Considering the key roles of macrophages in tissue repair and immune therapy, designing smart biomaterials able to harness macrophage phenotypes on demand during the healing process has become a promising strategy. Here, a novel “sandwich” cell culture platform with near-infrared (NIR) responsive dynamic stiffness was fabricated to polarize bone marrow-derived macrophages (BMDMs) in situ for revealing the relationship between the macrophage phenotype and substrate stiffness dynamically. Under NIR irradiation, calcium ions (Ca2+) diffused through the middle layer of the IR780-mixed phase change material (PCM) due to the photothermal effect of IR780, resulting in an increase of hydrogel stiffness in situ by the crosslinking of the upper layer of the hyaluronic acid-sodium alginate hydrogel (MA-HA&SA). The up-regulation of inducible nitric oxide synthase (iNOS) and tumor necrosis factor-α (TNF-α) was quantified by immunostaining and enzyme-linked immune sorbent assay (ELISA), respectively, indicating the transformation of macrophages from the anti-inflammatory to pro-inflammatory phenotype under dynamic stiffness. The nuclear Yes-associated-protein (YAP) ratio positively correlated with the shift of the macrophage phenotype. The modulation of macrophage phenotypes by stiffness-rise without the stimuli of cytokines offers an effective and noninvasive strategy to manipulate immune reactions to achieve optimized healing or therapeutic outcomes.

Encouraging progress in multifunctional nanotheranostic agents that combine photothermal therapy (PTT) and different imaging modalities has been made. However, rational designed and biocompatible multifunctional agents that suitfable for in vivo application is highly desired but still challenging. In this work, we rationally designed novel ultrasmall multifunctional nanodots (FS-GdNDs) by combining the bovine serum albumin (BSA)-based gadolinium oxide nanodots (GdNDs) obtained through a biomineralization process with a small-molecule NIR-II fluorophore (FS). The as-prepared FS-GdNDs with an ultrasmall hydrodynamic diameter of 9.3 nm exhibited prominent NIR-II fluorescence properties, high longitudinal relaxivity (10.11 mM −1 s −1 ), and outstanding photothermal conversion efficiency (43.99%) and photothermal stability. In vivo studies showed that the FS-GdNDs with enhanced multifunctional characteristics diaplayed satisfactory dual-modal MR/NIR-II imaging performance with a quite low dose. The imaging-guided PTT achieved successful ablation of tumors and effectively extended the survival of mice. Cytotoxicity studies and histological assay demonstrated excellent biocompatibility of the nanodots. Importantly, this novel FS-GdNDs can undergo efficient body clearance through both hepatobiliary and renal excretion pathways. The novel ultrasmall multifunctional FS-GdNDs with excellent features hold tremendous potential in biomedical and clinical applications.

Zn2Ga3−x−yCrxNdyGe0.75O8 (ZGGO:Crx,Ndy) persistent luminescent nanoparticles (PLNPs) (x = 0, 0.02; and y = 0, 0.01, 0.02, 0.03, 0.04) were synthesized via a hydrothermal method in combination with a following heat treatment under vacuum. The effects of Nd3+ concentration on chemical composition, size distribution, luminescence property, the persistent energy transfer from Cr3+ to Nd3+ and afterglow imaging were studied in detail. When the Nd3+ concentration was increased from 0 to 0.04, the particle size increased from 45.4 to 86.2 nm. The afterglow emissions in the NIR-I (696 nm) and NIR-II (1067 nm) regions, which are ascribed to the 2E, 4T2 → 4A2 transitions of Cr3+ and the 4F3/2 → 4I11/2 transition of Nd3+, respectively, were simultaneously acquired after stopping the 635 nm excitation. Among the nanoparticles with different concentrations, ZGGO:Cr0.02,Nd0.02 exhibits the strongest NIR-II afterglow intensity and the corresponding energy transfer efficiency from Cr3+ to Nd3+ is found to be 25.9%. In addition, enhanced X-ray excited afterglow imaging can be observed in Nd3+/Cr3+ codoped nanoparticles dispersed in water, human serum albumin solution and simulated lysosomal environment compared to the Cr3+ singly doped nanoparticles. Renewable NIR afterglow imaging was realized through an X-ray reexcitation strategy. In particular, both the X-ray excitation strategy accompanied by NIR-I afterglow emission and the red light (635 nm) excitation strategy accompanied by NIR-II afterglow emission exhibit high tissue penetration capability. This study provides a further understanding of how to develop a suitable strategy for realizing deep tissue autofluorescence-free bioimaging.

