Temperature is one of the physical parameters that affects the emission luminescence properties of upconversion (UC) materials. Temperature-dependent emission color tuning and good temperature sensitivity in multifunctional β-NaYF4:Yb/Er@β-NaYF4:Yb/Tm microcrystals were achieved under 980 nm excitation. Based on the different emission bands (blue, green and red bands) and spectral sensitivities of the core–shell structured microcrystals to temperature, intense white-light emission was obtained at room temperature and a real-time naked-eye recognizable emission color change from white to the purplish-pink region was realized with the temperature increasing from 300 to 500 K. Furthermore, optical temperature sensing properties of the sample with good sensitivity (from 0.0034 to 0.0044 K−1) were investigated by using a fluorescence intensity ratio between 521 nm (2H11/2 → 4I15/2 transition) and 540 nm (4S3/2 → 4I15/2 transition) in the temperature range from 300 to 500 K. The results indicate that the sample has high sensitivity in a relatively wide range of temperature intervals, especially in the low temperature region. We believe that the multifunctional core–shell structured microcrystals exhibit promising applications in white-light emitting diodes, the anti-counterfeiting field, and temperature sensing.

Endoplasmic reticulum (ER) stress is a cellular state that results from the overload of unfolded/misfolded protein in the ER that, if not resolved properly, can lead to cell death. Both acute lung infections and chronic lung diseases have been found related to ER stress. Yet no study has been presented integrating metabolomic and transcriptomic data from total lung in interpreting the pathogenic state of ER stress. Total mouse lungs were used to perform LC–MS and RNA sequencing in relevance to ER stress. Untargeted metabolomics revealed 16 metabolites of aberrant levels with statistical significance while transcriptomics revealed 1593 genes abnormally expressed. Enrichment results demonstrated the injury ER stress inflicted upon lung through the alteration of multiple critical pathways involving energy expenditure, signal transduction, and redox homeostasis. Ultimately, we have presented p-cresol sulfate (PCS) and trimethylamine N-oxide (TMAO) as two potential ER stress biomarkers. Glutathione metabolism stood out in both omics as a notably altered pathway that believed to take important roles in maintaining the redox homeostasis in the cells critical for the development and relief of ER stress, in consistence with the existing reports.

Septic cardiomyopathy is associated with mitochondrial damage and endoplasmic reticulum (ER) dysfunction. However, the upstream mediator of mitochondrial injury and ER stress has not been identified and thus little drug is available to treat septic cardiomyopathy. Here, we explored the role of B-cell receptor-associated protein 31 (BAP31) in septic cardiomyopathy and figure out whether melatonin could attenuate sepsis-mediated myocardial depression via modulating BAP31. Lipopolysaccharide (LPS) was used to establish the septic cardiomyopathy model. Pathway analysis was performed via western blot, quantitative polymerase chain reaction and immunofluorescence. Mitochondrial function and ER stress were detected via enzyme-linked immunosorbent assay, western blot, and immunofluorescence. After exposure to LPS, cardiac function was reduced due to excessive inflammation response and extensive cardiomyocyte death. Mechanistically, melatonin treatment could dose-dependently improve cardiomyocyte viability via preserving mitochondrial function and reducing ER stress. Further, we found that BAP31 transcription was repressed by LPS whereas melatonin could restore BAP31 expression; this effect was dependent on the MAPK-ERK pathway. Inhibition of the ERK pathway and/or knockdown of BAP31 could attenuate the beneficial effects of melatonin on mitochondrial function and ER homeostasis under LPS stress. Altogether, our results indicate that ERK-BAP31 pathway could be used as a critical mediator for mitochondrial function and ER homeostasis in sepsis-related myocardial injury. Melatonin could stabilize BAP31 via the ERK pathway and thus contribute to the preservation of cardiac function in septic cardiomyopathy.

