The preparation of multifunction chitosan adhesive hydrogel by “one- step” method

Ruixue Liu,Yingbo Li,Jichao Chen,Xiaojing Zhang,Zhibin Niu,Yadong Sun

Chitosan adhesive hydrogels have attracted interest because of their unique properties. However, it is still a challenge to use simple and unmodified chitosan to synthesize multifunctional hydrogels with adhesive, self-healing and strain sensing properties. Here, a multifunctional hydrogel of Ca2+-chitosan/polyacrylamide(Ca2+- CS/PAAm)with adhesive self-healing and strain sensing properties was carried out by one-step preparation method. The hydrogel has the ability to adhere to a variety of materials, wherein the adhesion strength to rubber surface is 48.4 kPa. The hydrogel has excellent strain sensing capability on finger bending or cyclic stretching, the sensitivity factor is 0.41 ± 0.015. The cut hydrogel healed completely at room temperature within 4 h, and the healing hydrogel can be stretched to 5 times of its original length. This makes Ca2+-CS/PAAm hydrogel to be great potential in the application of strain sensors, wound dressing and skin adhesives.

Antibiotic-loaded chitosan-gelatin scaffolds for infected seawater immersion wound healing

Qi Fang,Zexin Yao,Longbao Feng,Tian Liu,Shikun Wei,Pengcheng Xu,Rui Guo,Biao Cheng,Xinlu Wang

Skin damaged during sea battles is vulnerable to seawater immersion and bacterial infection. Scaffolds with effective biological function are highly desired for treatment of naval combat wound injuries. Herein, we prepared composite scaffolds of CS/GEL/GMs-CIP. The chitosan (CS) and gelatin (GEL) were cross-linked by genipin as matrix, and then gelatin microspheres loading ciprofloxacin hydrochloride (GMs-CIP) were add. From in vitro characterization results, CS/GEL/GMs-CIP had high water absorption ability, proper porosity, satisfactory fracture resistance, and flexibility. Furthermore, CS/GEL/GMs-CIP composite scaffold had excellent biocompatibility. Antibacterial experiments confirmed that CS/GEL/GMs-CIP had a significant inhibitory effect on E. coli , S. aureus and P. aeruginosa . The in vivo wound healing was evaluated using animal wound infection model of seawater immersion, and it was observed that the prepared composite scaffolds accelerated wound healing, reepithelialization, collagen deposition. Further analysis of wound tissue indicated that the expression of anti-inflammatory factor (TGF-β1) was up-regulated, but the serum endotoxin levels and expression of pro-inflammatory factor (TNF-a, IL-6, and IL-1β) were down-regulated. In summary, we believe that CS/GEL/GMs-CIP composite scaffold may serve as a promising multifunctional dressing for healing with open trauma wound infections and wound with seawater immersion.

Macroporous zwitterionic composite cryogel based on chitosan oligosaccharide for antifungal application

Peng Dong,Xiulin Shu,Ruqun Peng,Shunying Lu,Xiaobao Xie,Qingshan Shi

Chitosan oligosaccharide (COS), a time-dependent antimicrobial carbohydrate, is found antifungal active with a short duration of action due to excessive solubility. We attempted to address this issue by employing a hydrogel as a COS carrier. In this research, macroporous zwitterionic composite cryogels composed of COS and poly( N -methacryl arginine) (PMarg) were fabricated, serving as long-term antifungal dressings. Firstly, Marg was synthesized and characterized by Fourier transform infrared spectroscopy (FT-IR), 1 H and 13 C nuclear magnetic resonance (NMR), and high-resolution mass spectrometry (HRMS). Then, the COS/PMarg cryogels were prepared by redox initiation cryopolymerization. The macroporous morphology of the cryogels was confirmed by scanning electron microscope (SEM) with pore size varying from 20.86 to 50.87 μm. FTIR indicated that hydrogen bonding formed between COS and PMarg, and the interaction elevated thermal stability of the cryogels as evidenced by thermal-gravimetric analysis (TGA). Swelling capacity, mechanical properties, and COS release behavior of the COS/PMarg cryogels were investigated. With the release of COS, the antifouling activity of the cryogel increased. Antimicrobial tests indicated the COS/PMarg cryogel could effectively inhibit the proliferation of Candida albicans . It demonstrated that the macroporous zwitterionic COS/PMarg composite cryogel might be a potential antifungal dressing with sequential “sterilization-release” capacity.

