Antibody-Functional Microsphere-Integrated Filter Chip with Inertial Microflow for Size–Immune-Capturing and Digital Detection of Circulating Tumor Cells

Yi Su,Qingchang Tian,Dingyi Pan,Lanlan Hui,Yanni Chen,Qi Zhang,Wei Tian,Jie Yu,Shen Hu,Yang Gao,Dahong Qian,Tian Xie,Ben Wang

Circulating tumor cells (CTCs) in blood is the direct cause of tumor metastasis. The isolation and detection of CTCs in the whole blood is very important and of clinical value in early diagnosis, postoperative review, and personalized treatment. It is difficult to separate all types of CTCs that efficiently rely on a single path due to cancer cell heterogenicity. Here, we designed a new kind of “filter chip” for the retention of CTCs with very high efficiency by integrating the effects of cell size and specific antigens on the surface of tumor cells. The filter chip consists of a semicircle arc and arrays and can separate large-scale CTC microspheres, which combined with CTCs automatically. We synthesized interfacial zinc oxide coating with nanostructure on the surface of the microsphere to increase the specific surface area to enhance the capturing efficiency of CTCs. Microspheres, trapped in the arrays, would entrap CTCs, too. The combination of the three kinds of strategies resulted in more than 90% capture efficiency of different tumor cell lines. Furthermore, it is easy to find and isolate the circulating tumor cells from the chip as tumor cells would be fixed inside the structure of a filter chip. To avoid the high background contamination when a few CTCs are surrounded by millions of nontarget cells, a digital detection method was applied to improve the detection sensitivity. The CTCs in the whole blood were specifically labeled by the antibody–DNA conjugates and detected via the DNA of the conjugates with a signal amplification. The strategy of the antibody–functional microsphere-integrated microchip for cell sorting and detection of CTCs may find broad implications that favor the fundamental cancer biology research, the precise diagnosis, and monitoring of cancer in the clinics.

A Facile Droplet-Chip-Time-Resolved Inductively Coupled Plasma Mass Spectrometry Online System for Determination of Zinc in Single Cell

Han Wang,Beibei Chen,Man He,Bin Hu

Single cell analysis is a significant research field in recent years reflecting the heterogeneity of cells in a biological system. In this work, a facile droplet chip was fabricated and online combined with time-resolved inductively coupled plasma mass spectrometry (ICPMS) via a microflow nebulizer for the determination of zinc in single HepG2 cells. On the focusing geometric designed PDMS microfluidic chip, the aqueous cell suspension was ejected and divided by hexanol to generate droplets. The droplets encapsulated single cells remain intact during the transportation into ICP for subsequent detection. Under the optimized conditions, the frequency of droplet generation is 3–6 × 106 min–1, and the injected cell number is 2500 min–1, which can ensure the single cell encapsulation. ZnO nanoparticles (NPs) were used for the quantification of zinc in single cells, and the accuracy was validated by conventional acid digestion-ICPMS method. The ZnO NPs incubated HepG2 cells were analyzed as model samples, and the results exhibit the heterogeneity of HepG2 cells in the uptake/adsorption of ZnO NPs. The developed online droplet-chip-ICPMS analysis system achieves stable single cell encapsulation and has high throughput for single cell analysis. It has the potential in monitoring the content as well as distribution of trace elements/NPs at the single cell level.

High reliable and chromaticity-tunable flip-chip w-LEDs with Ce:YAG glass-ceramics phosphor for long-lifetime automotive headlights applications

Chaoyang Ma,Yongge Cao,Xiaofei Shen,Zicheng Wen,Ran Ma,Jiaqi Long,Xuanyi Yuan

Nowadays, major commercial w-LEDs fabricated by the traditionally gold-wire-welding packaging technology have undergone considerable development as indoor/outdoor lighting sources due to its high-energy utilization efficiency, long service life, environmental friendliness, and excellent chromatic stability. While, new generation applications in projections, automotive lighting, street lighting, plaza lighting, and high-end general lighting need further improvements in power handling and light extraction. Herein, transparent Ce:YAG glass-ceramics (GCs) phosphor was prepared by low-temperature co-sintering polycrystalline Ce:YAG phosphor powder and home-made PbO-B 2 O 3 -ZnO-SiO 2 glass powder. Thereafter, the flip-chip (FC) w-LEDs were fabricated with the GCs phosphor plates and FC blue chips. The GCs-based FC w-LEDs show not only excellent heat- and humidity-resistance characteristics, but also superior optical performances with an LE of 112.8 lm/W, a CRI of 71.2, a CCT of 6103 K as well as a chromaticity coordinate of (0.3202, 0.3298), under a high operation current of 400 mA. The technology route will open a practically commercial feasible approach to achieve excellent performances for advanced high-power FC w-LEDs.

