying technology. When using a cryoprotectant that contains 1.5% bovine serum albumin, 4.5% trehalose, 8.2% polyethylene glycol 8000, and 4.1% D-mannitol, the survival rate ofS. aureuscan reach 98.2 ± 2.6%. This cryoprotectant effectively preventsS. aureusfrom shrinking, deforming, and damaging cell walls. Additionally, it shows desirable protective efficiency for other Gram-positive bacteria. The molding of microspheres is efficient and cost-effective, demonstrating good uniformity and stability without the need for pre-freezing. The moisture content and the count ofS. aureusshowed no significant changes over 90 days at −20 °C. In the simulated contaminated sample, the recovery rate ofS. aureusin milk and green tea was 83.1–93.7%. This study could provide a practical approach to improve the monitoring efficiency ofS. aureusand shows potential as a generalized strategy for preparing ready-to-use strains related to food safety.

Inhibitory effect of licochalcone A on dual-species biofilms of Listeria monocytogenes and Staphylococcus aureus: mechanism involved in quorum sensing and application in beef

Ling Jiang,Yilin Han,Xianxuan Li,Kai Zhong,Yanping Wu,Hong Gao

This study aimed to investigate the antibiofilm activity and mechanism of licochalcone A (Lico A), a natural flavonoid compound, against biofilms of L. monocytogenes , S. aureus , and their dual-species, and evaluate the potential application of Lico A in beef. The minimum inhibitory concentration (MIC) of Lico A against both L. monocytogenes and S. aureus was 7.5 μg/mL. Crystal violet staining and confocal laser scanning microscopic analysis showed that Sub-MICs of Lico A remarkably inhibited the biofilm formation of mono- and dual-species of L. monocytogenes and S. aureus . Meanwhile, Lico A significantly reduced the extracellular polymeric substances (EPS) production and the metabolic activity of biofilms, causing decreases in surface hydrophobicity, auto-aggregation, and motility. Furthermore, Lico A inhibited autoinducer-2 signaling activity by binding to LuxS and suppressing luxS expression, which disturbed the interspecies communication and thereby impeded biofilm formation. Biofilm-related genes such as sigB , flaA , icaA , and SarA were also regulated by Lico A. Importantly, Lico A effectively inhibited the dual-species biofilms of S. aureus and L. monocytogenes on beef, decreasing by 1.1 and 1.0 log CFU/g, respectively, on day 7 of storage at 4 °C. These findings highlight the potential of Lico A as a natural antibiofilm agent, especially against mixed-species biofilms, in food industry applications.

Amplification-free and label-free rapid detection of Staphylococcus aureus using solution-gated graphene transistor-based DNA biosensor with hybridization enhancement by interface engineering

Jiayuan Zheng,Jinhua Li,Tianci Lin,Zhanpeng Ren,Fucheng Wang,Zhonghao Shi,Haiyang Yu,Wei Jiang,Wei Tang

Staphylococcus aureus ( S. aureus ) is one of the leading causes of foodborne illnesses worldwide, posing a significant risk to food quality and safety. However, conventional detection methods often require complex amplification and labeling procedures, which are time consuming and expensive. In this study, an extremely sensitive electrochemical biosensor using a solution-gated graphene transistor (SGGT) was constructed for the amplification-free and label-free rapid detection of S. aureus by utilizing specific single-stranded DNA (ssDNA) to modify the gold (Au) sensing gate. This modified ssDNA was designed to hybridize with the target S. aureus DNA sequence. Interface engineering of the ssDNA-modified sensing gate via optimization of specific target and probe sequences was proposed to improve its hybridization efficiency. This enabled the sensitive quantitative detection of S. aureus DNA. The resulting biosensor exhibited excellent specificity in distinguishing non-complementary, 3-base/9-base DNA mismatch targets of S. aureus DNA and the specific DNA sequences of other common pathogenic bacteria. It also enabled the rapid detection of extracted target DNA sequences in solution (within 27 min) and had a low limit of detection (LoD) of 10 −17 M over a wide linear detection range (10 −17 –10 −8 M). Furthermore, the newly-developed biosensor facilitated the real-time and on-site detection of the S. aureus genome (ATCC 6538) with LoD of 10 −17 M (10 3 CFU/mL) in a portable smart-sensing system. This suggests a promising future for the development of rapid, label-free, and amplification-free detection of foodborne illnesses using low-cost, highly sensitive SGGT DNA biosensors with appropriate functionalization and interface engineering of the sensing electrode.

