Icariin controlled release on a silk fibroin/mesoporous bioactive glass nanoparticles scaffold for promoting stem cell osteogenic differentiation

Xiaofeng Shen,Pengfei Yu,Hua Chen,Jiangping Wang,Binjie Lu,Xuefeng Cai,Chun Gu,Guoqiang Liang,Donglin Hao,Qihan Ma,Yuwei Li

The treatment of bone defects caused by various reasons is still a major problem in orthopedic clinical work. Many studies on osteogenic implant materials have used various biologically active factors such as osteogenic inducers, but these biologically active factors have various side effects. Therefore, in this study, silk fibroin (SF) was used as a scaffold material, mesoporous bioactive glass nanoparticles (MBGNs) as a sustained release carrier, and the traditional Chinese drug icariin (ICA) was loaded to promote bone formation. The experiments in this study have proven that SF/MBGNs-ICA scaffolds can successfully load and release ICA for a long time, and the sustained-release ICA can promote the proliferation and differentiation of BMSCs for a long time. This controlled-release ICA organic/inorganic two-component scaffold material is expected to become a new bone grafting solution.

Construction of covalent organic frameworks with alternating rigid and flexible units and their controlled release of active sites

Xiaofeng Li,Zhimin Jia,Jie Zhang,Ningning He,Honghan Long,Yue Qi,Jing Li,Ning Liu,Yang Li,Lijian Ma

Different from traditional covalent organic frameworks (COFs) with single structural mode, COFs constructed with unique alternating rigid and flexible structural elements can maximize the great advantages of COFs in both crystalline structure and application performance. Herein, a series of rigid & flexible COFs containing both rigid and flexible units were designed and synthesized. The rigid units can reduce the growth defects and maintain the regular crystalline structures. The flexible units endow COFs with abundant potential active sites that can be released through controlled dissociation reactions. Thus, the site-released multiple-site COFs exhibit significant improvement on adsorption performance towards mercury and uranium, with a maximum adsorption capacity increase of up to 61% compared to the original COFs. The strategy for the construction of rigid & flexible COFs and the controlled release of active sites points a new direction for the design and synthesis of COFs with multi-functional sites and outstanding application performance.

Synthesis and characterization of well-defined star PLLA with a POSS core and their microspheres for controlled release

Zhang Xiaojing,Wang Chaojun,Fang Shaoming,Sun Jiashu,Li Chong,Hu Yuelei

In this paper, a series of hybrid star PLLA (sPLLA) with different arm lengths was synthesized via the hydrosilylation between octakis(dimethylsiloxy) silsesquioxane (Q 8 M 8 ) and functionalized PLLA macromolecules with vinyl end groups (mPLLA). mPLLA was synthesized by ring-opening polymerization of l -lactide using 2-hydroxyethylmethacryl as an initiator in the presence of stannous 2-ethylhexanoate as a catalyst. The obtained sPLLA has low polydispersity with polydispersity index values from 1.29 to 1.30. The arm numbers of sPLLA vary from 5 to 7 and decrease with the increase in the length of the mPLLA arm due to the steric hindrance, which are estimated by 1 H NMR analysis. The branched structure of sPLLA is also evidenced by the lower intrinsic viscosity when compared with the linear mPLLA with similar molecular weight. Both the glass transition temperatures ( T g ’s) and melting temperatures ( T m ’s) of sPLLAs are higher than those of the mPLLA arms. The incorporation of polyhedral oligomeric silsesquioxane (POSS) does not change the crystalline structure of PLLA, while the crystallinity of sPLLA is enhanced as the result that the POSS core acts as a heterogeneous nucleating agent in the matrix to promote the crystallization ability of PLLA. High-resolution transmission electron microscopy observation suggests that POSS disperses in the crystalline PLLA matrix as 5–20 nm aggregates. Microspheres of sPLLA with mean diameter 1 to 2 μm were prepared via emulsion solvent evaporation method. The sPLLA microspheres have higher loading capacity and encapsulation efficiency and lower drug release rate than mPLLA microspheres.