Size-Dependent Biodistribution of Fluorescent Furano-Allocolchicinoid-Chitosan Formulations in Mice

Iuliia Gracheva,Maria Konovalova,Dmitrii Aronov,Ekaterina Moiseeva,Alexey Fedorov,Elena Svirshchevskaya

The aim of this study was to compare the biodistribution in mice of functionalized rhodamine B (Rh) labeled colchicine derivative furano-allocolchicinoid (AC,6) either conjugated to 40 kDa chitosan (AC-Chi,8) or encapsulated into chitosan nanoparticles (AC-NPs). AC-NPs were formed by ionotropic gelation and were 400–450 nm in diameter as estimated in mice by dynamic light scattering and confocal microscopy. AC-Chi and AC-NPs preserved the specific colchicine activity in vitro. AC preparations were once IV injected into C75BL/6 mice; muscles, spleen, kidney, liver, lungs, blood cells and serum were collected at 30 min, 2, 5, 10, and 20 h post injection. To analyze the distribution of the furano-allocolchicinoid preparations in body liquids and tissues, Rh was measured directly in sera or extracted by acidic ethanol from tissue homogenates. Preliminary Rh extraction rate was estimated in vitro in tissue homogenates and was around 25–30% from total quantity added. After in vivo injection, AC-NPs were accumulated more in liver and spleen, while less in kidney and lungs in comparison with free AC and AC-Chi. Therefore, incorporation of colchicine derivatives as well as other hydrophobic substances into nano/micro sized carriers may help redistribute the drug to different organs and, possibly, improve antitumor accumulation.Keywords:furano-allocolchicinoid;chitosan;nano/micro particles;biodistribution;Wnt-1 breast tumor

ROS-Eliminating Carboxymethyl Chitosan Hydrogel to Enhance Burn Wound-Healing Efficacy

Yang Cheng,Chen Yuhui,Huang Hai,Fan Shicai,Yang Chengliang,Wang Liping,Li Wenqiang,Niu Wenxin,Liao Jianwen

Overexpression of reactive oxygen species (ROS) can lead to chronic inflammation, which limits skin wound healing. Therefore, it is of great significance to develop materials that can locally control the adverse reactions caused by excessive ROS. In this research, an ROS-sensitive hydrogel with strong free radical scavenging ability was prepared by introducing the thione (Tk) group into carboxymethyl chitosan (CMCTS) hydrogel. CMCTS hydrogel was cross-linked by NH2-Tk-NH2 agent and loaded curcumin (Cur), which possessed favorable nontoxicity, water absorption, mechanical property, biodegradability, drug release behavior, the M2 phenotype, and inflammatory factor regulating the capacity of macrophages. It is worth noting that Cur@CMCTS-Tk hydrogel can significantly inhibit oxidative damage of human fibroblasts in the H2O2-induced microenvironment and protect their viability by reducing the production of intracellular ROS. In vivo, ROS-removing hydrogel effectively accelerated the process of wound healing and possessed good regenerative properties, including hair follicle formation, promotion of new blood vessel formation, and highly orderly arrangement of collagen fibers in the full-thickness skin burn defect rat model. Hence, we expect that the Cur@CMCTS-Tk hydrogel could be used for wound treatment and tissue regeneration due to the ability to scavenge excess ROS.

Adsorption of Pb2+, Cu2+ and Cd2+ by sulfhydryl modified chitosan beads

Yuru Yang,Lei Zeng,Zongkun Lin,Huabin Jiang,Aiping Zhang

A chitosan-based bead was synthesized by crosslinking as well as sulfhydryl modification reaction and its removal ability of Pb 2+ , Cu 2+ and Cd 2+ was investigated. The test results showed that the crystal structure of chitosan was destroyed completely and the specific surface area was greatly increased after modification. The adsorption of Pb 2+, Cu2+ and Cd2+ by the beads was carried out at different pH, ionic strength , contact time and initial concentration and the maximum adsorption capacities were 273.7 mg/g, 163.3 mg/g and 183.1 mg/g, respectively. Furthermore, due to the large ion radius of Pb 2+ , its adsorption was seriously disturbed by other ions in the competitive adsorption process. Finally, the adsorption processes of Pb 2+ , Cu 2+ and Cd 2+ were well fitted by the Langmuir isotherm model and the pseudo second-order kinetics model , respectively. Combined with the results of X-ray photoelectron spectroscopy, chemical coordination is the main adsorption mechanism.