Inactivation of Listeria monocytogenes, Escherichia coli O157:H7, and Staphylococcus aureus by sequential light-emitting diodes (LEDs) treatment at 365 nm and 420 nm

Jie Xiao,Hongchao Zhang

Frequent outbreaks caused by foodborne pathogens pose long-term risks to consumer health. To proactively reduce the load of pathogenic bacteria during food processing, a novel light-based antibacterial approach was developed by sequential application of 365 nm and 420 nm light-emitting diodes (LEDs). Results demonstrated that after treatment with 365 nm (480 J/cm 2 ) followed by 420 nm (307.2 J/cm 2 ), the reduction of Listeria monocytogenes reached 4.05 ± 0.31 log CFU/mL, significantly higher (an additional 1.8 log CFU/mL, P < 0.05) than cumulative reductions achieved by each 365 nm (2.25 ± 0.92 log CFU/mL) and 420 nm (0.02 ± 0.15 log CFU/mL) treatments. Further analysis revealed that the enhancement in bacterial reduction achieved through the sequential treatment with 365 nm and 420 nm was primarily driven by the exposure time to 365 nm. The inactivation mechanisms were investigated, considering possible photothermal, physical, and oxidative effects. Findings showed that the antibacterial effect of sequential treatment was mainly ascribed to intracellular oxidation generated by reactive oxidative species (ROS), namely hydrogen peroxide and superoxide anion. The antibacterial mechanism of two LEDs may result from the sensitization of bacterial cells to excessive ROS, as evidenced by fluorescent intensity measurements and chemical scavenger assays. This research provides new insight for improving the efficacy of UVA and blue light treatment to control food contamination by Listeria .

Near-infrared laser-controlled nitric oxide-releasing gold nanostar/hollow polydopamine Janus nanoparticles for synergistic elimination of methicillin-resistant Staphylococcus aureus and wound healing

Zhuoying Liang,Wenkang Liu,Ziqiang Wang,Peilian Zheng,Wei Liu,Jianfu Zhao,Yunlong Zhong,Yan Zhang,Jing Lin,Wei Xue,Siming Yu

Recently, nitric oxide (NO) has received increasing interest in combat against bacteria-induced infections because of its ability to sensitize and enhance the antibacterial effectiveness of many therapeutic approaches such as antibiotics. However, high-efficient loading and controlled release of NO remain a big challenge. In the present work, a type of gold nanostar/hollow polydopamine Janus nanostructure (GNS/HPDA JNPs) with precise near infrared (NIR)-controlled NO release property was fabricated using a facile seed-mediated method. Upon NIR laser irradiation, the NO-releasing GNS/HPDA JNPs (GNS/HPDA-BNN6) exhibited a synergistic photothermal and NO antibacterial effect by significantly inhibiting the growth and biofilm formation of both Gram-negative and Gram-positive bacterial strains , including methicillin-resistant Staphylococcus aureus (MRSA). An in-depth mechanism study revealed that two pathways were mainly involved in the synergistic photothermal and NO antibacterial effect. In one pathway, the synergistic effect severely destroyed the bacterial membrane by causing leakage of intracellular components such as DNA . In another pathway, the synergistic effect largely disturbed bacterial metabolism by regulating relative metabolic genes, followed by enhancing ROS generation to cause intracellular GSH depletion and DNA damage. More importantly, the synergistic effect significantly diminished the drug resistance of MRSA by downregulating the expression of the drug-resistant gene mecA and some relative multidrug efflux pumps ( e.g., SepA and Tet38 ). An in vivo evaluation using a rat model with MRSA-infected wounds indicated that the synergistic photothermal and NO effect of GNS/HPDA-BNN6 can effectively eliminate MRSA from wounds, thereby alleviating inflammation and promoting wound healing. Statement of significance Multidrug-resistant (MDR) bacteria have become a big threat to mankind, and therefore, the development of innovative antibacterial agents with high antibacterial efficiency is urgently required. Nanomaterial-mediated nitric oxide (NO) therapy is a promising strategy to effectively combat MDR bacteria through a synergistic antibacterial effect. Here, a gold nanostar/hollow polydopamine Janus nanostructure with precise near infrared (NIR) light-controlled NO release property (GNS/HPDA-BNN6) was developed. Both in vitro and in vivo evaluations demonstrated that GNS/HPDA-BNN6 could effectively eliminate methicillin-resistant Staphylococcus aureus (MRSA) from infected wounds and promote wound healing through a synergistic photothermal and NO therapeutic effect. Remarkably, the synergistic effect significantly diminished the drug resistance of MRSA by downregulating the expression of some drug-resistant genes and multidrug efflux pumps.