Planar integration of flexible micro-supercapacitors with ultrafast charge and discharge based on interdigital nanoporous gold electrodes on a chip

Chao Zhang,Jian Xiao,Lihua Qian,Songliu Yuan,Shuai Wang,Pengxiang Lei

Flexible, wearable, implantable and easily reconfigurable micro-fabricated pseudocapacitors with impressive volumetric stack capacitance and energy densities are desired for electronic devices. In this work, scratching technology at the micron-scale enables construction of the planar electrode systems directly based on nanoporous gold films. We demonstrate that both nanoporous channels with high ion-accessible ability and interconnected skeletons with high conductivity enable the design of pseudocapacitive micro-supercapacitors with high performances. These planar devices show several attractive features including ultrafast charge/discharge (high rate), large capacitance (1.27 mF cm−2, 127 F cm−3), and ultrahigh energy density (0.045 W h cm−3) while maintaining a high power density (22.21 W cm−3). Especially, the superb cyclability and mechanical flexibility give them great potential for future microelectronics with a tiny volume. The design concept reported here provides an avenue to integrate planar micro-supercapacitors into large-scale devices with a small environmental footprint.

Integrated microfluidic chip with a MIP-gated organic electrochemical transistor for continuous, selective, and label-free quantification of tumor marker

Qi Wang,Xiaoxia Bai,Fei Liu,Peipei Li,Qianlin Tang

Early diagnosis of cancer is crucial to effective treatment of the patients and to higher survival rates. Tumor marker detection as an effective and noninvasive cancer diagnosis method has important clinical significance. N 1 , N 12 -diacetylspermine (DAS) is a new tumor marker. The development of its detection methods is an urgent need for medical diagnosis and human health monitoring. The conventional detection methods of DAS are labor-intensive and time-consuming approaches. In this work, a novel electrochemical detection platform was established by integrating an organic electrochemical transistor (OECT) with a microfluidic chip and applied for trace measurement of DAS. The sensing unit is a gate electrode in the OECT modified with molecularly imprinted polymers (MIPs), which can be used as an artificial receptor to realize label-free detection. The high specific recognition ability of MIPs films combined with the amplified function of an OECT yielded a highly sensitive and selective electrochemical detection chip. The integrated microfluidic channel controlled   the exact amount of solution to be delivered and provided real-time detection of DAS. The experimental results indicated that changes in channel current was proportional to DAS concentrations from 1 nM to 10 μM, with the detection limits of 0.8 nM. It is the first demonstration of the integration of a microfluidic system with a MIP-gated OECT for real-time tumor marker detection. The study results of this work helped to develop Point-Of-Care (POC) tools for the early diagnosis of lung cancer, colon cancer and breast cancer.

Laser Construction TiOx-Ag heterostructure interface for high sensitive coupling resonance enhanced fluorescence in bacterial nucleic acid amplification sensor chip

Danhe Wang,Xiaomeng Xu,Tao Qiu,Na Ren,Haiyun Liu,Guoxin Fang,Zhen Liu

Coupled resonance at the noble metals and metal oxides heterogeneous interface can enhance the fluorescence of molecules. This fascinating direction provides high value for the preparation and engineering design of fluorescence sensing platforms in nucleic acid sensing, pathogen detection and biological imaging. Here, an integrated nucleic acid amplification sensor chip consists of an array of silver nanoparticles/oxygen vacancy titanium oxide (TiO x -Ag) heterostructure substrates. The femA gene of Staphylococcus aureus was amplified and detected by enhancement fluorescence on the integrated chip. The TiO x -Ag heterostructure array was prepared by a laser method, the enhanced fluorescence performance was investigated in detail using calcein as a probe. The optical, electronic structure and plasmonic local field analysis indicate the strong coupling between the fluorescence molecule and the TiO x -Ag crystal plane, which inspired the excellent fluorescence signal of calcein, with an enhancement factor of up to 5. Staphylococcus aureus was detected by the above chip with a detection limit as low as 157 cfu/mL. These methods were applied to the detection of spiked serum samples with good detected performances, providing a powerful tool for rapid and reliable diagnosis of pathogens in medical institutions.