Preparation and properties of caffeic-chitosan grafting fish bone collagen peptide

Xuqin Li,Zhou Lei,Jie Sheng,Yishan Song

In this study, a novel peptide grafted chitosan (CACS-FBP) with high peptide content, excellent moisture-absorption and moisture-retention abilities was prepared. Caffeic acid (CA) was used to modify chitosan, the highly water-soluble intermediate further reacted with fish bone collagen peptide to obtain the final product, and the synthesis of CACS-FBP was confirmed by the Fourier transform infrared spectroscopy (FT-IR), NMR, and UV-vis. The single-factor experiments indicated that the degree of substitution (DS) of CACS-FBP depended on the reaction temperature, reaction time, the mass ratio of fish bone collagen peptide to CACS (mFBP/mCACS) and the mass ratio of MTGase to CACS (mMTGase/mCACS). In addition, the antioxidant assay indicated that CACS-FBP had an excellent antioxidant capacity, and the CACS-FBP showed no cytotoxicity toward L929 mouse fibroblasts, all the results mean that the prepared peptide-containing chitosan derivative has potential application in pharmaceutical and biomedical fields.

Chitosan-Gelatin Scaffolds Incorporating Decellularized Platelet-Rich Fibrin Promote Bone Regeneration

Hui Chi,Xiangnan Song,Chengchao Song,Wei Zhao,Guanghua Chen,Anlong Jiang,Xiaoyan Wang,Tailong Yu,Leyu Zheng,Jinglong Yan

Platelet-rich fibrin (PRF), which functions as a growth factor carrier, has been extensively used to promote soft and hard tissue repair. However, whether decellularized PRF (DPRF) maintains its bioactive effects is unknown. Chitosan/gelatin(C/G) base scaffolds display appropriate biocompatibility and mechanical properties, but they lack biological activity. Thus, the incorporation of DPRF into the C/G scaffold can theoretically improve both the bioactivity of the C/G scaffold and the strength of PRF. In this study, DPRF was prepared using a method combining repeated freeze–thawing and enzymatic digestion. Also, DPRF-loaded chitosan-gelatin scaffolds (C/G/DPRF) were fabricated, using C/G scaffolds as controls. The osteogenic potential of scaffolds was investigated in vitro and in vivo. Compared with the C/G scaffold, C/G/DPRF had a larger pore size (280.8 ± 11.7 μm vs 235.0 ± 11.6 μm; P < 0.05), increased water uptake ratio (13.90 ± 0.09 vs 11.05 ± 0.10; P < 0.05), and similar porosity (90.50 ± 0.87 vs 90.65 ± 0.67; P > 0.05) but reduced compressive modulus (0.81 ± 0.02 MPa vs 1.17 ± 0.05 MPa; P < 0.05). In vitro, C/G/DPRF scaffolds accelerated attachment, proliferation, and osteogenesis-related marker expression of bone marrow stem cells. In vivo, C/G/DPRF scaffolds led to enhanced bone healing and defect closure in a rat calvarial defect model. Thus, we concluded that DPRF remains bioactive and the prepared C/G/DPRF scaffold is a promising material for bone regeneration.

Synthesis of gentamicin-grafted-chitosan with improved solubility and antibacterial activity

Tingting Yan,Chengpeng Li,Qianqian Ouyang,Dongying Zhang,Qingkun Zhong,Puwang Li,Sidong Li,Ziming Yang,Tao Wang,Qingxia Zhao

Due to superior properties, chitosan (CS) has been widely applied in health field. However its acid-solubility and poor antimicrobial activity compromise its safety and applications, respectively. To enhance its water-solubility and antimicrobial activity, gentamicin-grafted-chitosan (GT-CS) is synthesized for the first time, which is then confirmed by various characteristic methods and the graft ratio for gentamicin is about 22.1%. Particularly, it is the first study, disclosing there are degradation and crosslinking involved simultaneously during CS oxidation . Furthermore, GT-CS shows greatly improved water solubility (10.0w%). Compared to CS, the antibacterial activity of GT-CS is improved significantly, approaching the pure gentamicin. It should be noted that the antimicrobic mechanism may not be the target-specific interactions but the nonspecific interactions with the bacterial membrane as shown by scanning electron microscope. To sum up, GT-CS is a superior antimicrobial with good solubility.