Purpose It is demonstrated that unsaturated fatty acids can counteract saturated fatty acids-induced lipotoxicity, but the molecular mechanisms are unclear. In this study, we investigated the protective effects of monounsaturated oleic acid (OA) against saturated palmitic acid (PA)-induced cytotoxicity in rat β cells as well as islets, and mechanistically focused on its regulation on endoplasmic reticulum (ER) stress. Methods Rat insulinoma cell line INS-1E cells and primary islets were treated with PA with or without OA for 24 h to determine the cell viability, apoptosis, and ER stress. SD rats were fed with high-fat diet (HFD) for 16 w, then, HFD was half replaced by olive oil to observe the protective effects of monounsaturated fatty acids rich diet. Results We demonstrated that PA impaired cell viability and insulin secretion of INS-1E cells and rat islets, but OA robustly rescued cells from cell death. OA substantially alleviated either PA or chemical ER stressors (thapsigargin or tunicamycin)-induced ER stress. Importantly, OA attenuated the activity of PERK-eIF2α-ATF4-CHOP pathway and regulated the ER Ca 2+ homeostasis. In vivo, only olive oil supplementation did not cause significant changes, while high-fat diet (HFD) for 32 w obviously induced islets ER stress and impaired insulin sensitivity in SD rats. Half replacement of HFD with olive oil (a mixed diet) has ameliorated this effect. Conclusion OA alleviated PA-induced lipotoxicity in INS-1E cells and improved insulin sensitivity in HFD rats. The amelioration of PA triggered ER stress may be responsible for its beneficial effects in β cells.

Ethnopharmacological relevance Oxidative damage to osteoblasts was a key factor in the development of osteoporosis. Er-Xian Decotion (EXD) is widely used in China for the treatment of osteoporosis, which has a variety of antioxidant active ingredients. EXD may be an important source of protection against oxidative damage in osteoblasts, but the anti-osteoporotic active components of EXD is currently unclear. Aim of the study This work established an effective and reliable drug screening method to find main active ingredients in EXD (M-EXD) that can protect osteoblasts against oxidative stress and achieve anti-osteoporosis effects. Materials and methods H 2 O 2 -induced osteoblast cell fishing with UHPLC-QTOF/MS was firstly used to discover the potential active components from EXD. Afterword, the EXD compound-osteoporosis target network was constructed using network pharmacology , thus potentially anti-osteoporosis ingredients were founded, and their combination were defined as the M-EXD. Finally, pharmacology effects of M-EXD was evaluated by ovariectomized rats, prednisolone induced-zebrafish and H 2 O 2 -induced osteoblasts. Results 40 candidate active ingredients in EXD were initially screened out via pathological cell fishing. According to network pharmacology result, M-EXD consisted of 13 ingredients since they had a close relationship with 65 osteoporosis-related targets. Pharmacological evaluation showed that M-EXD significantly ameliorated oxidative stress in H 2 O 2 -induced osteoblast model, evidently reversed the activity of ALP , ROS , GSH-px, NO and MDA compared with the model group. M-EXD showed better anti-oxidative activities than individual ingredients, presenting obvious synergetic effects. In osteoporosis rat and zebrafish models, M-EXD also demonstrated good anti-osteoporotic properties by mitigating the osteoporosis bone loss and increasing serum bone morphogenetic protein 2 , and reversing osteocalcin expression in bone tissue. It significantly ameliorated oxidative stress in the in-vivo models. Moreover, M-EXD and EXD showed similar anti-osteoporotic and anti-oxidative properties, while the rest components of EXD had no satisfactory anti-osteoporotic efficacy. Conclusions Our work successfully identified the main active components in EXD, which could represent the efficacy of EXD on treating osteoporosis, and meanwhile, it also provided an effective strategy to investigate active ingredients from natural medicines, which might be helpful for drug development and application.

Autophagic degradation of the endoplasmic reticulum (ER-phagy) has been found to play a critical role in human sensory neuropathy. So far, however, specific and efficient intervention means for ER-phagy remain unexplored. Herein, brefeldin A (BFA), a blocking agent on protein transport between the ER and Golgi, was screened from ER stress inducers. BFA was then delivered to the perinuclear area co-localized with the ER by a mesoporous silica nanoparticle-based drug-carrier functionalized with autophagy-inducing peptides of TAT-beclin 1 (MSNs-BFA), to evoke a perturbation of ER-phagy. The molecular mechanism of ER-phagy regulated by BFA was explored by biochemical evaluation including time-lapse live-cell fluorescence imaging. We found that MSNs-BFA treatment caused a lower mRNA/protein expression level of FAM134b even under a compensation of autophagic flux in U2OS cells, and resulted in ER-expansion. The fragmentation of the ER was blocked as a response to ER stress mediated by inactivation of the AKT/TSC/mTOR pathway. Our work developed an efficient external manipulation strategy to regulate ER-phagy and may contribute to the therapeutic application of autophagy-related major human diseases.