Acoustofluidic bubble-driven assisted functionalized nano-array coated micro pin-fins surface for efficient liquid-vapor phase change chip cooling

Hongqiang Chen,Xiang Ma,Xiong Zhao,Quan Gao,Yonghai Zhang,Jinjia Wei

Embedded phase change boiling heat transfer is crucial for addressing the energy saving posed by high-power compact electronic devices. However, the endeavor to fabricate improved heat transfer structures in confined and constricted spaces remains a formidable challenge. Herein, we present a simple and straightforward acoustofluidics strategy to create stable, controllable, and efficient functional Zinc oxide (ZnO) nanoarray coatings in narrow micro-pin-fins chip surfaces with excellent phase change cooling performance. A robust acoustic micromixer combining bubble acoustic streaming and hydrodynamic effect provided by spiral microchannels is proposed to facilitate microscale fluid mixing across a broad flow rate range. The ZnO nanoarray-coated micro pin-fins chip, which allows for customizable lengths, densities, diameter, and morphology, is fabricated through straightforward adjustment of acoustofluidics micromixer parameters. Excellent phase change cooling performance is obtained on this surface, giving priority to nucleation (superheat≈ 4 °C, reduced by 50.2 %), low wall temperature (≤70.7 ℃) under the limit CHF, negligible pumping energy (≤3.5 kPa), and simultaneously enhancing the critical heat flux (CHF, from 45.1 to 77.2 W·cm −2 ) and heat transfer coefficient (HTC, from 5955 to 15552 W·m −2 ·K −1 ) by up to 71.3 % and 161.1 %, respectively, compared with the smooth chip surface. In situ observation and analysis of the dynamic wetting behavior and boiling bubble dynamics indicated ZnO nanoarray coated micro pin-fins chip promotes the phase change heat exchange process by massive nucleation sites, effective control of boiling bubbles, and ultra-fast two-stage liquid rewetting capability. These findings not only provide important guidelines for the precise control and rational design of functional nanomaterials, but also provide new insights for embedded cooling and significant energy savings on power devices.

A temperature-independent model of dual calibration standards for onsite and point-of-care quantification analyses via electrophoresis titration chip

Zehua Guo,Yiren Cao,Liuyin Fan,Weiwen Liu,Li Wei,Yixin Ma,Jicun Ren,Qiang Zhang,Chengxi Cao

Electrophoresis titration chip (ETC) is a versatile tool for onsite and point-of-care quantification analyses because it affords naked-eye detection and a straightforward quantification format. However, it is vulnerable to changes in environmental temperature, which regulates the electrophoretic migration by affecting the ion mobility and the target recognition by influencing the enzyme activity . Therefore, the quantification accuracy of the ETC tests was severely compromised. Rather than using the dry bath or heating/cooling units, we proposed a facile model of dual calibration standards (DCS) to mathematically eliminate the effects of temperature on quantification accuracy. To verify our model, we deployed the ETC device at different temperatures ranging from 5 to 40 °C. We further utilized the DCS-ETC to determine the protein content and uric acid concentration in real samples outside the laboratory. All the experimental results showed that our model significantly stabilized the quantification recovery from 35.31-153.44 % to 99.38–103.44 % for protein titration; the recovery of uric acid titration is also stable at 96.25–106.42 %, suggesting the enhanced robustness of the ETC tests. Therefore, DCS-ETC is a field-deployable test that can offer reliable quantification performance without extra equipment for temperature control. We envision that it is promising to be used for onsite applications, including food safety control and disease diagnostics.

Multiscale Anisotropic Scaffold Integrating 3D Printing and Electrospinning Techniques as a Heart-on-a-Chip Platform for Evaluating Drug-Induced Cardiotoxicity

Sitian Liu,Zihan Wang,Xinyi Chen,Mingying Han,Jie Xu,Ting Li,Liu Yu,Maoyu Qin,Meng Long,Mingchuan Li,Hongwu Zhang,Yanbing Li,Ling Wang,Wenhua Huang,Yaobin Wu

Cardiac safety assessments are significant in drug discovery, as drug-induced cardiotoxicity (DIC) is the primary cause of drug attrition. Despite heart-on-a-chip (HoC) technology becoming an increasingly popular tool for evaluating DIC, its development remains a challenge owing to the anisotropic cardiac structure of the native myocardium. Herein, an anisotropic multiscale cardiac scaffold is presented via a hybrid biofabrication method by combining 3D printing with electrospinning technology, where the 3D-printed micrometer-scale scaffold frames enable mimicking the interwoven myocardium anatomical structure and the branched-aligned electrospun nanofibers network is able to directionally guide cellular arrangements. The in vitro 3D bioengineered cardiac tissues are then fabricated by encapsulating three-layer multiscale scaffolds within a photocurable methacrylated gelatin hydrogel shell. It is demonstrated that such an anisotropic multiscale structure could contribute to enhancing cardiomyocyte maturation and synchronous beating behavior. More attractively, with the integration of 3D bioengineered cardiac tissues and a self-designed microfluidic perfusion system, a 3D anisotropic HoC platform is established for evaluating DIC and cardioprotective efficacy. Collectively, these results indicate that the HoC model developed by integrating the 3D bioengineered cardiac tissues could effectively recapitulate the clinical manifestations, thereby highlighting their efficacy as a valuable preclinical platform for testing drug efficacy and cardiotoxicity.