Combination of microbiological, spectroscopic and molecular docking techniques to study the antibacterial mechanism of thymol against Staphylococcus aureus: membrane damage and genomic DNA binding

Wang Lang-Hong,Zhang Zhi-Hong,Zeng Xin-An,Gong De-Ming,Wang Man-Sheng

Thymol (2-isopropyl-5-methylphenol) is a natural ingredient used as flavor or preservative agent in food products. The antibacterial mechanism of thymol against Gram-positive, Staphylococcus aureus was investigated in this work. A total of 15 membrane fatty acids were identified in S. aureus cells by gas chromatography–mass spectrometry. Exposure to thymol at low concentrations induced obvious alterations in membrane fatty acid composition, such as decreasing the proportion of branched 12-methyltetradecanoic acid and 14-methylhexadecanoic acid (from 22.4 and 17.3% to 7.9 and 10.3%, respectively). Membrane permeability assay and morphological image showed that thymol at higher concentrations disrupted S. aureus cell membrane integrity, which may decrease cell viability. Moreover, the interaction of thymol with genomic DNA was also investigated using multi-spectroscopic techniques, docking and atomic force microscopy. The results indicated that thymol bound to the minor groove of DNA with binding constant ( K a ) value of (1.22 ± 0.14) × 10 4  M −1 , and this binding interaction induced a mild destabilization in the DNA secondary structure, and made DNA molecules to be aggregated. Thymol exerts its antibacterial effect throught destruction of bacterial cell membrane and binding directly to genomic DNA

Integrated microneedle patch and machine learning in time-resolved fluorescent immunochromatography test strip for accurate and rapid detection of Staphylococcus aureus enterotoxin B in salmon

Hui Jiang,Hui Du,Donglei Jiang,Xinmei Liu,Haijing Wu,Mengjing Huang,Rui Sun,Xiaojie Sun,Siqi Zeng,Yanmin Ju,Jun Yang

Staphylococcus aureus enterotoxin B (SEB) is a leading cause of foodborne illness. In this study, we developed a time-resolved fluorescent immunochromatography test strip (TRFITS) combined with a hydrogel microneedle patch (HMNP) and machine learning (ML) for the rapid detection of SEB in salmon. The developed HMNP, fabricated from methacrylated gelatin, can automatically extract 37 mg of interstitial fluid within 6 min. We then conducted TRFITS analysis on salmon samples to detect SEB and ML algorithms so as to predict its concentration. This method enabled efficient sampling and accurate quantification of SEB within 32 min, with a limit of detection (LOD) of 4.63 ng/mL. In addition, ML-assisted TRFITS applied to salmon demonstrated that the YOLOv11 model achieved a prediction accuracy of 91.3% and a lower LOD of 2 ng/mL. These findings highlight a novel approach toward on-site SEB detection with significant implications for food safety monitoring and point-of-care diagnostic applications.