In this work, BiOCl x Br 1−x microflowers were synthesized as building blocks for superhydrophobic NIR reflective coatings. The molar ratio of Cl to Br had a great influence on the microstructure and optical properties of BiOCl x Br 1−x . As the increase of x value, the microflower structure tunes from loose to dense. The NIR solar reflectance of BiOCl x Br 1−x ranges from 0.860 to 0.935. The BiOCl x Br 1−x coatings reduced the inner temperature of the heat box by approximately 3.9 °C. Due to the three-dimensional hierarchical structure of BiOCl x Br 1−x microflowers, superhydrophobic cool coatings can be fabricated on various building materials, such as concretes, aluminum plates, and glasses. Moreover, the superhydrophobic coatings exhibit superior antifouling performance, contributing to retain high NIR reflectance of the coatings for a long time. Therefore, the prepared superhydrophobic NIR reflective coatings with superior antifouling property have great potential to be used for building energy-saving.

As a prominent member of two dimensional (2D) materials, MXenes have shown encouraging photophysical properties and so there is highly desirability to explore their phototherapeutic potential in cancer. Herein, we demonstrate a facile top-down strategy for the synthesis of nitride-based MXene, titanium nitride quantum dots (Ti 2 N QDs) in solution. With a size of around 5 nm, the obtained Ti 2 N QDs exhibit extraordinarily high photothermal conversion efficiency under laser irradiation in both first and second near-infrared (NIR) biowindows (NIR-I, 808 nm, 48.62%; and NIR-II, 1064 nm, 45.51%). In vitro and in vivo assessments demonstrate that the Ti 2 N QDs possess excellent biocompatibility, photoacoustic (PA) effect, and photothermal therapy (PTT) efficiency. Moreover, the Ti 2 N QDs showing obvious aggregation in tumors after 4 h post-injection, which enables facilitate PA imaging-guided PTT in both NIR-I/II biowindows without noticeable toxicity. Encouragingly, the Ti 2 N QDs with appropriate degradation property also show an applicable excretion rate from the body in vivo , which ensures sufficient stability in circulation to accomplish therapeutics roles and then can be smoothly discharged from the body after that. These results indicate the high potential of biodegradable Ti 2 N QDs in PA imaging-guided PTT in both NIR-I/II biowindows for precision cancer treatment.

Near-infrared (NIR) light-responsive, injectable hydrogels are among the most promising drug delivery systems for localized anticancer therapy owing to its minimally invasive administration and remote-controlled manner. However, most currently reported NIR-responsive hydrogels were usually generated through physical mixing of thermosensitive polymers and photothermal conversion agents. In this study, a novel type of dynamic-covalent hydrogel (GelPV-DOX-DBNP) with NIR light-triggered drug release behavior was rationally designed for chemo-photothermal combination treatment of tumors. Concretely, this NIR-responsive hydrogel was formed by specific benzoxaborole-carbohydrate interactions between benzoxaborole (BOB)-modified hyaluronic acid (BOB-HA) and fructose-based glycopolymer (PolyFru), where photosensitizer perylene diimide zwitterionic polymer (PDS), reductant ascorbic acid (Vc), anticancer drug doxorubicin (DOX) as well as photothermal nanoparticles (DB-NPs) were encapsulated, simultaneously. Upon 660 nm light irradiation, both PDS and Vc within the designed hydrogel can convert oxygen into hydrogen peroxide, which could make hydrogel be degraded through the breakage of dynamic covalent bonds based on benzoxaborole-carbohydrate interactions, leading to NIR light-activatable release of DOX and DB-NPs from GelPV-DOX-DBNP. Furthermore, the released DB-NPs can convert 915 nm light irradiation into heat, enabling the application of GelPV-DOX-DBNP as a NIR-responsive drug delivery platform for both chemotherapy and photothermal therapy (PTT). In vivo results prove that GelPV-DOX-DBNP exhibited a markedly enhanced chemo-photothermal synergistic therapy for 4T1 tumor model mice, compared to chemotherapy alone or PTT. This work presents a new strategy to construct NIR light-responsive hydrogel as one alternative drug delivery system for anticancer applications.