Naproxen Nanoparticle-Loaded Thermosensitive Chitosan Hydrogel for Prevention of Postoperative Adhesions

Yu Wang,XianLun Pang,Jia Luo,Qian Wen,ZhouXue Wu,QiuXia Ding,Ling Zhao,LingLin Yang,BiQiong Wang,ShaoZhi Fu

Postoperative adhesions are the most common complications of peri-abdominal surgery; they not only affect the patient’s quality of life but also increase the risk of a subsequent surgery. The use of implantable dressings to physically block surgical wounds is the primary solution to prevent postoperative adhesions. In this study, we prepared naproxen nanoparticles that were loaded with chitosan hydrogel (CS/Nap hydrogel) to prevent postoperative adhesions. Our data confirmed that the prepared CS/Nap hydrogel was thermosensitive and suitable for injection. The efficacy of anti-adhesion in a rat model revealed that the hydrogel effectively separated from the wounds of the abdominal wall and cecum. On day 7 postsurgery, the wounds were completely covered by a new epithelial layer, whereas wounds in the negative control group were glued together. Additionally, the in vivo toxicity study showed that the CS/Nap hydrogel had fewer toxic and side effects on major tissues and organs, including the liver, spleen, heart, lung, and kidney. We showed that a drug delivery system based on CS/Nap hydrogel has the potential not only to prevent postoperative abdominal adhesions and relieve pain but also to contribute to the administration of the hydrophobic drug naproxen.

Mesoporous silica nanoparticles carried on chitosan microspheres for traumatic bleeding control

Xun Sun,Yan Fang,Zonghao Tang,Zhengchao Wang,Xinqing Liu,Haiqing Liu

Chitosan has been made into various hemostats, but their hemostatic efficiency for controlling severe traumatic bleeding is still inadequate. The aim of this work is to make quick hemostats by incorporating mesoporous silica nanoparticles into chitosan. Porous chitosan-silica composite microspheres (CSMS-S) with high hemostatic efficacy were fabricated through a combination of the microemulsion , thermally induced phase separation, and surfactant templating method. A large number of mesoporous silica nanoparticles were formed on and within the CSMS-S microspheres, which had abundant surface and inner macropores. The synergetic two hemostatic mechanisms from chitosan and mesoporous silica nanoparticles let CSMS-S composite microspheres with proper amount of silica displayed better hemostatic potential than the single component porous chitosan microspheres (CSMS). Within a same time interval, the whole blood clotting kinetics showed that CSMS-S could form larger blood clots than CSMS. The hemostatic time of CSMS-S was down to 97 s from 114 s of CSMS in the rat liver laceration model. The cytotoxicity and histological analysis proved that CSMS-S was a safe hemostatic agent without noticeable adverse effects on tissues around the wound. Our results demonstrate that CSMS-K is a promising quick hemostatic agent for traumatic hemorrhaging control.

Preparation of collagen/chitosan microspheres for 3D macrophage proliferation in vitro

Di Wang,Meiyue Wang,Anhe Wang,Jieling Li,Xin Li,Honglei Jian,Shuo Bai,Jian Yin

Macrophages, as an important member of immune cells, have now become a research hotspot in the field of immunotherapy, which requires prominent proliferation in vitro for clinical use. Herein, we reported the fabrication of collagen/chitosan microspheres via emulsification for 3D macrophages proliferation. With glutaraldehyde (GA) as cross-linker, the microspheres have an average diameter of 200 μm and positively charged surfaces, which are of great stability and biocompatibility. Compared with 2D cell proliferation , the 3D microspheres could dramatically enhance macrophages proliferation rate and promote their biological functions. Furthermore, macrophages on the surface of microspheres could be easily collected by blowing and suction, without the need of trypsin digesting or the degradation of microsphere. Hence the microspheres could be reused for macrophage proliferation repeatedly. This study highlights opportunities for development of high-effective cell proliferation approach and provides a great promise in immune cell based immunotherapy.