Fabrication of core–shell microspheres using alginate and chitosan–polycaprolactone for controlled release of vascular endothelial growth factor

Hua Wu,Chunyan Liao,Qianyi Jiao,Zheng Wang,Wenze Cheng,Ying Wan

Alginate microspheres loaded with vascular endothelial growth factor (VEGF) were prepared via an emulsification method using calcium chloride as a crosslinker. The microspheres with encapsulation efficiency of about 80% were coated by chitosan–polycaprolactone (CH–PCL) with various PCL percentages changing from around 15 to 42   wt.% to fabricate core–shell alginate/CH–PCL microspheres with an average size of around 40   μm. It was found that the CH–PCL coating layer on the core–shell microspheres could have a sandwich-like structure. The PCL content in the CH–PCLs and the concentration of CH–PCL solutions in preparing the microsphere functioned as two key factors to regulate the release profiles of the microspheres. Some selected alginate/CH–PCL microspheres were further crosslinked using genipin as a crosslinker, and the amount of genipin was found to be another impactful factor to mediate the release patterns of the microspheres. In vitro release measurements revealed that VEGF-release from these core–shell microspheres was controlled either by Fickian diffusion or non-Fickian transport that involves both diffusion and swelling. Some optimized core–shell microspheres were capable of maintaining sustained VEGF-release in an approximately linear manner over a period of time longer than 4   weeks and did not involve a significant initial burst.

Biocompatible hydrophobic cross-linked cyclodextrin-based metal-organic framework as quercetin nanocarrier for enhancing stability and controlled release

Runan Zhao,Tao Chen,Yanfei Li,Lihang Chen,Yu Xu,Xuesong Chi,Songfeng Yu,Wenjun Wang,Donghong Liu,Beiwei Zhu,Jiangning Hu

Cyclodextrin-based metal-organic framework (CD-MOF) has been widely used in various delivery systems due to its excellent edibility and high drug loading capacity. However, its typically bulky size and high brittleness in aqueous solutions pose significant challenges for practical applications. Here, we proposed an ultrasonic-assisted method for rapid synthesis of uniformly-sized nanoscale CD-MOF, followed by its hydrophobic modification through ester bond cross-linking (Nano-CMOF). Proper ultrasound treatment effectively reduced particle size to nanoscale (393.14 nm). Notably, carbonate ester cross-linking method significantly improved water stability without altering its cubic shape and high porosity (1.3 cm 3 /g), resulting in a retention rate exceeding 90% in various media. Furthermore, the loading of quercetin did not disrupt cubic structure and showcased remarkable storage stability. Nano-CMOF achieved controlled release of quercetin in both aqueous environments and digestion. Additionally, Nano-CMOF demonstrated exceptional antioxidant (free radical scavenging 82.27%) and biocompatibility, indicating its significant potential as novel nutritional delivery systems in food and biomedical fields.

Formula and process optimization of controlled-release microcapsules prepared using a coordination assembly and the response surface methodology

BeiXing Li,Lei Guan,Kai Wang,DaXia Zhang,WeiChang Wang,Feng Liu

A model chlorpyrifos microcapsule was prepared using coordination assembly between Fe 3+ and tannic acid (TA). The influence of independent variables such as the dropping rate of TA ( X 1 ) and Fe 3+ ( X 3 ), the concentration of TA ( X 2 ) and Fe 3+ ( X 4 ), and the reaction temperature ( X 5 ) on the encapsulation efficiency ( R 1 ) and release characteristics ( R 2 ) of the microcapsule had been investigated, based on a central composite design with five factors and five levels. The results showed that the main factors influencing R 1 and R 2 were X 4 and X 2 , then the interaction between X 2 and X 4 , followed by X 5 and X 3 . The optimal formula mainly based on higher R 1 and lower R 2 were determined and then tested. The optimized conditions led to an encapsulation efficiency and cumulative release proportion of 97.12% ± 0.72% and 40.07% ± 0.53%, along with the average relative errors of predicted values being 1.78% and −1.60%, respectively. © 2015 Wiley Periodicals, Inc. Controlled release of vancomycin from 3D porous graphene-based composites for dual-purpose treatment of infected bone defects

Weizong Weng,Wei Nie,Qirong Zhou,Xiaojun Zhou,Liehu Cao,Fang Ji,Jin Cui,Chuanglong He,Jiacan Su