Chronic diabetic wounds are difficult to treat due to imbalanced inflammatory responses, high blood glucose levels, and bacterial infections. Novel therapeutic approaches based on nucleic acid analogues have been proposed, with unique advantages in improving angiogenesis, increasing collagen synthesis, and exerting anti-inflammatory effects. However, the inherent electronegativity of nucleic acids makes them less susceptible to cellular uptake. In this paper, a kind of near infrared (NIR)-responsive nanocomposite hydrogel loaded with nucleic acid vectors was proposed for promoting wound healing. The redox system composed of molybdenum disulphide nanosheets (MoS 2 NSs) initiated the copolymerization of quaternized chitosan containing double bonds and N -isopropylacrylamide (NIPAAm) to form the matrix. In addition, MoS 2 NSs with photothermal conversion performance endow the nanocomposite hydrogel to have NIR-response property and act as physical crosslinking points in the matrix. Polydeoxyribonucleotides (PDRN), which have the effect of promoting wound healing, were made into nucleic acid vectors, and loaded into the NIR-responsive hydrogel. MoS 2 NSs can convert NIR irradiation into heat, causing phase transitions of temperature-sensitive segments that trigger volume contraction of the hydrogel to extrude the nucleic acid vector. Promoting angiogenesis, slowing inflammation, and guiding tissue regeneration were demonstrated in the diabetic wound model treated with the NIR-responsive nanocomposite hydrogel.

Glaucoma is a leading cause of irreversible blindness worldwide and characterized by progressive loss of retinal ganglion cells (RGCs). As glaucoma progresses asymptomatically and the lack of specific biomarkers or noninvasive imaging tools during the early stage, it is often discovered at a relatively late stage without cure. Development of an effective tool for early diagnosis remains a necessity. As well known, endoplasmic reticulum (ER) stress and subsequent accumulation of reactive oxygen species (ROS) play a vital role in the development of glaucoma, with peroxynitrite (ONOO − ) as one of the most abundant cytotoxic ROS. In this work, we constructed a novel near-infrared (NIR) fluorescent nanoprobe named ER-ONOO-NPs. It specifically and sensitively targets ER-associated ONOO − with low biotoxicity in cultured RGCs. Systematically applied to the NMDA-induced mouse model of glaucoma, ER-ONOO-NPs performs well for real-time dynamic monitoring of ONOO − fluctuations at the Ganglion cell layer of mice prior to RGCs apoptosis. Thus, our results suggest that the ER-targeted NIR fluorescent nanoprobe could be a noninvasive imaging tool for early diagnosis of glaucoma in high-risk population.

The development of intelligent nanotheranostic technology that integrates diagnostic and therapeutic functions holds great promise for personalized nanomedicine. However, most of the nanotheranostic agents exhibit “always-on” properties and do not involve an amplification step, which may largely limit imaging contrast and restrict therapeutic efficacy. Herein, we construct a novel nanotheranostic platform (Hemin/DHPs/PDA@CuS nanocomposite) by assembling DNA hairpin probes (DHPs) and hemin on the surface of PDA@CuS nanosheets that enables amplified fluorescence imaging and activatable chemodynamic therapy (CDT) of tumors. The cancer-relevant APE1 triggers nucleic acid amplification with DHPs to generate activatable and amplified fluorescence signals for discriminating cancer cells from normal cells. Meanwhile, excessive G-quadruplex/hemin-based DNAzyme are also activated, and they function as Fenton-like catalysts to catalyze the production of highly toxic hydroxyl radicals (•OH) for CDT. Moreover, owing to the excellent photothermal conversion efficiency in the near-infrared-II (NIR-II) window, the PDA@CuS not only improves the catalytic performance of CDT but also furnishes PTT. A remarkable antitumor therapeutic effect is demonstrated both in vitro and in vivo. Therefore, the Hemin/DHPs/PDA@CuS nanocomposite is expected to provide a promising avenue for precise imaging-guided antitumor therapy.