Upconversion fluorescent nanostructures with controlled sizes and morphologies have attracted much attention due to their potential and important applications in bioimaging, photodynamic therapy, solar cells and so on. In this work, we describe a facile successive process to fabricate β-NaGdF4:Yb,Er nanocrystals coated with an ultrathin layer of CaF2 with enhanced upconversion fluorescence and stability. The as-prepared β-NaGdF4:Yb,Er@CaF2 nanoparticles were investigated by X-ray diffraction (XRD), transmission electron microscopy (TEM, JEM 21000), fluorescence spectroscopy (Hitachi High-Technology Corporation, F-2700) and inductively coupled plasma atomic emission spectroscopy (ICP-AES). The as-prepared core–shell nanoparticles show enhanced upconversion fluorescence and superior magnetic resonance (MR) relaxivity. In addition, the as-prepared β-NaGdF4:Yb,Er@CaF2 nanoparticles show enhanced chemical stability after surface functionalization and transfer into aqueous solutions.

A novel surface plasmon resonance (SPR) enhanced upconversion (UC) luminescence system was designed for the detection of α-fetoprotein (AFP) by using gold nanoparticle (NP) coated NaYF4:Yb,Er,Gd upconversion nanoparticles (UCNPs) as probes. The gold NP coated NaYF4:Yb,Er,Gd UCNPs had a strong emission at 665 nm and were modified with poly(acrylic acid) (PAA) before being conjugated with anti-AFP (Ab1). When the AFP antigens were added to the system, the binding affinity between AFP and Ab1 resulted in a reduced distance between the gold NP coated UCNPs. The UC luminescence intensity of this system was enhanced because of the SPR properties of the gold NPs. The sensing system exhibited a high sensitivity, good specificity, and wide linear range, 0.098–15.0 ng mL−1 for the detection of AFP (R = 0.99), with a detection limit as low as 0.095 ng mL−1 under optimal conditions. The method was successfully applied to AFP sensing in human serum samples.

Catalysts that effectively degrade organic pollutants and exhibit bactericidal properties are highly up-and-compromising for water treatment applications. To overcome the inherent limitation that the rapid recombination of the photogenerated carriers and to extend the practical utility of Bi 2 WO 6 , a flower-like structure of 7 wt.% Er 3+ -doped Bi 2 WO 6 (Er 7% -Bi 2 WO 6 ) capable of oxygen vacancy and crystal defect was synthesized using a straightforward hydrothermal method. Under visible light irradiation (λ > 420 nm), the Er 7% -Bi 2 WO 6 achieved a Rhodamine B (RhB) degradation efficiency of 92% within 80 min, significantly surpassing that of pristine Bi 2 WO 6 . The kinetic rate constant of Er 7% -Bi 2 WO 6 was determined to be 0.0288 min −1 , which is 5.9 times higher than the 0.0049 min −1 observed for Bi 2 WO 6 . Additionally, the bactericidal rate against Escherichia coli after 120 min of visible light exposure was 93.9%, nearly twice that of Bi 2 WO 6 at 49.8%. Density functional theory calculations and experimental results confirmed that doping with Er 3+ introduced lower band gap and more photogenerated carriers, enhanced visible light absorption, and ultimately improved the photocatalytic performance. Electron paramagnetic resonance and radical trapping experiments identified h⁺ and ·O 2 ⁻ as the primary active species generated during the photocatalytic process of Er 7% -Bi 2 WO 6 . The RhB removal rate remained above 90% after five degradation cycles, and the treatment efficacy on actual water samples was 76%. This study highlights the potential of Er-doped Bi 2 WO 6 to enhance both photocatalytic degradation of organic pollutants and bactericidal performance, thereby expanding the application scope of Bi 2 WO 6 in water treatment. Semiconductor photocatalytic technology is recognized as one of the effective technologies for using solar energy to solve current environmental problems. In this study, a Z-scheme heterojunction ZnFe 2 O 4 /Er-ZnO hollow nanotube composite was prepared by hydrothermal method. The results show that the degradation efficiency of tetracycline (TC) in ZnFe 2 O 4 is only 51 %, while the degradation efficiency of ZnFe 2 O 4 /2% Er-ZnO hollow tube composites is as high as 97 %. At the same time, ZnFe 2 O 4 /2% Er-ZnO has a larger specific surface area (specific surface area of 224.39 m 2 · g −1 ), which is 27 times that of ZnFe 2 O 4 (specific surface area of 8.30 m 2 ·g −1 ). Density functional theory (DFT) calculations and experimental analysis show that the doping of Er, the unique hollow structure and the formation of heterojunction make ZnFe 2 O 4 /2% Er-ZnO have a large specific surface area, efficient carrier separation ability and strong redox ability, so the Z-scheme heterojunction ZnFe 2 O 4 /2% Er-ZnO hollow tubular composites have strong degradation ability for tetracycline. In addition, the liquid chromatography-mass spectrometry (LC-MS) analysis technique was used to propose the possible degradation pathways of antibiotics, which provided ideas for the treatment of water pollution.