Infected bone defects (IBD) are a challenging problem in orthopedic practice. Biomaterials have attracted attention as a novel approach to overcome this challenge instead of using conventional strategies. Graphene, a two-dimensional carbon allotrope, has been shown to possess multiple advantages, such as osteogenesis-promoting activity, a large surface area for drug delivery, and antibacterial activities. However, these attractive merits have not been entirely explored or introduced for the management of IBD. Herein, a novel approach using a vancomycin-laden, self-assembled, graphene-based material against IBD was developed, and the approach uses a single-stage bone graft instead of multiple procedures. Owing to the π–π bonding with graphene, vancomycin can be promptly delivered in the initial stage, followed by a sustained release, which ensures the rapid elimination of an infection and provides a durable pathogen revival inhibition via the inherent antibacterial activity of graphene. Moreover, the 3D porous structure incorporated with nano-hydroxyapatite (nHA) provided a biomimetic microenvironment favored by cell adhesion and osteogenic differentiation. Further evaluation of this system in an IBD animal model demonstrated prompt control of infection and promotion of bone regeneration. These results revealed the potential of this approach to be an effectual solution for IBD treatment and a new area of graphene application in the future.

Synthesis and characterization of mesoporous magnetic nanocomposites wrapped with chitosan gatekeepers for pH-sensitive controlled release of doxorubicin

Juan Wu,Wei Jiang,Yewen Shen,Wei Jiang,Renbing Tian

Multifunctional nanocarriers based on the Fe 3 O 4 nanoparticles core and mesoporous silica shell (mSiO 2 ) were synthesized for controlled drug release through magnetic targeting and pH-sensitive performances. The developed Fe 3 O 4 @mSiO 2 nanocarriers exhibited a suitable size (63   nm) and good magnetic responsibility, doxorubicin (DOX) could be successfully loaded into the mesoporous of Fe 3 O 4 @mSiO 2 via electrostatic interaction, and the drug loading content and loading efficiency are 29.3% and 93.6%, respectively. The chitosan (CS) was employed to wrap the Fe 3 O 4 @mSiO 2 -DOX as the blocking agent to inhibit premature drug release, and the final CS/Fe 3 O 4 @mSiO 2 -DOX exhibited excellent pH-sensitivity, 86.1% DOX was released within 48   h at pH   4.0. Furthermore, all the release behaviors fit the Higuchi model very well and a purely diffusion-controlled process played a major role on DOX release from CS/Fe 3 O 4 @mSiO 2 -DOX. In addition, MTT assays in human liver hepatocellular carcinoma cells (HepG2) demonstrated that the CS/Fe 3 O 4 @mSiO 2 -DOX had high anti-tumor activity, while the Fe 3 O 4 @mSiO 2 nanocarriers were practically non-toxic. Thus, our results revealed that the CS/Fe 3 O 4 @mSiO 2 -DOX could play an important role in the development of intracellular delivery nanodevices for cancer therapy.

Redox-responsive PAEFc-b-PDMAEMA amphiphilic block copolymer self-assembly micelles: Physicochemical properties and anticancer drug controlled release

Yuan Wang,Yan-Ling Luo,Feng Xu,Ya-Shao Chen,Wei Tang

Ferrocene-containing amphiphilic block copolymers , poly(2-acryloyloxyethyl ferrocenecarboxylate)- block -poly(2-(dimethylamino) ethyl methacrylate) (PAEFc- b -PDMAEMA), were synthesized via sequential ATRP , and self-assembled into globular nanoscaled micelle aggregates. The copolymer micelles exhibited reversible redox on-off switch feature, which was mediated by using H 2 O 2 , KMnO 4 , NaClO and FeCl 3 as a model of oxidants and ascorbic acid as a model of reductants. The micelle nanoparticles were used to load and deliver anticancer drug, 10-hydroxycamptothecine, finding that the encapsulated drug was rapidly delivered by selectively responding to redox environments in cancer cells. MTT assays were performed to uncover cytotoxicity of the developed copolymer micelles and potentiality used for cancer therapy.