CeO2 nanozymes have demonstrated the potential to enhance biological scaffolds with chemodynamic therapy. However, their catalytic efficacy is limited by the slow conversion of Ce4+ to Ce3+ and the lack of substrates like H2O2 and H+. To address these challenges, we adopted a dual-pronged strategy that utilized the plasmonic resonance of Au nanoparticles and their glucose-oxidase mimicry to boost electron and hydrogen transfer. Specifically, we integrated Au/CeO2 nanozymes into poly-l-lactic acid scaffolds via selective laser sintering. This conversion of Ce3+ to Ce4+ in the scaffolds enhanced the reduction of H2O2 to a hydroxyl radical, inducing oxidative stress in tumor cells. The Au nanoparticles played a crucial role in boosting the Ce3+/Ce4+ catalytic cycle by providing both the energy and the catalytic substrates. They recycled Ce4+ back to Ce3+ by exploiting plasmonic-induced hot electrons and catalyzed glucose oxidation to supply H2O2 and H+. Our nanoscale and atomic-scale simulations confirmed that the Au/CeO2 hybrid structure utilized near-field coupling to amplify the plasmonic resonance and the Au–O–Ce bridge reduced the electron transfer barrier. Consequently, the Au/CeO2 scaffold decreased the activation energy from 22.57 to 9.92 kJ/mol. These findings highlight the significant promise of the Au/CeO2 nanozyme scaffold for NIR-enhanced chemodynamic therapy.

Amphiphilic random copolymer micelle of P(NVP- co -NHPM) that responds to both UV and NIR light was designed and investigated. N -hydroxyphthalimide methacrylate — bearing a photoacid generator functional group was synthesized and used to construct the hydrophobic segment of P(NVP- co -NHPM); the N–O bond can be photocleaved when exposing the polymer aqueous solution to 310 nm UV light irradiation, inducing the micellar dissociation and delivery of loaded substances. Dynamic light scattering (DLS), atomic force microscope (AFM), and transmission electron microscopy (TEM) were used to characterize the micellar dissociation and controlled release processes. Moreover, the upconverting nanoparticles (NaLuF 4 :Gd/Yb/Tm) that can effectively absorb NIR light and convert it into visible and UV light were coloaded into polymer micelles to activate the micellar dissociation and the delivery of loaded cargoes via 980 nm NIR light.

Near infrared (NIR) light-responsive polymeric nanomedicines might achieve spatiotemporal, pulsatile, and on-demand drug release profiles and are appealing as a noninvasive technology for various clinical therapies. Polypeptide copolymer composite nanoparticles with different amounts of upconversion nanoparticles (UCNPs) were successfully fabricated in aqueous solution, and the NIR sensitivity of the composite nanoparticles was characterized by time-resolved UV-vis spectroscopy, on-line dynamic light scattering, and transmission electron microscopy. The composite nanoparticles disassembled with the help of UCNPs converting NIR light into UV light, and the NIR-responsive time can be tuned by both the loaded amount of UCNPs and the polypeptide chain length. The composite nanoparticles loaded with the anticancer drug doxorubicin (DOX) can release DOX in a controllable and/or pulsatile manner, and the drug release profile can be manipulated by NIR light. The DOX-loaded composite nanoparticles can be quickly internalized by HeLa cells and then release DOX inside the cells, as evidenced by flow cytometry and confocal laser scanning microscopy. After 5 or 10 min of NIR irradiation, the half maximal inhibitory concentration (IC50) for the DOX-loaded composite nanoparticles dropped to 5.08 μg DOX equiv. mL−1 or 2.95 μg DOX equiv. mL−1, respectively, compared to 8.26 μg DOX equiv. mL−1 for the non-irradiated sample, demonstrating a tunable NIR-triggered cytotoxicity. This work provides a versatile platform for the fabrication of NIR-responsive polypeptide copolymer nanomedicines with the potential for on-demand drug delivery and cancer therapy.