The ordered and stable structure of inorganic lattice is the basis of constructing high-efficiency rare earth activated fluorescence, solid-state laser or other optical materials. The mixed sensitized and activated rare earth ions usually keeps a uniform space-occupying and the energy transfer depends much on the doping concentration, while the aggregation of rare earth dopants in order is rare. In this work, a series of LuMgGaO 4 phosphors were synthesized and the detail lattice parameters are analyzed through Vietveld refinement and the new CIF file is provided. The site-occupancy of Er 3+ , Yb 3+ shows clustered features and the Er–Yb sublattice structure with various Yb/Er ratios were constructed on the basis of planar triangular lattice. The long-distance excitation energy migration to lattice or defects can be effectively inhibited in case of the hexagonal circle Er-6Yb sublattice, in which the most intense green/red UCL was obtained. All the results demonstrate the novelty of regulating ordered site-occupancy of rare earth ions on a compact planar triangular lattice with higher efficiency of energy migration, which will pave a way for developing phosphor-converted materials or laser materials with better performance.

Lanthanide upconversion luminescent materials featuring multi-peak and tunable emissions allow for ratiometric luminescence thermometry in a remote detection manner. However, wide explorations are still required to develop relevant materials for efficient thermometric performances. Herein, Yb 3+ /Er 3+ co-doped non-stoichiometric analogues of bismuth oxyfluoride were obtained by simply tuning the F − /(Bi 3+ + Ln 3+ ) molar ratio of the reactants in a solid-state reaction method, for the first time. We achieved simultaneous regulations in composition , structure, morphology, and upconversion luminescence properties of BiO x F y :Yb/Er (2/1 mol%) microcrystals . Compositional analysis revealed not only the predictable increase of fluorine content, but also the constant O/(Bi + Ln) atomic ratios in all nominal BiO x F y :Yb/Er. Experimental results showed the structure and morphology evolutions from hexagonal-phase micro-flowers, to tetragonal-phase microplates , and further to orthorhombic-phase irregular microcrystals . Benefiting from adjustable composition, structure and morphology, up-conversion luminescence with adjustable intensity and color was obtained. Temperature dependent upconversion properties and ratiometric thermometric performances based on the thermally coupled levels, 2 H 11/2 and 4 S 3/2 of Er 3+ , were explored. BiOF:Yb/Er, BiO 0.67 F 1.66 :Yb/Er, and BiF 3 :Yb/Er microcrystals were found as suitable temperature sensing candidates with operating temperature range as wide as 298−673/723 K, high relative sensitivity up to 1.24% K -1 , and minimum temperature uncertainty of 0.15 K. These results benefit not only further explorations for high-performance lanthanide-based luminescence thermometers, but also the chemistry of fluorides made from the solid-state reaction method.