A tubular gelatin scaffold capable of the time-dependent controlled release of epidermal growth factor and mitomycin C

Jixiang Zhu,Fanwen Yang,Fupo He,Xiumei Tian,Shuo Tang,Xiaoming Chen

A tubular gelatin scaffold for the time-dependent controlled release of epidermal growth factor (EGF) and mitomycin C (MMC) was fabricated. EGF was incorporated using silk fibroin carriers, and MMC was planted using polylactide (PLA) microspheres. The relationship between scaffold properties and crosslinking degrees was evaluated. As the crosslinking degree was increased from 23.7% to 65.3%, the mechanical properties of the scaffold obviously improved, and the compressive modulus increased to approximately 65   kPa. The mass degradation of the scaffold was also controlled from 9 days to approximately 1 month. In vitro release tests indicated that the scaffold mainly released EGF in the early period and MMC in the later period. Urethral epithelial cells (UECs) and urethral scar derived fibroblast cells (UFCs) were coseeded in the scaffold at a ratio of 1:1. After 9 days of coculture, immunostaining results displayed that the proportion of UECs continuously increased to approximately 71%. These changes in cell proportion were confirmed by the results of Western blot analysis. Therefore, the scaffold promoted the growth but inhibited the regeneration of UFCs. This scaffold for time-dependent controlled release of multiple biofactors may be potentially useful in urethral reconstruction and other tissue engineering studies.

Development of phosphonate-terminated magnetic mesoporous silica nanoparticles for pH-controlled release of doxorubicin and improved tumor accumulation

Erxi Che,Long Wan,Ying Zhang,Qinfu Zhao,Xiling Han,Jia Li,Jia Liu,Siling Wang

In this study, phosphonate-terminated magnetic mesoporous nanoparticles (pMMSNs) was designed by incorporation of MNPs in the center of mesoporous silica nanoparticles (MSNs) and followed by grafting phosphonate group on to the surface of MMSNs. The carrier exhibited a typical superparamagnetic feature and the saturation magnetization was 4.89 emu/g measured by vibrating sample magnetometer (VSM). pMMSNs had a spherical morphology and a pore size of 2.2 nm. From N 2 adsorption-desorption analysis, pMMSNs had a surface area of 613.4 m 2 /g and a pore volume of 0.78 cm 3 /g. Phosphonate modification improved the colloidal stability of MMSNs, and the hydrodynamic diameter of pMMSNs was around 175 nm. The hydrophilic phosphonate group significantly enhanced the negative surface charge of MMSNs from −19.3 mV to −28.8 mV pMMSNs with more negative surface charge had a 2.3-fold higher drug loading capacity than that of MMSNs. In addition, the rate and amount of release of doxorubicin (DOX) from DOX/pMMSNs was pH-dependent and increased with the decrease of pH. At pH 7.4, the release amount was quite low and only approximately 17 wt% of DOX was released in 48 h. At pH 5.0 and 3.0, the release rate increased significantly and the release amount achieved 31 wt% and 60 wt% in 48 h, respectively. To evaluate the magnetic targeting performance of pMMSNs, FITC labeled pMMSNs was injected into mice bearing S180 solid tumor. FITC labeled pMMSNs controlled by an external magnetic field showed higher tumor accumulation and lower normal tissue distribution.

Thermally and magnetically dual-responsive mesoporous silica nanospheres: preparation, characterization, and properties for the controlled release of sophoridine

Liling Dong,Hailong Peng,Shenqi Wang,Zhong Zhang,Jinhua Li,Fanrong Ai,Qiang Zhao,Mei Luo,Hua Xiong,Lingxin Chen

Novel thermally and magnetically dual-responsive mesoporous silica nanoparticles [magnetic mesoporous silica nanospheres (M-MSNs)–poly( N -isopropyl acrylamide) (PNIPAAm)] were developed with magnetic iron oxide (Fe 3 O 4 ) nanoparticles as the core, mesoporous silica nanoparticles as the sandwiched layer, and thermally responsive polymers (PNIPAAm) as the outer shell. M-MSN–PNIPAAm was initially used to control the release of sophoridine. The characteristics of M-MSN–PNIPAAm were investigated by transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, thermogravimetry, N 2 adsorption–desorption isotherms, and vibrating specimen magnetometry analyses. The results indicate that the Fe 3 O 4 nanoparticles were incorporated into the M-MSNs, and PNIPAAm was grafted onto the surface of the M-MSNs via precipitation polymerization. The obtained M-MSN–PNIPAAm possessed superparamagnetic characteristics with a high surface area (292.44 m 2 /g), large pore volume (0.246 mL/g), and large mesoporous pore size (2.18 nm). Sophoridine was used as a drug model to investigate the loading and release properties at different temperatures. The results demonstrate that the PNIPAAm layers on the surface of M-MSN–PNIPAAm effectively regulated the uptake and release of sophoridine. © 2014 Wiley Periodicals, Inc. J. Appl. Polym.