A molecularly imprinted polymer-based triple-ratio fluorescence nanosensor integrated with a paper-based microfluidic chip for rapid detection of enoxacin

Chao Wang,Yuhao Wen,Siyu Wang,Jiawen Xiang,Die Hu,Mingsan Man,Xin Zhang,Bowei Li,Lingxin Chen

Enoxacin (ENX), a commonly used fluoroquinolone antibiotic, has raised environmental concerns due to its widespread use and resulting pollution. This study presents a novel approach for the rapid and sensitive detection of ENX in aquatic environments using molecularly imprinted polymer-based triple-ratio fluorescence (MIP-TRF) nanosensor. The nanosensor is constructed based on a photoinduced electron transfer (PET) mechanism, combining the high selectivity of molecular imprinting technology with the high sensitivity and anti-interference capability of triple-ratio fluorescence. By optimizing modifiers, eluents, and reaction conditions, the nanosensor demonstrates excellent selectivity and sensitivity, with a detection limit as low as 0.008 mg/L and good linearity over a concentration range of 0.05–7.0 mg/L. The recovery rate in river water samples was between 98.88 % and 102.76 %. To assess the potential application of the sensor, preliminary tests were also conducted by integrating the MIP-TRF nanosensor with a paper-based microfluidic chip, showing that the sensor offers higher detection speed, simplicity, and cost-effectiveness compared to traditional methods. This provides an efficient and convenient solution for the environmental monitoring of ENX and similar contaminants.

Effects of different fat addition amounts on the temporal dominance of sensations (TDS), texture characteristics and flavor of pork chip snacks

Mingcheng Zhang,Qiyuan Ni,Di Yu,Kaixian Zhu,Wensheng Yao,Dengyong Liu

Pork chip snacks (PCS) is a traditional Chinese delicacy originating from the southeastern coastal regions, renowned for its thin and brittle texture. To enhance the flavor profile of pork chip snacks, varying amounts of pork backfat (0%, 3%, 6%, 9%, 12%) were incorporated into the raw materials during production. TDS analysis was employed to evaluate how different fat levels influence the dominant sensory attributes during consumption. The results indicate that brittleness, hardness, dry, fatty, meaty, sweetness, and salty are the primary dominant sensations of pork chip snacks. The samples with added 6% (PCS 6 ) and 9% (PCS 9 ) fat showed a higher advantage in brittleness, mainly due to their low water activity and rich pore structure. PCS 9 has the highest volatile organic compound content and shows the highest meat flavor dominance rate (0.63). Therefore, adding 9% fat significantly improves both the flavor and brittleness of pork chip snacks.

Robust thermal performance and color purity of samarium based deep red emitting phosphor for near-UV chip-based high-color-rendering

Wasim Ullah Khan,Zhiqi Ye,Mebrouka Boubeche,Tong Liu,Zhenbin Guo,Yueli Zhang

In the progress of LED phosphors, the chemical stability, thermal stability, and color purity was not usually given due attention. However, the most critical problems of WLEDs are the deficit in the red-spectral region, stability, low color-rendering index (CRI) and high correlated color temperature (CCT) limit its efficiency for vivid application. Herein, the Ca 3 YGa 3 B 4 O 15 : x Sm 3+ (CYGB: x Sm 3+ ) were prepared by a solid-state route. X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FTIR), diffuse reflection spectroscopy (DRS), and as well as photoluminescence (PL) spectra are used to exemplify the resulting samples. The structure and the Sm 3+ dopants in the cation-borate phosphors were quantified, signifying the importance at Ca and Y sites. Under ultraviolet radiation-induced bright deep-red emissions with high color-purity up to 97.5%. The energy-migration (EM) between the Sm 3+ ion was investigated both empirically and theoretically in the region of Sm 3+ contents. Several theoretical-models were employed to simulate the lifetime of Sm 3+ at various-concentrations, and the processes were investigated as a forced-electric dipole. The chemical-stability of CY 0.95 GB:0.05Sm 3+ has also been investigated in this work. Interestingly, the CY 0.95 GB:0.05Sm 3+ showed exceptional thermal-quenching resistance of>91.2% intensity at 400 K. The WLED containing CY 0.95 GB:0.05Sm 3+ blended with the BAM:Eu 2+ and CYAB:Tb 3+ phosphor demonstrated low CCT ≈ 3843 K and high CRI ≈ 91.9. These findings were healthier than the industrial WLED having Y 3 Al 5 O 12 :Ce 3 + and blue-LED.
